<dcterms:abstract>Galactomannans are polysaccharides commonly extracted from the endosperm of various leguminous seeds, exhibiting variations in their man/gal ratio based on their botanical origin. These gums are highly valued for their biodegradability and excellent stability. However, their applications have been limited due to their high viscosity and molecular weight. In this study, enzymatic and ultrasound depolymerisation of galactomannans (guar gum, locust bean gum, and unconventional Neltuma ruscifolia seed gum) was carried out, resulting in hydrolysates, which were subsequently freeze-dried and charac terised. The hydrolysis process leads to a decrease in viscosity and molecular weight, accompanied by a noticeable increase in polydispersity through enzymatic treatment. Thin-layer chromatography (TLC) analysis indicates the generation of at least eight-membered subunits. Fourier-transform infrared (FT-IR) spectroscopy analysis demonstrates no significant changes in functional groups following ultrasound hydrolysis. Furthermore, in agreement with TLC results, enzymatic hydrolysates display partial reactivity towards non-enzymatic browning (NEB), yielding mostly intermediate products with limited brown ing development. This study diversifies potential applications of these gums in different industries.</dcterms:abstract>

<dcterms:abstract>Herbivorous insects assess and choose their potential host plants based on traits that may correlate with host suitability or quality. These traits may operate as cues for better resources, noxious chemicals, or fewer competitors and enemies. Interac tions between insects and their host plants may also be modulated by the nutritional value of the plant, by plant chemical stimuli, or by physical traits such as trichomes. Diferences in chemical or physical cues among plants may convey informa tion about diet suitability. Several studies have addressed the topic of diferences in plant traits between host and non-host plants; fewer have addressed these diferences among potential host plants. We have studied the efect of chemical and physi cal plant traits on the feeding and oviposition preferences of the oligophagous herbivore Epilachna paenulata (Coleoptera: Coccinellidae) and two of its host plants, Cucurbita maxima and Cucurbita moschata (Cucurbitaceae). We frst typifed the volatile and non-volatile chemical profles and the trichome distribution of the plants. Then, using behavioral assays, we evaluated feeding and oviposition preferences of E. paenulata. Further, to assess a correlation between oviposition prefer ences and ofspring survival, we measured larval performance as indicated by survival, mass, and time to complete each instar. Female longevity in both Cucurbita host plants was also evaluated. Our results show that the congeneric plants bear diferences in their chemical profles, including volatile organic compounds, leaf waxes, and leaf parenchyma metabolites. Trichome abundance was also diferent, with C. maxima presenting fewer hairs. Epilachna paenulata females laid signif cantly more eggs in C. maxima, regardless of the plant they were raised as larvae. Female longevity on C. maxima was higher, but larval performance showed no diference between both plants. The proximate causes mediating female oviposition preferences may then involve trichome abundance, oviposition deterrents, toxic substances, or a combination of these factors.</dcterms:abstract>

<dcterms:abstract>Doping with atom impurities is currently an attractive mechanism to enhance the optical absorption capacity of TiO2 and other inorganic oxide semiconductors. In this work, the electronic and optical properties of carbondoped TiO2(B) (100) ultrathin surfaces were investigated using density functional theory (DFT) calculations, with the aim of evaluating their potential as photoelectrode for dye-sensitized solar cells (DSSCs). Carbon impurities were introduced onto oxygen sites located in the proximity of the surface and catechol was employed as a sensitizer model, focusing our study mainly on the evolution of the semiconductor’s electronic structure. The reduction of the band gap energy resulting from impurity states within the valence band of C-doped TiO2(B) was confirmed. Moreover, our findings suggest that C doping not only induces a stronger interaction between the catechol molecule and the surface but also, could increase the light absorption capacity of the material. Thus, carbon-doped TiO2(B) could act as a promising semiconductor for DSSCs and other photochemical systems.</dcterms:abstract>

<dcterms:abstract>Eucalyptus species are among the most planted trees in forestry production, an ever-increasing commercial activity worldwide. Forestry expansion demands a continuous search for preventive and sanitary measures against pests and diseases. Massive application of phytosanitary products is incompatible with the forestry sector, so forest health management must be based on other principles. In this context, studies on insect plant relationships mediated by plant metabolites may contribute information relevant to plant resistance and genotype selection. In this study, we analyzed the leaf metabolome of four Eucalyptus species commonly planted in southern South America, to correlate this chemical information with feeding preference of Thaumastocoris peregrinus (Hemiptera: Thaumastocoridae), an important pest of eucalypt plantations. Gas chromatography mass spectrometry analyses were performed on polar and non-polar leaf extracts from Eucalyptus globulus, Eucalyptus grandis, Eucalyptus robusta, and Eucalyptus tereticornis (Myrtaceae). Feeding preferences were assessed in two-choice laboratory bioassays resulting in a preference gradient of the four plant species. Moreover, a performance bioassay where we contrasted survival and development time between the most and least preferred plants, showed a clear correlation with preference both in survival and developmental time of the most susceptible nymph instar. We found that species with high or low feeding preferences differ significantly in several foliar metabolites, which may be acting as feeding stimulants or deterrents for T. peregrinus. These findings may provide useful criteria for choosing Eucalyptus genotypes when planting in bronze bug infested areas.</dcterms:abstract>

<dcterms:abstract>Objective: To evaluate the prevalence of health-related cues as part of the digital marketing of ultrapro cessed foods on Instagram among food companies in Uruguay. Methods: Cross-sectional exploratory study. All content posted by 118 Instagram accounts of companies promoting ultraprocessed foods in Uruguay over 6 months (from August 2020 to February 2021) was retrieved. The content of 1,893 Instagram posts was coded considering visual and textual cues conveying health-related associations. The number and percentage of posts, including cues within each category and theme, were calculated. Results: More than half of all posts contained at least 1 visual or textual cue conveying health-related asso ciations. Three main themes emerged: i) product composition, ii) healthy lifestyle, and iii) health and health benefits. The prevalence of health-related cues differed dramatically across product categories, ranging from 100% to 1.5%. Conclusions and Implications: Health-washing constitutes an integral part of the digital marketing of ultraprocessed foods on Instagram in Uruguay. The inclusion of health-related cues can potentially under mine public health efforts targeted at reducing consumption of these foods. These findings suggest that strict and comprehensive regulations on the digital marketing of such products are needed in policies pro moting healthy eating habits globally.</dcterms:abstract>

<dcterms:abstract>Increasing consumption of energy drinks has raised public health concerns worldwide. The present study intended to contribute to the literature on policies to discourage consumption of these beverages by exploring the effect of front-of-package caffeine warning labels on the purchase intention of young Uruguayan adults. A mixed methods approach was used to address the research objectives in an online survey with 225 participants aged between 18- and 30-years old. The quantitative component of the survey intended to evaluate the effect of caffeine warnings on purchase intention using choice-based conjoint analysis, whereas the qualitative component explored participants’ perceived efficacy of the warnings using an open-ended question. The conjoint task involved a series of choice sets composed of two cans differing in three 2-level variables: octagonal caffeine warning featuring the expression ’Contains caffeine’, octagonal sugar warning, and brand. Results from the conjoint analysis showed that caffeine warnings increased choice likelihood, as did the brand conveying energy related associations. On the contrary, sugar warnings reduced choice likelihood. Responses to the open-ended question provided insights on the lack of efficacy of caffeine warnings and stressed the importance of raising awareness of the potential negative health consequences of energy drink consumption. Taken together results from the present work suggest the importance of designing effective front-of-package warning labels to discourage consumption of energy drinks among young adults.</dcterms:abstract>

<dcterms:abstract>Tissue transglutaminase (TG2) expressed in monocytes and macrophage is known to participate in processes during either early and resolution stages of inflammation. The alternative splicing of tissue transglutaminase gene is a mechanism that increases its functional diversity. Four spliced variants are known with truncated C terminal domains (TGM2_v2, TGM2_v3, TGM2_v4a, TGM2_v4b) but scarce information is available about its expression in human monocyte and macrophages. We studied the expression of canonical TG2 (TGM2_v1) and its short spliced variants by RT-PCR during dif ferentiation of TPH-1 derived macrophages (dTHP-1) using two protocols (condition I and II) that differ in Phorbol-12-myristate-13-acetate dose and time schedule. The production of TNF-α and IL-1β in supernatant of dTHP-1, measured by ELISA in supernatants showed higher proinflammatory milieu in condition I. We found that the expression of all mRNA TG2 spliced variants were up-regulated during macrophage dif ferentiation and after IFN-γ treatment of dTHP-1 cells in both conditions. Nevertheless, the relative fold increase or TGM2_v3 in relation with TGM2_v1 was higher only with the condition I. M1/M2-like THP-1 macrophages obtained with IFN-γ/IL-4 treatments showed that the up-regulation of TGM2_v1 induced by IL-4 was higher in relation with any short spliced variants. The qualitative profile of relative contribution of spliced variants in M1/M2-like THP-1 cells showed a trend to higher expression of TGM2_v3 in the inflammatory functional phenotype. Our results contribute to the knowledge about TG2 spliced variants in the biology of monocyte/macrophage cells and show how the differentiation conditions can alter their expression and cell function.</dcterms:abstract>

<dcterms:abstract>BACKGROUND: The global beef market demands the meat industry to ensure product quality and safety in markets that are often very distant. The present study aimed to evaluate the effects of chilled (CH, 120 d) and chilled-then-frozen (CHF, 28 d + 92 d) storage conditions of beef vacuum packaged (VP) and vacuum packaged with antimicrobial (VPAM) on meat quality, oxidative status and microbial loads. Treatments resulted from the combination of storage condition and packaging type: VP + CH, VP + CHF, VPAM + CH and VPAM + CHF. RESULTS: Warner–Bratzler shear force values decreased in all treatments after 28 d of chilling. Except for VP + CH, L* values (lightness) of meat color did not differ in each treatment as the storage time increased. Meat from VP + CH had greater a* values than CHF treatments on day 120 of storage. A consumer panel did not detect differences in tenderness, flavor and overall liking between VP and VPAM beef, but they preferred CHF steaks rather than CH beef. TBARS values did not differ between VP and VPAM and between CH and CHF at any time during the storage period. At the end of storage time, all treatments except VP + CHF presented a greater concentration of thiols than at 48 h post-mortem. On day 120 of storage, VP + CH had greater cat alase enzyme activity than CHF treatments while VP + CH and VP + CHF showed a greater superoxide dismutase activity than VPAM + CHF. Storage condition (CH or CHF) had a greater impact on microbial counts than the type of packaging. CONCLUSION: Freezing meat after an ageing period represents a suitable strategy to extend beef storage life without a detri mental impact on its quality.</dcterms:abstract>

<dcterms:abstract>Prenylated flavanones are a family of compounds with an important biological potential. Previously, anti tyrosinase activity, acetylcholinesterase inhibitory activity, and neuroprotective effects of several prenyl flava nones isolated from different American Dalea genus species were reported. The biological potency of these kinds of compounds, together with the particularity of their chemical structures, encouraged us to investigate them for in vitro and in silico anti-xanthine oxidase activity. So, five prenyl-flavanones obtained from different Dalea sp (Dalea elegans, Dalea boliviana, and Dalea pazensis) were studied and the relationships between the structure of these prenyl-flavanones and their inhibitory activity were evaluated. Molecular docking studies were performed in order to propose the binding mode of the most active natural compound. 2′,4′-dihydroxy-5′-(1‴,1‴-dimethy lallyl)-8-prenylpinocembrin (1) was the most active in this series showing an IC50 of 0.26 ± 0.07 µM comparable with the reference inhibitor, allopurinol. The presence of 5,7,2′,4′-tetrahydroxy substitution, accompanied by a prenyl group at 8-position in the A-ring, and a 5′ (1‴,1‴-dimethylallyl) were important to present a xanthine oxidase inhibitory activity. This fact was confirmed with molecular docking studies showing relevant interactions of 1 with the residues of the catalytic site of xanthine oxidase, and a binding energy of − 7.3297 kcal mol− 1 . These results contributed not only to understanding the binding mode but also to validating the in vitro results. The obtained findings lead us to propose these prenyl-flavanones as lead compounds for the design and devel opment of novel xanthine oxidase inhibitors for the treatment of diseases in which this enzyme is involved.</dcterms:abstract>

<dcterms:abstract>In this study, several approaches were tested to optimise the production and recovery of the widely used anticancer drug Taxol® (paclitaxel) from culturable vascular stem cells (VSCs) of Taxus baccata, which is currently used as a suc‑ cessful cell line for paclitaxel production. An in situ product recovery (ISPR) technique was employed, which involved combining three commercial macro-porous resin beads (HP-20, XAD7HP and HP-2MG) with batch and semi-continu‑ ous cultivations of the T. baccata VSCs after adding methyl jasmonate (Me-JA) as an elicitor. The optimal resin combi‑ nation resulted in 234±23 mg of paclitaxel per kg of fresh-weight cells, indicating a 13-fold improved yield compared to the control (with no resins) in batch cultivation. This resin treatment was further studied to evaluate the resins’ removal capacity of reactive oxygen species (ROS), which can cause poor cell growth or reduce product synthesis. It was observed that the ISPR cultivations had fourfold less intracellular ROS concentration than that of the control; thus, a reduced ROS concentration established by the resin contributed to increased paclitaxel yield, contrary to previous studies. These paclitaxel yields are the highest reported to date using VSCs, and this scalable production method could be applied for a diverse range of similar compounds utilising plant cell culture.</dcterms:abstract>

<dcterms:abstract>Green Analytical Chemistry (GAC) metrics include a variety of criteria, such as the regent amounts and toxicity, energy consumption, generated waste, among others. The analytical greenness metric (AGREE) and its variant for sample preparation (AGREEprep) cover different aspects that contribute to the environmental sustainability of sample preparation. White Analytical Chemistry (WAC) considers not only environmental aspects but also analytical and practical aspects with a holistic vision based on a Red-Green-Blue color model. A case study is presented to assess the green and white profile of a method based on ultrasoundassisted extraction and determination of Mn and Fe in beef using microwave-induced plasma atomic emission spectroscopy (MP AES). The method was validated and resulted simple, fast without external heating using diluted acids. It was concluded that we should think in green sample preparation with the AGREEprep tool, as well as in white holistic assessments (WAC) as both constitute complementary tools.</dcterms:abstract>

<dcterms:abstract>This narrative review revises the scientific evidence of recent years on healthy eating in children and adolescents, making sense of promising avenues of action, from a food system perspective. A conceptual framework is provided to better understand how eating habits of children and adolescents are shaped to identify key multisectoral approaches that should be implemented to promote healthier diets. The following influencing factors are discussed: individual factors (physiological and psychological factors, food preferences and food literacy competencies), factors within the personal and socio-cultural food environments, external food environments, and the supply chain. In each section, the main barriers to healthy eating are briefly discussed focussing on how to overcome them. Finally, a discussion with recommendations of actions is provided, anchored in scientific knowledge, and transferable to the general public, industry, and policymakers. We highlight that multidisciplinary approaches are not enough, a systems approach, with a truly holistic view, is needed. Apart from introducing systemic changes, a variety of interventions can be implemented at different levels to foster healthier diets in children through fostering healthier and more sustainable food environments, facilitating pleasurable sensory experiences, increasing their food literacy, and enhancing their agency by empowering them to make better food related decisions. Acknowledging children as unique individuals is required, through interpersonal interactions, as well as their role in their environments. Actions should aim to enable children and adolescents as active participants within sustainable food systems, to support healthier dietary behaviours that can be sustained throughout life, impacting health at a societal level.</dcterms:abstract>

<dcterms:abstract>The reduction of graphene oxide (GO) by means of thermal and/or chemical treatments leads to the production of reduced graphene oxide (rGO)—a material with improved electrical conductivity and considered a viable and low-cost alternative to pure graphene in several applications, including the production of supercapacitor electrodes. In the present work, GO was prepared by the oxidation of graphite recycled from spent Li-ion batteries using mixtures of sulfuric and phosphoric acids (with different H2SO4/H3PO4 ratios), leading to the production of materials with significant S and P contents. These materials were then thermally reduced, resulting in rGO papers that were investigated by solid-state 13C and 31P nuclear magnetic resonance, along with other methods. The electrochemical properties of the produced rGO papers were evaluated, including the recording of cyclic voltammetry and galvanostatic charge–discharge curves, besides electrochemical impedance spectroscopy analyses. The samples obtained by thermal reduction at 150 ◦C exhibited good rate capability at high current density and high capacitance retention after a large number of charge– discharge cycles. The results evidenced a strong relationship between the electrochemical properties of the produced materials and their chemical and structural features, especially for the samples containing both S and P elements. The methods described in this work represent, then, a facile and low-cost alternative for the production of rGO papers using graphite recycled from spent batteries, with promising applications as supercapacitor electrodes.</dcterms:abstract>

<dcterms:abstract>The aim of the present work was to explore children’s reaction to sugar reduction in the context of the imple mentation of nutritional warnings in Uruguay. The study was composed of two sessions involving three evalu ation conditions: tasting without package information, package evaluation without tasting, and tasting with package information. A total of 122 children, ages ranging between 6 and 13 years old (47% girls) were involved in the study. In the first session, children’s hedonic and emotional response to a regular and a sugar-reduced chocolate dairy dessert (without other sweeteners) was evaluated. In the second session, children first evalu ated their expected liking, emotional associations and choice of packages differing in the presence of warning labels for excessive sugar content and cartoon character (2x2 design). Finally, they tasted the selected sample in the presence of the package and evaluated their liking, emotional associations, and intention to re-taste it. Although sugar reduction led to a significant reduction in overall liking, the dessert with 40% sugar reduction showed a mean overall score of 6.5 in a 9-point hedonic scale and was described using positive emoji ( , , ). When the desserts were tasted with package information, no significant differences in the expected overall liking of the regular and sugar-reduced dessert were found. Regarding the effect of packaging elements, the presence of a warning label highlighting excessive content of sugar did not have a significant effect on children’s choice. Instead, children’s choices were defined by the presence of a cartoon character. Results from the present work provide additional evidence about the feasibility of reducing the sugar content and sweetness of dairy products targeted at children and stress the need to regulate the use of cartoon characters on products with an unfavorable nutritional profile.Methodological recommendations for sensory and consumer research with children are also discussed.</dcterms:abstract>

<dcterms:abstract>Oxydiacetate, iminodiacetate, and thiodiacetate are flexible dicarboxilates which have been used as chelating agents towards d-metal ions for decades. They differ among each other only in the central connecting group X: O, NH, or S. The interaction of these ligands with lanthanide ions has also received significant attention in recent years. This review deals with various aspects of the coordination chemistry of Ln ions with these three ligands. It includes a critical analysis of the stability studies carried out in solution, shedding light on the influence of X in the thermodynamic stability of the resulting complexes. In the solid state, the discovery of new materials by combination of the diverse coordination modes offered by the two carboxylate groups (or the protonated forms of them) with the fluxional geometry of the central hinge X, is explored. Luminescent properties both in solution and in the solid state are also discussed, again with regard to the coordination abilities of these ligands. Finally, the most relevant applications are discussed, including the design of white-light emitters and other optic ma terials and the preparation of MOFs with one-dimensional channels that are currently assayed as catalysts in several organic processes.</dcterms:abstract>

<dcterms:abstract>The serious widespread development of nematode resistance has motivated the use of combined anthelmintic formulations. However, the advantages/disadvantages of the combined use of anthelmintics require further scientific characterization. The goals of the current trial were a) to characterize the pharmacokinetics of closantel (CLO) and moxidectin (MXD) administered both subcutaneously (sc) and orally either separately or co administered (CLO + MXD) to lambs; b) to compare the nematodicidal activity of both molecules given indi vidually or co-administered to lambs infected with resistant nematodes. Seventy (70) Corriedale lambs naturally infected with multiple resistant gastrointestinal nematodes were involved in the pharmacokinetic and efficacy trials. The animals were allocated into six groups (n = 10) and treated with either CLO, MXD, or with the CLO + MXD combined formulation by both the oral and sc routes. Additionally, an untreated control group (n = 10) was included for the efficacy trial. The efficacy was estimated by the faecal egg count reduction test (FECRT). Higher systemic exposure of both CLO and MXD was observed after the sc compared to the oral administration in lambs. The combined administration of CLO + MXD did not markedly alter their disposition kinetics. At 13 days post treatment, the administration of both molecules as a single active principle reached efficacy levels ranging be tween 80% (MXDoral), 84% (CLOoral), 85% (CLOsc), and 92% (MXDsc). The combined oral and sc treatments reached 99% efficacy. No adverse effects were observed after the combined treatment of CLO + MXD, and their co-administration did not show any adverse pharmacokinetic interaction. The combined effect of CLO + MXD successfully restored the maximum efficacy levels, which were not reached by the individual active ingredients.</dcterms:abstract>

<dcterms:abstract>One hundred twenty-five yeast strains isolated from table grapes and apples were evaluated for the control Botrytis cinerea of in vitro and in vivo. Ten strains were selected for their ability to inhibit mycelial growth of B. cinerea in vitro. In the in vivo assays, these yeasts were tested at 20 ◦C on ‘Thompson Seedless’ berries for 7 days; only three were selected (m11, me99 and ca80) because they significantly reduced the incidence of gray mold. These three yeast strains were then evaluated at different concentrations (1 × 107 , 1 × 108 and 1 × 109 cells mL−1 ) on ‘Thompson Seedless’ grape berries at 20 ◦C. The strains m11, me99 and ca80 reduced the incidence of B. cinerea to 11.9, 26.1 and 32.1%, respectively, when the berries were submerged in a yeast suspension at a concentration of 1 × 109 cells mL−1 24 h before inoculation with B. cinerea. The most favorable pH for antifungal activity was 4.6 in the three isolates. The three yeast strains secreted the hydrolytic enzymes chitinase and β-1-glucanase, and two strains (me99 and ca80) produced siderophores. The three yeast strains exhibited low oxidative stress tolerance and only strain m11 had the ability to produce biofilms. The strains were identified using 5.8S-ITS rDNA PCR-RFLP and correspond to the Meyerozyma guilliermondii (m11) and Aureobasidium pullulans (me99 and ca80) species.</dcterms:abstract>

<dcterms:abstract>Objective: To study the quality of potato chips for sale in the Uruguayan market. Theoretical framework: For potato chips, the regulation controls moisture, peroxide value (VP) and acidity (%A). It was also considered important to evaluate the fatty material contained in the potatoes as frying oil, where polar compounds (PC) are controlled. The composition of CPs and total polymers (TP) was also evaluated. This set of measures took into account all the deterioration processes to which these lipids are subjected, so as to have a complete idea of their quality. Method: Six different brands of potato chips for sale in the Uruguayan market and the oil absorbed during their elaboration were evaluated. The lipid content and moisture content of the potatoes were measured. In the oil, VP, %A, CP and its composition, PT, the composition in fatty acids and the content of cabbage were measured. Results and conclusion: All the products analyzed comply with the current regulations, except for sample E, which presented high %A. Nevertheless, it was observed that in some cases the oils have an advanced deterioration. Likewise, the sample obtained through vacuum frying with a significantly lower lipid content than the others was highlighted. Implications of the research: This paper seeks to inform the quality of these foods that are for sale in the market. Originality/Value: The quality of food has a direct influence on the health of the population.</dcterms:abstract>

<dcterms:abstract>Antigen tests have been crucial for managing the COVID-19 pandemic by identifying individuals infected with SARS-CoV-2. This remains true even after immunity has been widely attained through natural infection and vaccination, since it only provides moderate protection against transmission and is highly permeable to the emergence of new virus variants. For this reason, the widespread availability of diagnostic methods is essential for health systems to manage outbreaks effectively. In this work, we generated nanobodies to the virus nucleocapsid protein (NP) and after an affinity-guided selection identified a nanobody pair that allowed the detection of NP at sub-ng/mL levels in a colorimetric two-site ELISA, demonstrating high diagnostic value with clinical samples. We further modified the assay by using a nanobody-NanoLuc luciferase chimeric tracer, resulting in increased sensitivity (detection limit = 61 pg/mL) and remarkable improvement in diagnostic performance. The luminescent assay was finally evaluated using 115 nasopharyngeal swab samples. Receiver Operating Characteristic (ROC) curve analysis revealed a sensitivity of 78.7% (95% confidence interval: 64.3%-89.3%) and specificity of 100.0% (95% confidence interval: 94.7%-100.0%). The test allows the parallel analysis of a large number of untreated samples, and fulfills our goal of producing a recombinant reagent-based test that can be reproduced at low cost by other laboratories with recombinant expression capabilities, aiding to build diagnostic capacity.</dcterms:abstract>

<dcterms:abstract>Introduction: The identification of chemical compounds that interfere with SARS CoV-2 replication continues to be a priority in several academic and pharmaceutical laboratories. Computational tools and approaches have the power to integrate, process and analyze multiple data in a short time. However, these initiatives may yield unrealistic results if the applied models are not inferred from reliable data and the resulting predictions are not confirmed by experimental evidence. Methods: We undertook a drug discovery campaign against the essential major protease (MPro) from SARS-CoV-2, which relied on an in silico search strategy –performed in a large and diverse chemolibrary– complemented by experimental validation. The computational method comprises a recently reported ligand-based approach developed upon refinement/learning cycles, and structure-based approximations. Search models were applied to both retrospective (in silico) and prospective (experimentally confirmed) screening. Results: The first generation of ligand-based models were fed by data, which to a great extent, had not been published in peer-reviewed articles. The first screening campaign performed with 188 compounds (46 in silico hits and 100 analogues, and 40 unrelated compounds: flavonols and pyrazoles) yielded three hits against MPro (IC50 ≤ 25 μM): two analogues of in silico hits (one glycoside and one benzo-thiazol) and one flavonol. A second generation of ligand-based models was developed based on this negative information and newly published peer-reviewed data for MPro inhibitors. This led to 43 new hit candidates belonging to different chemical families. From 45 compounds (28 in silico hits and 17 related analogues) tested in the second screening campaign, eight inhibited MPro with IC50 = 0.12–20 μM and five of them also impaired the proliferation of SARS-CoV-2 in Vero cells (EC50 7–45 μM). Discussion: Our study provides an example of a virtuous loop between computational and experimental approaches applied to target-focused drug discovery against a major and global pathogen, reaffirming the well-known “garbage in, garbage out” machine learning principle.</dcterms:abstract>

<dcterms:abstract>Global warming has a strong impact on the polar regions, in particular, the Antarctic Peninsula and nearby islands. Methane (CH4) is a major factor in climate change and mitigation of CH4 emissions can be accomplished through microbial oxidation by methanotrophic bacteria. Understanding this biological process is crucial given the shortage of research carried out in this geographical area. The aim of this study was to characterise psychrophilic enrichment cultures of aerobic methanotrophs obtained from lake sediments of the Fildes Peninsula (King George Island, South Shetland Islands) and revealing the distribution of the genus Methylobacter in different lake sediments of the peninsula. Four stable methanotrophic enrichment cultures were obtained and analysed by metagenome-assembled genomes (MAGs). The phylogeny of methanotroph MAGs recovered from these enrichment cultures based on the 16S rRNA gene showed that K-2018 MAG008 and D1-2020 MAG004Ts clustered within the Methylobacter clade 2, with high similarity to Methylobacter tundripaludum SV96T (97.88 and 98.56% respectively). However, the average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values with M. tundripaludum were < 95% (84.8 and 85.0%, respectively) and < 70% (30.2 and 30.3%, respectively), suggesting that they represent a putative novel species for which the name ‘Ca. Methylobacter titanis’ is proposed. This is the first species of clade 2 of the genus Methylobacter obtained from Antarctica. The bacterial diversity assessed by 16S rRNA gene sequencing of 21 samples of different lakes (water column and sediments) revealed 54 ASVs associated with methanotrophs and the genus Methylobacter as the most abundant. These results suggest that aerobic methanotrophs belonging to the Methylobacter clade 2 would be the main responsible for CH4 oxidation in these sediments.</dcterms:abstract>
