A comprehensive study of the different classes of cycloaddition reactions ([3+2], [2+2], and [2+1]) of SO2 to acetylene and ethylene has been performed using density functional theory (DFT) and composite wavefunction methods. The [3+2] cycloaddition reaction, that was previously explored in the context of the cycloaddition of thioformaldehyde S-methylide (TSM) to ethylene and acetylene, proceeds in a concerted way to the formation of stable heterocycles. In this paper, we extend our study to the [2+2] and [2+1] cycloadditions of SO2 to acetylene, which would produce 1,1-oxathiete-2-oxide and thiirene-1,1-dioxide, respectively. One of the main conclusions is that cyclic 1,1-oxathiete-2-oxide can open through a relatively easy breaking of the S O single bond and rearrange toward sulfinyl acetaldehyde (SA). The SA molecule can easily undergo several internal rearrangements, which eventually lead to sulfenic acid and sulfoxide derivatives of ethenone, 1,2,3-dioxathiole, and CO plus sulfinylmethane. The most probable path, however, produces 2-thioxoacetic acid, whose derivatives (or those of the corresponding acetate) are usually obtained by Willgerodt–Kindler-type sulfuration of acetates. This product can in turn decompose, leading to the final products CO2 and H2C S. Comparison of this decomposition path with that of 2-amino-2-thioxoacetic acid shows that the process occurs through different H-transfer processes. We have studied the reduction reactions of two cytosolic human peroxiredoxins (Prx) in their disulfide form by three thioredoxins (Trx; two human and one bacterial), with the aim of better understanding the rate and mechanism of those reactions, and their relevance in the context of the catalytic cycle of Prx. We have developed a new methodology based on stoppedflow and intrinsic fluorescence to study the bimolecular reactions, and found rate constants in the range of 105–106 M 1 s 1 in all cases, showing that there is no marked kinetic preference for the expected Trx partner. By combining experimental findings and molecular dynamics studies, we found that the reactivity of the nucleophilic cysteine (CN) in the Trx is greatly affected by the formation of the Prx–Trx complex. The protein–protein interaction forces the CN thiolate into an unfavorable hydrophobic microenvironment that reduces its hydration and results in a remarkable acceleration of the thiol-disulfide exchange reactions by more than three orders of magnitude and also produces a measurable shift in the pKa of the CN. This mechanism of activation of the thiol disulfide exchange may help understand the reduction of Prx by alternative reductants involved in redox signaling. This study investigated the in vitro bioaccessibility of aluminum, copper, iron, manganese, lead, selenium, and zinc in three important species of farmed insects: the yellow mealworm (Tenebrio molitor), the house cricket (Acheta domesticus) and the migratory locust (Locusta migratoria). Results show that all three insect species constitute excellent sources of essential elements (Fe, Cu and Zn) for the human diet, contributing to the recommended dietary allowance, i.e., 10%, 50%, and 92%, respectively. A higher accumulation of Se (≥1.4 mg Se/ kg) was observed with increasing exposure concentration in A. domesticus, showing the possibility of using insects as a supplements for this element. The presence of Al and Fe nanoparticles was confirmed in all three species using single particle-inductively coupled plasma-mass spectrometry and transmission electron microscopy. The results also indicate that Fe bioaccessibility declines with increasing Fe-nanoparticle concentration. These findings contribute to increase the nutritional and toxicological insights of farmed insects. Among them, CD5 and CD6 are lymphocyte scavenger receptors known to interact with different microbial-associated molecular patterns, and the administration of the recombinant soluble ectodomains of human CD5 (rshCD5) and/or CD6 (rshCD6) has shown therapeutic/prophylactic potential in experimental models of fungal, bacterial and echinococcal infections. The latter is a zoonosis caused by the larval stage of the cestode parasite Echinococcus granulosus sensu lato, which in humans can induce secondary cystic echinococcosis (CE) after the spillage of protoscoleces contained within fertile cysts, either spontaneously or during surgical removal of primary hydatid cysts. Herein, we have analysed the mechanisms behind the significant protection observed in the mouse model of secondary CE following prophylactic administration of rshCD5 or rshCD6. Our results show that both molecules exhibit intrinsic antiparasitic activities in vitro, as well as immunomodulatory functions during early secondary CE, mainly through Th1/Th17 cytokine bias and promotion of peritoneal polyreactive antibodies. These data support the relevance of the parasite components bound by rshCD5 and rshCD6, as well as the potential of their prophylactic administration as a useful strategy to reduce secondary CE in patients. Herein, using density functional calculations, we studied the substitutional doping in germanene with B, C, N, O, Al, Si, P, S, Ga, As, and Se. An Argentinean and high molecular weight (MW) chitosan (AC) was used and contrasted with a commercial type one (CC). FTIR and XRD were applied for the polysaccharide-vitamin dried mixed systems characterization. Shifts in bands of FA and AC or CC were observed in the mixed systems, confirming the interaction among the components, for example, the bands at 1382 and 1391 cm− 1 (AC and FA, respectively) shifted to 1387 cm–1 in the in AC-FA mixed systems. XRD data showed that the FA is in its protonated form as expected and is maintained in the different mixed systems obtained using either with CC or AC, demonstrating the robustness and applicability of the material to properly conserve the active ingredient. The ultrastructure and topography were studied by SEM and AFM, respectively. FA incrustations in a self-associated form were observed in the topography images. Variations in topography observed for the AC-FA and CC-FA mixed systems could be attributed to specific interactions between the components and the molecular features of each CS (i.e. the mean particle size was 63 ± 3 and 9 ± 1 nm and, roughness gave values of 0.47 and 0.41 nm, respectively). Vitamin bioaccessibility remained unchanged compared to the control (88.36% ±10.35%), being equal to 91.08% ± 5.02% for AC-FA and 71.56% ±14.08% for CC-FA, after submitted samples to an in vitro digestion process. In sum, mixed AC-FA systems obtained are promising materials for a possible additional application in coating matrices inspired by biopolymers to improve food preservation, maintaining the bioaccessibility of vitamins. Context Carbonyl compounds, especiallyaldehydes, emitted to the atmosphere, may suffer hydration in aerosols or water droplets in clouds. At the same time, they can react with hydroxyl radicals which may add or abstract hydrogen atoms from these species. The interplay between hydration and hydrogen abstraction is studied using density functional and quantum composite theoretical methods, both in the gas phase and in simulated bulk water. The H-abstraction from the aldehydic and geminal diol forms of formaldehyde, acetaldehyde, glycolaldehyde, glyoxal, methylglyoxal, and acrolein is studied to determine whether the substituent has any noticeable effect in the preference for the abstraction of one form or another. It is found that abstraction of the H-atom adjacent to the carbonyl group gives a more stable radical than same abstraction from the geminal diol in the case of formaldehyde, acetaldehyde, and glycolaldehyde. The presence of a delocalizing group in the Cα (a carbonyl group in glyoxal and methylglyoxal, and a vinyl group in acrolein), reverts this trend, and now the abstraction of the H-atom from the geminal diol gives more stable radicals. A further study was conducted abstracting hydrogen atoms from the other different positions in the species considered, both in the aldehydic and geminal diol forms. Only in the case of glycolaldehyde, the radical formed by H-abstraction from the –CH2OH group is more stable than any of the other radical species. Abstraction of the hydrogen atom in one of the hydroxyl groups in the geminal diol is equivalent to the addition of the •OH radical to the aldehyde. It leads, in some cases, to decomposition into a smaller radical and a neutral molecule. In these cases, some interesting theoretical differences are observed between the results in gas phase and (simulated) bulk solvent, as well as with respect to the method of calculation chosen. Methods DFT (M06-2X, B2PLYP, PW6B95), CCSD(T), and composite (CBS-QB3, jun-ChS, SCVECV-f12) methods using Dunning basis sets and extrapolation to the CBS limit were used to study the energetics of closed shell aldehydes in their keto and geminal-diol forms, as well as the radical derived from them by hydrogen abstraction. The Zn(II) compound is heteronuclear and the complex adopts a 1D cationic polymeric structure with zinc and sodium ions connected by two Hmidp– anions. The two other compounds are dinuclear, [Cu2(Hmidp)2(μ-H2O)2(H2O)2](CF3SO3)2⋅6H2O (4) and ([Ni2(midp)2(H2O)4] (5). Additionally, a Co(II) and a Cu(II) coordination compound were obtained with the same ligand and characterized by IR spectroscopy, thermal analysis and elemental analysis including metal quantification. For the copper compound the formula [Cu(Hmidp)](ClO4)⋅3H2O (7) is proposed while for the Co (II) compound (8) the proposed formula is [Co(midp)(H2O)2]. Although a veterinary vaccine exhibiting high levels of antibody-mediated protection has successfully reached the market, the large genetic diversity among parasite isolates and their particular host preferences, makes still necessary the search for novel vaccine candidates. Glutathione transferases (GSTs) constitute attractive targets for immunoprophylaxis due to their outstanding relevance in helminth detoxification processes, against both exogenous and endogenous stressors. Among the six GSTs known to be expressed in E. granulosus s.l., EgGST1 (Mu-class), EgGST2 (Sigma-class), and EgGST3 (a still non-classifiable isoenzyme), show the highest proteomic expression. Therefore, their recombinant forms -rEgGST1, rEgGST2 and rEgGST3- were herein analyzed regarding their potential to induce long-term antiparasite protection in mice. Only immunization with rEgGST1 induced long-lasting protection; and accordingly, rEgGST1-specific antibodies enhanced the parasite killing through both the classical activation of the host complement system and the antibody-dependent cellular cytotoxicity by macrophages. These results support further testing of rEgGST1 as a vaccine candidate in diverse hosts due to the broad expression of EgGST1 in different parasite stages and tissues. The growing evidence on the negative health effects of ultra-processed products indicates the need to develop strategies to discourage their consumption among infants and children. The development of such strategies requires a deeper understanding of the factors underlying children’s consumption of ultra-processed products. In this context, the present work aimed to analyze factors underlying mothers’ decision to offer meals composed of ultra-processed products during early childhood (6 months to 5 years old). An online survey with 888 mothers was conducted. Participants were shown a series of pictures of ultra-processed meals and were asked to indicate whether they would offer them to their children or not. A follow-up open-ended question was used to explore reasons underlying negative responses. A relevant proportion of mothers indicated that they would offer the ultra-processed meals to their children (48.5 % to 64.5 %). On the other hand, motives for not offering ultraprocessed meals to children were mainly related to food processing and nutritional composition. Several variables of the child (age, food fussiness), the mother (education level and socio-economic status), and the household (presence of children older than 5 years) had a significant effect on the likelihood of mothers offering ultra-processed meals and mentioning different reasons for not doing so. Results stress the need to implement strategies to increase perceived risk of ultra-processed products, particularly among parents with low educational level and from low socio-economic status. Recent empirical evidence suggests that gut dysbiosis is one of the negative health outcomes potentially associated with chronic consumption of some food additives. In this context, the present study aimed at analyzing the disclosure of food additives associated with gut dysbiosis in the labels of products commercialized in the Uruguayan market. A cross-sectional survey of packaged products commercialized in nine supermarkets was conducted between August and September 2021. All packaged processed and ultra-processed products available in each data collection site were surveyed using a cellphone app. The information available on the labels was manually extracted and the disclosure of food additives was analyzed using a computer assisted approach. Results showed that 38.1% of the products disclosed at least one food additive associated with gut dysbiosis. Disclosure was most frequent in ice-cream and popsicles, beverages, meat products and analogues, desserts, and fats, oils and fat and oil emulsions. Potassium sorbate was the individual additive associated with gut dysbiosis most frequently disclosed on the labels, followed by mono- and di-glycerides of fatty acid, sucralose, carboxymethylcellulose, acesulphame potassium, carrageenan, and sodium benzoate. These food additives frequently cooccurred and network analysis enabled the identification of patterns of co-occurrence. Taken together, results from the present work suggest the need to conduct additional research to assess the intake of food additives associated with gut dysbiosis at the population level, as well as to evaluate potential synergistic effects of food additives. To study the effect of acyl group donor and water, different chain length of saturated fatty acids (ranging from 6:0 to 18:0) and water content (ranging from 0, 1, 2 or 3 molar ratio of water/ChCl) were tested in the lipase-catalyzed esterification with n-butanol in ChCl:urea (1:2, molar ratio). When the incubation was performed in the absence of water, the shorter the fatty acid chain length, the higher the conversion. Upon water addition, a drastic increase in the esterification rate of all the fatty acids was observed and, conversely to what occurred in the anhydrous media, the longer the fatty acid chain length, the higher the conversion. Results suggest that water concentration in the DES medium possesses a multifactorial effect, as it influences the enzyme activity, the solvent viscosity, the DES structure, and the interaction between DES components, substrates, and enzymes, thus determining the efficiency and yield of the esterification reaction. Backgrounds: Sleep restriction is considered a stressful condition itself, causing a wide variety of physiological alterations, from cognitive and hormonal to immunological status. In addition, it is established that stress in mother rats can modify milk ejection, milk composition, and maternal care of the pups. Also, sleep disturbances during the early stages of motherhood are a common feature of all studied species. In this context, while the impacts of sleep disruption in non-lactating animals were extensively investigated, its repercussions during the initial phases of motherhood have been poorly explored. Therefore, we wonder if maternal behavior, milk ejection and its macronutrient composition would be disrupted when mother rats are subjected to an additional acute or chronic sleep restriction to the already existing sleep disturbances. Methods: Lactating rats were implanted with unilateral electrodes for polysomnographic recordings and for deep brain electrical stimulation into mesopontine waking-promoting area (for sleep deprivation). During the early postpartum period (postpartum day 5–9), mother rats were randomly assigned into one of three groups: chronic sleep restriction group (CSR; 6 h of sleep deprivation/day for five consecutive days), acute sleep restriction group (ASR; 6 h of sleep deprivation only for one day), or undisturbed group (control group). Active maternal behaviors (retrievals of the pups into the nest, mouthing, lickings [corporal and anogenital] and sniffing the pups) and passive maternal behaviors (kyphotic and supine nursing postures) were evaluated during a 30 min period without sleep restriction immediately after the sleep restriction or control period. The litter weight gain was assessed every day, and on the last experimental session mothers were milked for posterior macronutrients analysis (protein, carbohydrates and fat).Results: When compared to control group, CSR decreased the amount of milk ejected in the middle days of the sleep restriction period, while ASR did not affect this parameter. Moreover, ASR reduced milk protein content compared to control and CSR groups. Finally, compared to the control group, CSR reduced active maternal behaviors towards the end of the treatment days. Conclusions: We demonstrated that not only acute but also chronic sleep restriction impacts on the postpartum period, each one affecting different aspects of maternal behavior and lactation. Our results suggest the existence of a homeostatic recovery mechanism in breastfeeding during CSR, possibly ensuring the survival of the litter, while the decline in active maternal behaviors appears to be cumulative. An in-depth understanding of how citizens make their food choices is necessary to develop targeted strategies to promote healthier dietary habits. However, studies with vulnerable populations in non-WEIRD (Western- Educated-Industrialized-Rich-Democratic) societies are still scarce. In this context, the present study aimed at exploring the food purchase decisions of adults in charge of children and adolescents living in households experiencing food insecurity in Uruguay, an emerging Latin American country. Semi-structured interviews were conducted with beneficiaries of food assistance programs from both the state and civil society in four Uruguayan cities. The transcripts were analyzed using content analysis based on inductive coding. Three main themes related to food purchase decisions were identified: frequency of food purchasing, food purchase locations, and motives underlying food choice. Income was identified as the key determinant of food choice. Participants reported experiencing challenges for managing their limited household budget, struggling to purchase food throughout the month. They relied on an intricate set of multiple strategies to maximize the quantity of foods they can purchase with their limited household budget. Despite extreme focus on the immediate lack of money, food purchase decisions seemed to be highly rational, instead of driven by automatic or impulsive processes. Results have relevant implications for the development of strategies to promote healthier eating patterns among socioeconomically vulnerable sectors of the population in Uruguay and similar settings. The IGP-InN was characterized via density functional theory (DFT) simulations. The phonon dispersion shows the dynamic stability of IGP-InN, and molecular dynamics simulations confirm its thermal stability up to 700 K. Besides the electronic properties, the indirect band gap transition with energy (Egap) of 2.49 eV makes IGP-InN suitable for optoelectronic applications under UV–visible. Also, the Egap tunability with mechanical strain was analyzed, with a decrease of 1.19 eV in the Egap for tensile strains. The structural buckling plays an important role, with a transition to a planar structure for tensile strains. This work reveals a new class of 2D materials, buckled inorganic graphenylenes, and provides valuable insights into designing and optimizing graphenylene-like materials. This computational study investigated germanene nanotubes (GeNTs) using density functional theory (DFT) simulations to comprehend their structural, mechanical, and electronic properties. The analysis includes armchairs and zigzag GeNTs with diameters from ~7 to ~221 Å. It explores their relative stabilities, band structures, density of states, effective mass carriers, and piezoelectric and elastic constants. As a result, it is highlighted that smaller nanotube diameters exhibit higher instability than larger ones due to increased structural strain produced by higher curvature. An intriguing behavior is observed for diameters greater than 25 Å, with negative strain energies. The quantum confinement effect significantly influences the electronic properties of GeNTs, leading to increased band gap energy for smaller nanotubes. The band gap energy of armchair nanotubes has a decreasing behavior as the diameter increases. In contrast, for zigzag nanotubes, the band gap energy increases to ~13 Å diameter and then decreases, reaching a band gap energy of 0.08 eV. Regarding mechanical properties, smaller GeNTs exhibit significantly lower elastic constants (C11). As the diameter increases, C11 values converge around 114 GPa for both armchair and zigzag GeNTs. Only the zigzag GeNTs exhibit piezoelectricity. Carrier effective masses decrease with increasing diameter, enhancing carrier mobility for larger-diameter GeNTs. The hereinreported findings provide valuable insights into the properties of germanene nanotubes, demonstrating their promising potential for nanoscale device applications and inspiring further research and synthesis of germanenerelated systems. Alzheimer’s disease (AD) involves a neurodegenerative process that has not yet been prevented, reversed, or stopped. Continuing with the search for natural pharmacological treatments, flavonoids are a family of compounds with proven neuroprotective effects and multi-targeting behavior. The American genus Dalea L. (Fabaceae) is an important source of bioactive flavonoids. In this opportunity, we tested the neuroprotective potential of three prenylated flavanones isolated from Dalea species in a new in vitro pre-clinical AD model previously developed by us. Our approach consisted in exposing neural cells to conditioned media (3xTg-AD ACM) from neurotoxic astrocytes derived from hippocampi and cortices of old 3xTg-AD mice, mimicking a local neurodegenerative microenvironment. Flavanone 1 and 3 showed a neuroprotective effect against 3xTg-AD ACM, being 1 more active than 3. The structural requirements to afford neuroprotective activity in this model are a 5’-dimethylallyl and 4’-hydroxy at the B ring. In order to search the mechanistic performance of the most active flavanone, we focus on the flavonoid-mediated regulation of GSK-3β-mediated tau phosphorylation previously reported. Flavanone 1 treatment decreased the rise of hyperphosphorylated tau protein neuronal levels induced after 3xTg- AD ACM exposure and inhibited the activity of GSK-3β. Finally, direct exposure of these neurotoxic 3xTg-AD astrocytes to flavanone 1 resulted in toxicity to these cells and reduced the neurotoxicity of 3xTg-AD ACM as well. Our results allow us to present compound 1 as a natural prenylated flavanone that could be used as a precursor to development and design of future drug therapies for AD.
