A summary of the resource.
Two multiple response optimization methodologies were applied and compared to obtain an optimum combination medium formulation for culturing the yeast Rhodotorula graminis: (1) desirability function, and (2) pareto-optimal front by means of genetic algorithms (POF-GA). The concentration of microbial lipids, a raw material for the production of biodiesel, was modelled by the response surface methodology and then maximized through the application of a methodology based in genetic algorithms. The results obtained by applying POF-GA (one of the first times this approach was applied to a microbiological process) were confirmed by using the well-known Derringer's desirability function and by carrying out independent experiments. Then, it was verified that POF-GA is a viable option to be applied in biotechnological processes and that molasses may be employed as a source for culture media employed to produce microbial lipids, representing important economical savings since molasses is a raw material from sugarcane industrialization.

Polyamides-based compounds related to the Streptomycetal distamycin and netropsin are potent cytostatic molecules that bind to AT-rich regions of the minor groove of the DNA, hence interfering with DNA replication and transcription. Recently, derivatives belonging to this scaffold have been reported to halt the proliferation of deadly African trypanosomes by different and unrelated mechanisms. Here we describe the synthesis and preliminary characterization of the anti-trypanosomal mode of action of new potent and selective distamycin analogues. Two tri-heterocyclic derivatives containing a central N-methyl pyrrole ring (16 and 17) displayed high activity (EC50 < 20 nM) and selectivity (selectivity index >5000 with respect to mammalian macrophages) against the infective form of T. brucei. Both compounds caused cell cycle arrest by blocking the replication of the mitochondrial DNA but without affecting its integrity. This mode of action clearly differs from that reported for classical minor groove binder (MGB) drugs, which induce the degradation of the mitochondrial DNA. In line with this, in vitro assays suggest that 16 and 17 have a comparatively lower affinity for different template DNAs than the MGB drug diminazene. Therapeutic efficacy studies and stability assays suggest that the pharmacological properties of the hits should be optimized. The compounds can be rated as excellent scaffolds for the design of highly potent and selective anti-T. brucei agents.

A summary of the resource.
In this work we present the synthesis, structural, morphological, optical and electrochemical characterization of sodium titanate nanotubes (NaNT) obtained by hydrothermal means. According to our detailed structural study, we found that a Na2Ti2O5·H2O layered crystalline phase with [TiO5] pyramidal square base coordination, utilized as a building block of the nanotubes, explains satisfactorily the experimental results. This is a very important aspect, since the bibliography does not always provide structural information about titanate nanotubes. With respect to the formation mechanism we highlight the crucial role of the post treatment washing in the final crystalline structure of the nanotubes. Simulated X-rays powder diffraction patterns (XRPD), based on real size nanotubes models, were refined with experimental data. Building blocks for the NaNT model were inferred from computational simulations, by means of Density Functional Theory. The obtained tube's model characteristic dimensions were also consistent with SAXS and TEM analysis. In addition, our SAXS analysis suggested that the presence of humidity in the cavities of titanate nanotubes is progressively lost at the 80–100 °C temperature range, in good agreement with thermogravimetric analysis. Electrochemical impedance spectroscopy analysis for dried samples in the temperature range, studied under cooling conditions, revealed a single ionic transport mechanism, probably associated to the in-between [TiO5] layers ion transport. However, a secondary process was observed for non-dried samples, during heating conditions, that could be associated to the ionic transport along the water filled tube's cavity, consistent with SAXS analysis. In summary, this work shows an integral approach, through theory and experiment, in order to understand the effect of the structure on the physical and electronic properties of the system.

Nutritional warnings are rapidly gaining relevance, particularly in the region of the Americas, as a front-of-package nutrition labelling scheme that facilitate the identification of products with high content of nutrients associated with non-communicable diseases (NCDs). Several graphic designs are being developed as countries discuss the implementation of nutritional warnings, which makes it necessary to develop research to evaluate their efficacy. In this context, the aim of the present work was to compare the efficacy of a series of nutritional warnings that are being considered by Brazilian national authorities for the country with two of the most studied schemes: the guidelines daily amounts (GDA) and the traffic light system. Two studies were conducted. In the first one, visual search was conducted with 62 participants to evaluate the time needed by them to identify the schemes on food labels and to identify whether a product had high nutrient content. In the second study, an online survey with 1932 participants was used to evaluate their ability to use FOP nutrition labelling schemes to correctly identify the most healthful product in a set, as well as high nutrient content in a product. In addition, the influence of FOP nutrition labelling schemes on perceived healthfulness was evaluated. Finally, consumers’ perception of the schemes was gathered using an open-ended question. Results confirmed the advantages of nutritional warnings compared to the GDA and the traffic-light system to facilitate the identification of products with high content of nutrients associated with NCDs. In addition, the use of familiar signs frequently used to convey a ‘warning message’ outperformed other unfamiliar signs in terms of their ability to facilitate the interpretation of nutrition information. Pseudocapacitive materials can enhance the energy storage performance of supercapacitors by making use of surface redox reactions. In recent years, different iron compounds have been investigated as pseudocapacitive materials, showing promising features for supercapacitor electrode applications. Carbon nanocomposites containing iron/phosphorus compounds have been prepared from porous carbon, followed by thermal treatment at different temperatures (700°C to 1000°C). The obtained supercapacitor electrodes were evaluated by electrochemical analyses using sulfuric acid electrolyte. The as-prepared nanocomposite contained nanostructured iron oxides or oxyhydroxides, whereas the nanocomposites prepared at 700°C to 900°C were composed of nanostructured iron phosphates. On the other hand, heat treatment at 1000°C caused the formation of nanocrystalline iron phosphides (mostly Fe2P nanoparticles). The Fe-containing samples showed enhanced specific capacitance (246 F g−1 to 447 F g−1 at 10 A g−1), which can be related to the pseudocapacitive contribution of the iron compounds. The sample heat treated at 1000°C exhibited favorable electrochemical performance, showing high electrical capacitance and good rate capability at 40 A g−1. These results reveal that porous carbon/iron phosphide nanocomposites are promising materials for use in supercapacitor electrode applications.

A summary of the resource.
Vitis vinifera flowers and grape fruits are one of the most interesting ecosystem niches for native yeasts development. There are more than a 100 yeast species and millions of strains that participate and contribute to design the microbial terroir. The wine terroir concept is understood when grape and wine micro-regions were delimited by different quality characteristics after humans had been growing vines for more than 10,000 years. Environmental conditions, such as climate, soil composition, water management, winds and air quality, altitude, fauna and flora and microbes, are considered part of the “terroir” and contribute to a unique wine style. If “low input winemaking” strategies are applied, the terroir effect will be expected to be more authentic in terms of quality differentiation. Interestingly, the role of the microbial flora associated with vines was very little study until recently when new genetic technologies for massive species identification were developed. These biotechnologies allowed following their environmental changes and their effect in shaping the microbial profiles of different wine regions. In this chapter we explain the interesting positive effects on flavor diversity and wine quality obtained by using “friendly” native yeasts that allowed the microbial terroir flora to participate and contribute during fermentation.

The aim of this work was to develop and evaluate a 99mTc‐labeled neuropeptide Y derivative with affinity toward Y1‐receptor. The selected amino acid sequence included nine amino acids derived from the C‐terminal portion of the NPY complemented with the addition of one cysteine‐mercaptoacetic acid moiety to bind the radiometal. Labeling was achieved through the preparation of a 3 + 1 nitrido complex. Physicochemical evaluation, cell uptake, internalization and externalization studies, and competitive assays were performed. Biodistribution experiments were carried out in normal and tumor‐bearing mice. A single product with radiochemical purity >90% and high stability was obtained. In vitro analysis showed specific cellular uptake, IC50 of 73.2 nM, and a high internalization rate (80%). Biodistribution studies showed low blood and renal uptake and combined hepatobiliary and urinary elimination. Preliminary studies in mice bearing induced breast tumors rendered promising uptake values.

Neurodegenerative diseases affect millions of people around the world. Several studies point out caspase-3 as a key player in the development and progression of neurological disorders including amyotrophic lateral sclerosis, Alzheimer's, Parkinson's and Huntington's diseases. Furthermore, oxidative stress and mitochondrial dysfunction plays an important role in neurodegenerative pathologies leading to neuronal damage and cell death. Pharmacological properties of nitrones such as free radical trapping and neuroprotection has been previously described. In the present work, we have assessed ten non-cytotoxic nitrones for their ability to inhibit apoptosis plus their potential to reduce active caspase-3 and oxidative stress in the hippocampal neuronal cell line HT22. Our results highlight the faculty of nitrones to inhibit apoptosis by a mechanism that involves active caspase-3 reduction and decrease of reactive oxygen species. Moreover, docking and molecular dynamics approaches lead to a detailed analysis at the atomic level of the nitrones binding mode to caspase-3 suggesting that compounds bind in a region close to the catalytic site. All these data place these molecules as excellent hits for further efforts to redesign novel compounds in the search of a new therapy against neurodegenerative disorders.

The objective of this study was to determine the activity of pyridine-2-thiol 1-oxide sodium salt (Na mpo) and its complex with iron [Fe(mpo)3] against Mycobacterium tuberculosis. The compounds were tested against a standard strain of M. tuberculosis H37Rv (ATCC 27294), with minimal inhibitory concentrations (MIC90) of 7.20 and 1.07 μM to Na mpo and [Fe(mpo)3], respectively, and against three clinical isolates with different genotypic profiles, with MIC values ranging from 0.74 to 6.52 and 0.30 to 2.25 μM to Na mpo and [Fe(mpo)3], respectively. [Fe(mpo)3] was more effective against susceptible strains but both compounds were effective in inhibiting MDR and XDR-TB clinical strains. The profile activity was determined through the methodology of a time-kill curve against standard and clinical strains of M. tuberculosis. Time-kill studies indicated that Na mpo had an early bactericidal activity against H37Rv and clinical isolates, with sterilizing effects observed in 5 and 7 days, respectively, at its MIC90. The anti MDR and XDR-M. tuberculosis activity and bactericidal effect of Na mpo and [Fe(mpo)3] demonstrate their potential as new compounds for the treatment of tuberculosis.

BACKGROUND Rice bran (RB), a by‐product of the rice milling industry, constitutes around 10% of the total weight of rough rice. The interest in the use of RB is centered on its nutritional quality, its low cost, and its extensive worldwide production. As RB is commonly used for oil extraction, the defatted rice bran (DRB) is obtained as a second by‐product. The aim of this work was to obtain a defatted rice bran concentrate (DRBC), enriched in protein and fiber, from defatted rice bran flour (DRBF) and to determine its physicochemical and emulsifying properties. RESULTS To obtain the DRBC, the starch was efficiently hydrolyzed (> 98%) with α‐amylase and amyloglucosidase, with a concomitant increase in the proportions of crude protein (from 154.7 to 274.3 g kg−1) and total dietary fiber (from 276.1 to 492.3 g kg−1). Defatted rice bran concentrate exhibited a loss of protein solubility and increased surface hydrophobicity compared with DRBF. Defatted rice‐bran concentrate dispersions with and without previous ultrasound treatment were prepared. The sonication led to an increase in the apparent viscosity. Emulsions were prepared with dispersions with and without previous ultrasound treatment and showed high stability in quiescent conditions over 28 days. However, the emulsions prepared with dispersions treated with ultrasound resulted in lower D4,3 values and higher elastic and viscous moduli. CONCLUSION The rice bran concentrate can be used to obtain stable oil‐in‐water (O/W) emulsions, including both soluble and insoluble fractions, in acidic and neutral conditions. These innovative findings thus contribute to increasing the added value of this important by‐product of the rice‐milling industry. © 2019 Society of Chemical Industry

Although several efforts to promote breastfeeding have been implemented worldwide, the prevalence of exclusive breastfeeding in the first 6 months of life remains low. One of the factors responsible for this trend is the widespread sales and marketing of breast-milk substitutes. In this context, the present work aimed at exploring mothers and health professionals' social representations of breastfeeding and infant formula. The study was conducted in Montevideo (Uruguay), as part of the periodic assessment performed by the Uruguayan government to monitor the marketing of breast-milk substitutes according to the OMS/UNICEF NetCode toolkit. A total of 330 mothers of children under 24 months old and 154 health professionals were recruited at 33 health care facilities. Participants were asked to complete two word association tasks, one with breastfeeding and the other with infant formula. Inductive coding was used to classify the responses into categories and dimensions. The structure of the social representations was explored considering the frequency of mention and the average rank of appearance of the categories. Results showed that mothers’ social representations mainly positioned breastfeeding as a feeding practice, whereas infant formulas were mainly perceived as a complement to breast-milk that provides support. In the case of health professionals, the social representations of breastfeeding and infant formulas were in line with scientific evidence and health recommendations. These results suggest that efforts to promote breastfeeding should focus on raising public awareness of the risks associated with breast-milk substitutes and on overcoming the systemic barriers to breastfeeding, particularly among working mothers.

This work presents the synthesis and characterization of eight copper complexes [Cu(L-dipeptide)(neo)]·nH2O (neo = neocuproine) and their cytotoxic activities against tumor cell lines. The crystalline structure of [Cu(gly-val)(neo)]·3H2O, [Cu(gly-leu)(neo)]·H2O, [Cu(ala-gly)(neo)]·4H2O, [Cu(val-phe)(neo)]·4.5H2O and [Cu(phe-phe)(neo)]·3H2O were determined by single crystal X-ray diffraction. In all of them, the Cu(II) is pentacoordinated, in a square pyramidal environment. The coordination observed in solid state was retained in the major species in aqueous solution, as suggested by Electronic Paramagnet Resonance and UV–vis spectroscopies. The complexes were shown to have affinity for isolated DNA, as determined by Circular Dichroism experiments. Furthermore, biological experiments showed that all the complexes present high cytotoxic activity against the cell lines: MDA-MB-231, MCF-7 (human metastatic breast adenocarcinomas, the first triple negative), MCF-10A (human normal breast cells), A549 (human lung epithelial carcinoma) and MRC-5 (human lung epithelial cells). Together, these results suggest that these compounds are promising steps towards new effective drugs to treat cancer.

New thiazolyl derivatives (BT and TT) and their copper (II) complexes [Cu2Cl2(BT)2] (Cu-BT) and [Cu4ClO2(TT)2]PF6⋅3.5H2O (Cu-TT) were synthesized and characterized by elemental analysis, 1H NMR and 13C NMR, HRMS, X-ray diffraction, IR and UV–Vis spectroscopies. The crystal structure of Cu-BT shows the formation of a dinuclear complex where each copper(II) center is bonded to two thiazol N atoms, from different BT ligands, one deprotonated amide N atom, an O atom from the ester terminal groups and a chlorine atom. The structure found for Cu-TT is a positively charged tetranuclear moiety containing two deprotonated TT ligands, a chlorine anion, two hydroxide anions acting as bridges between the copper centers and a water molecule. The cytotoxic activity of both copper complexes was evaluated on metastatic breast cancer cell lines, characterized for its rapidly dividing behavior. Both, Cu-BT and Cu-TT, show higher cytotoxic activity against these tumor cells than free BT and TT and also than cisplatin. In addition, we found that both complexes interact with DNA. Consistently, they also show cytotoxicity against a rapidly dividing non-tumor cell line, although with higher IC50, being such interaction and selectivity an indicator of the possible coexistence of more than one mechanism of action.

Fifteen new allyldithiocarbimates were prepared from different allylic bromides and various potassium dithiocarbimates, yielding (Z)-2-(methoxycarbonyl)-3-(X-nitrophenyl)allyl-(N-R-sulfonyl)dithiocarbimates (where X = 2, 3 and 4; R = phenyl, 4-fluorophenyl, 4-chlorophenyl, 4-bromophenyl and 4-iodophenyl). These anions were isolated as tetraphenylphosphonium salts and characterized by HRMS, infrared, 1H and 13C NMR spectroscopies. Molecular electrostatic potentials were used to evaluate intermolecular interactions present in the new substances and to explain variations observed on their melting points. Single crystal X-ray diffraction experiments confirmed the Z stereochemistry of the allyldithiocarbimate anions. C–HO, C–HN, C–HS and C–Hπ intermolecular interactions in the solid state were studied by X-ray diffraction and Hirshfeld surface analyses. The new compounds inhibited the mycelial growth of various fungi species responsible for severe plant diseases. The allylithiocarbimates were especially active against Botrytis cinerea, with IC50 values as low as 20 μM, being more effective than the active principals of the commercial fungicides Ziram and Mancozeb.

Temporal Dominance of Sensations (TDS) and Temporal Check-all-that-Apply (TCATA) from three different case studies are compared by means of canonical correlation analysis, orthogonalization and principal component analysis of the vertically unfolded data (which means that the matrices compared have samples*timepoints in the rows and attributes in the columns). The multivariate analyses decompose the datasets into common and distinct components. The results showed that the major part of the variation is common between the two methods for the cases investigated, but that there were subtle differences showing better discrimination for TCATA than TDS. TDS showed a more complex data structure and more unique variation. The unique variation in TDS is, however, difficult to interpret. The methods are more different towards the end of the mastication, this can be explained both by the difficulty of assessors to agree on the dominant attributes at the bolus stage for TDS, and that assessors may forget to unclick attributes in TCATA. This work builds on recent methodological studies on temporal methods that aim to better understand differences among methodologies and ultimately to identify what methods could be better for answering different objectives.

African trypanosomiasis is a major problem for human and animal health in endemic countries, where it threatens millions of people and affects economic development. New drugs are needed to overcome the toxicity, administration, low efficacy, and resistance issues of the current chemotherapy. Robust, simple, and economical high‐throughput, whole‐cell‐based assays are required to accelerate the identification of novel chemical entities. With this aim, we generated a bioluminescent cell line of the bloodstream stage of Trypanosoma brucei brucei and established a screening assay. Trypanosomes were stably transfected to constitutively express a thermostable red‐shifted luciferase. The growth phenotype and drug sensitivity of the reporter cell line were essentially identical to that of the parental cell line. The endogenous luciferase activity, measured by a simple bioluminescence assay, proved to be proportional to parasite number and metabolic status. The assay, optimized to detect highly potent compounds in a 96‐well‐plate format, was validated by screening a small compound library (inter‐assay values for Z' factor and coefficient variation were 0.77 and 5.8%, respectively). With a hit‐confirmation ratio of ~97%, the assay was potent enough to identify several hits with EC50 ≤ 10 μM. Preliminary tests indicated that the assay can be scaled up to a 384‐well‐plate format without compromising its robustness. In summary, we have generated reporter trypanosomes and a simple, robust, and affordable bioluminescence screening assay with great potential to speed up the early‐phase drug discovery against African trypanosomes.

Food waste is a major barrier to a sustainable society. Cleaner production is most effectively approached by amongst others avoiding waste. A lot of food waste is due to consumer perception of sub-optimality of foods, and supermarkets can offer ‘imperfect’ products to consumers at a reduced price in order to fight food waste. However, little is known about how this is perceived by consumers and whether it backfires as negative associations about the store, this is particularly under-researched for consumers in the global south. An online experimental survey was conducted in Uruguay to qualitatively explore consumer’s associations with stores selling suboptimal food, and to quantitatively compare the associations generated by three types of sub-optimality. Findings revealed that associations were related to four main dimensions. Consumers majorly thought of which retail practices and personnel behaviour cause the sub-optimality and the relationship between consumers and company, as well as which signal the products send, and whether or not they consider choiceof the item. Few positive associations were mentioned. However, they emerged more often for fresh produce suboptimal in appearance, particularly when price is reduced, and among deal prone consumers. Associations on relationship and signal were primarily negative and in reaction to food with package damage. Associations on choice were more frequent for products nearing expiration date. Price reduction appeared to send a mixed signal to consumers. Results suggest that retailers should improve their overall image and communicate their precise reason for offering suboptimal food, in order to avoid a negative impact on store image.

In this study, activated biochar was produced using pilot-scale technologies of fast pyrolysis and activation to create desirable morphology, surface chemistry, and adsorptive properties for application in supercapacitors. First, residues from white birch were converted into biochar by fast pyrolysis (~ 450 °C). Then, physical (using CO2) or chemical (using KOH) activation was carried out in a homemade pilot-scale furnace at 900 °C. These synthesized materials presented distinct porosity structures: micro-/mesoporous (CO2 material) and highly microporous (KOH material), reaching surface areas of up to 1700 m2 g−1. Electrochemical results showed that KOH-activated biochar had higher specific electrical capacitance in both acidic and neutral electrolytes with a maximum specific capacitance value of 350 and 118 F g−1 at 1 A g−1, respectively; while, for CO2-activated biochar, the maximum obtained values were 204 and 14 F g−1. The greater proportion of oxygenated and nitrogenated functional groups on the surface of the KOH activated biochar, along with its high surface area (with wider porosity), improved its performance as a supercapacitor electrode. Specifically, the low proportion of ultramicropores was determinant for its better electrochemical behavior, especially in the neutral electrolyte. Indeed, these results are similar to those found in the literature on the electrical capacitance of carbonaceous materials synthesized in a small-scale furnace. Thus, the chemical-activated biochar made from wood residues in pilot-scale furnaces is a promising material for use as electrodes for supercapacitors.

A summary of the resource.
β-galactosidase is an enzyme responsible for catalyzing the hydrolysis of the lactose β-1,4 linkage into α-D-glucose and β-D-galactose. β-galactosidase is found in plants, animals and microorganisms. In Beta-Galactosidase: Properties, Structure and Functions, the authors discuss the main microorganisms that produce β-galactosidase, the characteristics of the culture media, bioprocessing parameters, the most relevant downstream steps used in the recovery of microbial β-galactosidase, as well as the main immobilization techniques. Next, this compilation examines β-galactosidases classification, mechanisms, characterization and applications. Recent advances in galacto-oligosaccharides production and purification are also covered, and different challenges and future perspectives are discussed. In the closing study, strains with β-Galactosidase activity were isolated from 25 Yak Yoghourt samples collected from the Gannan pasturing area of Gansu Province. An efficient β-galactosidase producing strain SYA2 screened out from 21 strains was identified as Enterobacter sp. by means of morphological feature observation, physiological and biochemical characteristics measurement, and 16S rDNA sequence analysis.
