A summary of the resource.
A novel method for the determination of trace cadmium in honey is presented. It consists of a preconcentration step based on dispersive liquid-liquid microextraction (DLLME) and measurement by flame AAS. Samples are dissolved in water. A mixture of carbon tetrachloride (extraction solvent) and acetonitrile (dispersion solvent) containing ammoniun pyrrolidine dithiocarbamate (APDC) as complexing agent is injected rapidly into the dissolved sample by means of a syringe pump. After centrifugation, an aliquot of the organic phase is injected in the nebuliser of an atomic absorption spectrometer by means of a lab-made microsampling device. Response (integrated absorbance) was linear with concentrations up to 12 μg L−1. Precision (sr(%)) at the 5 μg L−1 level was < 3.1%. Detection and quantification limits were 1.6 and 5.5 μg per 100 g of honey respectively. Six commercial samples were spiked at the 10 μg per 100 g level and analysed by the proposed method and by electrothermal atomic absorption spectrometry. The results did not differ at the 5% significance level, with recoveries for the DLLME/FAAS method ranging from 82 to 125%. The proposed method requires simple sample preparation and does not use expensive equipment, thus it could be used as screening method for the content of cadmium in honey.

In this work, we report a novel polymorph of AgBiW2O8, which was synthesized by a simple arc melting method. The structural details of this compound were determined by X-ray powder diffraction, and elemental composition was confirmed with ICP-AES. β-AgBiW2O8 adopts the scheelite structure type with tetragonal space group I41/a and lattice parameters a = b = 5.3019(3) and c = 11.678(2) Å. Structural details and optical properties are discussed in detail. Diffuse reflectance spectroscopy (DRS) measurements of β-AgBiW2O8 showed light absorption profile extended to the visible region, which suggests the material has a smaller band gap energy than Bi2WO6, as expected due to contribution of Ag d-orbitals to the electronic structure.

Ibogaine is an atypical psychedelic alkaloid, which has been subject of research due to its reported ability to attenuate drug-seeking behavior. Recent work has suggested that ibogaine effects on alcohol self-administration in rats are related to the release of Glial cell Derived Neurotrophic Factor (GDNF) in the Ventral Tegmental Area (VTA), a mesencephalic region which hosts the soma of dopaminergic neurons. Although previous reports have shown ibogaine's ability to induce GDNF expression in rat midbrain, there are no studies addressing its effect on the expression of GDNF and other neurotrophic factors (NFs) such as Brain Derived Neurotrophic Factor (BDNF) or Nerve Growth Factor (NGF) in distinct brain regions containing dopaminergic neurons. In this work, we examined the effect of ibogaine acute administration on the expression of these NFs in the VTA, Prefrontal Cortex (PFC), Nucleus Accumbens (NAcc) and the Substantia Nigra (SN). Rats were i.p. treated with ibogaine 20 mg/kg (I20), 40 mg/kg (I40) or vehicle, and NFs expression was analyzed after 3 and 24 h. At 24 h an increase of the expression of the NFs transcripts was observed in a site and dose dependent manner. Only for I40, GDNF was selectively upregulated in the VTA and SN. Both doses elicited a large increase in the expression of BDNF transcripts in the NAcc, SN and PFC, while in the VTA a significant effect was found only for I40. Finally, NGF mRNA was upregulated in all regions after I40, while I20 showed a selective upregulation in PFC and VTA. Regarding protein levels, an increase of GDNF was observed in the VTA only for I40 but no significant increase for BDNF was found in all the studied areas. Interestingly, an increase of proBDNF was detected in the NAcc for both doses. These results show for the first time a selective increase of GDNF specifically in the VTA for I40 but not for I20 after 24 h of administration, which agrees with the effective dose found in previous self-administration studies in rodents. Further research is needed to understand the contribution of these changes to ibogaine's ability to attenuate drug-seeking behavior.

A summary of the resource.
Lapachol acetate [systematic name: 3-(3-methyl­but-2-en­yl)-1,4-dioxonaph­thalen-2-yl acetate], C17H16O4, was prepared using a modified high-yield procedure and its crystal structure is reported for the first time 80 years after its first synthesis. The full spectroscopic characterization of the mol­ecule is reported. The mol­ecular conformation shows little difference with other lapachol derivatives and lapachol itself. The packing is directed by inter­molecular π–π and C—H...O inter­actions, as described by Hirshfeld surface analysis. The former inter­actions make the largest contributions to the total packing energy in a ratio of 2:1 with respect to the latter.

Workflows based on gas and liquid chromatography coupled to mass spectrometry for the identification of toxic pyrrolizidine alkaloids present in plants were developed and applied to Echium plantagineum L., Boraginaceae, extracts. GC–MS based determinations need reduction and derivatization steps prior to MS analysis, which is performed using a Full Scan and Single Ion Monitoring sequence for screening, identification and quantification purposes. The LC–(ESI)–MS/MS determination was performed directly from the extract without derivatization. Acetyl lycopsamine, echimidine, echimidine N-oxide, echiumine, echiumine N-oxide, lycopsamine, lycopsamine N-oxide, 7,9-ditigloylretronecine N-oxide and a not reported PA of m/z 466, were identified and quantified in E. plantagineum extracts, through three operating modes of LC-QTRAP: precursor ion scan, enhanced product ion scan and multiple reaction monitoring. Precursor ion scan detects all the ions that give rise to a daughter ion at m/z 120, the presence of the parent pyrrolizidine alkaloid is confirmed through its MS2 spectrum (enhanced product ion) and quantified by multiple reaction monitoring. These workflows are general approaches to study chemical families using GC/LC-MS. For extracts suspicious of containing pyrrolizidine alkaloids, they are suitable tools for the quality and safety control of food, feed as well as phytotherapeutics.

The design of white-light emitting materials for solid-state lighting technology is governed by the combination of independent red, green and blue emitting centers. Many mixed-lanthanide single phase white-light source materials have been produced so far by a careful trial and error combination of lanthanide ions during the synthetic procedure. In this work we propose a simple and efficient method for the direct preparation of the desired material with the optimal combination of metal ions, avoiding the trial and error multiple synthetic approach. In the proposed strategy, the emission profiles of the isostructural complexes of each lanthanide ion for a chosen excitation wavelength are compared relative to their ion density and the necessary final composition of the material is calculated in order to obtain the white-light CIE coordinates. Using this method, we prepared a white-light emitting material which is a metal-organic framework built from oxydiacetate ligand, lanthanide ions (Eu, Tb and Dy emitting red, green and blue respectively) and zinc (a closed shell metal ion that has proven to be ideal for optical applications). The compound [Dy1.60Eu0.38Tb0.02Zn3(oda)6(H2O)6]·12H2O was prepared and fully characterized by single crystal x-ray diffraction, its optical properties were also measured. The proposed rational mixing of lanthanide ions could be useful for the preparation of white-light source materials in analogous systems, provided the single-lanthanide compounds are isostructural along the series and no optical interaction is operative among ions.

The title complex, {[Ni(hab)]K[(hab)Ni]}·SCN (H 2 hab is bis(2-hydroxy-3-methoxybenzylidene)-1,2-diaminobenzene) crystallizes in the Monoclinic space group P2 1 /c with unit cell dimensions a = 11.5891 (13) Å, b = 17.307 (2) Å, c = 19.781 (2) Å,β = 92.368 (10)°, V = 3964.2 (8) Å 3 , Z = 4, R 1 = 0.106 and wR 2 = 0.305. The Ni II cation is in a N 2 O 2 inner and has a distorted square-planar coordination environment. The Ni II cation is linked by two imino nitrogen atoms and two bridged phenolate oxygen atoms. The potassium cation is octacoordinated in O 8 donor site and linked by four bridged phenolate oxygen atoms and four methoxy oxygen atoms. In the crystal, the {[Ni(hab)]K[(hab)Ni]} + cations form layers parallel to the ab plane. The SCN-anions are located in the interlayer space and connect the layers via C-H…N and C-H…S hydrogen bonds, forming three dimensional network.

The crystal structure of the title compound, [Ni3(C8H4O4)3(C3H7NO)4], is a two-dimensional coordination network formed by trinuclear linear Ni3(tp)3(DMF)4 units (tp = terephthalate = benzene-1,4-di­carboxyl­ate and DMF = di­methyl­formamide) displaying a characteristic coordination mode of acetate groups in polynuclear metal–organic compounds. Individual trinuclear units are connected through tp anions in a triangular network that forms layers. One of the DMF ligands points outwards and provides inter­actions with equivalent planes above and below, leaving the second ligand in a structural void much larger than the DMF mol­ecule, which shows positional disorder. Parallel planes are connected mainly through weak C—H⋯O, H⋯H and H⋯C inter­actions between DMF mol­ecules, as shown by Hirshfeld surface analysis.

Development of new antimicrobial agents, capable of combating resistant and multidrug-resistant fungal and bacterial clinical strains, is necessary. This study presents the synthesis and antimicrobial screening of 42 2-substituted-1,4-benzenediols, being 10 novel compounds. In total, 23 compounds showed activity against fungi and/or bacteria. Benzenediol compounds 2, 5, 6, 8, 11, and 12 demonstrated broad spectrum antimicrobial actions, including resistant and multidrug-resistant species of dermatophytes (Trichophyton mentagrophytes), Candida spp. and the ESKAPE panel of bacteria. Minimum inhibitory concentrations of these compounds for fungi and bacterial strains ranged from 25 to 50 µg/ml and 8–128 µg/ml, respectively. The antifungal mechanism of action is related to the fungal cell wall of dermatophytes and membrane disruption to dermatophytes and yeasts, in the presence of compound 8. Specific structural changes, such as widespread thinning along the hyphae and yeast lysis, were observed by scanning electron microscopy. The effects of compound 8 on cell viability are dose-dependent; however they did not cause genotoxicity and mutagenicity in human leukocyte cells nor haemolysis. Moreover, the compounds were identified as nonirritant by the ex-vivo Hen’s egg test-chorioallantoic membrane (HET-CAM). The furan-1,4-benzenediol compound 5 showed in vivo efficacy to combat S. aureus infection using embryonated chicken eggs. Therefore, the compounds 8, and 5 are promising as hits for the development of new antimicrobial drugs with reduced toxicity.

Este trabajo describe la optimización de un método espectrofotométrico clásico (“método Hornsey”) para determinar hierro hemo en carnes. Este método extrae hierro hemo de la carne mediante agitación con acetona ácida, maceración en la oscuridad y determinación espectrofotométrica a 640nm sobre el filtrado. Es ampliamente utilizado, pero su descripción original no detalla la influencia de algunas variables. En numerosos trabajos se determina hierro hemo según el “método Hornsey” utilizando diferentes estrategias de agitación y tiempos de incubación diversos. Estas variables se optimizaron mediante un diseño multivariado de tipo central compuesto. Con el método optimizado se analizaron distintos cortes de carne vacuna para evaluar su valor nutricional en cuanto a niveles de hierro biodisponible. A partir de una optimización racional, se obtuvieron las condiciones experimentales adecuadas para realizar un método analítico, considerado de referencia, de forma sistemática y protocolizada en los laboratorios para evaluar el contenido de hierro hemo en carnes.

The hexanuclear compounds [Fe6O2Cl2{(py)2CO2}2(pyOH)2(pyCOO)2(ButCOO)6] (1) and [Fe6O2{(py)2CO2}2(pyCOO)4(ButCOO)6] (2) were obtained by condensation of the trinuclear μ3-oxo-centered iron(III) pivalate [Fe3O(ButCOO)6(H2O)3]+ in the presence of di(2-pyridyl)ketone [(py)2CO] in MeCN at ambient temperature. Both complexes contain an analogous core which can be described as two {Fe3O}7+ units joined by gem-diolato (py)2CO22− bridges. Variable-temperature magnetic susceptibility measurements in polycrystalline samples of 1 and 2 reveal strong antiferromagnetic couplings between the iron(III) ions leading to S = 0 ground states.

The parallel segmented flow (PSF) chromatography column was applied for the first time to trace residue analyses with mass spectral detection. Three problematic pesticides, notorious for poor peak shape and ionization issues were selected to illustrate the advantages and disadvantages of utilizing the PSF column as an alternative front-end separation interface for LC-MS approaches. Assessment of performance for each pesticide was based on a spiked lemon matrix prepared via a QuEChERS (quick, easy, cheap, rugged and safe) sample preparation approach. Comparisons were made between assays conducted using a conventional column and a PSF column (operated with a 40% segmentation ratio), each packed with identical stationary phases and of the same column format. Both columns were connected to the MS detector using the exact same connective tubing and electrospray (ESI) conditions. The primary advantage of the PSF column was that it enabled an increase in throughput by at least 2.5 times compared to the conventional HPLC column. At the same time, the volumetric flow to the MS was exactly the same for both the PSF column and the conventional column. Effectively there was no difference in the sensitivity provided by peak areas between the PSF and conventional columns: peak area LOQ values were comparable, 4–9 μg/kg for the PSF and 4–8 μg/kg for the conventional column. This is a vital requirement for food control methods to secure absence at default maximum residue limit (MRL) of 10 μg/kg (10 ppb) level. Since the efficiency of the PSF was superior to that of the conventional column, the peak width of eluting components was reduced by as much as 70% in the former compared to the peak widths on the latter. Hence, the PSF column provides greater peak capacity and this will enable faster analysis of complex samples.

Freshwater harmful algal blooms produce a broad array of bioactive compounds, with variable polarity. Acute exposure to cyanotoxins can impact the liver, nervous system, gastrointestinal tract, skin, and immune function. Increasing evidence suggests chronic effects from low-level exposures of cyanotoxins and other associated bioactive metabolites of cyanobacterial origin. These sundry compounds persist in drinking and recreational waters and challenge resource managers in detection and removal. A systematic approach to assess the developmental toxicity of cyanobacterial metabolite standards was employed utilizing a robust and high throughput developmental Danio rerio embryo platform that incorporated a neurobehavioral endpoint, photomotor response. Subsequently, we applied the platform to cyanobacterial bloom surface water samples taken from temperate recreational beaches and tropical lake subsistence drinking water sources as a model approach. Dechorionated Danio rerio embryos were statically immersed beginning at four to six hours post fertilization at environmentally relevant concentrations, and then assessed at 24 h and 5 days for morbidity, morphological changes, and photomotor response. At least one assessed endpoint deviated significantly for exposed embryos for 22 out of 25 metabolites examined. Notably, the alkaloid lyngbyatoxin–a resulted in profound, dose-dependent morbidity and mortality beginning at 5 μg/L. In addition, hydrophobic components of extracts from beach monitoring resulted in potent morbidity and mortality despite only trace cyanotoxins detected. The hydrophilic extracts with several order of magnitude higher concentrations of microcystins resulted in no morbidity or mortality. Developmental photomotor response was consistently altered in environmental bloom samples, independent of the presence or concentration of toxins detected in extracts. While limited with respect to more polar compounds, this novel screening approach complements specific fingerprinting of acutely toxic metabolites with robust assessment of developmental toxicity, critical for chronic exposure scenarios.

BACKGROUND AND PURPOSE: Atherosclerosis is characterized by chronic low-grade inflammation with concomitant lipid accumulation in the arterial wall. Anti-inflammatory and anti-atherogenic properties have been described for a novel class of endogenous nitroalkenes (nitrated-unsaturated fatty acids), formed during inflammation and digestion/absorption processes. The lipid-associated antioxidant α-tocopherol is transported systemically by LDL particles including to the atheroma lesions. To capitalize on the overlapping and complementary salutary properties of endogenous nitroalkenes and α-tocopherol, we designed and synthesized a novel nitroalkene-α-tocopherol analogue (NATOH) to address chronic inflammation and atherosclerosis, particularly at the lesion sites. EXPERIMENTAL APPROACH: We synthesized NATOH, determined its electrophilicity and antioxidant capacity and studied its effects over pro-inflammatory and cytoprotective pathways in macrophages in vitro. Moreover, we demonstrated its incorporation into lipoproteins and tissue both in vitro and in vivo, and determined its effect on atherosclerosis and inflammatory responses in vivo using the Apo E knockout mice model. KEY RESULTS: NATOH exhibited similar antioxidant capacity to α-tocopherol and, due to the presence of the nitroalkenyl group, like endogenous nitroalkenes, it exerted electrophilic reactivity. NATOH was incorporated in vivo into the VLDL/LDL lipoproteins particles to reach the atheroma lesions. Furthermore, oral administration of NATOH down-regulated NF-κB-dependent expression of pro-inflammatory markers (including IL-1β and adhesion molecules) and ameliorated atherosclerosis in Apo E knockout mice. CONCLUSIONS AND IMPLICATIONS: In toto, the data demonstrate a novel pharmacological strategy for the prevention of atherosclerosis based on a creative, natural and safe drug delivery system of a non-conventional anti-inflammatory compound (NATOH) with significant potential for clinical application.

Plant defensins comprise a large family of antimicrobial peptides (AMP) with a wide range of biological functions. They are cysteine-rich molecules, highly sequence diverse but with a conserved and stable structure. In this work, a defensin gene (EcgDf1) was isolated from Erythrina crista-galli, a legume tree native from South America. The predicted peptide presents eight cysteines, with a γ-core motif GXCX3-9C and six cysteines distributed like the typical defensin αβ motif. The mature EcgDf1 coding sequence was heterologously expressed in Escherichia coli strains and purified by affinity chromatography. Possible dimer and oligomers of EcgDf1 were visible in SDS electrophoresis. Moreover, its 3D structure, determined by homology modeling, docking, and molecular dynamics simulations, was found to be compatible with the formation of homodimers between the β3 and β1-loop-α1, leaving the β2-loop-β3 free to interact with lipid membranes. The purified recombinant peptide inhibited the growth of several critical plant and human pathogens, like the opportunistic fungi Candida albicans and Aspergillus niger and the plant pathogens Clavibacter michiganensis ssp. michiganensis, Penicillium expansum, Botrytis cinerea, and Alternaria alternata. EcgDf1 is a promising candidate for the development of antimicrobial products for use in agriculture and medicine.

Background The timing of milk donations to human milk banks ranges from a few days to more than 1 year after delivery, and the Holder method is used for pasteurization. We evaluated the effect of temporal variation and thermal treatment on the immunological properties of milk. Methods We analyzed 73 milk samples, raw and after pasteurization, donated at different lactation stages. We studied antibodies, lysozyme, cytokines, soluble receptors, and factors with impact on barrier function. We also evaluated in vitro the capacity of milk to modulate nuclear factor-κB (NF-κB) signaling in an HT-29 epithelial cell line stimulated with tumor necrosis factor-α (TNF-α). Results With few exceptions, immune components exhibited their highest levels in colostrum, and were stable in the various stages of mature milk. Pasteurization altered the immunological composition of milk, and very drastically for some components. Raw milk of the first year reduced NF-κB activation in HT-29 cells treated with TNF-α to approximately the same extent, and Holder pasteurization significantly affected this capacity. Conclusions Overall, the present work reports that mature donated milk is equally valuable over the first year of lactation, but warns about drastic losses of anti-inflammatory properties during Holder pasteurization that could be critical for the health of preterm infants.

In the search for a more effective chemotherapy for the treatment of Chagas’ disease and human African trypanosomiasis, caused by Trypanosoma cruzi and Trypanosoma brucei parasites, respectively, the use of organometallic compounds may be a promising strategy. In this work, eight new heterobimetallic compounds are described including four 5-nitrofuryl containing thiosemicarbazones as bioactive ligands (HL1–HL4) and dppf = 1,1′-bis(diphenylphosphino) ferrocene as an organometallic co-ligand. Complexes of the formula [MII(L)(dppf)](PF6) with M = Pd or Pt were synthesized and fully characterized in the solid state and in solution, including the determination of the molecular structure of four of them by single crystal X-ray diffraction methods. Most compounds showed activity in the low micromolar or submicromolar range against both parasites, with the platinum compounds being more active than the palladium analogues. Activity was significantly increased by generation of the M-dppf compounds (3–24 fold increase with respect to free ligands HL for T. cruzi and up to 99 fold increase with respect to HL for T. brucei). The inclusion of the organometallic co-ligand also led to lower toxicity in mammalian cells and higher selectivity towards both parasites when compared to the free HL compounds. The complexes interact with DNA and affect the redox metabolism of the parasites. Furthermore, the most active and selective compound of the new series showed no in vivo toxicity in zebrafish embryos.

Supercapacitors have received great attention in science and technology, due to its important qualities as the high cycle life, high power density, and variety of applications. The main challenge for the development of better devices is the electrode material. Developing electrode materials with high specific electrical capacitance and low electrical resistance enables an increase of energy accumulated in the supercapacitor. In addition, it is expected that the electrode material presents a simple procedure for preparation with low production cost and be friendly to the environment. In this work, a cheap and simple electroless process was involved to deposit chemical species of iron on a no expensive textile activated carbon fiber felt with interesting properties for application as supercapacitor electrode. The resulting binary composite (Fe@ACF) supercapacitor electrode possesses a high specific capacitance of 237 F g−1 at a current density of 0.2 A g−1 and only 3% of loss of its initial specific capacitance after 3000 charge-discharge cycles. These results reveal that the activated carbon fiber felt/iron composites are promising materials for supercapacitor electrode applications.

There are three basic steps to activated carbon fibers (ACF) manufacturing, from PAN fiber: oxidation/stabilization, carbonization and activation. Carbon material, specially ACF is a very attractive material to be used as supercapacitor electrode. The literature describes carbon material surface chemistry importance for supercapacitors application, mainly nitrogen groups by N-doping. Oxidation/stabilization is an important non-explored factor that influence the surface chemical functionality. This work describes the influence of oxidation/stabilization process on ACF production, from textile PAN fiber, and the non-doping nitrogen surface chemistry characteristic caused by the oxidation process design. Its textural, structural and surface was evaluated for supercapacitor electrode. The results show that the oxidation degree can be used as a mechanism of textural and surface chemistry control. The surface chemistry is the key of this work, different oxidation conditions can produce nitrogen compounds that help to increase specific capacitance. The tests showed an increase in capacitance higher than 100% in comparison to the standard oxidation.

A key metabolic adaptation of some species that face hypoxia as part of their life cycle involves an alternative electron transport chain in which rhodoquinone (RQ) is required for fumarate reduction and ATP production. RQ biosynthesis in bacteria and protists requires ubiquinone (Q) as a precursor. In contrast, Q is not a precursor for RQ biosynthesis in animals such as parasitic helminths, and most details of this pathway have remained elusive. Here, we used Caenorhabditis elegans as a model animal to elucidate key steps in RQ biosynthesis. Using RNAi and a series of C. elegans mutants, we found that arylamine metabolites from the kynurenine pathway are essential precursors for RQ biosynthesis de novo. Deletion of kynu-1, encoding a kynureninase that converts l-kynurenine (KYN) to anthranilic acid (AA) and 3-hydroxykynurenine (3HKYN) to 3-hydroxyanthranilic acid (3HAA), completely abolished RQ biosynthesis but did not affect Q levels. Deletion of kmo-1, which encodes a kynurenine 3-monooxygenase that converts KYN to 3HKYN, drastically reduced RQ but not Q levels. Knockdown of the Q biosynthetic genes coq-5 and coq-6 affected both Q and RQ levels, indicating that both biosynthetic pathways share common enzymes. Our study reveals that two pathways for RQ biosynthesis have independently evolved. Unlike in bacteria, where amination is the last step in RQ biosynthesis, in worms the pathway begins with the arylamine precursor AA or 3HAA. Because RQ is absent in mammalian hosts of helminths, inhibition of RQ biosynthesis may have potential utility for targeting parasitic infections that cause important neglected tropical diseases.

Aromatized olive oils or olive oil based dressings have gained interest from the food industry since they expand the flavors offered to consumers. The present work compared the results obtained from the Word Association (WA) technique with those obtained from a Focus Group (FG) about olive oil, extra virgin olive oil and olive oil-based dressings, and analyzed the advantages of performing a FG prior to data analysis of WA. The results of the FG analyzed prior to the application of the WA technique allowed a better interpretation and understanding of the words associated by the consumers with each WA stimulus. The WA technique clearly differentiated preparations prepared from olive oil with the oils themselves, mainly in terms of taste, texture and combinations allowed.

Searching for a more effective chemotherapy for the treatment of Human African trypanosomiasis, the disease caused by the parasite Trypanosoma brucei, and cancer, in the current work five new [PtII(L)(dppf)](PF6) compounds, with HL = 8-hydroxyquinoline derivatives and dppf = 1,1′-bis(diphenylphosphino)ferrocene, were synthesized and fully characterized. Crystal structures of three compounds were solved by XRD. The compounds displayed fairly good activity against bloodstream T. brucei, with IC50 values in the submicromolar range (IC50: 0.14–0.93 μM), and good selectivity towards the pathogen (SI: 11 - 48) with respect to mammalian macrophages (cell line J774). Coordination to the {Pt-dppf} moiety led, in most cases, to an enhancement of the activity in respect to the bioactive ligands (11 to 41 fold). Cytotoxicity was assessed against wildtype (A2780) and cisplatin-resistance (A2780cisR) ovarian cancer cell lines. Four [PtII(L)(dppf)](PF6) compounds were more active (IC50: 1.2–4.4 μM) than cisplatin (IC50: 26.0 μM) on A2780 cells and showed far superior activity than the reference drug against A2780cisR cells. Platinum levels in A2780 cells showed poor correlation between cellular uptake and the cytotoxic activity. All the complexes interacted with DNA and the most active ones induced reactive oxygen species (ROS) formation which suggested that the mechanism of action for these complexes may be mediated by oxidative stress and interaction with DNA that could act as a potential molecular target for this type of complexes. Some complexes of this series could be considered new hits for the development of prospective agents against trypanosomatid parasites and ovarian cancer.

Nematode parasites have a profound impact on humankind, infecting nearly one-quarter of the world’s population, as well as livestock. There is a pressing need for discovering nematicides due to the spread of resistance to currently used drugs. The free-living nematode Caenorhabditis elegans is a formidable experimentally tractable model organism that offers key advantages in accelerating nematicide discovery. We report the screening of drug-like libraries using an overnight high-throughput C. elegans assay, based on an automated infrared motility reader. As a proof of concept, we screened the “Pathogen Box” library, and identical results to a previous screen using Haemonchus contortus were obtained. We then screened an in-house library containing a diversity of compound families. Most active compounds had a conjugation of an unsaturation with an electronegative atom (N, O, or S) and an aromatic ring. Importantly, we identified symmetric arylidene ketones and aryl hydrazine derivatives as novel nematicides. Furthermore, one of these compounds, (1E,2E)-1,2-bis(thiophen-3-ylmethylene)hydrazine, was active as a nematicide at 25 µm, but innocuous to the vertebrate model zebrafish at 50 µm. Our results identified novel nematicidal scaffolds and illustrate the value of C. elegans in accelerating nematicide discovery using a nonlabor-intensive automated assay that provides a simple overnight readout.

Para extender y fortalecer la Mejora Continua en la “Unidad de Educación Permanente” (UNEP) de la Facultad de Química (FQ) de la Universidad de la República de Uruguay (UdelaR) se emplearon herramientas clásicas de ”Gestión de Calidad” que han demostrado su eficacia en distintas ramas de las actividades Empresarial y Educativa. La experiencia permitió identificar “oportunidades de mejora” e implementar las acciones de seguimiento correspondientes, La experiencia realizada es considerada una muy buena práctica de trabajo en consideración a los resultados obtenidos. Además, su ejecución implicó un uso mínimo de recursos disponibles, se aplicó fácilmente la metodología de trabajo, se logró una comprensión rápida de los participantes y gran empatía por parte de los actores involucrados. En resumen, se consiguió un excelente cumplimiento de los objetivos propios establecidos por el Consejo de Facultad de Química (CFQ) para la función de “Educación Permanente” (EP) de la FQ. La divulgación de esta experiencia puede servir como ejemplo para su aplicación en múltiples áreas de la actividad docente y no docente de la UdelaR.
