<dcterms:abstract>The nitroheterocycle nifurtimox, as part of a nifurtimox-eflornithine combination therapy, represents one of a limited number of treatments targeting Trypanosoma brucei, the causative agent of human African trypanosomiasis. The mode of action of this prodrug involves an initial activation reaction catalyzed by a type I nitroreductase (NTR), an enzyme found predominantly in prokaryotes, leading to the formation of a cytotoxic unsaturated open-chain nitrile metabolite. Here, we evaluate the trypanocidal activities of a library of other 5-nitrofurans against the bloodstream form of T. brucei as a preliminary step in the identification of additional nitroaromatic compounds that can potentially partner with eflornithine. Biochemical screening against the purified enzyme revealed that all 5-nitrofurans were effective substrates for T. brucei NTR (TbNTR), with the preferred compounds having apparent kcat/Km values approximately 50-fold greater than those of nifurtimox. For several compounds, in vitro reduction by this nitroreductase yielded products characterized by mass spectrometry as either unsaturated or saturated open-chain nitriles. When tested against the bloodstream form of T. brucei, many of the derivatives displayed significant growth-inhibitory properties, with the most potent compounds generating 50% inhibitory concentrations (IC50s) around 200 nM. The antiparasitic activities of the most potent agents were demonstrated to be NTR dependent, as parasites having reduced levels of the enzyme displayed resistance to the compounds, while parasites overexpressing TbNTR showed hypersensitivity. We conclude that other members of the 5-nitrofuran class of nitroheterocycles have the potential to treat human African trypanosomiasis, perhaps as an alternative partner prodrug to nifurtimox, in the next generation of eflornithine-based combinational therapies.</dcterms:abstract>

<dcterms:abstract>Introduction: The use of [99mTc]glucarate has been reported as an infarct-avid agent with the potential for very early detection of myocardial infarction. [99mTc]Glucarate has also been postulated as an agent for non-invasive detection of tumors. The aim of our study was to develop a Glucarate kit and evaluate [99mTc]glucarate as a potential cancer imaging agent in female SCID mice bearing humanMDA-MB-435 breast tumors. Methods: Glucarate in a kit formulation was labeled with 99mTc and evaluated for radiolabelling efficiency and radiochemical purity. The Glucarate kit stability was assessed by monthly quality controls. The pharmacokinetics of [99mTc]glucarate were determined in female SCID mice bearing MDA-MB-435 human breast carcinoma tumors at 0.5, 1, 2, 4 and 24 h. Nuclear imaging studies were performed with a microsingle photon emission tomography (SPECT)/computed tomography (CT) system at 2 h post injection, while magnetic resonance imaging (MRI) was employed for tumor morphology analysis and metastatic deposit localization. Results: The Glucarate kits exhibited a stable shelf life of 6 months. [99mTc]Glucarate was obtained with radiochemical purity greater than 95%. Biodistribution studies demonstrated moderate tumor uptake coupled with high renal clearance. Tumor-to-muscle ratios were 4.85 and 5.14 at 1 and 4 h post injection. MRI analysis showed tumors with dense cellular growth and moderate central necrosis. [99mTc]Glucarate uptake in the primary MDA-MB-435 shoulder tumors and metastatic lesions were clearly visualized with micro-SPECT/CT imaging. Conclusions: Selective tumor uptake and rapid clearance from nontarget organs makes [99mTc]glucarate a potential agent for breast cancer imaging that awaits validation in a clinical trial.</dcterms:abstract>

<dcterms:abstract>The current research explored the possibility of using attribute ratings as a variant (RATA: rate-allthat- apply) to CATA questions (check-all-that-apply), in order to improve sample description and discrimination and to engage participants in greater cognitive processing. The RATA question variant was implemented by asking participants, for the terms they ticked as ‘‘apply’’, to indicate intensity (using a 3-pt scale with anchors ‘low’, ‘medium’ or ‘high’) or rate applicability (using a 5-pt scale anchored at ‘slightly applicable’ and ‘very applicable’). A total of four studies with 328 consumers were conducted. Studies 1–3 involved the consumption of products (milk desserts, bread and gummy lollies), whereas Study 4 was performed with yogurt labels. A between-subjects design was used in all studies to compare product characterizations from CATA and RATA questions. Across the four studies, compared to the simple CATA questions, the RATA variant led to an increase in the total number of selected terms and a small increase in the percentage of terms for which significant differences among samples were identified. Although the stability of sample and term configurations from CATA and RATA questions, calculated using a bootstrapping re-sampling approach, were similar, for two of the four studies RATA questions provided more stable sample and term configurations. Results from the present work reveal the potential of intensity-based CATA variants with consumers for sensory product characterization, but also suggest that these may be study and sample specific.</dcterms:abstract>

<dcterms:abstract>Whey proteins are known for their high nutritional value and bioactivity, notably great potential is found on α-lactalbumin. The objective of this study was to evaluate antioxidant, antiglycant and ACE (angiotensin converting enzyme)-inhibitory activity of α-lactalbumin peptides obtained from enzymatic hydrolysis with Alcalase through different hydrolysis conditions. The optimization of enzymatic hydrolysis was studied by response surface methodology variating enzyme:substrate ratio (0.0050%, 0.0525% and 0.1000% w/w) and time (0, 30 and 60 minutes) measuring antioxidant activity by ABTS and ORAC-FL (response variables), founding an augment of this activity with time. Characterization of seven samples showed increasing hydrolysis with time. Hydrolysate obtained with 0.1000% w/w and 60 minutes demonstrated higher antioxidant activity related to greater hydrolysis. This hydrolysate did not show antiglycant activity but promoted advanced glycation end products formation with methylglyoxal and glucose. In contrast, this sample showed 30% of ACE inhibition when compared to Captopril, having great potential by enhancing hydrolysis.</dcterms:abstract>

<dcterms:abstract>Searching for prospective vanadium-based drugs for cancer treatment, a new series of structurally related [VIVO(L − 2H)(NN)] compounds (1–8) was developed. They include a double deprotonated salicylaldimine Schiff base ligand (L-2H) and different NN-polypyridyl co-ligands having DNA intercalating capacity. Compounds were characterized in solid state and in solution. EPR spectroscopy suggests that the NN ligands act as bidentate and bind through both nitrogen donor atoms in an axial-equatorial mode. The cytotoxicity was evaluated in human tumoral cells (ovarian A2780, breast MCF7, prostate PC3). The cytotoxic activity was dependent on type of cell and incubation time. At 24 h PC3 cells presented low sensitivity, but at 72 h all complexes showed high cytotoxic activity in all cells. Human kidney HEK293 and ovarian cisplatin resistant A2780cisR cells were also included to evaluate selectivity towards cancer cells and potency to overcome cisplatin resistance, respectively. Most complexes showed no detectable interaction with plasmid DNA, except 2 and 7 which depicted low ability to induce single strand breaks in supercoiled DNA. Based on the overall cytotoxic profile, complexes with 2,2´-bipyridine and 1,10-phenanthroline ligands (1 and 2) were selected for further studies, which consisted on cellular distribution and ultrastructural analyses. In the A2780 cells both depicted different distribution profiles; the former accumulates mostly at the membrane and the latter in the cytoskeleton. Morphology of treated cells showed nuclear atypia and membrane alterations, more severe for 1. Complexes induce different cell death pathways, predominantly necrosis for 1 and apoptosis for 2. Complexes alternative mode of cell death motivates the possibility for further developments.</dcterms:abstract>

<dcterms:abstract>PURPOSE: To study the rhenium-188 labeling of polyamidoamine (PAMAM) generation 4 (G4) dendrimer and its evaluation on biodistribution and chromosomal aberrations in melanoma cells induced by ionizing radiation as potential treatment agent. MATERIALS AND METHODS: Dendrimers were first conjugated with Suc-HYNIC (succinimidyl 6-hydrazinopyridine-3-carboxylic acid hydrochloride). Dendrimer-HYNIC was then incubated with 188ReO4-. Biodistribution was performed administrating 188Re-dendrimer to normal (NM) or melanoma-bearing mice (MBM). Chromosome aberration test was conducted in order to measure treatment capacity of 188Re-dendrimer in melanoma cells. RESULTS: Radiolabeling yield of dendrimer was approx. 70%. Biodistribution studies in NM showed blood clearance with hepatic and renal depuration. MBM showed a similar pattern of biodistribution with tumor uptake of 6% of injected dose. Aberrant metaphases quantified in control cells were 7%, increasing to 29.5% in cells treated with 15μCi (0.555 MBq) of 188Re-dendrimer for 24 h. CONCLUSIONS: 188Re-dendrimer can produce double-stranded breaks in DNA induced by ionizing radiation in melanoma cells in vitro.</dcterms:abstract>

<dcterms:abstract>Limited shelf life of bakery products, caused by microbial deterioration, is a concern for industries due to economic losses. Fungal spoilage of sponge cakes industrially produced in Montevideo was caused mainly by Penicillium species, in particular by Penicillium crustosum. The combination of different hurdles was studied to inhibit P. crustosum growth in sponge cakes. A full factorial design was performed to study the effect of the concentration of potassium sorbate, pH, packaging atmosphere and storage time. The results showed that packaging atmosphere and storage time were the significant factors in the ranges tested. No growth was detected in cakes stored in modified atmosphere packaging (MAP) (N2:CO2 50:50) at room temperature (25 °C) for 15 days. The effect of MAP on P. crustosum growth in cakes at room temperature was compared with the effect of air-packaging and storage at low temperature (4 °C) for 30 days. P. crustosum growth was not detected in cakes packaged in MAP, whereas it was detected after 20 days in cakes packaged in air and stored at 4 °C. This growth was quantified by a specific real time PCR developed in this work. Specific primers were designed using the sequence of β-tubulin gene of P. crustosum as a target and PCR conditions were adjusted to ensure specificity. PCR efficiency was 107%, with a detection limit of 0.0014 ng of DNA. The qPCR method presented here, resulted specific and sensitive enough to detect the growth of P. crustosum even before biodeterioration signs were visible.</dcterms:abstract>

The results shows that dendrimers with amine terminal groups (PAMAM-AT G = 3) and dendrimers with carboxylate terminal groups (PAMAM-CT G2.5 and G4.5) produces triazine derivative hydrolysis, even in buffered medium, and are not suitable as carriers. In contrast, dendrimers with neutral end groups (PAMAM-OHT) shows stable association with the active compound, making this dendrimer a possible medium for triazine carriage.

</dcterms:abstract>

<dcterms:abstract>Calendula officinalis L. (marigold) is a medicinal plant consumed at ton scale worldwide. For that reason the raw material supply cannot rely solely on the collection of wild specimens. C. officinalis is cultivated all over the world not only to cover the increasing demand but also to ensure its regular quality. When conventional cropping systems are employed, the use of pesticides to protect the cultures against pests is a common practice. The presence of contaminants in herbs for medicinal purposes is strictly ruled by Pharmacopoeias. The European pharmacopoeia, as well as the MERCOSUR, require validated methods to determine the content of pesticide residues in each specific medicinal plant. As a contribution to accomplish such requirements, in the present work a comparison of the performance of three modern multirresidue methods based on QuEChERS methodology was conducted. Aiming to find a method fit for that purpose, the original, unbuffered version and variationss of the AOAC 2007.01 and the EN 15662 protocols were evaluated in marigold for the determination of 24 pesticide residues using LC–MS/MS. The three evaluated methods where accurate and precise for all the studied compounds but the original QuEChERS extracts causes less matrix effects over the studied pesticides. After the evaluation of the figures of merits, the selected methodology was validated according to the SANTE guidelines. LOQs were determined in the range 10–100 μg kg−1 showing a linear behaviour (r2 > 0.993) from LOQ to 400 μg kg−1. The method was applied in commercial samples where atrazine, imidacloprid, pirimiphos-methyl and methalaxyl residues were detected.</dcterms:abstract>

<dcterms:abstract>The aim of the present work was to evaluate the possibility of a fast, simple, and greener sample preparation method using electrospun fibers as SPE sorbents and its application for doping control analysis. Electrospun polymer fibers of polystyrene with smooth morphology (1.3 μm average diameter) were used as sorbent materials for the fabrication of miniaturized solid phase extraction devices. A microcolumn SPE device (10 mg of electrospun fibers packed in a 200 μL pipette tip) and a SPE disk device (10 mg of electrospun fibers packed in a 500 μL polypropylene barrel) were used. Their performance was evaluated using dexamethasone and betamethasone as model analytes for the analysis of drug residues in biological samples. A qualitative confirmatory method for the detection of these glucocorticosteroids by HPLC-MS/MS in human urine was developed using electrospun microfibers for sample clean-up and pre-concentration. The limit of detection for both analytes was 10 ng mL−1 in urine matrix, and the resulting analytical method is at the same time fit for purpose (complying with strict requirements of doping control international organizations), simple (using tip-based microcolumns) and has a greener footprint (using very small amounts of organic solvents).</dcterms:abstract>

<dcterms:abstract>The influence of the cooking method on the elemental composition of popcorn, prepared according to the way it is consumed, was evaluated. Arsenic, calcium, cadmium, copper, iron, lead, magnesium, manganese, potassium, selenium, sodium and zinc levels were determined using atomic absorption spectrometry. Results showed that the losses after cooking varied considerably (2.9%–48.3%) depending on the element. A bioaccessibility study of cooked samples was also performed, using an in vitro approach. The most bioaccessible elements were sodium (77.4%–88.5%) and potassium (63.7%–81.8%), while considerably low values were observed for calcium (5.5%–13.7%) probably due to strong interactions with the matrix. The contribution to the Recommended Dietary Allowance was evaluated, finding an important coverage for selenium (36.7%– 50.0%). Positive Pearson correlation coefficients were found for the pairs Cu/Fe, Cu/Mn, Fe/Mn Cu/K, Fe/K, K/Mn which can be explained by natural physiological mechanisms of the plant. To the best of our knowledge, this is the first study of bioaccessibility of essential elements in Uruguayan popcorn

<dcterms:abstract>Two straightforward methods for glyphosate and AMPA analysis in honey through ion chromatography coupled to Q-Orbitrap accurate high-resolution mass spectrometry as well as their application to real samples are presented. The sample preparation step is accomplished through dilution with methanol–water or water. No pH adjustment is necessary. Recoveries were in the range 80 to 110% with RSDs < 20%. The LOQ of glyphosate was 0.005 mg kg−1, far below the allowed EU-MRL (0.05 mg kg−1) in honey, showing a linear range of 0.005–0.5 mg kg−1 and medium matrix effect. LOQ of AMPA was 0.02 mg kg−1. Glyphosate quantitation was performed with internal or external calibration yielding identical results, thus broadening the applicability of the method as isotopically labelled standards are no longer mandatory. The method was applied to 32 honey samples from different origins. Glyphosate was found in 81% of the samples and 41% were above the EU-MRL showing its applicability in routine work. AMPA was not detected in any of the samples.</dcterms:abstract>

<dcterms:abstract>The objectives of this study were: to assess the efficiency of high hydrostatic pressure or ultra-high pressure homogenization against Mycobacterium smegmatis in milk and to discuss whether M. smegmatis can be considered a suitable surrogate for other Mycobacterium spp. in high pressure inactivation trials using milk. Three strains of this specie (CECT 3017, 3020 and 3032) were independently inoculated into both skimmed (0.2% fat) and whole milk (3.4% fat) at an approximate load of 6.5 Log CFU/ml and submitted to HHP treatments at 300, 400 or 500 MPa for 10 m at 6°C and 20°C. Evolution of the surviving cells of the inoculated strains was evaluated analysing milk immediately after the treatments and after 5 and 8 d of storage at 6°C. HHP treatments at 300 MPa were seldom efficient at inactivating M. smegmatis strains, but lethality increased with pressure applied in all cases. Generation of sub-lethal injured cells was observed only after 400 MPa treatments since inactivation at 500 MPa was shown to be complete. Significant differences were not observed due to either temperature of treatment or fat content of milk, except for strain CECT3032, which was shown to be the most sensitive to HHP treatments. Milk inoculated with strain CECT3017 was submitted to ultra-high pressure homogenization (UHPH) treatments at 200, 300 and 400 MPa. Maximum reductions were obtained after 300 and 400 MPa treatments, although less than 3.50 Log CFU/ml were inactivated. UHPH did not cause significant number of injured cells. The usefulness of this species as a marker for pressure-based processing seems limited since it showed greater sensitivity than some pathogenic species including other Mycobacteria reported in previous studies.</dcterms:abstract>

<dcterms:abstract>Astringency has been reported to be strongly time-dependent and to exhibit buildup on repeated ingestions. A common approach to deal with this phenomenon during astringency evaluation is the use of palate cleansers. The aim of this work was to evaluate palate cleansers for red wine astringency evaluation, considering both reduction of astringency buildup and sample discrimination. Fourteen trained panelists evaluated two sets of four Tannat wine samples using time-intensity methodology. In the first sample set the same wine was presented four times, while the second one comprised four samples with different added concentrations of added grape seed tannins. Each assessor evaluated both sample sets with five different palate cleansers (still mineral water, plain unsalted crackers, skimmed milk, drinkable plain sweetened yogurt and 2 g/L pectin solution) in triplicate. None of the evaluated palate cleansers could prevent the occurrence of astringency buildup, while sample discrimination ability differed among the five palate cleansers considered.</dcterms:abstract>

The purpose of this study was to explore the potential of this tracer to evaluate malignant head and neck tumors by means of SPECT/CT software fusion imaging. Methods: Eleven male patients with advanced head and neck carcinoma were included in the study: 9 with locally advanced disease and 2 with clinical suspicion of local relapse. Scanning started 3–6 h after the injection of 1,110 MBq of 99mTc-glucarate. Planar and SPECT images of the head, neck, and thorax were acquired. Three-dimensional images were also coregistered with CT. Results: We found 99mTc-glucarate uptake in all suspected lesions. SPECT/CT fusion imaging was helpful in all cases for topographically localizing the tracer foci. Conclusion: 99mTcglucarate can be considered a potential tracer for the evaluation of patients with head and neck tumors.</dcterms:abstract>

<dcterms:abstract>The goals of the current trial were (a) to characterize the plasma disposition kinetics of levamisole (LEV), albendazole (ABZ) and ivermectin (IVM), each administered either alone (single active ingredient) or as a combined formulation to lambs; (b) to compare the clinical anthelmintic efficacy of the same drugs given either separately or co-administered to lambs infected with resistant nematodes. Fifty Corriedale lambs naturally infected with multiple resistant gastrointestinal nematodes were involved in the following experimental trials: (a) "Pharmacokinetic trial": the animals were allocated into five groups (n=10 each) and intraruminally treated with either LEV (8 mg/kg), ABZ (5mg/kg), IVM (0.2mg/kg), or with a LEV+ABZ+IVM combined formulation, where each active ingredient was administered at the same dose. Blood samples were collected over 15 days post-treatment and drug plasma concentrations measured by HPLC. (b) "Efficacy trial": the same treated groups plus an untreated control group were used to assess the comparative anthelmintic efficacy by the faecal egg count reduction test (FECRT). Although the overall LEV disposition kinetics was unaffected, significantly lower (61%) ABZ-sulphoxide and higher (71%) IVM systemic availabilities were obtained after administration of the combined formulation in comparison to those obtained after treatment with each drug alone. A multiple drug resistance situation was observed for Haemonchus spp. The observed efficacies were 52% (LEV), 72% (ABZ), 80% (IVM) and 87% (triple combined formulation). The results reported here contribute to the pharmaco-therapeutic knowledge on drug combinations. This type of research is crucial before further development of combined anthelmintic preparations reaches the market to deal with resistant nematode control. The co-administration of LEV+ABZ+IVM did not result in a significant advantageous anthelmintic effect compared to the treatment with IVM alone. The simultaneous/combined administration of LEV, ABZ and IVM may account for a drug-drug pharmacological interaction in infected lambs. The pharmacokinetic interaction accounted for a reduced ABZ-sulphoxide and enhanced IVM systemic exposure following the combined treatment.</dcterms:abstract>

<dcterms:abstract>Herbal teas and yerba mate infusions are an important part of the human diet that may contain toxic contaminants due to the industrial processing of the herbal material. The presence of the lipophilic compounds polycyclic aromatic hydrocarbons (PAHs) was investigated in dry yerba mate samples and how they transfer into the infusions. Samples were selected among eight different brands, and three types of yerba mate products (aged, green, and roasted). Synchronized fluorescence scanning (SFS) was used as primary screening method for PAHs in dried yerba mate, and high-per formance liquid chromatography with fluorescence detection for the ana lysis of the infusions. SFS screening showed presence of PAHs in the dry herbal material, and the PAHs ratios suggest that the contamination origi nates from pyrogenic sources. Using an experimental setup to simulate the traditional yerba mate consumption (successive extractions with small amounts of water), the sum of the six PAHs in each infusion of yerba mate sample ranged from 4.8 to 30.2 ng g 1 . Benzo [a] pyrene concentration ranged from n.d. to 1.1 ng g 1 while phenanthrene reached a maximum of 59 ng g 1 in the yerba mate samples tested. An estimated 0.11% to 0.54% of the six PAH analytes were transferred into the infusion and the SFS method can be used to screen the presence of PAHs in the dry mater ial. The chemical profile of the most abundant PAHs found in the infusions indicate a low total equivalent exposure index.</dcterms:abstract>

<dcterms:abstract>Biorsorbents can be used to pack minicolumns for the preconcentration of trace elements and thus improve the detection limits of some techniques. A biosorbent is efficient when presents binding sites as carbonyls, amines, and carboxylic groups, among others. Thus, natural materials as the sisal fiber (Agave sisalana) may be a good candidate. Sisal fiber demonstrated good performance for Cu preconcentration when it was impregnated with alizarine fluorine blue (AFB). However, very good results were reported for the first time by our group with this column, without the use of additional reagents for both, Mn and Cu determinations in water samples by using microwave plasma atomic emission spectroscopy (MP AES). In this paper a comparison of the sisal fiber preparation with and without impregnation with AFB is presented and discussed in terms of the figures of merit, including precision, trueness, limits of detection and quantification. In addition to that, the number of determinations without the need of replacement of the solid phase was evaluated. Results demonstrated that the impregnation of sisal fiber with AFB does not leads to an improvement in the analytical performance. Analytical Greenness Metric Approach (AGREE) was used to evaluate the greenness of both methods and results obtained were similar. Despite this, the method without impregnation of the fiber has some remarkable advantages, that contributes to Green Analytical Chemistry (GAC), related to the lifetime of the sorbent, not considered in the AGREE tool.</dcterms:abstract>

<dcterms:abstract>The superoxide dismutase-like activity of a series of copper(II) complexes of the aminoacids glycine, alanine, valine, isoleucine, and serine has been investigated using the nitroblue tetrazolium/superoxide reduction assay. The results show that all the investigated complexes possess the capability to dismutate the superoxide anion generated in the xanthine/xanthine oxidase system. The results are compared with those obtained for the native enzyme superoxide dismutase, tested under the same experimental conditions. The lipophilicity of the complexes was tested determining their partition coefficients between physiological solution and n-octanol. The results show an important increment in lipophilicity with increasing complexity of the ligands and also in comparison with that of the free aminoacids.</dcterms:abstract>
