<dcterms:abstract>We studied the accumulation and depuration of microcystin-LR (MCLR) in the hepatopancreas of the crab Neohelice granulata fed twice weekly with either non toxic or MCLR-producing Microcystis aeruginosa (strain NPDC1 or NPJB, respectively) during seven weeks. We also analyzed MCLR effects on the oxidative stress- and detoxification-related variables, superoxide dismutase and glutathione-S-transferase activities, and the levels of reduced glutathione and lipid peroxidation (as thiobarbituric acid reactive substances, TBARS).

Hepatopancreas MCLR content slightly increased during the first three weeks, up to 8.81±1.84 ng g−1 wet tissue mass (WTM) and then started to decrease to a minimum of 1.57±0.74 ng g−1 WTM at the seventh week (p<0.05 with respect to that in the first week). TBARS levels were about 55% higher in treated than in control N. granulata (p<0.001 and p<0.05) during the first three weeks of the experimental period. GSH content became 50% lower than in control individuals (p<0.01) during weeks 6 and 7. SOD activity was increased by about 2-fold (p<0.05 or p<0.001) from week 3 to 7 in treated crabs with respect to control ones, while GST activity was about 70% higher in treated than in control crabs from week 4 to week 7 (p<0.05).

Our data suggest that in the hepatopancreas of N. granulata MCLR accumulation and oxidative damage are limited and reversed by detoxification-excretion and antioxidant mechanisms. The activation of these defensive mechanisms becomes evident at 3-4 weeks after the start of the intoxication.

</dcterms:abstract>

A total of 51 patients (15 women, 36 men) with different characteristics (pathologies, comedications, age, habits, etc) and taking a standard dose of omeprazole for more than three months were included. 17.6 % of the patients showed mild hypomagnesemia but we concluded that the observed low levels of this electrolyte could be attributed to comedications, age and different pathologies rather than the 20-mg dose of omeprazole. So, hypomagnesemia does not eliminate proton-pump inhibitors as a reasonable option; it just requires clinicians to be aware of this problem and use them safely at conventional doses.</dcterms:abstract>

    <dcterms:abstract>The Baccharis genus has more than 400 species of aromatic plants. However, only approximately 50 species have been studied in oil composition to date. From these studies, very few take into consideration differences between male and female plants, which is a significant and distinctive factor in Baccharis in the Asteraceae family. Baccharis articulata is a common shrub that grows wild in south Brazil, northern and central Argentina, Bolivia, Paraguay and Uruguay. It is considered to be a medicinal plant and is employed in traditional medicine. We report B. articulata male and female volatile composition obtained by simultaneous distillation–extraction technique and analyzed by gas chromatography with mass spectrometry. Also, an assessment of aromatic differences between volatile extracts was evaluated by gas chromatography with olfactometry. The results show a very similar chemical composition between male and female extracts, with a high proportion of terpene compounds of which β-pinene, limonene and germacrene D are the main components. Despite the chemical similarity, great differences in aromatic profile were found: male plant samples exhibited the strongest odorants in number and intensity of aromatic attributes. These differences explain field observations which indicate differences between male and female flower aroma, and might be of ecological significance in the attraction of pollinating insects.</dcterms:abstract>

    <dcterms:abstract>Five novel ruthenium(II)–arene complexes with polycyclic aromatic ligands were synthesized, comprising three compounds of the formula [RuCl(η6-p-cym)(L)][PF6], where p-cym = 1-isopropyl-4-methylbenzene and L are the bidentate aromatic ligands 1,10-phenanthroline-5,6-dione, 1, 5-amine-1,10-phenanthroline, 4, or 5,6- epoxy-5,6-dihydro-phenanthroline, 5. In the other two complexes [RuCl2(η6-p-cym)(L′)], the metal is coordinated to a monodentate ligand L′, where L′ is phenanthridine, 2, or 9-carbonylanthracene, 3. All compounds were fully characterized by mass spectrometry and elemental analysis, as well as NMR and IR spectroscopic techniques. Obtained ruthenium compounds as well as their respective ligands were tested for their antiparasitic and antitumoral activities. Even though all compounds showed lower Trypanosoma brucei activity than the free ligands, they also resulted less toxic on mammalian cells. Cytotoxicity assays on HL60 cells showed a moderate antitumoral activity for all ruthenium compounds. Compound 1 was the most potent antitumoral (IC50 = 1.26±0.78 μM) and antiparasitic (IC50=0.19±0.05 μM) agent, showing high selectivity towards the parasites (selectivity index N100). As complex 1was themost promising antitumoral compound, its interactionwith ubiquitin as potential target was also studied. In addition, obtained ruthenium compounds were found to bind DNA, and they are thought to interact with this macromolecule mainly through intercalation of the aromatic ligand.</dcterms:abstract>

    <dcterms:abstract>The concurrent use of CATA questions (check-all-that-apply) with hedonic scaling in product research with consumers has gained acceptance in recent years. However, CATA questions do not enable product developers to identify consumers’ preferred intensity of a sensory attribute directly, and for this reason just-about-right (JAR) questions are sometimes used together with questions that quantify attribute presence and/or intensity. Using JAR questions, consumers have to describe each of the samples in terms of each specific attribute, typically: 1 = ‘Not enough…’, 3 = ‘Just about right’, and 5 = ‘Much too…’. Past research has suggested that co-elicitation of hedonic and JAR responses bias the hedonic data. This undesired effect could extend to the case where JAR questions are used concurrently with hedonic and CATA questions. In seven studies (N = 762) across seven product categories no such evidence was found. Additionally, there was some evidence pointing to an increase in product discrimination based on hedonic and CATA responses. Preliminary data indicated that the inclusion of JAR questions in product tests also eliciting hedonic and CATA responses did not negatively affect consumers’ perception of the task, which, on average, was still regarded as easy and not tedious. Future research is required to validate the findings across more varied experimental settings.</dcterms:abstract>

    <dcterms:abstract>Concurrent elicitation of hedonic and sensory information from consumers helps to identify the sensory attributes that drive consumer preferences. However, there is a risk that asking consumers about specific sensory characteristics can bias their hedonic responses. This research extends previous work by investigating if concurrent use of rate-all-that-apply (RATA) questions, a variant of CATA questions is associated with bias of the hedonic scores. RATA questions extend CATA questions by requiring consumers to also rate the intensity of the terms that are applicable for describing samples (‘high’, ‘medium’ and ‘low’). In eight consumer studies (n = 560) across five product categories there was no evidence of concurrent use of RATA questions influencing hedonic scores. When RATA questions were used concurrently with a hedonic question there was a strong tendency for greater hedonic discrimination between samples (5 of 8 studies). This suggested that the discriminative capacity of the hedonic question was increased by inclusion of the RATA question, a result that could be attributable to the RATA question increasing consumers’ engagement and attention to the task. Further research is required to confirm this result and tentative explanation.</dcterms:abstract>

    <dcterms:abstract>Intermittent drainage is one of the most promising approaches to mitigate methane (CH 4 ) emission from paddy fi elds. Irrigated rice fi elds in Uruguay are temporarily established on soils used as cattle pastures. We studied soil from the pasture-rice rotation (UR) as well as soil from a permanent cattle pasture (UT) hypothesizing that activity and structure of the bacterial and archaeal communities involved in pro- duction of CH 4 change systematically with intermittent drainage. Methane production started after 7 days and 16 days of anoxic incubation in UR and UT soil, respectively. Then, production rates of CH 4 were higher in UT than UR soil. Intermittent drainage signi fi cantly decreased the rates of CH 4 production. Analysis of d 13 C indicated that CH 4 was mainly produced from acetate both in UR (73 e 98%) and UT (51 e 80%) soil. Intermittent drainage did not change the pathway of CH 4 production. Quantitative PCR showed that methanogenic archaeal gene copy numbers (16S rRNA, mcrA ) were much lower in UT than UR soil, but increased upon incubation under anoxic conditions. Terminal restriction fragment length polymorphism (T-RFLP) and pyrosequencing of bacterial and archaeal 16S rRNA genes showed that the communities were clearly different between UR and UT soil. The bacterial community consisted of 9 phyla with relative abundance of > 1% in both soils. Whereas the archaeal community in UR soil was dominated by Methanocellales and Methanosarcinaceae , that in UT soil was dominated by Crenarchaeota . Anoxic incubation affected the composition of the bacterial and archaeal communities in UT soil, but not so much in UR soil. In UT soil, the relative abundance of Clostridiales increased to 19%, and the archaeal community changed to dominance by Methanosarcinaceae and Methanobacteriales . Subsequent drainage and re- fl ooding, however, had comparatively little effect on the composition, although it decreased the rates of CH 4 production in both soils. Difference in previous soil management and in the structures of the microbial communities apparently only affected their dynamics and functioning after the fi rst fl ooding but not upon subsequent drainage and re- fl oodin</dcterms:abstract>

    <dcterms:abstract>Plant growth-promoting bacteria of the genus Azospirillum are present in the rhizosphere and as endophytes of many crops. In this research we studied 40 Azospirillum strains isolated from different plants and geographic regions. They were first characterized by 16S rDNA restriction analysis, and their phylogenetic position was established by sequencing the genes 16S rDNA, ipdC, hisC1, and hisC2. The latter three genes are involved in the indole-3-pyruvic acid (IPyA) biosynthesis pathway of indole-3-acetic acid (IAA). Furthermore, the suitability of the 16S-23S rDNA intergenic spacer sequence (IGS) for the differentiation of closely related Azospirillum taxa and development of PCR protocols allows for specific detection of strains. The IGS-RFLP analysis enabled intraspecies differentiation, particularly of Azospirillum brasilense and Azospirillum lipoferum strains. Results demonstrated that the ipdC, hisC1, and hisC2 genes are highly conserved in all the assessed A. brasilense isolates, suggesting that these genes can be used as an alternative phylogenetic marker. In addition, IAA production determined by HPLC ranged from 0.17 to 98.2 μg mg(-1) protein. Southern hybridization with the A. brasilense ipdC gene probe did not show, a hybridization signal with A. lipoferum, Azospirillum amazonense, Azospirillum halopreferans and Azospirillum irakense genomic DNA. This suggests that these species produce IAA by other pathways. Because IAA is mainly synthesized via the IPyA pathway in A. brasilense strains, a species that is used worldwide in agriculture, the identification of ipdC, hisC1, and hisC2 genes by PCR may be suitable for selecting exploitable strains</dcterms:abstract>

    <dcterms:abstract>While it is known that toxic metals contribute individually to child cognitive and behavioral deficits, we still know little about the effects of exposure to multiple metals, particularly when exposures are low. We studied the association between children’s blood lead and hair arsenic, cadmium, and manganese and their performance on the Bayley Scales of Infant Development III. Ninety-two preschool children (age 13–42 months) from Montevideo, Uruguay, provided a hair sample and 78 had a blood lead level (BLL) measurement. Using latent class analysis (LCA), we identified four groups of exposure based on metal concentrations: (1) low metals, (2) low-to-moderate metals, (3) high lead and cadmium, and (4) high metals. Using the four-group exposure variable as the main predictor, and fitting raw scores on the cognitive, receptive vocabulary, and expressive vocabulary scales as dependent variables, both complete-case and multiple imputation (MI) analyses were conducted. We found no association between multiple-metal exposures and neurodevelopment in covariate-adjusted models. This study demonstrates the use of LCA together with MI to determine patterns of exposure to multiple toxic metals and relate these to child neurodevelopment. However, because the overall study population was small, other studies with larger sample sizes are needed to investigate these associations.</dcterms:abstract>

    <dcterms:abstract>Children´s exposure to inorganic arsenic (iAs) is a concern because of its effects on growth and development, and its carcinogenic properties. Low level As exposure is prevalent but its sources are understudied. We investigated the contribution of diet to iAs exposure and arsenic methylation in 1st-graders (6-8 y, 57.3% boys) from Montevideo. First-morning urine samples were analyzed for iAs metabolites (MMA, DMA and remaining iAs) by high-pressure liquid chromatography with hydride generation and inductively coupled mass spectrometry. Caregivers provided two 24-hr recalls of children’s diets. Principal component analysis identified 6 dietary patterns, explaining 41% of variability in food intake: 1) high intake of red meat, yogurt and sweetened beverages, 2) breads, processed meats and fats, 3) dark green leafy vegetables and white meat, 4) vegetables and eggs, 5) sweets, chips and fried foods, and 6) fruit. Consumption of rice, corn, and grain-based foods was also analyzed. Sp. gravity adjusted urinary arsenic metabolite concentrations (U-As) and metabolite fractions (%MMA, %DMA and %iAs) were modeled as dependent variables with dietary components as predictors, adjusted for sex and BMI- for-age Z score. Models with metabolite fractions were adjusted for U-As. 192 children had urine samples, 188 had diet recall. U-As was fairly low (median, range: 9.93, 2.8-48.1 µg/L). Children scoring high on the fruit pattern had higher U-As (ß±SE: 1.0±04 p<0.05). Children consuming more rice and grains also had higher U-As (0.1±0.03 and 0.02±0.01, respectively, p<0.01). Interestingly, children scoring high on the red meat, yogurt and sweetened beverages, and dark green vegetables and white meats pattern had higher %DMA (0.5±0.3, and 0.9±0.3, respectively, p<0.01) and lower %iAs (-0.4±0.2 and -0.5±0.2, respectively, p<0.1). Children’s food patterns and consumption of specific foods may contribute to inorganic As exposure and affect their ability to methylate arsenic.
<dcterms:abstract>In the search for new anthelmintics able to overcome the resistance problem against all available drugs in livestock, the synthesis of novel valerolactam-benzimidazole hybrid compounds was reported. This allowed us to obtain these in vitro and in vivo bioactive compounds using Nippostrongylus brasiliensis rat model by integrating physiology-based assays and ex vivo diffusion studies. In order to further study those novel hybrid molecules, Haemonchus contortus (a sheep gastrointestinal nematode of interest) and Mesocestoides vogae tetrathyridia (a useful system to study the efficacy of anthelmintic drugs against cestoda) were used as parasite models to compare the ex vivo patterns of diffusion and biotransformation of benzimidazoles and their valerolactam-benzimidazole hybrid derivatives. On average, a nine-fold higher intraparasitic concentration of compounds was found in M. vogae compared with H.contortus, with similarities regarding the order of entry of compounds, highlighting febendazole (FEB) and its hybrid compound 10, while valerolactam compound 2 practically did not penetrate the parasites. Interestingly, sulphoxidation drug metabolism was observed and measured, revealing percentages of oxidation of 8.2% and 14.5% for albendazole (ABZ) and febendazole respectively in M. vogae, while this effect was more relevant in H. contortus parasite. More importantly, significant differences were observed between anthelmintic-susceptible adult parasites (Hc S) and those from sheep farms (Hc U). In fact, the percentages of oxidation of FEB and the hybrid compound 8 were higher in Hc U (25.5%, 54.1%, respectively) than in Hc S (8.8%, 38.2%). Interestingly, sulphoxidation of hybrid compound 10 was neither observed in M. vogae nor in H. contortus parasites, suggesting that increased drug metabolism (oxidation reactions) could not be used by these parasites as a defense mechanism against this novel drug.</dcterms:abstract>

    <dcterms:abstract>Reducing the sugar content of processed products has been claimed to be one of the most efficient strategies for decreasing sugar intake. The present work aimed at studying the influence of sugar reduction on the dynamic sensory profile and consumers' liking of probiotic chocolate-flavored milks using a novel temporal methodology, and to evaluate two alternatives (vanilla flavor and thaumatin) to attenuate the sensory changes caused by sugar reduction. Probiotic chocolate-flavored milks were formulated with different reductions in added sugar (0, 20, 40 and 60%). Vanilla flavor and thaumatin were added to the sugar-reduced samples at two concentrations. Samples were evaluated by trained assessors using Temporal check-all-that-apply (TCATA). Additionally, consumers evaluated the dynamic sensory profile of a subset of the samples using TCATA and indicated their overall liking using a 9-point hedonic scale. Results from the present work showed that the main effect of sugar reduction on the dynamic sensory profile of the probiotic chocolate-flavored milks was related to changes in sweetness, bitterness and thickness. A reduction in added sugar of 20% led to changes in sweetness intensity, which were perceived by both trained assessors and consumers. However, consumers' liking was not significantly affected by sugar reduction up to 40%. The addition of vanilla flavor at suprathreshold concentrations was not efficient in increasing sweetness perception in chocolate-flavored milks with the lowest sugar reduction percentage, suggesting that it may not be a feasible alternative for reducing sugar in this product category. These results suggest that in many situations sugar content of food products could be decreased without a relevant impact on consumers' sensory and hedonic perception.</dcterms:abstract>

The structure of 1 consists of [Re(NO)Br4(Hnic)]− anions and NBu4+ cations. Each Re(II) is six-coordinate with four bromide ligands, a linear nitrosyl group and a nitrogen atom from the Hnic molecule, in a distorted octahedral surrounding. The structures of 2–5 are made up of discrete heterodinuclear Re(II)M(II) units where the fully deprotonated [Re(NO)Br4(nic)]2− entity acts as a didentate ligand through the carboxylate group towards the [Ni(dmphen)2]2+ (2), [Co(dmphen)2]2+ (3), [Mn(dmphen)(H2O)2]2+ (4) and [Cu(bipy)2]2+ (5) fragments, the Re–M separation across the nic bridge being 7.8736(8) (2), 7.9632(10) (3), 7.7600(6) (4) and 8.2148(7) Å (5). The environment of the Re(II) ion in 2–5 is the same as that in 1 and all M(II) are six-coordinate in highly distorted octahedral surroundings, the main source of the distortion being due to the reduced bite of the chelating carboxylate. The magnetic properties of 1–5′ were investigated in the temperature range 1.9–300 K. 1 behaves as a quasi-magnetically isolated spin doublet with very weak antiferromagnetic interactions through space Br⋯Br contacts. Its magnetic susceptibility data were successfully modeled through a deep analysis of the influence of the ligand field, spin–orbit coupling, tetragonal distortion and covalence effects as variable parameters. Compounds 2–5′ exhibit weak antiferromagnetic interactions. The intramolecular exchange pathway in this family being discarded because of the symmetry of magnetic orbitals of the Re(II) ion (dxy) precludes any spin delocalization on the bridging nic orbitals, the observed magnetic interactions are most likely mediated by π–π type interactions between the peripheral ligands which occur in them. Only in the case of 4, short through space Br⋯Br contacts of ca. 4.03 Å (values larger than 5.5 Å in 2, 3 and 5) could be involved in the exchange coupling.</dcterms:abstract>

    <dcterms:abstract>Synopsis
Introduction

The use of bioadhesive hydrogels for skin care presents important advantages such as long residence times on the application site and reduced product administration frequency.
Object

The aim of the present work was to develop bioadhesive hydrogels for skin application, using caffeine as a model active ingredient.
Methods

Eight hydrogels were formulated using binary combinations of a primary polymer (carbomer homopolymer type C (Carbopol® 980) or kappa carrageenan potassium salt (Gelcarin® GP-812 NF)) and a secondary polymer (carbomer copolymer type B (Pemulen™ TR-1), xanthan gum or guar gum). Hydrogels were characterized by means of physico-chemical (dynamic rheological measurements, spreadability and adhesion measurements) and sensory methods (projective mapping in combination with a check-all-that-apply (CATA) question). Caffeine hydrogels were formulated using two of the most promising formulations regarding adhesion properties and sensory characteristics. In vitro active ingredient release studies were carried out.
Results

Hydrogel formulations showed a prevalently elastic rheological behaviour. Complex viscosity of carbomer homopolymer type C hydrogels was higher than that of the kappa carrageenan hydrogels. Besides, complex viscosity values were dependent on the secondary polymer present in the formulation. Significant differences among hydrogels were found in detachment force, work of adhesion and spreading diameter results. Association of projective mapping with CATA allowed to determine similarities and dissimilarities among samples. Cluster analysis associated the samples in two groups. Two hydrogels were selected to study the release of caffeine. Both hydrogels presented similar release profiles which were well described by the Higuchi model. Caffeine release was exclusively controlled by a diffusive process.
Conclusion

Physico-chemical and sensory techniques enabled the identification of bioadhesive hydrogel formulations with positive characteristics for cosmetic applications. Formulations which combined carbomer homopolymer type C with xanthan gum or with carbomer copolymer type B were the most promising for bioadhesive skin products. Caffeine release profiles of selected formulations were not statistically different. Both hydrogels gradually released the active ingredient, reaching approximately 80% within the first 5 h, and their profiles were well described by the Higuchi model. In this context, it could be concluded that the selected hydrogels are suitable bioadhesive hydrogel formulations for cosmetic application on the skin.

</dcterms:abstract>

    <dcterms:abstract>A simple and fast ultrasound-assisted procedure for the determination of iron and zinc in infant formulas is presented. The analytical determinations were carried out by flame atomic absorption spectrometry. Multivariate experiments were performed for optimization; in addition, a comparative study was carried out using two ultrasonic devices. A method using an ultrasonic bath was selected because several samples can be prepared simultaneously, and there is less contamination risk. Analytical precision (sr(%)) was 3.3% and 4.1% for iron and zinc, respectively. Trueness was assessed using a reference material and by comparison of the results obtained analyzing commercial samples using a reference method. The results were statistically equivalent to the certified values and in good agreement with those obtained using the reference method. The proposed method can be easily implemented in laboratories for routine analysis with the advantage of being rapid and in agreement with green chemistry.</dcterms:abstract>

    <dcterms:abstract>Pathogenic Gram-negative bacteria resistant to almost all β-lactam antibiotics are a major public health threat. Zn(II)-dependent or metallo-β-lactamases (MBLs) produced by these bacteria inactivate most β-lactam antibiotics, including the carbapenems, which are “last line therapies” for life-threatening Gram-negative infections. NDM-1 is a carbapenemase belonging to the MBL family that is rapidly spreading worldwide. Regrettably, inhibitors of MBLs are not yet developed. Here we present the bisthiazolidine (BTZ) scaffold as a structure with some features of β-lactam substrates, which can be modified with metal-binding groups to target the MBL active site. Inspired by known interactions of MBLs with β-lactams, we designed four BTZs that behave as in vitro NDM-1 inhibitors with Ki values in the low micromolar range (from 7 ± 1 to 19 ± 3 μM). NMR spectroscopy demonstrated that they inhibit hydrolysis of imipenem in NDM-1-producing Escherichia coli. In vitro time kill cell-based assays against a variety of bacterial strains harboring blaNDM-1 including Acinetobacter baumannii show that the compounds restore the antibacterial activity of imipenem. A crystal structure of the most potent heterocycle (L-CS319) in complex with NDM-1 at 1.9 Å resolution identified both structural determinants for inhibitor binding and opportunities for further improvements in potency.</dcterms:abstract>
