<dcterms:abstract><![CDATA[The existent research literature of Dye Sensitized Solar Cells (DSSC) is mainly focused on the dye but there are very few studies focused in the semiconductor. Here, we present the performance of hydrogen titanate nanotubes (HTNT), showing an enhancement of the solar cell efficiency from 5.95% to 7.63%, respect to anatase as photoelectrode. This enhancement could be explained in terms of open-circuit voltage, directly associated to the value of the HTNT band gap (Eg = 3.3 eV), the relative position of the edge conduction band; and the increase of the short-circuit current, directly associated to the enhancement of the specific area of the photoelectrode. All these characteristics are in very good concordance with our theoretical modelling of the system, based on First Principles calculations. We expect that this system could be further characterized, utilizing different dyes or absorbers, in order to evaluate the potential use of hydrogen titanate nanotubes for solar cell applications.]]></dcterms:abstract>

Broccoli is a highly perishable vegetable with unique nutritional characteristics. Development of minimally processed broccoli products demands varietal-specific knowledge as to which are the packaging conditions that preserve quality throughout shelf life. ‘Legacy’ cultivar broccoli florets were washed, disinfected, packaged in polypropylene and stored at 4, 8 and 15 °C for 21 d. Weight loss, internal atmosphere composition, respiration rate, color, texture, glucosinolate content, antioxidant capacity (AOC) and sensory attributes were evaluated throughout storage time. Results showed that 4 °C helped preserve sensory quality, texture, total glucosinolate content and AOC for 21 d. Temperature fluctuations reaching 15 °C resulted in loss of total glucosinolate content and unacceptable sensory quality. MAP helped mitigate temperature effects, especially at 8 °C. MAP is therefore an appropriate technology which can be applied to extend the shelf-life of ready-to-eat broccoli florets.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[The preparation of oxyfluoroborate glasses and glass–ceramics doped with Yb3+ and Er3+ with improved luminescent properties is reported. Glasses with compositions (100 − x − y) (MO·2B2O3) + xPbF2 + y(YbF3:ErF3) (% in mol), where M is Sr, Ba, Ca, x = 10–40% and y = 0, 1 or 5% were prepared by the melt/quenching technique. Glass crystallization was studied using thermal analysis and X-ray diffraction techniques. Optical absorption and infrared up-conversion were studied on both glasses and glass–ceramics. The incorporation of a fluoride compound into the borate glass resulted to depend on the ionic radius of the glass modifier: as it increases, glasses become more stable against crystallization. On the other hand, alkali–fluoride compounds such as BaF2 and SrF2 can be crystallized from these systems by selecting an appropriate proportion between their components. Furthermore, the up-conversion response can be tuned by changing the glass modifiers (Sr, Ba and Ca) of the borate matrix, which also influence the type of fluoride crystallized compound. The strontium glass–ceramics have the highest luminescence response due to the crystallization of SrF2 compound in the system SrO–B2O3–PbF2. Meanwhile, the lowest luminescence signal was obtained for samples in the system BaO–B2O3–PbF2 where the Pb1.33Ba2.66B11FO20 phase crystallizes. With this strategy, new materials with improved luminescence properties that can be used as up converters, were obtained.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Glycans present in biological glycoconjugates have several structural and functional roles. Elucidation of glycan structure and biological function is critical to understand their role in physiological and pathogenic process, enabling the development of diagnostic methods and disease treatment. Immobilized glycosidases are powerful tools for glycan analysis, as they are able to remove specific carbohydrates without altering the protein structure. Here we describe the individual immobilization of Aspergillus oryzae β-galactosidase and Canavalia ensiformis α-mannosidase onto agarose and silica magnetic nanoparticles activated with cyanate ester groups. High immobilization yields (70–90%) were achieved, keeping above 60% of its original activity. Immobilized glycosidases were effective in the selective deglycosylation of model glycoproteins and a Fasciola hepatica lysate, evidenced by a decrease in specific lectin recognition of 40–50% after enzymatic deglycosylation. Immobilized glycosidases were reused for several deglycosylation cycles without loss of effectiveness. Their use was extended to the elucidation of the glycan role of native glycoconjugates. A decrease in the recognition of lactoferrin treated with α-mannosidase by a C-type lectin receptor, DC-SIGN was found. Also the specific deglycosylation of a F. hepatica lysate demonstrated the relevance of mannosylated glycans in the induction of Th2/Treg immune responses during the infection. Our results show successful immobilization of specific glycosidases in nano-supports and validate their utility to identify glycans biological functions.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Bardet-Biedl syndrome (BBS) is a ciliopathy characterized by retinal degeneration, obesity, polydactyly, renal disease and mental retardation. CCDC28B is a BBS-associated protein that we have previously shown plays a role in cilia length regulation whereby its depletion results in shortened cilia both in cells and Danio rerio (zebrafish). At least part of that role is achieved by its interaction with the mTORC2 component SIN1, but the mechanistic details of this interaction and/or additional functions that CCDC28B might play in the context of cilia remain poorly understood. Here we uncover a novel interaction between CCDC28B and the kinesin 1 molecular motor that is relevant to cilia. CCDC28B interacts with kinesin light chain 1 (KLC1) and the heavy chain KIF5B. Notably, depletion of these kinesin 1 components results in abnormally elongated cilia. Furthermore, through genetic interaction studies we demonstrate that kinesin 1 regulates ciliogenesis through CCDC28B. We show that kinesin 1 regulates the subcellular distribution of CCDC28B, unexpectedly, inhibiting its nuclear accumulation, and a ccdc28b mutant missing a nuclear localization motif fails to rescue the phenotype in zebrafish morphant embryos. Therefore, we uncover a previously unknown role of kinesin 1 in cilia length regulation that relies on the BBS related protein CCDC28B.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Inflammation plays a major role in the onset and development of chronic non-communicable diseases like obesity, cardiovascular diseases and cancer. Combined, these diseases represent the most common causes of death worldwide, thus development of novel pharmacological approaches is crucial. Electrophilic nitroalkenes derived from fatty acids are formed endogenously and exert anti-inflammatory actions by the modification of proteins involved in inflammation signaling cascades. We have developed novel nitroalkenes derived from α-tocopherol aiming to increase its salutary actions by adding anti-inflammatory properties to a well-known nutraceutical. We synthesized and characterized an α-tocopherol-nitroalkene (NATOH) and two hydrosoluble analogues derived from Trolox (NATxME and NATx0). We analyzed the kinetics of the Michael addition reaction of these compounds with thiols in micellar systems aiming to understand the effect of hydrophobic partition on the reactivity of nitroalkenes. We studied NATxME in vitro showing it exerts non-conventional anti-inflammatory responses by inducing Nrf2-Keap1-dependent gene expression and inhibiting the secretion of NF-κB dependent pro-inflammatory cytokines. <dcterms:abstract><![CDATA[ETHNOPHARMACOLOGICAL RELEVANCE: Ophidian accidents are a serious public health problem in Argentina; the Bothrops species is responsible for 97% of these accidents, and in particular, B. diporus is responsible for 80% of them. In the northeast of the country (Corrientes Provinces), Cissampelos pareira L. (Menispermaceae) is commonly used against the venom of B. diporus; its use is described in almost all ethnobotanical literature from countries where the plant grows. AIM OF THE STUDY: In this study, the in vitro and in vivo antivenom activities of C. pareira extracts were evaluated against B. diporus venom, with a particular focus on the local effects associated with envenoming. The seasonal influence on the chemical composition of the active extracts was also studied, in order determine the associated range of variability and its influence on the antivenom activity. MATERIALS AND METHODS: This research was conducted using aerial parts (leaves, flowers, tender stems) and roots of Cissampelos pareira collected from two different phytogeographic regions of Corrientes (Argentina); Paso de la Patria and Lomas de Vallejos. In addition, to perform a seasonal analysis and to evaluate the metabolic stability, material was collected at three different growth stages. In vivo and in vitro anti-snake venom activities were tested, and a bio-guided chromatographic separation was performed in order to determine the active chemicals involved. The fractions obtained were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and the chemical profile of the most active constituent was analyzed by ultra high-performance liquid chromatography coupled to quadrupole/high-resolution mass spectrometry (Q-Orbitrap). (UHPLC-MS). RESULTS: The alcoholic extract was found to be the most active The bio-guided fractionation allowed selection one fraction to be analyzed by UHPLC-MS in order to identify the components responsible for the activities found; this identified five possible flavonoids. CONCLUSIONS: Our studies of the activity of C. pareira against the venom of B. diporus have confirmed that this species possesses inhibitory effects in both in vitro and in vivo models. Moreover, the present data demonstrate that certain flavonoids may mitigate some of the venom-induced local tissue damage.]]></dcterms:abstract>

Complexes and ligands were evaluated against the bloodstream form of T. brucei, L. infantum amastigotes, M. tuberculosis (MTB) sensitive strain and MTB clinical isolates. Complexes showed a significant increase of the anti-T. brucei activity with respect to the free ligands (>28- and >46-fold for Trop and 6- and 22-fold for Hino coordinated to Pt-dppf and Pd-dppf, respectively), yielding IC50 values < 5 μM. The complexes proved to be more potent than the antitrypanosomal drug Nifurtimox. The new ferrocenyl derivatives were more selective towards the parasite than the free ligands. The Pt compounds were less toxic on J774 murine macrophages (mammalian cell model), than the Pd ones, showing selectivity index values (SI = IC50 murine macrophage/IC50T. brucei) up to 23. Generation of the {M-dppf} compounds lead to a slightly positive impact on the anti-leishmanial potency. Although the ferrocenyl derivatives were more active on sensitive MTB than the free ligands (MIC90 = 9.88-14.73 μM), they showed low selectivity towards the pathogen. Related to the mechanism of action, the antiparasitic effect cannot be ascribed to an interference of the compounds with the thiol-redox homeostasis of the pathogen. Fluorescence measurements pointed at DNA as a probable target of the new compounds. [Pt(Trop)(dppf)](PF6) and [Pt(Hino)(dppf)](PF6) could be considered prospective anti-T. brucei agents that deserve further research.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Snakins are antimicrobial peptides (AMPs) found, so far, exclusively in plants, and known to be important in the defense against a wide range of pathogens. Like other plant AMPs, they contain several positively charged amino acids, and an even number of cysteine residues forming disulfide bridges which are considered important for their usual function. Despite its importance, studies on snakin tertiary structure and mode of action are still scarce. In this study, a new snakin-like gene was isolated from the native plant Peltophorum dubium, and its expression was verified in seedlings and adult leaves. The deduced peptide (PdSN1) shows 84% sequence identity with potato snakin-1 mature peptide, with the 12 cysteines characteristic from this peptide family at the GASA domain. The mature PdSN1 coding sequence was successfully expressed in Escherichia coli. The purified recombinant peptide inhibits the growth of important plant and human pathogens, like the economically relevant potato pathogen Streptomyces scabies and the opportunistic fungi Candida albicans and Aspergillus niger. Finally, homology and ab initio modeling techniques coupled to extensive molecular dynamics simulations were used to gain insight on the 3D structure of PdSN1, which exhibited a helix–turn–helix motif conserved in both native and recombinant peptides. We found this motif to be strongly coded in the sequence of PdSN1, as it is stable under different patterns of disulfide bonds connectivity, and even when the 12 cysteines are considered in their reduced form, explaining the previous experimental evidences.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[The objectives of the present study were to compare the diet quality, nutritional status and satisfaction with life, food related life and family life between mothers and adolescent children from single and dual-headed households. In Temuco, Chile, families composed of single mothers and married or cohabiting mothers with one child aged between 10 and 17 years answered a questionnaire that included the Adapted Healthy Eating Index (AHEI), Satisfaction with Life Scale (SWLS), Satisfaction with Food-related Life (SWFoL) scale and Satisfaction with Family Life (SWFaL) scale. Continuous variables were analyzed using a dyadic hierarchical multilevel mixed model for distinguishable dyads. To analyze nutritional status, the between-dyads independent variable procedure for categorical data was performed using a Mann-Withney U test. Mothers and adolescents from single-headed and dual-headed families did not significantly differ in their AHEI scores. The proportion of overweight and obese mothers and adolescent children was higher in dual-headed households. Type of household did not influence the SWLS, SWFoL, and SWFaL of mothers, whereas adolescents from dual-headed households had higher SWFaL. Strategies to improve diet quality should be developed and implemented regardless of the family structure, yet interventions to improve nutritional status should be targeted at dual-headed households. Also, special emphasis should be given to improving the satisfaction with family life in adolescents from single-headed households.]]></dcterms:abstract>

This study reports on the production and characterization of Issatchenkia terricola, beta-glucosidase and its potential to release red-wine aromatic compounds. The enzyme, a monomer of 48 kDa with an isoelectric point of 3.5 is tolerant to glucose and ethanol, properties compatible with enological use. Although fed-batch is usually the most suitable system for enzyme production in submerged culture, in this case the yield was practically the same as in batch culture. Enzyme productivity was increased 2-fold in synthetic medium with glucose with respect to the YPG and 3–8-fold with respect to other media assayed. After enzymatic treatment, GC-MS analysis of the released aglycones demonstrated significant increases in the concentration of phenols (83%) and norisoprenoids (65%). According to the judges of the sensory panel, the treatment resulted in a wine with dried fruits and raisins notes, as compared to the control, which was found more sweet and fruity. This, together with the lack of activity on anthocyanin glycosides, highlights the potential of this enzyme in enology, since its high selectivity allowed the development of aroma without compromising wine color.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Adulteration is a common practice in the illicit drugs market, but the psychoactive and toxic effects provided by adulterants are clinically underestimated. Coca-paste (CP) is a smokable form of cocaine which has an extremely high abuse liability. CP seized samples are sold adulterated; however, qualitative and quantitative data of CP adulteration in forensic literature is still scarce. Besides, it is unknown if adulterants remain stable when CP is heated. This study was designed to report the chemical content of an extensive series of CP seized samples and to demonstrate the stability (i.e., chemical integrity) of the adulterants heated. To achieve this goal, the following strategies were applied: (1) a CP adulterated sample was heated and its fume was chemically analyzed; (2) the vapor of isolated adulterants were analyzed after heating; (3) plasma levels of animals exposed to CP and adulterants were measured. Ninety percent of CP seized samples were adulterated. Adulteration was dominated by phenacetin and caffeine and much less by other compounds (i.e., aminopyrine, levamisole, benzocaine). In the majority of CP analyzed samples, both cocaine and caffeine content was 30%, phenacetin 20% and the combination of these three components reached 90%. Typical cocaine pyrolysis compounds (i.e., BA, CMCHTs, and AEME) were observed in the volatilized cocaine and CP sample but no pyrolysis compounds were found after isolated adulterants heating. Cocaine, phenacetin, and caffeine were detected in plasma. We provide current forensic data about CP seized samples and demonstrated the chemical integrity of their adulterants heated.]]></dcterms:abstract>

MATERIALS AND METHODS: Dendrimers were first conjugated with Suc-HYNIC (succinimidyl 6-hydrazinopyridine-3-carboxylic acid hydrochloride). Dendrimer-HYNIC was then incubated with 188ReO4-. Biodistribution was performed administrating 188Re-dendrimer to normal (NM) or melanoma-bearing mice (MBM). Chromosome aberration test was conducted in order to measure treatment capacity of 188Re-dendrimer in melanoma cells. RESULTS: Radiolabeling yield of dendrimer was approx. 70%. Biodistribution studies in NM showed blood clearance with hepatic and renal depuration. MBM showed a similar pattern of biodistribution with tumor uptake of 6% of injected dose. Aberrant metaphases quantified in control cells were 7%, increasing to 29.5% in cells treated with 15μCi (0.555 MBq) of 188Re-dendrimer for 24 h. CONCLUSIONS: 188Re-dendrimer can produce double-stranded breaks in DNA induced by ionizing radiation in melanoma cells in vitro.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Soybean glycinin [11S] was modified by an acidic pH treatment to improve its emulsifying properties. Glycinin was obtained by isoelectric precipitation (11Sn) and then treated with acid (11St). The oil–water interfacial tension and rheology were measured. The rate constants of adsorption (ka) and rearrangement (kr) of proteins at the interface and the dilational (E), elastic (Ed), and viscous modulus (Ev) of the protein interfacial film were determined. Particle size distribution (PSD), interfacial protein concentration (Γ), and the creaming destabilization rate constants ks (for the smaller droplets) and kl (for the larger droplets) were analyzed in oil‐in‐water emulsions (25% v/v of oil and 75% v/v of 1 mg/mL protein solution in 10 mM sodium phosphate buffers, pH 7.0 and 2.5, respectively). Compared to the native protein, the acid treatment caused irreversible denaturation of 11S and significantly increased Г, ka, E, Ed, and Ev (P ≤ 0.05), resulting in a greater rate of protein adsorption to the interface and a stronger interfacial film. PSD showed a bimodal distribution with peaks above and below 4 μm. Smaller droplets moved toward smaller diameters for 11St. It also showed lower values of creaming destabilization constants ks (d ≤ 4 μm) and kl (d ≥ 4 μm) than 11Sn. In conclusion, acid treatment of 11S enhances the creaming stability of its emulsions by improving the interfacial properties and reducing the droplet size.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Significance: Major pathogenic enterobacteria and protozoan parasites from the phylum Euglenozoa, such as trypanosomatids, are endowed with glutathione (GSH)-spermidine (Sp) derivatives that play important roles in signaling and metal and thiol-redox homeostasis. For some Euglenozoa lineages, the GSH-Sp conjugates represent the main redox cosubstrates around which entire new redox systems have evolved. Several proteins underwent molecular adaptations to synthesize and utilize the new polyamine-based thiols. Recent Advances: The genomes of closely related organisms have recently been sequenced, which allows mining and analysis of gene sequences that belong to these peculiar redox systems. Similarly, the three-dimensional structures of several of these proteins have been solved, which allows for comparison with their counterparts in classical redox systems that rely on GSH/glutaredoxin and thioredoxin. Critical Issues: The evolutionary and structural aspects related to the emergence and use of GSH-Sp conjugates in Euglenozoa are reviewed focusing on unique structural specializations that proteins developed to use N1,N8-bisglutathionylspermidine (trypanothione) as redox cosubstrate. An updated overview on the biochemical and biological significance of the major enzymatic activities is also provided. Future Directions: A thiol-redox system strictly dependent on trypanothione is a feature unique to trypanosomatids. The physicochemical properties of the polyamine-GSH conjugates were a major driving force for structural adaptation of proteins that use these thiols as ligand and redox cofactor. In fact, the structural differences of indispensable components of this system can be exploited toward selective drug development. Future research should clarify whether additional cellular processes are regulated by the trypanothione system. Antioxid. Redox Signal. 28, 463–486.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Chagas disease is an endemic illness in Latin America caused by the parasite Trypanosoma cruzi. Current chemotherapies are old and inadequate, and the emergence of drug-resistant strains underscores the need of new drugs. Platinum-based complexes have been shown to be a promising approach against parasitic diseases. In this work, the effect of 1,1'-bis(diphenylphosphino)ferrocene pyridine-2-thiolate-1-oxide Pt(II) hexafluorophosphate, Pt-dppf-mpo, was studied on T. cruzi. A promising antitrypanosomal activity was determined for the CL Brener strain with a low cytotoxicity determined using in vitro-cultured mammal cells. The compound uptake in parasites treated with concentrations of 1× and 10× the IC50 value reached ~75% and 19%, respectively. Pt-dppf-mpo induced necrosis after 24 hr of parasite incubation. This event was preceded by depolarization of mitochondrial membrane potential. Cell vitality assays showed high esterase activity in treated parasites. However, despite this increase in metabolic activity, treated epimastigotes showed rounded morphology and loss of flagellum with a reduction in mobility as compound concentration and/or time of incubation was increased. At last, we demonstrate that Pt-dppf-mpo incubation also affects the trypomastigote infection process as well as the infection persistence evaluated as the number of amastigotes per cell in a dose-dependent manner.]]></dcterms:abstract>
