    <dcterms:abstract><![CDATA[Background Cadmium (Cd) exposure has adverse health effects in children. Diet contributes to Cd exposure, but dietary components could affect body Cd levels. Objective To examine associations between diet and urinary Cd (U-Cd) in children. Methods In this cross-sectional study conducted in Montevideo, Uruguay, Cd exposure of 5&ndash;8 year old children (n = 279 with complete data) was assessed in first morning urine (U-Cd), a marker of long-term exposure, measured by ICP-MS and adjusted for specific gravity. Distribution of U-Cd was (median [5%, 95%]: 0.06 [0.02, 0.17] &mu;g/L); data were natural-log-transformed (ln) for statistical analyses. Serum ferritin (SF), an indicator of iron stores, was measured in fasting samples. Trained nutritionists completed two non-consecutive 24-h dietary recalls with both child and caregiver present. Measures of iron, zinc, calcium and fiber intake, and the consumption of grains, root vegetables, milk, and foods rich in heme iron (white and read meats) and non-heme iron (legumes, spinach, broccoli, tomatoes, dried fruit) were derived. Multivariable ordinary least squares (OLS) and ordinal regressions were used to examine associations among tertiles of water Cd, SF, diet, and U-Cd. OLS models were further stratified by sex. Results In covariate-adjusted models, SF was not related to ln-U-Cd. Children in highest tertile of iron and zinc intake had lower ln-U-Cd: (&minus;0.23 [&minus;0.42, &minus;0.03]) and (&minus;0.25 [&minus;0.44, &minus;0.05]), respectively, compared to the reference group. Children consuming higher amounts of foods rich in heme iron had slightly lower ln-U-Cd (&minus;0.17 [&minus;0.36, 0.03]). High grain consumption was related to higher ln-U-Cd (0.25 [0.06, 0.45]). Conclusions Diets rich in grains were related to higher urinary Cd levels among children living in the context of low Cd pollution. Higher intake of iron and zinc was related to lower Cd levels. Given that urinary Cd is mainly a marker of long-term exposure, these findings should be further corroborated.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The electrochemical adsorption characteristics of 3-buten-2-one and 2-butanone on porous platinum electrodes were investigated by differential electrochemical mass spectrometry (DEMS) in aqueous 0.5 M H2SO4. The study was conducted through the oxidation and reduction of residues formed at different adsorption potentials (Ead). The maximum adsorption for both organic substances was observed at the potential of zero charge of the interface, that is, ca 0.20 V. At this potential an adsorbate species with the C&ndash;O bond lying parallel to the surface is proposed. The sole oxidation product for both molecules was carbon dioxide. Anodic current transients were detected for 3-buten-2-one at adsorption potentials higher than 0.20 V. Between 0.40 and 0.60 V, it is possible to admit a 1&ndash;3e&minus; loss during adsorption with a progressive fragmentation of the original molecule at increasing Ead. After three cycles in the H-adatom region, about 75% of the 3-buten-2-one adsorbate is desorbed; the main products being butane, 1-butene and propane. The ratio of these products depends on Ead, as is expected for the fragmentation of the molecule upon increasing potentials. On the other hand, 2-butanone was adsorbed in the 0.10&ndash;0.60 V potential range without current transients associated with the dissociative reactions of the molecule. The surface coverage diminished drastically at both sides of the maximum adsorption (0.20 V). Butane was the main reduction product of 2-butanone.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[We describe a novel pericyclic/anionic cascade reaction on tropolonic substrates that combines a Diels-Alder reaction, an oxa-Michael addition, and an acyloin rearrangement to afford tricyclic &alpha;-hydroxy-&beta;-methoxyketones. Spectroscopic, crystallographic, and mechanistic studies indicate that the process requires stabilization of reaction intermediates through intramolecular H-bonding to take place, and suggest that the conjugate addition step involves a catalytic cycle with initial formation of an ammonium enolate and sustained by an alkoxide ion pair. Given the rich functionality and structural complexity generated in a single step, the process could be exploited in the preparation of natural product-like compound libraries.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[turgeons are chondrostean fish of high economic value and critically endangered due to anthropogenic activ ities, which has led to sturgeon aquaculture development. Russian sturgeon (Acipenser gueldenstaedtii), the second most important species reared for caviar, is successfully farmed in subtropical countries, including Uruguay. However, during the Uruguayan summer, sturgeons face intolerable warmer temperatures that weaken their defences and favour infections by opportunistic pathogens, increasing fish mortality and farm economic losses. Since innate immunity is paramount in fish, for which the liver plays a key role, we used deep RNA sequencing to analyse differentially expressed genes in the liver of Russian sturgeons exposed to chronic heat stress and challenged with Aeromonas hydrophila. We assembled 149.615 unigenes in the Russian sturgeon liver tran scriptome and found that metabolism and immune defence pathways are among the top five biological processes taking place in the liver. Chronic heat stress provoked profound effects on liver biological functions, up regulating genes related to protein folding, heat shock response and lipid and protein metabolism to meet en ergy demands for coping with heat stress. Besides, long-term exposure to heat stress led to cell damage triggering liver inflammation and diminishing liver ability to mount an innate response to A. hydrophila challenge. Accordingly, the reprogramming of liver metabolism over an extended period had detrimental effects on fish health, resulting in weight loss and mortality, with the latter increasing after A. hydrophila challenge. To our knowledge, this is the first transcriptomic study describing how chronic heat-stressed sturgeons respond to a bacterial challenge, suggesting that liver metabolism alterations have a negative impact on the innate anti bacterial response.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The reaction of [ReOCl3(PPh3)2] with 1,2-bis(2,3-dimercaptobenzamido) ethane (H4-1) in the presence of Na2CO3 in methanol under anaerobic conditions affords the dinuclear ReV oxido complex [PPh4]2[ReO(1)]2 containing two distorted square-pyramidal {ReVOS4} units bridged by the ligand strands in a double-stranded fashion. The coordination geometry around the metal centers is similar to the one observed for [ReO(bdt)2]&ndash;. The ReS4 planes are arranged in a coplanar fashion and are not twisted around the metal&ndash;metal vector, which prevents the complex to adopt a helical structure. Luminescence studies show the presence of emission bands, which are assigned to singlet-singlet transitions exhibiting very fast decays (ca. 10 ns).     <dcterms:abstract><![CDATA[The recently recommended NIST-JANAF Thermochemical Table values for the thermochemical properties of oxygen fluorides are examined critically on the basis of high-level density functional and ab initio G2 calculations. Special attention is given to the enthalpies of formation and it is concluded that, although the proposed values for OF(g), FOO(g) and FOF(g) are reasonable and in agreement with our previously recommended values, the enthalpy of formation recommended for FOOF is too low. A value about 50% larger is proposed and it is recommended that the original 1959 experimental determination of this enthalpy of formation be repeated.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[Immunochromatographic strips (ICS) attract much attention as practical analytical tools, particularly for point of care testing. However, ICS suffer from insufficient sensitivity, especially for small molecules. Competitive assays developed with analyte peptidomimetics obtained from phage display peptide libraries, usually enable to attain lower detection limits than the widely used chemical haptens, but are rarely used in ICS assay because of the shortcomings of the phage particle. In this work, we used a phage-free fluorescent peptide tracer (C2-15-EmGFP) for ICS, which consists of the emerald green fluorescent protein (EmGFP) fused to a peptidomimetic of imidaclothiz (C2-15). C2-15-EmGFP works as fluorescent donor and colloidal gold nanoparticles (AuNPs) coated with an anti-imidaclohtiz antibody as quencher, which allowed to develop a fluorescence quenching-based ICS (FQICS) for the detection of imidaclothiz in agricultural and environmental samples. The colorimetric and fluorescent signals were determined by both naked eye and imageJ software. Quantitative image analysis showed that the sensitivities obtained using the colorimetric (50% inhibitory concentration (IC50): 9.62&thinsp;ng&thinsp;mL&minus;1) and fluorescent (50% saturation concentration (SC50): 10.1&thinsp;ng&thinsp;mL&minus;1) signals were similar. The visual detection by the naked eye showed a sensitivity of 8.00&thinsp;ng&thinsp;mL&minus;1 with fluorescent readout which was 8-fold lower than that of the colorimetric readout (64.0&thinsp;ng&thinsp;mL&minus;1). The FQICS performed with excellent recoveries and good correlation with high-performance liquid chromatography in different matrices.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[Nitro-fatty acids (NO2-FA) are electrophilic lipid mediators derived from unsaturated fatty acid nitration. These species are produced endogenously by metabolic and inflammatory reactions and mediate anti-oxidative and anti-inflammatory responses. NO2-FA have been postulated as partial agonists of the Peroxisome Proliferator-Activated Receptor gamma (PPAR&gamma;), which is predominantly expressed in adipocytes and myeloid cells. Herein, we explored molecular and cellular events associated with PPAR&gamma; activation by NO2-FA in monocytes and macrophages. NO2-FA induced the expression of two PPAR&gamma; reporter genes, Fatty Acid Binding Protein 4 (FABP4) and the scavenger receptor CD36, at early stages of monocyte differentiation into macrophages. These responses were inhibited by the specific PPAR&gamma; inhibitor GW9662. Attenuated NO2-FA effects on PPAR&gamma; signaling were observed once cells were differentiated into macrophages, with a significant but lower FABP4 upregulation, and no induction of CD36. Using in vitro and in silico approaches, we demonstrated that NO2-FA bind to FABP4. Furthermore, the inhibition of monocyte FA binding by FABP4 diminished NO2-FA-induced upregulation of reporter genes that are transcriptionally regulated by PPAR&gamma;, Keap1/Nrf2 and HSF1, indicating that FABP4 inhibition mitigates NO2-FA signaling actions. Overall, our results affirm that NO2-FA activate PPAR&gamma; in monocytes and upregulate FABP4 expression, thus promoting a positive amplification loop for the downstream signaling actions of this mediator.]]></dcterms:abstract>

    <dcterms:title><![CDATA[<strong>A family of diverse Kunitz inhibitors from Echinococcus granulosus potentially involved in host-parasite cross-talk</strong>]]></dcterms:title>

    <dcterms:abstract><![CDATA[The cestode Echinococcus granulosus, the agent of hydatidosis/echinococcosis, is remarkably well adapted to its definitive host. However, the molecular mechanisms underlying the successful establishment of larval worms (protoscoleces) in the dog duodenum are unknown. With the aim of identifying molecules participating in the E. granulosus-dog cross-talk, we surveyed the transcriptomes of protoscoleces and protoscoleces treated with pepsin at pH 2. This analysis identified a multigene family of secreted monodomain Kunitz proteins associated mostly with pepsin/H+-treated worms, suggesting that they play a role at the onset of infection. We present the relevant molecular features of eight members of the E. granulosus Kunitz family (EgKU-1 &ndash; EgKU-8). Although diverse, the family includes three pairs of close paralogs (EgKU-1/ EgKU-4; EgKU-3/EgKU-8; EgKU-6/EgKU-7), which would be the products of recent gene duplications. In addition, we describe the purification of EgKU-1 and EgKU-8 from larval worms, and provide data indicating that some members of the family (notably, EgKU-3 and EgKU-8) are secreted by protoscoleces. Detailed kinetic studies with native EgKU-1 and EgKU-8 highlighted their functional diversity. Like most monodomain Kunitz proteins, EgKU-8 behaved as a slow, tight-binding inhibitor of serine proteases, with global inhibition constants (KI *) versus trypsins in the picomolar range. In sharp contrast, EgKU-1 did not inhibit any of the assayed peptidases. Interestingly, molecular modeling revealed structural elements associated with activity in Kunitz cation-channel blockers.     <dcterms:abstract><![CDATA[Fijiviruses replicate and package their genomes within viroplasms in a pro cess involving RNA-RNA and RNA-protein interactions. Here, we demonstrate that the 24 C-terminal residues (C-arm) of the P9-1 major viroplasm protein of the mal de R&iacute;o Cuarto virus (MRCV) are required for its multimerization and the formation of viroplasm like structures. Using an integrative structural approach, the C-arm was found to be dis pensable for P9-1 dimer assembly but essential for the formation of pentamers and hexamers of dimers (decamers and dodecamers), which favored RNA binding. Although both P9-1 and P9-1DC-arm catalyzed ATP with similar activities, an RNA-stimulated ATPase activity was only detected in the full-length protein, indicating a C-arm-mediated interaction between the ATP catalytic site and the allosteric RNA binding sites in the (do)decameric assemblies. A stronger preference to bind phosphate moieties in the dec amer was predicted, suggesting that the allosteric modulation of ATPase activity by RNA is favored in this structural conformation.     <dcterms:abstract><![CDATA[The interaction between alkyl radicals and graphene was studied by means of dispersion-corrected density functional theory. The results indicate that isolated alkyl radicals are not likely to be attached onto perfect graphene. It was found that the covalent binding energies are low, and because of the large entropic contribution, DGA 298 is positive for methyl, ethyl, isopropyl, and tert-butyl radicals. Although the alkylation may proceed by moderate heating, the desorption barriers are low. For the removal of the methyl and tert-butyl radicals covalently bonded to graphene, 15.3 and 2.4 kcalmol1 are needed, respectively. When alkyl radicals are agglomerated, the binding energies are increased. For the addition in the ortho position and on opposite sides of the sheet, the graphene&ndash; CH3 binding energy is increased by 20 kcalmol1, whereas for the para addition on the same side of the sheet, the increment is 9.4 kcalmol1. In both cases, the agglomeration turns the DGA 298&lt;0. For the ethyl radical, the ortho addition on opposite sides of the sheet has a negative DGA 298, whereas for isopropyl and tert-butyl radicals the reactions are endergonic. The attachment of the four alkyl radicals under consideration onto the zigzag edges is exergonic. The noncovalent adsorption energies computed for ethyl, isopropyl, and tert-butyl radicals are significantly larger than the graphene&ndash;alkyl-radical covalent binding energies. Thus, physisorption is favored over chemisorption. As for the DGA 298 for the adsorption of isolated alkyl radicals, only the tertbutyl radical is likely to be exergonic. For the phenalenyl radical we were not able to locate a local minimum for the chemisorbed structure since it moves to the physisorbed structure.     <dcterms:abstract><![CDATA[Peptides isolated from phage display libraries are powerful reagents for small-molecule immunoassay; however, their application as phage-borne peptides is significantly limited by the biological nature of the phage. Here, we present the use of lysine scaffold to prepare a series of different valence peptides to serve as replacements for phage-borne peptides. Benzothiostrobin was selected as a model analyte, the cyclic benzothiostrobin-peptidomimetic in the form of monomer, dendrimer-like dimer, and tetramer were designed and synthesized. Compared with the monomer, the affinity of dendrimer-like dimer and tetramer increased 1.87 and 13.6 times, respectively, as determined by isothermal titration calorimetry (ITC). A novel inner filter effect immunoassay (IFE-IA) with positive readout was developed for benzothiostrobin detection utilizing the peptidomimetics attached to upconversion nanoparticles (UCNPs) as energy donor and monoclonal antibody (mAb)-labeled urchin-like gold nanoflowers (AuNFs) as energy absorber, respectively. The sensitivity of the assay based on dendrimer-like tetramer was approximately 6 and 3 times higher than monomer and dendrimer-like dimer, respectively. After optimization, 50% saturation of the signal (SC50) and detection range (SC10 to SC90) of the IFE-IA based on dendrimer-like tetramer were 11.81 ng mL&ndash;1 and 2.04&ndash;106.17 ng mL&ndash;1, respectively. The IFE-IA also shows good accuracy for the detection of benzothiostrobin in authentic samples.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[A simple, fast, and environmentally friendly method for sample preparation and subsequent determination of Cu, Fe, Mn, and Zn in wheat flour using total reflection X-ray fluorescence (TXRF) was developed and validated. Furthermore, TXRF and two other analytical techniques currently used for elemental analysis in foodstuffs, namely flame atomic absorption spectrometry (FAAS) and electrothermal atomic absorption spectrometry (ETAAS), were compared to confirm the suitability of TXRF for routine and screening analyses of metallic elements in wheat flour. The TXRF sample preparation procedure takes around 6 min and does not require a mineralization step. Although the detection limits of the TXRF technique were higher than those of FAAS and ETAAS, they were suitable for the proposed purpose. The advantages of the proposed method are also discussed. The TXRF method was applied to the analysis of five commercial wheat flour samples representing all brands available in Montevideo, Uruguay. TXRF proved to be suitable for the determination of Cu, Fe, Mn, and Zn in wheat flour, and is a promising technique for routine and screening analyses of bakery foodstuffs.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[A one-pot efficient, practical and eco-friendly synthesis of tocopherol analogues has been developed using water or solvent free conditions via Passerini and Ugi multicomponent reactions. These reactions can be optimized using microwave irradiation or ultrasound as the energy source. Accordingly, a small library of 30 compounds was prepared for biological tests. The evaluation of the antiproliferative activity in the human solid tumor cell lines A549 (lung), HBL-100 (breast), HeLa (cervix), SW1573 (lung), T-47D (breast), and WiDr (colon) provided lead compounds with GI50 values between 1 and 5 &mu;M. A structure&ndash;activity relationship is also discussed. One of the studied compounds comes up as a future candidate for the development of potent tocopherol-mimetic therapeutic agents for cancer.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[Single phase BaCe0.8Pr0.2O3&minus;&delta; perovkite was obtained by the modified Pechini method followed by self-combustion and sintering at 1350◦C. Crystal structure, thermodynamic stability, linear expansion and electrical properties were characterized as a function of temperature, under oxidizing and reducing atmospheres. BaCe0.8Pr0.2O3&minus;&delta; presents phase transitions under both atmospheres, which do not compromise the transport properties. The electrical conductivity showed a mixed character, dominated by electronic and oxygen vacancies transport in O2-containing atmosphere. Pr+4 reduction was observed above 400◦C under H2-containing atmosphere. This reduction increases the oxygen non-stoichiometry and OH&bull; charge carriers concentration, unit cell volume and thermal expansion coefficients (TECs). Pr+4 reduction suppresses the electronic conduction and hence the transport number for OH&bull; would be dominant in wet reducing atmosphere]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The &beta;-lactoglobulin protein from cow&rsquo;s milk is a common allergen present in human milk that leads to serious allergic reactions in susceptible infants. This creates a serious safety issue in Human Milk Banks, and the best practice to assure the safety of susceptible newborns would be to classify the donated human milk according to its content of &beta;-lactoglobulin. To provide a simple and fast assay to detect this molecule in human milk we developed a nanobody-based sandwich ELISA. The capture and detection nanobodies were isolated by phage display from a llama immunized with the antigen using a sequential selection strategy for antibody pair iden tification. This allowed to attain a highly sensitive test with a limit of quantitation of 40 pg/mL and extended lineal range of up to 3000 pg/mL, showing an excellent performance with human milk. For these characteristics, the ELISA developed might be useful for the classification of donated human milk according to their &beta;-lacto globulin content, improving the quality of milk destined to susceptible newborns.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[Antigen tests have been crucial for managing the COVID-19 pandemic by identifying individuals infected with SARS-CoV-2. This remains true even after immunity has been widely attained through natural infection and vaccination, since it only provides moderate protection against transmission and is highly permeable to the emergence of new virus variants. For this reason, the widespread availability of diagnostic methods is essential for health systems to manage outbreaks effectively. In this work, we generated nanobodies to the virus nucleocapsid protein (NP) and after an affinity-guided selection identified a nanobody pair that allowed the detection of NP at sub-ng/mL levels in a colorimetric two-site ELISA, demonstrating high diagnostic value with clinical samples. We further modified the assay by using a nanobody-NanoLuc luciferase chimeric tracer, resulting in increased sensitivity (detection limit = 61 pg/mL) and remarkable improvement in diagnostic performance. The luminescent assay was finally evaluated using 115 nasopharyngeal swab samples. Receiver Operating Characteristic (ROC) curve analysis revealed a sensitivity of 78.7% (95% confidence interval: 64.3%-89.3%) and specificity of 100.0% (95% confidence interval: 94.7%-100.0%). The test allows the parallel analysis of a large number of untreated samples, and fulfills our goal of producing a recombinant reagent-based test that can be reproduced at low cost by other laboratories with recombinant expression capabilities, aiding to build diagnostic capacity.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The main objective of this work was to develop a kinetic model to describe the variation of the surface tension in an air-water interface due to the adsorption of proteins from different origins and to identify quantitatively the relevant parameters. It was considered that the processes of adsorption, unfolding and reordering of the protein molecule in the interface occur simultaneously. The model used in the present work to calculate the surface tension postulates the existence of two simultaneous processes, adsorption and protein rearrangement represented with an equation of first order with two exponential components. The relevant parameter of the equation are ka and kr&mdash;the rate constants of the two first order kinetic phases that correspond to both conformational states of the protein, adsorption and rearrangement during the process of variation of the surface tension, and the amplitude parameters Aa and Ar. The results suggest that the kinetic model for the variation of the surface tension of protein solutions proposed in this work, with two simultaneous first order processes, is more appropriate than previous models to describe such variation.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The objectives of the present study were to identify adolescent classes based on perceptions of their family's eating habits and to determine whether these classes differ in terms of family meal frequency and source, parents and adolescent diet quality, nutritional status, satisfaction with life, family life and food-related life. Questionnaires were administered to a sample of 300 two-parent families with one child between 10 and 17 years in Chile. Questionnaires included the Satisfaction with Life Scale (SWLS), the Satisfaction with Food-related Life scale (SWFoL), the Satisfaction with Family Life scale (SWFaL) and the Adapted Healthy Eating Index (AHEI). Adolescents also answered the Family Eating Habits Questionnaire (FEHQ). Latent class (LC) analysis was used to estimate the number of classes associated with the adolescent's perceptions about their family's eating habits, based on the three factors detected in the FEHQ. The LC analysis yielded three classes: &ldquo;medium cohesion around family meals/healthy food-related parenting practices&rdquo; (Class 1, 50.4%), &ldquo;high cohesion around family meals/healthy food-related parenting practices&rdquo; (Class 2, 25.9%) and &ldquo;high cohesion around family meals/unhealthy food-related parenting practices&rdquo; (Class 3, 23.7%). Classes also differed in the adolescents AHEI score, frequency and source of family meals; body mass index, SWLS and SWFaL scores in mothers, fathers and adolescents, adolescent sex and household financial situation. These findings suggest that frequent and cohesive family meals improve the well-being in both parents and adolescents, but are not enough to achieve healthier diets and weight statuses in adolescents.]]></dcterms:abstract>
