    <dcterms:abstract><![CDATA[<p>En este art&iacute;culo se presentar&aacute; un resumen de algunas de las l&iacute;neas de investigaci&oacute;n que se vienen desarrollando en el Centro NanoMat, y que son consideradas como estrat&eacute;gicas tanto desde el punto de vista de su relevancia y potencialidades como porque pueden ser llevadas adelante exitosamente por el Centro con sus actuales capacidades. Sin embargo, cabe se&ntilde;alar que dichas capacidades est&aacute;n en permanente crecimiento desde su instalaci&oacute;n, en el 2008, lo que permite el abordaje de nuevos emprendimientos.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>La nanotecnolog&iacute;a (NT) es una rama de la tecnolog&iacute;a en la que se realizan procesos con la materia a una escala nanom&eacute;trica. Un nanometro o nan&oacute;metro es la millon&eacute;sima parte de un mil&iacute;metro. Dicho de esta manera, es muy dif&iacute;cil aquilatar la dimensi&oacute;n de referencia, la que da origen al mismo nombre de esta tecnolog&iacute;a.<br />Se podr&iacute;a agregar que casi se ingresa en la zona del tama&ntilde;o de los propios &aacute;tomos, pero si bien esta aclaraci&oacute;n suena a algo muy peque&ntilde;o, probablemente no sea suficiente para darse cuenta de la real magnitud de lo que hablamos. Sin embargo, todos tenemos una noci&oacute;n de la peque&ntilde;ez de las c&eacute;lulas, que se definen como la menor unidad de los organismos vivos y<br />que todos entendemos como algo muy peque&ntilde;o. A las c&eacute;lulas las entendemos como parte de nosotros, pero tambi&eacute;n hablamos de g&eacute;rmenes y contagios o infecciones, t&eacute;cnicamente provocadas por bacterias, que son en s&iacute; mismas seres vivos unicelulares. Ahora bien, un nanometro es entre mil y diez mil<br />veces m&aacute;s peque&ntilde;o que una c&eacute;lula promedio, lo que nos dimensiona el tema m&aacute;s claramente.<br />Cuando hablamos de NT estamos incluyendo todas las tecnolog&iacute;as que tratan de procesos o productos en esta escala, ya sea productos finales o productos intermedios que se usen como precursores, o incluso sustancias que a esa escala favorezcan que tenga lugar una reacci&oacute;n que de otra manera<br />no ocurrir&iacute;a.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p><em>Streptococcus thermophilus</em> relies heavily on two type II-A CRISPR-Cas systems, CRISPR1 and CRISPR3, to resist siphophage infections. One hallmark of these systems is the integration of a new spacer at the 50 end of the CRISPR arrays following phage infection. However, we have previously shown that ectopic acquisition of spacers can occur within the CRISPR1 array. Here, we present evidence of the acquisition of new spacers within the array of CRISPR3 of <em>S. thermophilus</em>. The<br />analysis of randomly selected bacteriophage-insensitive mutants of the strain Uy01 obtained after phage infection, as well as the comparison with other S. thermophilus strains with similar CRISPR3 content, showed that a specific spacer within the array could be responsible for misguiding the adaptation complex. These results also indicate that while the vast majority of new spacers are added at the 50 end of the CRISPR array, ectopic spacer acquisition is a common feature of both<br />CRISPR1 and CRISPR3 systems in <em>S. thermophilus</em>, and it can still provide phage resistance. Ectopic spacer acquisition also appears to have occurred naturally in some strains of<em> Streptococcus pyogenes</em>, suggesting that it is a general phenomenon, at least in type II-A systems.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>We present the first worldwide study on the apple (Malus &times; domestica) fruit microbiome that examines questions regarding the composition and the assembly of microbial communities on and in apple fruit. Results revealed that the composition and<br />structure of the fungal and bacterial communities associated with apple fruit vary and are highly dependent on geographical location. The study also confirmed that the spatial variation in the fungal and bacterial composition of different fruit tissues exists at a global level. Fungal diversity varied significantly in fruit harvested in different geographical locations and suggests a potential link between location and the type and rate of postharvest diseases that develop in each country. The global core microbiome of apple fruit was represented by several beneficial microbial taxa and accounted for a large fraction of the fruit microbial community. The study provides foundational information about the apple fruit microbiome that can be utilized for the development of novel approaches for the management of fruit quality and safety, as well as for reducing<br />losses due to the establishment and proliferation of postharvest pathogens. It also lays the groundwork for studying the complex microbial interactions that occur on apple fruit surfaces.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>The present research contributed to the growing body of research regarding the use of emoji in sensory and consumer research by filling an acknowledged knowledge gap. Its specific aim was to explore the meaning of emoji in a food-related context. The empirical approach involved 357 consumers who were tasked with writing a social media post or text message relating to a situation involving eating/drinking which was to be centered around a specific emoji. A total of 21 facial emoji were<br />included in the research. As expected, emoji were used to convey different meanings, including degree of liking/disliking, emotional associations, gestures and expressions.<br />The emoji were also used as part of messages describing negative and positive disconfirmation of expectations, which points to the importance of measuring expectations in a food-related context. Two emoji were identified as unsuitable for future research due to their nonconsensual meaning ( and ).</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>Sugar intake among children has raised concern worldwide as it exceeds nutritional recommendations.<br />Sugar contributes to the daily energy intake, without providing additional nutritional value and is associated with several negative health outcomes. Sugars added to foods during industrial processing have been identified as the main source of sugar in children's diets. The present work critically discusses the role of the food industry in contributing to children's excessive sugar intake worldwide, and the strategies that have<br />been encouraged or implemented to reduce the sugar content in products targeted at children. The risk of the current sugar reduction practices in products targeted at children is discussed based on recent scientific evidence. Children's heightened preference for sweetness may not justify the availability of highly sugary products for children. Although research suggests that children readily accept less sweet products, there is still<br />some wariness in the food industry to reduce sweetness intensity. This has strengthened the use of non-nutritive sweeteners (NNS), focusing on maintaining the sweetness level.<br />However, emerging evidence suggests that this may not be the best approach.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p><em>Lithraea molleoides</em> (Anacardiaceae) is a tree with a broad area of distribution in the southern region of South America, including Southern Brazil, Southern, and Eastern Bolivia, Southern Paraguay, Northern, and Central Argentina, and<br />reaches its southernmost distribution limit in Northern and Northeastern Uruguay. It presents considerable morphological variability. It is used as medicinal plant and as a sweetener in traditional infusions. There have been reports on its antibacterial, antiviral, antioxidant, anti-inflammatory and antinociceptive activities. It is traditionally known that it may induce allergic contact dermatitis in sensitive persons by<br />direct and/or airborne contact, i.e. when used as firewood. Many of its biological activities have been associated with the reported presence of 5-alkyl/alkenyl resorcinols. Its allergenic activity has been ascribed to 3-alkyl/alkenyl catechols, as it<br />is well known for other species of the Anacardiaceae family. The wide distribution of the species, its morphological plasticity, the traditional medicinal uses, and toxic properties indicate the necessity of a more extensive and systematic study of the<br />species to assess its medicinal and biological potential.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>Ilex paraguariensis St. Hil., from the family of holy plants, Aquifoliaceae, is a native South American tree that can reach 18 m in height and is used to produce &ldquo;yerba mate&rdquo;. It is found primarily in the southern regions of South America, namely, Brazil (Mato Grosso do Sul, Minas Gerais, Parana, Rio Grande do Sul, Rio de Janeiro, Santa Catarina, Sao Paulo), Argentina (Corrientes, Misiones), Paraguay (Alto Paran&aacute;, Amambay, Caaguaz&uacute;, Canendiy&uacute;, Central, Guaira, Itapu&aacute;, Misiones,<br />San Pedro), and Uruguay. &ldquo;Yerba mate&rdquo; tea, an infusion made from the leaves of this tree, is a widely consumed nonalcoholic beverage in South America which is gaining rapid introduction into the world market, either as tea itself or as ingredient<br />in formulated foods or dietary supplements. The indigenous people have used it for centuries as a social and medicinal beverage. The container in which it is generally served is a gourd made from the dry and hollowed fruit of Lagenaria vulgaris Ser. (Cucurbitaceae), whose dimensions vary according the region and uses. It is called &ldquo;mate&rdquo; in the region. &ldquo;Mate&rdquo; has a very important social role. The act of offering and sharing it has connotations like those of the tea ceremony for some Oriental cultures.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>Eupatorium buniifolium Hook. ex Hook. &amp; Arn. (Asteraceae) is an aromatic and medicinal plant. Its area is spreading from the Southern parts of Bolivia, Brazil, Paraguay and Uruguay to the northern and central parts of Argentina. It is known as &ldquo;chilca negra&rdquo;, &ldquo;romerillo&rdquo;, &ldquo;chilca&rdquo;, &ldquo;chirca&rdquo;, &ldquo;romero colorado&rdquo;. The<br />infusion of leaves and stems is used as a digestive for stomachache, against diarrhea and for liver inflammation. Some studies have confirmed the digestive, hepatoprotective,<br />anti-inflammatory and antioxidant potential of this plant. The main chemical components responsible for its known biological activities have been identified as flavonoids. From E. buiniifolium leaves also an essential oil has been obtained<br />that is useful due to its potential insecticidal, antifungal and varroacide activities, both by direct contact and their vapors against the tested organisms.     <dcterms:abstract><![CDATA[<p><em>Cissampelos pareira</em>L. (Menispermaceae) is a widely used medicinal plant, also used to treat bites of venomous animals, particularly snakes. Ophidian accidents are a serious public health problem in Argentina where the Bothrops genus of snakes is responsible for 97% of these accidents, peculiarly B. diporus (&ldquo;yarar&aacute; chica&rdquo;) responsible for 80% of them. In the northeast of the country (Corrientes Province), C. pareira is commonly used against the venom of B. diporus; its use is described in almost all-ethnobotanical literature of the countries where the plant grows. In vitro and in vivo antivenom activities of C. pareira extracts from plants collected at two different phytogeographic regions of Corrientes (Argentina) were evaluated against B. diporus venom. The seasonal influence on the chemical composition of extracts was also studied to determine the associated range of variability and its influence on the antivenom activity. Besides, an evaluation of the chemical composition of the volatile extract was performed to assess its chemical stability. These results support the ethnopharmacological use of this species. Moreover, the presented data demonstrate that certain flavonoids may mitigate some of the venom-induced local tissue damages.</p>]]></dcterms:abstract>

It belongs to the small Martyniaceae family and it is used, along with less common species of Ibicella such as I. parodii and I. nelsoniana, in remedies of popular medicine. The phytochemical analysis of the plants showed the presence of several fatty<br />acid glycosides with antibacterial activity against Staphylococcus aureus, as well as several strains of uropathogenic Proteus mirabilis.     <dcterms:abstract><![CDATA[<p>Medicinal plants encompass a rich source of active compounds that can neutralize snake venoms or toxins. Nectandra angustifolia (Schrad.) Nees &amp; Mart. (Lauraceae), commonly known as &ldquo;laurel amarillo&rdquo;, &ldquo;aju&rsquo;yh&ucirc;&rdquo;, &ldquo;laurel-hu&rdquo;, &ldquo;laurel<br />del r&iacute;o&rdquo;, is used in traditional medicine as a digestive, for the treatment of rheumatism, arthritis, pain, and as antivenom. Snakebite envenomation was included in category A of neglected tropical diseases (NTD), in 2017, as they affect millions of people in the world (Chippaux, J Venom Anim Toxins 23:38&ndash;39: 2017). In Argentina, the snake genus Bothrops is responsible for the greatest number of accidents. Presently, the application of serum antivenom is the only specific treatment. This treatment does not protect over local effects, which are the main cause of morbidity. In the search for effective antivenoms, extracts of different polarity obtained from Nectandra angustifolia were evaluated both, in vitro and in vivo, against the venom of B. diporus "yarar&aacute; chica". The effect of seasonal variability in this activity was also studied, to optimize both, the harvest period and the most appropriate type of extract. By successive bio-guided fractionations of the ethanol extract of N. angustifolia a fraction enriched in flavonoids responsible for the alexiteric activity of the species has been isolated. The phytochemical study of the volatile fraction of N. angustifolia and N. megapotamica allowed for the phytochemical characterization of farther interesting compounds.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>Passiflora caerulea L. (Passifloraceae) (&ldquo;mburucuy&aacute;&rdquo;, &ldquo;maracuj&aacute;-laranja&rdquo;), a widely distributed species in South America, has been traditionally considered as a medicinal, ornamental, and edible plant. Its fruits, besides being well<br />appreciated for their flavor and sweetness, have valuable nutritional properties. The ethnopharmacological use as anxiolytic, antistress and sedative of the non-reproductive<br />aerial parts (leaves and tendrils) from P. caerulea is the main reason for its commercialization as an infusion herb. In general, its preparations are rich in flavonoids (chrysin and C-glycosides such as vitexin, isovitexin, orientin, isoorientin, and vicenin-2, among others), which were confirmed as responsible for the anxiolytic effect. The Argentine Pharmacopeia includes a monograph on P. caerulea, while other Pharmacopeias (Brazilian, European, and British) consider it as a potential<br />adulterant of P. incarnata and P. edulis. Harmala alkaloids from P. caerulea might also contribute to the anxiolytic effect, but no preclinical assays have been conducted to confirm it. Pharmacological research confirmed other bioactivities associated with the aerial parts of P. caerulea, such as antimicrobial, gastroprotective, analgesic, and anticonvulsant. Even though, no systematic toxicological assessments have been conducted in order to evaluate the safety in the consumption, especially regarding the presence of cyanogenic glycosides both in the vegetal drug and the unripe fruits.</p>]]></dcterms:abstract>

Don (Dicotyledoneae) family. Recently, remarkable developments have been achieved regarding its domestication,<br />propagation, and biochemical study. The first results on the structural study of the complex mixture of saponins produced by different organs of the tree have been published. Analogously to the related species Q. saponaria Molina, its saponins, either alone or in colloidal formulations, have been proved to be very effective as adjuvants in experimental vaccines, being able to elicit an early humoral and cellular response against the co-administered antigens.     <dcterms:abstract><![CDATA[<strong><strong></strong></strong>Smilax campestris Griseb. (Smilacaceae) (&ldquo;zarzaparrilla&rdquo;) is a medicinal plant species widely used by traditional communities in South America. In general, the plant parts used by popular medicine are the rhizophore (underground stem) and roots, in the form of infusions or decoctions. Their main indications are: antirheumatic, antisyphilitic, diuretic, diaphoretic, anti-inflammatory, antiseptic, and to treat skin conditions (i.e. psoriasis) and gout; among others. There is lacking scientific evidence regarding the phytochemistry of such aqueous extracts, as well as their biological and pharmacological properties. Both aerial and underground organs have been described to contain polyphenolic acids, flavonoids (aglycones, glycosides and proanthocyanidin oligomers/polymers) and saponins; but no systematic research on the isolation of other families of natural products has been conducted, yet. The production of secondary metabolites (in particular flavonoids) is highly dependent on both environmental conditions (solar radiation, herbivores, etc.) of habitats and the phenological stage of the plant itself. The scientific pharmacological in vitro tests performed on S. campestris extracts have confirmed their antioxidant and anti-inflammatory activities, as well as the lack of acute toxicity. However, they have failed to demonstrate diuretic effects in in vivo models; results on the antimicrobial potential are controversial. Further research is needed to evaluate the other ethnomedicinal properties attributed to the vegetal drug. This chapter presents a critical review of the special literature on S. campestris including the most updated information available in journals, books and the internet.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>Objective: To evaluate if the inclusion of nutritional warnings in food ordering web-sites can discourage consumers from purchasing foods with excessive content of nutrients associated with non-communicable diseases (NCD).</p>
<p>Design: Participants were randomly assigned to one of the two experimental conditions: control (n 225) or nutritional warnings (n 222). Nutritional warnings corresponded to separate black octagonal signs containing the word &lsquo;Excess&rsquo; followed by the corresponding nutrient: total fat, saturated fat, sugars and sodium.</p>
<p>Participants were asked to purchase a lunch for themselves using a simulated food ordering website.</p>
<p><br />Setting: Online study in Uruguay.</p>
<p><br />Participants: Convenience sample of 447 Uruguayan participants, recruited using social media.</p>
<p>Results: In the control condition, 76 % of the participants selected a dish or a beverage with excessive content of at least one nutrient in the simulated food ordering website. When nutritional warnings were included, this percentage significantly decreased to 62 % (P = 0&middot;002). In addition, nutritional warnings caused a significant reduction in the percentage of participants who selected dishes with excessive content of total fat: 50 % v. 62 % (P = 0&middot;012).</p>
<p>Conclusions: Results from the present work provide preliminary evidence that the inclusion of nutritional warnings in food ordering websites could discourage consumers from selecting dishes and beverages with excessive content of nutrients associated with NCD.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>Increasing consumption of ultra-processed products from early age has raised growing concerns. In this context, the aims of the present work were: i) to explore the automatic mental associations raised by packages of ultra-processed products among parents of children under 12 months of age, ii) to estimate the valence of the affective reactions elicited by ultra-processed products, and iii) to explore the influence of socio-demographic variables and feeding styles on the valence of parents&rsquo; associations with ultra-processed products. A total of 419 parents of children under 12 months of age were recruited at 20 health facilities that offer well-child services, selected using probability proportional to size sampling based on the number of births at each institution. Participants were presented with the labels of six ultra-processed products (corn snack, frankfurters, milk dessert, orange juice, wafer cookies, yogurt) and were asked to indicate the first words that came to their mind. The products raised mixed mental associations, which were mainly related to pleasure and health. Responses from the majority of participants (61&ndash;82%) evidenced negative associations with corn snacks, frankfurters and wafer cookies, whereas responses from 35 to 45% of the participants suggested a positive affective reaction towards orange juice, milk dessert and yogurt. Likelihood of having a positive affective reaction towards the evaluated ultra-processed products was higher for parents of 1 to 4 months infants, as well as those from low educational level and low socio-economic status. In addition, beliefs about three feeding practices (restriction on diet quality, indulgence in screen use and pressure for finishing) significantly influenced the valence of the associations. Results from the present work suggest the need to implement regulatory measures to reduce positive associations with ultra-processed products among parents and contribute to reduce their consumption among infants.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>The aim of the present work was to explore the influence of consumer characteristics on purchase decisions when facing<br />products with nutritional warnings a few days after their implementation in Uruguay. A non-probabilistic sample of 917<br />participants was obtained using an advertisement on social media. Participants were asked if they had seen the warning<br />signs when making their food purchases and if they had seen the warnings on any product they intended to buy. Participants<br />who answered affirmately (n= 616) were asked about their purchase decision by answering the question &ldquo;What have you<br />done with the product?&rdquo; using the following response options: &lsquo;I purchased it anyway&rsquo;, &lsquo;I purchased a similar product with<br />fewer excess signs&rsquo;, &lsquo;I purchased a similar product without excess signs&rsquo;, &lsquo;I didn&rsquo;t purchase the product or any similar one&rsquo;.<br />Univariate and multivariate multinomial logistic regression models were used to explore the influence of individual variables on participants&rsquo; likelihood of having taken different decisions when facing a product with warnings. Results showed that participants older than 55 years were more likely to react to the warnings by purchasing a similar product with fewer warnings or by not purchasing any product. Likelihood of modifying purchase decisions due to the inclusion of the warnings was associated with a frequent consumption of natural and minimally processed foods and a low consumption frequency of ultra-processed products. These results provide insights to target efforts to promote the use of nutritional warnings in decision making.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<span>Infections caused by multidrug resistant (MDR) bacteria are a major public health threat. Carbapenems are among the most potent antimicrobial agents that are commercially available to treat MDR bacteria. Bacterial production of carbapenem-hydrolysing metallo-&beta;-lactamases (MBLs) challenges their safety and efficacy, with subclass B1 MBLs hydrolysing almost all &beta;-lactam antibiotics. MBL inhibitors would fulfil an urgent clinical need by prolonging the lifetime of these life-saving drugs. Here we report the synthesis and activity of a series of 2-mercaptomethyl-thiazolidines (MMTZs), designed to replicate MBL interactions with reaction intermediates or hydrolysis products. MMTZs are potent competitive inhibitors of B1 MBLs&nbsp;</span><em>in vitro</em><span>&nbsp;(</span><em>e.g.</em><span>,&nbsp;</span><em>K</em><small><span>i</span></small><span>&nbsp;= 0.44 &mu;M&nbsp;</span><em>vs.</em><span>&nbsp;NDM-1). Crystal structures of MMTZ complexes reveal similar binding patterns to the most clinically important B1 MBLs (NDM-1, VIM-2 and IMP-1), contrasting with previously studied thiol-based MBL inhibitors, such as bisthiazolidines (BTZs) or captopril stereoisomers, which exhibit lower, more variable potencies and multiple binding modes. MMTZ binding involves thiol coordination to the Zn(</span><small>II</small><span>) site and extensive hydrophobic interactions, burying the inhibitor more deeply within the active site than&nbsp;</span><small>D</small><span>/</span><small>L</small><span>-captopril. Unexpectedly, MMTZ binding features a thioether&ndash;&pi; interaction with a conserved active-site aromatic residue, consistent with their equipotent inhibition and similar binding to multiple MBLs. MMTZs penetrate multiple Enterobacterales, inhibit NDM-1&nbsp;</span><em>in situ</em><span>, and restore carbapenem potency against clinical isolates expressing B1 MBLs. Based on their inhibitory profile and lack of eukaryotic cell toxicity, MMTZs represent a promising scaffold for MBL inhibitor development. These results also suggest sulphur&ndash;&pi; interactions can be exploited for general ligand design in medicinal chemistry.</span>]]></dcterms:abstract>

Macrophages are among the first cells to be parasitized and, after activation by inflammatory stimuli, they participate in the control of infection. However, some parasites manage to evade the immune response and establish a chronic infection in differentiated cells. L-arginine is located at the crossroads of divergent routes that produce metabolites, including nitric oxide and polyamines, which influence the outcome (i.e. resolution or progression) of infection.     <dcterms:abstract><![CDATA[Los hígados de merluza (Merluccius hubbsi) son un subproducto de la industria fileteadora y se utilizan como fuente de aceite. En este trabajo se estudia el aceite contenido en dichos hígados durante su almacenamiento a 4oC y a -20o C frente a la del aceite extraído de ellos y guardado en iguales condiciones. El aceite contenido en los hígados almacenados a -20o C, se oxida en un par de semanas; el aceite extraído de ellos y almacena do en iguales condiciones se conserva durante varios meses. El problema principal radica en que, una vez que el aceite se deteriora en los hígados, no es posible estabilizarlo por el agregado de antioxidantes. Para obtener un aceite de buena calidad es imprescindible que los hígados se almacenen a una temperatura menor de -20oC, que su aceite se extraiga lo antes posible, que se le adicione de antioxidantes adecuados y que se le almacene refrigerado.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>Baccharis dracunculifolia DC. (Asteraceae) is an aromatic plant species widely distributed across and along South American countries where it grows in different environments, even being considered, in some cases, as a hard to control weed. Its essential oil is highly appreciated in the fragrance/perfumery industry by its woody, foral, and green notes; mainly attributed to (E)-nerolidol and other sesquiterpene alcohols. Recent research has demonstrated a wide range of applications for the essential oil besides its aromatic properties. Although B. dracunculifolia cannot be considered as a medicinal plant for most of the indigenous communities in South America (except for Brazil), modern pharmacological research has demonstrated its high potential for the development of new therapeutic drugs, mainly with antiulcer, anti-infammatory, antiparasitic and anticancer effect. The special literature regarding B. dracunculifolia is profuse and thus, this chapter attempts to summarize the principal knowledge about this species.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p>Background: Sialic acids are widely distributed in nature and have biological relevance owing to their varied structural and functional roles. Immobilized neuraminidase can selectively remove terminal N-acetyl neuraminic acid from glycoproteins without altering the protein backbone while it can be easily removed from the reaction mixture avoiding sample contamination. This enables the evaluation of changes in glycoprotein performance upon desialylation.</p>
<p><br />Methods: Neuraminidase was immobilized onto agarose activated with cyanate ester groups and further used for<br />desialylation of model glycoproteins, a lysate from tumour cells and tumour cells. Desialylation process was analysed by lectin binding assay, determination of sialyl-Tn or flow cytometry.</p>
<p><br />Results: Clostridium perfringens neuraminidase was immobilized with 91 % yield and expressed activity yield was<br />of 41%. It was effective in the desialylation of bovine fetal serum fetuin, bovine lactoferrin and ovine submaxilar mucin. A decrease in sialic-specific SNA lectin recognition of 83% and 53 % was observed for fetuin and lactoferrin with a concomitant increase in galactose specific ECA and PNA lectin recognition. Likewise, a decrease in the recognition of a specific antibody (82%) upon mucin desialylation was observed. Moreover, desialylation of aprotein lysate from the sialic acid-rich cell line TA3/Ha was also possible leading to a decrease in 47 % in SNA recognition. Immobilized neuraminidase kept 100% of its initial activity upon five desialylation cycles.</p>
<p>Conclusions: Immobilized neuraminidase is an interesting as well as a robust biotechnological tool for enzymatic<br />desialylation purposes.</p>
<p>General significance: Immobilized neuraminidase would contribute to understand the role of sialic acid in biological processes.</p>]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[<p style="text-align: left;">The elucidation of glycans biological function is essential to<br />understand their role in biological processes, both normal and<br />pathological. Immobilized glycoenzymes are excellent tools for this purpose as they can selectively release glycans from glycoproteins without altering their backbone. They can be easily removed from the reaction mixture avoiding their<br />interference in subsequent experiments. Here, we describe the<br />immobilization of peptide-N-glycosidase F (PNGase F) onto<br />silica magnetic nanoparticles with immobilization yields of<br />86% and activity yields of 12%. Immobilized PNGase F showed<br />higher thermal stability than its soluble counterpart, and could<br />be reused for at least seven deglycosylation cycles. It was<br />efficient in the deglycosylation of several glycoproteins (ribonuclease B, bovine fetuin, and ovalbumin) and a protein lysate from the parasite Fasciola hepatica under nativeconditions, with similar performance to that of the solubleenzyme. Successful deglycosylation was evidenced by adecrease in specific lectin recognition of the glycoproteins (40%&ndash;80%). Moreover, deglycosylated F. hepatica lysate allowed us to confirm the role of parasite N-glycans in the inhibition of the lipopolysaccharide-induced maturation of dendritic cells. Immobilized PNGase F probed to be a robust biotechnological tool for deglycosylation of glycoproteins and complex biological samples under native conditions.</p>]]></dcterms:abstract>
