<dcterms:abstract>
Purpose Instrumental capabilities and software tools of modern hybrid mass spectrometry (MS) instruments such as high-resolution mass spectrometry (HRMS), quadrupole time-of-flight (QTOF), and quadrupole linear ion trap (QLIT) were experimentally investigated for the study of emerging contaminants in Henares River water samples. Methods Automated screening and confirmatory capabilities of QTOF working in full-scan MS and tandem MS (MS/MS) were explored when dealing with real samples. Investigations on the effect of sensitivity and resolution power influence on mass accuracy were studied for the correct assignment of the amoxicillin transformation product 5(R) amoxicillin-diketopiperazine-2′,5′ as an example of a nontarget compound. On the other hand, a comparison of quantitative and qualitative strategies based on direct injection analysis and off-line solid-phase extraction sample treatment were assayed using two different QLIT instruments for a selected group of emerging contaminants when operating in selected reaction monitoring (SRM) and information-dependent acquisition (IDA) modes. Results and discussion Software-aided screening usually needs a further confirmatory step. Resolving power and MS/MS feature of QTOF showed to confirm/reject most findings in river water, although sensitivity-related limitations are usually found. Superior sensitivity of modern QLIT-MS/MS offered the possibility of direct injection analysis for proper quantitative study of a variety of contaminants, while it simultaneously reduced the matrix effect and increased the reliability of the results. Confirmation of ethylamphetamine, which lacks on a second SRM transition, was accomplished by using the IDA feature. Conclusion Hybrid MS instruments equipped with high resolution and high sensitivity contributes to enlarge the scope of targeted analytes in river waters. However, in the tested instruments, there is a margin of improvement principally in required sensitivity and data treatment software tools devoted to reliable confirmation and improved automated data processing.
</dcterms:abstract>

    <dcterms:abstract>The Latin American flora contains one of the greatest biodiversities in the world. Thus renewable natural resources found in this region acquire an outstanding importance in the search for new useful raw materials for the fragrance and flavor industry. Many chemical and pharmacological studies have been carried on these aromatic resources. However, aromatic quality evaluations are quite scarce. Actually, several analytical technologies are applied to profiling aromatic. These are key issues that influence the development of new agrobusiness ventures. Sensory profiling by different analytical techniques in addition to the chemical characterization can contribute to a better knowledge of these native aromatic plant resources and encourage their potential industrial purposes. Such has been the case of Adesmia boronioides of Patagonia, its essential oils has been used to launch a perfume for a major international cosmetics company in its “Aromas de América” campaign. Actually we have studied other aromatic species of South America, Mulinum spinosum, motivated by previous literature proposing this plant as a surrogate of galbanum, a raw material highly appreciated in perfumery. Chemical analysis was applied by GC-FID-MS and sensory characteristics of the essential oil of this species were evaluated by GC-O and descriptive analysis and compared with that of a commercial sample of galbanum. An experienced panel of reviewers smelled the effluent gas chromatograph using an olfactometer, identifying and describing significant aromas related to chromatographic peaks. The aromagrams obtained from both samples differed significantly, however showed the same descriptors, derivatives of 2-methoxypyrazines were identified as responsible for the characteristic scent note in both products giving the olfactory similarity. Despite this, the overall flavor profile of both products differs substantially, giving Mulinum spinosum own olfactory and promising features. <dcterms:abstract>Monitoring economic access to food is fundamental to implement strategies to reduce food insecurity and enhance the affordability of healthy diets. The aims of the present study were to develop a healthy food basket (HFB) and analyze its cost from 2011 to 2024, in Uruguay, a highincome Latin American country. The HFB was conceptualized as a set of foods that allows the achievement of the nutritional needs of a person at the lowest possible cost, considering the dietary patterns of the local population. Its construction was based on national and international dietary intake recommendations, local food acquisition patterns, and food prices. The daily cost of the HFB in May 2024 was USD 7.5, considering purchasing power parity (PPP) conversion factors. The cost of the basket was higher than the Cost and Affordability of a Healthy Diet elaborated by FAO for the country and the region and higher than the cost of the official food baskets of the country. The cost of the HFB increased 202.3% from January 2011 to May 2024, while inflation increased by 178.4%. Fruits and vegetables, meat, dairy products, and processed cereals made the largest contribution to the cost of the HFB. Results highlight the need to reassess the country’s poverty estimation criteria and implement strategies to improve the affordability of healthy foods, such as adjusting cash transfer amounts and introducing targeted subsidies. The proposed methodological approach can contribute to the standardization of approaches to measure and monitor the cost of healthy diets in other countries.</dcterms:abstract>

    <dcterms:abstract>Nowadays, fertilization and pest control are carried out using chemical compounds that contaminate soil and deteriorate human health. Plant growth promoting bacteria endophytes (PGPBEs), are a well-studied group of bacteria that offers benefits to the host plant, such as phytostimulation, biofertilization, and protection against other microorganisms. The study of Gluconacetobacter diazotrophicus–which belongs to PGPBEs-aids the development of alternative strategies of an integrated approach for crop management practices. Ralstonia solanacearum is responsible for bacterial wilt disease. This phytopathogen is of great interest worldwide due to the enormous economic losses it causes. In this study the action of G. diazotrophicus as a growth promoting bacterium in Arabidopsis thaliana seedlings is analyzed, evaluating the antagonistic mechanisms of this beneficial endophytic bacterium during biotic stress produced by R. solanacearum. Effective colonization of G. diazotrophicus was determined through bacterial counting assays, evaluation of anatomical and growth parameters, and pigments quantification. Biocontrol assays were carried out with Ralstonia pseudosolanacearum GMI1000 model strain and R. solanacearum A21 a recently isolated strain. Inoculation of A. thaliana (Col 0) with G. diazotrophicus Pal 5 triggers a set of biochemical and structural changes in roots, stems, and leaves of seedlings. Discrete callose deposits as papillae were observed at specific sites of root hairs, trichomes, and leaf tissue. Upon R. pseudosolanacearum GMI1000 infection, endophyte-treated plants demonstrated being induced for defense through an augmented callose deposition at root hairs and leaves compared with the non-endophyte-treated controls. The endophytic bacterium appears to be able to prime callose response. Roots and stems cross sections showed that integrity of all tissues was preserved in endophyte-treated plants infected with R. solanacearum A21. The mechanisms of resistance elicited by the plant after inoculation with the endophyte would be greater lignification and sclerosis in tissues and reinforcement of the cell wall through the deposition of callose. As a consequence of this priming in plant defense response, viable phytopathogenic bacteria counting were considerably fewer in endophyte-inoculated plants than in not-inoculated controls. Crystal structure parameters were accurately determined using 10 000 eV photons, and the Pr occupancy was refined using data collected with 5962 eV photons, close to the Pr LI I I absorption edge. BaCe1ı xPrxO3ı ı crystallizes in the Pnma (No. 62) space group for all x values. Pr cations are mainly located at the Ce sites (perovskitesB site), but a small fraction of them increasingly substitute some of the Ba ionsat the A site asPr content increases. The Pr doping introduces electronic defects (Pr+3/Pr+4) and oxygen vacancies needed for H2O incorporation and H-ionic conductivity. A decrease in the orthorhombic distortion would produce the opposite effects on the electronic and ionic mobility. The electronic mobility should increase due to an improvement in the overlap of the (Ce/Pr)4f–O2p orbital, while the proton mobility should decrease as a consequence of a larger hopping distance</dcterms:abstract>

    <dcterms:abstract>Despite the great advances in chemotherapeutics, infectious diseases are still one of the leading causes of death worldwide. Among some of the clinically relevant pathogens, methicillin-resistant Staphylococcus aureus (MRSA) ranks as one of the most difficult bacteria to treat. It is a common cause of skin, soft-tissue, and endovascular infections, as well as pneumonia, septic arthritis, endocarditis, osteomyelitis, and sepsis. The research on Basidiomycota is extensive; many species show a broad spectrum of pharmacological activities, including antimicrobial activity. The vast majority of the literature to date generally focuses on screening the antibacterial properties of mushroom extracts. A gap still exists in the identification of the individual compounds responsible for these properties, and few low molecular weight compounds have been described. Gymnopilus junonius, the big laughter mushroom, grows wild in Uruguay, especially on Eucalyptus spp. plantations; it is known as the "eucalyptus fungus." In this work, we report the bioguided isolation, structural elucidation, and antistaphylococcal activity of the main antimicrobial components of fresh basidiocarps of G. junonius.</dcterms:abstract>

    <dcterms:abstract>Chagas’ disease is the major endemic disease in South and Central America caused by a trypanosomatid parasite (Trypanosoma cruzi). The current treatment relies on two old and nonspecific chemotherapeutic agents, Nifurtimox and Benznidazole. Despite the major advances that have been made in the identification of specific targets that afford selectivity, the drugs used today have serious side effects. Furthermore, differences in drug susceptibility among different T. cruzi isolates have led to varied parasitological cure rates depending on the geographical region. There is, therefore, an urgent need for the development of new antichagasic drugs. In this regard we have spent more than a decade in the search for more effective agents able to compromise the proliferation of T. cruzi. We began our research with our own compounds and then continued with compounds from other researcher groups. We systematically characterized representatives of a wide range of different chemical families. In this review we summarize our ongoing efforts to identify potential anti-T. cruzi agents using our compound-library. It is discussed and presented the structure- activity relationship observed among the different groups of chemical families.</dcterms:abstract>

    <dcterms:abstract>In a continuing effort to identify new active compounds for combating Chagas disease and other neglected diseases, our research group synthesized and evaluated 23 3-arylquinoxaline-2-carbonitrile di-N-oxides against Trypanosoma cruzi. Five of them presented IC50 values of the same magnitude as the standard drug Nifurtimox, making them valid as new lead compounds. The optimized molecular structures of 23 derivatives represented by 1497 types of DRAGON descriptors were subjected to linear regression analysis, and the derived QSAR was shown to be predictive. In this way, we achieved a rational guide for the proposal of new candidate structures whose activities still remain unknown.</dcterms:abstract>

    <dcterms:abstract>Autoimmunity is a feature of celiac disease (CD) with tissue transglutaminase (tTG) as a major autoantigen. A correlation between gynecological-obstetric disorders in CD patients and the presence of circulating antibodies anti-tTG that inhibited tTG activity was reported. Serum anti-tTG antibodies were detected in a non-obese diabetic (NOD) mouse model of type I insulin-dependent diabetes mellitus and Sjögren's syndrome, two comorbid states with CD. Since pregnancy complications have been described in NOD mice, we evaluated the ability of anti-tTG antibodies to affect the functions of tTG relevant to the normal course of an early pregnancy like extracellular matrix assembling and apoptotic cell phagocytosis by macrophages. Circulating IgG antibodies against tTG were detected in NOD mice with titers that decreased at early pregnancy; interestingly, the in vitro transamidating activity of tTG was reduced by NOD serum samples. Particularly, anti-tTG antibody inhibited apoptotic cell phagocytosis by peritoneal macrophages from pregnant NOD mice that express the enzyme on surface. Evidence provided support for a role for anti-tTG antibodies through reduced transamidating activity and reduced apoptotic cell clearance by the macrophages of pregnant NOD mice.</dcterms:abstract>

The synthetic route to access to these derivatives was an efficient Wittig reaction performed in mild conditions with polymer-supported triphenylphosphine (PS-TPP). Additionally, the benzofurozan analogues, deoxygenated benzofuroxans, were prepared using PS-TPP as reductive reagent in excellent yields. The trypanosomicidal and leishmanocidal activities of the benzofuroxan derivatives were measured and also some aspects of their mechanism of action studied. In this sense, inhibition of mitochondrial dehydrogenases activities, production of intra-parasite free radicals and cruzipain inhibition were studied as biological target for the anti-trypanosomatid identified compounds. The trypanosomicidal activity could be the result of both the parasite-mitochondrion function interference and production of oxidative stress into the parasite</dcterms:abstract>

    <dcterms:abstract>Crude extract (CE) from pulp of Bromelia antiacantha Bertol. mature fruit, contains at least 3 cysteine proteases with proteolytic activity. By single step cation exchange chromatography (Hi-trap SP-HP) of partially purified CE, the protease with the lowest pI, Antiacanthain A (AntA), was isolated. It showed maximum activity at pH9, and 75% of remaining activity was maintained over a wide pH range (pH6-10). The AntA activity exhibits a constant increase up to 70°C. Maintains almost 100% of its activity at 45 at pH6 and 9. A 60% of AntA was active by titration with specific inhibitor, E64. Amidasic activity was studied with pyroglutamyl-phenyl-leucyl-paranitroaniline (PFLNA) substrate having higher AntA catalytic efficiency of (kcat/Km=470s-1M-1) relative to stem bromelain (kcat/Km=305s-1M-1). Esterase activity using p-nitrophenyl esters of N-α-CBZ-l-Lysine (z-L-LysONp) showed a 10-fold higher catalytic efficiency for AntA (kcat/Km=6376s-1M-1) relative to stem bromelain (kcat/Km=688s-1M-1). Incubation with 8M Urea did not affect AntA activity and remained unchanged for 18h, with 6M GndHCl resulted in a 41% decrease in activity after 30min incubation, maintained this activity 18h. AntA exhibits high sequence identity with proteases of the Bromeliaceae family.</dcterms:abstract>

    <dcterms:abstract>In spite of the great advances in chemotherapeutics, infectious diseases are still one of the leading causes of death in the world. Amongst the most problematic clinically relevant pathogens at present, methicillin–resistant Staphylococcus aureus (MRSA) ranks as one of the most difficult bacteria to treat. Interest in plants with antimicrobial properties has revived as a consequence of current problems associated with the use of antibiotics. The objective: was to evaluate the antibacterial activity of a compound isolated from Ibicella lutea (Lindl.) Van Eselt. (Martyniaceae) and some of its derivatives, against methicillin–sensitive and MRSA. Compound 1 was isolated and identified as previously described and minimal inhibitory concentration (MIC) of 1 and simple analogs were determined by the microdilution technique. Compound 1 (11–hydroxystearic acid 11–0–(6–0–acetyl–β–D–grucopyranoside) showed a MIC of 0.06 mM for S. aureus ATCC 6538p. All the synthetic analogs and precursors presented higher MIC values. Compound 1 showed antimicrobial activity against different strains of MRSA. These results suggest that the glycosidic and acidic moieties are necessary for the antimicrobial activity under study.</dcterms:abstract>

The heterocyclic N atom nearest to the Cl atom and the Nsulfonamide atom could be coordination sites for the silver ion in the Ag–SCP–SCN complex. The Ag–SCP complex is a polymeric compound with metal–metal bonds, and the heterocyclic and sulfonamide N atoms are points of coordination for AgI. Both complexes showed activity against all the tested bacteria, and in the cases of Escherichia coli and Pseudomonas aeruginosa, the action was better than that of SCP. In all cases, both silver–SCP complexes showed better antifungal activity than SCP, which was inactive against the tested fungi. Notably, the activity against P. aeruginosa, a nosocomial multidrug-resistant pathogen, was better than that of the reference antibiotic cefotaxim. Both silver–sulfa complexes displayed moderate activity against the tested yeast, especially for C. neoformans, which is an important fact considering the incidence of cryptococcosis, mainly in immune-deficient patients. No chromosomal aberrations were observed with the Allium cepa test, which is auspicious for further study of these complexes as potential drugs.</dcterms:abstract>

    <dcterms:abstract>he aims of this study were to investigate whether a Th1-or a Th2-type response is stimulated in the first stages of experimental infection with Echinococcus granulosus, and to determine whether live or dead protoscoleces equally contribute to such Th1/Th2-type polarization. Live parasites stimulated the production of IL-10, IL-4 and IL-5 as early as week 1 postinoculation. The levels of IL-10 and IL-4 decreased towards week 4 p.i. and that of IFNγ increased. The production of specific antibodies was characterized by high levels of systemic IgG1 and local IgM and IgG3 (measured in peritoneal lavages). In contrast, dead parasites induced elevated levels of IL-4, IFNγ, IL-10 and IL-5 on week 1 postinoculation followed by a decrease of IFNγ and an increase of IL-4. Low levels of specific antibodies were stimulated by dead parasites both systemically and in the peritoneal cavity. These results show that E. granulosus infection induced an early Th2-type response and that live parasites stimulated stronger antibody responses than dead parasites. In addition, they strongly suggest that both phenomena were modulated by live protoscoleces.</dcterms:abstract>

    <dcterms:abstract>BACKGROUND: When feeding preterm infants, donor milk is preferred if the mother's own milk is unavailable. Pasteurization may have detrimental effects on bioactivity, but more information is needed about its effects on the immunological compounds. Research aim: This work has two main aims: evaluate the antibody profile of colostrum and study the quantitative variations in the antibodies' level and specific reactivity after undergoing Holder pasteurization. The authors focused on immunoregulatory components of colostrum (antidietary antibodies and TGF-β2) in the neonatal gut. METHODS: This is a descriptive cross-sectional study of a convenience sample of 67 donated colostrum samples at different days after delivery, both raw and pasteurized. Antibody profiles were analyzed at different times during breastfeeding, and total and specific antibodies (IgM, IgA, and IgG subclasses) were compared with tetanus toxoid and ovalbumin using enzyme-linked immunosorbent assay. The processing effect on total and specific antibodies, as well as TGF-β2, was evaluated by paired analyses. RESULTS: No variations in immunological compounds were observed throughout the colostrum stage. The TGF-β2, antibodies' concentrations, and antibodies' specific reactivity after pasteurization did not vary significantly as days of lactation varied. Changes in antibody levels were dependent on isotype and IgG subclass, and IgG4 showed remarkable resistance to heating. Moreover, the effect of the pasteurization on specific reactivity was antigen dependent. CONCLUSION: The supply of relevant immunological components is stable throughout the colostrum stage. The effects of pasteurization on antibodies depend on isotype, subclass, and specificity. This information is relevant to improving the immunological quality of colostrum, especially for preterm newborns.</dcterms:abstract>

    <dcterms:abstract>Cystic echinococcosis is a zoonotic disease caused by the cestode parasite Echinococcus granulosus. In endemic regions, seropositive individuals to E. granulosus usually and markedly outnumber image-confirmed cases of cystic echinococcosis, suggesting that some parasite challenges derive in unsuccessful infection establishments. However, it is still unknown whether such parasite-specific antibodies in healthy individuals might play a role in resistance/susceptibility to the infection. Therefore, we have here analysed the profile of antibodies recognizing E. granulosus antigens in seropositive but ultrasound normal individuals, as well as in surgery-confirmed patients and healthy donors. Our results showed that ultrasound normal individuals exhibited low avidity IgG antibodies, as well as low levels of parasite-specific IgG1 and IgG4 antibodies. In addition, they displayed significant levels of specific IgE, and thus, they revealed a uniquely high IgE:IgG4 ratio. Moreover, high levels of parasite-specific IgM were detected in such individuals, which showed characteristics of natural cross-reacting antibodies. Therefore, our results indicate that ultrasound normal individuals but seropositive for E. granulosus antigens exhibit a distinctive antibody profile. In this regard, possible associations between their antiparasite antibodies and potential resistance mechanisms to cystic echinococcosis are discussed.</dcterms:abstract>

    <dcterms:abstract>The aims of this work were to investigate the existence of Tindependent antigens in Echinococcus granulosus protoscoleces and to evaluate the relative contribution of T-independent stimulation to the overall antibody response in early infection. Mice depleted of CD4þ-cells were immunized with protoscolex somatic antigens (PSA) or infected with E. granulosus protoscoleces (PSC). Results showed that the response of CD4-depleted immunized mice had the expected characteristics of a T-independent stimulation and that such T-independent stimulation was important mainly during primary response. During infection absence of CD4þ-cells affected mainly the secretion of all IgG subclasses with the exception of IgG3 and IgM. To carry out a preliminary isolation of PSC T-independent antigens we prepared a carbohydrate enriched fraction from protoscolex antigens, using a monoclonal antibody specific for the carbohydrate moiety Gala(1,4)Gal highly expressed in PSC. This fraction was mitogenic for naive mouse splenocytes and was recognized by a high percentage of the specific antibodies secreted by CD4-depleted immunized or infected mice. In summary, these results suggest that E. granulosus protoscoleces contain immunogenic T-independent antigens. Primary antibody responses to protoscolex somatic antigens and the production of IgM and IgG3 in early infection would be mainly stimulated by a T-independent mechanism.</dcterms:abstract>
