<dcterms:abstract>The volatile compounds of 18 yerbamate (YM) commercial brands fromArgentina, Brazil, P.araguay and Uruguay were extracted using a Likens–Nickerson simultaneous distillation–extraction (SDE) apparatus, and analyzed by gas chromatography–mass spectrometry (GC–MS). Results showed that a total of 35 major volatile compounds were identified by GC–MS; among them, norisoprenoid compounds. A principal component analysis (PCA) was carried out in order to ascertain the degree of differentiation between samples and which compounds were involved. The results showed threemain groups of YM corresponding to the places where they are commercialized. The first group (samples from Paraguay and Argentina) correlated positively with 6-methyl-5-hepten-2-one, octanal and 1-octanol. The second group (YM marketed in Uruguay but produced in Brazil) correlated positively with α- and β-ionone, linalool, nerol, α-terpineol, (Z)-linalool oxide and the aliphatic hydrocarbons undecane, dodecane and heptadecane. The third group (YM produced and marketed in Brazil) correlated positively with 2,4-heptadienal isomers, (E)-linalool oxide and β-cyclocitral. Through a preference test, one commercial brand was selected to be analyzed by gas chromatography–olfactometry (GC–O). Sixteen odor active compounds were perceived by GC–O analysis. It seems that the norisoprenoid compounds and components produced by fatty acid degradation play a vital role in the special aroma of yerba mate.</dcterms:abstract>

    <dcterms:abstract>Glutathione transferases (GSTs) perform several catalytic and non-catalytic roles in the defense against toxicities of electrophile compounds and oxidative stress, and therefore are involved in stress-response and cell detoxification. Previously, we have provided evidence indicating that EgGST2 and EgGST3, two phylogenetically distant Echinococcus granulosus GSTs, can naturally form a heterodimeric structure (EgGST2-3). In the present work, the recombinant heterodimer GST (rEgGST2-3) is characterized. Hence, rEgGST2-3 was able to conjugate GSH to three substrates: 1-chloro-2,4-dinitrobenzene (CDNB, general substrate for GSTs), 1,2-dichloro-4-nitrobenzene (specific substrate for mammalian Mu class) and trans,trans-deca-2,4-dienal (reactive carbonyl). The canonical activity was considerably reduced by all the conventional inhibitors (cybacron blue, triphenylthin chloride and bromosulfophthalein) and by other inhibitors (ellagic acid, alizarin and chenodeoxycholic acid). Besides this, rEgGST2-3 activity was inhibited by a number of anthelmintic drugs, where the halogenated phenolic drugs (mainly bithionol and hexachlorophene) acted as stronger inhibitors, suggesting they may bind to the EgGST2-3. Moreover, rEgGST2-3 exhibited glutathione-peroxidase activity, and its specific constant (kcat/KM) was calculated. Finally, rEgGST2-3 displayed the ability to bind non-substrate molecules, particularly anthelmintic drugs, suggesting that ligandin activity may have potential to act as a passive protection parasite mechanism. Overall, the rEgGST2-3 behavior was shown to be both complementary and redundant to that reported for rEgGST1, another characterized GST from E. granulosus. It would be appropriate that different enzymes in the same organism do not have exactly the same functional properties to develop a better adaptation to life in the host.</dcterms:abstract>

    <dcterms:abstract>In social insects, task-related recognition plays an important role in the coordination and cohesion between members of the colony. Tetragonisca angustula is an eusocial stingless bee that presents a sophisticated system of defense involving two complementary groups of guards: hovering and standing guards. We identified, quantified, and compared the cuticular compounds of worker bees captured within the nest, and bees performing tasks outside: foragers and guards. In addition to cuticular hydrocarbons (CHCs), we identified abundant di- and triterpenes. Among the CHCs, we found a mixture of n-alkanes, methyl-branched alkanes, alkenes, and alkadienes. Significant differences in the relative abundance of CHCs between behavioral groups were found. Particularly, guards present high amounts of branched alkanes relative to nest bees and foragers. Differential CHC profiles associated with behavioral groups could imply a mechanism for caste recognition.</dcterms:abstract>

    <dcterms:abstract>The synthesis of novel galactosides is interesting because of their important role in several biological processes. Their properties greatly depend upon the configuration and type of galactoside. Therefore, to study biological activity, it is essential to elucidate the structure of the products. Glycosidases are capable of catalyzing glycosidic linkages with absolute stereoselectivity of the anomeric center. We report the enzymatic synthesis of galactosyl-ethylene glycol, galactosylglycerol, and galactosyl-erythritol by immobilized β-galactosidase from Aspegillus oryzae. The obtained galactosides were isolated and fully characterized by an extensive nuclear magnetic resonance (NMR) study. Complete structure elucidation and full proton and carbon assignments were carried out using 1D (1H and 13C) and 2D (gCOSY, TOCSY, multiplicity-edited gHSQC, and gHMBC) NMR experiments. The β-galactosidase from A. oryzae showed a strong preference for primary alcohols. <dcterms:abstract>The objective measure of grape quality is an important requirement to enhance wine quality. Particularly, the quantitation of glycosylated aroma compounds could be a useful index for winemakers. In the present work, we evaluated the bound compounds profile on Tannat grapes, and the potential of near infrared (NIR) spectroscopy coupled to chemometric techniques to determine the concentration of the different glycosylated aroma compounds present in this grape variety. The results showed phenols and norisoprenoids as the quantitative most important group of compounds, representing almost 80 % of the total glycosidic components present in Tannat grapes. The glycosidic form of ethyl-β-(4-hydroxy-3- methoxy-phenyl)-propionate was identified for the first time in grapes. The results suggested NIR technique as a screening tool for a fast analysis of the grape aroma potential. The application of such methodology would be ideal for fast grape screening purposes, contributing to the winemaking decisions, classification of the fruit, and perhaps maturity monitoring.</dcterms:abstract>

    <dcterms:abstract>Kefir is a fermented probiotic drink obtained by placing kefir granules in a suitable substrate. The kefir granules are a consortium of bacteria and yeasts embedded in a exopolysaccharide matrix. The aim of this research was the isolation and identification of yeasts from kefir of different origin, the evaluation of their antifungal capacity against Aspergillus spp. and the characterization of virulence related traits. Using RFLP of ITS1/ITS4 region, D1/D2 region sequencing and RAPD techniques, 20 kefir isolates were identified as Geotrichum candidum, Pichia kudriavzevii, P. membranifaciens, Saccharomyces cerevisiae and Candida ethanolica. Their antifungal capacity was evaluated by their conidia germination reduction, which allowed the selection of eight isolates with high to moderate conidia germination reduction against A. flavus and A. parasiticus. Furthermore, these selected isolates showed growth inhibition on contact in the dual culture assay for both Aspergillus species and 3 of them ‒belonging to S. cerevisiae and P. kudriavzevii species‒ generated volatile organic compounds which significantly affected the growth of both fungi. For the evaluation of virulence related traits, growth at high temperatures, enzymatic activities and the adhesion to Caco-2 cells were analyzed. The isolates did not present more than one positive virulence-related trait simultaneously. In particular, it is important to highlight that the adhesion capacity to the model of intestinal barrier was extremely low for all of them. According to the results obtained, further studies would be of interest for the possible use of these promising yeasts as biocontrol agents against fungi in food.</dcterms:abstract>

    <dcterms:abstract>To identify molecular markers of cell differentiation in developing nervous tissue, monoclonal antibodies against chick embryo neural retina were made. One of them, 3C3mAb, recognized a developmentally regulated antigen present in several organs of the CNS. Data from MALDI-TOF mass spectrometry and peptide sequencing of the immuno-affinity purified protein indicated identity of the antigen with MARCKS. The immunoreactive material was always found as a unique polypeptide (Mr 71 kDa) in SDS-PAGE, however isoelectrofocusing revealed the existence of several bands (pI ranging from 4.0 to 4.5). Interestingly some retinal cell types, as photoreceptors, exhibited an extremely significant decrease in the intensity of the immunoreactive material during the final phases of terminal differentiation while others, as some retinal neurons, maintained the immunoreactivity when fully differentiated. Taken together these results indicate that MARCKS, a protein susceptible of several posttranslational modifications as myristoylation and phosphorylation at variable extent, may act differently in neural retina cell types.</dcterms:abstract>

    <dcterms:abstract>
The effect on color of the initial pH employed in dulce de leche (DL) production was evaluated through physicochemical and spectroscopical characterization of the melanoidins formed in the process. Melanoidins originated at pH values of 6.5, 7.0,
and 7.5, and they were released by the enzymatic hydrolysis of the protein backbone and purified by gel filtration. They showed a significant degree of polydispersity, in general, with molecular weights (MWs) below 1,800 Da. DL produced at a higher pH
released melanoidins with higher average MW after the enzymatic hydrolysis. They also presented darker colors (dE∗ab, C∗), more closely resembling those typical of the commercial product. Analysis of the fractions isolated by gel filtration using HPLC-DAD and multinuclear NMR showed an heterogeneous and complex composition. Even though structurally related, the 1H NMR spectra of melanoidins showed a higher degree of aromaticity at higher pH values. In conclusion, the pH employed in DL production affects the amount and structure of the colored products originated by MR reactions, and thus the color of the final product.
</dcterms:abstract>

    <dcterms:abstract>Blue mold caused by Penicillium spp. is the most important postharvest disease of apple in Uruguay. Fourteen isolates of Penicillium were recovered from rotten apple and pear fruit with blue mold symptoms, and from water from flotation tanks in commercial apple juice facilities. Phenotypic identification to species level was performed, and the isolates were tested for sensitivity to commonly used postharvest fungicides. Genetic characterization of the isolates was performed with restriction fragment length polymorphism of the region including the internal transcribed spacer (ITS) ITS1 and ITS2 and the 5.8SrRNA gene (ITS1-5.8SrRNA gene-ITS2) ribosomal DNA region and with random amplified polymorphic DNA (RAPD) primers. Both techniques were able to differentiate these isolates at the species level. RAPD analysis proved to be an objective, rapid, and reliable tool to identify Penicillium spp. involved in blue mold of apple. In all, 11 isolates were identified as Penicillium expansum and 3 as P. solitum. This is the first report of P. solitum as an apple pathogen in Uruguay.</dcterms:abstract>

    <dcterms:abstract>Cystic echinococcosis (CE) is a zoonotic disease worldwide distributed, caused by the cestode Echinococcus granulosus sensu lato (E. granulosus), with an incidence rate of 50/100,000 person/year and a high prevalence in humans of 5-10%. Serology has variable sensitivity and specificity and low predictive values. Antigens used are from the hydatid fluid and recombinant antigens have not demonstrated superiority over hydatid fluid. A cell line called EGPE was obtained from E. granulosus sensu lato G1 strain from bovine liver. Serum from CE patients recognizes protein extracts from EGPE cells with higher sensitivity than protein extracts from hydatid fluid. In the present study, EGPE cell protein extracts and supernatants from cell colonies were eluted from a protein G affinity column performed with sera from 11 CE patients. LC-MS/MS proteomic analysis of the eluted proteins identified four E. granulosus histones: one histone H4 in the cell extract and supernatant, one histone H2A only in the cell extract, and two histones H2A only in the supernatant. This differential distribution of histones could reflect different parasite viability stages regarding their role in gene transcription and silencing and could interact with host cells. Bioinformatics tools characterized the linear and conformational epitopes involved in antibody recognition. The three-dimensional structure of each histone was obtained by molecular modeling and validated by molecular dynamics simulation and PCR confirmed the presence of the epitopes in the parasite genome. The three histones H2A were very different and had a less conserved sequence than the histone H4. Comparison of the histones of E. granulosus with those of other organisms showed exclusive regions for E. granulosus. Since histones play a role in the host-parasite relationship they could be good candidates to improve the predictive value of serology in CE.</dcterms:abstract>

    <dcterms:abstract>Expression of Tk antigen, a truncated carbohydrate antigen, was examined in helmith parasites. Using the monoclonal antibody LM389, this antigen was detected in extracts from Taenia hydatigena, Mesocestoides vogae (syn corti), and Taenia crassiceps. No reactivity was observed in Thysanosoma spp., Dipylidium caninum, Fasciola hepatica, and Nyppostrongylus brasiliensis. On the basis of their electrophoretic mobility, diVerent patterns of Tk-bearing glycoproteins were observed among T. hydatigena, M. corti and T. crassiceps by immunoblotting, with certain components resolved as broad bands typical of mucin-like glycoproteins. Most Tk-reactive material remained in the 0.6N perchloric acid-soluble fraction, conWrming that Tk epitopes are carried by mucin-type glycoproteins. Immunohistochemical analysis revealed that in T. hydatigena, Tk antigen is mainly expressed in the tegument, whereas in M. corti the reactivity was principally observed in the subtegumental parenchyma. The presence of a novel tumor-associated carbohydrate antigen in invertebrates, contributes to strengthen the notion that truncated mucin-type O-glycosylation is a normal phenomenon in parasitic worms and may help identify new biological characteristics of helminth parasites.</dcterms:abstract>

    <dcterms:abstract>Flavobacterium sp. AUG42 is a cellulase-producing bacterium isolated from the Antarctic oligochaete Grania sp. (Annelida). In this work, we report that AUG42 produces a glycoside hydrolase cocktail with CMCase, PASCase and cellobiase activities (optimum pHs and temperatures ranging from 5.5 to 6.5 and 40 to 50 °C, respectively). The time-course analyses of the bacterial growth and cellulase production showed that the cocktail has maximal activity at the stationary phase when growing at 16 °C with filter paper as a cellulosic carbon source, among the tested substrates. The analyses of the CAZome and the identification of secreted proteins by shotgun Mass Spectrometry analysis showed that five glycoside hydrolyses are present in the bacterial secretome, which probably cooperate in the degradation of the cellulosic substrates. Two of these glycoside hydrolyses may harbor putative carbohydrate binding modules, both with a cleft-like active site. The cellulolytic cocktail was assayed in saccharification experiments using carboxymethylcellulose as a substrate and results showed the release of glucose (a fermentable sugar) and other reducing-sugars, after 24 h incubation. The ecological relevance of producing cellulases in the Antarctic environment, as well as their potential use in the bio-refinery industry, are discussed.</dcterms:abstract>

Sucrose gradient fractionation demonstrated that these forms of MT4-MMP are all present in lipid rafts.
Mutational and computational analyses revealed that Cys564, which is present within the stem region, mediates MT4-MMP homodimerization by forming a disulfide bond.
Substitution of Cys564 results in a more rapid MT4-MMP turnover, when compared to the wild-type enzyme, consistent with a role for dimerization in protein stability. Expression of MT4-MMP in MDCK cells enhanced cell migration and invasion of Matrigel, a process that requires catalytic activity. However, a serine substitution at Cys564 did not reduce MT4-MMP-stimulated cell invasion of Matrigel suggesting that homodimerization is not required for this process. Deglycosylation studies showed that MT4-MMP is modified by N-glycosylation. Moreover, inhibition of Nglycosylation by tunicamycin diminished the extent of MT4-MMP dimerization suggesting that N-glycans may confer stability to the dimeric form. Taken together, the data
presented here provide a new insight into the characteristics of MT4-MMP, and highlight the common and distinct properties of the GPIanchored MT-MMPs.
</dcterms:abstract>

    <dcterms:abstract>Ultrasonication is known to induce cavitation and can thus accelerate extraction, dissolution, digestion, and other processes, while the intense mixing caused by the propagation of ultrasound waves enhances analyte transfer to the extraction medium. Consequently, ultrasound-assisted extraction (UAE) is viewed as a simple procedure well suited for the extraction and subsequent determination of trace elements in food. Herein, we use different techniques to characterize several processes involved in probe- and bath-based UAE, apply them to the determination of trace metals (Cd, Cu, Ni, Pb, and Zn) in globe artichoke leaves and soybean seeds, and closely monitor the extraction rates of selected analytes. The developed UAE-based method is found to comply with the criteria of Green Chemistry and is concluded to be a reliable, simple, and cheap alternative to standard food analysis techniques.</dcterms:abstract>

    <dcterms:abstract>The larval stage of Echinococcus granulosus causes the chronic infection known as cystic echinococcosis, deploying strong inhibitory mechanisms on host immune responses. Using experimental intraperitoneal infection in C57BL/6 mice, we carried out an in-depth analysis of the local changes in macrophage populations associated with chronic infection. In addition, we analyzed T cells and relevant soluble mediators. Infected animals showed an increase in local cell numbers, mostly accounted for by eosinophils, T cells, and macrophages. Within macrophage populations, the largest increases in cell numbers corresponded to resident large peritoneal macrophages (LPM). Monocyte recruitment appeared to be active, as judged by the increased number of monocytes and cells in the process of differentiation towards LPM, including small (SPM) and converting peritoneal macrophages (CPM). In contrast, we found no evidence of macrophage proliferation. Infection induced the expression of M2 markers in SPM, CPM, and LPM. It also enhanced the expression of the co-inhibitor PD-L1 in LPM, SPM, and CPM and induced the co-inhibitor PD-L2 in SPM and CPM. Therefore, local macrophages acquire M2-like phenotypes with probable suppressive capacities. Regarding T cells, infection induced an increase in the percentage of CD4+ cells that are PD-1+, which represent a potential target of suppression by PD-L1+/PD-L2+ macrophages. In possible agreement, CD4+ T cells from infected animals showed blunted proliferative responses to in vitro stimulation with anti-CD3. Further evidence of immune suppression in the parasite vicinity arose from the observation of an expansion in FoxP3+ CD4+ regulatory T cells and increases in the local concentrations of the anti-inflammatory cytokines TGF-β and IL-1Ra.</dcterms:abstract>

    <dcterms:abstract>Characterization studies were performed to a proteolytic extract, prepared from latex of Ficus luschnathiana (Miq.) Miq., a Moraceae family tree. The mechanism of action was determined, corresponding to serine proteinases behaviour. Studies of optimum pH and temperature showed a maximum catalytic activity at pH 8.0 and 75°C, respectively. The proteolytic activity exhibited good stability when incubated at different pH and temperature conditions. At 7°C and pH 7.5, the extract kept a 100% of the initial activity for 5 months. When comparing the effect of pH on the stability, maximum values were obtained at pH 6.5, with 98% of the initial activity remaining after 5 h of incubation at 60°C. After 1 min at 100°C total inactivation occurred. Regarding the potential use in the dairy industry, milk clotting activity was determined obtaining a very effective coagulant action. Analysis protocols which included molecular exclusion chromatography, electrophoretic methods and mass spectrometry revealed a complex protein profile. It was found that the molecules responsible for the proteolytic activity of the extract were several proteins of different molecular weight and isoelectric points, most of which still retained the activity in presence of SDS and reducing agents. These proteic components displayed a pH-dependant tendency to form aggregates. Using the proteolytic activity of the crude extract under controlled hydrolysis conditions on whey proteins, yielded peptidic fractions with antimicrobial activity against a S. aureus strain. It was found that fractions of peptide concentration of 0.96 μmol mL-1 inhibited growth by 94%.</dcterms:abstract>

    <dcterms:abstract>In this work we present the synthesis and full structural characterization of hydrothermally synthesized titanate nanotubes (TNTs) for energy applications. It is of interest to characterize the building block of certain nanomaterials and their surface termination. This task is particularly difficult for nanotubes, where the exhibited surface curvature is a limitation for its study. We present the synthesis and thorough structural characterization through several experimental techniques for morphology and compositional characterization. Simulated X-rays powder diffraction patterns (XRPD) of real size nanotubes models are refined with experimental data. The building blocks for the TNT models were obtained from computational simulations by means of Density Functional Theory for H2Ti2O5·0.5H2O, H2Ti2O5·H2O, H2Ti3O7 and H2Ti3O7·0.67H2O. Our procedure confirms that open‐ended TNTs are obtained, with ∼100 nm of length and 6(1) nm of inner diameter. By interpreting the XRPD data, the TNT building block was obtained and found to derive from H2Ti3O7, where the distance between [TiO6] octahedral layers is increased in accordance with H2Ti3O7·0.67H2O. This model fully agrees with all the structural characterizations, validating this methodology and suggesting its potential use to study other nanomaterials.</dcterms:abstract>

    <dcterms:abstract>A southwest Atlantic croaker protease was purified from the pyloric caeca by ammonium sulfate fractionation, acetone precipitation and affinity chromatography. The enzyme was classified as a trypsin on the basis of molecular weight, its ability to hydrolyze synthetic substrates N-α-benzoyl-arginine-p-nitroanilide (BAPA) and tosylarginine methyl ester (TAME), and its inhibition by known trypsin inhibitors. The isolated enzyme has a single band on SDS-PAGE with an estimated molecular mass of 24 kDa. Croaker trypsin activity was stable between pH 5 and pH 11 for 30 min at 0C, 10C and 25C and its maximal activity against BAPA was at pH 9.5. Thermostability was observed to about 55C for 30 min at pH 7.8 and its temperature optimum was 60C. Substrate turnover number was 850 BAPA units per μmol trypsin, and the Kmwas 0.081 mM at 25C. For the hydrolysis of TAME, Vmaxwas 9273 units per μmol trypsin and the Km was 0.155 mM at 25C. The catalytic efficiency Vmax/Kmwas higher than that of trypsin from fish living in colder waters.</dcterms:abstract>

    <dcterms:abstract>Honeybees (Apis mellifera) adjust their time and effort during foraging activity. Their metabolic rates together with body temperature rise while gathering profitable resources. These physiological changes may result in a differential cuticular profile, which in turn may bear communicational value. We evaluated if sucrose concentration of collected food affects the cuticular chemistry of honeybees during foraging. We trained bees to artificial feeders with high (2 M) and low (0.5 M) sucrose concentrations, and captured the active foragers for surface extraction of cuticular compounds. We sampled foragers just after feeding, before taking-off towards the hive, and upon landing at the hive entrance, before entering the hive. Through gas chromatography-mass spectrometry analysis of cuticular extracts, we identified and quantified 48 compounds, including cuticular hydrocarbons (CHCs) and volatiles associated with exocrine glands. We found that higher sucrose concentrations resulted in increased amounts of alkanes and alkenes in the surface extracts of foragers captured at the hive entrance, but not at the feeding site. Our results suggest that the differences that have been reported for CHCs in waggle-dancing honey bees can be already found once they return to the hive from profitable food sources.</dcterms:abstract>
