<dcterms:abstract>In this work we present the preparation and characterization of new fluoroborate glasses doped with Yb3 + and Er3 + and modified by Ca, Sr and Ba oxides. Glasses with compositions (100-x-y) (MO ⋅ 2B2O3) + x NaF + y (YbF3:ErF3) (% in mol), where M is Sr, Ba, Ca, x = 10–30% and y = 0, 1 or 5% were prepared by the melt/quenching technique. Glass forming ability and stability were studied by thermal analysis and it is shown that calcium samples presented the highest stability. The nature of the modifier cation has also shown a direct influence on the absolute and relative intensities of the rare earth ions transitions in the visible and near-infrared regions. Though the calcium samples present lower absorption intensities, they display the best up- and down-conversion efficiencies. There are evidences that the incorporation of the fluoride compound leads to a fluorine rich environment on the first coordination sphere of the RE, decreasing the probability of OH– groups – well known quenchers of RE emissions. Though preliminary, the results in this study can become of relevance for the field of solar cell technology improvement via betterment of solar light harvesting in the infrared and ultraviolet regions.</dcterms:abstract>

In 2011, nitrogen treatment involved both foliar urea sprayings and soil application at two different levels, with two controls - irrigated without nitrogen and no irrigation nor nitrogen. In 2012, the same grapevines received either soil or foliar nitrogen using the same controls. Results showed that foliar application increased the amino acid content to a greater extent than soil application, but that a combination of both was the most effective. For the first time, significantly elevated proline levels in response to drought were demonstrated for the grapevine. Increased contents of aromatic compounds and glycosylated precursors closely mirrored the applied nitrogen dose. Wines produced from N-fertilized Syrah grapes in 2011 showed a statistically significant effect of irrigation and fertilization on positive sensorial perception.</dcterms:abstract>

<dcterms:abstract>Background The horn fly Haematobia irritans is a blood-sucking ectoparasite responsible for substantial economic loss of livestock. Like other hematophagous arthropods species, the successful blood-feeding of H. irritans is highly dependent on the modulation of the host’s hemostasis and immune system. Here, we evaluated the biological activity of hematobin (HTB), a protein recently identified in the H. irritans saliva, on macrophage biology. The goal was to understand the putative interactions between the components of H. irritans saliva and the early host immune responses. Results Thioglycolate-elicited peritoneal macrophages from BALB/c mice were stimulated by lipopolysaccharide (LPS) plus interferon-γ (IFN-γ) in the presence or absence of recombinant HTB. The presence of the salivary protein in the cultures inhibited nitric oxide production and decreased the inducible nitric oxide synthase (iNOS) expression induced by LPS plus IFN-γ. The tumor necrosis factor-α (TNF-α) and interleukin-12p40 (IL-12p40) levels were also reduced in the macrophages pre-incubated with HTB; these findings correlated to the decreased NF-κB expression. The biological activities described here were not associated with changes in annexin V binding to macrophages suggesting that HTB does not induce cell death. In addition, the activity of HTB seems to be specific to macrophages because no changes were observed in lymphocyte proliferation or cytokine production. Conclusions We describe here the first bioactive salivary protein of H. irritans. We characterized its ability to modulate macrophage inflammatory response, and the results can help explain how horn flies modulate the host immune system to feed on blood.</dcterms:abstract>

<dcterms:abstract>Eukaryotic diphthine synthase, Dph5, is a promiscuous methyltransferase that catalyzes an extraordinary N,O-tetramethylation of 2-(3-carboxy-3-aminopropyl)-l-histidine (ACP) to yield diphthine methyl ester (DTM). These are intermediates in the biosynthesis of the post-translationally modified histidine residue diphthamide (DTA), a unique and essential residue part of the eukaryotic elongation factor 2 (eEF2). Herein, the promiscuity of Saccharomyces cerevisiae Dph5 has been studied with in silico approaches, including homology modeling to provide the structure of Dph5, protein–protein docking and molecular dynamics to construct the Dph5-eEF2 complex, and quantum mechanics/molecular mechanics (QM/MM) calculations to outline a plausible mechanism. The calculations show that the methylation of ACP follows a typical SN2 mechanism, initiating with a complete methylation (trimethylation) at the N-position, followed by the single O-methylation. For each of the three N-methylation reactions, our calculations support a stepwise mechanism, which first involve proton transfer through a bridging water to a conserved aspartate residue D165, followed by a methyl transfer. Once fully methylated, the trimethyl amino group forms a weak electrostatic interaction with D165, which allows the carboxylate group of diphthine to attain the right orientation for the final methylation step to be accomplished.</dcterms:abstract>

<dcterms:abstract>Background Uruguay, a South American country of 3.4 million inhabitants that has already banned tobacco advertising, prohibited such terms as light, mild and low-tar and required graphic warnings covering 80% of cigarette packs, is considering the imposition of plain, standardised packaging.

Methods We conducted an experimental choice-based conjoint analysis of the impact of alternative cigarette package designs on the risk perceptions of 180 adult current Uruguayan smokers. We compared plain packaging, with a standardised brand description and the dark brown background colour required on Australian cigarette packages, to two controls: the current package design with distinctive brand elements and colours; and a modified package design, with distinctive brand elements and the dark brown background colour. Graphic warnings were also varied.

Results Plain packaging significantly reduced the probability of perceiving the stimulus cigarettes as less harmful in comparison to the current package design (OR 0.398, 95% CI 0.333 to 0.476, p<0.001) and the modified package design (OR 0.729, 95% CI 0.626 to 0.849, p<0.001).

Conclusions Plain packaging enhanced the perceived risk of cigarette products even in a highly regulated setting such as Uruguay. Both the elimination of distinctive brand elements and the use of Australia’s dark brown background colour contributed to the observed effect.</dcterms:abstract>

Among them three parasitic diseases caused by genetically related protozoa belonging to the trypanosomatid genus and kinetoplastida order are major health concerns in the developing world: American Trypanosomiasis (Chagas disease), Human African Trypanosomiasis and Leishmaniasis. This review focuses on efforts carried out to develop bioorganometallic compounds as prospective agents for the treatment of these diseases. It is divided in sections devoted to efforts grouped by type of organometallic core. Those series of compounds for which efforts have been done to modify structures through rational design, to get at least qualitative structure–activity relationships and to identify molecular targets and mode of action will be preferentially highlighted. Comments on potentialities, perspectives and tasks of bioorganometallic chemistry for the field of development of drugs against trypanosomatid parasites are also included.</dcterms:abstract>

<dcterms:abstract>The bronze bug, Thaumastocoris peregrinus Carpintero & Dellape (Heteroptera: Thaumastocoridae), is an exotic emerging pest in Eucalyptus commercial forests in South America, Africa and southern Europe. Information on the chemical communication system and reproductive ecology of this insect is scant, and it may be relevant for designing management strategies for eucalypt plantations. Adults and nymphs usually aggregate in the field, possibly by means of chemical signals. Males emit large amounts of 3-methyl-2-butenyl butyrate, which attracts conspecific adult males but not females. The ecological role of this putative male aggregation pheromone remains unknown. Here, we report olfactometer bioassays showing that late-instarmale nymphs are also attracted to synthetic 3-methyl-2-butenyl butyrate and to adult male volatile extracts, which contain this compound as the major component. As previously shown for adult females, nymphs that moulted into females were not attracted to either volatile stimulus. The intra-gender attraction of nymphs and adults may be related to the exploitation of food resources, or as a reproductive strategy for newly emerged males. Further studies on the reproductive behaviour and mating system of T. peregrinus will contribute to understanding the ecological significance of male-male, adultnymph attraction, as well as the practical applications that may result from these findings.</dcterms:abstract>

<dcterms:abstract>Many metabolites naturally occur as glycosides, since sugar moieties can be crucial for their biological activity and increase their water solubility. In the plant kingdom they may occur as glycosides or sugar esters, depending on precursor chemical structure, and in wine they have traditionally attracted attention due to their organoleptic properties, such as astringency and bitterness, and because they affect the colour and aroma of wines. A new approach directed at detailed description of glycosides in a large selection of monovarietal wines (8 samples each of Pinot Blanc, Muller Thurgau, Riesling, Traminer, Merlot, Pinot Noir and Cabernet Sauvignon) was developed by combining high performance liquid chromatography with high resolution tandem mass spectrometry. Analytical separation was performed on an Accucore™ Polar Premium LC column, while mass analysis was performed in negative ion mode with an non-targeted screening approach, using a Full MS/AIF/NL dd-MS2 experiment at a resolving power of 140,000 FWHM. Over 280 glycoside-like compounds were detected, of which 133 (including low-molecular weight phenols, flavonoids and monoterpenols) were tentatively identified in the form of pentose (6), deoxyhexose (17), hexose (73), hexose-pentose (16), hexose-deoxyhexose (7), dihexose (5) and hexose ester (9) derivatives. It was not possible to univocally define the corresponding chemical structure for the remaining 149 glycosides. Non-parametric statistical analysis showed it was possible to well characterise the glycosylated profile of all red and Traminer wines, while the identified glycosides were almost entirely lacking in Pinot Blanc, Riesling and Muller Thurgau wines. Also Tukey’s Honestly Significant Difference test (p < 0.05) and Principal Component Analysis confirmed that it was possible to almost entirely distinguish the selected red wines from each other according to their glycosylated profile.</dcterms:abstract>

<dcterms:abstract>In social insects, task-related recognition plays an important role in the coordination and cohesion between members of the colony. Tetragonisca angustula is an eusocial stingless bee that presents a sophisticated system of defense involving two complementary groups of guards: hovering and standing guards. We identified, quantified, and compared the cuticular compounds of worker bees captured within the nest, and bees performing tasks outside: foragers and guards. In addition to cuticular hydrocarbons (CHCs), we identified abundant di- and triterpenes. Among the CHCs, we found a mixture of n-alkanes, methyl-branched alkanes, alkenes, and alkadienes. Significant differences in the relative abundance of CHCs between behavioral groups were found. Particularly, guards present high amounts of branched alkanes relative to nest bees and foragers. Differential CHC profiles associated with behavioral groups could imply a mechanism for caste recognition.</dcterms:abstract>

<dcterms:abstract>Cross-cultural research is becoming increasingly relevant in sensory and consumer science. The design of cross-cultural studies involves several methodological challenges that are not commonly faced in studies involving a single culture. However, several of these challenges have not yet received enough attention in the field, which poses several limitations to the validity and generalizability of the findings of cross-cultural studies. In this context, the aim of the present work is to review some of the most relevant methodological issues that should be considered when designing cross-cultural studies. In particular, five topics are addressed: sampling procedures, conceptual equivalence, linguistic equivalence, data collection procedures and cultural differences in response style. Suggestions and recommendations are discussed for each of the topics, which are expected to encourage greater methodological rigor and contribute to more theoretical robust cross-cultural studies in the field.</dcterms:abstract>

<dcterms:abstract>A novel approach for the analysis of phenolics in tea was developed using multiplexed simultaneous detection. This is the first study employing a short reaction flow (RF) chromatography column (length 50 mm) for the analysis of phenolics in complex samples (not a standard mixture of compounds). RF separation efficiency was high as the detectors are multiplexed simultaneously, rather than sequentially. The delay time between RF detection responses is minor (1.2 s) providing simple peak assignment between the phenolic detection and conventional detectors, such as the UV and fluorescence detectors (FLD). Thirty-nine phenolics were detected amongst the five different tea samples (three from Camellia sinensis and two herbal teas). The highest number of phenols detected was 24 for English Breakfast tea (four highly abundant), 19 for Earl Grey (six highly abundant), 13 in Pure Green (two highly abundant) and 9 in both Fruit of the Forest (two highly abundant) and Camomile (none highly abundant) herbal teas. Numerous low concentration phenolic compounds in the Fruit of the Forest tea were not included due to their abundance being only slightly above the noise level of the detection process. The relative response factor (RRF) analysed at 520 and 500 nm aided to identify unique phenolics to each sample with similar retention times; three phenolics were unique to the Pure Green tea (2.30, 4.11 and 5.80 min) and two to the Earl Grey Tea (4.11 and 5.80 min). The Fruit of the Forest tea and the Camomile tea were both distinctly different from all other teas. This approach may be used for phenolic profiling of food, beverages or natural products.</dcterms:abstract>

<dcterms:abstract>The study of two commonly used post-column derivatisation (PCD) approaches for the selective detection of “bioactive” compounds in natural products were investigated – the ‘phenolic’ assay as the name suggests was responsive to phenolic compounds, while the Ferric Reducing Antioxidant Power (FRAP) assay was responsive to antioxidants. Detailed assessment of the peaks (based largely on retention time) that were observed in both detection modes for five beverage samples (four teas and one coffee) revealed the presence of a total of 56 compounds. Eight compounds were not detected by the FRAP assay and 13 were not observed by the phenolic assay. A geometric approach to factor analysis (GAFA) was applied to the detection sensitivity, and this analysis showed the correlation between these two assays was 0.67 and the spreading angle between the detection vectors was 47°. Such outcomes are usually indicative of a multidimensional separation approach, but in this study this was achieved only from the detection protocol. Hence, when employing PCD to determine the ‘activity’ of samples derived from natural origin, testing should be conducted using multiple selective detection processes.</dcterms:abstract>

<dcterms:title>Narrow bore post column derivatisation assays via reaction flow chromatography</dcterms:title>
<dcterms:abstract>This study highlights the capability to conduct a post column derivatisation (PCD) process with a narrow bore, low volume column. A reaction flow (RF) HPLC column (30 mm × 2.1 mm internal diameter (i.d.), 3 μm particle diameter) enabled the selective detection of phenols for both a simple standard mixture and a complex sample represented by green tea. The resolving power and sensitivity obtained were similar to an analytical scale (4.6 mm i.d.) RF HPLC column of the same format. However, the baseline noise experienced was higher when the assay was scaled down from the RF 4.6 mm i.d. to the RF 2.1 mm i.d. narrow bore column. The PCD mixing and reaction speed inside the RF column outlet (frit and head-fitting design) enabled the coupling of PCD protocols with “UHPLC-type” low volume columns and systems without significant loss in separation performance. In comparison to a conventional analytical scale PCD approach the scaled down narrow bore RF approach demonstrated 40 times greater sensitivity in terms of S/N. Scaled down PCD is vital for decreased consumption and cost without significant loss in separation performance.</dcterms:abstract>

UV-C treated juice resulted in 2.33 log and 2 log reductions in aciduric microorganisms and yeast and mould count, respectively. After UV-C treatment, there were no significant changes (p > 0.05) in pH, acidity and soluble solids; however antioxidant activity and the ascorbic acid content were affected by this process. The losses in ascorbic acid were 27.5 and 42% after UV-C treatment necessary to decrease 95 and 99% of the microbial load, respectively. Based on sensory analysis results, no significant differences were detected between fresh and UV-C treated juices.
