<dcterms:abstract>Recently developed semiempirical methods (AM1, MNDO-PM3 and MNDO/M) are tested by comparison with ab initio Hartree-Fock and Møller-Plesset 4-31G and 6-311G (d,p) calculations on cuts of the potential energy hyperfaces of the hydrogen bonded dimers H2O·H2O, NH3·NH3 and NH3·H2O. AM1 gives a bifurcatedly hydrogen-bonded complex for the water dimer, as already published, whereas it gives trifurcated structures for the other two dimers. There is no indication of a quasi-linear hydrogen bonded dimer in these last two cases, in agreement with the ab initio calculations. Moreover, there is a secondary minimum in the ammonia dinner surface, with a cyclic structure, which closely resembles that obtained in the ab initio calculations. MNDO-PM3 gives the quasi-linear hydrogen bonded structure of the water dimer as the correct one. For NH3·NH3 the global minimum is no longer the trifurcated structure (which is, however, a secondary minimum) but a cyclic one. However, the ring is so closed that hydrogen atoms of both monomers are almost head-to-head. For NH3·H2O a further minimum appears, which is now the global one, corresponding to the linearly hydrogen-bonded structure of the dimer. The aspect of this attractor, as well as the aspect of the cut in the PES of the water dimer, suggests that undue importance has been given to linear hydrogen bonds in the parametrization of MNDO-PM3. MNDO/M behaves in a very similar way to MNDO-PM3 and, owing to its better definition, the latter should be preferred. In conclusion, none of these methods is truly reliable for calculations on weakly bound dimers. MNDO-PM3 is better than AMI for the water and the ammonia-water dimers but not for the ammonia dimer; MNDO/M is no better than MNDO-PM3.</dcterms:abstract>

    <dcterms:abstract>Traumatic brain injury (TBI) remains as one of the leading causes of mortality and morbidity worldwide and there are no effective treatments currently available. Gene therapy applications have emerged as important alternatives for the treatment of diverse nervous system injuries. New strategies are evolving with the notion that each particular pathological condition may require a specific vector. Moreover, the lack of detailed comparative studies between different vectors under similar conditions hampers the selection of an ideal vector for a given pathological condition. The potential use of lentiviral vectors versus several modular protein-based nanovectors was compared using a controlled cortical impact model of TBI under the same gene therapy conditions. We show that variables such as protein/DNA ratio, incubation volume, and presence of serum or chloroquine in the transfection medium impact on both nanovector formation and transfection efficiency in vitro. While lentiviral vectors showed GFP protein 1 day after TBI and increased expression at 14 days, nanovectors showed stable and lower GFP transgene expression from 1 to 14 days. No toxicity after TBI by any of the vectors was observed as determined by resulting levels of IL-1β or using neurological sticky tape test. <dcterms:abstract>The serodiagnosis of hydatid disease is a valuable instrument for clinical diagnosis and epidemiological surveillance of high-risk populations. In the past decade a wealth of reports on the diagnostic performance of numerous antigens have been produced. However, their diagnostic value has been estimated under different conditions, using different serum collection, therefore precluding their direct comparison. Here we report an unbiased comparison of the same batch of six major E. granulosus antigens, namely, hydatid cyst fluid (HCF), native antigen B (AgB), two recombinant AgB subunits, an AgB-derived synthetic peptide, and recombinant cytosolic malate dehydrogenase from E. granulosus (EgMDH), against the same serum collection. The doubleblind analysis was performed using a standardized protocol and receiver operating characteristic (ROC) data analysis by a network of six South American laboratories. High intercenter reproducibility was attained, and the intralaboratory analysis allowed the comparative ranking of the antigen panel. HCF, AgB, and its AgB8/1 subunit exhibited equivalent diagnostic efficiencies, 81.4% 0.5%, 81.3% 0.6%, and 81.9% 2.0%, respectively; with a more favorable balance toward specificity in the case of the last antigen. The diagnostic efficiencies for the other three antigens were 76.8% 6.8%, 69.1% 2.7%, and 66.8% 2.1%, for the peptide, the AgB8/2 subunit, and the EgMDH, respectively. The study also included an analysis of batch-to-batch variation in the diagnostic performance of different HCF regional preparations. Based on these results, a suggested recommendation on the use of these antigens was drawn.</dcterms:abstract>

    <dcterms:abstract>Two different Echinococcus granulosus antigen B subunits (AgB8:1 and AgB8:2) were characterized and the structure of the genes encoding these two proteins were compared. DNA sequences were expressed in Escherichia coli and the antigens’ diagnostic value was then assessed. The genomic sequence of AgB8:1 has a 92 bp intron in the position corresponding to amino acid 16; the AgB8:2 genomic sequence presents a 68 bp intron in the position corresponding to amino acid 20. Both introns are located between the putative N-terminal hydrophobic sequence and the secreted peptide. A comparison between the AgB8:1 and AgB8:2 nucleotide sequences showed a 53.5% identity among exons and a 50% identity between introns. According to the molecular diversity analysis, the elapsed time since both genes shared a common ancestor would be around 4.2107 years. When the native AgB and the two recombinant antigens (rAgB8:1 and rAgB8:2) were tested in an anti-IgG ELISA, the sensitivity of the native antigen B was 77.41% and its specificity was 81.9%, while rAgB8:1 showed 54.84% of sensitivity and 80.17% of specificity and rAg138:2 had an 83.87% sensitivity and a 98.28% specificity. Statistical analysis confirms that rAgB8:2 has a better performance than rAgB8:1 and native AgB in ELISA</dcterms:abstract>

    <dcterms:abstract>Benzimidazole (BZD) anthelmintic compounds are widely used in antiparasite therapy in both veterinary and human medicine. The poor water solubility of BZD limits their development under alternative pharmaceutical formulations. Pharmaceutical formulations of BZD molecules, which enhance their bioavailability or extend their residence times in monogastrics, could improve their efficacy in these species. Gelucire lipid matrix was developed and studied for use in human medicine, particularly Gelucire 44/14 which is composed of a well-defined mixture of mono-, di- and triglycerides and mono- and difatty acid esters of polyethyleneglycol. They are characterized either by their melting temperature (44°C) or their hydrophiliclipophilic balance (HLB) values (14 in this case) (Aungst et al., 1997). Gelucire increases the solubility of poorly hydrosoluble drugs and produces stable slim dispersions on the gastrointestinal walls, giving enhanced drug absorption (Esquizabel et al., 1996; Aïnaoui et al., 1997; Aungst et al., 1997). <dcterms:abstract>A pharmaco-parasitological assessment of four different albendazole (ABZ) formulations was carried out in lambs infected with multiple resistant gastrointestinal (GI) nematodes. The comparative drug systemic exposure profiles (ABZ sulphoxide plasma concentrations) and anthelmintic efficacies (clinical endpoint measured through the faecal nematode eggs reduction counts) were determined for a reference formulation (RF) and three different test (T1, T2, T3) generic ABZ preparations. Fifty (50) Corriedale lambs naturally infected with multiple resistant GI nematodes were allocated into five experimental groups (n = 10). Animals in each group received treatment with either the RF, one of the test ABZ formulations (5 mg⁄ kg by the intraruminal route) or were kept as untreated control. Blood samples were collected over 48 h post-treatment. ABZ parent drug was not recovered in the bloodstream. The ABZ sulphoxide (ABZSO) and sulphone (ABZSO2) metabolites were measured in plasma by ultraviolet high-performance liquid chromatography over 36–48 h post-treatment. A faecal nematode egg count reduction test (FECRT) was performed at day 10th post-treatment to lambs from all treated and untreated groups, which indicated the predominance of nematodes with high level of resistance to ABZ. Both ABZSO Cmax and AUC0–LOQ values obtained for the RF (pioneer product) were significantly higher (P < 0.05) than those obtained for the T1 and T3 preparations. Based on the currently available bioequivalence criteria, the test (generic) ABZ formulations under evaluation could not be considered equivalent to the RF regarding the rate (Cmax) and extent (AUC0–LOD) of drug absorption (indirectly estimated through the ABZSO metabolite). A large variation in nematode egg counts did not permit to obtain statistically significant differences among formulations. However, a favourable trend in the efficacy against the most resistant nematodes was observed for the formulations with the highest ABZSO systemic exposure.</dcterms:abstract>

    <dcterms:abstract>There is an urgent need to develop new drugs against Chagas’ disease. In addition, the mechanisms of action of existing drugs have not been completely worked out at the molecular level. High throughput approaches have been demonstrated to be powerful tools not only for understanding the basic biology of Trypanosoma cruzi, but also for the identification of drug targets such as proteins or pathways that are essential for parasite infection and survival within the mammalian host. Here, we have applied these tools towards the discovery of the effects of two organometallic compounds with trypanocidal activity, Pd–dppf–mpo and Pt–dppf–mpo, on the transcriptome and proteome of T. cruzi epimastigotes. These approaches have not yet been reported for any other prospective metal-based anti T. cruzi drug. We found differentially expressed transcripts and proteins in treated parasites. Pd–dppf–mpo treatment resulted in more modulated transcripts (2327 of 10 785 identified transcripts) than Pt–dppf–mpo treatment (201 of 10 773 identified transcripts) suggesting a mechanism of action for Pd–dppf–mpo at the transcriptome level. Similar numbers of differentially expressed proteins (342 and 411 for Pd–dppf–mpo and Pt–dppf–mpo respectively) were also observed. We further functionally categorized differentially expressed transcripts and identified cellular processes and pathways significantly impacted by treatment with the compounds. Transcripts involved in DNA binding, protein metabolism, transmembrane transport, oxidative defense, and the ergosterol pathways were found to be modulated by the presence of the compounds. Our transcriptomic dataset also contained previously validated essential genes. These data allowed us to hypothesize a multimodal mechanism of action for the trypanocidal activity of Pd–dppf–mpo and Pt–dppf–mpo, and a differential contribution of the metal moiety of each compound.

The CoMFA model generated in the study has been used to estimate the namatocide activity (MIC50) of seven 6,7-diarylpteridines related to those previously studied. The model is highly predictive for all pteridine derivatives in the test set. Moreover, this model also predicts satisfactorily the nematocide activity of other [6] + [6] fused pyrazines (quinoxalines and pyridopyrazines) which were not represented in the training set. The analysis revealed the importance of steric factors (64.8%) and then the electrostatic ones (35.8%). Most pteridines under study were previously synthesized and tested as nematocide agents. <dcterms:abstract>Different interpretative front-of-pack (FOP) nutrition labelling schemes have recently been implemented in several countries but it is still unclear which is the most effective. The present work compared three interpretative schemes (Nutri-score, health star rating and nutritional warnings) in terms of attentional capture, processing time, influence on perceived healthfulness and purchase intention of products with different nutritional profile. Two studies were conducted. In the first study, attention to and processing time for interpretation of FOP labels was evaluated using a visual search task with 112 participants. In the second study, an online survey with 892 participants was conducted to evaluate the influence of interpretive FOP labels on purchase intention and perceived healthfulness of a series of products. A between-subjects design was implemented to compare a control condition (without front-of-pack nutrition information) and the three interpretive FOP schemes. The health star rating was found to perform worse than the other two schemes in terms of capturing attention and altering perceived healthfulness and purchase intention. The latter effect depended on the degree of healthfulness of the food products in question, but the effect on consumer behaviour towards unhealthful product categories was more pronounced for the warning label scheme. From a nutrition policy effectiveness point of view, results suggest that nutritional warnings may have advantages over Nutri-score and the health star rating in the context of the current food environment, characterized by the wide availability of products with high content of nutrients associated with non-communicable diseases.</dcterms:abstract>

The reactivity of the nitro-anion radical for these derivatives with glutathione, a biological relevant thiol, was also studied by cyclic voltammetry. <dcterms:abstract>
In a search for new antitumoral agents, a series of homoleptic copper(II) complexes with amino acids and dipeptides, as well as heteroleptic complexes containing both dipeptides and
1,10-phenanthroline, were studied. Furthermore, a single-crystal structure containing alanyl-leucinato ([Cu3(AlaLeu)3 (H2O)3(CO3)]PF6H2O), which is the first homotrinuclear carbonato-bridged copper(II) complex with a dipeptide moiety, is presented. To assess possible antitumor action mechanisms,
we focused on the comparative analysis of pro- and antioxidant behaviors. Pro-oxidant activity, in which the reactive oxygen species (ROS) formed by the reaction of the complexes with H2O2 produce oxidative damage to 2-deoxy-D-ribose, was evaluated using the TBARS method. Additionally, the antioxidant action was quantified through the superoxide dismutase (SOD)-like activity, using a protocol based on the inhibitory effect of SOD on the reduction of nitrobluetetrazolium (NBT) by the superoxide anion generated by the xanthine/ xanthine oxidase system. Our findings show that Cu–amino acid complexes are strong ROS producers and moderate SOD mimics. Conversely, Cu–dipeptide–phen complexes are good SOD mimics but poor ROS producers. The activity of Cu–dipeptide complexes was strongly dependent on the dipeptide. A DFT computational analysis revealed that complexes with high SOD-like activity tend to display a large dipole moment and condensed-to-copper charge, softness and LUMO contribution. Moreover, good ROS producers have higher global hardness and copper electrophilicity, lower copper softness and flexible and freely accessible coordination polyhedra.
</dcterms:abstract>

    <dcterms:abstract>Lead does not fulfill any physiological function in the human body. It is generally accepted that the blood lead level (BLL) is the best exposure index, as there is an excellent correlation between the actual exposure of the individual and the concentration of lead in blood. In Uruguay, lead contamination becomes a matter of public concern in 2001, giving rise to a sensitization in the population, which in many cases brought about a change in hygienic and dietary habits of the children. In 2004, after the leaded gasoline phasing out process in Uruguay was completed, we studied non-exposed children to correlate BLL with variables such as age, sex, area of residence, and available environmental lead data and compared these results with those from our similar screening studies 10years ago. The main result of this comparison is that BLL from children in our country had a significant decrease between 1994 and 2004.</dcterms:abstract>

    <dcterms:abstract>We have applied dispersion corrected density functional theory to gauge the reactivity of the most common defects found in graphene. Specifically, we investigated single vacancies, 585 double vacancies, 555−777 reconstructed double vacancies, Stone− Wales defects, and hydrogenated zigzag and armchair edges. We found that the extent to which defects increase reactivity is strongly dependent on the (a) functional group to be attached and (b) number of functional groups attached. For the addition of one H, F, and phenyl groups to defective graphene, we found the following decreasing order of reactivity: single vacancy > hydrogenated zigzag edge > 585 double vacancy > 555−777 reconstructed double vacancy > Stone−Wales > hydrogenated armchair edge > perfect graphene. However, when two phenyl groups are attached, the Stone−Wales defect becomes more reactive than the 585 double vacancy and 555−777 reconstructed double vacancy. The largest increase of reactivity is observed for the functional groups whose binding energy onto perfect graphene is small. In contrast with recent experimental results, we determined that the reactivity of edges in comparison with perfect graphene is much higher than the reported value. When two groups are attached onto a 585, 555−777, or Stones−Wales defect, they prefer to be paired on the same CC bond on opposite sides of the sheet. However, for the single vacancy, this is not the observed behavior as the preferred addition sites are those carbon atoms that were previously bonded to the missing carbon.</dcterms:abstract>

    <dcterms:abstract>The catalytic performance of trimetallic oxides MeLaZr (Me: Ni, Co) was evaluated in the steam reforming of ethanol for hydrogen production. Rh doped and non doped catalysts were prepared by the pseudo sol–gel like method, characterized by X-ray diffraction (XRD), thermo-programmed reduction (TPR), micro-homogeneity analysis (TEM-EDXS), specific surface areas (BET). A phase-segregation process takes place during the calcination for both trimetallic systems leading to the formation of La2Zr2O7 pyrochloretype compound in coexistence with the Me-oxide. NiLaZr and CoLaZr catalysts show high activity and high selectivity for hydrogen production. Addition of Rh promotes reducibility at lower temperatures for both trimetallic catalysts and remarkably improves the activity and selectivity for H2. The H2 selectivity is lowered by CO2 injection because of the increase of the rate of the reverse WGSR and/or methanation reaction. The CH4 selectivity is lowered by the dry reforming reaction. The key-role played by oxycarbonate species formed during the reforming tests leads to a highly efficient gasification of carbon deposits by the Boudouard reaction.</dcterms:abstract>

    <dcterms:abstract>Consumption of natural orange juice is considered a healthy habit. Despite the wide offer of options such as fruit, vegetable or healthy smoothies to start the day, orange juice continues to be one of the favourite drinks to “start with energy”. Currently, a concern among general population, regarding nutritional importance and knowledge about how the foods consumed influence health is growing. Uruguay is the second largest exporter in Latin America and one of the twenty largest citrus exporters in the world, thus, analytical methods to monitor their nutritional value and quality are always being revised and modernized. This study focused on examining the correlation between different parameters assessed using standard methods and determined through near infrared spectroscopy (NIR) for Citrus sinensis Valencia orange juice. Besides, several parameters to characterize the nutritional value of this juice were determined and reported. The findings showed a novel correlation between NIR spectroscopy data and results obtained for calcium (Slope > 0.94 and Residual Predicted Deviation > 3) compared with standard methods which are discussed. Notably, the outcomes highlight NIR spectroscopy as a valuable approach for streamlining analyses, in this case calcium determination, significantly reducing the required resources-such as time and costs of parameters pivotal for assessing juice quality. The results indicate that the orange juice studied showed good nutritional quality parameters, besides calcium content can also be evaluated by NIR, being this technique in more agreement with the principles of Green Analytical Chemistry.</dcterms:abstract>

    <dcterms:abstract>Herein, we performed a theoretical investigation of the reactivities of graphene and 2D-BN. The results indicate that 2D-BN can display chemistry comparable to that of graphene. As observed experimentally, we found that the addition of carbenes, nitrenes and amines is possible. In addition, [2+2] cycloadditions are more likely to occur on 2D-BN rather than on graphene, while the 1,3 cycloaddition of azomethine ylides is also possible. The nitrogen atoms tend to be less reactive than the boron ones. Nevertheless, this situation is modified once a boron atom is functionalized. Indeed, if two fluorine atoms are added to a BN sheet, they prefer to be attached on a BN bond, in place of two boron atoms. This result opens the possibility to synthesize fully hydrogenated, fluorinated or hydroxylated 2D-BN. Although great efforts have been made to explore to chemistry of 2D-BN, there is much more room to expand its chemistry.</dcterms:abstract>

The most promising compound (an iminoquinone derivative) has undergone National Cancer Institute (NCI) 60 cell line (single and five dose) screening, and using the NCI COMPARE program, has shown correlation to NQO1 activity and to other NQO1 substrates. Common structural features suggest that the iminoquinone moiety is significant with regard to NQO1 specificity. Computational docking into the active site of NQO1 was performed, and the first comprehensive mitomycin C (MMC)-NQO1 docking study is presented. Small distances for hydride reduction and high binding affinities are characteristic of MMC and of iminoquinones showing correlations with NQO1 via COMPARE analysis. Docking also indicated that the presence of a substituent capable of hydrogen bonding to the His194 residue is important in influencing the orientation of the substrate in the NQO1 active site, leading to more efficient reduction.
</dcterms:abstract>

Within this context, the aim of the present study was to analyze how two age cohorts of users perceive products of different familiarity grade (mobile phones and watches), using check-all-that-apply (CATA) questions and preference mapping, techniques not commonly used in the industrial design field. Two studies were carried out in which young and older users assessed a series of product images by answering a CATA question and rating their overall liking using a 9-point hedonic scale. Results demonstrated how certain terms apply only to a specific age group, how these correlated to particular design features and how these data constructed different preference patterns. <dcterms:abstract>Microwave Assisted Extraction and a modified CEN-QuEChERS methodology were evaluated as extraction and clean up procedures for the simultaneous analysis of 42 organophosphate pesticides in yerba mate (Ilex paraguaiensis). The obtained extracts were analyzed by gas chromatography using a flame photometric detector. Linearity, recovery percentages, relative standard deviations, detection and quantification limits and matrix effects were determined according to DG-SANCO guidelines for both methods. At 0.2 and 0.5 mg/kg the evaluated methods showed percentages recoveries between 70 and 120% for most of the analytes. Using Microwave Assisted Extraction methodology, 33 pesticide residues could be properly analyzed whereas only 27 could be determined with the proposed modified QuEChERS. All relative standard deviation were below 18% except for omethoate and disulfoton sulfone when evaluated by the modified QuEChERS. The limits of detection in both methodologies were 0.2 mg/kg for most of the analyzed compounds. The average detection limit for QuEChERS was 0.04 mg/kg. For 19 of the analytes determined through Microwave Assisted Extraction the lowest validated level were 0.004 mg/kg. Signal suppression/enhancement was observed for most of the pesticides, thus matrix-matched calibration curves were used for quantification. The Microwave Assisted Extraction and QuEChERS procedures studied could detect the organophosphate pesticides above the MRL fixed for “mate” by the European Union. They have been successfully applied for the determination of organophosphate pesticide residues in commercial samples and the positives were confirmed through GC–(ITD)-MS.</dcterms:abstract>
