<dcterms:abstract>Polynuclear complexes are widely regarded as potential materials in separation, catalysis, gas storage, and molecular recognition. Those complexes including lanthanide (Ln) ions in the heterobimetallic assembly enhance the structural flexibility and increase the thermodynamic stability. Up to now, the studies on these heteropolynuclear complexes have been mostly focused on lanthanide-copper compounds in the solid state. We have expanded the studies to solution chemistry, to know if such polynuclear species can also exist in aqueous solution. In this work, potentiometric titrations (25.0 °C, and I = 0.50 M Me4NCl) were carried out for systems containing a Ln ion (La, Ce, Sm), a bridging ligand (oda or ida), and a second metal ion. We found that polynuclear units can be formed with MII (Mn, Fe, Co, Ni, Cu, Zn, Cd, and VO2+) and MIII (Fe, Al) ions, and can be predominant in aqueous solution. Species like [Ln2M3L6] or [Ln2M3L6(OH)n]n- (n = 3, 6) can exist in solution, representing the same 2:3:6 stoichiometry found in the solid state. The presence of an amino group in the ida ligand makes the mononuclear complexes with 3d metal ions more stable, preventing to some extent the formation of polynuclear complexes.</dcterms:abstract>

<dcterms:abstract>Uncontrolled combustion due to garbage recycling is a widespread activity among slum dwellers in distressed economy countries and has been indicated as a major source of dioxin contamination. However, because of the high cost and complexity of gas chromatography/high-resolution mass spectrometry (GC-HRMS) analysis, the magnitude of the problem remains largely unknown. The present study describes a first approach toward the use of a dioxin antibody-based enzyme-linked immunosorbent assay (ELISA) as the basis for a sustainable, simple, and low-cost monitoring program to assess the toxicological impact of uncontrolled combustion in slums. A panel of 16 samples was analyzed by GC-HRMS and ELISA on split extracts. Close to 20% of the analyzed samples showed dioxin concentrations up to almost twice the guidance level for residential soil in several countries, pointing out the need for performing a large-scale monitoring program. Despite the potential for variations in dioxin congener distribution due to the mixed nature of the incinerated material, there was a good correlation between the toxic equivalents as determined by GC-HRMS and ELISA. Furthermore, an interlaboratory ELISA validation showed that the capacity to perform the dioxin ELISA was successfully transferred between laboratories. It was concluded that the ELISA method performed very well as a screening tool to prioritize samples for instrumental analysis, which allows cutting down costs significantly.</dcterms:abstract>

The hypoxic selective cytotoxicity towards V79 cells and the superoxide dismutase-like activity of the complexes were determined and related to physicochemical properties of the compounds. In particular, the copper(II) complex with 3-amino-6-chloro-7-fluoroquinoxaline-2-carbonitrile N1,N4-dioxide showed cytotoxic selectivity in hypoxia being the most lipophilic compound of the series. On the contrary, the complex with 3-aminoquinoxaline-2-carbonitrile N1,N4-dioxide was cytotoxic but not selective and that with 3-amino-7-chloro-6-methoxy-quinoxaline-2-carbonitrile N1,N4-dioxide was not cytotoxic towards V79 cells neither in oxia nor in hypoxia in the assayed conditions. The σm Hammett substituent electronic descriptor was related to the effect in hypoxic conditions and the SOD-like activity was correlated to the effect in normoxia.</dcterms:abstract>

The katG gene of 62 INH-resistant strains was analysed by an RFLP-PCR assay. PCR products were digested with MspI to detect Ser315Thr and Arg463Leu substitutions. A total of 16 of the 62 (26 %) INH-resistant strains analysed had a Ser315Thr substitution. Only one INH-resistant strain had an Arg463Leu substitution and two strains had a deletion in katG. Of the 16 strains with Ser315Thr, 15 showed different profiles using a double-repetitive-element PCR assay, demonstrating that there was no local dissemination of any particular strain. These findings are in agreement with published data from regions where the prevalence of tuberculosis (TB) is intermediate and may be due in part to the success of the local TB control programme.</dcterms:abstract>

<dcterms:abstract>Background: The sudden changes (increase in capillar permeability, edema formation, vasodilation and hypotension) observed in septic patients and the measurements taken in order to revert this situation make vancomycin concentrations difficult to interpret. Therapeutic drug monitoring (TDM) of vancomycin is routinely performed at steady state, target concentrations are peaks between 20 and 40 mg/l and troughs of 5 – 10 mg/l. Lately, continuous infusion of vancomycin (CIV) has been used as an alternative mode of administration mainly in critically ill patients with sepsis or septic shock. Despite this novel mode of administration, the need of drug monitoring in this population is under discussion. Objective: The aim of our study was to test the usefulness of a multi-compartment model in order to understand the rapid changes that occur in critically ill patients. Materials and methods: A prospective, cohort study was carried out in the intensive care unit of the University Hospital. 25 intensive care unit adults patients with severe sepsis or septic shock receiving vancomycin in CIV modality for documented gram-positive infections were included in the study. Once the infusion was started, blood samples were drawn periodically and analized by fluorescence polarisation immunoassay (FPIA, TDx, Abbott Laboratories, Chicago, IL, USA). A multi-compartment model was used to predict vancomycin level evolution throughout the treatment of patients with sepsis. Results: High doses of vancomycin were administered in order to rescue patients from septic shock. Plasma drug concentration dropped while clinical condition of patients worsened. Conversely, drug levels increased spontaneously once the infection was reverted. The theoretical model provided greater insight into pharmacokinetic features related with the use of vancomycin in septic patients. Conclusions: There was consistency between the model based prediction and the experimental data so dose adjustment was performed in order to reach target concentrations above 20 mg/l and an initial dose of 3 grams of vancomycin per day was recommended to reach these levels.</dcterms:abstract>

The molecule exists as a dimer with each CuII ion coordinated to five ligands, forming a distorted square-based pyramid. The only chemical connection between magnetically non-equivalent copper ions is a very long path comprising 17 diamagnetic atoms and a weak putative hydrogen bond. <dcterms:abstract>In the search for new therapeutic tools against Chagas’ disease (American Trypanosomiasis) two series of new platinum(II) complexes with bioactive 5-nitrofuryl containing thiosemicarbazones as ligands were synthesized, characterized and in vitro evaluated. Most of the complexes showed IC50 values in the μM range against two different strains of Trypanosoma cruzi, causative agent of the disease, being as active as the anti-trypanosomal drug Nifurtimox. In particular, the coordination of L3 (4-ethyl-1-(5-nitrofurfurylidene)thiosemicarbazide) to Pt(II) forming [Pt(L3)2] lead to almost a five-fold activity increase in respect to the free ligand. Trying to get an insight into the trypanocidal mechanism of action of these compounds, DNA and redox metabolism (intra-parasite free radical production) were evaluated as potential parasite targets. Results suggest that the complexes could inhibit parasite growth through a dual mechanism of action involving production of toxic free radicals by bioreduction and DNA interaction.</dcterms:abstract>

<dcterms:abstract>Due to their size, small molecules cannot be simultaneously bound by two antibodies, precluding their detection by noncompetitive two-site immunoassays, which are superior to competitive ones in terms of sensitivity, kinetics, and working range. This has prompted the development of anti-immune complex antibodies, but these are difficult to produce, and often exhibit high crossreactivity with the unliganded primary antibody. This work demonstrates that anti-immune complex antibodies can be substituted by phage particles isolated from phage display peptide libraries. Phages bearing specific small peptide loops allowed to focus the recognition to changes in the binding area of the immune complex. The concept was tested using environmental and drug analytes; with improved sensitivity and ready adaptation into on-site formats. Peptides specific for different immune complexes can be isolated from different peptide libraries in a simple and systematic fashion allowing the rapid development of noncompetitive assays for small molecules</dcterms:abstract>

<dcterms:abstract>In the search for new therapeutic tools against Chagas disease (American trypanosomiasis) palladium and platinum complexes of the bioactive ligand pyridine-2-thiol N-oxide were exhaustively characterized and evaluated in vitro. Both complexes showed high in vitro growth inhibition activity (IC50 values in the nanomolar range) against Trypanosoma cruzi, the causative agent of the disease. They were 39–115 times more active than the antitrypanosomal drug Nifurtimox. The palladium complex showed an approximately threefold enhancement of the activity compared with the parent compound. In addition, owing to their low unspecific cytotoxicity on mammalian cells, the complexes showed a highly selective antiparasite activity. To get an insight into the mechanism of action of these compounds, DNA, redox metabolism (intraparasite free-radical production) and two parasite-specific enzymes absent in the host, namely, trypanothione reductase and NADH-fumarate reductase, were evaluated as potential parasite targets. Additionally, the effect of metal coordination on the free radical scavenger capacity previously reported for the free ligand was studied. All the data strongly suggest that trypanocidal action of the complexes could mainly rely on the inhibition of the parasite-specific enzyme NADH-fumarate reductase.</dcterms:abstract>

The aims of the present work were to study the influence of different package attributes on consumer willingness to purchase regular and functional chocolate milk desserts; and to assess if the influence of these attributes was affected by consumers’ level of involvement with the product. A conjoint analysis task was carried out with 107 regular milk desserts consumers, who were asked to score their willingness to purchase of 16 milk dessert package concepts varying in five features of the package, and to complete a personal involvement inventory questionnaire. Consumers’ level of involvement with the product affected their interest in the evaluated products and their reaction towards the considered conjoint variables, suggesting that it could be a useful segmentation tool during food development. Package colour and the presence of a picture on the label were the variables with the highest relative importance, regardless of consumers’ involvement with the product. The importance of these variables was higher than the type of dessert indicating that packaging may play an important role in consumers’ perception and purchase intention of functional foods.</dcterms:abstract>

Seventy-five consumers participated in the study and were asked to complete a word association task before the evaluation. Then, consumers tried six milk desserts with different polyphenolic concentration, scored their overall liking and willingness to purchase and provided up to four words to describe each of the samples. Cluster analysis performed on consumer-elicited terms in the word association task allowed the identification of three consumer segments with different expectations and motivations toward chocolate milk desserts enriched with antioxidants. These groups also differed in their evaluation of the desserts when tasting them, showing different overall liking, willingness to purchase and sensory description of the samples. This suggests that consumers’ prior expectations and motivations significantly affected their response when tasting the desserts.</dcterms:abstract>

<dcterms:abstract>Many dairy industries have been using SBR wastewater treatment plants because they allow optimal working condition to be reached. However, to take advantage of SBR capabilities, strong process automation is needed. The aim of this work is to study the factors that influence SBR performance to improve modelling and control. To better understand the whole process we studied the kinetic modelling, the carbon removal mechanism and the relation between reactor performance, aerobic heterotrophic activity and bacterial population dynamics (by terminal restriction fragment length polymorphisms of 16S rDNA, T-RFLP). The heterotrophic activity values presented high variability during some periods; however, this was not reflected on the reactor performance. As sludge health indicator, the average activity in a period was better than individual values. Although all the carbon removal mechanisms are still unclear for this process, they seemed to be influenced by non-respirometric ways (storage, biosorption, accumulation, etc.). The variability of heterotrophic activity could be correlated with the bacterial population diversity over time. Despite the high variability of the activity, a simple kinetic model (pseudo ASM1) based on apparent constant parameters was developed and calibrated. Such modellisation provided a good tool for control purposes.</dcterms:abstract>

<dcterms:abstract>B-Galactosidases from Escherichia coli, Kluyveromyces lactis and Aspergillus oryzae were used to characterize the potential for enzyme stabilization of a two-step strategy: (i) immobilization on glutaraldehyde-agarose (Glut90), (ii) subsequent generation of a hydrophilic nano-environment by reaction with polyaldehyde-dextran polymer (Glut90-Pal), followed by polyamine-dextran polymer (Glut90-Pal-Pam). The derivatives were characterized by kinetics parameters, co-solvent (ethanol and acetone) and temperature stability. Hydrophilization achieved important co-solvent stabilization in all cases. One of the most remarkable results obtained was a 25-fold increase in the half-life of the A. oryzae Glut90-Pal-Pam derivative in 50% (v/v) acetone. Stabilization achieved in very drastic co-solvent concentrations is directly related to the hydrophilization of the nano-environment. The KM values show that the hydrophilic shell appears to behave as an open structure and may create a “partition effect” that protects the enzymes from denaturation. These results show the potential of hydrophilization for building up additional stabilization of immobilized enzymes which would make possible the development of industrial applications</dcterms:abstract>

This procedure allowed
obtaining concentrated Rangpur lime juices between 23 and 51.3 °Brix, without affecting their organoleptic
and nutritional characteristics. The efficiency of the procedure was inversely related to the film thickness
of the matrix to be lyophilised. The sublimation speed per unit area diminished with the reduction in
the residual moisture content of the juice. The mathematical model that fitted to the concentration procedure
allowed to predict the time required for concentrating Rangpur lime by freeze-drying starting from
samples of different thickness. When compared with other citric juices evaluated, the Rangpur lime juice
was the most suitable for freeze-drying concentration. The results could be used to devise optimal industrial
freeze-drying cycles for processing Rangpur lime juices at different concentrations.</dcterms:abstract>

<dcterms:abstract>A theoretical docking study was conducted on a sample of previously reported nitrofuran derivatives, at the binding sites of Trypanosoma cruzi trypanothione reductase (TR) and human erythrocyte glutathione reductase (GR), in order to examine interaction energies (affinities) towards the parasite enzyme and check for selectivity with respect to the human counterpart. A large proportion of nitrofurans were previously shown to be TR inhibitors and some of them have in vitro trypanocidal action as well. The analysis of data collected from the docking procedure was undertaken both from the numeric and graphical standpoints, including the comparison of force field energies, molecular contacts and spatial location of the different orientations that ligands acquired at the binding sites. The results clearly suggest that nitrofurans are not able to selectively bind at the active site of the parasite TR, attaining even larger interaction energies at GR active site. The reason for inhibitor specificity is two-fold: (a) the nitrofuran group acts as a strong electrostatic anchor in GR; (b) the compounds form more stable complexes when binding at the non-selective dimmer interface. Therefore, these two aspects should be specially considered when seeking good candidates for leaders in regards the development of drugs with selective inhibition of TR.</dcterms:abstract>

<dcterms:abstract>The solubility of different ethyl esters derivatized from hake liver oil in supercritical carbon dioxide was studied. A selectivity factor was used to determine optimal conditions to fractionate the ethyl ester mixture. A strong influence of solvent pressure and temperature was observed within8.63–18.04 MPa and 40–70 C. The lowest total solubility of the ethyl ester mixture was obtained when using supercritical carbon dioxide at the lowest density (the lowest pressure and the highest temperatures value tested). The highest discrimination against long-chain polyunsaturated fatty acids (e.g. EPA and DHA) was also obtained at these above conditions. Conversely, higher solubility and lower selectivity were obtained when solvent density increased. Considering this inverse correlation between selectivity and solubility, a single-step batch-fractionation process was designed to increase the 22:6 ethyl ester content from an initial value of 17.5% in the starting material to 55% in the final extract.</dcterms:abstract>

<dcterms:abstract>In this study sludge wash-out was evaluated as a strategy to start-up the Anammox process in order to establish it in a shorter period of time. Sludge from a domestic wastewater treatment plant (WTP) was used to seed two (RI and RII) anaerobic sequencing batch reactors (ASBR). During the start-up period RI was operated as a continuous stirred tank reactor (CSTR) using a dilution rate of 0.2d-1, which promoted the sludge wash-out. After this period, the remaining sludge was retained in the reactor. The reactor RII was operated as an ASBR throughout the study period with a high cell retention. The performance of the two reactors in terms of nitrogen removal was compared over a period of 380 days. During the last RI operation phase the specific nitrogen removal rate increased exponentially, attaining values of 85mgN/gTSS.d. However, a rate of 190mgN/gTSS.d in the batch test under optimal conditions was achieved. The specific nitrogen removal rate remained almost constant for RII with a mean value of 6mgN/gTSS.d being observed during the operation period. The rate for the RII batch test was 20mgN/gTSS.d. These results confirm that the higher TSS in RII (reactor with high cell retention) was not effective in terms of N removal improvement. Anammox-like bacteria were found using fluorescence in situ hybridization (FISH) in reactor RI after 225 days and a new Anammox species was identified.</dcterms:abstract>

<dcterms:abstract>The barrier for internal rotation around the OOCH3 bond in 2,3,5,6- tetrafluoroanisole was calculated using the density functional theory (DFT) and secondorder Møller–Plesset (MP2) methods with Pople’s basis sets up to 6-311G(3df,2p) and Jensen basis sets up to pc-3. The results are converged only if fairly large basis sets are used (at least 6-311G(3df,2pd) or pc-2). Both the DFT and MP2 potential energy curves show internal structure. Two minima and three maxima are observed on the curves, arising from the interplay between lone-pair delocalization and changes in the hybridization around the oxygen atom, together with the attraction between the positively polarized hydrogens in the methyl group and the negatively polarized fluorine atom at the ortho position. These critical points are somehow ironed out by the addition of zero-point and thermal corrections to the energy curves. At this level, the MP2 method can describe reasonably well the previously determined single-well experimental rotational barrier, 2.7 2.0 kcal/mol at 298 K, while all DFT methods yield a much smaller result. As observed experimentally, the OOCH3 group is perpendicular to the aryl ring in the equilibrium structure, although two very close minima with an intermediate hump at 90° are still observable. The theoretical free energy barrier of rotation at the MP2(full)/pc-2 level is 2.0 1.0 kcal/mol, in reasonable agreement with the experimental determination.</dcterms:abstract>

The lectin agglutinates mouse red cells exhibiting broad specificity towards several monosaccharides including the N-acetylneuraminic acid. Agglutination was also inhibited by the glycoproteins: fetuin, lactoferrin, and recombinant erythropoietin. GSL is a glycoprotein possessing 16 % of carbohydrates; the SDS-PAGE showed two bands with molecular mass of 52.1 and 64.4 kDa. Isoelectric focusing displayed microheterogeneity, with two bands at pIs 5.1 and 5.3. The lectin was stable between pH 2 and pH 8 while at pH 10, the agglutination decayed to 50%of initial activity. Incubation at 40 and 80 °C led to 50 and 100 % loss in activity of the lectin, respectively. Synthesized GSL-Sepharose interacts with serum pregnant mare gonadotropin, and at least two subpopulations of this glycoprotein were separated. There was no interaction between transferrin and soluble GSL while a partial recognition was achieved with GSL-Sepharose. The terminal sialic acid seems to play an active role in modifying the interaction with GSL, depending if the lectin is in a soluble or immobilized form. In HA inhibition assays, 43 carbohydrates including mono-, di-, tri-, tetrasaccharides, glycoproteins, and polysaccharides were tested as haptens, to determine the saccharide-binding specificities of the lectins. A novel lectin with specificity towards N-acetyl-glucosamine was purified from mycelia of Punctularia atropurpurascens using affinity chromatography on chitosan-Sepharose. The lectin has a subunit molecular mass of 67 kDa determined by SDS-PAGE and a pI of 5.0.</dcterms:abstract>

<dcterms:abstract>OBJECTIVE: There is little evidence regarding the role of (68)Ga-DOTATATE PET/CT for the identification of primary tumors in patients with metastatic neuroendocrine carcinoma of unknown primary. The aim of this study is to assess the value of this technique in the mentioned clinical scenario. METHODS: We retrospectively studied twenty-nine patients (mean age 59.5 ± 10.6 years; female 17) with pathologically proven neuroendocrine metastases. In all cases conventional imaging was negative for primary tumor identification. (68)Ga-DOTATATE PET/CT was performed with a mean dose of 104.2 ± 18.8 MBq, using a 64-slice PET/CT with time-of-flight correction. A team of an experienced radiologist and a nuclear medicine physician evaluated the images. The maximum SUV (SUVm) was measured in all abnormal foci. Histopathology (when available) and/or clinical follow-up with correlative imaging was considered as reference standard. RESULTS: (68)Ga-DOTATATE PET/CT identified the primary tumor in 17/29 (59%) patients in the following locations: pancreas (n = 7), ileum (n = 7), duodenum (n = 1), colon (n = 1) and stomach (n = 1). In this population a significant correlation was found between SUVm of primary tumor and metastases (r = 0.815, P < 0.0001). Furthermore, additional sites of unsuspected metastases were demonstrated in 9 patients of this group and in 6 patients in whom no primary tumor was localized, mainly in lymph nodes and mesentery. Pathology confirmation was obtained in 7 patients who underwent surgery, whereas in the remaining 10 patients, correlative imaging and follow-up confirmed primary tumor localization. CONCLUSIONS: (68)Ga-DOTATATE PET/CT is a clinically useful imaging technique for the localization of primary tumors in patients with neuroendocrine metastatic carcinoma of unknown origin with the potential of having a significant impact in patient management and therapy planning.</dcterms:abstract>
