    <dcterms:abstract><![CDATA[A multiproxy analysis of diatoms, chrysophyte cysts, opal phytoliths, and palynomorphs was undertaken in Negra Lagoon, to decipher Holocene changes in paleobotanical proxies associated with sea-level changes and paleolimnological conditions. Before the Holocene transgression (7000 yr BP), a terrestrial system was inferred as no aquatic palynomorphs or biogenic silica remains were recorded. During the sea-level maximum (5200 yr BP), marine/ brackish conditions were established as indicated by diatoms, Chenopodiaceae, and a high content of cysts of Peridinioideae. The catchment consisted of grasslands and wetlands as indicated by the opal phytolith data. The Holocene transgression was followed by a sea-level decrease, which led to the onset of brackish/freshwater conditions as inferred from the co-dominance of freshwater and marine/ brackish diatoms. This is also supported not only by the concomitant increase in non-siliceous freshwater microalgae and emergent macrophytes, but also by the presence of Myriophyllum. As sea level continued to decrease during late Holocene, a freshwater system was observed because of the complete separation from the ocean. Chrysophyte cysts were consistently recorded and brackish diatoms exhibited reduced abundances. The observed limnological changes are consistent with the alternative states hypothesis of Scheffer&rsquo;s model (1998). That is, the clear water phases were explained because of high macrophyte abundances, low phytoplankton frequencies, high cyst to diatom ratios, and increase in frequencies of benthic diatom species. Conversely, turbid phases were likely to occur when low cyst to diatom ratios together with increase in planktonic diatoms and decrease in macrophytes values were detected. This highlights the importance of this approach to detect long-term changes in shallow lake alternative states.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[Chagas disease represents a relevant health problem in Central and South America. The first line of treatment is Nifurtimox and Benznidazole which have a great deal of disadvantages that demands the rapid generation of therapeutic alternatives. Based in our research on aza-thiaheterocycles as anti-Trypanosoma cruzi agents we identified pharmacophores that act through oxidative stress. Here, we describe the synthesis and the activity of new containing bioactive-heterocycles analogues of naftifine as potential T. cruzi membrane sterol biosynthesis inhibitors. Benzimidazole 1,3-dioxides (11 and 13) and quinoxaline 1,4-dioxides (22 and 23) displayed excellent parasite/mammal selectivity indexes. Analysis of the free sterols from parasite incubated with the compounds showed that any of them are able to accumulate squalene suggesting that in the anti-T. cruzi mechanism of action is not involved the inhibition of sterol biosynthesis. Some derivatives were also tested as antifungal agents. The results obtained in the present work open potential therapeutic possibilities of new compounds for these infectious diseases.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[Many neurodegenerative diseases are characterized by the conformational change of normal self-proteins into amyloidogenic, pathological conformers, which share structural properties such as high b-sheet content and resistance to degradation. The most common is Alzheimer&rsquo;s disease (AD) where the normal soluble amyloid b (sAb) peptide is converted into highly toxic oligomeric Ab and fibrillar Ab that deposits as neuritic plaques and congophilic angiopathy. Currently, there is no highly effective treatment for AD, but immunotherapy is emerging as a potential disease modifying intervention. A major problem with most active and passive immunization approaches for AD is that both the normal sAb and pathogenic forms are equally targeted with the potential of autoimmune inflammation. In order to avoid this pitfall, we have developed a novel immunomodulatory method that specifically targets the pathological conformations, by immunizing with polymerized British amyloidosis (pABri) related peptide which has no sequence homology to Ab or other human proteins. We show that the pABri peptide through conformational mimicry induces a humoral immune response not only to the toxic Ab in APP/PS1 AD transgenic mice but also to paired helical filaments as shown on AD human tissue samples. Treated APP/PS1 mice had a cognitive benefit compared to controls (p,0.0001), associated with a reduction in the amyloid burden (p = 0.0001) and Ab40/42 levels, as well as reduced Ab oligomer levels. This type of immunomodulation has the potential to be a universal b-sheet disrupter, which could be useful for the prevention or treatment of a wide range of neurodegenerative diseases.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The objective of this work was to develop a novel 99mTc complex bearing the 5-nitroimidazol-1-yl moiety with recognised selectivity towards hypoxic tissue, as a potential radiopharmaceutical for imaging tumour hypoxia. The new metronidazole derivative (2-amine-3-[2-(2-methyl-5-nitro-1H-imidazol-1-yl)ethylthio]propanoic acid) (L) containing adequate groups to coordinate technetium through the formation of a Tc(I)-tricarbonyl complex was synthesised with adequate yield (33%) and characterised by spectroscopy. Labelling was performed by substitution of three labile water molecules of the technetium tricarbonyl precursor, fac-[99mTc(CO)3(H2O)3]+ with the ligand. A radiochemical purity higher than 90% was achieved and remained unchanged for more than 4&thinsp;h. The complex has a high stability in plasma, a moderate plasma protein binding and a moderate hydrophilicity. In vitro cell uptake studies showed a ratio between the activity taken up by cells in hypoxia/normoxia of 1.6&thinsp;&plusmn;&thinsp;0.4 (p&thinsp;&lt;&thinsp;0.5). Biodistribution in normal mice showed rapid depuration and low uptake in all organs and tissues except liver. Biodistribution in mice bearing induced tumours showed a low tumour uptake, but tumour/muscle ratio was favourable thanks to depuration. Comparison with biological results of other metronidazole derivatives clearly shows that modifications of the chelator are very important and contribute to improve the biological behaviour.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The mononuclear Cu(II) complex, [Cu(o-Va)2(H2O)2] (o-HVa = o-vanillin, 2-hydroxy-3-methoxybenzaldehyde) has been synthesized and characterized by elemental analyses, FTIR, FT-Raman, and electronic spectroscopies and compared with the results obtained for the free ligand. The optimized geometry, the harmonic vibrational frequencies and the electronic transitions of the complex and the ligand were calculated using methods based on the density functional theory. Antimicrobial activity against Escherichia coli and Staphylococcus aureus and SOD-mimic activities of the complex were studied and compared with the analogous copper complex with vanillin, [Cu(Va)2(H2O)2]. Stability of the compounds in the essayed solution and with time was determined by means of conductimetric measurements. Their redox behavior was studied by cyclic voltammetry and was compared with that observed for the ligands. The complexes undergo two main reductions and one oxidation processes involving the metal center and the coordinated ligand, respectively.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[Logos on environmental sustainability could consist of an effective strategy to provide consumers with accurate, understandable and trustworthy information to encourage them to buy environmentally sustainable wines. However, the large number of different logos indicating environmental sustainability available in the market raises the question ofwhether their associated messages are successfully conveyed to consumers. In this context, the aim of the present exploratory study was to investigate how Burgundy wine consumers perceive a series of logos indicating environmental sustainability in wine production. Fourteen logos available in the French market were selected: three logos being specific to wine and eleven nonspecific. The logos were presented to 127 wine consumers from Dijon area (France), following an incomplete balanced block design. For each logo, participants had to answer the question: &ldquo;What does a bottle of wine with this logo suggest to you?&rdquo;. Responses were qualitatively analyzed and grouped into different categories. Chi-square tests and Correspondence analysiswere used to identify the relationship among logos and categories. Results showed large differences in how consumers perceived the logos. Biodyvin, the former European AB and the French AB organic logos were the logos that most successfully conveyed their messages, being strongly associated to organic wine. Most logos did not communicate a message related to environmental sustainability, which reaffirms the need to provide consumers with adequate information on environmental sustainability and to conduct further research on this subject.]]></dcterms:abstract>

Interestingly, phenoxyindole-4,9-dione 9d displayed excellent nanomolar inhibitory activity, IC50 = 20 nM, and high selectivity index, SI = 625.     <dcterms:abstract><![CDATA[The goals of the current trial were (a) to characterize the plasma disposition kinetics of levamisole (LEV), albendazole (ABZ) and ivermectin (IVM), each administered either alone (single active ingredient) or as a combined formulation to lambs; (b) to compare the clinical anthelmintic efficacy of the same drugs given either separately or co-administered to lambs infected with resistant nematodes. Fifty Corriedale lambs naturally infected with multiple resistant gastrointestinal nematodes were involved in the following experimental trials: (a) "Pharmacokinetic trial": the animals were allocated into five groups (n=10 each) and intraruminally treated with either LEV (8 mg/kg), ABZ (5mg/kg), IVM (0.2mg/kg), or with a LEV+ABZ+IVM combined formulation, where each active ingredient was administered at the same dose. Blood samples were collected over 15 days post-treatment and drug plasma concentrations measured by HPLC. (b) "Efficacy trial": the same treated groups plus an untreated control group were used to assess the comparative anthelmintic efficacy by the faecal egg count reduction test (FECRT). Although the overall LEV disposition kinetics was unaffected, significantly lower (61%) ABZ-sulphoxide and higher (71%) IVM systemic availabilities were obtained after administration of the combined formulation in comparison to those obtained after treatment with each drug alone. A multiple drug resistance situation was observed for Haemonchus spp. The observed efficacies were 52% (LEV), 72% (ABZ), 80% (IVM) and 87% (triple combined formulation). The results reported here contribute to the pharmaco-therapeutic knowledge on drug combinations. This type of research is crucial before further development of combined anthelmintic preparations reaches the market to deal with resistant nematode control. The co-administration of LEV+ABZ+IVM did not result in a significant advantageous anthelmintic effect compared to the treatment with IVM alone. The simultaneous/combined administration of LEV, ABZ and IVM may account for a drug-drug pharmacological interaction in infected lambs. The pharmacokinetic interaction accounted for a reduced ABZ-sulphoxide and enhanced IVM systemic exposure following the combined treatment.]]></dcterms:abstract>

<h4>Problem</h4>
<div class="para">
<p>Women with celiac disease (CD) are often affected by atypical presentations of the disease associated with reproductive disorders as a main extra-digestive complaint. Here, we analyzed if autoantibodies against tissue transglutaminase (tTG) in sera from CD patients with reproductive disorders could display direct effects through their interaction with tTG expressed on trophoblast cells and phagocytes inducing tissue damage and interfering in the clearance of trophoblast apoptotic bodies.</p>

<h4>Method of study</h4>
<div class="para">
<p>Sera from CD women with reproductive disorders were obtained, and their ability to induce apoptosis of Swan-71 (cytotrophoblast cell line) and to modulate the wound-healing and phagocytes process was tested.</p>

<h4>Results</h4>
<div class="para">
<p>Swan-71 cells expressed tTG and CD sera displayed a significant decrease in trophoblast cell migration and a delay in injury healing on trophoblast cells, compared with those observed with control sera. Moreover, CD sera significantly reduced trophoblast cell proliferation and increased apoptosis levels in comparison with those observed in the control sera. Finally, autoantibodies against tTG interfere in the clearance of trophoblast apoptotic bodies through a mechanism involving MFG-E8 (milk fat globulin-EGF factor 8)&ndash;tTG binding.</p>

<p>The anti-tTG antibodies might contribute to trophoblast damage and disrupt the phagocytosis process of apoptotic bodies that could promote a pro-inflammatory microenvironment.</p>

    <dcterms:abstract><![CDATA[Autoimmunity is a feature of celiac disease (CD) with tissue transglutaminase (tTG) as a major autoantigen. A correlation between gynecological-obstetric disorders in CD patients and the presence of circulating antibodies anti-tTG that inhibited tTG activity was reported. Serum anti-tTG antibodies were detected in a non-obese diabetic (NOD) mouse model of type I insulin-dependent diabetes mellitus and Sj&ouml;gren's syndrome, two comorbid states with CD. Since pregnancy complications have been described in NOD mice, we evaluated the ability of anti-tTG antibodies to affect the functions of tTG relevant to the normal course of an early pregnancy like extracellular matrix assembling and apoptotic cell phagocytosis by macrophages. Circulating IgG antibodies against tTG were detected in NOD mice with titers that decreased at early pregnancy; interestingly, the in vitro transamidating activity of tTG was reduced by NOD serum samples. Particularly, anti-tTG antibody inhibited apoptotic cell phagocytosis by peritoneal macrophages from pregnant NOD mice that express the enzyme on surface. Evidence provided support for a role for anti-tTG antibodies through reduced transamidating activity and reduced apoptotic cell clearance by the macrophages of pregnant NOD mice.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[This study provides insights into the pathogenesis of Ralstonia solanacearum, in particular with regards to strains belonging to phylotype IIB, sequevar 1 (IIB-1) and their interaction with potato, its natural host. We performed a comparative genomic analysis among IIB-1 R. solanacearum strains with different levels of virulence in order to identify candidate virulence genes. With this approach, we identified a 33.7-kb deletion in a strain showing reduced virulence on potato. This region contains a cluster of six genes putatively involved in type IV pili (Tfp) biogenesis. Functional analysis suggests that these proteins contribute to several Tfp-related functions such as twitching motility and biofilm formation. In addition, this genetic cluster was found to contribute to early bacterial wilt pathogenesis and colonization fitness of potato roots.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[Parasitic illnesses are major causes of human disease and misery worldwide. Among them, both amebiasis and Chagas disease, caused by the protozoan parasites, Entamoeba histolytica and Trypanosoma cruzi, are responsible for thousands of annual deaths. The lack of safe and effective chemotherapy and/or the appearance of current drug resistance make the development of novel pharmacological tools for their treatment relevant. In this sense, within the framework of the medicinal inorganic chemistry, metal-based drugs appear to be a good alternative to find a pharmacological answer to parasitic diseases. In this work, novel ruthenium complexes [RuCl2(HL)(HPTA)2]Cl2 with HL=bioactive 5-nitrofuryl containing thiosemicarbazones and PTA=1,3,5-triaza-7-phosphaadamantane have been synthesized and fully characterized. PTA was included as co-ligand in order to modulate complexes aqueous solubility. In fact, obtained complexes were water soluble. Their activity against T. cruzi and E. histolytica was evaluated in vitro. [RuCl2(HL4)(HPTA)2]Cl2 complex, with HL4=N-phenyl-5-nitrofuryl-thiosemicarbazone, was the most active compound against both parasites. In particular, it showed an excellent activity against E. histolytica (half maximal inhibitory concentration (IC50)=5.2 &mu;M), even higher than that of the reference drug metronidazole. In addition, this complex turns out to be selective for E. histolytica (selectivity index (SI)&gt;38). The potential mechanism of antiparasitic action of the obtained ruthenium complexes could involve oxidative stress for both parasites. Additionally, complexes could interact with DNA as second potential target by an intercalative-like mode. Obtained results could be considered a contribution in the search for metal compounds that could be active against multiple parasites.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[In this study, we report a strategy using dynamic combinatorial chemistry for targeting the thioredoxin (Trx)-reductase catalytic site on Trx glutathione reductase (TGR), a pyridine nucleotide thiol-disulfide oxido-reductase. We chose Echinococcus granulosus TGR since it is a bottleneck enzyme of platyhelminth parasites and a validated pharmacological target. A dynamic combinatorial library (DCL) was constructed based on thiol-disulfide reversible exchange. We demonstrate the use of 5-thio-2-nitrobenzoic acid (TNB) as a non-covalent anchor fragment in a DCL templated by E. granulosus TGR. The heterodimer of TNB and bisthiazolidine (2af) was identified, upon library analysis by HPLC (IC[Formula: see text] = 24 [Formula: see text]M). Furthermore, 14 analogs were synthetically prepared and evaluated against TGR. This allowed the study of a structure-activity relationship and the identification of a disulfide TNB-tricyclic bisthiazolidine (2aj) as the best enzyme inhibitor in these series, with an IC[Formula: see text] = 14 [Formula: see text]M. Thus, our results validate the use of DCL for targeting thiol-disulfide oxido-reductases.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The freshwater cyanotoxins, microcystins (MCs), pose a global public health threat as potent hepatotoxins in cyanobacterial blooms; their persistence in drinking and recreational water has been associated with potential chronic effects in addition to acute intoxications. Rapid and accurate detection of the over 80 structural congeners is challenged by the rigorous and time consuming clean up required to overcome interference found in raw water samples. MALDI-MS has shown promise for rapid quantification of individual congeners in raw water samples, with very low operative cost, but so far limited sensitivity and lack of available and versatile internal standards (ISs) has limited its use. Two easily synthesized S-hydroxyethyl-Cys(7)-MC-LR and -RR ISs were used to generate linear standard curves in a reflectron MALDI instrument, reproducible across several orders of magnitude for MC-LR, -RR and -YR. Minimum quantification limits in direct water samples with no clean up or concentration step involved were consistently below 7 &mu;g/L, with recoveries from spiked samples between 80 and 119%. This method improves sensitivity by 30 fold over previous reports of quantitative MALDI-TOF applications to MCs and provides a salient option for rapid throughput analysis for multiple MC congeners in untreated raw surface water blooms as a means to identify source public health threats and target intervention strategies within a watershed. As demonstrated by analysis of a set of samples from Uruguay, utilizing the reaction of different MC congeners with alternate sulfhydryl compounds, the m/z of the IS can be customized to avoid overlap with interfering compounds in local surface water samples.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[In freshwater, harmful cyanobacterial blooms threaten to increase with global climate change and eutrophication of surface waters. In addition to the burden and necessity of removal of algal material during water treatment processes, bloom-forming cyanobacteria can produce a class of remarkably stable toxins, microcystins, difficult to remove from drinking water sources. A number of animal intoxications over the past 20 years have served as sentinels for widespread risk presented by microcystins. Cyanobacterial blooms have the potential to threaten severely both public health and the regional economy of affected communities, particularly those with limited infrastructure or resources. Our main objectives were to assess whether existing water treatment infrastructure provides sufficient protection against microcystin exposure, identify available options feasible to implement in resource-limited communities in bloom scenarios and to identify strategies for improved solutions. Finally, interventions at the watershed level aimed at bloom prevention and risk reduction for entry into potable water sources were outlined. We evaluated primary studies, reviews and reports for treatment options for microcystins in surface waters, potable water sources and treatment plants. Because of the difficulty of removal of microcystins, prevention is ideal; once in the public water supply, the coarse removal of cyanobacterial cells combined with secondary carbon filtration of dissolved toxins currently provides the greatest potential for protection of public health.     <dcterms:abstract><![CDATA[Glass ceramics of PbB4O7 doped with 0.15 and 0.50% molar concentration of Ce and Eu were prepared by means of controlled crystallization of the glass powder. Crystallization was carried out at 590 &deg;C for the undoped and at 640 &deg;C for the doped glass ceramic samples. Heat treatment time was 60 and 180 min for both kinds of samples. Crystalline phase analysis reveals that the only present phase was lead diborate. Thermal analysis characterization shows that Europium may act as nucleation promoter, lowering the energy barrier for crystallization. On the other hand Cerium acts as inhibitor of the crystallization process. Optical characterization was done by means of absorption spectroscopy and luminescence measurements at room temperature. A shift in absorption edges was observed for both samples; this shift could be attributed to a large amount of BO4 groups as dopant concentration increases. From the luminescence spectra it was showed that the 5D0 to 7F2 red emission (613 nm) occurs through an electric dipole mechanism which is dominant in all the Europium doped samples, while in Cerium doped samples the emission peaks are due to the 5d&ndash;2F5/2 (528 nm) and 5d&ndash;2F7/2 (472 nm) transitions. Thermoluminescence analysis was performed with different irradiation doses (1 and 10 Gy) on samples at different concentrations. Results show the incorporation of localized states near the conduction band.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The syringyl/guaiacyl (S/G) ratio of Eucalyputs grandis lignin was determined and its relation with kraft pulping parameters was studied. Twenty one wood samples obtained from 10-year-old trees grown in the same place were analyzed using Py-GC-MS to determine the syringyl/guaiacyl (S/G) ratio. The samples were pulped to the same final lignin content (Kappa number 18) obtaining pulp yields between 48.8% and 54.3%. Relationships were observed between pulp yield and S/G ratio (r = 0.51) also between alkali charge used in pulping and S/G ratio (r = - 0.60).]]></dcterms:abstract>
