In a previous work we have identified two possible structures for the radical cation obtained by ionization of hydrogen‐bonded formaldehyde–water complexes [Coitiño et al., J. Am. Chem. Soc. 115, 9121 (1993)], a hydrogen‐bonded and an addition‐like complexes. We observed that the results were highly dependent on the method of calculation employed. Inclusion of correlation was crucial for obtaining the correct structures of some of the complexes. In this work we used high‐level ab initio calculations in order to predict the equilibrium structure of these two complexes, the possibility of its existence in gas phase, and the infrared spectrum to be expected in each case. A series of progressively more sophisticated basis sets was used to assess the effect of the quality of the calculations on the expected results. Also, full geometry optimization with each basis set was performed at the second‐order Mo/ller–Plesset level, and correlation energy was calculated at the fourth‐order Mo/ller–Plesset level to assess the contribution of this factor to the global result. Confirming our previous results, we found that correlation affects the hydrogen‐bonded radical‐cation complex more than the addition one, due to the different bonding patterns in each of them. Both complexes are stable—toward decomposition to the fragments or to CO+H+H3O+—by several kcal/mol at all levels of theory. The hydrogen‐bonded complex is more stable than the additional one by a respectable amount (13 kcal/mol at the highest level used here), lending support to our previous analysis of the reactions of the former as the main channels for evolution of the formaldehyde–water radical cation. The H‐bonded complex [H3O+...HCO⋅] presents two characteristics, very intense absorptions which should allow identification of this radical cation if present in the experimental setup. These transitions are also present in the HCO⋅ radical but their intensity is enhanced by an order of magnitude due to the coupling with the proton in H3O+. We conclude that the combination of stability and characteristic infrared transitions should make this radical‐cation complex a relatively easy target for experimental determination.

A summary of the resource.
The condensation of an aldehyde (formaldehyde or acetaldehyde) with vinyl alcohol has been studied using quantum-mechanical methods. Reaction models with and without an additional water molecule have been examined in parallel. An analysis of the electronic changes during the reaciton has been performed, using a localized orbital approach. The effect of the solvent has been introduced, making use of a continuous dielectric model developed in our laboratory. Analysis of the reaction mechanism shows that a water molecule eases the condensation process, and may be considered as a catalyst.

It has been reported that restricted and unrestricted MP2 ab initio calculations of the energy difference between the formaldehyde (H2CO+) and hydroxymethylene (HCOH+) radical cations are seriously in error because of poor convergence of the perturbation series. We had reported a study of the formaldehyde—water radical cation and found MP2 to be reliable. A study of the correlation energy contribution to the isomeric structures formaldehyde—water and hydroxymethylene—water radical cations is reported. Both perturbation theory (from MP2 to MP4SDTQ) and configuration interaction (CISD and CISD(Q)) calculations were performed with the 6-311 + G(d, p) basis set at the MP2/6-311 + G(d, p) optimum geometries. The MP2 flaw in formaldehyde/hydroxymethylene is not present in the radical cations derived from the complexes with water. Analysis of our results for formaldehyde/hydroxymethylene shows that the flow is mainly connected with poor convergence of the perturbation series for the formaldehyde radical cation (and not for the hydroxymethylene). Since proton transfer occurs in the complexes with water, this species is not present in our calculations, so the results are reliable. The hydroxymethylene—water radical cation is predicted to be more stable than the formaldehyde—water radical cation by 29 kJ/mol on the basis of CISD(Q)/6-311 + G(d, p)//MP2/6-311 + G(d, p) calculations.

A summary of the resource.
A new integrated technique employing the capped subsystem strategy of the IMOMO method is used as a way to include electron correlation (configuration mixing) in a molecular subsystem without requiring a transformation to localized orbitals. We obtain good results for bond energies that require a correlated treatment of at least part of the molecule, and we include the effect of substituents in any part of the molecule. An especially important result is that the method provides a convenient and practical way to include electron correlation in a localized subsystem, while retaining the quantum mechanical electronic effects of uncorrelated substituents.

A new formulation (CLS-PCM) for the calculation of the apparent surface charges in the framework of the ab initio polarizable continuum model of the solvent (PCM) is introduced. Its performance is compared with that of the original iterative version (ITER-PCM) of the method as well as with a matricial alternative formulation (matrix-BEM-PCM) of the same problem. Both CLS-PCM and matrix-BEM-PCM have shown to be computationally more efficient than ITER-PCM without presenting any problems associated with the convergence of the process. Although for small and medium-size solutes the use of matrix-BEM-PCM is recommended, for neutral solutes of larger size the use of CLS2 becomes computationally more convenient. Finally, for very large-size systems, compromise between matrix storage requirements, time of calculation, and exactness of the results may make preferable the use of the more approximate CLS1 formalism, possibly in conjunction with semiempirical or semiclassical descriptions of the solute.

Sin embargo la planta de soja tiene una extraordinaria capacidad para compensar la defoliación provocada por esta clase de insectos. Para evitar que este tipo de plagas ocasionen daños que afecten la producción del cultivo se hace necesario recurrir a la aplicación de productos químicos.

A summary of the resource.
The parallel segmented flow (PSF) chromatography column was applied for the first time to trace residue analyses with mass spectral detection. Three problematic pesticides, notorious for poor peak shape and ionization issues were selected to illustrate the advantages and disadvantages of utilizing the PSF column as an alternative front-end separation interface for LC-MS approaches. Assessment of performance for each pesticide was based on a spiked lemon matrix prepared via a QuEChERS (quick, easy, cheap, rugged and safe) sample preparation approach. Comparisons were made between assays conducted using a conventional column and a PSF column (operated with a 40% segmentation ratio), each packed with identical stationary phases and of the same column format. Both columns were connected to the MS detector using the exact same connective tubing and electrospray (ESI) conditions. The primary advantage of the PSF column was that it enabled an increase in throughput by at least 2.5 times compared to the conventional HPLC column. At the same time, the volumetric flow to the MS was exactly the same for both the PSF column and the conventional column. Effectively there was no difference in the sensitivity provided by peak areas between the PSF and conventional columns: peak area LOQ values were comparable, 4–9 μg/kg for the PSF and 4–8 μg/kg for the conventional column. This is a vital requirement for food control methods to secure absence at default maximum residue limit (MRL) of 10 μg/kg (10 ppb) level. Since the efficiency of the PSF was superior to that of the conventional column, the peak width of eluting components was reduced by as much as 70% in the former compared to the peak widths on the latter. Hence, the PSF column provides greater peak capacity and this will enable faster analysis of complex samples.

Abstract
A summary of the resource.
Chondriamide C (3), a new bis(indole) amide, was isolated from the red alga Chondria atropurpurea, and its structure was established from spectroscopic data and chemical transformations. A new natural product, 3-indoleacrylamide (4), and the previously described chondriamides A and B (1, 2) and 3-indoleacrylic acid (5) were also isolated. The anthelmintic activities of compounds 1, 3, 4, and 6 (the O,N1,N1'-trimethyl derivative of compound 2) against Nippostrongylus brasiliensis in vitro were evaluated.

Abstract
A summary of the resource.
Porphyra drewiana Coll et Oliveira, sp. nov., is described from plants collected on the south-east coast of Brazil. The species proposed is monostromatic, monoecious, monoplastidial, without marginal microscopic teeth and does not produce monospores. Both phases, leafy and filamentous, have three chromosomes. Morphologically the most similar species is Porphyra spiralis Oliveira et Coll var. amplifolia Oliveira et Coll, from which it differs by: (i) thallus gross morphology; (ii) scattered pluristromatic areas of vegetative cells; (iii) division of the plastids prior to the nucleus at the first division of the carpospores mother cell; (iv) the number of carpospores and spermatia produced per mother cell; and (v) morphology and behavior of the filamentous phase in cultures. An identification key for the species referred to Brazil is included.

Several breeding programs are under way to introduce resistance to bacterial wilt caused by Ralstonia solanacearum in solanaceous crops. The lack of screening methods allowing easy measurement of pathogen colonization and the inability to detect latent (i.e. symptomless) infections are major limitations when evaluating resistance to this disease in plant germplasm. We describe a new method to study the interaction between R. solanacearum and potato germplasm that overcomes these restrictions. The R. solanacearum strain UY031 was genetically modified to constitutively generate light from a synthetic luxCDABE operon stably inserted in its chromosome. Colonization of this reporter strain on different potato accessions was followed using life imaging. Bacterial detection in planta by this non-disruptive system correlated with the development of wilting symptoms. In addition, we demonstrated that quantitative detection of the recombinant strain using a luminometer can identify latent infections on symptomless potato plants. We have developed a novel, unsophisticated and accurate method for high-throughput evaluation of pathogen colonization in plant populations. We applied this method to compare the behavior of potato accessions with contrasting resistance to R. solanacearum. This new system will be especially useful to detect latency in symptomless parental lines before their inclusion in long-term breeding programs for disease resistance.

With the aim of monitoring multiclass semi-polar pesticide residues in freshwater fatty fish, two QuEChERS approaches (so-called acetate buffered and unbuffered versions) were evaluated for the determination of 77 pesticide residues. Compounds were selected according to the dominant rainfed agriculture activities in South America. Unbuffered QuEChERS was finally chosen for validation purposes owing that it provided the best results in terms of recovery yields. Method performance was evaluated in two instrumental systems, liquid chromatography – tandem mass spectrometry (LC–MS/MS) in Scheduled MRM™ algorithm available on hybrid quadrupole – linear ion trap (QLIT) instrument, and gas chromatography – mass spectrometry (GC–MS) under selected ion monitoring (SIM) mode. Spiking experiments were carried out to determine the trueness, precision, linearity, limit of quantification of the method as well as matrix effect. The Unbuffered QuECHERS method described here: • Was validated for the analysis of 67 pesticide residues in fish muscle tissue. • Presented quantification limits in the range 1–15 μg kg−1 for the vast majority of the studied compounds. • Enable environmental monitoring of pesticide residues in fish due to their low LOQs.

Valero-fenbendazole (VAL-FBZ) is a novel hybrid compound with in vitro anthelmintic activity, designed and synthesized to address the global problem of resistance to anthelmintic compounds. This new molecule derives from fenbendazole (FBZ), a wellknown commercially available benzimidazole used in veterinary medicine despite its poor water solubility. In this work, we report for the first time a strategy to solve the solubility problems of FBZ and VAL-FBZ by means of self-dispersible nanocrystals (SDNC). Nanocrystals were prepared by media milling followed by a spray-drying step, and a comprehensive and exhaustive structural and physicochemical characterization was carried out, in order to understand the systems and their behavior. The formulation poloxamer 188 (P188):FBZ 1:1 turned out with the best process yield (53%) and re-dispersability properties, particle size average of 258 nm, and polydispersity index of 0.2 after redispersion in water. The dissolution profile showed a markedly increased dissolution rate compared with the simple mixture of the components (80% FBZ dissolved in 15 min from the SDNC vs 14% from the control formulation). FTIR spectroscopy, thermal analysis, and X-Ray Powder Diffraction (XRPD) studies showed no chemical interactions between components and an extensive confocal Raman microscopy analysis of the formulations showed very homogeneous spatial distribution of components in the SDNC samples. This manufacturing process was then successfully transferred for preparing and characterizing VAL-FBZ:P188 (1:1) SDNC with similar results, suggesting the promising interest of a novel anthelmintic with improved biopharmaceutical behavior. The maintenance of thiol–redox homeostasis is vital to the survival of living organisms. Sulfur-based low-molecular weight compounds and proteins synthesized by cells provide efficient and specific ways to counteract oxidative stress and regulate cellular processes. For these tasks, most organisms share the glutathione and thioredoxin NADPH-dependent redox systems. However, in certain lineages, evolution has taken different paths that led to the emergence of novel cysteine-based low-molecular weight redox cofactors, around which new redox systems evolved. These include the sugar-based cysteinyl derivatives mycothiol and bacillithiol, and ergothioneine (EGT), which are present in different phyla from bacteria. Within Eukarya, some fungi contain EGT, whereas trypanothione is unique to species from the Euglenozoa family. This Forum compiles the state-of-the-art knowledge about these noncanonical redox systems of pathogenic organisms. The functions in physiology and pathogenicity, as well as structural and biochemical specializations that these system components evolved, are thoroughly discussed.

Polyamides-based compounds related to the Streptomycetal distamycin and netropsin are potent cytostatic molecules that bind to AT-rich regions of the minor groove of the DNA, hence interfering with DNA replication and transcription. Recently, derivatives belonging to this scaffold have been reported to halt the proliferation of deadly African trypanosomes by different and unrelated mechanisms. Here we describe the synthesis and preliminary characterization of the anti-trypanosomal mode of action of new potent and selective distamycin analogues. Two tri-heterocyclic derivatives containing a central N-methyl pyrrole ring (16 and 17) displayed high activity (EC50 < 20 nM) and selectivity (selectivity index >5000 with respect to mammalian macrophages) against the infective form of T. brucei. Both compounds caused cell cycle arrest by blocking the replication of the mitochondrial DNA but without affecting its integrity. This mode of action clearly differs from that reported for classical minor groove binder (MGB) drugs, which induce the degradation of the mitochondrial DNA. In line with this, in vitro assays suggest that 16 and 17 have a comparatively lower affinity for different template DNAs than the MGB drug diminazene. Therapeutic efficacy studies and stability assays suggest that the pharmacological properties of the hits should be optimized. The compounds can be rated as excellent scaffolds for the design of highly potent and selective anti-T. brucei agents.

<p>The first stage of the drug discovery process involves the identification of small compounds with biological activity. Iboga alkaloids are monoterpene indole alkaloids (MIAs) containing a fused isoquinuclidine-tetrahydroazepine ring. Both the natural products and the iboga-inspired synthetic analogs have shown a wide variety of biological activities. Herein, we describe the chemoenzymatic preparation of a small library of novel N-indolylethyl-substituted isoquinuclidines as iboga-inspired<br />compounds, using toluene as a starting material and an imine Diels–Alder reaction as the key step in the synthesis. The new iboga series was investigated for its potential to promote the release of glial cell line-derived neurotrophic factor (GDNF) by C6 glioma cells, and to inhibit the growth of infective trypanosomes. GDNF is a neurotrophic factor widely recognized by its crucial role in development, survival, maintenance, and protection of dopaminergic neuronal circuitries affected in<br />several neurological and psychiatric pathologies. Four compounds of the series showed promising activity as GDNF releasers, and a leading structure (compound 11) was identified for further studies. The same four compounds impaired the growth of bloodstream Trypanosoma brucei brucei (EC50<br />1–8 uM) and two of them (compounds 6 and 14) showed a good selectivity index.</p>

Propolis samples from north‐west Argentina (Amaicha del Valle, Tucumán,) were evaluated by palynology, FT‐IR spectra, and RP‐HPTLC. In addition, the volatile fraction was studied by HS‐SPME‐GC‐MS. The botanical species most visited by Apis mellifera L. near the apiaries were collected and their RP‐HPTLC extracts profiles were compared with propolis samples. In addition, GC‐MS was performed for volatile compounds from Zuccagnia punctata Cav. (Fabaceae). FT‐IR spectra and RP‐HPTLC fingerprints of propolis samples showed similar profiles. In RP‐HPTLC analyses, only Z. punctata presented a similar fingerprint to Amaicha propolis. The major volatile compounds present in both were trans‐linalool oxide (furanoid), 6‐camphenone, linalool, trans‐pinocarveol, p‐cymen‐8‐ol, and 2,3,6‐trimethylbenzaldehyde. Potential variations for the Amaicha del Valle propolis volatile fraction as consequence of propolis sample preparation were demonstrated.

A summary of the resource.
The goal of our study was to improve Tannat grape and wine composition by achieving vine balance in high-capacity conditions. The current Uruguayan grape-growing paradigm accepts unpredictable periods of water deficit or excess in non-irrigated vineyards, only applying herbicides to eliminate weed competition (H). We used an under-trellis cover crop (UTCC) to limit vine water availability and to reduce vine growth rate and final canopy size. However, to avoid excessive vine water stress due to UTCC competition, irrigation was used as needed during water deficit periods. The experiment was conducted over three consecutive growing seasons from 2011 to 2013, in two experimental vineyards located in southern Uruguay (34° S 56° W). Treatments were: UTCC (full cover of the vineyard soil with tall fescue) versus conventional alleyway tall fescue with a 1.0 m-wide weed-free strip under the trellis. Deficit drip irrigation was provided at mid-day stem water potential (SWP) thresholds of −0.9 MPa early in the season and −1.1 MPa later. Shoot growth rate, SWP, berry size and berry composition (Brix, organic acids, total anthocyanins) were monitored over the season as well as final yield, cluster and pruning weights and wine sensory attributes. UTCC regulated vine vegetative growth and final canopy size and reduced bunch rot incidence as well as increasing fruit Brix and anthocyanin concentration in grapes and wines. Wines from UTCC treatments had an increased fruity aroma and overall aroma intensity levels and had distinctive sensory characteristics that exceeded those of H wines during overall palatability tests (liking tests).

A summary of the resource.
Elevated wine pH is one of the main problems affecting the quality of Vitis vinifera L. Tannat wines in Uruguay. High soil fertility, promoting vigorous vines, and high potassium availability result in high pH grapes and wine. The main objective of this work was to determine the effects of partial leaf and lateral shoot removal practices on the reduction of juice and wine pH over three growing seasons. Partial defoliation in shaded canopies significantly reduced grape potassium accumulation, malic to tartaric acid ratio, and wine pH without negatively affecting other quality parameters of soluble solids, organic acids, anthocyanins, and phenolics.
