Five C13-norisoprenoid glycosides, isolated from lulo (Solanum quitoense) leaves, were subjected to enzymatic hydrolysis with a commercial glucosidase (emulsin) and also with an enzymatic preparation having glycosidase activity, isolated from lulo fruit pulp. The volatile compounds generated after reaction were characterized by capillary GC and capillary GC&ndash;MS. Lulo fruit glycosidases were extracted by ammonium sulfate precipitation at pH 6.5 and subjected to fractionation by hydrophobic interaction chromatography (HIC) on phenyl Sepharose1 gel, and some of their properties were determined. As the result of enzymatic hydrolysis with exogenous enzymes (emulsin), glycosides isolated from lulo leaves produced only the aglycone (glycosidically bound volatile); in contrast, enzymatic hydrolysis of glycosides, using endogenous enzymes (lulo fruit glycosidases), generated additional volatile compounds, some recognisable as components of fruit flavour. The role of lulo leaf glycosides as flavour precursors was confirmed. [ReOCl2(PPh3)(3-(5-Nitrofuryl)acroleine semicarbazone)] crystallizes in the space group P-1 with a � 11.2334(2), b � 11.3040(2), c � 12.5040(2) A &deg; , &alpha; � 81.861(1), &beta; � 63.555(1), &gamma; � 83.626(1)&deg;, and Z � 2. The Re(V) ion is in a distorted octahedral environment, equatorially coordinated to a deprotonated semicarbazone molecule acting as a bidentate ligand through its carbonylic oxygen and azomethynic nitrogen atoms, to an oxo ligand and a chlorine atom. The six-fold coordination is completed by another chlorine atom and a triphenylphosphine ligand at the axial positions. Chagas&rsquo; disease, considered incurable, is a major third world parasitosis that affects millions of people in Latin America. Previous work has shown that ruthenium clotrimazole complexes are more active against Trypanosoma cruzi , causative agent of Chagas&rsquo; disease, than the corresponding free ligand. In this work, the synthesis and characterization of a series of new Ru(II) complexes with different antitrypanosomal active compounds is presented. Complexes of general formulae [RuIICl2(dmso)2L], where dmso�/ dimethylsulfoxide and L�/5-nitro-2-furaldehyde semicarbazone (L1), N4-n-butyl-5-nitro-2-furaldehyde semicarbazone (L2) or 3- (5-nitrofuryl)acroleine semicarbazone (L3), were prepared in good yields by reaction of [RuIICl2(dmso)4] with L in ethanol or toluene solutions. Complexes were characterized by elemental analyses and electronic, FTIR, 1H and 13C NMR spectroscopies. Crystal and molecular structures of [RuCl2(dmso)2L1] and [RuCl2(dmso)2L2] were determined by X-ray diffraction methods. In both crystals the ruthenium metal atom is in a quite similar elongated octahedral environment, equatorially coordinated to the semicarbazone molecule, acting as a bidentate ligand through its azomethynic nitrogen and carbonylic oxygen atoms. The sixfold coordination is completed with the sulfur atoms of two dimethylsulfoxide ligands at cis positions and two chlorine ions at the axial positions. The proposed formula for L3 complex was supported by FTIR, NMR and theoretical studies. NOE�/NMR experiments allowed to assign L3 spatial distribution in the complex. Word association, a projective qualitative technique, was applied to the development of olive oil-based and olive leaf extract-supplemented cosmetic creams. One hundred twenty female respondents presented with five stimulus terms &ndash; namely, &ldquo;nourishing cream,&rdquo; &ldquo;antiaging cream,&rdquo; &ldquo;olive oil-based cream,&rdquo; &ldquo;olive leaf extract-supplemented cream&rdquo; and &ldquo;antioxidant-rich cream&rdquo; &ndash; were asked to write down four images, associations, thoughts or feelings evoked by each of the different stimuli. Word associations made by the surveyed sample were then grouped into 14 categories. For each of the proposed stimuli, the frequency of mention of elicited words was analyzed according to and within each category. The use of word association enabled the identification of expected or desired sensory properties and efficacy of the studied cosmetic creams, as well as the concepts associated &ndash; in the minds of respondents &ndash; with each of the creams. The resulting information may be used to optimize the selection of new products and their denominations. These two molecules behave very differently from lower polyoxides, given that some isomers present unusual bonding. In the case of H2O6, the central OO bond is predicted, to be 1.909A&deg; , at the CCSD(T)/cc-pVTZ level. Two conformational isomers of H2O6 display nearly the same enthalpy of formation, but only one of them has extremely long OO bonds. For H2O7, the most stable isomer also has two unusually long OO bonds. At the CCSD(T) level, and after extrapolation to the complete basis set limit, this isomer is predicted to be 5.37 kcal mol&ndash;1 more stable than the one with short OO bonds, and the longest OO bond distance is expected to be close to 1.96A&deg; . Analysis of correlation energies indicated that the new isomers found for H2O6 and H2O7 are among the most strongly correlated molecules that can be formed with first-row atoms. In this study we investigated the formation of isomers of lycopene, phytoene, and phytofluene from tomato and their theoretical energy. The results indicated that certain (Z)-isomers are favored thermodynamically and/or kinetically over their (all-E)-counterparts. The relative percentages of (5Z)-lycopene in either thermodynamic or kinetic equilibria were approximately 33%, and those of (all-E)-lycopene were only approximately 22%. Most strikingly (15Z)-phytoene was the major isomer (&gt;90%) when the thermodynamic or the kinetic equilibria were reached. These observations can explain the high levels of lycopene (Z)-isomers found in humans and their rapid formations upon additions of oil to tomato products. In addition, the results can be useful to predict the isomeric forms of lycopene, phytoene, and phytofluene expected in foods as well as in plasma and tissues upon ingestion. In light of the data in the present study, the use of certain geometrical isomers of phytoene, phytofluene and lycopene on their own or as mixtures is recommended in future studies aimed at assessing their possible bioactivity. Nanoparticles functionalized with carbohydrates are one of the most powerful systems for studying carbohydrate&ndash;lectin interactions, because they mimic the multivalent presentation of carbohydrates encountered in nature, for example when viruses and bacteria bind to cells. On the basis of the model system glucose&ndash;Concanavalin A (ConA), we explore the application of Transient Magnetic Birefringence (TMB) to study these weak interactions, using glucose-functionalized colloidal magnetite nanoparticles (NPs) as probes. We demonstrate that the binding dynamics can be monitored and derive a model to obtain the apparent cooperativity. For our studies, we use nanoparticles of 6 and 8 nm in diameter. The ConA-generated response shows apparent cooperativity, due to the cross-linking of nanoparticles by the ConA tetramer which has four binding sites. Cooperativity is higher for 6 nm NPs, possibly due to a better accessibility of all four ConA binding sites on smaller NPs, enhancing cross-linking. For this system, we find a detection limit of 3&ndash;23 nM. The aim of this work was the development, synthesis and evaluation of a potential radiopharmaceutical enable the rapid and accurate diagnosis by scintigraphic images of fungal infection using caspofungin, a lipopeptide, radiolabelled with (99m)Tc. Caspofungin was radiolabeled with (99m)Tc-tricarbonyl precursor. The complex was assessed for in vitro stability, lipophilicity, plasma protein binding and plasma stability. Biological evaluation was conducted in four groups of CD1 female mice. G1 healthy animals, G2 was induced sterile inflammation with turpentine oil. G3 and G4 were infected with Candida albicans and Aspergillus Niger. Scintigraphicimages were acquired before sacrifice. The Caspofungin - tricarbonyl complex was obtained with RCP higher than 95%, it was stable in labeling milieu for at least 20 hours, and in plasma for 4 hours. Challenge with competitive agents showed no ligand exchange during 200 min. The product was well tolerated by mice and showed mainly hepatobiliar excretion. Lesion uptake was markedly higher in infected tissues than in sterile inflammation. Scintigraphic images clearly distinguished inflammation from infection. The high RCP yields and in vitro stability, the targeted biodistribution profile and good T/NT ratios, outlines this complex as a potential agent for rapid and specific diagnosis of infections caused by pathogenic yeasts. At day 4 PHT plasma concentrations were measured and, obtained several organs, brain, parotid gland, liver and duodenum in which were analyzed for the Pgp expression. Results: An induction of the expression of efflux transporter mediated by phenytoin in a concentration- and-time dependent manner was found when increasing oral and ip doses of phenytoin, One week after the interruption of ip treatment a basal expression of transporters was recovered. The presence of carbohydrate-binding proteins, namely lectins, &szlig;galactosidases and amylases, was determined in aqueous extracts of plants collected in Uruguay. Twenty-six extracts were prepared from 15 Uruguayan plants belonging to 12 Phanerogam families. Among them, 18 extracts caused hemagglutination (HAG) that was inhibited by mono- and disaccharides in 13 cases, indicating the presence of lectins. The other 8 extracts did not cause any HAG with the four systems used to detect HAG activity (rabbit and mouse red cells, trypsin-treated rabbit and mouse red cells). For the extracts prepared from Solanum commersonii, HAG activity and HAG inhibition were similar for those prepared from tubers, leaves and fruits, with the chitocompounds being responsible for all the inhibitions. Purification of the S. commersonii tuber lectin was carried out by affinity chromatography on asialofetuin-Sepharose, and SDS-PAGE under reducing conditions gave a single band of Mr of approximately 80 kDa. The monomer N-acetylglucosamine did not inhibit HAG induced by the purified lectin, but chitobiose inhibited HAG at 24 mM and chitotriose inhibited it at 1 mM. &szlig;-Galactosidase activity was detected in leaves and stems of Cayaponia martiana, and in seeds from Datura ferox. The aim of the present work was to apply survival analysis to estimate equivalent sweet concentration of low-calorie sweeteners in orange juice. Five low-calorie sweeteners were considered: sucralose and four samples of stevia. For each sweetener, a series of six orange juice samples with different sweetener concentration were prepared. Groups of 34 consumers evaluated the series of six pairs of samples, each composed of one reference sample (sweetened with 10% sucrose) and one sample with low-calorie sweetener. Consumers were asked to complete a paired-comparison test and to indicate which of the samples was sweeter. The elicited data were analyzed using survival analysis. For each sweetener, the percentage of assessors perceiving the sample with the low-calorie sweetener as sweeter than the sample with sucrose as a function of sweetener concentration was determined. Equivalent sweet concentrations were determined for each sweetener and compared with estimations obtained using logistic regression and published data. The influence of two sample presentation and instruction delivery protocols on the results of consumer-based sensory evaluations was studied. In two independent experiments, five peach jams and four Colonia-type cheese (from the Colonia region in Uruguay) were taste-tested. In the first experiment, one group of 54 consumers used the sequential monadic (SM) and the other group of 54 consumers used the rank-rating (RR) sample presentation protocols for the jam sample evaluation. In the second experiment, one group of 54 consumers used the SM and the other group of 54 consumers used the RR sample presentation protocols for the cheese sample evaluation. All samples were evaluated using hedonic scales and CATA (check-all-that-apply) questions. Overall liking scores were found not to depend on the sample presentation protocol. Although the protocol was found to affect the choice and number of attributes listed in the CATA question used by respondents to differentiate the samples, the spatial configurations of samples derived by multiple factor analysis showed a high degree of correlation between the two protocols for most of the CATA attributes. Based on these results, the SM and RR protocols were found to be equally sensitive, for the products tested, in the conditions of this study. The melanoma targeting peptides (Ala-triazol)Ac-Re(Arg(11))CCMSH and N4-CO-Re(Arg(11))CCMSH were radiolabeled with [(99m)Tc(CO)3](+) and [(99m)TcO2](+), respectively, and examined for in vitro cell binding, in vivo biodistribution and imaging properties. The (Ala-triazol)Ac-Re(Arg(11))CCMSH and N4-CO-Re(Arg(11))CCMSH were synthesized as protected peptides on resin followed by rhenium cyclization with [(C6H5)3P]2ReOCl3 in DMF. The peptides were labeled with (99m)Tc and examined for radiochemical stability and melanoma cell binding. In vivo biodistribution and SPECT/CT imaging studies were performed in B16/F1 melanoma tumor bearing C57 mice. (99m)Tc(CO)3-(Ala-Triazol)Ac- Re(Arg(11))CCMSH and (99m)TcO2-N4-CO-Re(Arg(11))CCMSH were stable and internalized in B16/F1 melanoma cells upon binding. In vivo biodistribution studies revealed that tumor uptake of (99m)Tc(CO)3-(Ala-Triazol)Ac-Re(Arg(11))CCMSH was 6.08&plusmn;1.06% ID/g and 7.05&plusmn;1.48% ID/g at 2 h and 4 h post injection, respectively. Tumor uptake of (99m)TcO2-N4-CORe(Arg(11))CCMSH was 7.54&plusmn;1.82% ID/g and 2.28&plusmn;0.22% ID/g at 1 h and 2 h post injection, respectively.
