La falta de tratamientos eficientes para gliomas de alto grado, como lo es el GBM, conduce a la necesidad de la I+D en nuevas estrategias farmacológicas eficaces. En esa línea, en el terreno de la terapia oncológica se ha experimentado una rápida evolución hacia la llamada terapia dirigida a blancos moleculares (en inglés, targeted therapy). Esta modalidad de tratamiento utiliza fármacos que van específicamente dirigidos a dianas moleculares que han demostrado desempeñar un rol crucial en la supervivencia, proliferación o capacidad invasiva de las células tumorales. En el caso de GBM, los receptores celulares con actividad tirosina quinasa (EGFR, VEGFR, PDGFR, entre otros) son claros ejemplos de dianas prometedoras. Por otro lado, actualmente se encuentra en estudio la terapia para GBM por captura neutrónica de boro (BNCT, del inglés Boron Neutron Capture Therapy) técnica que involucra radioterapia in situ. Se basa en la inusual capacidad del nucleido 10B (un isotopo estable de boro) en capturar neutrones térmicos de baja energía produciéndose partículas alfa y núcleos de litio los que depositarán la energía en la célula tumoral. En la presente tesis se abordó la modificación estructural de compuestos con bioactividad reconocida, los fármacos erlotinib y sunitinib, mediante hibridación molecular con entidades con alto contenido de boro, entre ellos carboranos y metalacarboranos. El arroz (Oryza sativa) es el cereal cultivado más importante a nivel mundial (672×106 toneladas /año). Su destino prioritario es la alimentación humana como arroz blanco. El Uruguay es un exportador neto de arroz blanco, generando anualmente 130 mil toneladas de salvado de arroz como residuo. Aunque el 10% de este salvado tiene como destino la producción de aceite y una parte se utiliza para la alimentación animal, es de gran interés estudiar su uso potencial en la alimentación humana. Las enfermedades no transmisibles son responsables de la muerte de 40 millones de personas en el mundo cada año. Uno de los factores de riesgo más relevante son el sobrepeso y la obesidad. Estos están directamente relacionados un mayor riesgo de padecer diabetes tipo 2, enfermedades cardiovasculares, entre otras. Dados los actuales hábitos alimenticios, de alto consumo de azúcares libres se prevé un incremento de la incidencia global de estas patologías en los próximos años. La presente tesis doctoral se enfoca en la posibilidad de aprovechar una materia prima sub-valorada como el salvado de arroz, con el objetivo de desarrollar nuevos ingredientes alimentarios para el consumo humano que contribuyan a la reducción del riesgo de padecer estas patologías crónicas no trasmisibles. Para ello, se desarrolló un extracto rico en antioxidantes polifenólicos cuyas condiciones de producción fueron optimizadas a través de un diseño experimental central compuesto (CCD) 2k = 23 = 16 tratamientos, con 3 variables independientes (% de etanol, tiempo y temperatura) a 3 niveles (-1, 0 y +1) y utilizando tecnologías simples y fácilmente escalables. El residuo insoluble generado tras la obtención de dicho extracto fue caracterizado por análisis de su composición nutricional y capacidad antioxidante demostrando ser una fracción rica en fibra dietética antioxidante. x Las condiciones de extracción optimizadas (maceración simple con 35% de etanol en la solución extractora, a 20°C por 105 minutos) permitió obtener un extracto de salvado de arroz principalmente rico en proteínas (12,74 g/100g de MS) y en carbohidratos (77,0 g/100g MS). Bajo dichas condiciones, se recuperó el total de fibra dietética soluble (2,23 ±0,10 g/100g MS) que compone el salvado de arroz; así como, un contenido de lípidos totales de 3,38 ±0,07 g/100g MS y de cenizas de 4,82±0,06 g/100g MS. El extracto antioxidante resultó ser rico en polifenoles (71,42±1,60 mg de GAE/ g de extracto) y presentó elevada capacidad antioxidante (451±14 μmol de TE/g muestra frente a radical ABTS.+ y 912±31 μmol de TE/g muestra frente a radicales peroxilos). El análisis cromatográfico por UHPLC indicó que los componentes fenólicos mayoritarios presentes en extracto son ácido clorogénico (44, 84 ± 2, 25 mg / 100 g de materia seca) y ácido ferúlico (12,44 ± 0,60 mg / 100 g de materia seca). El extracto presentó valores IC50 de 0,14 ± 0,01 y 4,50 ± 0,08 mg/mL contra radicales peroxilos y ABTS.+, respectivamente. Para su mejor caracterización el extracto seleccionado se fraccionó mediante ultrafiltración empleando una membrana con tamaño de poro uniforme de 3 kDa. Aplicando este procedimiento se encontró que el extracto está constituido por un 68% de compuestos con tamaño molecular mayor a 3 kDa (FAPM) y 32% de compuestos con tamaño menor a este valor (FBPM). El análisis cromatográfico de ambas fracciones indica que la FAPM contiene principalmente ácido ferúlico mientras que el compuesto fenólico mayoritario que compone a la FBPM es ácido clorogénico. El estudio de bioaccesibilidad de los componentes bioactivos que forman el extracto antioxidante de salvado de arroz llevado a cabo por simulación gastrointestinal in vitro indicó una pérdida significativa (p6g/100g de galleta). Se utilizó stevia como edulcorante no nutritivo resultando ser un alimento que no contiene azúcar añadido y; por lo tanto, presenta potencial para reducir el riesgo de diabetes. El alimento presentó una capacidad antioxidante de 3653 ± 150 μmol TE/ porción considerando los compuestos xii extraíbles e hidrolizables presentes y de 1125 ± 73 μmol de TE/porción de galleta (30g) por el método de ABTS-QUENCHER. El % de bioaccesibilidad de los polifenoles presentes en las galletas fue de 17% respecto a los polifenoles totales presentes en una porción de 30g de alimento (33,07±1,59 mgGAE/porción). El promedio de aceptabilidad general de las nuevas galletas libres de gluten, fuente de fibra dietética y antioxidantes fue de 51% (88 evaluaciones). La formulación debe considerarse un prototipo inicial sobre el cual debe trabajarse para obtener un alimento de óptima calidad sensorial. En conclusión, se puede afirmar con un 95% de confianza que es posible obtener 2 nuevos ingredientes alimentarios de calidad nutricional y funcional a partir de salvado de arroz: extracto antioxidante (fracción soluble) y la fracción insoluble rico en fibra dietética antioxidante. El proceso de extracción propuesto es de gran simplicidad y bajo costo de inversión por lo que se alinean al concepto de “extracción verde” destacándose el hecho de no generar nuevos residuos de producción siendo innovadora, eco-sostenible y de promisoria implementación en la industria uruguaya. Las galletas elaboradas permiten afirmar que se ha logrado un alimento tradicional de gran consumo regional que reúne varias de las condiciones recomendadas sobre una dieta saludable por la OMS (2014 y 2015) para la reducción del riesgo de padecer enfermedades crónicas no transmisibles en un solo alimento. Las galletas no contienen gluten, ni azúcar añadido (endulzado con stevia como edulcorante no nutritivo), son fuente de fibra alimentaria antioxidante (3 g /porción) y presentan capacidad antioxidante siendo un primer prototipo de alimento funcional que valida la recuperación del salvado de arroz. The present study aimed to obtain novel information regarding the pathways of a polyphenol-rich coffee silverskin extract (CSE) for inhibiting the formation of advanced glycation products (AGE). Chlorogenic acid (CGA) is the main phenol present in coffee beans. The contribution of CGA to the antiglycoxidative properties of CSE was also evaluated by analysis of CGA standard. The CGA (90%) and CSE (49%) significantly inhibited (p < 0.001) the formation of fluorescent AGE in a protein–methylglyoxal (MGO) model system (37 °C for 96 h). The lack of MGO-derivative AGE in the presence of CGA and CSE was verified using liquid chromatography–tandem mass spectrometry techniques, which evidenced the almost complete absence of arginine adducts after 24 h of reaction. The inhibitory effect of polyphenols present in CSE may be associated to its carbonyl trapping capacity as well as to its ability to react with side-chains of protein amino residues blocking the reaction sites. In conclusion, aqueous CSE can be considered a natural source of various inhibitors of in vitro formation of AGE acting by different pathways. The objective measure of grape quality is an important requirement to enhance wine quality. Particularly, the quantitation of glycosylated aroma compounds could be a useful index for winemakers. In the present work, we evaluated the bound compounds profile on Tannat grapes, and the potential of near infrared (NIR) spectroscopy coupled to chemometric techniques to determine the concentration of the different glycosylated aroma compounds present in this grape variety. The results showed phenols and norisoprenoids as the quantitative most important group of compounds, representing almost 80 % of the total glycosidic components present in Tannat grapes. The glycosidic form of ethyl-β-(4-hydroxy-3- methoxy-phenyl)-propionate was identified for the first time in grapes. The results suggested NIR technique as a screening tool for a fast analysis of the grape aroma potential. The application of such methodology would be ideal for fast grape screening purposes, contributing to the winemaking decisions, classification of the fruit, and perhaps maturity monitoring. Bacterial canker caused by Clavibacter michiganensis subspecies michiganensis (Cmm) has been a threat to tomato production in Uruguay for several years. In this study, 39 Cmm strains were collected from several outbreaks and production areas in Uruguay, and were identified by molecular assays and pathogenicity tests on a susceptible cultivar of tomato. In addition, a TaqMan assay targeting a putative two-component system sensor kinase gene demonstrated good specificity with all strains tested and gave no false negative results. The first epidemiological study of Cmm in this country was carried out in order to elucidate the origin of outbreaks and sources of infection and dissemination of the pathogen. Strains from Uruguay showed high genetic diversity based on a Multi Locus Sequence Typing analysis of five housekeeping genes. This approach revealed 36 sequence types (STs) within a worldwide collection of 108 Cmm strains. Ten STs correspond to strains solely isolated in Uruguay, including eight novel STs for the subspecies michiganensis. This high diversity reflects the introduction of new strains from different origins that most probably results from seed importation. This study provides relevant information about the distribution and origin of strains causing bacterial canker in Uruguay and will pave the way for the establishment of preventive measures to control the disease. Actinomycetes are generally recognized as a diverse group of gram-positive, myceliumforming, soil bacteria that play an important role in mineralization processes and plant health, being Streptomyces the most well-known genus from this group. Although plant pathogenicity is a rare attribute in this genus, some species have significant impact worldwide due to their ability to cause important crop diseases such as potato common scab (CS). In this work, an integrative approach was applied to investigate the pathogenic potential of Streptomyces spp. isolates obtained from a local collection of actinomycetes isolated from potato fields. Secretion of phytotoxic compounds was verified in most pathogenic strains from our collection (27 out of 29), and we followed metabolomic analysis to investigate those phytotoxins. We first evaluated the production of the known phytotoxins thaxtomin A (TXT) and desmethylmensacarcin (DMSN) in phytotoxic Streptomyces spp. by HPLC analysis, resulting in 17 TXT and 6 DMSN producers. In addition, NMR-based metabolomic models were able to classify strains according to their phytotoxicity, and metabolomic data was also used to infer chemotaxonomy within pathogenic species. A correlation between phylogeny and the production of distinct phytotoxins was found, supporting the idea that there are “species specific” metabolites produced by this genus. The recently discovered polyketide DMSN was associated unequivocally with S. niveiscabiei strains and was not produced by other species in the growth conditions employed. Two S. niveiscabiei and two S. puniciscabiei phytotoxic strains that did not produce TXT nor DMSN suggest the production of other kind of metabolites involved in phytotoxicity, and allowed the prioritization of these strains for further chemical studies. Indeed, we found two S. niveiscabiei strains whose supernatants were not phytotoxic in the radish assay, suggesting other pathogenic mechanisms involved. We believe our work will be useful to help understand relations between metabolites and phylogenetic clades within actinomycetes. The increasing concern about accidents involving radioactive materials has led to the search of alternative methods to quickly identify and quantify radionuclides in workers and in the population. One of the options to face up an eventual demand for mass monitoring of internal contamination is the use of a nuclear medicine diagnostic equipment known as gamma camera, a device used to scan patients who have been administered specific amounts of radioactive materials for medical purposes. The components of a gamma camera are the collimator, detector crystal, photomultiplier tube array, position logic circuits, and the data analysis computer. A Thallium-activated Sodium Iodide [NaI (Tl)] detector crystal is generally used in Gamma cameras. This is due to this crystal's optimal detection efficiency for the gamma ray energies of radionuclide emission common to Nuclear Medicine. Although the gamma camera is used for image diagnosis it can be calibrated with anthropomorphic phantoms or point sources for the quantification of radionuclide activities in the human body. Therefore, a wide range of radioactive materials can be detected by means of this technique. This work presents a protocol for the calibration of gamma cameras for such application. In order to evaluate the suitability of this type of equipment, a gamma camera available in a public hospital located in Rio de Janeiro was calibrated for the in vivo measurement of 131I. The calibration includes the determination of detection efficiencies and minimum detectable activities for each radionuclide. The results show that the gamma camera presents enough sensitivity to detect activity levels corresponding to effective doses bellow 1 mSv. The protocol is the basis to establish a network of nuclear medicine centers, located in public hospitals in 8 countries of South America (Argentina, Brazil, Colombia, Cuba, Chile, Mexico, Peru, Uruguay) and in Spain that could be requested to collaborate in remediation actions in the event of an accident involving incorporation of radioactive materials. En esta tesis se prepararon y caracterizaron diferentes materiales de carbono para ser usados como electrodos de supercondensadores. Los supercondensadores son un tipo de condensador eléctrico con una capacidad eléctrica muy superior a cualquier otro tipo de condensador. En su constitución básica, están formados por dos electrodos (separados por un aislante eléctrico) y un electrolito iónico. Los desafíos actuales en el desarrollo de supercondensadores se focalizan en aumentar la densidad de energía del dispositivo. Por el lado de los materiales de electrodo, esto se puede lograr con el desarrollo de materiales que tengan una elevada capacidad eléctrica específica. El trabajo experimental de esta tesis se focalizó en la preparación y caracterización de carbones en polvo activados, y monolitos de carbón obtenidos a partir de diferentes cortes (longitudinal al eje de crecimiento del tronco del árbol y transversal al eje del tronco) de madera Eucaliptus Grandis. Los materiales fueron preparados mediantes diferentes métodos y condiciones de activación (generación de porosidad) y fueron caracterizados mediante análisis textural, análisis elemental, Desorción Térmica Programada (TPD), difracción de rayos X (DRX), microscopía electrónica de barrido (SEM), medidas de conductividad eléctrica y electroquímicas. Las caracterizaciones electroquímicas (curvas galvanostáticas de carga y descarga, voltametría cíclica e impedancia electroquímica) se llevaron a cabo utilizando una celda de laboratorio simétrica tipo Swagelok usando dos tipos de electrolitos: (i) solución acuosa H2SO4 2M (electrolito ácido) y (ii) Et4NBF4 disuelto en acetonitrilo 1M (electrolito orgánico). IV Los resultados obtenidos para los carbones en polvo indican que estos tienen áreas superficiales que varían en un amplio rango dependiendo del método y condiciones de preparación. De acuerdo con los resultados del TPD, el contenido de grupos funcionales oxigenados superficiales de los carbones varía también según el método y condiciones de preparación. La conductividad eléctrica de los materiales depende marcadamente de la temperatura máxima de preparación siendo prácticamente independiente del método o agente activante utilizado. Se determinó la existencia de una correlación directa de la capacidad eléctrica específica de los materiales con el área superficial y el contenido de grupos funcionales oxigenados superficiales determinados mediante TPD. Los mejores resultados en cuanto a capacidad eléctrica específica en electrolito ácido se determinó para el carbón activado con ZnCl2 a 900 °C seguido de una posterior oxidación con ácido nítrico. Se comprobó que la energía acumulada para un mismo carbón es mayor en el electrolito orgánico que en el electrolito ácido. El análisis de los monolitos demuestra que existe una incidencia directa de la anisotropía de la madera en las propiedades texturales, eléctricas y electroquímicas de los monolitos de carbón obtenidos. Los monolitos obtenidos a partir del corte transversal presentaron una estructura más abierta con mayor desarrollo de porosidad (la más alta obtenida para los activados con CO2), mayor capacidad eléctrica específica gravimétrica, mayor energía gravimétrica y mayor conductividad eléctrica. Sin embargo no se encontraron diferencias sustanciales en la capacidad y energía volumétrica para los dos tipos de monolitos estudiados. Los mejores resultados de capacidad eléctrica y energía acumulada obtenidos para los materiales de carbón preparados en esta tesis fueron mejores a los obtenidos para un carbón comercial de referencia y otros materiales similares reportados en la bibliografía. The work reports the growth of single BI3 crystals with platelets habit. Platelets were grown by physical vapor deposition (PVD) in a high vacuum atmosphere and with argon, polymer or iodine as additives. Crystals grew in the zone of maximum temperature gradient, perpendicular to the ampoule wall. Crystals grown with argon as additive show a very shining surface, have hexagonal (0 0 l) faces, sizes up to 20 x 10 mm2 and thicknesses up to 100 μm. They were characterized by optical microscopy and scanning electron microscopy (SEM). Dendritic-like structures were found to be their main surface defect. SEM indicates that they grow from the staking of hexagonal unities. Electrical properties of the crystals grown under different growth conditions were determined. Resistivities up to 2 x 1012 Ωcm (the best reported value for monocrystals of this material) were obtained. X-ray response was measured by irradiation of the platelets with a 241Am source of 3.5 mR/h. A comparison of results according to the growth conditions was made. Properties of the crystals grown by this method are compared with the ones measured for others previously grown from the melt. Also, results for bismuth tri-iodide platelets are compared with the ones obtained for mercuric and lead iodide platelets. Biocarbon monoliths were obtained from Eucalyptus grandis and the influence of wood anisotropy on the electrical and electrochemical performance as supercapacitor electrodes was studied. They were produced from wood pieces cut along the transversal and longitudinal direction of the tree trunk, followed by pyrolysis and, for some of them, also by activation with CO2. Monoliths with drilled channels were also obtained. All the monoliths were characterized by SEM, nitrogen adsorption/desorption isotherms, electrical conductivity measurements and electrochemical measurements, the latter in 2M aqueous H2SO4 electrolyte. Electrical conductivity and specific capacitance are higher for the transversal carbon monoliths than for the longitudinal ones. The electrical conductivity reaches values up to 27 S cm−1 for the transversal monolith. The specific capacitance reaches values up to 260 F g−1 for the transversal monolith that was activated and drilled. However, the highest volumetric capacitance, of 90 F cm−3, is found for the longitudinal monolith that was activated and non-drilled. The energy density and power density, both referred to gravimetric and volumetric magnitudes, reach values as high as 36 Wh kg−1 and 12 Wh L−1, and 2181 W kg−1 and 783 W L−1, respectively. There are three basic steps to activated carbon fibers (ACF) manufacturing, from PAN fiber: oxidation/stabilization, carbonization and activation. Carbon material, specially ACF is a very attractive material to be used as supercapacitor electrode. The literature describes carbon material surface chemistry importance for supercapacitors application, mainly nitrogen groups by N-doping. Oxidation/stabilization is an important non-explored factor that influence the surface chemical functionality. This work describes the influence of oxidation/stabilization process on ACF production, from textile PAN fiber, and the non-doping nitrogen surface chemistry characteristic caused by the oxidation process design. Its textural, structural and surface was evaluated for supercapacitor electrode. The results show that the oxidation degree can be used as a mechanism of textural and surface chemistry control. The surface chemistry is the key of this work, different oxidation conditions can produce nitrogen compounds that help to increase specific capacitance. Reduction and covalent immobilization of Kluyveromyces lactis �-galactosidase through disulfide bonds onto thiolsulfinate-agarose was performed using two fixed-bed mini-reactors connected in series, one packed with thiopropyl-agarose (a solid phase reducing agent) and the other with thiolsulfinate-agarose (a thiol-reactive support). With the aim of optimizing the whole process, two reactor systemswere assessed. In System I, the percolate from thiopropyl-agarose containing the reduced enzyme was re-circulated through the thiolsulfinate-agarose reactor alone. In System II, re-circulation was performed through both the reactors, improving the immobilization yield from 17% (System I) to 42% and the expressed activity from 25% (System I) to 56%. When the bio-reactor achieved with System II was fed with skimmed milk at 22 ◦C at a flow rate of 48 ml/h, steady state lactose hydrolysis reached 80%. In addition, it could be reused four times without losing its lactose hydrolysis capacity. Six new polynuclear coordination compounds based on 3d transition metal ions (Cu(II), Ni(II)) and Zn(II) with the flexible dicarboxylate compound N-methyliminodipropionic acid (H2midp) were prepared and characterized by IR spectroscopy, elemental analysis and single-crystal X-ray diffraction. Four of these compounds are 1D coordination polymers, namely [Cu(midp)(H2O)] (1), [Cu3(midp)2(H2O)4](NO3)2⋅6H2O (2) [Cu3(midp)2(H2O)4] (CF3SO3)2⋅4H2O (3) and [NaZn(Hmidp)2](CF3SO3) (6). The structural analysis revealed that the chain structure of 1 is composed of [Cu(midp)(H2O)] units connected by the free carboxylato oxygen atom of neighbor units, in an alternate fashion. In 2 and 3, on the other hand, there are two [Cu(midp)H2O] units, connected by a central Cu(II) ion coordinating again to the free oxygen atom of the carboxylato groups. The Zn(II) compound is heteronuclear and the complex adopts a 1D cationic polymeric structure with zinc and sodium ions connected by two Hmidp– anions. The two other compounds are dinuclear, [Cu2(Hmidp)2(μ-H2O)2(H2O)2](CF3SO3)2⋅6H2O (4) and ([Ni2(midp)2(H2O)4] (5). Additionally, a Co(II) and a Cu(II) coordination compound were obtained with the same ligand and characterized by IR spectroscopy, thermal analysis and elemental analysis including metal quantification. For the copper compound the formula [Cu(Hmidp)](ClO4)⋅3H2O (7) is proposed while for the Co (II) compound (8) the proposed formula is [Co(midp)(H2O)2]. Solution studies of the ligand–M(II) systems (M (II) = Mn, Fe, Co, Ni, Cu, Zn, Cd and Pb) were also carried out in order to help the selection of the best synthetic conditions and also to complement the structural knowledge in the exploration of the potential formation of coordination polymers in these systems. The aetiological agent of cystic hydatid disease, the platyhelminth parasite Echinococcus granulosus, undergoes a series of metamorphic events during its complex life cycle. One of its developmental stages, the protoscolex, shows a remarkable degree of heterogeneous morphogenesis, being able to develop either into the vesicular or strobilar direction. Another level of complexity is added by the existence of genotypes or strains that differ in the range of intermediate hosts where they can develop and form fertile cysts. These features make E. granulosus an interesting model for developmental studies. Hence, we focused on the study of the regulation of gene expression by microRNAs (miRNAs), one of the key mechanisms that control development in metazoans and plants and which has not been analysed in E. granulosus yet. In this study, we cloned 38 distinct miRNAs, including four candidate new miRNAs that seem to be specific to Echinococcus spp. Thirty-four cloned sequences were orthologous to miRNAs already described in other organisms and were grouped in 16 metazoan miRNA families, some of them known for their role in the development of other organisms. The expression of some of the cloned miRNAs differs according to the parasite life cycle stage analysed, showing differential developmental expression. We did not detect differences in the expression of the analysed miRNAs between protoscoleces of two parasite genotypes. This work sets the scene for the study of gene regulation mediated by miRNAs in E. granulosus and provides a new approach to study the molecules involved in its developmental plasticity and intermediate host specificity. Understanding the developmental processes of E. granulosus may help to find new strategies for the control of cystic hydatid disease, caused by the metacestode stage of the parasite.
