The interest in non-conventional yeasts for innovation and a unique selling feature in beer fermentation is increasing. This work studied the autochthonous Saccharomyces and non Saccharomyces yeasts, isolated from various food sources, with the ability to modify and improve the fermentative and aromatic profiles during alcoholic fermentation. The ability to ferment maltose and produce desirable aroma compounds were considered as the key characters for the screening selection. A synthetic beer wort was developed for this purpose, to simulate beer wort composition.</p> <p>A total of forty-seven yeast strains belonging to different genera were analysed according to their fermentation profile, volatile compounds production and sensory analysis. Three native strains of Saccharomyces cerevisiae, Zygoascus meyerae and Pichia anomala were selected to evaluate their aromatic profile in single and mixed fermentations. The strains produced 4-vinylguaiacol, &beta;-phenylethyl alcohol, and isoamyl alcohol at levels significantly above the sensory threshold, making them interesting for wheat and blond craft beer styles. Different commercial Saccharomyces cerevisiae strains have been applied at the winemaking level, trying to establish a dominant population of selected strains from the start of fermentation and ensuring the complete consumption of sugars. Although a large population of active yeast cells can be introduced in the inoculated wines, resulting in a complete fermentation, this does not necessarily mean an improvement of the sensory characteristics of the wines. The impact of the size of the inocula in wine quality parameters has been very little studied, and in no case the nutrient balance of the grape must utilized was taken into account. In this work we present results obtained for wine aroma compounds at three inoculum levels (104, 105 and 106 cells/mL), and two different yeast assimilable nitrogen (YAN) in a white grape must, using two S. cerevisiae strains commonly used for winemaking. A significant effect in the final concentrations of higher alcohols, esters, fatty acids, free monoterpenes and lactones was attributed to the size of inoculum in both strains but not in an easily predictable way. However, a consistent increase of desired aroma compounds (esters, lactones and free monoterpenes), and a decrease of less desired compounds for white wine (higher alcohols and medium chain fatty acids), was shown at inoculum sizes of 105 cells/mL for both strains in real winemaking conditions. In a discriminant analysis six aroma compounds discriminate the three inoculum sizes for all wine samples: 1,8-terpine, hodiol I (trans-3,7-dimethyl-1,5-octadiene-3,7-diol), isobutyl alcohol, iso C4 acid, ethyl C6 ester and C8 acid. Macrosiphum euphorbiae, also known as the potato aphid, is a potato virus vector, associated with decreased tuber production. Feeding, toxicity, and reproductive effects of the major potato glycoalkaloids (&alpha;-chaconine and &alpha;-solanine), their common aglycone (solanidine), and a nonpotato glycoalkaloid (&alpha;-tomatine) and its aglycone (tomatidine) onM. euphorbiae were studied. Common Solanum glycosides were tested against a non-Solanum pest, the greenbug, Schizaphis graminum, fed by artificial diet supplemented with the tested compounds. In contrast with the potato aphid Macrosiphum euphorbiae, S. graminum is sensitive to both potato and tomato glycoalkaloids, as well as to all the different Solanum steroidal glycosides tested. S. graminum was less sensitive to glycoalkaloids than to the steroidal glycosides. Laxumin A (LC50 4.3 &mu;M) and laxumin B (LC50 6.1 &mu;M), isolated from Solanum laxum, were the most active of the tested compounds. Ready-to-eat fruit and vegetables comprise a group of increasingly demanded value-added products. Broccoli is a highly perishable vegetable with unique nutritional characteristics. Development of minimally processed broccoli products demands varietal-specific knowledge as to which are the packaging conditions that preserve quality throughout shelf life. &lsquo;Legacy&rsquo; cultivar broccoli florets were washed, disinfected, packaged in polypropylene and stored at 4, 8 and 15 &deg;C for 21 d. Weight loss, internal atmosphere composition, respiration rate, color, texture, glucosinolate content, antioxidant capacity (AOC) and sensory attributes were evaluated throughout storage time. Results showed that 4 &deg;C helped preserve sensory quality, texture, total glucosinolate content and AOC for 21 d. Temperature fluctuations reaching 15 &deg;C resulted in loss of total glucosinolate content and unacceptable sensory quality. MAP helped mitigate temperature effects, especially at 8 &deg;C. MAP is therefore an appropriate technology which can be applied to extend the shelf-life of ready-to-eat broccoli florets. Chagas disease, caused by the protozoan parasite Trypanosoma cruzi, is a major health problem worldwide. In this work, we report the development of palladium and platinum metal complexes with 5-nitrofuryl-containing thiosemicarbazones (L) as bioactive ligands against T. cruzi and PTA (1,3,5-triaza-7-phosphaadamantane) as co-ligand. Eight new complexes of the formula [MCl(L)(PTA)] with M = Pd or Pt were synthesized and fully characterized. Most complexes showed similar activities against T. cruzi to those of the corresponding free thiosemicarbazone ligands. No significant differences between palladium and platinum complexes were observed. Metal compounds with the phenylthiosemicarbazone derivative were the most active ones (IC50 = 9.84&thinsp;&plusmn;&thinsp;0.32 and 4.94&thinsp;&plusmn;&thinsp;0.24 &mu;M for Pd2+ and Pt2+, respectively). The prepared complexes were not toxic on mammalian cells, showing selective indexes of more than 10&ndash;20. The ability of the complexes to be reduced in the parasite, which leads to toxic free radical species, was confirmed by the detection of OH&middot; and nitroanion free radical species by ESR spectroscopy experiments. Gel electrophoresis and fluorescence experiments were consistent with an intercalating-like mode of DNA interaction for the complexes, but DNA interaction does not seem to be the main mechanism of anti T. cruzi action for these compounds. The results obtained show that complexation of the bioactive ligands with the selected metals is a valid strategy to obtain improved metal-based antiparasitic compounds. In this work we studied the electrochemical performance of Ba0.5Sr0.5Fe0.8Cu0.2O3-&delta; (BSFCu) as cathode for Intermediate Temperature Solid Oxide Fuel Cells (IT-SOFC) with Ce0.9Gd0.1O1.95 (CGO) electrolyte and the effect of the symmetric cell preparation temperature on the oxygen reduction reaction (ORR) activity. Symmetrical cells with the configuration BSFCu/CGO/BSFCu were prepared at 900 &deg;C, 950 &deg;C and 1000 &deg;C to perform the electrochemical characterization in the 500&ndash;700 &deg;C temperature range. The resultant area specific resistance (ASR) of the cells with different preparation temperatures followed the tendency: ASR900&deg;C &lt; ASR950&deg;C &lt; ASR1000&deg;C. The symmetric cell constructed at 900 &deg;C showed ASR values of 0.18, 0.078 and 0.035 &Omega; cm2 at 600, 650 and 700 &deg;C respectively, which demonstrated superior electrochemical activities than previous reports. Additional, X-ray diffraction (XRD), scanning and transmission electron microscopies (SEM and TEM) techniques were used to characterize the microstructure of the original and fired BSFCu materials and correlate it with the cell preparation temperature. Four Deep Eutectic Solvents (DES) based on choline chloride (ChCl) and different hydrogen bond donors (1,4-butanediol, glycerol, isosorbide, and urea) were tested as reaction medium for the immobilized Candida antarctica B (iCALB) lipase-catalyzed esterification of fatty acids with n-butanol. Although the strong hydrogen bonds between DES components were expected to lower their reactivity, all except urea, competed in the esterification reaction. To study the effect of acyl group donor and water, different chain length of saturated fatty acids (ranging from 6:0 to 18:0) and water content (ranging from 0, 1, 2 or 3 molar ratio of water/ChCl) were tested in the lipase-catalyzed esterification with n-butanol in ChCl:urea (1:2, molar ratio). When the incubation was performed in the absence of water, the shorter the fatty acid chain length, the higher the conversion. Upon water addition, a drastic increase in the esterification rate of all the fatty acids was observed and, conversely to what occurred in the anhydrous media, the longer the fatty acid chain length, the higher the conversion. Results suggest that water concentration in the DES medium possesses a multifactorial effect, as it influences the enzyme activity, the solvent viscosity, the DES structure, and the interaction between DES components, substrates, and enzymes, thus determining the efficiency and yield of the esterification reaction. The present study investigated the effects of thinking style, health-related consumption purpose and their interaction on food choice, using yogurt packages (plain and strawberry). For each yogurt, four 2-level label variables were used in a discrete choice experiment: image (simple vs. complex), taste description (present vs. absent), probiotics claim (detailed vs. shallow) and nutritional information (healthful vs. regular). The choice experiment was completed by 148 female study participants while their eye movements were recorded using a remote eye-tracker. The expanded Cognitive Reflection Test (CRT) was used to evaluate their thinking style. The participants&rsquo; self-reports on the purpose of their yogurt consumption were used to evaluate whether or not the purpose of product consumption was associated with health, per participant. Results from the choice task showed that both CRT and health-related consumption purpose influenced the effect of image and nutritional information variables. For the participants&rsquo; visual attention, the CRT significantly influenced all four label variables, while health-related consumption purpose only significantly influenced the image variable. Visual attention towards image decreased when the purpose of product consumption was associated with health. In addition, the attention towards and the importance attached to nutritional information increased with increasing CRT scores, but these effects were diminished when the participants&rsquo; purpose of product consumption was related to health. The aim of this work was to assess the influence of treatment with U-Caspofungin, on the quality of diagnostic scintigraphic images of induced lesions in nude mice undergoing both bacterial and fungal infections and to determine the level of specificity of 99mTc-tricarbonyl-Caspofungin to discriminate between fungal or bacterial infections. In vitro studies on the behaviour of the 99mTc-tricarbonyl-Caspofungin complex binding percentage at different yeast concentrations of Candida albicans or Staphyolococcus aureus were determined. The incubation was performed with and without U-Caspofungin. In vivo evaluation was performed of 6 groups of athymic mice: sham, inflammation (LPS), fungal infection with Candida albicans (CA) and bacterial infection with Staphylocuccus aureus (SA). In vitro studies showed that the uptake of the complex by both yeasts and bacteria, depends on the concentration of colony forming units (cfu), and that this uptake is favoured by the presence of UCaspofungin that increases the membrane permeability to the 99mTc-tricarbonyl-Caspofungin complex. In vivo evaluation showed low uptake in sterile inflammation model and moderate to high uptake in infections models both treated or not with U-Caspofungin. The results of biodistributions were compatible with scintigraphic images. Conclusion: The uptake of the complex depends on the load of microorganisms, but it presents high sensibility, even at low concentrations of the infecting agent. The treatment with U-Caspofungin has no influence on the quality of the scintigraphic images used for diagnosis and localization of infection foci. Galactomannans are polysaccharides commonly extracted from the endosperm of various leguminous seeds, exhibiting variations in their man/gal ratio based on their botanical origin. These gums are highly valued for their biodegradability and excellent stability. However, their applications have been limited due to their high viscosity and molecular weight. In this study, enzymatic and ultrasound depolymerisation of galactomannans (guar gum, locust bean gum, and unconventional Neltuma ruscifolia seed gum) was carried out, resulting in hydrolysates, which were subsequently freeze-dried and charac terised. The hydrolysis process leads to a decrease in viscosity and molecular weight, accompanied by a noticeable increase in polydispersity through enzymatic treatment. Thin-layer chromatography (TLC) analysis indicates the generation of at least eight-membered subunits. Fourier-transform infrared (FT-IR) spectroscopy analysis demonstrates no significant changes in functional groups following ultrasound hydrolysis. Furthermore, in agreement with TLC results, enzymatic hydrolysates display partial reactivity towards non-enzymatic browning (NEB), yielding mostly intermediate products with limited brown ing development. This study diversifies potential applications of these gums in different industries. The optimal conditions were analyzed over 8 weeks. A quadratic model was obtained that predicted the L. monocytogenes log reduction in vacuum-packed beef treated with LA and UV-C. The maximum log reduction for L. monocytogenes (1.55&nbsp;<span>&plusmn;</span> 0.41 log CFU/g) and<br />LAB (1.55&nbsp;<span>&plusmn;</span>&nbsp;1.15 log CFU/g) with minimal impact on meat color was achieved with 2.6% LA and 330 mJ/cm2 UV-C. These conditions impaired L. monocytogenes growth and delayed LAB growth by 2 weeks in vacuum-packed meat samples throughout 8 weeks at 4&deg; C. In recent years vacuum frying was developed as an alternative methodology to traditional frying. In this study, sunflower oil thermoxidation was evaluated using conventional process conditions (180 &deg;C and atmospheric pressure) and vacuum technology conditions (130 &deg;C and 0,1 bar). Traditional thermoxidation lasted 20 h while vaccum thermoxidation was completed after 56 h. Total polar compounds reached 23 and 7,1 % at the end of atmospheric and vacuum thermoxidation respectively, while polymers content was 9,3 and 2,2 % for each oil. Tocopherols contents decreased 45 % for atmospheric thermoxidized oil and were reduced to 17 % for vacuum thermoxidized oil. These results clearly proved vacuum thermoxidation achieved a significantly lower deterioration rate than atmospheric thermoxidation of sunflower oil, conferring it much longer useful life and better nutritional qualities. Accordingly, a singnificantly slower vanishing rate of tocopherols was observed in vacuum thermoxidation. In several grape varieties, the dominating aryl alkyl alcohols found are the volatile group of phenylpropanoid-related compounds, such as glycosylated benzyl and 2-phenylethyl alcohol, which contribute to wine with floral and fruity aromas after being hydrolysed during fermentation. Saccharomyces cerevisiae is largely recognized as the main agent in grape must fermentation, but yeast strains belonging to other genera, including Hanseniaspora, are known to predominate during the first stages of alcoholic fermentation. Although non-Saccharomyces yeast strains have a well-recognized genetic diversity, understanding of their impact on wine flavour richness is still emerging. In this study, 11&thinsp;Hansenisapora vineae strains were used to ferment a chemically defined simil-grape fermentation medium, resembling the nutrient composition of grape juice but devoid of grape-derived secondary metabolites. GC&ndash;MS analysis was performed to determine volatile compounds in the produced wines. Our results showed that benzyl alcohol, benzyl acetate and 2-phenylethyl acetate are significantly synthesized by H. vineae strains. Levels of these compounds found in fermentations with 11&thinsp;H. vineae different strains were one or two orders of magnitude higher than those measured in fermentations with a known S. cerevisiae wine strain. The implications for winemaking in response to the negative correlation of benzyl alcohol, benzyl acetate and 2-phenylethyl acetate production with yeast assimilable nitrogen concentrations are discussed. An increasing global concern regarding energy saving and a growing pharmaceutical interest towards the adoption of industrial continuous processing generate an appropriate scenario for the development of solid dosage forms by moisture activated dry granulation (MADG) processes, which eliminate granulate drying and sizing, and minimize granulation end point control by making it less sensitive to shear and processing time. The aim of this study was to contribute to further enlighten formulation aspects of an attractive granulation process such as MADG, using statistical techniques. A 2^(4-1) fractional factorial design was introduced to study the effect of formulation variables on granulates obtained and on their tablets. Multivariate techniques were applied in the evaluation of the products. Lactose monohydrate and mannitol, povidone K15 and copovidone, and microcrystalline cellulose, partially pregelatinized corn starch and amorphous silica were selected, as diluents, binders and moisture absorbents, respectively. Contact angle and drop penetration time of diluents and moisture absorbents were used to study their wettability. All of the granulates yielded tablets with appropriate aspect and tensile strength, low friability, short disintegration time and in most of the cases, good weight uniformity. The statistical techniques showed the influence of excipient selection in the granulation process. Lactose produced granulates which presented lower median size and more rounded granules, leading to tablets which showed greater weight uniformity than the other diluent mannitol. The binder PVP K15 increased granule mechanical strength, producing finished tablets with higher tensile strength and longer disintegration time than copovidone. The use of a second moisture absorbent improved granulate flowability and contributed to prevent granulate caking. Wettability studies provided useful information for guidance on preliminary excipient selection. In summary, MADG is an innovative and economical process which facilitates quality by design approach. Treated wood samples were subjected to leaching tests. Scanning electron microscopic images of wood samples were examined to analyze the distribution of zinc borate within the wood. The micronized zinc product kept its crystal structure intact, and it increased the proportion of particles with diameters below 100 nm by 25% when compared to the unmilled product. Malt extract-agar medium supplemented with 2.5% of w/w milled and unmilled zinc borate inhibited fungal growth tested. Both milled and unmilled zinc borate protected the wood when not subjected to leaching. The milled sample of zinc borate improved resistance to leaching, which would allow its application in environments of high moisture content; however, it did not improve the fungicidal action against decay fungi. Among many sources of natural bioactive substances, mushrooms constitute a huge and mostly unexplored group. Biologically active secondary metabolites of Ganoderma, a group of wood-degrading mushrooms, have recently been reviewed. Our previous study revealed the antimicrobial activity of extracts from G. resinaceum grown in submerged culture against phytopathogens. Different factors can influence the production of secondary metabolites, including nutritional factors. In this study we evaluated the influence of different culture conditions on the antimicrobial activity of extracts from liquid cultures of G. resinaceum, through use of a factorial design. Minimum inhibitory concentrations for extracts produced under different culture conditions were determined against Staphylococcus aureus and Xanthomonas vesicatoria. Based on the results of these assays, larger-scale cultures in malt extract broth supplemented with 20 g/L glucose and a 15-day incubation time should be performed in order to isolate from G. resinaceum antibiotic compound(s) that are potentially useful against S. aureus. In addition, pH 5 should be considered for the production of antimicrobial metabolites against X. vesicatoria from supernatant broths or extracts from G. resinaceum. They were exposed to two-dimensional images of 12 mock-up products presented in random and balanced order. Participants assigned to the intervention groups were exposed to one FOPL scheme: black octagonal warning labels (OWL), magnifying glass high-in single icon (MGG) or traffic-light labelling (TFL). The control group was exposed to the nutrition facts up front.</p> <p>Outcome measures: OR for correctly understanding nutritional information (correctly selecting the least harmful option, correctly identifying sugars, sodium and/or saturated fats found to be in excess) and choosing to purchase the least harmful option (purchase intention), more often.</p> <p>Results: Compared with the control group, the odds for correctly selecting the least harmful option more often were 107% higher in the OWL group (OR 2.07, 95% CI 1.54 to 2.78; p&lt;0.001), whereas the MGG (1.18, 95% CI 0.89 to 1.57; p=0.24) and the TFL (1.13, 95% CI 0.85 to 1.51; p=0.39) were inefficacious in improving such odds. Propanil was only detected at the inner portion of the Andreoni canal (IAC), whereas quinclorac and clomazone were found at the IAC and at the canal mouth (CM). A major propanil metabolite was detected at the CM. Herbicides were undetectable at 13 km from the CM. MTB showed an increased susceptibility to propanil with decreasing crab sizes. The MFS showed a drastic decrease in abundance towards the freshwater discharge, concurrently with decreasing salinities. The triad approach that included water analyses, toxicological experiments and long-term field sampling allowed rejecting relationships between herbicide exposure and mole crab lethal effects. Introduction: The gastrointestinal and immune systems of premature infants are not fully developed, rendering them more vulnerable to severe complications like necrotizing enterocolitis. Human milk offers a rich array of bioactive factors that collectively contribute to reducing the incidence of gut infections and inflammatory conditions. When a mother's milk is unavailable, preterm infants are often provided with donor human milk processed in Human Milk Banks. However, it remains uncertain whether pasteurized milk confers the same level of risk reduction as unprocessed milk. This uncertainty may stem from the welldocumented adverse effects of heat treatment on milk composition. Yet, our understanding of the comprehensive impact on protective mechanisms is limited. Methods: In this study, we conducted a comparative analysis of the effects of raw versus pasteurized milk and colostrum versus mature milk on cellular functions associated with the gut epithelial barrier and responses to inflammatory stimuli. We utilized THP-1 and HT-29 cell lines, representing monocyte/macrophages and gut epithelial cells, respectively. Results: Our observations revealed that all milk types stimulated epithelial cell proliferation. However, only raw colostrum increased cell migration and interfered with the interaction between E. coli and epithelial cells. Furthermore, the response of epithelial and macrophage cells to lipopolysaccharide (LPS) was enhanced solely by raw colostrum,with a milder effect observedwith mature milk. In contrast, both raw and pasteurized milk diminished the LPS induced response in monocytes. Lastly, we examined how milk affected the differentiation of monocytes into macrophages, finding that milk reduced the subsequent inflammatory response of macrophages to LPS.Discussion: Our study sheds light on the impact of human milk on certain mechanisms that potentially account for its protective effects against necrotizing enterocolitis, highlighting the detrimental influence of pasteurization on some of these mechanisms. Our findings emphasize the urgency of developing alternative pasteurization methods to better preserve milk properties. Moreover, identifying the key components critically affected by these protective mechanisms could enable their inclusion in donor milk or formula, thereby enhancing immunological benefits for vulnerable newborns. The EEG recordings were taped and processed off-line to calculate power spectra, autocorrelation functions, and averages. In addition, the times at which spike-and-wave complexes appeared were identified and autocorrelation histograms and cross correlations (with the EEG) were made. <em>Naja</em> toxin and <em>d</em>-tubocurarine provoked a 3.5- to 5-Hz theta rhythm in both hippocampi. Higher doses elicited rhythmic epileptic spike and wave complexes which appeared at a preferred phase of theta rhythm. Atropine and medial septal lesions blocked the rhythm and disrupted the rhythmicity of epileptiform activity. We conclude that different neural subsystems sustain the theta rhythm and epileptiform spikes, and discuss the possible mechanisms involved. We performed spin-polarized ab initio calculations substituting an Mg atom by an Nb impurity. Then, some charged vacancies were included in the MgH2 + Nb system (VH, VMg, or VMg&ndash;H). In each case, three possible charge states were considered (+1, 0, or &minus;1). We computed cohesion and formation energies, band gap, and magnetic moment. We also calculated the transition level energy value and the density of states. Nb states are located in the gap, and a magnetic moment is induced. In the case of the system with charged vacancies, we found that VH+ and VH0 are the more probable vacancies formed, and the states near the Fermi level (EF) are filled, thus getting an important reduction in the band gap.
