The flavor of eight samples of commercial strawberry yogurt was studied by Free-Choice Profile analysis (FCP). Generalized Procrustes Analysis (GPA) applied to FCP allowed differentiation between samples and highlighted flavor attributes responsible for the observed differences. The relation between sensory and physicochemical datasets was studied by means of GPA. Those samples with higher carbohydrate content were perceived as sweeter, having stronger strawberry flavor, and with more dairy and yogurt flavors. Samples with higher titratable acidity, ash and protein content were perceived as more acidic and higher in intensity of “faulty” or “defective” flavors. Higher moisture content was associated with lower intensity of “dairy” flavors (creamy, dairy, and yogurt) and greater intensity of rancid flavor. It is concluded that, though not often used to this end, GPA is a suitable method to study the relationship of sensory and instrumental measurements. Two studies, one in Uruguay using Uruguayan white pan bread (WPB) and another in Spain using Spanish WPB, were carried out with trained judges and consumers from the country in question. Survival analysis methodology was used to estimate the shelf life of alfajor (a chocolate-coated individually wrapped cake) at 20 and 35C by using results obtained from consumers when asked if they would accept or reject samples with different storage times. Sensory acceptability (measured by consumers), off-flavor (measured by a trained panel) and moisture content were linearly related to time. Lead bromide polycrystalline films were grown by the physical vapor deposition method (PVD). Glass 1″x1″ in size, uncoated, and coated with Indium Tin Oxide (ITO), was used as substrate and rear contact. The starting material was evaporated at temperatures from 395°C to 530°C under high vacuum atmosphere (6 x 10-3 Pa) and during 8 days. The substrate temperature was prefixed from 190°C to 220°C. Film thickness yielded values from 40 to 90 μm. Optical microscopy and scanning electron microscopy (SEM) were performed on the films. Grain size resulted to be from 1.0 to 3.5 μm. SEM and X-ray diffraction indicate that films grow with a preferred orientation with the (0 0 l) planes parallel to the substrate. The Texture Coefficient (TC) related to the plane (0 0 6) was 7.3. Resistivity values in the order of 1012 Ωcm were obtained for the oriented samples, but a strong polarization indicates severe charge transport problems in the films. Film properties were correlated with the growth temperature and with previous results for films of other halides. The results of computer models for colloidal aggregation, that consider both Brownian motion and gravitational drift experienced by the colloidal particles and clusters, are extended to include concentrations spanning three orders of magnitude. In previous publications and for a high colloidal concentration, it was obtained that the aggregation crosses over from diffusion-limited colloidal aggregation (DLCA) to another regime with a higher cluster fractal dimension and a speeding up followed by a slowing down of the aggregation rate. In the present work we show, as the concentration is decreased, that we can still cross over to a similar regime during the course of the aggregation, as long as the height of the sample is increased accordingly. Among the differences between the mentioned new regimes for a high and a low colloidal concentration, the cluster fractal dimension is higher for the high concentration case and lowers its value as the concentration is decreased, presumably reaching for low enough concentrations a fixed value above the DLCA value. It is also obtained the fractal dimension of the sediments, arising from the settling clusters that reach the bottom and continue a 2D-like diffusive motion and aggregation, on the floor of the container. For these clusters we now see two and sometimes three regimes, depending on concentration and sedimentation strength, with their corresponding fractal dimensions. The first two coming from the crossover already mentioned, that took place in the bulk of the sample before the cluster deposition, while the third arises from the two-dimensional aggregation on the floor of the container. Reversible binding, if possible, is desirable to allow matrix reuse. Usually these aims come into conflict: strong binding may imply low expressed activity and poor reversibility. Immobilisation of Aspergillus oryzae β-galactosidase to conventional ionic exchangers is efficient but the derivatives exhibit noticeable enzyme leakage when used for the hydrolysis of lactose from whey. This problem can be avoided by covalent cross-links to the matrix, at the expense of losing reversibility. We have optimised the conditions for polyethylenimine (PEI)-coating of agarose supports to achieve a β-galactosidase derivative that allows a high lactose conversion from whey in a steady bed-reactor with no enzyme leakage, together with good elution properties. Therefore, PEI-coating appears to be a very suitable strategy to convert the agarose surface to an adequate support for a reversible, but strong and non-distorting, ionic protein immobilisation. Tannat (Vitis vinifera) were analyzed at 89 microsatellite loci. Only one, VMCNg 1d12, showed a differential pattern that separated the clones in two groups. The statistical analysis of concentrations for aroma compounds from microvinifications also resulted in the same two groupings of clones. Many analyzed microsatellite loci amplified only one allele, implying that Tannat is a highly homozygous variety. For a given set of 15 micro-satellites the level of homozygosity was 53 % for Tannat, in contrast to 6 % for Pinot, 20 % for both Cabernet Franc and Chardonnay and 33 % for Cabernet Sauvignon. We provide molecular data for Tannat, originating from south-western France and nowadays becoming the emblematic cultivar of Uruguayan fine red wines. We also report a correlation between aroma-related compounds and molecular markers within clones of a cultivar. The ubiquitous intracellular molecule myo-inositol hexakisphosphate (IP6) is present extracellularly in the hydatid cyst wall (HCW) of the parasitic cestode Echinococcus granulosus. This study shows that extracellular IP6 is present as its solid calcium salt, in the form of deposits that are observed, at the ultrastructural level, as naturally electron dense granules some tens of nanometers in diameter. The presence of a calcium salt of IP6 in these structures was determined by two different electron microscopy techniques: (i) the analysis of the spatial distribution of phosphorus and calcium in the outer, acellular layer of the HCW (the laminated layer, LL) through electron energy loss spectroscopy, and (ii) the observation, by transmission electron microscopy, of HCW that were selectively depleted of IP6 by treatment with EGTA or phytase, an enzyme that catalyses the dephosphorylation of IP6. The deposits of the IP6-Ca(II) salt are also observed inside membrane vesicles in cells of the germinal layer (the inner, cellular layer of the HCW), indicating that IP6 precipitates with calcium within a cellular vesicular compartment and is then secreted to the LL. Thus, much as in plants (that produce vesicular IP6 deposits), the existence of transporters for IP6 or its precursors in internal membranes is needed to explain the compound's cellular localisation in E. granulosus. A flow injection method is proposed for the determination of phenylephrine hydrochloride in pharmaceutical dosage forms. The method involves the use of on-line solid-phase extraction by means of a microcolumn containing Dowex 50W X8 ion-exchange resin for the separation of the analyte prior to colour development and spectrophotometric detection in the visible region. The influence of preconcentration flow, preconcentration pH and elution volume was studied. The method exhibits appropriate linearity (r(2) = 0.9999) which was proved statistically by means of the "F"-test. When applied to commercial samples containing several active ingredients and excipients, a significant reduction of interferences was found. Accuracy, evaluated by means of the spike recovery method was in the range 99.7-100.8%, with precision (R.S.D., %) better than 1%. In order to achieve the automation the system was controlled from a notebook computer by means of a program written in QuickBASIC language. Under these conditions, a sampling frequency of 40 samples per hour could be attained. Insolubles determination is one of the parameters usually recommended to evaluate the residual life of oil because their presence at elevated levels in diesel lubricating oil changes the viscosity, prematurely clogs filters and is one of the major factors in causing abrasive engine wear. The proposed method employs visible spectrophotometric detection in association with flow injection analysis. The results obtained by this method were compared with the ones obtained by Fourier transform infrared spectrometry (FT-IR) since this is the most employed method for insolubles determination. The proposed method presented a linear response from 0 to 3% (w/w) of insolubles in pentane (ASTM D-893). The sampling frequency was about 30 samples h−1, with a relative standard deviation (n=5) of 2.4% or better. Accuracy was evaluated analysing 98 real samples and the results obtained with the FIA-spectrophotometric method were plotted against those obtained by the FT-IR method by means of linear regression. Slope and intercept of the straight line obtained were compared with the theoretical values of 1 and 0 by means of the joint-confidence ellipse F-test. At the confidence level of 95% no evidence of a difference was found between both methods. The development of 99mTc complexes for imaging 5-HT1A neural system receptors using the 3 + 1 mixed ligand approach is described. Six novel complexes (I-VI) were designed using two different strategies. In complexes I-IV the pharmacophore 1-(2-methoxyphenyl)piperazine was attached to a monodentate thiol used as co-ligand and combined with tridentate dianionic aminothiols (SNS and NNS). On the other hand, complexes V and VI were obtained using thiophenol and 4-methoxy-thiophenol as co-ligand and a tridentate ligand (SNS) with the pharmacophore bound to the nitrogen through an alkyl chain. All complexes were prepared at tracer level using 99mTc-glucoheptonate as precursor. Ligand and co-ligand concentration, reaction time and temperature were optimized to achieve high substitution yield and radiochemical purity. Structure was studied at carrier level through the corresponding rhenium complexes. Complexes I and II presented the expected ReOLK structure and a distorted trigonal bipyramidal geometry. The structure of the other four complexes has not been completely elucidated yet. Biodistribution studies of all the complexes demonstrated selective brain uptake and retention. Uptake of complex I in receptor-rich hippocampus was significantly higher than that of the cerebellum (P = 0.05) 1 h post-injection. Oxorhenium complexes I and II showed affinity for the 5-HT1A receptor binding sites, with IC50 values in the nanomo with IC50 values in the nanomolar range. The results demonstrate the potential of the mixed ligand approach for the design of 99mTc complexes with the ability to bind neuroreceptors. However, the goal of imaging 5-HT1A receptors with technetium requires further development of complexes with improved biological profiles. A biotype of clary sage (Salvia sclarea L.) growing in Uruguay was evaluated for its suitability to be cultivated in Uruguay. Essential oils from the inﬂorescences and leaves of plant harvested at full ﬂowering and early seed ripeness stages were obtained by steam distillation and characterized by GC and GC–MS (quadrupole). Twenty-seven components (91–98% of the total composition) were identiﬁed. The oil was characterized by a high content of linalool (8–22%), linalyl acetate (39–48%) and sesquiterpenes, with germacrene D (8–20%) and (E)-β-caryophyllene (3–5%) as the main components. The enantiomeric distribution of -pinene, sabinene, β-pinene, limonene, linalool, linalyl acetate and germacrene D was studied by multidimensional HRGC–HRGC. The development stage did not inﬂuence the oil composition or the enantiomeric distribution of the studied components of the clary sage biotype over the period of evaluation.
