Methodologies that provide reliable information about the sensory characteristics of the developed products in short time frames can speed up the product development process and contribute to the success of the developed products in the marketplace. A panel of 9 trained assessors evaluated samples using descriptive analysis. Besides, the yogurts were evaluated by 3 groups of 81 consumers using three rapid methodologies: check-all-that-apply (CATA) questions, projective mapping and polarized sensory positioning. The three rapidmethodologies provided similar information on themain differences among samples. However, several differences can be highlighted. Sample configurations from CATA questions were the most similar to those provided by descriptive analysis, whereas projective mapping provided the least similar configurations. The three methodologies also differed in their ability to detect differences among samples due to formulation variables and the stability of sample configurations. The estimation of equivalent sweet concentrations of high-intensity sweeteners is one of the first steps for the substitution of sucrose during the development of low-calorie products. The aim of this work was to compare two sensory methodologies (paired comparison and magnitude estimation) and two data analysis approaches (logistic regression and survival analysis) to estimate equivalent sweet concentration of high-intensity sweeteners, using orange/pomegranate juice as case study. Three low-calorie sweeteners were considered: sucralose, stevia and monk fruit extract. For each sweetener, a series of six orange/pomegranate juice samples with different sweetener concentration were prepared. Three groups of 100 consumers participated in the study. Each group was randomly divided into two groups of 50, each of which evaluated samples using one of the two methodologies (paired comparison and magnitude estimation). Paired comparison and magnitude estimation provided similar estimations for the three high-intensity sweeteners which were in agreement with published data, regardless of the statistical approach used for data analysis. However, logistic regression and survival analysis differed in the accuracy of the estimations. New processing technologies are gaining popularity worldwide due to several advantages related to food safety, shelf life extension, nutritional and sensory quality. However, consumers can be cautious of food products produced using these technologies. In this context, the objectives of the study were to explore consumers' spontaneous associations with fruit juices processed using different technologies, and to study the influence of food technology neophobia on those associations. A study with 423 Brazilian consumers was conducted to evaluate their perception of five types of juices (fresh juice, cold pressed juice, pasteurized juice, pressurized juice and non-pressurized juice) using word association. Participants also answered the Food Technology Neophobia Scale (FTNS) and a series of socio-demographic questions. Results showed that consumers' attitudes towards processing technologies were mainly defined by top-down processes. Fresh, cold pressed and non-pressurized juice were mainly associated with healthy and natural products, whereas concepts including references to processing technologies were associated with processed products and unhealthiness. Food technology neophobia moderated consumers' perceptions of juice processing technologies. Interestingly, consumers with high technology neophobia perceived juices processed by both conventional and innovative technologies more negatively than those with low or medium levels of neophobia. Nutritional warnings are rapidly gaining relevance, particularly in the region of the Americas, as a front-of-package nutrition labelling scheme that facilitate the identification of products with high content of nutrients associated with non-communicable diseases (NCDs). Several graphic designs are being developed as countries discuss the implementation of nutritional warnings, which makes it necessary to develop research to evaluate their efficacy. In this context, the aim of the present work was to compare the efficacy of a series of nutritional warnings that are being considered by Brazilian national authorities for the country with two of the most studied schemes: the guidelines daily amounts (GDA) and the traffic light system. Two studies were conducted. In the first one, visual search was conducted with 62 participants to evaluate the time needed by them to identify the schemes on food labels and to identify whether a product had high nutrient content. In the second study, an online survey with 1932 participants was used to evaluate their ability to use FOP nutrition labelling schemes to correctly identify the most healthful product in a set, as well as high nutrient content in a product. In addition, the influence of FOP nutrition labelling schemes on perceived healthfulness was evaluated. Finally, consumers&rsquo; perception of the schemes was gathered using an open-ended question. Results confirmed the advantages of nutritional warnings compared to the GDA and the traffic-light system to facilitate the identification of products with high content of nutrients associated with NCDs. In addition, the use of familiar signs frequently used to convey a &lsquo;warning message&rsquo; outperformed other unfamiliar signs in terms of their ability to facilitate the interpretation of nutrition information. So far, sugar reduction strategies have been targeted at minimizing the sensory changes associated with product reformulation, mainly through the use of non-nutritive sweeteners. However, emerging evidence has shown that we may have been following the wrong path by focusing on reducing sugar while maintaining sweetness. The aim of this review is to critically discuss sugar reduction strategies in the light of recent evidence related to consumers&rsquo; hedonic sensitivity to sugar reduction, the potential negative effects of uncoupling sweet taste from energy load, and the limited effectiveness of voluntary agreements for sugar reduction. A large body of evidence assessing the effectiveness of front-of-package (FOP) nutrition labeling exists. Most experimental studies have been conducted with fictitious products. However, consumers&rsquo; perception depends on several products extrinsic factors such as brand. Understanding how strong brand associations influence the effectiveness of FOP nutrition labeling schemes may be crucial to informing policymaking. In this context, the aim of this work was to evaluate the effect of five different variants of nutritional warnings labels (black magnifier, red magnifier, black octagon, black triangle, and red circle) on consumers&rsquo; choice of commercial products, compared with two FOP nutrition labeling schemes: the guidelines daily amounts (GDAs) system and the traffic light system (TLS). An online randomized controlled trial with 1,932 participants was used to evaluate the effect of FOP nutrition labeling on participants&rsquo; choices in eight sets of three commercial products, available in the Brazilian marketplace. A multinomial logistic regression model was used to evaluate the influence of FOP nutrition labeling on participants&rsquo; likelihood of selecting the different products in the choice task. Results showed that nutritional warnings and the TLS significantly increased the likelihood of selecting none of the products instead of the least healthful product, or a healthier product, in at least one of the product categories compared with the GDA. Warnings tended to have a larger effect, suggesting their potential to encourage healthier food choices. Se estima que cerca de 7% son causados por mutaciones en el &aacute;cido desoxirribonucleico germinal. Los costos de la secuenciaci&oacute;n gen&eacute;tica han descendido dram&aacute;ticamente gracias a la aparici&oacute;n de la secuenciaci&oacute;n de nueva generaci&oacute;n (NGS). El cambio tecnol&oacute;gico abri&oacute; una nueva etapa en el estudio del c&aacute;ncer hereditario en nuestro pa&iacute;s. Objetivo: comunicar los resultados de la utilizaci&oacute;n de tecnolog&iacute;a NGS y paneles multig&eacute;nicos en familias uruguayas con alto riesgo de c&aacute;ncer de mama hereditario. Pacientes y m&eacute;todo: se secuenciaron 135 familias de alto riesgo que proven&iacute;an de la consulta de consejer&iacute;a gen&eacute;tica que funciona en el Grupo Colaborativo Uruguayo: Investigaci&oacute;n de afecciones oncol&oacute;gicas hereditarias. Cuando la historia familiar suger&iacute;a claramente un s&iacute;ndrome de c&aacute;ncer de mama y ovario hereditario se efectu&oacute; secuenciaci&oacute;n NGS exclusiva de los genes BRCA1 y 2; cuando el patr&oacute;n familiar no configuraba claramente se utiliz&oacute; un panel multig&eacute;nico. Resultados: se efectu&oacute; NGS exclusiva de genes BRCA1 y 2 en 62 familias y un panel multig&eacute;nico en 73 familias. Se identificaron en total 29 mutaciones pat&oacute;genas (21 en genes BRCA y 8 en otros genes). Dos de ellas fueron noveles y tres pueden considerarse recurrentes en la poblaci&oacute;n uruguaya. Conclusiones: este trabajo es el primero en Uruguay en reportar el resultado de esta nueva tecnolog&iacute;a en el c&aacute;ncer de mama hereditario. El hallazgo de 29 mutaciones pat&oacute;genas nos ayuda a delinear el perfil mutacional de nuestro pa&iacute;s. Pachyrhizus ahipa is an Andean leguminous plant which has purple-pigmented edible roots. The content and distribution of the phenolic compounds in the skins and flesh of P. ahipa roots as well as their antioxidant capacity were studied. Total phenols, flavonoids, tannins and anthocyanins were quantified and the effect of the extraction solvent on the maximum absorbance wavelength (&lambda;vis-max) was measured. The ATR-FTIR spectrum and stability of the purified anthocyanin extracts were analyzed. Individual anthocyanins were partially characterized using high-performance liquid chromatography-mass spectrometry and absorbance spectra. Ahipa skins had much more phenols (3.25 mg gallic acid equivalents/g), flavonoids (3.0 mg catechin equivalents/g) and tannins (4.6 mg tannic acid equivalents/g) than the flesh, which was reflected in a higher antioxidant capacity. Changing the extraction solvent from acidified methanol to acidified water shifted the &lambda;vis-max 16 nm lower, suggesting copigmentation reactions. Anthocyanin in the skins was &gt;8.7 content in the flesh, and the chromatographic analysis showed at least 6 different anthocyanins with identical retention times for skins and flesh, including delphinidin, pelargonidin and cyanidin as aglycons. The absorbance spectra showed that two of them are acylated structures. Pigments were stable for at least 180 min at pH 1 and 25 &deg;C, but they were affected by increased pH and temperature. Ethnopharmacological relevance: Lauraceae family includes Nectandra angustifolia a species widely used in the folk medicine of South America against various maladies. It is commonly used to treat different types of processes like inflammation, pain, and snakebites. Snakes of the Bothrops genus are responsible for about 97% of the ophidic accidents in northeastern Argentina. Aim of the study: To evaluate the anti-snake activity of the phytochemicals present in N. angustifolia extracts, identify the compounds, and evaluate their inhibitory effect on phospholipase A2 (PLA2) with in vitro and in silico assays. Methods: Seasonal variations in the alexiteric potential of aqueous, ethanolic and hexanic extracts were evaluated by inhibition of coagulant, haemolytic, and cytotoxic effects of B. diporus venom. The chemical identity of an enriched fraction obtained by bio-guided fractioning was established by UPLC-MS/MS analysis. Molecular docking studies were carried out to investigate the binding mechanisms of the identified compounds to PLA2 enzyme from snake venom. Results: All the extracts inhibited venom coagulant activity. However, spring ethanolic extract achieved 100% inhibition of haemolytic activity. Bio-guide fractioning led to an enriched fraction (F4) with the highest haemolytic inhibition. Five flavonoids were identified in this fraction; molecular docking and Molecular Dynamics (MD) simulations indicated the binding mechanisms of the identified compounds. The carbohydrates present in some of the compounds had a critical effect on the interaction with PLA2. Conclusion: This study shows, for the first time, which compounds are responsible for the anti-snake activity in Nectandra angustifolia based on in vitro and in silico assays. The results obtained in this work support the traditional use of this species as anti-snake in folk medicine. Hanseniaspora vineae exhibits extraordinary positive oenological characteristics contributing to the aroma and texture of wines, especially by its ability to produce great concentrations of benzenoid and phenylpropanoid compounds compared with conventional Saccharomyces yeasts. Consequently, in practice, sequential inoculation of H. vineae and Saccharomyces cerevisiae allows to improve the aromatic quality of wines. In this work, we evaluated the impact on wine aroma produced by increasing the concentration of phenylalanine, the main amino acid precursor of phenylpropanoids and benzenoids. Fermentations were carried out using a Chardonnay grape juice containing 150 mg N/L yeast assimilable nitrogen. Fermentations were performed adding 60 mg/L of phenylalanine without any supplementary addition to the juice. Musts were inoculated sequentially using three different H. vineae strains isolated from Uruguayan vineyards and, after 96 h, S. cerevisiae was inoculated to complete the process. At the end of the fermentation, wine aromas were analysed by both gas chromatography– mass spectrometry and sensory evaluation through a panel of experts. Aromas derived from aromatic amino acids were differentially produced depending on the treatments. Sensory analysis revealed more floral character and greater aromatic complexity when compared with control fermentations without phenylalanine added. Moreover, fermentations performed in synthetic must with pure H. vineae revealed that even tyrosine can be used in absence of phenylalanine, and phenylalanine is not used by this yeast for the synthesis of tyrosine derivatives. Polyreactive antibodies (pAb) bind to a broad range of unrelated structures, providing<br />hosts with functional components able to rapidly recognize and protect against different<br />pathogens. However, their roles against helminth parasites are still unexplored.<br />Here, pAb profiles were analysed in cystic echinococcosis (CE), a zoonosis caused by<br />the cestode Echinococcus granulosus sensu lato. Levels of anti-DNP (2,4-dinitrophenyl-<br />hapten) antibodies were measured as a surrogate parameter of pAb in different<br />biological settings. Firstly, levels of serum and peritoneal pAb were measured during<br />early experimental secondary CE, using both high (Balb/c) and low (C57Bl/6) susceptible<br />mouse strains. Among them, CD5 and CD6 are lymphocyte scavenger receptors known to interact with different microbial-associated molecular patterns, and the administration of the recombinant soluble ectodomains of human CD5 (rshCD5) and/or CD6 (rshCD6) has shown therapeutic/prophylactic potential in experimental models of fungal, bacterial and echinococcal infections. The latter is a zoonosis caused by the larval stage of the cestode parasite Echinococcus granulosus sensu lato, which in humans can induce secondary cystic echinococcosis (CE) after the spillage of protoscoleces contained within fertile cysts, either spontaneously or during surgical removal of primary hydatid cysts. Herein, we have analysed the mechanisms behind the significant protection observed in the mouse model of secondary CE following prophylactic administration of rshCD5 or rshCD6. Our results show that both molecules exhibit intrinsic antiparasitic activities in vitro, as well as immunomodulatory functions during early secondary CE, mainly through Th1/Th17 cytokine bias and promotion of peritoneal polyreactive antibodies. These data support the relevance of the parasite components bound by rshCD5 and rshCD6, as well as the potential of their prophylactic administration as a useful strategy to reduce secondary CE in patients. La mitocondria es un el sitio principal de generacion de superoxido y otras especies reactivas y es uno de los principales sitios de formacion de 3-nitrotirosina proteica tanto en condiciones basales como patologicas. En este trabajo se estudiaron dos proteinas mitocondriales, el citocromo c y la superoxido dismutasa de manganese (MnSOD) y de cobre y cinc (CuZnSOD), estudiando tanto la nitracion de estas proteinas como cambios estructurales en condiciones similares alas encontradas in vivo. En el caso del citocromo c se estudio la nitracion de tirosinas proteicas evaluando la presencia de un lipide mitocondrial, la cardiolipina. En los experimentos realizados se observo por diversas tecnicas que en presencia de este Iipido la nitracion de citocromo c expuesto a peroxinitrito se encuentra aumentada, siendo este aumento mediado por un cambio conformacional mediado por la cardiolipina. A nivel estructural, se observaron los cambios sufrido por citocromo 0 en presencia de cardiolipina por tecnicas de NMR para y diamagnetico. Estos experimentos, si bien no fueron concluyentes mostraron que la proteina sufre un cambio conformacional global. Estudios de la reactividad de cada tirosina de citocromo c utilizando mutantes mostro que, en presencia de cardiolipina, tirosinas que no son preferencialmente nitradas 0 modificadas en ausencia de este Iipido estan involucradas en procesos oxidativos tales como la actividad peroxidasa de citocromo c mediada por cardiolipina. Esta actividad peroxidasa se estudio tam bien utilizando un derivado carboximetilado en me\ionina de ci\ocmmo c que, a di~efencia de\ ci\ocmmo c sin modi~icaf pefmi\e \a deteccion de un Compuesto I de peroxidasa. EI agregado de cardiolipina a citocromo c carboximetilado expuesto a peroxido de hidrogeno aumenta la formacion de compuesto I de citocromo c, y por 10 tanto su actividad peroxidasa. Para profundizar el estudio de los cambios estructurales sufridos por citocromo c frente a diferentes oxidantes se sintetizo y purifico citocromo c mononitrado a nivel de Tyr 97 y 74, para generar anticuerpos monoclonales contra formas conformacionalmente modificadas de esta proteina que permitan la deteccion de las mismas tanto en estudios in vitro como in vivo En el caso de las superoxido dismutasas (SOD) se estudiaron las modificaciones sufridas en presencia de flujos de oxido nitrico y superoxido evaluando la capacidad de esta enzima de competir con el oxido nitrico por su sustrato, observandose que en presencia de estos flujos se forma peroxinitrito en concentraciones capaces de inactivar a la enzima y en el caso de MnSOD capaz de nitrarla. Por otra parte se reeestudio la reaccion de MnSOD con peroxinitrito determinando un nuevo valor para la constante de reaccion de 1,9 x 104 M&middot;1s&middot;1 por tetramero. A nivel estructural se estudio el efecto de la nitracion en la accesibilidad del sustrato al sitio activo utilizando metodos in silico donde se determino que la nitracion de Tyr 34, responsable de la inactivacion de MnSOD por peroxinitrito genera un aumento en la barrera de energia libre para el acceso del superoxido al sitio activo, 10 cual impide el acceso al sustrato, tanto por repulsion esterica como electrostatica. Tambien a nivel estructural, se determino el efecto de mutaciones puntuales de MnSOD en la estructura cuaternaria de la enzima, evaluando, a su vez, el efecto de estas mutaciones en la sensibilidad de sus tioles. ----- No se encontraron virus en este mensaje. Human Mn-containing superoxide dismutase (hMnSOD) is a mitochondrial enzyme that metabolizes superoxide radical (O2(&bull;-)). O2(&bull;-) reacts at diffusional rates with nitric oxide to yield a potent nitrating species, peroxynitrite anion (ONOO(-)). MnSOD is nitrated and inactivated in vivo, with active site Tyr34 as the key oxidatively modified residue. We previously reported a k of &sim;1.0 &times; 10(5) M(-1) s(-1) for the reaction of hMnSOD with ONOO(-) by direct stopped-flow spectroscopy and the critical role of Mn in the nitration process. In this study, we further established the mechanism of the reaction of hMnSOD with ONOO(-), including the necessary re-examination of the second-order rate constant by an independent method and the delineation of the microscopic steps that lead to the regio-specific nitration of Tyr34. The redetermination of k was performed by competition kinetics utilizing coumarin boronic acid, which reacts with ONOO(-) at a rate of &sim;1 &times; 10(6) M(-1) s(-1) to yield the fluorescence product, 7-hydroxycoumarin. Time-resolved fluorescence studies in the presence of increasing concentrations of hMnSOD provided a k of &sim;1.0 &times; 10(5) M(-1) s(-1), fully consistent with the direct method. Proteomic analysis indicated that ONOO(-), but not other nitrating agents, mediates the selective modification of active site Tyr34. The nutritional composition of the sapucaia nut, cake and shell, the nut and cake minerals content and the lipid profile of the nut oil (fatty acids, tocopherols, phytosterols and triacylglycerols) were determined. The nuts and cake exhibited a high content of lipid (47.9 to 60.8 mg 100 g&minus;1), protein (15.8 to 19.5 mg 100 g&minus;1), dietary fiber (16.5 to 22.6 mg 100 g&minus;1) and provided an excellent source of selenium (26.4 to 46.94 &mu;g g&minus;1). The oil contained a high amount of unsaturated fatty acids (39.7 to 45.4% of oleic and 32.2 to 46.6% of linoleic acids) and presented a high Oxidative Stability Index (8.57&ndash;12.95 h) indicating the presence of antioxidant compounds in the oil. The major triacylglycerols in the sapucaia oil were LLO, PLO, LOO, POO, OOO, PLL and LLL. The main bioactive lipids identified in the oil were &gamma;-tocopherol (19.2 to 28.5 mg 100 g&minus;1) and &beta;-sitosterol (92.8 to 194 mg 100 g&minus;1). The results showed that the sapucaia nut and its by-products are a promising natural source of bioactive and nutritional compounds and when present in the diet can contribute to the maintenance of human health. In addition, the nut and by-product represents a promising raw material for the food industry. To face the high costs of developing new drugs, researchers in both industry and academy are looking for ways to repurpose old drugs for new uses. In this sense, bisphosphonates that are clinically used for bone diseases have been studied as agents against Trypanosoma cruzi, causative parasite of Chagas disease. In this work, the development of first row transition metal complexes (M = Co2+, Mn2+, Ni2+) with the bisphosphonate ibandronate (iba, H4iba representing the neutral form) is presented. The in-solution behavior of the systems containing iba and the selected 3d metal ions was studied by potentiometry. Mononuclear complexes [M(Hxiba)](2&minus;x)&minus; (x = 0&ndash;3) and [M(Hiba)2]4&minus; together with the formation of the neutral polynuclear species [M2iba] and [M3(Hiba)2] were detected for all studied systems. In the solid state, complexes of the formula [M3(Hiba)2(H2O)4]&middot;6H2O were obtained and characterized. All obtained complexes, forming [M(Hiba)]&minus; species under the conditions of the biological studies, were more active against the amastigote form of T. cruzi than the free iba, showing no toxicity in mammalian Vero cells. In addition, the same complexes were selective inhibitors of the parasitic farnesyl diphosphate synthase (FPPS) enzyme showing poor inhibition of the human one. However, the increase of the anti-T. cruzi activity upon coordination could not be explained neither through the inhibition of TcFPPS nor through the inhibition of TcSPPS (T. cruzi solanesyl-diphosphate synthase). The ability of the obtained metal complexes of catalyzing the generation of free radical species in the parasite could explain the observed anti-T. cruzi activity.
