Infection of Balb/c mice with Echinococcus granulosus protoscoleces constitutes the model for secondary hydatid infection. The immune response of Balb/c mice infected with E. granulosus is characterized by secretion of antibodies specific for carbohydrate epitopes and production of type-2 cytokines. A role for glycoconjugates in the induction of type-2 responses has been suggested in other host--parasite systems. Although glycoconjugates are immunogenic in E. granulosus infection, the role of these molecules in the establishment of the type-2 response has never been analysed. In this study, a carbohydrate rich fraction (E4+) from E. granulosus protoscoleces was obtained using the monoclonal antibody E492/G1 specific for the moiety Galalpha(1,4)Gal which is widely represented in protoscoleces and other E. granulosus antigenic preparations. The results showed that E4+ was immunogenic in Balb/c mice evoking an antibody response mainly directed against carbohydrate epitopes. In addition, splenocytes from E4+-immunized mice showed suppressed proliferative responses to Con A and E4+ induced IL-10 secretion by E4+-primed and naive splenocytes. The fraction E4+ also was immunogenic in infected mice during early infection. In this case also, splenocytes from infected mice as well as peritoneal cells from infected or naive mice, when stimulated in vitro with E4+, secreted IL-10. Collectively, these results suggest that E4+ may be involved in immunosuppression phenomena and, by stimulating IL-10 secretion, may contribute to the induction and sustaining of the type-2 cytokine response established in early experimental infection. Moreover, it is known that tumour cells show enhanced intake of α-amino acids (Hnaa). With this in mind, the study of samarium complexes with these ligands has obvious interest. In this work, potentiometric studies of Sm(III) in the presence of glycine, alanine, proline, tryptophane, valine, glutamic acid and cysteine have been carried out. Experiments were performed in aqueous solution ([Sm3+]=5.0–60.0 mM) at 37°C and 0.15 M NaClO4. Mono and dinuclear species were detected in the pH range 1.5–5.3. For higher pH values, Sm(OH)3 competes with the formation of Sm complexes. In addition, Sm(III)–Hnaa complexes were isolated and characterized at solid state. The structure of [Sm2(Hala)4(H2O)8](ClO4)4(Cl)2 is reported. In this complex, two Sm centres are joined by four μ-COO bridges. Four molecules of water complete the positions of each eight-coordinate Sm atom. This antigen was detected in both larval and adult worm extracts, with the highest specific activity observed in the adult excretion/secretion preparation. Histochemical analysis showed that Tn is preferentially expressed in the parenchyma in both parasite stages and the external part of tegument in adult worms. A similar pattern was observed for sialyl-Tn, a related O-linked antigen. Tn glycoproteins from protoscoleces were resolved by SDS-PAGE in two main components of 43 and 49 kDa. After purification, this material was reactive with lectins which bind GlcNAc/sialic acid, GalNAc, and T antigen. In a preliminary evaluation, high levels of Tn antigen were detected in serum samples from patients with hydatid cyst, suggesting that the measure of Tn in serum could be a biomarker of this disease, although extensive work is necessary in order to determine the clinical usefulness of this assay. The results reported here, the first evidence of O-glycosylation pathways in E. granulosus and the presence of Tn antigen in cestodes, suggest that the evaluation of O-glycosylated antigens might give new insights in the host-parasite relationship. A group of Gram-negative denitrifying bacteria has been isolated from a denitrifying reactor treating landfill leachate. The new isolates produced both oxidase and catalase and showed growth on acetate, butyrate, n-caproate, i-butyrate, i-valerate, propionate, n-valerate, lactate, alanine, benzoate, phenylalanine and ethanol. No growth was observed on sugars. The bacteria could perform anoxic reduction of nitrate, nitrite and nitrous oxide to nitrogen, coupled to the oxidation of the same substrates as those used under aerobic conditions, except for aromatic compounds. They were very efficient denitrifiers, as estimated from the specific rate of N2 gas production. All the strains showed the same 16S rDNA restriction profile and one of them, designated 23310T, was selected for phylogenetic analysis. The organism clustered within the family Comamonadaceae, being related to Comamonas terrigena (95.8% sequence similarity). On the basis of the phylogenetic analysis, physiological characterization and the ability to efficiently reduce nitrate to N2, it is proposed that the bacterium be assigned to a new species, Comamonas nitrativorans. Mercuric iodide platelets were grown in the HgI 2 }HI}H 2 O system by solvent evaporation at constant temperature. Di!erent solution temperatures, growing times and stirring conditions were studied. In order to determine the best growing parameters the chemistry of the HgI 2 }HI}H 2 O system was studied. Complexes [HgI 3 ]~ and [HgI 4 ]~2 were identi"ed and quanti"ed in the solution by visible and UV spectrometry, according to the R ratio ([HgI 2 ]/[HI]). Solution oxidation and I~3 interference were eliminated performing growth in high purity Argon atmosphere. Good mercuric iodide platelets, up to 6.0]7.0]1.5mm3 in size, were obtained with appropriate argon #ow, temperature and growing time, for several independent experiences. Platelets were observed by optical microscopy and the absence of possible dislocations was con"rmed by studying etch pits density after etching with ethanol}trichloroethylene. Nonexistence of occluded water and hydrocarbons was con"rmed by Fourier transform infrared spectroscopy and was related to results obtained for mercuric iodide crystals grown by the physical vapor transport method. Detectors were made with the platelets and their apparent resistivity, current density and X-ray performance were measured. Discussions about future improvements of growing mercuric iodide platelets in the HgI 2 }HI}H 2 O system and comparison of this method with others are included. The animals were euthanized and the intestines were dissected out 72 h postinfection and were incubated on a Baermann's apparatus to recover L4 stage larvae. No larvae were recovered from the 20 mg/kg albendazole-treated group, and means of 15, 33, and 175 L4 larvae were recovered from the 15, 10, and 5 mg/kg albendazole-treated groups, respectively. Levamisole and fenbendazole were also tested using the same procedure. Means of 100 and 1887 L4 larvae were obtained from the 50 and 10 mg/kg of levamisole-treated groups respectively; while, 191 and 583 L4 larvae were recovered from rats treated with 50 and 10 mg/kg of fenbendazole, respectively. These results indicate that the L4 stage of N. brasiliensis could be useful for in vivo screening of new nematocide drugs. The larval stage of the parasite Echinococcus granulosus causes hydatid disease. The hydatid cyst is potentially capable of activating host complement, since it is a large, persistent, carbohydrate-rich structure, coated with host immunoglobulins, and localized in the host's internal organs. Nonetheless, in vitro studies have suggested that the cyst surface, the hydatid cyst wall (HCW), is a poor complement activator. In this study, we assessed the occurrence of in vivo complement activation on the hydatid cyst by measuring the levels of two complement activation products, C3d and complexes bearing a C9 activation neoepitope (TCC/MAC), in extracts from HCW of human origin. Low amounts of C3d and TCC/MAC were found in HCW in comparison with their levels in normal human plasma and activated human sera, suggesting that in vivo complement activation on HCW is efficiently down-regulated. This regulation may contribute to limit host inflammation which has been observed to correlate with parasite degeneration and death. Lead iodide purification was performed by repeated sublimation at 390°C. Studies of purity and stoichiometry were done by Inductively Coupled Plasma and Differential Scanning Calorimetric methods. These results were compared with those obtained from the same starting material through zone melting purification and with previous reports. Palladium contacts were attached to Teflon substrates with areas of 2 cm2 and lead iodide films 50 to 500 μm in thickness were deposited onto it by sublimation of the purified material at near 390°C. Deposition conditions such as source state (powder, bulk), source–substrate distance and deposition time were optimized. Front palladium thermal deposition contacts and acrylic encapsulation were done on the radiation penetrating surface and electrical properties such as apparent resistivity and current density were measured. The X-ray film detection capabilities were checked by light pulses, X and gamma irradiation. Finally, the film properties were correlated with the starting material, the deposition process and previous data. Polycrystalline films were grown by physical vapor deposition using lead iodide purified by evaporation as starting material. Purity and stoichiometry of starting material were determined by Inductively Coupled Plasma and wet procedures. Palladium film was thermal deposited as rear contact onto glass and alumina substrates 5x5 cm2 in size. Onto it, lead iodide films were grown by evaporation with source temperatures from 430 to 450 degree(s)C in a 500 mmHg Argon atmosphere. Substrate temperature were from 200 to 250 degree(s)C and deposition times from 2 to 10 days. Film thickness was measured by 59.5 keV (241Am) emission absorption, resulting values up to 50micrometers . The films were characterized by optical and atomic force microscopy, giving an average grain size up to 2micrometers . Film's low temperature photoluminescence confirmed the purity of the starting material. X-ray diffraction measurements of film's reflections show an intensity relation [(Sigma) I(0 0 l)] / [(Sigma) I(h k l)] from 0.2 to 0.9 that correlates to the film deposition temperature. For determining electrical and spectrometric properties, front palladium thermal deposition contacts and acrylic encapsulation were done onto the lead iodide films. Apparent resistivities from 1012 to 1015 (Omega) .cm and current densities in the order of 6 pA/cm2 (50 V) were determined. X-ray film response and uniformity was checked by irradiating with an X-ray medical equipment. Film properties and performance were correlated with starting material and substrate temperature, with previous results for lead iodide films grew by other methods and with similar results for mercuric iodide films. Radiation detectors were fabricated with them and detector current density and apparent resistivity were determined by I-V curves, giving results 300 pA/cm2 (30V) and 1013 (Omega) .cm. X-ray spectra were performed determining detector's energy resolution for different emissions, giving a resolution of 1 keV in the range of 10 - 20 keV radiations. According to electrical and spectrometric performance, optical microscopy and production yields, it was not necessary to use high purity starting material for obtaining excellent platelets. Correlation between optical and electrical properties of the crystals grown from this method and others previously grown from the melt are also discussed. The solvent effect was considered for three solvents (chloroform, methanol, and water).
