The current limitations on bioconversion of lignocellulosic biomass include poorenzyme stability and inhibition by secondary or final products. Here, we report the biochemical char-acterization of a novel, rumen metagenome-derived glucanase and xylanase, named EndoG. From thededuced amino acid sequence it was assigned to glycoside hydrolase family 5. EndoG showed similarityto non-characterized proteins, and its parental organism is likely related to the genus Prevotella. The1146pb ORF encoding EndoG was over-expressed in Escherichia coli and the protein purified. The recom-binant EndoG displayed a wide range of pH activity with a maximum at pH 5.0 and at least 65% activityat pH between 4.5 and 7.5. The enzyme was highly stable at 55◦C for 1 h, and retained 81% activity at 4 MNaCl. EndoG was also active in the presence of diverse divalent cations, detergents, EDTA, acetate, fur-fural, imidazolium ionic liquids, and ethanol. The biotransformation of (4R)-(-)-carvone by Mentha pulegium (pennyroyal) leaves and its endophytic bacteria was performed in order to search for novel biocatalysts with enoate reductase activity. The obtained results clearly indicated that endophytes play an important role in the biotransformation of (4R)-(-)-carvone with pennyroyal plant tissues. P. proteolytica FM18Mci1 had preference for the 4S-(-)-carvone, reaching a conversion 95% in 24 h. In contrast, Bacillus sp. FM18civ1 had preference for (4R)-(-)-carvone. The results obtained in the kinetic resolution of carvone indicated that the Bacillus strain could be useful for resolving a racemic mixture of carvone. Bull’s-eye rot, produced by Phlyctema vagabunda, is an important postharvest disease in apples. Current measures to control infection include synthetic fungicides, in addition to the application of copper hydroxide and potassium phosphite. However, growing public concern regarding fungicide residues in food has generated interest in developing non-chemical alternative control methods; biological control is one of the most promising alternatives. In this research, native endophytic yeasts were isolated and evaluated for the biocontrol of P. vagabunda in apples. The mechanisms of action involved were also determined. Our research found 2 isolates, Vishniacozyma victoriae EPL4.5 and EPL29.5, which exhibited biocontrol activity against P. vagabunda at 20 �C in apples, the incidence of bull’s-eye rot was reduced by 39% and 61%, respectively, and the severity of the disease was decreased by 67% and 70%, respectively, when apples were inoculated with these yeasts 24 h before applying the pathogen. The main mechanisms that could be involved in the observed biocontrol activity are the ability to form biofilms and the production of volatile organic compounds. Context Carbonyl compounds, especiallyaldehydes, emitted to the atmosphere, may suffer hydration in aerosols or water droplets in clouds. At the same time, they can react with hydroxyl radicals which may add or abstract hydrogen atoms from these species. The interplay between hydration and hydrogen abstraction is studied using density functional and quantum composite theoretical methods, both in the gas phase and in simulated bulk water. The H-abstraction from the aldehydic and geminal diol forms of formaldehyde, acetaldehyde, glycolaldehyde, glyoxal, methylglyoxal, and acrolein is studied to determine whether the substituent has any noticeable effect in the preference for the abstraction of one form or another. It is found that abstraction of the H-atom adjacent to the carbonyl group gives a more stable radical than same abstraction from the geminal diol in the case of formaldehyde, acetaldehyde, and glycolaldehyde. The presence of a delocalizing group in the Cα (a carbonyl group in glyoxal and methylglyoxal, and a vinyl group in acrolein), reverts this trend, and now the abstraction of the H-atom from the geminal diol gives more stable radicals. A further study was conducted abstracting hydrogen atoms from the other different positions in the species considered, both in the aldehydic and geminal diol forms. Only in the case of glycolaldehyde, the radical formed by H-abstraction from the –CH2OH group is more stable than any of the other radical species. Abstraction of the hydrogen atom in one of the hydroxyl groups in the geminal diol is equivalent to the addition of the •OH radical to the aldehyde. It leads, in some cases, to decomposition into a smaller radical and a neutral molecule. In these cases, some interesting theoretical differences are observed between the results in gas phase and (simulated) bulk solvent, as well as with respect to the method of calculation chosen. Methods DFT (M06-2X, B2PLYP, PW6B95), CCSD(T), and composite (CBS-QB3, jun-ChS, SCVECV-f12) methods using Dunning basis sets and extrapolation to the CBS limit were used to study the energetics of closed shell aldehydes in their keto and geminal-diol forms, as well as the radical derived from them by hydrogen abstraction. This article analyses mathematical modelling from several different perspectives and contrasts modelling with problem solving. Then it describes the ways in which modelling, applications, and problem solving are approached in several Latin American countries. It further describes workshops for secondary schools and for university teachers in which participants engaged in interdisciplinary modelling activities, and the impact these workshops had on modifying their teaching. Based on the experience of these workshops, recommendations are made with respect to promoting modelling in the curricula. Bacterial wilt (BW) caused by Ralstonia solanacearum is responsible for substantial losses in cultivated potato (Solanum tuberosum) crops worldwide. Resistance genes have been identified in wild species; however, introduction of these through classical breeding has achieved only partial resistance, which has been linked to poor agronomic performance. The Arabidopsis thaliana (At) pattern recognition receptor elongation factor-Tu (EF-Tu) receptor (EFR) recognizes the bacterial pathogen-associated molecular pattern EF-Tu (and its derived peptide elf18) to confer anti-bacterial immunity. Previous work has shown that transfer of AtEFR into tomato confers increased resistance to R. solanacearum. Here, we evaluated whether the transgenic expression of AtEFR would similarly increase BW resistance in a commercial potato line (INIA Iporá), as well as in a breeding potato line (09509.6) in which quantitative resistance has been introgressed from the wild potato relative Solanum commersonii. Resistance to R. solanacearum was evaluated by damaged root inoculation under controlled conditions. Both INIA Iporá and 09509.6 potato lines expressing AtEFR showed greater resistance to R. solanacearum, with no detectable bacteria in tubers evaluated by multiplex-PCR and plate counting. Notably, AtEFR expression and the introgression of quantitative resistance from S. commersonii had a significant additive effect in 09509.6-AtEFR lines. These results show that the combination of heterologous expression of AtEFR with quantitative resistance introgressed from wild relatives is a promising strategy to develop BW resistance in potato. In this work an extensive characterization of Co doped Cu2O thin films grown by electrochemical deposition is presented. The doped films showed ferromagnetic behavior, with impressive Curie temperature up to 700 K. Undoped samples also presented magnetic moments at room temperature, but much reduced when compared to doped ones. The observed magnetism in undoped and doped samples was taken as an indicative of a defect-mediated ferromagnetism. Moreover, the sample magnetization was reduced as a consequence of a decreasing in defect density due to thermal annealing. Polarization-dependent X-ray absorption measurements demonstrated that oxygen derived states are the source of the observed magnetic moment and its magnetic dichroism depends on the amount of point defects. Structural, optical and electrical characterization provides information that confirms the incorporation of Co atoms in the Cu2O lattice, without the formation of secondary segregated Co phases. In addition, Electron Paramagnetic Resonance and Glow Discharge Optical Emission Spectroscopy measurements reject the existence of contaminants in the films, dismissing them as the source for the observed magnetism. Computational calculations have shown that Co doping shall locally increases the amount of defects, such as interstitial O, leading to highly oxidized regions from where the unpaired spin arises. We report the effect of La0.57Li0.3TiO3 (LLTO) nanoparticles addition, with different crystallite size, on the lithium conductivity of LLTO-PMMA nanocomposites. X-ray powder diffraction and differential scanning calorimetry analyses showed a moderate increase of the disorder on the polymer chains with the addition of LLTO nanoparticles. Confocal Raman spectroscopy analysis evidenced lithium nitrate dissociation for both LLTO-loaded and unloaded polymer. In the present work, we report the enhancement of lithium-ion dissociation and transport in poly(acrylonitrile) host promoted by the addition of hydrogen titanate nanotube fillers for solid polymer electrolytes. We show experimental and theoretical evidence of lithium perchlorate dissociation due to the presence of the acidic hydrogen titanate nanotubes embedded in the polymer matrix. We performed confocal Raman microscopy analysis to reveal the presence of lithium perchlorate dissociation at the interface of polymer and nanotube fillers. The large affinity of perchlorate anions at the hydrogen titanate nanotube surface, as envisaged from the ab initio molecular dynamics simulations, could be responsible for the enhancement of more than 2 orders of magnitude in the lithium conductivity, reaching &sim;4 × 10–4 S·cm−1 for a certain amount of nanotube fillers addition. The aim of this work was to develop nanomaterials that can be used as sensitizers in radiotherapy treatment. The synthesis of bismuth sulfide (Bi2S3) particles with nanometer size by the hot injection method was studied as well as their behavior when coated with a biocompatible agent. The hot injection method allowed us to synthesize Bi2S3 nanorods measuring in average 4.2 ± 1.4 nm in width and 27.5 ± 16.3 nm in length. Moreover, to improve their biocompatibility, these nanorods were coated with polyvinylpyrrolidone with the purpose of studying the behavior of nanorods in the bloodstream. The results indicated that the suspensions were more stable when the media had protein components which may facilitate the blood circulation of the particles. The toxicity of the nanorods and their radiosensitization potential were evaluated and determined in breast cancer cells (MCF7). These results show that the synthesized nanorods did not evidence cytotoxicity up to a concentration of 2500 μg/mL, over 72 h of incubation. On the other hand, the sensitivity of MCF7 cells with and without nanorods in the culture medium, afterward irradiating them with a dose of 2 Gy 60Co gamma radiation source, showed a 38% increase in cell death when compared to cells that were only irradiated. Our results show that Bi2S3 nanorods are very promising as anticancer agents improving the efficiency of radiotherapy. Response surface methodology (RSM) was applied to optimise laccase production by a new strain of Pycnoporus sanguineus, isolated from the Uruguayan forests, grown on bark shavings under semi-solid-state fermentation conditions. Enhancement of laccase production was studied, with a full 24 factorial design with two replicates and four central points. Operating under the optimal conditions obtained (pH 3.0, yeast extract 10 g L−1, glucose 5 g L−1 and bark shavings 3 g), a laccase activity of nearly 3000 U L−1 was achieved. Subsequently, the effect of different potential inducers on laccase production (Cu+2, Mn+2 and lignin) was tested using the one-factor-at-a-time approach. It was found that the addition of 0.5 mM Cu+2 into the cultures considerably increased laccase production and, thus, maximum activity values of about 16 000 U L−1 were achieved. Further, the optimised culture broth (containing mainly laccase) showed high capacity to decolourise different recalcitrant synthetic dyes such as Acid Red 88 (AR88), Reactive Black 5 (RB5) and Lanaset Grey G (LG). After selecting enzyme concentration and time of reaction, almost 70% of AR88 could be decolourised in the absence of redox mediators. However, mediators were necessary to bleach RB5 and LG. Methyl syringate was the most effective mediator for AR88 (reaching a decolouration degree over 90%) and RB5 (nearly 70%). Around 80% of LG could be decolourised in the presence of violuric acid. These promising results make this enzyme a very useful tool for developing wastewater treatments. Major drawbacks for enzyme-catalyzed transesterification of vegetable oil triglycerides to produce biodiesel (esters of short chain alcohols) are the high concentration of alcohol in the reaction mixture and the deposition of the produced glycerol on the enzyme support. Although stepwise addition of alcohol to the reaction mixture solves the first problem, an effective way to remove glycerol from the reaction system is needed. In the present study the efficiency of three systems for removal of glycerol from the reaction mixture during the enzymatic synthesis of sunflower oil methyl esters was investigated. By coupling a single cold column to the reactor 87.6% of total glycerol produced by the reaction was removed, while 10.9% was still retained by the enzyme. The percentage of glycerol separated from the reaction system increased to 93.1% when a second separation column packed with Amberlite BD10DRY resin was included, reducing the percentage of glycerol retained by the enzyme to 5.8%. Finally, changing the geometry of cold separation column in which a conical decanter was used, the percentage of glycerol separated increased to 99.2%, while only 0.4% was retained by the enzyme. Results demonstrated that use of a convenient separation system such as those described before, glycerol produced during transesterification of sunflower oil can be efficiently removed from the reaction mixture. El diseño y construcción de estructuras supramoleculares polinucleares de iones lantánidos es un campo de investigación de gran interés. Ellas presentan interesantes arquitecturas y propiedades, ya sea magnéticas, ópticas, eléctricas, etc. Los lantánidos (Ln) en su estado de oxidación +3, son iones ideales para el diseño de estas estructuras poliméricas, debido a sus altos índices de coordinación que permiten construir fascinantes estructuras térmicamente estables. El futuro de este tipo de complejos depende de continuar la búsqueda de nuevos compuestos y del cuidadoso diseño y control de la arquitectura final generada. La selección del ligando utilizado es crucial para el diseño de estos compuestos. Un tipo de ligandos de amplio estudio por formar complejos de gran estabilidad y ofrecer diversos modos de coordinación son los ligandos policarboxílicos. En particular el ácido iminodiacético (H2ida) ha mostrado una sorprendente versatilidad gracias a su gran flexibilidad y diversos modos de coordinación. Varias estructuras polinucleares se han reportado con este ligando, siendo relevante la serie [Ln(Hida)3]·xH2O. La particularidad de estos compuestos es la formación de estructuras tridimensionales que contienen canales de unos 11 Å de diámetro. Es de destacar que en estas estructuras se observa la capacidad de este ligando de pasar de ser un ligando tridentado ONO, a un ligando ditópico, que puede coordinar centros metálicos alejados. La protonación del átomo de N es la responsable de estos cambios. En esta misma línea, otras estructuras porosas han sido reportadas para complejos heteropolinucleares conteniendo iones lantánidos, metales de transición 3d y el ligando ida. Un ejemplo de estos son los complejos isoestructurales del tipo [Ln2Cu3(ida)6]·xH2O. Con estos antecedentes, el objetivo de este trabajo es sintetizar nuevas series de complejos polinucleares conteniendo iones lantánidos y ligandos aminodicarboxílicos N-sustituidos derivados del iminodiacetato y caracterizarlos del punto de vista estructural y de sus propiedades. Los ligandos aquí empleados son el ácido N-hidroxietiliminodiacético (H2hida), el ácido N-benciliminodiacético (H2bzlida), el ácido N-metiliminodiacético (H2mida) y el ácido Nmetiliminodipropiónico (H2midp). Con el fin de determinar las mejores condiciones de síntesis se realizaron estudios potenciométricos en solución acuosa. A partir de estos fue posible determinar las especies principales en solución y el rango de pH en el cual pueden ser aislados al estado sólido. Luego se pasó a la etapa de síntesis, donde se obtuvieron nuevos complejos polinucleares con todos los ligandos: [Ln(bzlida)(Hbzlida)]·H2O (Ln = La, Nd), [Ln2(bzlida)3]·3H2O (L n= Nd, Sm, Eu, Gd), [Ho(hida)(H2O)2](CF3SO3)·xH2O, [Sm(Hmida)2(H2O)2]·ClO4·H2O·1/6NaClO4 y [Ln2(Hmidp)3(H2O)3](ClO4)3·3H2O (Ln = Nd, Sm, Eu, Gd, Tm, Dy, Ho). Los complejos fueron caracterizados completamente incluyendo su elucidación estructural mediante difracción de rayos X. Finalmente se evaluaron las propiedades ópticas y magnéticas de una selección de los compuestos obtenidos. Esto permitió relacionar las estructuras obtenidas con las propiedades al estado sólido. El estudio de sus propiedades ópticas permitió determinar que es posible una Resumen 2 transferencia de energía desde los ligandos a los centros metálicos, aunque la eficiencia de la transferencia es baja, siendo los compuestos de Eu los que arrojan los mejores resultados. Al evaluar las propiedades magnéticas se pudo racionalizar el origen del acoplamiento antiferromagnético en el compuesto [Gd2(bzlida)3]·3H2O.
