Furthermore, a single-crystal structure containing alanyl-leucinato ([Cu3(AlaLeu)3 (H2O)3(CO3)]�PF6�H2O), which is the first homotrinuclear carbonato-bridged copper(II) complex with a dipeptide moiety, is presented. To assess possible antitumor action mechanisms, we focused on the comparative analysis of pro- and antioxidant behaviors. Pro-oxidant activity, in which the reactive oxygen species (ROS) formed by the reaction of the complexes with H2O2 produce oxidative damage to 2-deoxy-D-ribose, was evaluated using the TBARS method. Additionally, the antioxidant action was quantified through the superoxide dismutase (SOD)-like activity, using a protocol based on the inhibitory effect of SOD on the reduction of nitrobluetetrazolium (NBT) by the superoxide anion generated by the xanthine/ xanthine oxidase system. Our findings show that Cu–amino acid complexes are strong ROS producers and moderate SOD mimics. Conversely, Cu–dipeptide–phen complexes are good SOD mimics but poor ROS producers. The activity of Cu–dipeptide complexes was strongly dependent on the dipeptide. A DFT computational analysis revealed that complexes with high SOD-like activity tend to display a large dipole moment and condensed-to-copper charge, softness and LUMO contribution. Moreover, good ROS producers have higher global hardness and copper electrophilicity, lower copper softness and flexible and freely accessible coordination polyhedra. El aceite de oliva virgen es el zumo oleoso de las aceitunas que se separa de los demás componentes del fruto. Cuando se obtiene por sistemas de elaboración adecuados y procede de frutos frescos de buena calidad, sin defectos ni alteraciones, y con la adecuada madurez, el aceite posee excepcionales características organolépticas. Es prácticamente el único entre los aceites vegetales que puede consumirse crudo, conservando íntegra su composición en ácidos grasos y el contenido en componentes menores, de elevada importancia saludable-nutricional, destacando el contenido en vitaminas liposolubles y polifenoles (Alba, 2008). Los lípidos son nutrientes necesarios ya que proveen al organismo de energía, de ácidos grasos esenciales (linoleico y linolénico) y esteroides precursores de hormonas, entre otros compuestos de interés, que ayudan a mantener la temperatura corporal y participan en el recambio celular (Valenzuela y Morgado, 2005) En la actualidad Uruguay consume poco aceite de oliva virgen y se pretende aumentarlo, sobretodo el consumo nacional, aunque a nivel mundial existe la tendencia del aumento de dicho consumo. Hay varias causas que justifican dicho aumento, entre las que más se destaca es el deseo de la población por consumir productos saludables. Uruguay quiere caracterizar sus aceites de oliva vírgenes para competir internamente con los aceites extranjeros y para poder exportarlos con perfil propio; algo ya se ha hecho porque se han ganado premios en diferentes concursos a nivel internacional; es por esos postulados que se pretende caracterizar los AOV y ver qué zonas hay que potenciar. La caracterización se realizaría tanto en forma sensorial como química: por estudio del perfil de ácidos grasos y del contenido de antioxidantes. El objetivo general de esta Tesis es el estudio de aceites de oliva vírgenes, variedades Arbequina y Picual, obtenidos a partir de aceitunas provenientes de dos zonas geográficas diferentes del país, con distintos índices de madurez, durante tres cosechas, años 2011, 2012 y 2013 y el estudio de la percepción del consumidor uruguayo sobre el aceite de oliva virgen. 7 Para la escritura de dicha tesis primero se exponen los estudios químicos realizados a los aceites de oliva vírgenes extraídos, luego los estudios sensoriales obtenidos por medio de jueces sensoriales realizados a los mismos aceites y por último los estudios realizados con consumidores de la percepción del consumidor uruguayo sobre el aceite de oliva virgen. Según los resultados de los análisis tanto químicos como sensoriales realizados a todos los aceites de oliva vírgenes estudiados en esta Tesis se concluye que los mismos pertenecen a la categoría aceite de oliva virgen extra ya que cumplieron los requisitos establecidos en la legislación. The proliferation of extra virgin olive oil produced in Uruguay makes necessary the study of Uruguayan consumers’ needs and expectations for olive oils. In this context, the aims of the present work were to gather information about consumers’ perception of olive oil and to compare those perceptions with those of common vegetable oils commonly found in Uruguayan market. A total of 168 consumers completed a word association task about five types of vegetable oil: olive oil, corn oil, sunflower oil, soybean oil and rice bran oil. Consumers were asked to write down the first four images, associations, thoughts or feelings that came to their minds when thinking of each of the oils. The different associations were grouped into eighteen categories and the frequency of mention of each of them was determined for each of the evaluated oils. The evaluated vegetable oils raised clearly different associations in consumers’ mind, suggesting that the personal constructs behind their consumption might differ. Olive oil was perceived as clearly different from the other oils and was described using categories such as Dressing, Good quality, Positive effect on health, and Positive feelings. On the other hand, soybean oil showed the opposite profile, being associated with the categories Bad quality, Price, Negative feelings. The other three oils were perceived as multipurpose common oils and were associated with categories such as Frying, Ingredient in food products, Nutritional content, Consumption occasions, Packaging material, Raw material, Appearance and Texture. Consumption frequency of olive oil strongly affected consumers’ associations of this product, suggesting the importance of familiarity and previous experiences with the product in determining consumers’ perception. The findings of this qualitative study provide a valuable insight for developing marketing strategies for promoting the consumption of olive oil among Uruguayan consumers. Phycocyanin, a blue protein extracted from Spirulina spp, shows promising characteristics that made it suitable for its use as natural dye in photovoltaic devices as the dye sensitized solar cells. In this work, a study of the aqueous solution-phase photochemistry, photophysics, spectroscopy, voltammetry and thermal stability of phycocyanin is presented. Suitable redox potentials (Eox = 1.2 V vs. Ag/AgCl) and a value of 1.96 V for E0,0 (i.e., the energy difference between the vibrationally relaxed levels of the first electronic excited state, S1, and the ground state, S0 of phycocyanin), allows the calculation of energetic profiles that in comparison with the conduction band of the anatase-TiO2 and I-/I3- electrolyte, could predict electron transfer with these components of the cell. The data reported herein should not only help to evaluate the potential use of phycocyanin as sensitizer for solar cells, but should also help in the development of novel solar cells where the photoinduced behavior of this protein can be controlled. En esta Tesis de Doctorado se trabajó sobre diferentes fotosensibilizadores para su uso en celdas solares sensibilizadas con pigmentos (DSSC). Estos dispositivos son celdas electroquímicas que imitan el proceso natural de fotosíntesis, utilizando pigmentos orgánicos que captan la luz incidente y provocan una separación de cargas, seguida por un flujo de electrones, representando una forma alternativa de generación de energía fotovoltaica a bajo costo. En particular, en este trabajo de investigación se estudiaron tres pigmentos naturales diferentes: las antocianinas provenientes de la flor del ceibo, la proteína ficocianina proveniente del alga Spirulina spp y la proteína ficoeritrina proveniente de algas rojas antárticas. Se llevaron adelante procesos de extracción y purificación de los pigmentos a través de cromatografía de exclusión molecular y fase reversa con columnas C18, así como también su caracterización por espectrofotometría UV-visible, espectroscopía de fluorescencia, voltamperometría cíclica y estudios de estabilidad frente a distintos parámetros. Además, se realizaron medidas en microbalanza de cuarzo para determinar la adsorción de los pigmentos al semiconductor de la celda. Por último, se armaron celdas con los distintos fotosensibilizadores y en diferentes condiciones, evaluando su funcionamiento a través de perfiles J vs E y estudios de espectroscopía de impedancia electroquímica para determinar parámetros fotovoltaicos, utilizando un simulador solar. Destacar que varios de estos estudios fueron posibles gracias a la cooperación con el Instituto Tecnológico de Chascomús (Argentina), el Karlsruher Institute für Technologie (Alemania) y la École Politechnique Fédérale de Lausanne (Suiza). We propose 3-(benzyloxy)-1-(5-[18F]fluoropentyl)-5-nitro-1H-indazole, [18F]-IND1, structurally related to the AChE-inhibitor CP126,998, as a new positron emission tomography-radiotracer. Experimental: Radiosynthesis, with 18F, stability, lipophilicity and protein binding of [18F]-IND1 were studied. In vivo behavior, in normal mice and on AD mice models, were also analyzed. Results: [18F]-IND1 was obtained in good radiochemical yield, was stable for at least 2 h in different conditions, and had adequate lipophilicity for blood–brain barrier penetration. Biodistribution studies, in normal mice, showed that [18F]-IND1 was retained in the brain after 1 h. In vivo tacrine-blocking experiments indicated this uptake could be specifically due to AChE interaction. Studies in transgenic AD mice showed differential, compared with normal mice, binding in many brain regions. Conclusion: [18F]-IND1 can be used to detect AChE changes in AD patients. Initiation of celiac disease is triggered in the gastrointestinal tract by transglutaminase 2 (TG2) assisted deamidation of gluten peptides. Deamidation is a side-reaction to transamidation and occurs if primary amines are absent. In contrast to deamidation, transamidation does not trigger an immune response. The aim of the study was to identify a suitable food additive that interacts with TG2 binding motives in gluten-derived peptides to prevent deamidation/transamidation. Homology modelling of α2-gliadin and computational screening of compounds for their binding affinity to a common TG2 binding motive (P)QLP were done by using computational approaches followed by experimental testing of TG2 activity. A database containing 1174 potential food grade ligands was screened against the model of α2-gliadin (27 out of 33 aa). Out of the five best ligands, ascorbyl palmitate, was observed to decrease TG2 transamidation of gliadin by 82% ± 2%. To completely silence the transamidation, we added zinc chloride (ZnCl2), and thereby reached a 99% ± 1% inhibition of TG2 activity. In addition, we conducted a pilot experiment in which ascorbyl palmitate was observed to decrease TG2 deamidation of gliadin completely. We propose ascorbyl palmitate in combination with ZnCl2 with the future perspective to become an additive in celiac-safe foods. Bicarbonate ion reacts with aqueous hydrogen peroxide, yielding monoperoxocarbonate (HCO4 �, PCA�). This species is a better oxidizing agent than hydrogen peroxide itself and has been shown to lead to double bond epoxidation of alkenes under mild conditions. The structure of this anion as well as that of the parent monoperoxocarbonic acid (H2CO4, HPCA) are not known experimentally. The structures of the neutral, anionic and radical species derived from H2CO4 are reported in this paper employing DFT and MP2 methods, both in gas phase and simulating the bulk water solvent using the PCM method. Several conformers are found, of which the most stable one in gas phase has a planar, hydrogen-bonded structure similar to the one observed for peroxyformic acid (PFA). However, optimization of this structure in the bulk solvent leads to a non-planar conformation with a gauche disposition of the bonds in the COOH group. A new CYP26A1 homology model was built based on the crystal structure of cyanobacterial CYP120A1. The model quality was examined for stereochemical accuracy, folding reliability, and absolute quality using a variety of different bioinformatics tools. Furthermore, the docking capabilities of the model were assessed by docking of the natural substrate all-trans-retinoic acid (atRA), and a group of known azole- and tetralone-based CYP26A1 inhibitors. The preferred binding pose of atRA suggests the (4S)-OH-atRA metabolite production, in agreement with recently available experimental data. The distances between the ligands and the heme group iron of the enzyme are in agreement with corresponding distances obtained for substrates and azole inhibitors for other cytochrome systems. The calculated theoretical binding energies agree with recently reported experimental data and show that the model is capable of discriminating between natural substrate, strong inhibitors (R116010 and R115866), and weak inhibitors (liarozole, fluconazole, tetralone derivatives). Environmental sustainability of South American rainfed agroecosystems is of current concern. In this work, we evaluate the occurrence of multiple pesticide residues in muscle tissue of wild fish species from two large rivers in South America (Uruguay and Negro Rivers). Two sampling campaigns (representing summer and winter crops) were performed during 2015 targeting a wide biodiversity of fish species used for human consumption (ranging from migratory to non-migratory and from detritivorous to top-predators). Three different localities associated to rainfed agriculture were assessed, two of them enclosed to a RAMSAR site (National Park “Esteros de Farrapos e Islas del Rio Uruguay”). Pesticide residues occurred in muscle tissue of 143 from 149 sampled fishes (96%). Thirty different pesticides were detected at concentrations from <1 to 194 μg kg−1. Incidence of pesticides in fish were tightly related to: i) features of the contaminant: (Kow, environmental persistence and mobility) and ii) intensity of use of particular pesticides and land dedicated to rainfed agriculture. Trifloxystrobin, metolachlor and pyraclostrobin showed the highest rates of occurrence. Of great concern is that strobirulins have highest toxicity to fish from those detected compounds. From the pattern of pesticides occurring for non-migratory fish species it was possible to trend important spatial differences related to the intensity of rainfed agriculture. Results suggest a regular exposition of aquatic wild biota to sublethal concentrations of multiple semi-polar pesticides. Según estudios recientes realizados en Europa y América, la mayoría de las opiniones de la población sobre las aplicaciones de la radiactividad son negativas y están influenciadas muchas veces por los accidentes nucleares ocurridos en la historia, el desconocimiento o la falta de información confiable sobre el tema y las concepciones previas erróneas (Montes, Damonte, Errico y Taylor, 2015). Respecto a la enseñanza de la radiactividad y las ideas de los estudiantes, estudios realizados en Europa y Argentina reflejaron el escaso abordaje de esta temática en la currícula (García-Carmona y Criado, 2008), la falta de conocimiento (Lijnse, Eijkelhof, Klaasen y Scholte, 1990; Millar, Klaasen y Eijkelhof, 1990; Gutiérrez, Capuano, Perrotta, De la Fuente, y Follari, 2000; Cornejo, Speltini, Roble y Santilli, 2009) y las ideas previas erróneas de los estudiantes (Corbelle y Domínguez, 2015; Corbelle y Domínguez, 2016; Lavín y Mínguez, 2016). En Uruguay no hay antecedentes de revisiones de los libros de texto, de los contenidos que se enseñan, ni de las metodologías aplicadas. Teniendo en cuenta los antecedentes descritos anteriormente, esta investigación tuvo como objetivo analizar las posibilidades de construcción de la imagen perceptiva del estudiante de Enseñanza Media uruguayo sobre la radiactividad, a partir del conocimiento de la temática. En una primera etapa se analizó el enfoque de la radiactividad en la currícula de Educación Media y en los libros de texto recomendados por la inspección docente, y se indagó sobre la imagen mental del estudiante uruguayo sobre la radiactividad. En una segunda etapa, el docente generó instancias de trabajo en los entornos virtuales de aprendizaje (en el contexto de la pandemia por Covid-19), en dónde orientó a los estudiantes en la selección de los contenidos y en la elaboración de los recursos educativos abiertos; para mejorar la comprensión de la temática y promover la construcción de una imagen perceptiva fundamentada. La metodología empleada en la elaboración de los recursos fue el Aprendizaje Basado en Proyectos (Rodríguez-Sandoval, Vargas-Solano y Luna-Cortés, 2010, Aristizabal, 2012). Los recursos elaborados por los estudiantes fueron publicados en el repositorio institucional y compartidos a través de las plataformas educativas utilizadas en Enseñanza Media. En una tercera etapa, el docente aplicó un cuestionario con contenidos conceptuales de la temática y actitudinales del proyecto desarrollado, para analizar las posibilidades de construcción de la imagen perceptiva del estudiante sobre la radiactividad. Se concluyó que tanto la currícula como los libros de texto no consideran a la radiactividad un tema fundamental y no profundizan algunos conceptos claves relacionados a la temática (Cabanyes, 2000). La currícula actual de Bachillerato sugiere estrategias de aprendizaje generales para potenciar la construcción de los saberes. La mayoría de los estudiantes indagados, tuvieron una imagen mental negativa de la radiactividad. Los estudiantes que participaron en la elaboración de los recursos educativos abiertos mejoraron la comprensión de la temática de la radiactividad y consideraron tener una percepción más favorable, una vez finalizado el Proyecto. Por lo anterior, se recomienda continuar trabajando en el diseño y difusión de los recursos educativos abiertos en entornos virtuales, promoviendo la interacción de los estudiantes con el docente y mejorando la calidad de los aprendizajes de la Química y otras disciplinas. This antigen was detected in both larval and adult worm extracts, with the highest specific activity observed in the adult excretion/secretion preparation. Histochemical analysis showed that Tn is preferentially expressed in the parenchyma in both parasite stages and the external part of tegument in adult worms. A similar pattern was observed for sialyl-Tn, a related O-linked antigen. Tn glycoproteins from protoscoleces were resolved by SDS-PAGE in two main components of 43 and 49 kDa. After purification, this material was reactive with lectins which bind GlcNAc/sialic acid, GalNAc, and T antigen. In a preliminary evaluation, high levels of Tn antigen were detected in serum samples from patients with hydatid cyst, suggesting that the measure of Tn in serum could be a biomarker of this disease, although extensive work is necessary in order to determine the clinical usefulness of this assay. The results reported here, the first evidence of O-glycosylation pathways in E. granulosus and the presence of Tn antigen in cestodes, suggest that the evaluation of O-glycosylated antigens might give new insights in the host-parasite relationship. The syntheses and biological evaluation of the first anti-protozoa imidazole N-oxide and benzimidazole N-oxide and their derivatives are reported. They were tested in vitro against two different protozoa, Trypanosoma cruzi and Trichomonas vaginalis. The aroma profile of five premium red wines has been studied by sensory descriptive analysis, quantitative gas chromatography-olfactometry (GC-O), and chemical quantitative analysis. The most relevant findings have been confirmed by sensory analysis. Forty-five odorants, including the most intense, were identified. At least 37 odorants can be found at concentrations above their odor threshold. A satisfactory agreement between GC-O and quantitative data was obtained in most cases. Isobutyl- 2-methoxypyrazine, (E)-whiskey lactone, and guaiacol were responsible for the veggie, woody, and toasted characters of the wines, respectively. The sweet-caramel notes are related to the presence of at least five compounds with flowery and sweet notes. The phenolic character can be similarly related to the presence of 12 volatile phenols. The berry fruit note of these wines is related to the additive effect of nine fruity esters. Ethanol exerts a strong suppression effect on fruitiness, whereas norisoprenoids and dimethyl sulfide enhance fruity notes.
