In spite of high plant growth regulator application rates, little has been reported in the literature on determination of their residues in fruits and vegetables. This would be useful in monitoring good manufacturing practices and overall safety through the enforcement of maximum residue levels (MRLs). The present work describes method validation for the determination of 1-naphthylacetic acid (NAA) and 2(1-naphthyl)acetamide (NAAm) in tomato and zucchini using the mini-Luke, ethyl acetate (EtOAc) and acetate-buffered quick, easy, cheap, effective, rugged, and safe (QuEChERS) methods. Samples were spiked at two different levels: 50 and 100 &mu;g/kg for NAA and 20 and 100 &mu;g/kg for NAAm. These compounds were analyzed within the same chromatographic run with LC coupled to triple quadrupole MS (LC/(QqQ)MS/MS) in positive and negative electrospray ionization [ESI(+) and ESI(&ndash;)] modes for NAAm and NAA, respectively. For analyte confirmation, LC/ESI(-)QTOF-MS was also investigated given that NAA has only one multiple reaction monitoring transition (185.1&rarr;140.9 m/z). These three common methods were used to determine linearity, recoveries, precision (RSD), matrix effects, repeatability, and reproducibility (n = 5) for the selected matrixes. In terms of the Directorate-General for Health and Consumers (DG-SANCO) guidelines, only insignificant differences were found for the multiresidue methods tested, regardless of the commodity. Matrix-matched calibration was used, and LODs were below 10.1 &mu;g/kg for NAA and 6.0 &mu;g/kg for NAAm, which were lower than the MRLs established in current European Union legislation for these compounds. Obtained recoveries for NAA ranged from 87 to 107% with RSD values below 10% for mini-Luke, 83 to 107% with RSD 0.996) for all methods used. Good repeatability and reproducibility in terms of RSD were obtained (11.3&ndash;17.8%) for these analytical methods. The results were acceptable within the studied ranges for both analytes in all methods. As these multiresidue methods are used at the present time in many laboratories, this survey indicates that these analytes should be included in their monitoring scope. Manuka honey is a valuable commodity produced by bees foraging the flowers of Leptospermum scoparium, a bush native to New Zealand and Australia. Due to its high value and proven health benefits, authenticity fraud in the sale of this food is a significant risk, as recounted in the literature. Four compulsory natural products must be present at minimum concentrations to authenticate manuka honey (3-phenyllactic acid, 2&prime; -methox yacetophenone, 2-methoxybenzoic acid, and 4-hydroxyphenyllactic acid). However, spiking other kinds of honey with these compounds and/or the dilution of manuka honey with other varieties may result in fraud going undetected. In this work, liquid chromatography coupled with high-resolution mass spectrometry and a metabolomics-based strategy has allowed us to tentatively identify 19 natural products &ndash;putative manuka honey markers&ndash;, nine of which are reported for the first time. Chemometric models applied to these markers allowed the detection of both spiking and dilution fraud attempts of manuka honey, even at 75% manuka honey purity. Thus, the herein-reported methodology can be employed in the prevention and detection of manuka honey adulteration even at low levels, and the tentatively identified markers presented in this work proved valuable for manuka honey authentication procedures. On the other hand, a comparison of quantitative and qualitative strategies based on direct injection analysis and off-line solid-phase extraction sample treatment were assayed using two different QLIT instruments for a selected group of emerging contaminants when operating in selected reaction monitoring (SRM) and information-dependent acquisition (IDA) modes. Results and discussion Software-aided screening usually needs a further confirmatory step. Resolving power and MS/MS feature of QTOF showed to confirm/reject most findings in river water, although sensitivity-related limitations are usually found. Superior sensitivity of modern QLIT-MS/MS offered the possibility of direct injection analysis for proper quantitative study of a variety of contaminants, while it simultaneously reduced the matrix effect and increased the reliability of the results. Confirmation of ethylamphetamine, which lacks on a second SRM transition, was accomplished by using the IDA feature. Conclusion Hybrid MS instruments equipped with high resolution and high sensitivity contributes to enlarge the scope of targeted analytes in river waters. Considering as a whole, these results may imply a great antidiabetic and antiobesity potential. Moreover, EHAA showed to have anti-inflammatory capacity (IC50 587 &micro;g/mL) on RAW 264.7 cells. The incubation was performed with and without U-Caspofungin. In vivo evaluation was performed of 6 groups of athymic mice: sham, inflammation (LPS), fungal infection with Candida albicans (CA) and bacterial infection with Staphylocuccus aureus (SA). In vitro studies showed that the uptake of the complex by both yeasts and bacteria, depends on the concentration of colony forming units (cfu), and that this uptake is favoured by the presence of UCaspofungin that increases the membrane permeability to the 99mTc-tricarbonyl-Caspofungin complex. In vivo evaluation showed low uptake in sterile inflammation model and moderate to high uptake in infections models both treated or not with U-Caspofungin. The results of biodistributions were compatible with scintigraphic images. Conclusion: The uptake of the complex depends on the load of microorganisms, but it presents high sensibility, even at low concentrations of the infecting agent. The treatment with U-Caspofungin has no influence on the quality of the scintigraphic images used for diagnosis and localization of infection foci. The production, under environmentally benign conditions, of efficient and more cost-effective anti-infective agents (available to the whole mankind) is one of most ambitious dreams of the industrial medicinal chemistry. Semi-synthetic b-lactam antibiotics are very effective anti-infective agents. They are very stable and can be used via oral delivery. They exhibit a very wide spectrum of anti-bacterial activity and minimal side effects after being massively used for a very long time. In this way, we can assume that semi-synthetic b-lactam antibiotics are going to be one of the key anti-infective agents in the next years. The condensation of natural or modified antibiotic nuclei with different acyl donor chains is one of the key steps for the industrial synthesis of these anti-infective agents. Up to now, these condensations are mainly carried out through classical chemical methods and this implies a number of economical, ecological and technological drawbacks (high energy requirements, many protection and deprotection steps, utilization of toxic methylene chloride as solvent, etc). Enzyme biocatalysts may be very useful to catalyse these selective condensations under very mild experimental and environmental conditions. In fact, the possibility of using enzymes to carry out such biotransformations, at laboratory scale, has been discussed and demonstrated a long time ago. However, industrial synthesis of b-lactamic antibiotics is still carried out via unfavorable chemical routes. In fact, enzymes have not been produced in the nature to act in industrial reactors; so they are usually very unstable, inhibited by substrates and products and they may not have ideal catalytic properties for industrial uses (high reaction rates, required selectivity, ability to reach quantitative synthetic yields, stable enough to run a number of reaction cycles, etc). These limitations of enzyme biocatalysis become even more significant mainly when the enzyme is going to be used with non-natural substrates, catalysing non-natural processes and working under non-conventional conditions. However, in the last ten years, a great number of papers have reported substantial improvement of these enzymatic synthetic approaches (new enzymes, better enzyme derivatives, improved reaction designs and so on) and it seems that a massive industrial implementation of enzymes in antibiotic synthesis is approaching. The surface carboxylic groups of penicillin G acylase and glutaryl acylase were chemically aminated in a controlled way by reaction with ethylenediamine via the 1-ethyl-3-(dimethylamino-propyl) carbodiimide coupling method. Then, both proteins were immobilized on glyoxyl agarose. In both cases, the immobilization of the chemically modified enzymes improved the enzyme stability compared to the stability of the immobilized but non-modified enzyme (by a four-fold factor in the case of PGA and a 20-fold factor in the case of GA). The chemical modification presented a deleterious effect on soluble enzyme stability. Therefore, the improved stability should be related to a higher multipoint covalent attachment, involving both the lysine amino groups and also the new amino groups chemically introduced on the enzyme. Moreover, the lower pKa of the new amino groups permitted to immobilize the enzyme under milder conditions. In fact, the aminated proteins could be immobilized even at pH 9, while the non-modified enzymes could only be immobilized at pH over 10. Este art&iacute;culo presenta reflexiones sobre la experiencia de trabajo de la red conformada por la Universidad Nacional de Tierra del Fuego, Ant&aacute;rtida e Islas del Atl&aacute;ntico Sur de Argentina y el Grupo de Educaci&oacute;n Ambiental de la Red Tem&aacute;tica de Medio Ambiente de UdelaR, en el marco del programa: &ldquo;N&uacute;cleo de Estudios e Investigaci&oacute;n en Educaci&oacute;n Superior del MERCOSUR (NEIES-MERCOSUR)&rdquo;. El proyecto apunta a repensar el papel de las universidades del MERCOSUR ante la actual crisis ambiental, en el cruce problem&aacute;tico entre los procesos de globalizaci&oacute;n y el desarrollo sustentable. La produccion biologica de metano a partir de materia organica es un procedimiento eficaz para reducir la contaminacion de efluentes domesticos e industriales. Las bacterias que intervienen en el proceso componen diferentes grupos troficos que actuan en forma secuencial, la ultima etapa de las cuales es la metanogenesis. En esta tesis se investigo el efecto del Cr+3, un toxico potencial presente en las aguas residuales y en los lodos de las curtiembres, sobre las bacterias de estos diferentes grupos troficost Se establecio un orden de sensibilidad y se determino que las mas afectadas fueron las bacterias metanogenicas degradadoras de acetato. Se encontro correlacion entre la actividad metabolica de estos microorganismos en cultivo puro y formando parte de un ecosistema bacteriano complejo. Ademas de la ,concentracion de cromo, totros parametros como la concentracion de biomasa 0 la presencia de ion ferrico modificaron el comportamiento del sistema y podrfan manejarse para mejorar la eficiencia del proceso Se establecio que las bacterias metanogenicas acetotrofas sobreviven a concentraciones de cromo muy superiores alas inhibitorias de la metanogenesis y que tienen cierta capacidad de adaptacion. Estos resultados tambien pueden contribufr a minimizar la inhibicion de la metanogenesis en un digestor anaerobio. Los ensayos con cultivos puros de bacterias metanogenicas acetotrofas permitieron inferir algunos aspectos del mecanisme de acci6n del cromo. La inhibici6n de la metanogenesis depende de la concentraci&oacute;n de i&oacute;n libre 0 quelado, no interfiere con la captura de metales esenciales, y difiere cuantitativamente segun el sustrato utilizado. La diferencia de tolerancia que se observ&oacute; entre las bacterias metanogenicas de los dos unicos Generos capaces de degradar acetato permite predecir el comportamiento de un digestor anaerobio alimentado con este tipo de efluente y configurar una hip&oacute;tesis sobre el desarrollo de estos microorganismos en presencia de un grupo mas amplio de t&oacute;xicos. In this work, we model TiO2(B) angstrom-scale wires derived from TiO2(B) (001) ultrathin sheets. By means of density functional theory (DFT) calculations, we study their structural, electronic and thermodynamic properties, comparing with anatase angstrom scale wire with z as growing direction, of which there is experimental evidence. We found locally stable structures. TiO2(B) atomic wire oriented along y direction exhibits significantly lower formation energy (0.33 eV) than the other studied systems (1.05 eV for TiO2(B) wire oriented along x- and 0.88 eV for anatase wire oriented along z-direction). Additionally, we present the simulated X-ray powder diffraction diagrams in order to help the experimental identification of this potential titanium dioxide polymorph&rsquo;s nanostructures. These results could be interesting for their potential applications, such as energy storage and photovoltaic applications, and for fundamental studies as well. In this work we present the synthesis and full structural characterization of hydrothermally synthesized titanate nanotubes (TNTs) for energy applications. It is of interest to characterize the building block of certain nanomaterials and their surface termination. This task is particularly difficult for nanotubes, where the exhibited surface curvature is a limitation for its study. We present the synthesis and thorough structural characterization through several experimental techniques for morphology and compositional characterization. Simulated X-rays powder diffraction patterns (XRPD) of real size nanotubes models are refined with experimental data. The building blocks for the TNT models were obtained from computational simulations by means of Density Functional Theory for H2Ti2O5&middot;0.5H2O, H2Ti2O5&middot;H2O, H2Ti3O7 and H2Ti3O7&middot;0.67H2O. Our procedure confirms that open‐ended TNTs are obtained, with &sim;100 nm of length and 6(1) nm of inner diameter. By interpreting the XRPD data, the TNT building block was obtained and found to derive from H2Ti3O7, where the distance between [TiO6] octahedral layers is increased in accordance with H2Ti3O7&middot;0.67H2O. This model fully agrees with all the structural characterizations, validating this methodology and suggesting its potential use to study other nanomaterials. The polarity and accessibility to the interior of several dendrimers using phenanthrene, anthracene and tetrabenzonaphtalene as probe molecules have been investigated. In addition the prospective application of the dendrimers as drug carriers was evaluated by incorporating 5(6)-methylbenzo[1,2-c]1,2,5-oxadiazole N 1-oxide] (1) and 2&prime;-(benzo[1,2-c] 1,2,5-oxadiazol-5(6)-yl(N1-oxide) methylidene]-1-methoxy methane hydrazide (2). These compounds have antichagasic therapeutic activity but very low water solubility, which limits their application. Polypropylene imine dendrimers with amine terminal groups (DAB-16AT and DAB-32AT) and polyamide amine (PAMAM) dendrimers with carboxylate terminal groups (PAMAM-32CT), with amine terminal groups, (PAMAM-8AT and PAMAM-32AT) and with hydroxyl terminal groups (PAMAM-32OHT) were chosen for this study. Approximately one molecule of phenanthrene or anthracene was encapsulated in PAMAM-32CT, PAMAM-32AT, PAMAM-32OHT and DAB-32AT dendrimers. However, slight encapsulation was observed working with PAMAM-8AT and DAB-16AT. The studies with tetrabenzonaphtalene show that the guest molecule might only be partially caged within the dendrimer host. However, for relatively insoluble solutes the efficiency to encapsulate can be dictated by the saturation in the aqueous phase besides the dendrimer capacity to dissolve it. These dendrimers are also able to encapsulate and consequently solubilize 1 and 2 oxadiazol. However, PAMAM dendrimers are better for encapsulation and retention due to guest-host specific interactions. El objetivo de esta Tesis fue encontrar nuevas lipasas, extraidas de semillas germinantes que presentaran una actividad hidrolitica importante frente a un a~ Se parti&oacute; de la hip&oacute;tesis de trabajo de que las semillas que tuvieran un porcentaje de aceite elevado como reserva energetica podrian poseer lipasas activas para hidrolizarlo in situ durante su germinaci&oacute;n. En primer lugar se hizo una busqueda de semillas de distintas familias. Las semillas estudiadas fueron achicoria de raiz (Cichorium intybus), cardo de Castilla (Cynara cardunculus ), girasol (Helianthus annuus L.), colza variedad 00 (Brassica napus cv. Ceres), nabiza (Brassica napus L.), nabo (Brassica napus L. cultivada), repollo blanco (Brassica oleracea L. var. capitata L.), repollo crespo (Brassica oleracea L.), rabanito de punta blanca (Raphanus sativus L. var. Acanthiformis Makino), morr6n (Capsicum annum L.), tomate perita (Lycopersicon esculentum L.), zanahoria (Daucus carota), zapallo de cascara dura (Cucurbita pepo), remolacha (Beta vulgaris L. ). Se analiz&oacute; su contenido en aceite, asi como otros parametros tales como el contenido de agua y la velocidad de germinaci6n. Esta se consider&oacute; como un indice indirecto de la actividad de las lipasas presentes. Una vez que se encontraron las semillas con las caracteristicas buscadas (alto contenido en aceite y periodo de germinaci&oacute;n de duraci6n intermedia, menor de 16 dias) se determin&oacute;, para cada uno de sus distintos estadios de germinaci6n, la actividad hidrolitica end&oacute;gena o in vivo y la actividad hidrolitica ex&oacute;gena o in vitro de los crudos preparados con ellas. Todas las semillas presentaron una disminuci6n poco significativa (menos dell 0%) de su contenido en aceite y un aumento de masa apreciable antes del estadio E2 . Todas las semi lIas presentaron algo de actividad hidrolitica in vitro, desde el estadio E1 de su germinaci6n, aunque esta estuvo ausente en todas las semillas sin germinar. En general, no se not&oacute; un aumento de la actividad hidrolitica in vitro (medida por el ensayo descripto en la Tesis) a medida que avanz6 la germinaci6n yen la mayoria de los casos, mas bien esta decreci&oacute; (10 cual no era esperable, de acuerdo con e1 metabolismo normal de las semillas). Solamente las semillas de colza, repollo blanco y tomate perita tuvieron una tendencia clara a alcanzar un maximo en el estadio E4. Los maximos grados de hidr61isis ex&oacute;gena obtenidos, en general, fueron muy bajos (excepto para colza: 47.6%) : achicoria de raiz 4.6%, cardo 11.5%, girasol 17.1 %, morr6n 3.5%, nabiza 9.7%, nabo 13.8%, rabanito 7.3%, repollo blanco 19.4%, repollo crespo 7.0%, tomate perita 17.2%, zanahoria 2.6% y zapallo de cascara dura 2.3%. Se realiz6 un estudio comparativo de las semillas de la familia Brassica: colza variedad 00 (Brassica napus cv. Ceres), nabiza (Brassica napus L.), nabo (Brassica napus L. cultivada), repollo blanco (Brassica oleracea L. var. capitata L.), repollo crespo (Brassica oleracea L.), rabanito de punta blanca (Raphanus sativus L. var. Acanthiformis Makino) ). Dicho estudio indic6 que las semillas no presentaron un comportamiento similar durante la germinaci6n. En particular, las de nabiza, nabo y repollo crespo dieron lugar a crudos con baja capacidad de hidr&oacute;lisis ex&oacute;gena y las de colza y repollo blanco, con aha capacidad. Luego de realizar los estudios comparativos de la capacidad de hidr61isis end&oacute;gena y ex6gena, en funci6n de los resultados obtenidos se eligieron las semillas cuyos crudos presentaron mayor capacidad de hidr61isis ex&oacute;gena. Estas semillas fueron cardo, repollo blanco y tomate perita. Con los crudos de estas semillas se optimizaron las condiciones del ensayo de actividad ex6gena: pH, relaci6n gramos semillas I mL buffer, relaci6n gramos crudol mL aceite, tiempo de reacci6n. En base a ello se pudo concIuir que la sensibilidad del grado de hidr&oacute;lisis ex&oacute;gena respecto de cada uno de estos parametros dependi&oacute; de la semilla estudiada. Sin embargo, en val ores absolutos, la optimizaci&oacute;n de estos parametros no modific6 de manera importante el grado de hidr&oacute;lisis ex&oacute;gena. Se realiz&oacute; la concentraci6n de las lipasas contenidas en las semillas de cardo, repollo blanco, repollo crespo, nabiza y tomate perita por el metoda de "acetone powder". Se estudi6 el comportamiento de los polvos de acetona frente a distintos sustratos para evaluar la existencia de alglin tipo de especificidad. La comparaci&oacute;n del comportamiento de estas lipasas frente a los distintos aceites de origen vegetal (coco, girasol y oliva) no indic&oacute; ninguna especificidad, ya que la fracci&oacute;n hidrolizada present6 una composici6n igual a la del aceite original. Cuando las lipasas actuaron sobre un aceite de pescado se observ6 que no habia hidr61isis de los acidos grasos con mas de tres insaturaciones. Las lipasas estudiadas bajo forma de "acetone-powder" de semillas germinantes de nabiza, repollo blanco, repollo crespo, tomate y cardo en el estadio E4 presentaron un comportamiento similar. Se estudi6 el comportamiento de las lipasas purificadas por el metodo de "acetone powder" frente al agregado de distintos cofactores: cloruro de calcio, cloruro de magnesio y Trit6n X 100. Se pudo concluir, en forma general, que los cationes calcio y magnesio y el tensoactivo Trit6n Xl 00 utilizados en las concentraciones indicadas en esta tesis, produjeron una marcada disminuci6n en la actividad de todas las lipasas estudiadas. We compared chemical profiles and in vitro biological activity of extracts obtained through maceration with ethanol or supercritical fluid extraction with CO2 (scCO2), from two different cannabis inflorescences chemotypes, one with high total tetrahydrocannabinol (THC) and other with high total cannabidiol (CBD) content. scCO2 was performed to obtain extracts with highest total THC and CBD recovery using ethanol as a co-solvent. In addition, a short maceration with ethanol was performed for both chemotypes. Cannabinoid and terpene composition of the extracts were determined using chromatographic and spectroscopic methods, and their cytotoxicity and cell proliferation effects were evaluated in vitro using HT-29 colon adenocarcinoma cell line. In vitro biological activity confirmed that cannabis extracts from both chemotypes have anti-proliferative activity but the scCO2 extract from high total THC chemotype was more cytotoxic. These results underscore the importance of chemotype and extraction method selection in the preparation of cannabis extracts for different therapeutic applications. While Cannabis sativa L. varieties have been traditionally characterized by their major cannabinoid profile, it is now well established that other plant metabolites can also have physiological effects, including minor cannabinoids, terpenes, and flavonoids. Given the multiple applications of cannabis in the medical field, it is therefore critical to characterize it according to its chemical composition (i.e., its metabolome) and not only its botanical traits. With this in mind, the cannabinoid and metabolomic profiles from inflorescences of two C. sativa varieties with either high D9 -tetrahydrocannabinolic acid (THCA) or high cannabidiolic acid (CBDA) contents harvested at different times were studied. According to results from HPLC and NMR-based untargeted metabolomic analyses of organic and aqueous plant material extracts, we show that in addition to expected variations according to cannabinoid profiles, it is possible to distinguish between harvests of the same variety. In particular, it was possible to correlate variations in the metabolome with presence of powdery mildew, leading to the identification of molecular markers associated with this fungal infection in C. sativa. With the aim to develop new potential 99mTc-radiopharmaceuticals for imaging hypoxia based on the formation of Tc-nitrido complexes, two novel dithiocarbamate containing metronidazole derivatives (L1 and L2) have been prepared and characterised. The synthesis of L1 and L2 was achieved in excellent yield and high purity. Labelling with 99mTc was successfully performed using a low ligand concentration (approximately 2&ndash;3 mg) and the desired products were obtained with high radiochemical purity (&gt;90%). Lipophilicity, plasma protein binding, and biodistribution in normal- and tumour-bearing-CD1 mice studies were performed to asses the potentiality for nuclear medicine oncology. According to the physicochemical and biological behaviour both in healthy animals and in animals bearing solid tumours complex dtcTc1 could be considered as a starting point for the development of new radiopharmaceuticals for imaging hypoxia.
