The Aclepiadaceae family has been reported by its use in ethnomedicine. In the northeas of Argentina, A. mellodora and A. curassavica are traditionally used in ofidic accidents as poultices. In this work, aqueous, alcoholic and hexanoic extracts were analyzed by SDS-PAGE to determine their anti-snake activity. The in vitro study of the inhibitory ability of the following activities: proteolytic, indirect hemolytic activity and inhibition of the coagulant activity, allowed demonstrating that both species were active against venom, being A. mellodora the most active. Statistically, all extracts of A. mellodora were active against venom in the inhibition of the coagulant activity, without significant differences with respect to the organ used; which is consistent with the traditional use as external poultice. The alcoholic extract of A. mellodora roots was subjected to a bio-guided separation. The fractions obtained were enriched in compounds which could probably be responsible for the activity against venom. Two fatty acid binding proteins, MvFABPa and MvFABPb were identified in the parasite Mesocestoides vogae (Platyhelmithes, Cestoda). Fatty acid binding proteins are small intracellular proteins whose members exhibit great diversity. Proteins of this family have been identified in many organisms, of which Platyhelminthes are among the most primitive. These proteins have particular relevance in flatworms since de novo synthesis of fatty acids is absent. Fatty acids should be captured from the media needing an efficient transport system to uptake and distribute these molecules. While HLBPs could be involved in the shuttle of fatty acids to the surrounding host tissues and convey them into the parasite, FABPs could be responsible for the intracellular trafficking. In an effort to understand the role of MvFABPs in fatty acid transport of M. vogae larvae, we analysed the intracellular localization of both MvFABPs and the co-localization with in vivo uptake of fatty acid analogue BODIPY FL C16. Immunohistochemical studies on larvae sections using specific antibodies, showed a diffuse cytoplasmic distribution of each protein with some expression in nuclei and mitochondria. MvFABPs distribution was confirmed by mass spectrometry identification from 2D-electrophoresis of larvae subcellular fractions. This work is the first report showing intracellular distribution of MvFABPs as well as the co-localization of these proteins with the BODIPY FL C16 incorporated from the media. Our results suggest that fatty acid binding proteins could target fatty acids to cellular compartments including nuclei. In this sense, M. vogae FABPs could participate in several cellular processes fulfilling most of the functions attributed to vertebrate’s counterparts. La química inorgánica médica constituye un área joven y en creciente expansión que ha conducido al desarrollo de gran número de compuestos metálicos bioactivos, varios de los cuales ya se encuentran en uso clínico o en fase preclínica. La gran diversidad de la química de coordinación y organometálica de los metales de transición ofrece amplias posibilidades para el desarrollo de fármacos metálicos. En particular, los compuestos de coordinación muestran variados números de coordinación y geometrías, y estados de oxidación y potenciales de reducción accesibles in vivo, junto con interesantes características termodinámicas y cinéticas que pueden sufrir un ajuste fino por modificación de la esfera de coordinación del metal. Además, la coordinación a metales puede cambiar de forma significativa las propiedades biológicas de los ligandos orgánicos, pudiendo conducir a modificaciones favorables en la actividad biológica de los mismos. La presencia del metal en estos compuestos determina que puedan ejercer su acción terapéutica por medio de mecanismos no convencionales, actuando a nivel de nuevas dianas. Como consecuencia de un esfuerzo innovador relativamente reciente, el desarrollo de compuestos metálicos bioactivos constituye una estrategia atractiva y prometedora en la búsqueda de nuevas herramientas terapéuticas para el tratamiento de la enfermedad de Chagas. En el presente capítulo se describen esfuerzos realizados para desarrollar este tipo de compuestos. In the search for new metal-based drugs against diseases produced by trypanosomatid parasites, four organoruthenium(II) compounds [Ru2(p-cymene)2(L)2] X2, where L are bioactive 5-nitrofuryl-containing thiosemicarbazones and X=Cl or PF6, had been previously obtained. These compounds had shown activity on Trypanosoma brucei, the etiological agent of African trypanosomiasis. Because of genomic similarities between trypanosomatides, these ruthenium compounds were evaluated, in the current work, on Trypanosoma cruzi, the parasite responsible of American trypanosomiasis (Chagas disease). Two of them showed significant in vitro growth inhibition activity against the infective trypomastigote form of T. cruzi (Dm28c clone, IC50=11.69 and 59.42 μM for [Ru2(p-cymene)2(L4)2]Cl2 and [Ru2(p-cymene)2(L1)2]Cl2, respectively, where HL4=5- nitrofuryl-N-phenylthiosemicarbazone and HL1=5- nitrofurylthiosemicarbazone), showing fairly good selectivities toward trypanosomes with respect to mammalian cells (J774 murine macrophages). Moreover, [Ru2 (p-cymene)2(L2)2]Cl2, where HL2=5-nitrofuryl-Nmethylthiosemicarbazone, was synthesized in order to evaluate the effect of improved solubility on biological behavior. This new chloride salt showed higher activity against T. cruzi than that of the previously synthesized hexafluorophosphate one (Dm28c clone, IC50=14.30 μM for the former and 231.3 μM for the latter). In the search for drugs with anti-trypanosome activity, we had previously synthesized two series of platinum and palladium analogous compounds of the formula [MIICl2(HL)], where HL were bioactive 5-nitrofuryl or 5- nitroacroleine thiosemicarbazone derivatives. In this work, we thoroughly characterized [MIICl2(HL)] complexes interaction with DNA by using different techniques: gel electrophoresis, DNA viscosity measurements, circular dichroism (CD) and atomic force microscopy (AFM). Electrophoresis results showed that all complexes induced a withdrawal of DNA superhelicity demonstrated by a decrease in electrophoretic mobility of supercoiled DNA form. This effect on migration was dependent on dose but also on the nature of both the metal and the ligand. In general, the effect produced by palladium complexes was significantly more intense than that observed for the corresponding platinum analogs. Differences between palladium and platinum complexes were also observed in CD experiments. While palladium complexes induce evident calf thymus (CT)-DNA profile changes compatible with B-DNA to Z-DNA conformational transition, no clear effect was observed for platinum ones. Additionally, AFM studies showed that changes in the shape of plasmid DNA, like supercoiling, kinks and thickness increase resulted more intense for the former. In addition, either Pd or Pt complexes increased the viscosity of CT DNA solutions in a concentration dependent manner. Vanadium compounds display important pharmacological actions in vivo and in vitro systems. Semicarbazones are versatile ligands with therapeutic effects. Herein, we report the effects of VVO2(salicylaldehydesemicarbazone) (V(V)-Salsem) on two osteoblast cell lines in culture (MC3T3-E1 and UMR106). V(V)-Salsem inhibited cell proliferation in a dose response manner. At 100 lM, the complex caused an inhibition of ca. 48% and 38% for the normal and the tumoral osteoblasts, respectively (p < 0.001). This inhibition could be partially reversed to 35% and 28% by NAC (N-acetylcysteine) and a mixture of vitamins E and C. Changes in cell proliferation correlated with morphological alterations and the disruption of actin cytoskeleton fibers. The complex also enhanced the level of ROS (reactive oxygen species) up to ca. 100% over basal in both cell lines. Activation of ERK signalling cascade was also observed. These events led to apoptosis (up to 44% in MC3T3-E1 and 33% in UMR106 cells). Scavengers of ROS and inhibitors of ERK cascade allowed to elucidate the mechanisms involved in the cytotoxicity. In conclusion, V(V)-Salsem displayed cytotoxic effects on osteoblasts in culture through the production of free radicals and the activation of ERK cascade. Four new palladium(II) complexes with the formula Pd(L)2, where L are quinoxaline-2-carbonitrile N1,N4- dioxide derivatives, were synthesized as a contribution to the chemistry and pharmacology of metal compounds with this class of pharmacologically interesting bioreductive prodrugs. Compounds were characterized by elemental, conductometric and thermogravimetric analyses, fast atom bombardment mass spectrometry (FAB-MS) and electronic, Fourier transform infrared (FTIR) and 1H-nuclear magnetic resonance spectroscopies. The complexes were subjected to cytotoxic evaluation on V79 cells in hypoxic and aerobic conditions. In addition, a preliminary study on interaction with plasmid DNA in normoxia was performed. Complexes showed different in vitro biological behavior depending on the nature of the substituent on the quinoxaline ring. Pd(L1)2 and Pd(L2)2, where L1 is 3-aminoquinoxaline-2-carbonitrile N1,N4-dioxide and L2 is 3-amino-6(7)-methylquinoxaline-2-carbonitrile N1,N4-dioxide, showed non selective cytotoxicity, being cytotoxic either in hypoxic or in aerobic conditions. On the other hand, Pd(L3)2, where L3 is 3-amino-6(7)-chloroquinoxaline-2-carbonitrile N1,N4-dioxide, resulted in vitro more potent cytotoxin in hypoxia (P = 5.0 lM) than the corresponding free ligand (P = 9.0 lM) and tirapazamine (P = 30.0 lM), the first bioreductive cytotoxic drug introduced into clinical trials. In addition, it showed a very good selective cytotoxicity in hypoxic conditions, being non-cytotoxic in normoxia. Its hypoxic cytotoxicity relationship value, HCR, was of the same order than those of other hypoxia selective cytotoxins (i.e., Mitomycine C, Misonidazole and the N-oxide RB90740). Interaction of the complexes with plasmid DNA in normoxia showed dose dependent ability to relax the negative supercoiled forms via different mechanisms. Pd(L2)2 introduced a scission event in supercoiled DNA yielding the circular relaxed form. Meanwhile, both Pd(L1)2 and Pd(L3)2 produced the loss of negative supercoils rendering a family of topoisomers with reduced electrophoretic mobility. Pd(L3)2 showed a more marked effect than Pd(L1)2. Indeed, for the highest doses assayed, Pd(L3)2 was even able to introduce positive supercoils on the plasmid DNA. Eight new platinum(II) complexes with 3-(5-nitrofuryl)acroleine thiosemicarbazones showing antitrypanosomal activity were synthesized, characterized and in vitro evaluated. Most of the complexes showed IC50 values in the micromolar range against two different strains of Trypanosoma cruzi, causative agent of Chagas disease (American Trypanosomiasis). In addition, most of the newly developed complexes, together with the analogous platinum 5-nitrofuraldehyde containing thiosemicarbazones previously reported, resulted more active than the reference trypanocidal drug nifurtimox on the infective trypomastigote form of the parasite. Their capacity to produce free radicals that could lead to parasite death was evaluated by ESR experiments in the parasite and by respiration measurements. Compounds were tested for their DNA interaction ability. Results showed that some of the compounds could act as dual inhibitors in the parasite, through production of toxic free radicals and interaction with DNA. Several strains of endophytic bacteria and yeast were isolated from Daucus carota, Erythrina crista-galli, Curcubita maxima and Fortunella margarita, and tested for the reduction of two model �-alkyl-�- ketoesters. The yeast strains resulted better biocatalysts for the reduction of the tested substrates with the exception of Enterobacter agglomerans C8 strain, isolated from Erythrina crista-galli, that provided the best results for the preparation of syn (2R,3S) �-alkyl �-hydroxyesters. Among the isolated yeast strains, the Pichia sp. strain isolated from D. carota, was the best biocatalyst for production of anti (2S,3S) �-alkyl �-hydroxyesters. Our results show that the microbial endophytic community provides an interesting niche on the search for novel biocatalysts. Aim We sought to determine variations in fiber organization at the molecular level using x-ray diffraction analyses on human blood vessel specimens after cryopreservation processes. Materials and Methods Diffractometric profiles were performed on aortic and carotid cryopreserved-thawed vessel samples (CVS) versus the same fresh vessel samples (FVS). X-ray diffraction was performed on vascular tissues from 17 cadaveric donors after informed consent. Measurements utilized a Seifert Scintag PAD-II powder diffractometer with CuKa radiation; λ = 1.5418 Å. Scans were evaluated in the 5° to 60° range in theta −2theta mode, in the 5° to 60° range in 2-theta, with steps 0.1° and 10 seconds per step. Ten aortic and 8 carotid diffractometric profiles were analyzed, using differential planimetric surfaces measured under x-ray diffraction curve. Diffractographic profiles were analyzed according to intervals based upon the ages of the donors. An ordering profile coefficient (OPC) was obtained as the quotient between the differential planimetric surface (DPS) of FVS versus CVS vessel ordering diffraction. Results There was a decreased ordering profile according to age: older donors showed less ordering than younger ones. Clear peaks at d-spacing of 2.86 Å and 2.15 Å (2-theta = 31.3° and 42.0°, respectively) were always confirmed despite the different profiles of samples. OPC showed a higher ordering profile among the CVS than FVS: 70% aortas and 62.5% carotids. Conclusion The cryopreserved-thawed procedure does not damage the fibrillar organization of vessels.
