Leaf removal is beginning to receive consideration for many growers as an important cultural practice for bunch rots control. It is normally applied after veraison and almost only on red varieties because of the high sunburn susceptibility of white grapes varieties under Uruguayan climatic conditions. The aim of this study was to determine the time and intensity of leaves removal impact on bunch rots and grapes quality on Sauvignon blanc. In parallel, the effects of kaolin (Surround® WP) applications in reducing the negative impact of excessive exposure to high temperatures were evaluated. Different intensities and times of defoliation, in combination with kaolin applications, were evaluated over 2006/07, 2007/08 and 2008/09 seasons. All defoliation treatments and mainly those made after fruit set, significantly reduced the occurrence of bunch rots, having a critical impact on those metabolites responsible for the final taste of wines. Early defoliation was significantly more effective controlling Botrytis incidence and improving berry quality than later in the season. Leaf removal had a determining impact over fruit quality and primary and secondary metabolites contents. An increase in soluble solids and glycosides concentrations was observed, while IBMP grape content was reduced, thus affecting the flavor of Sauvignon blanc wines. Kaolin applications significantly reduced berries temperature, sunburn and other berry damages associated with clusters solar exposure. In sum, leaves removal and exogenous kaolin application in Sauvignon blanc showed great potential as Botrytis mitigation strategy because it positively impacts berry quality as a result of many molecular and biochemical changes in key secondary metabolic pathways. Composition and antioxidant properties of sixteen pecan [Carya illinoinensis (Wangenh) K. Koch] cultivars grown simultaneously in a single experimental orchard in Uruguay were evaluated to characterize their nutritional properties and nutraceutical potential. The percentage of oil, moisture, ash, minerals, and proteins were determined and also the fatty acid profile. Total phenolic compounds [18–41 mg gallic acid equivalents (GAEs)/g], condensed tannin [2–12 mg catechin equivalents (CEs)/g], and tocopherols (110–163 µg/g) contents were estimated in nut kernels. Total phenolic compounds (32–117 mg GAE/g), condensed tannins (130–357 mg CE/g), and total anthocyanins (1–3 mg 3-glucoside cyanidin/g) were also determined for pecan shells. The antioxidant activity in shells [57.15–578.88 µmol Trolox equivalents (TEs)/g] was 5 times higher compared with the kernels (23.15–156.60 µmol TEs/g) measured with hydrophilic ORAC. Bioactive compounds concentrations present statistically significant genetic variability between cultivars studied (p < 0.05). The presence of phenolic compounds was related with high-antioxidant capacity in kernels and shells, and a strong correlation between content of total phenolic compounds and condensed tannins in pecan shells was found. Principal component analysis (PCA) and hierarchical cluster analysis (HCA) show association between cultivars and the observed variables. The nutritional profile in the different cultivars showed the trends described in other countries, but this work shows some significant differences that could be attributed to the specific edaphoclimatic conditions of cultivation in Uruguay. Actualmente existen a nivel mundial un gran número de infecciones parasitarias producidas por helmintos, con gran incidencia tanto en la producción ganadera como en la salud humana. La fascioliasis, cuyo agente etiológico es el trematodo Fasciola hepatica, es una parasitosis que afecta a bovinos y ovinos ocasionando pérdidas económicas mundiales estimadas en 3.2 billones de dólares anuales. El único fármaco activo tanto contra el estadio adulto como juvenil del parásito, es el triclabendazol, con la problemática de la aparición de aislamientos resistentes del parásito en varios países. Fasciola hepatica se caracteriza por tener un ciclo de vida complejo que involucra varios estadios y cuyo hospedero definitivo es el ganado y eventualmente el hombre. Dada la necesidad de desarrollar nuevos tratamientos para esta parasitosis, en este trabajo se avanzó en la búsqueda de inhibidores de dos enzimas de la familia de las catepsinas, FhCL1 y FhCL3, que cumplen roles esenciales para la supervivencia del parásito, Se realizó un screening con dos series de compuestos, 1,4-di-N-óxido de quinoxalina y moléculas híbridas flavonoide-quinoxalina, abordando el estudio de la inhibición de los blancos moleculares desde una óptica multidisciplinaria que combinó estudios in vitro e in silico. Se identificaron once derivados de 1,4-di-N-óxido de quinoxalina y catorce compuestos híbridos con un porcentaje de inhibición igual o mayor a 50% para al menos una de las enzimas a 10 μM. De ellos, cinco presentaron una inhibición entre 75%-100% para ambos blancos moleculares evaluados (C17, C23, C24, H4 y H14). Describimos comportamientos diferenciales en cuanto a reversibilidad y cinética de unión, y caracterizamos mediante modelado computacional la interacción entre las catepsinas y los compuestos inhibidores, estudiando la contribución de diferentes sustituyentes del core 1,4-di-N-óxido de quinoxalina a la estructura farmacofórica. Se evaluó además la selectividad de la inhibición utilizando la catepsina L humana, la actividad fasciolicida sobre los parásitos juveniles cultivados in vitro y la citotoxicidad sobre células HepG2 y espermatozoides bovinos. Los compuestos evaluados que presentaron mejor perfil como potenciales fármacos fueron los derivados C17 y H4, los cuales resultaron excelentes inhibidores de ambas catepsinas, mostrando selectividad frente a las enzimas parasitarias. Asimismo, presentaron actividad fasciolicida sobre el estadio juvenil del parásito y citotoxicidad comparable al triclabendazol, sugiriendo que son buenos candidatos para el desarrollo de fármacos para el control de esta parasitosis. The aim of this work was to evaluate the potential of lufenuron, a benzylphenylurea with ability to interfere with the formation of insect exoskeleton, as a therapeutic drug for larval echinococcosis (hydatid disease). For this purpose lufenuron, alone or in combination with albendazole, was administered to CD1 mice bearing Echinococcus granulosus hydatid cysts in the peritoneal cavity. Neither of the drugs alone was able to exert parasiticidal effects. However, in combination with albendazole, lufenuron reduced the growth of cysts (30–40% in cyst diameter respect to control, p < 0.05). This effect was associated with ultrastructural alterations of the hydatid cyst wall and a reduction of the content of myo-inositol-hexakisphosphate, the major component of the electron dense granules of the laminated layer. Overall, this work provides evidence that lufenuron could represent a useful compound for the use in chemotherapy against larval echinococcosis, by enhancing albendazole parasiticidal activity. Chronic stress deteriorates the immune function of fish, thereby increasing their vulnerability to infections. However, the molecular and cellular mechanisms underlying stress-mediated immunosuppression and infection susceptibility in fish remain largely unknown. Understanding these mechanisms will contribute to improving fish welfare and their farm production. Herein, we review the challenges of sturgeon aquaculture in subtropical countries, where current climate change has giving rise to significant temperature increments during summer. This leads to the exposure of fish to stressful conditions during these months. Chronic heat stress deserves attention considering the rapid warming rate of the planet. It is already affecting wild fish populations, with disastrous consequences for sturgeons, which are one of the most endangered fish species in the world. In this context, we discuss the most recent advances through the studies on the effects of chronic heat stress on the innate immune components of sturgeons. To this end, we summarise the findings of studies focusing on the aquaculture of Russian sturgeons and observations made on other Acipenser species. Special attention is given to acute-phase proteins, as they might be valuable biomarkers of heat stress and infection, with applicability in monitoring the fish health status in farms. Thus, we compared the systemic and local in¯ammatory responses induced by the parasite, and the outcome of infection, between congenic C5-suf®cient (B10.D2 n/SnJ) and C5-de®cient (B10.D2 o/SnJ) mice challenged with protoscoleces. Indirect evidence of in-vivo complement activation during the establishment phase was obtained; infection induced serum amyloid P and eosinophil responses which were dependent on C5. Early recruitment of polymorphonuclear cells was not dependent on the presence of C5. The higher capacity of C5-suf®cient mice to recruit eosinophils was also observed during the cystic phase of infection, and mice recruiting more eosinophils developed lower parasite masses. Analysis of the outcome of infection after 8 months showed that C5-suf®cient mice were more resistant to infection than C5-de®cient mice in terms of individuals with no cysts; this trend was not statistically signi®cant. In addition, C5-de®cient mice developed higher numbers of large (> 5mm in diameter) cysts and higher cyst weights than C5-suf®cient mice indicating that C5-mediated mechanisms are detrimental for parasite growth. Taken together, our results suggest that complement, through C5-mediated effectors, contributes to host defences by both restricting the establishment of infection and controlling the growth of established cysts. This contribution may, at least partially, be associated with the ability of C5a to promote eosinophil in®ltration. The hydatid cyst is potentially capable of activating host complement, since it is a large, persistent, carbohydrate-rich structure, coated with host immunoglobulins, and localized in the host's internal organs. Nonetheless, in vitro studies have suggested that the cyst surface, the hydatid cyst wall (HCW), is a poor complement activator. In this study, we assessed the occurrence of in vivo complement activation on the hydatid cyst by measuring the levels of two complement activation products, C3d and complexes bearing a C9 activation neoepitope (TCC/MAC), in extracts from HCW of human origin. Low amounts of C3d and TCC/MAC were found in HCW in comparison with their levels in normal human plasma and activated human sera, suggesting that in vivo complement activation on HCW is efficiently down-regulated. This regulation may contribute to limit host inflammation which has been observed to correlate with parasite degeneration and death. Nitroalkene derivatives of fatty acids act as adaptive, antiinflammatory signalling mediators, based on their high-affinity PPARγ (peroxisome-proliferator-activated receptor γ ) ligand activity and electrophilic reactivity with proteins, including transcription factors. Although free or esterified lipid nitroalkene derivatives have been detected in human plasma and urine, their generation by inflammatory stimuli has not been reported. In the present study, we show increased nitration of cholesteryllinoleate by activated murine J774.1 macrophages, yielding the mononitrated nitroalkene CLNO2 (cholesteryl-nitrolinoleate). CLNO2 levels were found to increase &sim;20-fold 24 h after macrophage activation with Escherichia coli lipopolysaccharide plus interferon-γ ; this response was concurrent with an increase in the expression of NOS2 (inducible nitric oxide synthase) and was inhibited by the •NO (nitric oxide) inhibitor L-NAME (NG-nitro- L-arginine methyl ester). Macrophage (J774.1 and bone-marrowderived cells) inflammatory responses were suppressed when activated in the presence of CLNO2 or LNO2 (nitrolinoleate). This included: (i) inhibition of NOS2 expression and cytokine secretion through PPARγ and •NO-independent mechanisms; (ii) induction of haem oxygenase-1 expression; and (iii) inhibition of NF-κB (nuclear factor κB) activation. Overall, these results suggest that lipid nitration occurs as part of the response of macrophages to inflammatory stimuli involving NOS2 induction and that these by-products of nitro-oxidative reactions may act as novel adaptive down-regulators of inflammatory responses. In this work we studied the evolution of early inflammation, complement activation and parasite survival/death along the establishment phase of Echinococcus granulosus metacestode. Using a chamber model of infection in mice, we examined cell infiltration and C3 deposition on individual parasites during their development from protoscoleces to cystic forms. We found that the intensity of the initial inflammation decreased around undamaged but not around damaged parasites: at 43 dpi undamaged parasites were mostly associated with poor/mild inflammation while damaged parasites with a strong inflammation. In addition, whereas complement activation participated in the induction of early inflammation, the deposition of C3 on undamaged parasites progressively diminished. Interestingly, we observed some parasites in pre-cystic stage with no/poor C3 deposition at 3 dpi. Overall, these results indicated that the establishment and survival of the hydatid cyst is associated with the control of complement and, consequently, of local inflammation at the initial phases of infection. Antigen B (EgAgB) is the most abundant and immunogenic antigen produced by the larval stage (metacestode) of Echinococcus granulosus. It is a lipoprotein, the structure and function of which have not been completely elucidated. EgAgB apolipoprotein components have been well characterised; they share homology with a group of hydrophobic ligand binding proteins (HLBPs) present exclusively in cestode organisms, and consist of different isoforms of 8-kDa proteins encoded by a polymorphic multigene family comprising five subfamilies (EgAgB1 to EgAgB5). In vitro studies have shown that EgAgB apolipoproteins are capable of binding fatty acids. However, the identity of the native lipid components of EgAgB remains unknown. The present work was aimed at characterising the lipid ligands bound to EgAgB in vivo. EgAgB was purified to homogeneity from hydatid cyst fluid and its lipid fraction was extracted using chloroform:methanol mixtures. This fraction constituted approximately 40–50% of EgAgB total mass. High-performance thin layer chromatography revealed that the native lipid moiety of EgAgB consists of a variety of neutral (mainly triacylglycerides, sterols and sterol esters) and polar (mainly phosphatidylcholine) lipids. Gas-liquid chromatography analysis showed that 16:0, 18:0 and 18:1(n-9) are the most abundant fatty acids in EgAgB. Furthermore, size exclusion chromatography coupled to light scattering demonstrated that EgAgB comprises a population of particles heterogeneous in size, with an average molecular mass of 229 kDa. Our results provide the first direct evidence of the nature of the hydrophobic ligands bound to EgAgB in vivo and indicate that the structure and composition of EgAgB lipoprotein particles are more complex than previously thought, resembling high density plasma lipoproteins. Results are discussed considering what is known on lipid metabolism in cestodes, and taken into account the Echinococcus spp. genomic information regarding both lipid metabolism and the EgAgB gene family. As tyrosine phosphatases are capable of using multiple substrates, we developed an improved methodology to pull down novel PtpA substrates from an enriched P-Y macrophage extract using the mutant PtpA D126A. This methodology reduced non-specific protein interactions allowing the identification of four novel putative PtpA substrates by MALDI-TOF-MS and nano LC-MS: three mitochondrial proteins - the trifunctional enzyme (TFP), the ATP synthase, and the sulfide quinone oxidoreductase - and the cytosolic 6-phosphofructokinase. All these proteins play a relevant role in cell energy metabolism. Using surface plasmon resonance, PtpA was found to bind immunopurified human TFP through its catalytic site since TFP-PtpA association was inhibited by a specific phosphatase inhibitor. Moreover, PtpA wt was capable of dephosphorylating immunopurified human TFP in vitro supporting that TFP may be a bona fide PtpA susbtrate. Overall, these results suggest a novel scenario where PtpA-mediated dephosphorylation may affect pathways involved in cell energy metabolism, particularly the beta oxidation of fatty acids through modulation of TFP activity and/or cell distribution. Common Solanum glycosides were tested against a non-Solanum pest, the greenbug, Schizaphis graminum, fed by artificial diet supplemented with the tested compounds. In contrast with the potato aphid Macrosiphum euphorbiae, S. graminum is sensitive to both potato and tomato glycoalkaloids, as well as to all the different Solanum steroidal glycosides tested. S. graminum was less sensitive to glycoalkaloids than to the steroidal glycosides. Laxumin A (LC50 4.3 μM) and laxumin B (LC50 6.1 μM), isolated from Solanum laxum, were the most active of the tested compounds. The different sensitivity of both aphids against Solanum glycosides could be related to a possible adaptation of the insects to their host plants secondary metabolites. Background: The cestode Echinococcus granulosus - the agent of cystic echinococcosis, a zoonosis affecting humans and domestic animals worldwide - is an excellent model for the study of host-parasite cross-talk that interfaces with two mammalian hosts. To develop the molecular analysis of these interactions, we carried out an EST survey of E. granulosus larval stages. We report the salient features of this study with a focus on genes reflecting physiological adaptations of different parasite stages. Methodology/Principal Findings: We generated ,10,000 ESTs from two sets of full-length enriched libraries (derived from oligo-capped and trans-spliced cDNAs) prepared with three parasite materials: hydatid cyst wall, larval worms (protoscoleces), and pepsin/H+-activated protoscoleces. The ESTs were clustered into 2700 distinct gene products. In the context of the biology of E. granulosus, our analyses reveal: (i) a diverse group of abundant long non-protein coding transcripts showing homology to a middle repetitive element (EgBRep) that could either be active molecular species or represent precursors of small RNAs (like piRNAs); (ii) an up-regulation of fermentative pathways in the tissue of the cyst wall; (iii) highly expressed thiol- and selenol-dependent antioxidant enzyme targets of thioredoxin glutathione reductase, the functional hub of redox metabolism in parasitic flatworms; (iv) candidate apomucins for the external layer of the tissuedwelling hydatid cyst, a mucin-rich structure that is critical for survival in the intermediate host; (v) a set of tetraspanins, a protein family that appears to have expanded in the cestode lineage; and (vi) a set of platyhelminth-specific gene products that may offer targets for novel pan-platyhelminth drug development.
