La estrategia adoptada por el programa nacional de mejoramiento gen&eacute;tico de papa desarrollado en INIA, implica la introgresi&oacute;n de genes de resistencia a partir de la especie nativa silvestre Solanum commersonii, ampliamente distribuida y adaptada a nuestras condiciones ambientales. El objetivo general de esta tesis fue generar conocimiento sobre el patosistema R. solanacearum - S. commersonii para contribuir al desarrollo de cultivares con resistencia a esta bacteria fitopat&oacute;gena y al control integrado de la marchitez bacteriana. Se evalu&oacute; la resistencia en germoplasma derivado del programa de mejoramiento y se seleccionaron genotipos con resistencia parcial altamente tolerantes que fueron utilizados como progenitores en los sucesivos cruzamientos. Adem&aacute;s, se seleccionaron genotipos con diferentes niveles de resistencia para continuar con los estudios de las interacciones planta-pat&oacute;geno. Para realizar el seguimiento de la colonizaci&oacute;n de la bacteria in planta, se construyeron y validaron cepas reporteras de R. solanacearum con actividad fluorescente (prote&iacute;na verde fluorescente, GFP) y luminiscente (oper&oacute;n luxCDABE, LUX). Los resultados obtenidos sugieren que la resistencia a la marchitez bacteriana en papa est&aacute; relacionada con la capacidad de la planta de restringir la colonizaci&oacute;n y multiplicaci&oacute;n del pat&oacute;geno, particularmente limitando la diseminaci&oacute;n a lo largo del tallo. A su vez, se observaron varias respuestas de defensa inducidas luego de la infecci&oacute;n con la bacteria en los genotipos con resistencia parcial como deposici&oacute;n de lignina, producci&oacute;n de especies reactivas del ox&iacute;geno, t&iacute;lides, un aumento del intercambio de electrolitos a trav&eacute;s de la membrana y cambios a nivel de la expresi&oacute;n de genes de defensa. Tambi&eacute;n se analiz&oacute; la composici&oacute;n mineral de la savia xilem&aacute;tica, hoja, tallo y ra&iacute;z de los genotipos seleccionados encontrando mayores concentraciones de Ca a nivel de tallo y savia xilem&aacute;tica en el genotipo parcialmente resistente. Este mineral tiene un efecto inhibidor del crecimiento de R. solanacearum y sobre caracter&iacute;sticas asociadas a la virulencia como la formaci&oacute;n de biofilm, adhesi&oacute;n y la movilidad tipo twitching. El agregado ex&oacute;geno de sales de Ca retras&oacute; y disminuy&oacute; la severidad de s&iacute;ntomas de marchitamiento en plantas susceptibles y resistentes inoculadas con R. solanacearum. Potato (Solanum tuberosum L.) is one of the main hosts of Ralstonia solanacearum, the causative agent of bacterial wilt. This plant pathogen bacteria produce asymptomatic latent infections that promote its global spread, hindering disease control. A potato breeding program is conducted in Uruguay based on the introgression of resistance from the wild native species S. commersonii Dun. Currently, several backcrosses were generated exploiting the high genetic variability of this wild species resulting in advanced interspecific breeding lines with different levels of bacterial wilt resistance. The overall aim of this work was to characterize the interaction of the improved potato germplasm with R. solanacearum. Potato clones with different responses to R. solanacearum were selected, and colonization, dissemination and multiplication patterns after infection were evaluated. A R. solanacearum strain belonging to the phylotype IIB-sequevar 1, with high aggressiveness on potato was genetically modified to constitutively generate fluorescence and luminescence from either the green fluorescence protein gene or lux operon. These reporter strains were used to allow a direct and precise visualization of fluorescent and luminescent cells in plant tissues by confocal microscopy and luminometry. Based on wilting scoring and detection of latent infections, the selected clones were classified as susceptible or tolerant, while no immune-like resistance response was identified. Typical wilting symptoms in susceptible plants were correlated with high concentrations of bacteria in roots and along the stems. Tolerant clones showed a colonization pattern restricted to roots and a limited number of xylem vessels only in the stem base. Results indicate that resistance in potato is achieved through restriction of bacterial invasion and multiplication inside plant tissues, particularly in stems. Tolerant plants were also characterized by induction of anatomical and biochemical changes after R. solanacearum infection, including hyperplasic activity of conductor tissue, tylose production, callose and lignin deposition, and accumulation of reactive oxygen species. This study highlights the potential of the identified tolerant interspecific potato clones as valuable genetic resources for potato-breeding programs and leads to a better understanding of resistance against R. solanacearum in potato. Hemos caracterizado la cinética de reacción de un conjunto de porfirinas de manganeso (MnPorfirinas) solubles en agua frente a peroxinitrito (ONOO–) y radical carbonato (CO3·–). La reacción, entre ONOO– y el complejo de MnIII, tiene constantes de velocidad entre 105 y 107 M-1 s-1 a 37 ºC con un comportamiento sigmoide en función del pH, pKa aparentes cercanos al del par ONOOH/ONOO– y máximos a pH alcalino. La reacción produce el complejo O=MnIV y ·NO2. Los valores de estas constantes de velocidad tienen una relación lineal de energía libre con parámetros fisicoquímicos de la porfirina tales como el potencial redox del par MnIII/MnII o la acidez de Brønsted de los nitrógenos pirrólicos o de las moléculas de agua axiales. La reacción, entre ONOO– y el complejo de MnII, tiene constantes de velocidad <106 M-1 s-1 a pH 7.4 y 37 ºC y produce estequiométricamente O=MnIV y NO2–. La reacción, entre CO3·– y el complejo de MnIII tiene constantes de velocidad extrapoladas a pH neutro entre 2 y 12 x 108 M-1 s-1 con un comportamiento ácido base congruente con la ionización de las moléculas axiales de agua presentes en el complejo. La reacción entre CO3·– y el complejo de MnII tiene constantes de velocidad a pH = 10.4 entre 1 y 5 x 109 M-1 s-1. En ambos casos el CO3·– produce oxidaciones por un electrón. Los complejos de O=MnIV se reducen en forma rápida con ascorbato o urato hasta MnIII en presencia de oxígeno y, con bajas concentraciones de O2, la reducción puede llegar hasta MnII si los reductores son flavoenzimas reducidas, entre ellas succinato deshidrogenasa y NADH deshidrogenasa de la cadena mitocondrial de transporte de electrones. Estas reacciones de reducción completan dos posibles ciclos para catalizar la reducción de ONOO– o CO3·– con una eficiencia suficiente para proteger blancos biológicos en concentraciones micromolares de MnPorfirinas. Se presentan resultados de uso del ciclo catalítico MnIII/MnIV con urato como reductor en la protección de LDL in vitro y del ciclo catalítico MnII/MnIV con succinato/succinato deshidrogenasa como reductor para proteger actividades enzimáticas de partículas submitocondriales in vitro. In this paper, the influence of several operational parameters on a well established multiresidue LC&ndash;MS/MS method has been studied in relation to the analysis of 150 pesticides commonly present in vegetable samples. The operational parameters investigated are: (i) the influence of different modifiers (0.1% formic acid; 5mMammonium formiate; 5mMammonium acetate in aqueous phase) &ndash; both on the retention time and on the analytical response of the studied compounds; (ii) the effect of the analytical column&rsquo;s temperature on the retention time and on the analytical response of the pesticides investigated; (iii) the effects of co-elution in mixture containing 150 pesticides and, additionally, (iv) the carrying out of a study about the common transitions obtained by LC&ndash;MS/MS. Various common transitions were found among the 150 pesticides, but there were only two problematic cases, the pairs diuron&ndash;fluometuron and prometryn&ndash;terbutryn, which have common scanned transitions and have very close retention times. The use of ammonium salts as modifier instead of formic acid reports enhancement or suppression of the response depending on the pesticides. No great influence on the retention time or on the response of the pesticides and commodities studied was observed with relation to the column temperature. Two different columns: an HPLC (5�m particle size) and an UHPLC analytical column (1.8�m particle size) have been used. As was expected, shorter run times and lower peak width was achieved with the UHPLC column. In this paper, the effect of the compounds on each other in the MS analysis when the number of coeluting compounds is quite high is also described. Mainly small suppression or enhancement co-elution effect was observed, but some particular pesticides presented high sensitivity (&gt;&plusmn;60% effect) when they elute together with others. This is an important factor and it has to be taken into account when performing multiresidue pesticide analysis. Inositol poly- and pyrophosphates (InsPs and PP-InsPs) are central eukaryotic messengers. These very highly phosphorylated molecules can exist in two distinct conformations, a canonical one with five phosphoryl groups in equatorial positions, and a &ldquo;flipped&rdquo; conformation with five axial substituents. Using 13C-labeled InsPs/PP-InsPs, the behavior of these molecules was investigated by 2D-NMR under solution conditions reminiscent of a cytosolic environment. Remarkably, the most highly phosphorylated messenger 1,5(PP)2 -InsP4 (also termed InsP8 ) readily adopts both conforma tions at physiological conditions. Environmental factors&mdash;such as pH, metal cation composition, and temperature&mdash;strongly influence the conformational equilibrium. Thermodynamic data revealed that the transition of InsP8 from the equatorial to the axial conformation is, in fact, an exothermic process. The speciation of InsPs and PP-InsPs also affects their interaction with protein binding partners; addition of Mg2+ decreased the binding constant Kd of InsP8 to an SPX protein domain. The results illustrate that PP-InsP speciation reacts very sensitively to solution conditions, suggesting it might act as an environment-responsive molecular switch. The present work deals with a new methodology to produce a sustainable carbon material through a simple synthetic route, and its performance as Ni-based electrocatalyst support. Such porous carbon was easily synthesized from the raw Kraft black liquor, a by-product rich in lignin, by chemical polymerization in alkaline medium with a nickel source and subsequent activation by CO2. Nickel nanoparticles were homogeneously distributed in the samples, as revealed by STEM images. The antioxidant capacity of propolis from the southern region of Uruguay was evaluated using in vitro as well as cellular assays. Free radical scavenging capacity was assessed by ORAC, obtaining values significantly higher than those of other natural products (8000 &mu;mol Trolox equiv/g propolis). ORAC values correlated well with total polyphenol content (determined by Folin�Ciocalteu method) and UV absorption. Total polyphenol content (150 mg gallic acid equiv/g propolis) and flavonoids (45 mg quercetin equiv/g propolis) were similar to values reported for southern Brazilian (group 3) and Argentinean propolis. Flavonoid composition determined by RP-HPLC indicates a strong poplar-tree origin. Samples high in polyphenols efficiently inhibit low-density lipoprotein lipoperoxidation and tyrosine nitration. In addition, Uruguayan propolis was found to induce the expression of endothelial nitric oxide synthase and inhibit endothelial NADPHoxidase, suggesting a potential cardiovascular benefit by increasing nitric oxide bioavailability in the endothelium. [99mTc]Glucarate has also been postulated as an agent for non-invasive detection of tumors. The aim of our study was to develop a Glucarate kit and evaluate [99mTc]glucarate as a potential cancer imaging agent in female SCID mice bearing humanMDA-MB-435 breast tumors. Methods: Glucarate in a kit formulation was labeled with 99mTc and evaluated for radiolabelling efficiency and radiochemical purity. The Glucarate kit stability was assessed by monthly quality controls. The pharmacokinetics of [99mTc]glucarate were determined in female SCID mice bearing MDA-MB-435 human breast carcinoma tumors at 0.5, 1, 2, 4 and 24 h. Nuclear imaging studies were performed with a microsingle photon emission tomography (SPECT)/computed tomography (CT) system at 2 h post injection, while magnetic resonance imaging (MRI) was employed for tumor morphology analysis and metastatic deposit localization. Results: The Glucarate kits exhibited a stable shelf life of 6 months. [99mTc]Glucarate was obtained with radiochemical purity greater than 95%. Biodistribution studies demonstrated moderate tumor uptake coupled with high renal clearance. Tumor-to-muscle ratios were 4.85 and 5.14 at 1 and 4 h post injection. MRI analysis showed tumors with dense cellular growth and moderate central necrosis. [99mTc]Glucarate uptake in the primary MDA-MB-435 shoulder tumors and metastatic lesions were clearly visualized with micro-SPECT/CT imaging. Conclusions: Selective tumor uptake and rapid clearance from nontarget organs makes [99mTc]glucarate a potential agent for breast cancer imaging that awaits validation in a clinical trial. American trypanosomiasis or Chagas disease, caused by the protist parasite Trypanosoma cruzi (T. cruzi), is a major health concern in Latin America. In the search for new bioactive compounds, eight Pd(II) and Pt(II) complexes of thiosemicarbazones derived from 1-indanones (HL) were evaluated as potential anti-T. cruzi compounds. Their unspecific cytotoxicity was determined on human erythrocytes. Two physicochemical features, lipophilicity and redox behavior, that could be potentially relevant for the biological activity of these complexes, were determined. Crystal structure of [Pd(HL1)(L1)]Cl&middot;CH3OH, where HL1 = 1-indanone thiosemicarbazone, was solved by X-ray diffraction methods. Five of the eight metal complexes showed activity against T. cruzi with IC50 values in the low micromolar range and showed significantly higher activity than the corresponding free ligands. Four of them resulted more active against the parasite than the reference antitrypanosomal drug Nifurtimox. Anti-T. cruzi activity and selectivity towards the parasite were both higher for the Pd(II) compounds than for the Pt(II) analogues, showing the effect of the metal center selection on the biological behavior. Among both physicochemical features tested for this series of compounds, lipophilicity and redox behavior, only the former seemed to show correlation with the antiproliferative effects observed. Metal coordination improved bioactivity but lead to an increase of mammalian cytotoxicity. The present study describes the analgesic activity of extracts and some fractions obtained from Erythrina crista-galli leaves in different in vivo analgesic models, using mice as experimental animals. The results showed that extract E2 was the most active, inhibiting 48% of the abdominal constrictions when evaluated against the writhing test at 10 mg kg�1, intraperitoneal. It also caused dose-dependent inhibition in the same model, with a calculated ID50 value and respective confidence interval of 10 (9&ndash;14)mg kg�1, and was more potent than reference drugs. Administered orally, E2 caused potent antinociceptive action, with a calculated ID50 value of 35 (26&ndash;47) mgkg�1. The fractions F1 and F2 obtained from E2 were evaluated against the writhing test at 10 mg kg�1, causing inhibitions of 41 and 88%, respectively. The most active fraction, F2, presented ID50 calculated value of 3 (2&ndash;4) mg kg�1, being about 7-fold more active than the reference drugs (acetyl salicylic acid and acetaminophen). In the formalin test, F2 inhibited both phases of pain (44%, first phase; 58%, second phase). However, in contrast to the results observed for E2, it was not active against the hot-plate test. The phytochemical results showed that at least four main components are present in F2, which show a positive reaction of terpenes with TLC spray reagents. Considering the great impact of texture on consumers&rsquo; liking of several products, it is important for food companies to understand how consumers describe the texture of food products. The aim of the present study was to get an insight on consumers&rsquo; texture vocabulary in three different Spanish-speaking countries: Argentina, Spain and Uruguay. A free listing task was carried out in each country with 107&ndash;120 consumers. Participants were asked to list all the texture characteristics of food products they knew about. Between 80 and 112 terms were elicited by consumers, comprising mainly words related to texture characteristics of food products. By simultaneously considering frequency of mention and average order of elicited terms, the most familiar texture terms in each country were identified, being the most frequently used texture terms similar. Results from the present work would contribute to a greater knowledge of the vocabulary used by consumers to describe the texture of food products and show the existence of crosscultural differences in word usage within a same language. The present study compared three methods for generating sensory vocabulary with consumers: repertory grid (RG), comparison of the sample set (CW), and individual sample description (ID). A consumer study was carried out with two groups of 90 participants, randomly divided into 3 subgroups (n = 30) which evaluated one of the two types of satiating product (chocolate-flavoured shakes and biscuits) using one of the three methods to generate sensory vocabulary. The methods differed in their ability to encourage consumers to verbalize their sensory perception in detail, and also in the total number of terms generated and the frequency with which consumers generated some terms. The lowest number of terms, at both individual and aggregate level, was elicited when consumers described similarities and differences in the whole sample set. However, the most frequent terms were similar in the three methods, indicating their convergent validity. Results from the present work suggest that ID and RG are preferable to CW. Considering practical aspects related to their implementation, ID shows advantages over RG for identifying the consumers&rsquo; vocabulary without seriously compromising data quality. Texture attributes, which have been related to satiating capacity, were frequently mentioned to describe the evaluated products. Terms related to viscosity were the most frequently elicited, whereas varied textural features were mentioned for biscuits. We previously reported a multigene family of monodomain Kunitz proteins from Echinococcus granulosus (EgKU-1-EgKU-8), and provided evidence that some EgKUs are secreted by larval worms to the host interface. In addition, functional studies and homology modeling suggested that, similar to monodomain Kunitz families present in animal venoms, the E. granulosus family could include peptidase inhibitors as well as channel blockers. Using enzyme kinetics and whole-cell patch-clamp, we now demonstrate that the EgKUs are indeed functionally diverse. In fact, most of them behaved as high affinity inhibitors of either chymotrypsin (EgKU-2-EgKU-3) or trypsin (EgKU-5-EgKU-8). In contrast, the close paralogs EgKU-1 and EgKU-4 blocked voltage-dependent potassium channels (Kv); and also pH-dependent sodium channels (ASICs), while showing null (EgKU-1) or marginal (EgKU-4) peptidase inhibitory activity. We also confirmed the presence of EgKUs in secretions from other parasite stages, notably from adult worms and metacestodes. Interestingly, data from genome projects reveal that at least eight additional monodomain Kunitz proteins are encoded in the genome; that particular EgKUs are up-regulated in various stages; and that analogous Kunitz families exist in other medically important cestodes, but not in trematodes. Members of this expanded family of secreted cestode proteins thus have the potential to block, through high affinity interactions, the function of host counterparts (either peptidases or cation channels) and contribute to the establishment and persistence of infection. From a more general perspective, our results confirm that multigene families of Kunitz inhibitors from parasite secretions and animal venoms display a similar functional diversity and thus, that host-parasite co-evolution may also drive the emergence of a new function associated with the Kunitz scaffold. Thiosemicarbazones have been described as cruzipain inhibitors. While searching for new antichagasic drugs, we synthesized a series of selenosemicarbazone analogs and demonstrated that the isosteric replacement of the sulfur atom with selenium resulted in an enhancement of the cysteine protease inhibitory effect. Three selenosemicarbazones were characterized enzymatically and proved to be reversible, slow-binding inhibitors for the Z-Phe-Arg- AMC substrate. Their KI values were in the low nM range (3.7 to 29.7 nM), suggesting a strong interaction with the enzyme, approaching a tight binding definition. All selenosemicarbazones tested showed better activities against epimastigotes than Benznidazole, the currently used drug, with IC50 values ranging from 1.2 to 5.9 mM (Bnz IC50 &frac14; 12.5 mM). Three of these compounds showed a better Selectivity Index (SI) than Benznidazole. These compounds also displayed activity against the infective intracellular amastigote form at low micromolar range. Cyclic voltammetry and electron spin resonance techniques were used in the investigation of novel 3-alkoxy- and 3-hydroxy-1-[&omega;-(dialkylamino)alkyl]-5-nitroindazole derivatives. A self-protonation process involving the protonation of the nitro group was observed. The reactivity of the nitro-anion radical for these derivatives with glutathione, a biological relevant thiol, was also studied by cyclic voltammetry. These studies demonstrated that glutathione could react with radical species from 5-nitroindazole system.
