OBJECTIVES: Lead (Pb) and manganese (Mn) are confirmed neurotoxins but it is unclear to what extent low-level exposure produces a unique behavioral signature. The objective of this study was to investigate latent cognitive profiles among children (6-8 years) from Montevideo, Uruguay co-exposed to these metals. METHOD: Among 345 children, blood Pb and hair Mn were measured using atomic absorption spectroscopy and ICP-MS, respectively. Sixteen measures, reflecting multiple domains of cognitive functioning were gathered: (1) three tests from Cambridge Neuropsychological Test Automated Battery (CANTAB): Intra-Extra Dimensional Shift (IED), Spatial Span (SSP) and Stockings of Cambridge (SOC), (2) ten tasks from Woodcock-Muñoz Achievement Battery, Revised (WM): Visual-Motor Integration, Verbal Comprehension (Vocabulary, Synonyms, Antonyms, Analogies), Visual-Auditory Comprehension, Concept Formation, Visual Spatial Thinking, Number Inversion and Spatial Relations, (3) Bender Gestalt task, and (4) Weschler block design task. Scores were modeled using latent profile analysis (LPA). Association between blood Pb and hair Mn on performance profiles was assessed using ordinal regression, controlling for confounders. An interaction between Pb and Mn was tested. RESULTS: Mean ± SD of blood Pb was 4.1 ± 2.1 μg/dL and 35% of children had blood Pb ≥ 5 μg/dL. Median [5%, 95%] hair Mn level was 0.8 [0.3, 4.1] ppb. Three latent cognitive performance profiles were identified: high (n = 46, 13%), average (n = 209, 61%) and low (n = 90, 26%). Each one-unit increase in blood Pb was associated with a 28% greater likelihood of belonging to a poorer-performing profile. The association was non-linear, with the effect of Pb on profile membership strongest at lower levels of exposure. There was no meaningful interaction between Pb and Mn. CONCLUSIONS: A behavioral signature for low-level Pb & Mn exposure was not identified, but the likelihood of membership in low-performing profile was higher at lowest levels of blood Pb. There was no effect measure modification between Pb and Mn. Future research should address how complex environments created by chemical exposures and the social context relate to cognitive performance in young children

A summary of the resource.
Metarhizium rileyi is an entomopathogenic fungus of Lepidoptera and infects many noctuids, which are key pests of agricultural crops. In this literature review, we summarise the state of art and use of this potential biological pest control agent. Pathogen–host interactions include conidial adhesion, invasion process, and enzymatic activity. Environmental factors that impact the productivity associated with this fungus such as environment temperature, humidity, and availability of spores are discussed. Recent research has focused on enzymatic aspects related to pathogenicity, in which the cause is not fully elucidated yet. More than 60 known host species, mainly lepidopterans, are listed. Non-susceptible species, including predatory insects, are also presented. The difficulty of mass production and the formulation are the principal obstacles in use of M. rileyi. We summarise the challenges necessary to better utilise this agent in biocontrol and suggest novel research approaches, including the development of commercial and technically viable formulations.

Abstract
A summary of the resource.
Often referred to as the “king of cancers” pancreatic cancer is a serious threat to public health, as most cases are diagnosed at an advanced stage. Although tumor immunotherapy is a major advancement in cancer treatment, its effectiveness in pancreatic cancer remains limited, particularly owing to the unique characteristics of the tumor microenvironment. Nano-delivery systems offer a promising approach for overcoming the dense stromal barrier, which is crucial for enhancing the therapeutic efficacy. In this review, we systematically summarize key immunotherapeutic targets, including cancer-associated fibroblast (CAF), immune cells (tumor-associated macrophage (TAM), myeloid-derived suppressor cell (MDSC), regulatory T cells (Treg), tumor-associated neutrophil (TAN), CD4+T, CD8+T cells), extracellular matrix (ECM), and discussed regulatory strategies in pancreatic cancer based on its pathogenesis and recent clinical advances to guide target selection. We then detail the physicochemical properties of nano-delivery systems suitable for pancreatic cancer and outline how tumor microenvironment-responsive linkers can enable particle size or charge conversion. Furthermore, we review the latest research progress on various nano-delivery systems (Lipid-based nanoparticle, Hydrogel/microneedle, Inorganic nanoparticle, Cancer vaccine, Adoptive cell therapy, Exosome/vesicle, Bacteria/virus) to inform the design of pancreatic cancer nanocarriers. Finally, we discuss future directions and summarize the challenges in translating pancreatic cancer nano-delivery systems into clinical practice. By integrating target identification, nanocarrier design, and clinical applications, we aimed to provide a cohesive framework that offers insights to accelerate the development of effective pancreatic cancer therapies.

Cystic echinococcosis (CE) is a zoonotic disease worldwide distributed, caused by the cestode Echinococcus granulosus sensu lato (E. granulosus), with an incidence rate of 50/100,000 person/year and a high prevalence in humans of 5-10%. Serology has variable sensitivity and specificity and low predictive values. Antigens used are from the hydatid fluid and recombinant antigens have not demonstrated superiority over hydatid fluid. A cell line called EGPE was obtained from E. granulosus sensu lato G1 strain from bovine liver. Serum from CE patients recognizes protein extracts from EGPE cells with higher sensitivity than protein extracts from hydatid fluid. In the present study, EGPE cell protein extracts and supernatants from cell colonies were eluted from a protein G affinity column performed with sera from 11 CE patients. LC-MS/MS proteomic analysis of the eluted proteins identified four E. granulosus histones: one histone H4 in the cell extract and supernatant, one histone H2A only in the cell extract, and two histones H2A only in the supernatant. This differential distribution of histones could reflect different parasite viability stages regarding their role in gene transcription and silencing and could interact with host cells. Bioinformatics tools characterized the linear and conformational epitopes involved in antibody recognition. The three-dimensional structure of each histone was obtained by molecular modeling and validated by molecular dynamics simulation and PCR confirmed the presence of the epitopes in the parasite genome. The three histones H2A were very different and had a less conserved sequence than the histone H4. Comparison of the histones of E. granulosus with those of other organisms showed exclusive regions for E. granulosus. Since histones play a role in the host-parasite relationship they could be good candidates to improve the predictive value of serology in CE.

This paper describes the application of LC/Q-Orbitrap MS for the analysis of pesticide residues in fruit and vegetable commodities. LC/Q-Orbitrap MS working in full scan simultaneously with a single MS/MS scan was used to analyse 139 pesticide residues in QuEChERS extracts of tomato, pepper, orange and green tea. Full scan data were obtained at a resolution of 70,000 whereas MS/MS data were obtained at a resolution of 17,500. Quantitation and detection was carried out using full scan data while MS/MS data were used only for identification. MS/MS scans did not have a negative influence on quantitation under the applied conditions. Some peak area reproducibility problems were the consequence of the low sensitivity for some compounds (aldicarb, chlorpyriphos methyl, fenitrothion and fipronil) under the applied conditions. The relation between the operational parameters (viz. automatic gain control (AGC) target, maximum injection time (IT), underfill ratio, isolation window and apex trigger) and the number of automatically identified compounds was investigated. Mass error and minimal intensity of selected fragment ions were also studied. Various working modes were compared, such as full scan with single MS/MS scan and full scan with multiple MS/MS scans. In both cases, the number of automatically reported pesticides was the same. However full scan with single MS/MS scan ensured more points per peak in full scan mode and better peak area reproducibility. The evaluation of the identification and quantitation capabilities of the instrument was performed through the analysis of 100 real samples. The samples were also analysed by LC–QqQ MS/MS and the results of both analytical systems were compared. The comparison revealed that the two instruments were consistent with each other. They found the same pesticides and neither false positive nor false negatives were reported. Nevertheless the Q-Orbitrap MS allowed one to work in high resolution mass spectrometry, increasing the selectivity and, in full scan mode, permitting the retrospective analysis of the data feature that cannot be achieved with QqQ.

The myo-inositol phosphates (InsPs) are specific signalling metabolites ubiquitous in eukaryotic cells. Although Ins(1,3,4,5,6)P5 is the second most abundant member of the InsPs family, its certain biological roles are far from being elucidated, in part due to the large number of species formed by Ins(1,3,4,5,6)P5 in the presence of metal ions. In light of this, we have strived in the past to make a complete and at the same time “biological-user-friendly” description of the Ins(1,3,4,5,6)P5 chemistry with mono and multivalent cations. In this work we expand these studies focusing on the inframolecular aspects of its protonation equilibria and the microscopic details of its coordination behaviour towards biologically relevant metal ions. We present here a systematic study of the Ins(1,3,4,5,6)P5 intrinsic acid–base processes, in a non-interacting medium, and over a wide pH range, analyzing the 31P NMR curves by means of a model based on the Cluster Expansion Method. In addition, we have used a computational approach to analyse the energetic and structural features of the protonation and conformational changes of Ins(1,3,4,5,6)P5, and how they are influenced by the presence of two physiologically relevant cations, Na+ and Mg2+.

Baccharis trimera (Less.) DC is a South American plant that in folk medicine is considered to produce reduction in blood pressure. One aspect of this putative effect is the vasorelaxation. The aim of this work was to evaluate the ability of a B. trimera extract to relax rat aortic rings precontracted with noradrenaline. As the infusion is the usual way of intake of this plant, an infusion of B. trimera was prepared using 100g of the plant (leaves) boiled in water, frozen and lyophilized. Working solutions were prepared using different concentrations of the dried extract diluted in Krebs Henseleit solution. It was proved that the infusion relaxed the aortic rings in a dose dependent manner 100 minutes after adding the extract to the bath. Considering as 100% the maximum contraction achieved with noradrenaline, a relaxation of 101.1±2.3% was observed with the highest dose of the infusion used in these experiments (0.32mg/mL). While in control rings relaxation was 12.9±2.4%. In aortic rings denuded from endothelium the percentage of vasoralaxation did not show statistically significant differences when compared to intact rings. These data support the hypothesis of a vasorelaxant effect of this plant and constitutes the first approach to the scientific basis of a potential antihypertensive effect. The profiles of tocopherols and tocotrienols (vitamin E) in the seed oils recovered from apple (Malus domestica Borkh.), canary melon (Cucumis melo L.), gooseberry (Ribes uva-crispa L.), grape (Vitis vinifera L.), Japanese quince (Chaenomeles japonica), pomegranate (Punica granatum L.), red currant (Ribes rubrum L.), sea buckthorn (Hippophae rhamnoides L.), and watermelon (Citrullus lanatus) by-products of the fruit industry were studied. The highest concentration of tocochromanols was found in pomegranate seed oil (398.73 mg/100 g) and the lowest in gooseberry seed oil (69.13 mg/100 g). The γ and α homologues were the predominant forms of tocopherol and tocotrienol in all fruit seed oils studied. The pomegranate, canary melon, watermelon, gooseberry and red currants seed oils contained mainly γ-tocopherol (382.69, 63.08, 111.01, 60.35, and 156.39 mg/100 g, respectively). In the apple, sea buckthorn and Japanese quince seed oils, α-tocopherol was the predominant tocochromanol (58.77, 121.79, and 198.94 mg/100 g, respectively). High concentrations of tocotrienol homologes (γ and α) (49.49 and 34.53 mg/100 g, respectively) were found only in grape seed oil. A significant correlation (r = 0.994, p < 0.00001) between the total content of tocochromanols in the seed oils and scavenging of the free radical DPPH was found

Composition of tocochromanols in kernels recovered from 16 different apricot varieties (Prunus armeniaca L.) was studied. Three tocopherol (T) homologues, namely α, γ and δ, were quantified in all tested samples by an RP-HPLC/FLD method. The γ-T was the main tocopherol homologue identified in apricot kernels and constituted approximately 93% of total detected tocopherols. The RP-UPLC-ESI/MSn method detected trace amounts of two tocotrienol homologues α and γ in the apricot kernels. The concentration of individual tocopherol homologues in kernels of different apricots varieties, expressed in mg/100 g dwb, was in the following range: 1.38–4.41 (α-T), 42.48–73.27 (γ-T) and 0.77–2.09 (δ-T). Moreover, the ratio between individual tocopherol homologues α:γ:δ was nearly constant in all varieties and amounted to approximately 2:39:1.

Tocopherol separations employing the same Kinetex™ C18 column via supercritical fluid chromatography (SFC) and reversed-phase liquid chromatography (RP-LC) were compared. The application of the SFC system with UV diode array detection (DAD) resulted in rapid separation of all four tocopherol homologues with a total analysis time below 2 min. The RP-LC approach could not separate the isomers β and γ. The developed SFC-DAD method was precise, accurate, and most importantly more environmentally friendlier compared to the RP-LC method due to the 125-fold decrease in methanol consumption. The present study illustrated the selectivity differences between LC and SFC and how the C18 column can be used for tocopherol characterization. The optimized SFC method was successfully applied for the tocopherol determination in the seeds of nine different fruit species.

Abstract
A summary of the resource.
Macrosiphum euphorbiae, also known as the potato aphid, is a potato virus vector, associated with decreased tuber production. Feeding, toxicity, and reproductive effects of the major potato glycoalkaloids (α-chaconine and α-solanine), their common aglycone (solanidine), and a nonpotato glycoalkaloid (α-tomatine) and its aglycone (tomatidine) onM. euphorbiae were studied. As expected, neither α-chaconine nor α-solanine had a strong lethal effect on the aphid, but α-chaconine stimulated feeding at low concentrations. Both aglycones were deterrent and lethal at high concentrations and, like α-chaconine and α-tomatine, they delayed the appearance and decreased the number of nymphs.

Abstract
A summary of the resource.
We report the deterrent, toxic, and antireproductive effect of several Solanum glycoalkaloids on the potato aphid, Macrosiphum euphorbiae, and discuss the structure–activity relationship of the tested compounds. The results indicate a structure-dependent biological effect of the glycoalkaloids, suggesting that, while the structure of the aglycone defines a basal biological effect, the carbohydrate moiety is crucial for the overall biological effect.

Abstract
A summary of the resource.
The design and the execution of a multi-generation approach to pyrrolizidine alkaloids is summarized. A historical perspective and the various solutions to the problems of regio-, stereo-, and enantiocontrol in the preparation of these ubiquitous natural products are provided. The details of the fourth generation approach based on the resolution of ß-oxo esters of prochiral alcohols by yeast are disclosed. OVULACION
Abstract
A summary of the resource.
Ovulation was induced in rabbits by the administration of human chorionic gonadotropin (hCG) [9002-61-3]. A group of rabbits was treated with 2 different concns. (1/5 diln. and undild.) of a specific antiserum to oxytocin [50-56-6] injected immediately before the administration of hCG. The no. of ruptured follicles was detd. 12 h after administration of hCG. The no. of ovulation points in control rabbits was 7.7, whereas the group treated with the highest concn. of antiserum showed a mean of 3.8. Other groups of animals were treated with the prostaglandin synthesis inhibitor indomethacin or with the combined administration of indomethacin and oxytocin antiserum. The no. of ovulation points obsd. in the animals treated with indomethacin and antiserum was not significantly different from that of the group of animals in which those agents were administered sep.
