Abstract
A summary of the resource.
Gas phase conversion of ethanol–water mixtures on copper-based catalysts leads to a significant yield of acetone. Copper catalysts containing lanthanum oxide stabilized with zirconia, in the form of the so-called La2Zr2O7 pyrochlore compound, exhibit the best performance for the selective conversion of ethanol into acetone with yields of 96% of the stoichiometric amount at 400°C. Acid–base properties of the pyrochlore in addition to its higher specific surface area and copper dispersion can explain the better catalytic behavior in comparison to the other tested copper catalysts. The most plausible reaction mechanism involves ethanol dehydrogenation to acetaldehyde followed by oxidation to acetic acid and further ketonisation to yield acetone.

A summary of the resource.
A radical cyclization approach to the complete skeleton of morphine was investigated in several iterations. The first attempt at a radical cascade via a Bergman-type intermediate derived from ene-diyne 10 failed during a model study in which 10-membered silicon-tethered ene-diyne 17 proved inert to Bergman cyclization conditions. A second model study involving ene-diyne 27 , functionalized with an allyl group, underwent Claisen rearrangement to 32 in preference to a Bergman-type cyclization. Several simple model studies were performed with bromophenols appended to protected diols 40 and 50 , respectively, to determine the feasibility of C12–C13 bond formation in the former case and the cascade closure C12–C13/C14–C9 in the latter via radical species generated from the aryl halides. The second-generation approach employed the diene diol 7a derived biocatalytically from b -bromoethylbenzene via oxidation with E. coli JM109(pDTG601), its conversion to cyclization precursor 55 , and the radical cyclization to 56a , b . The conditions and the outcome of this process are discussed in detail along with the rationalization of stereochemistry of the cyclization, which furnished C14- epi configuration in 56a in low yield. The third-generation synthesis relied on stepwise radical cyclization of vinyl bromide 67 derived from obromo- b -ethylbenzene (also by biocatalytic means) and equipped with an oxazolidone as the radical acceptor group. Isoquinoline derivatives 68a and 68b were obtained as a mixture of isomers, the major of which, 68a , was converted via a second tin-mediated cyclization to the pentacyclic compound 78 , also possessing C14- epi configuration. The stepwise radical cyclization proceeded in higher yields, produced cleaner reaction mixtures, and was also performed with the more flexible alcohol 87 , whose tin-mediated closure produced a 1:1 mixture of C14 epimers, tetracyclic compounds 81 and 89 . Finally, tetracycle 80 or pentacycle 79 was converted to oxo aldehyde 83 and cyclized to the complete morphinan skeleton, 84 , in the ent -C14- epi series. Additionally, preliminary studies were performed on direct closures of chloride 82 to 85 , via a C10/C11 alkylation of a sp 3 -hybridized center. The three generations of synthetic effort are discussed in detail and physical and spectral data are provided for all new compounds. The relative merits of tandem vs . stepwise radical cyclization are evaluated and projections for future work are indicated. Abstract
A summary of the resource.
The search for protective antigens of intestinal parasites is conditioned by the methodology used to induce a relevant local immune response against them. The present work describes the use of immuno stimulating complexes (iscoms) from tegumental antigens from protoscoleces (PSC) of the cestode Echinococcus granulosus as immunogens in dogs by the intranasal route. It also describes the evaluation of the immune response evoked at the antibody level (systemically and at a distant mucosal location) as well as at the level of antibody secreting cells in peripheral blood. Iscoms from both E. granulosus tegumental antigens and hen ovalbumin (OVA), given at 50 μg doses by intranasal route, evoked significant secretory IgA antibody responses detected in saliva. Specific IgA secreting cells in peripheral blood also increased 10–20-fold, although transiently, after primary and secondary stimulation, whereas specific IgG secreting cells in peripheral blood were only detected in some individuals after the second antigenic exposure. Generation of immune responses at a related mucosal site provides evidence of localised immunity. No significant increase in systemic antibody titers of either IgM, IgG or IgA isotype was detected in plasma as a result of the immunisation. This fact could reflect that the nasopharyngeal mucosal associated lymphoid tissue of dogs is more strictly compartmentalised than that of other mammals.

A summary of the resource.
The reaction of reactive oxygen and nitrogen species with the [4Fe–4S]2+cluster of mitochondrial (m-) and cytosolic (c-) aconitases leads to loss of catalytic activity and, in the case of the c-aconitase, triggers total cluster disruption to yield the iron-regulatory protein-1 (IRP-1). Herein we have studied the relative contribution and interplay of reactive oxygen species (O [equation] and H2O2), nitric oxide (•NO), and peroxynitrite in the modulation of m- and c-aconitase and IRP-1 activities in V79-M8 mammalian fibroblasts, identifying key variables that control the various reactivities at the cellular level. Extracellular production of H2O2led to inactivation of both m- and c-aconitase and IRP-1 activation, while extracellular [equation] had no effect. However, increased intracellular production of [equation] caused a loss in m- and c-aconitase activity and IRP-1 activation. Nitric oxide released from NOC-12 had a more complex effect on aconitase and IRP-1 activities. Mitochondrial aconitase was more sensitive than c-aconitase to•NO-mediated inactivation and minimal activation of IRP-1 was observed during a 30-min exposure to the•NO donor. The action of•NO was down- or upregulated by the presence of extra- or intracelular [equation], respectively. Extracellular [equation] decreased the•NO-mediated inactivation of aconitases, due to the preferential extracellular decomposition and the lower diffusivity of peroxynitrite compared to•NO. On the other hand,•NO exposure concomitant with enhanced intracellular [equation] fluxes lead to intracellular peroxynitrite formation as evidenced by Western blot analysis of nitrated proteins, which increased the effects observed with•NO alone. Peroxynitrite-mediated aconitase inactivation, IRP-1 activation, and cellular protein nitration were more pronounced in cells with low GSH content such as V79-M8 glutathione-depleted cells as well as in pGSOD4 cells which contain 32% of the GSH of the parental strain. Mechanistically, our results imply that the differential actions of the studied reactive species toward cellular aconitases depend on at least three critical factors: (i) their reaction rates with aconitases, (ii) the cellular compartment where they are formed, and (iii) the intracellular status of glutathione.

A summary of the resource.
Solubility tests of copper(ll) complexes with amino acids, of potential pharmacologic interest, were performed using a series of twelve solvents with dielectric constants ranging between 111 and 2.24. The lipophilicity of these complexes were also tested, determining their partition coefficients between physiologic solution and n-octanol. For those complexes containing water of crystallization, the stability of the hydrates was determined from thermogravimetric measurements.

Abstract
A summary of the resource.
Several novel semicarbazones derivatives were prepared from 5-nitro-2-furaldehyde or 5-nitrothiophene-2-carboxaldehyde, and tested in vitro as potential anti-trypanosomal agents. The compounds were prepared in good to excellent yields in 2–3 steps from readily available starting materials. Some derivatives were found to be active against Trypanosoma cruzi with an activity similar to that of Nifurtimox.

Abstract
A summary of the resource.
The purpose of this study is to assess the environmental lead exposure of a group of people living near a lead smelter aiready closed. Blood lead levels of 84 adults and 11 children living at a residential complex (Euskal Ema) situated 2 km far from the source were determined. Personal data collected for each subject was concomitant with the taking of blood samples. The data collected included sex, age, time of residence and symptoms that could be related to lead exposure. Lead in blood was determined by flame atomic'absoprtion spectrophotometry after complexation of lead with DDDC and direct extraction into meíhylisobuíhyl- ketone using as a Perkin Elmer 2380 equipment. The analytical method was validated by an international intercalibration program. A control population of 29 unexposed adults and 34 unexposed children from different places of Montevideo was included in the study. The average blood lead concentration for adults was 9.3 µg % (range 6.1-15.0) and 10.0 µg % for children (range 6.0-16.9).For the population under study the average blood lead concentration obtained was 8.6 µg % for adults (range 3.1-24.8) and 9.3 µg % for children (range 6.1-15.0). The results obtained were beneath the limits established by WHO, and could be compared with the control population. They also show that there are no other lead sources in the residential environment considered.

A summary of the resource.
Chondriamide C (3), a new bis(indole) amide, was isolated from the red alga Chondria atropurpurea, and its structure was established from spectroscopic data and chemical transformations. A new natural product, 3-indoleacrylamide (4), and the previously described chondriamides A and B (1, 2) and 3-indoleacrylic acid (5) were also isolated. The anthelmintic activities of compounds 1, 3, 4, and 6 (the O,N1,N1'-trimethyl derivative of compound 2) against Nippostrongylus brasiliensis in vitro were evaluated.

The infrared spectra of the bis-chelated Cu(I1) complexes of the amino acids glycine, alanine, leucine, isoleuceine, valine and phenylalanine, which posses different kind of pharmacologic activity, were recorded and analyzed in relation to its structural peculiarities. Some comparisons between the recorded spectra are also presented and the characteristics of the carboxylate motions as well as those of the metal-to-ligand vibrations are discussed in detail.

A summary of the resource.
We analysed specific antibody (Ab) and circulating antigen (CAg) profiles along experimental mouse infection using as control a group of mice immunized with intact but dead parasites. Results from this experiment showed an early major CAg peak followed by a larger Ab peak which partially overlaps with other minor CAg peaks. These results suggest that CAg may be a marker of early mouse infection. In order to study the relevance of these findings in humans we similarly analysed by ELISA 148 sera provided by retrospective post-surgical follow-up of 19 patients. Available records showed that 14 patients developed new cysts one to ten years after surgery while no new disease was observed in the other five. Some of the former patients showed CAg as early as two months after surgery while no CAg was observed in the other five patients at any time. In addition, a collection of 38 sera obtained before surgery were similarly tested and five of them showed only CAg, while 18 showed only Ab and 12 sera showed Ab&CAg. These results in humans are consistent with the findings in the mouse experimental model and suggest that CAg may be an early marker of hydatid infection, thus being relevant for post-surgical follow-up.

Abstract
A summary of the resource.
Macrocycles possess desirable properties that make them promising candidates for the discovery of novel drugs. They present structural features to favor bioactive conformations, selectivity to the receptors, cell permeability and metabolic stability. More effective and nontoxic drugs to cure human African trypanosomiasis, Leishmaniasis and Chagas disease are needed, especially because resistance has been detected. Therefore, major efforts should be made for investigation in new bioactive compounds exhibiting different mechanisms of action. Macrocycles might fulfill the expectations for the development of new drugs to treat those diseases. In the current review, we focus on macrocycles exhibiting biological activities as antitrypanosomal and/or antileishmanial. The isolation, synthetic and biological studies of this class of compounds published from 2005 to 2014 are summarized.

A summary of the resource.
As albendazole sulphoxide (ABZS) shows better dissolution properties than albendazole (ABZ), a lipidic matrix with this drug was formulated in order to evaluate if its absorption and so systemic infection chemotherapy could be improved. A cross-over, randomised study in 8 healthy volunteers was carried out, after single administration of 1 g of albendazole or albendazole sulphoxide in lipidic matrix of Gelucire 44/14 (ABZLM and ABZSLM). Absorption was followed performing albendazole sulphoxide dosage in urine samples by high pressure liquid chromatography analysis, during 48 hours. Significant differences were found (p = 0.02) between the urinary recoveries (% E48), being 1.74% and 0.19% the percentage of dose recovered when ABZSLM or of ABZLM were respectively administered. In a previous study of our group similar values were obtained of urinary recovery percentages after albendazole sulphoxide powder administered to another group of healthy volunteers. Lipidic matrix does not improve the physicochemical properties of albendazole sulphoxide powder.

Using two approaches, reaction-path dynamics (which uses information on electronic structure energy and energy derivatives calculated ab initio along the minimum energy path) and dual-level dynamics (in which the reaction-path is calculated at a low level of theory, and stationary point information from a high level of theory is used to interpolate corrections to energy quantities, vibrational frequencies, and moments of inertia), the reaction-path for the CH3F + OH reaction was traced. Qualitatively, both methods yield similar results, showing the usefulness of the more economical dual-level approach. The H2O product may be vibrationally excited due to the nonadiabatic flow of energy between the reaction coordinate and the O−H bound mode. The rate coefficents were calculated for the temperature range 200−1000 K using the variational transition-state theory. The tunneling effect was included in the first approach using the small-curvature tunneling method, and in the second using the microcanonical optimized multidimensional tunneling method. Tunneling plays an important role in this reaction and the calculated rate coefficients show a more pronounced curvature in the Arrhenius plot than the experimental data.

Abstract
A summary of the resource.
A new anaerobic, proteolytic, moderately thermophilic bacterium, strain 3RT, was isolated from a methanogenic mesophilic reactor treating protein-rich wastewater. The cells were Gram-negative, non-spore-forming, non-motile rods. The DNA base composition was 43 mol% G + C. The optimum pH and temperature for growth were 7.0 and 55 degrees C respectively. The bacterium fermented gelatin, casein, bovine albumin, peptone and yeast extract. Glucose, fructose, sucrose, maltose and starch were poorly fermented. The major fermentation products from glucose were acetate, CO2 and H2 and, from gelatin, propionate was also detected. Growth on glucose was stimulated by thiosulfate, which was reduced to sulfide. Sulfate and nitrate were not reduced. 16S rRNA gene analysis revealed that the isolated bacterial strain was phylogenetically related to Coprothermobacter proteolyticus (96.3% sequence similarity), the only known species within the genus. DNA-DNA hybridization analysis demonstrated a very low level of homology, indicating that the isolated strain and C. proteolyticus were not related at species level. Enzymatic dioxygenation of benzyl azide by toluene dioxygenase produces the expected enantiopure cis-cyclohexadienediol along with an exocyclic diene formed by a spontaneous sequence of two [3,3] sigmatropic shifts. This novel dienediol presents high synthetic potential for natural product synthesis. The sigmatropic rearrangements can be reversed by protection of the diol moiety. An optimized production protocol for either of these valuable diols is presented.

Substitution of residues Y29 and W310 by smaller amino acids provided increased activity on C18-substrates. The crystal structure of the binuclear Cu(II) complex [Cu(mef)2DMSO]2 (mef = deprotonated N-2,3-dimethylphenyl-anthranilic acid; DMSO = dimethylsulfoxide) has been determined by single-crystal X-ray diffractometry. It crystallizes in the triclinic space group P1̄ with Z = 1. IR and electronic spectra of the compound are also discussed briefly.

Major drawbacks for enzyme-catalyzed transesterification of vegetable oil triglycerides to produce biodiesel (esters of short chain alcohols) are the high concentration of alcohol in the reaction mixture and the deposition of the produced glycerol on the enzyme support. Although stepwise addition of alcohol to the reaction mixture solves the first problem, an effective way to remove glycerol from the reaction system is needed. In the present study the efficiency of three systems for removal of glycerol from the reaction mixture during the enzymatic synthesis of sunflower oil methyl esters was investigated. By coupling a single cold column to the reactor 87.6% of total glycerol produced by the reaction was removed, while 10.9% was still retained by the enzyme. The percentage of glycerol separated from the reaction system increased to 93.1% when a second separation column packed with Amberlite BD10DRY resin was included, reducing the percentage of glycerol retained by the enzyme to 5.8%. Finally, changing the geometry of cold separation column in which a conical decanter was used, the percentage of glycerol separated increased to 99.2%, while only 0.4% was retained by the enzyme. Results demonstrated that use of a convenient separation system such as those described before, glycerol produced during transesterification of sunflower oil can be efficiently removed from the reaction mixture. In this way the well-documented inhibitory effects of the glycerol product are controlled and enzymatic catalysis remains fully functional

A summary of the resource.
The crystal structure of the binuclear Cu(II) complex [Cu(flu)2DMF]2 (flu = deprotonated N-3-trifluromethylphenylanthranilic acid: DMF = dimethylformamide) has been solved by single crystal X-ray diffractometry. It crystallizes in the monoclinic space group P21/c with Z = 2. The infrared and electronic spectra of the compound are also briefly discussed. Kristallstruktur und spektroskopisches Verhalten eines zweikernigen Kupfer(II)-Komplexes von Flufenam-Säure und Dimethylformamid Die Kristallstruktur des zweikernigen Kupfer(II)-Komplexes [Cu(flu)2DMF]2 (flu = deprotonierte N-3-trifluoromethylphenylanthranilsäure; DMF = Dimethylformamid) wurde röntgenographisch an Einkristallen bestimmt. Er kristallisiert in der monoklinen Raumgruppe P21/c mit Z = 2. Die Infrarot- und Elektronen-Spektren der Verbindung werden kurz besprochen.

Sample configurations from check-all-that-apply (CATA) questions are obtained using Correspondence Analysis (CA). Classical CA is based on chi-square distance, which has been reported to be strongly affected by infrequently selected terms. The Hellinger distance has been proposed as an alternative distance metric, and the aim of the present work was to compare product spaces from CATA questions obtained using CA based on chi-square and Hellinger distances. Data sets from 71 studies (5121 consumers), differing by product category, number of consumers, number of samples and number of terms included in the CATA question, as well as frequency of infrequently used terms, were analyzed. For each of the studies, frequency tables were input to CA based on chi-square and Hellinger distances. Sample and term configurations in the first two dimensions were compared using the RV coefficient. Furthermore, the stability of sample and term configurations for each type of distance was evaluated by simulating repeated experiments using a bootstraping resampling approach. Sample and term configurations obtained using Hellinger and chi-square distances were similar (average RV coefficients for sample configurations = 0.99; average RV coefficients between term configurations = 0.89). The stability of sample and term configurations were not largely affected by the type of distance used to analyze frequency tables. Results from the present work suggest that CA based on chi-square and Hellinger distances provide similar results. Contributing to guidelines for practitioners, this research therefore supports classical CA analysis as an acceptable approach to the analysis of sensory-specific CATA data.

Bioavailability and bioequivalence evaluations of drug products carried out using the experimental maximum concentration (Cmax) and the experimental time to reach Cmax (Tmax), are not advisable for slow-release formulations and for trials performed with saliva as biologic fluid. When slow-release curves are considered the drug concentration profiles usually show multiple peaks, making it difficult to compute a Cmax, Tmax value. The saliva profile throughout time shows a high variability observed as more than one peak in the saliva concentration versus time curves. In both cases, even if there is a major peak, when the statistical analysis of the data is performed, an important variability in Cmax results in high values in the residual variance of the ANOVA test. Consequently, the power of the bioequivalence test decreases and sometimes it is not possible to conclude on bioequivalence. The average concentration (Cav), the average maximum concentration (Cmax, av) and Cmax,av/Cav x 100 (%Cmax,av) are proposed in this paper as possible parameters in order to evaluate the profile of the concentration-time curves, as they reduce the residual variability in bioequivalence studies.

In order to assess the extent and the rate of absorption in bioavailability studies, area under the curve (AUC), experimental maximum concentration (Cmax) and experimental time to reach Cmax (Tmax), are used. But when slow-release formulations are considered, the drug concentration-time curves usually show multiple peaks, and it is difficult to compute a Cmax and Tmax value. In case a Cmax value is computed, important variability in this parameter results in high values in the residual variance of the ANOVA test. So in order to decrease the high variability, average parameters: average concentration (Cav), average maximum concentration (Cmax,av) and Cmax,av x 100/Cav (%Cmax,av), are proposed. These new parameters were applied in a bioavailability study of slow-release amitriptyline formulation.

Yet, key questions for this approach remain unaddressed – how to generate the sensory terms that populate CATA questions and how many terms should be used. The second of these questions was addressed in seven consumer studies, involving a total of 735 consumers and five product categories (crackers, cheese, fruit-flavored drinks, chocolate, milk desserts). Sensory product characterizations elicited with ‘‘short’’ and ‘‘long’’ CATA questions (10–17 terms vs. 20–28 terms) were compared on a number of criteria such as frequency of CATA term use, product differences, spatial configurations (samples and terms) and task perceptions. Two strategies for generating ‘‘long’’ lists of CATA terms were examined: adding synonym terms to those already featuring on the ‘‘short’’ list (e.g., ‘hard’ and ‘firm’), and adding antonym terms to those already featuring on the ‘‘short’’ list (e.g., ‘hard’ and ‘not hard’ or ‘natural’ and ‘artificial’). Between-subjects experimental designs were used to compare product characterizations from ‘‘short’’ and ‘‘long’’ CATA questions. Results revealed that ‘‘short’’ and ‘‘long’’ lists of CATA terms generated largely similar results. In general, sample configurations were very similar, as were task perceptions. However, there were, at times, differences in frequency of CATA term use and term configurations, as well as instances where conclusions about sample differences depended on whether ‘‘short’’ or ‘‘long’’ CATA lists were used. Additionally, here was some evidence that CATA questions with ‘‘long’’ lists of synonym or antonym terms may cause a ‘‘dilution’’ effect of the responses. This fits expectations of idiosyncrasy in consumer perception/expression of sensory stimuli, but may be associated with reduced discriminatory ability of the CATA question. How to best balance these opposing considerations is deserving of further investigation.
