Several radiolabeled alpha-melanocyte stimulating hormone (α-MSH) analogs have been studied for their abilities to target melanoma tumor cells through specific recognition and binding to the melanocortin receptor 1 (MCR1). In this work, a lactam bridgecyclized α-MSH analog was labeled with 99m via the hydrazinonicotinamide (HYNIC) chelator and characterized for its melanoma tumor targeting properties. The bifunctional chelating agent HYNIC-Boc was attached to the N-terminus of the MSH peptide followed by the lactam cyclization, resulting in the HYNIC-cyc-MSH analog. The lactam cyclized peptide displayed high affinity and specificity for MC1-receptors present on B16/F1 melanoma tumor cells, exhibiting an IC50 of 6.48 nM. HYNIC-cyc-MSH was radiolabeled with 99mTc using two common co-ligands, tricine and EDDA. In vitro, the radiochemical stability, cell binding and efflux properties were similar between the peptides radiolabeled with tricine and EDDA as co-ligands. In vivo, biodistribution studies (n=4) demonstrated that 99mTc- HYNIC-cyc-MSH/tricine had superior tumor to muscle and tumor to blood ratios than 99mTc-HYNIC-cyc-MSH/EDDA at early time points. OBJECTIVE: There is little evidence regarding the role of (68)Ga-DOTATATE PET/CT for the identification of primary tumors in patients with metastatic neuroendocrine carcinoma of unknown primary. The aim of this study is to assess the value of this technique in the mentioned clinical scenario. METHODS: We retrospectively studied twenty-nine patients (mean age 59.5 ± 10.6 years; female 17) with pathologically proven neuroendocrine metastases. In all cases conventional imaging was negative for primary tumor identification. (68)Ga-DOTATATE PET/CT was performed with a mean dose of 104.2 ± 18.8 MBq, using a 64-slice PET/CT with time-of-flight correction. A team of an experienced radiologist and a nuclear medicine physician evaluated the images. The maximum SUV (SUVm) was measured in all abnormal foci. Histopathology (when available) and/or clinical follow-up with correlative imaging was considered as reference standard. RESULTS: (68)Ga-DOTATATE PET/CT identified the primary tumor in 17/29 (59%) patients in the following locations: pancreas (n = 7), ileum (n = 7), duodenum (n = 1), colon (n = 1) and stomach (n = 1). In this population a significant correlation was found between SUVm of primary tumor and metastases (r = 0.815, P < 0.0001). Furthermore, additional sites of unsuspected metastases were demonstrated in 9 patients of this group and in 6 patients in whom no primary tumor was localized, mainly in lymph nodes and mesentery. Pathology confirmation was obtained in 7 patients who underwent surgery, whereas in the remaining 10 patients, correlative imaging and follow-up confirmed primary tumor localization. CONCLUSIONS: (68)Ga-DOTATATE PET/CT is a clinically useful imaging technique for the localization of primary tumors in patients with neuroendocrine metastatic carcinoma of unknown origin with the potential of having a significant impact in patient management and therapy planning. In the search of new therapeutic tools for the treatment of American Trypanosomiasis, the largest parasitic disease burden in the American continent, three series of novel ruthenium complexes of the formula [RuCl2(HL)2], [RuCl3(dmso)(HL)] and [RuCl(PPh3)(L)2] with bioactive 5-nitrofuryl containing thiosemicarbazones as ligands (HL neutral, L monoanionic) were synthesized and characterized. A nivel internacional existen importantes avances en el estudio de granos y harinas de diferentes cereales. En particular el estudio en trigo ha estado vinculado a las modificaciones en las propiedades reológicas de las masas formadas a partir de las harinas de este en función del contenido proteico. Existen también estudios que vinculan a componentes minoritarios, los arabinoxilanos, en relación a las propiedades anteriormente indicadas, donde las evidencias según los diferente autores, resulta contradictoria. Estas contradicciones posiblemente se deben a que las investigaciones generalmente estudian el efecto del agregado de compuestos aislados en lugar de estudiar el efecto en las variaciones de los mismos presentes en las harinas. En nuestro país, este trabajo es la primera aproximación a evaluar la influencia de la composición química de los carbohidratos no amiláceos sobre los parámetros tradicionalmente asociados a la calidad panadera de las harinas de variedades locales. Se seleccionaron harinas representativas de la variabilidad local con respecto a los principales parámetros predictores de calidad, con contenidos de proteínas de acuerdo a los requerimientos para su comercialización, de las cuales se encontró que, en su mayoría, desarrollarán el gluten de manera adecuada durante el amasado. Se encontró que el volumen del pan, la formación del gluten y la proporción de proteínas que lo forman son afectados negativamente por los arabinoxilanos, posiblemente por la competencia en la absorción de agua durante el amasado la cual es afectada por las propiedades estructurales de éstos últimos, tanto por sus ramificaciones como por la presencia de ácidos fenólicos que influyen sobre su capacidad para formar agregados durante el amasado. Se determinaron asimismo variaciones estructurales en los arabinoxilanos por el proceso de amasado y panificación con el agregado de enzimas específicas confirmando la influencia sobre las interacciones que ocurren entre estos componentes minoritarios y el gluten. Se encontró que las muestras demasiado elásticas que resisten a ser extendidas presentan mayores contenidos de arabinoxilanos insolubles. Both complexes including dipyrido[3,2-a: 2',3'-c]phenazine (dppz) as polypyridyl coligand showed IC(50) values in the muM range against Dm28c strain (epimastigotes) of Trypanosoma cruzi, causative agent of the disease, being as active as the anti-trypanosomal reference drug Nifurtimox. To get an insight into the trypanocidal mechanism of action of these compounds, DNA was evaluated as a potential parasite target and EPR, and (51)V NMR experiments were also carried out upon aging aerated solutions of the complexes. In the search for new therapeutic tools against Chagas disease (American trypanosomiasis) palladium and platinum complexes of the bioactive ligand pyridine-2-thiol N-oxide were exhaustively characterized and evaluated in vitro. Both complexes showed high in vitro growth inhibition activity (IC50 values in the nanomolar range) against Trypanosoma cruzi, the causative agent of the disease. They were 39–115 times more active than the antitrypanosomal drug Nifurtimox. The palladium complex showed an approximately threefold enhancement of the activity compared with the parent compound. In addition, owing to their low unspecific cytotoxicity on mammalian cells, the complexes showed a highly selective antiparasite activity. To get an insight into the mechanism of action of these compounds, DNA, redox metabolism (intraparasite free-radical production) and two parasite-specific enzymes absent in the host, namely, trypanothione reductase and NADH-fumarate reductase, were evaluated as potential parasite targets. Additionally, the effect of metal coordination on the free radical scavenger capacity previously reported for the free ligand was studied. All the data strongly suggest that trypanocidal action of the complexes could mainly rely on the inhibition of the parasite-specific enzyme NADH-fumarate reductase. In order to characterize GATA transcription factors in Echinococcus granulosus, a PCR-based cloning strategy was developed. Degenerate oligonucleotides were designed for the most conserved sequence in GATA proteins that include 20 amino acids of the zinc domain. A 60 bp fragment was isolated that had high homology among this sequence and those reported in other species. An analogous sequence was obtained by performing the same procedure with DNA from the free living platyhelminth Dugesia tigrina. Searching for new promising metal-based drugs for the treatment of parasitic diseases against Trypanosoma cruzi, three related oxidovanadium(IV) complexes, [VIVO(SO4)(H2O)2(NN)], with the phenanthroline derivatives (NN) 1,2,5thiadiazolo[3,4-f]1,10phenanthroline (tdzp), 1,10-phenanthroline-5, 6-dione (phendione), and 5,6-epoxy-5,6-dihydro-1,10-phenanthroline (epoxyphen) are synthesized, characterized, and evaluated in vitro as anti-T. cruzi agents. The compounds are characterized in the solid state and in solution by elemental analysis, electrospray ionization mass spectrometry (ESI-MS), conductimetric measurements, and infrared (FTIR), UV/Vis, and electronic paramagnetic resonance (EPR) spectroscopy. EPR spectroscopy suggests that the ligands act as bidentate, binding through both nitrogen donor atoms in an axial-equatorial mode. DFT calculations corroborate the structural assignments. The stability of the complexes in solution is evaluated by EPR and 51V- NMR spectroscopy and all complexes show reasonable stability. The anti-T. cruzi activity of the complexes was tested by measuring the growth inhibitory effect on the epimastigote life cycle form of the parasite (Dm28c strain). All complexes show IC50 values in the micromolar range against T. cruzi and display activities of the same order of that of Nifurtimox, but lower than that of the previously reported analogue [VIVO(SO4)(H2O)2(dppz)] (dppz = dipyrido[3, 2-a:2′,3′-c]phenazine). Furthermore, DNA was evaluated as a potential target by using atomic force microscopy (AFM), showing that the complexes display ability to interact with this biomolecule. Biological results of these copper compounds have deserved some of them entering clinical trials. Significant efforts have been devoted to the in-depth identification of their mechanism of action. Using gel electrophoresis analysis, we have previously shown that the interaction of the Casiopeínas® Cas II-gly, [Cu(4,7-dimethyl-1,10- phenanthroline)(glycinate)]NO3 with DNA, triggers the cleavage of the biomolecule by a free radical mechanism. In this work, we further study the behavior of different complexes of the same Casiopeínas® series also including glycinate as co-ligand {Cas VI-gly (5,6 dimethyl-1,10- phenanthroline glycinato copper(II) nitrate), Cas VII-gly (1,10-phenanthroline glycinato copper(II) nitrate), and Cas IX-gly (2,2′-bipyridine glycinato copper(II) nitrate)} and of a Casiopeínas® with a different co-ligand (Cas III-Cs; 4,7-dimethyl- 1,10-phenanthroline salicylaldehydato-copper(II) nitrate). While all of them produce DNA degradation, the performance in the presence of a radical scavenger suggests the existence of differences in their mechanism of interaction with DNA. Particularly, citrus peel is a source of cold-pressed EO, where limonene is its major component, and can be used as an additional food component for ruminants. The aim of the present work was to determine the in vitro anthelmintic activity of EO from Citrus bergamia (EOB), C. x paradisii (EOG) and limonene against the benzimidazole-susceptible Kirby isolate of H. contortus, using the egg hatch test (EHT) and the exsheathed third stage larval motility test (XLMT) using a WMicroTracker equipment. Albendazole (ABZ) and monepantel (MON) were used as positive controls. The 50% inhibitory concentrations (IC50) in XLMT were 8.77 and 13.88 μg/ml for EOB and EOG respectively, after an incubation of 72 h. An interesting observation on XLMT resulted when the positive controls were tested on the same plate, but in different well of the EOB. The volatile components of the EO significantly influenced (P < 0.05) the percentage of larval motility, reducing values from 66.9 to 19.6% for ABZ, and from 72.8 to 33.7% for MON, when comparing the activity of positive controls in a control plate without EO. The in vitro anthelmintic activity of EOB and EOG shows that they could be interesting candidates for nematode control. It is still necessary additional studies against the adult stage of H. contortus in efficacy trials in infected animals to validate their anthelmintic activity. (Martyniaceae) and some of its derivatives, against methicillin–sensitive and MRSA. Compound 1 was isolated and identified as previously described and minimal inhibitory concentration (MIC) of 1 and simple analogs were determined by the microdilution technique. Compound 1 (11–hydroxystearic acid 11–0–(6–0–acetyl–β–D–grucopyranoside) showed a MIC of 0.06 mM for S. aureus ATCC 6538p. All the synthetic analogs and precursors presented higher MIC values. Compound 1 showed antimicrobial activity against different strains of MRSA. These results suggest that the glycosidic and acidic moieties are necessary for the antimicrobial activity under study. Low force stereolithography is exploited for the first time for one-step facile fabrication of chemiluminescence (CL) flow-through cells that bear unrivalled features as compared to those available through milling or blowing procedures or alternative 3D printing technologies. A variety of bespoke cross-section geometries with poly hedral features (namely, triangular, square, and five-side polygon) as well as semicircular cross-section are herein critically evaluated in terms of analytical performance against the standardcircular cross-section in a flat spirally-shape format. The idea behind is to maximize capture of elicited light by the new designs while leveraging 3D printing further for fabrication of (i) customized gaskets that enable reliable attaching of the active mixing zone of the CL cell to the detection window, (ii) in-line 3D-printed serpentine reactors, and (iii) flow confluences with tailorable shapes for enhancing mixing of samples with CL reagents. Up to twenty transparent functional cells were simultaneously fabricated without inner supports following post-curing and surface treat ment protocols lasting less than 5 h. In fact, previous attempts to print spirally-shaped cells in one-step by resorting to less cost effective photopolymer inkjet printing technologies were unsuccessful because of the requirement of lengthy procedures (>15 days) for quantitative removal of the support material. By exploiting the phthalazinedione-hydrogen peroxide chemistry as a model reaction, the five-side irregular pentagon cell exhibited superior analytical figures of merit in terms of LOD, dynamic range and intermediate precision as compared to alternative designs.Computational fluid dynamic simulations for mapping velocities at the entry region of the spiral cell corroborated the fact that the 5-side polygon cross-section flow-cell with Y-type confluence permitted the most efficient mixing of reagents and sample while enabling larger flow velocities near the inlet that contribute to a more efficient capture of the photons from the flash-type reaction. The applicability of the 3D-printed 5-side polygon CL cell for automatic determination of hydrogen peroxide using a computerized hybrid flow system was demonstrated for the analysis of high matrix samples, viz., seawater and saliva, with relative recoveries ranging from 83 to 103%. Paleolimnological data are presented on trophic development, climatic change and sea level variations in Negra Lagoon, a 142 km2 coastal lagoon in southern Uruguay. The strong correspondence between the spatial arrangement of archaeological sites and wetland environments is one of the main axes of the Merín Lagoon basin archaeology. Without implying a deterministic simplification from the epistemological point of view, the cultural and environmental history of the region hold evidence suggesting a more complex scenario, where cultural responses to climate change were not so mechanical or direct as previously argued. The relationship between paleoclimate and prehistoric mound builders development between 7.0 and 0.6 ka 14C BP was studied in SE Uruguay. Paleoclimatic data were inferred from the phytolith record of three lagoon sediment cores. Four paleoclimatic periods were identified according to temperature/humidity changes. The first period, dated at 7.0e4.5 ka 14C BP, was characterized by a warm/humid climate. Between 4.5 and 3.5 ka 14C BP, the onset of a cool/dry climate was inferred. A hiatus between 3.5 and 2.6 ka 14C BP was identified. In the third period, 2.5e1.2 ka 14C BP, similar climate conditions to those of the present were inferred. Finally, the period 1.2 and 0.6 ka 14C BP, was characterized by warmer and wetter conditions than those of the present. According to these data, it was determined that mounds were first observed during the transition from warm/humid to cool/dry conditions, but the complexity of the mounds increased after the full establishment of cool/dry conditions. With the inception of warm/humid conditions, an increased number of mounds was observed in the plains located close to the Merín Lagoon littoral. The mounds showed lower density and height than those recorded for the cool/dry period. A multiproxy analysis of diatoms, chrysophyte cysts, opal phytoliths, and palynomorphs was undertaken in Negra Lagoon, to decipher Holocene changes in paleobotanical proxies associated with sea-level changes and paleolimnological conditions. Before the Holocene transgression (7000 yr BP), a terrestrial system was inferred as no aquatic palynomorphs or biogenic silica remains were recorded. During the sea-level maximum (5200 yr BP), marine/ brackish conditions were established as indicated by diatoms, Chenopodiaceae, and a high content of cysts of Peridinioideae. The catchment consisted of grasslands and wetlands as indicated by the opal phytolith data. The Holocene transgression was followed by a sea-level decrease, which led to the onset of brackish/freshwater conditions as inferred from the co-dominance of freshwater and marine/ brackish diatoms. This is also supported not only by the concomitant increase in non-siliceous freshwater microalgae and emergent macrophytes, but also by the presence of Myriophyllum. As sea level continued to decrease during late Holocene, a freshwater system was observed because of the complete separation from the ocean. Chrysophyte cysts were consistently recorded and brackish diatoms exhibited reduced abundances. The observed limnological changes are consistent with the alternative states hypothesis of Scheffer’s model (1998). That is, the clear water phases were explained because of high macrophyte abundances, low phytoplankton frequencies, high cyst to diatom ratios, and increase in frequencies of benthic diatom species. Conversely, turbid phases were likely to occur when low cyst to diatom ratios together with increase in planktonic diatoms and decrease in macrophytes values were detected. This highlights the importance of this approach to detect long-term changes in shallow lake alternative states. Las experiencias didácticas vividas en formación docente inicial tienen un fuerte impacto en la conformación del perfil profesional del profesor . En estas experiencias se deben ver claramente trabajados los grandes fines en la enseñanza de las ciencias: formar ciudadanos críticos, formar para la participación ciudadana, alfabetizar cientí ficamente. La teniosis por solium, cisticercosis y triquinosis, son parasitosis en los humanos que se adquieren por consumo de carne de cerdo. En la formación inicial de docentes de Biología pa ra la enseñanza media, la s especies causantes de las parasitosis se estudian en la asignatura Zoología. Quebrando con el clásico abordaje de temas programáticos siguiendo la lógica de la disciplin a, se realizaron en la as ignatura trabajos con un enfoque de tipo CTS. Se organizó la conformación de equipos en el grupo clase, uno de los cuales abordó los helmintos parásitos transmitidos al hu mano por el cerdo. El trabajo incluyó la realización de una investigación bibliográfica, re colección de información en industrias locales que crían cerdo, la producción de una monogr afía y la presentación del tema a los demás compañeros de clase. Se vio el vínculo existente en tre la práctica religiosa judía y la ausencia en dicha comunidad de parasitosis por helmintos q ue se transmiten con el consumo de carne suina. Se analizaron cuestiones relacionadas con las costumbres alimenticias de la comunidad religiosa (su filosofía, sus libros sagrados), utilizándose lo s conocimientos de Zoolog ía y Parasitología para dar fundamento científico al vínculo señalado. Con esto se reconocieron diferencias entre preceptos religiosos y construcciones científica s, así como los fundamentos sanitarios de la adecuada tecnología en la cría del cerdo. La experiencia educativa realizada confirma que el tratamiento de temas disciplinares bajo un enfoque CTS es una forma de integrar el conoc imiento, promover la interacción e intercambio disciplinar y humanizar la cien cia. El enfoque apunta a la confluencia de propuestas que promueven la participación de los ciudadanos en los problemas sociales y ambientales. The 5-(n)-alkylresorcinol fraction of the epicuticular waxes of Hordeum vulgare seeds appeared to be responsible for their in-born resistance to pathogenic fungi such as Aspergillus niger and Penicillium crysogenum. The antifungal properties of this fraction were evaluated qualitatively and quantitatively with a novel bioassay where the extreme lipophilicity of these compounds was taken into account. The minimum inhibitory concentration in the fungi tested ranged from 5.6 to 10 micrograms cm-2 for the alkyresorcinols. The behaviour of the different cultivars against these fungi could be predicted by measuring the natural amount of resorcinols of each variety by TLC-scanning densitometry.
