Las bacterias endófitas son bacterias que viven dentro de los tejidos vegetales sin causar daño al organismo hospedero. Se ha postuladoque estas bacterias pueden aumentar la productividad de los cultivos por distintos mecanismos relacionados al aporte de nutrientes, producción de hormonas, y protección contra stress biótico y abiótico. En esta tesis se llevó a cabo el estudio de las comunidades de bacterias endófitas presentes en los distintos tejidos vegetales de arroz utilizando principalmente herramientas moleculares independientes del cultivo de microorganismos en el laboratorio. Se seleccionó este cultivo porque es de gran importancia económica y social para nuestro país y porque la planta y el suelo experimentan un cambio drástico durante el ciclo, como es la inundación. Se estudiaron la comunidad total y dos grupos fisiológicos particulares con relevancia biotecnológica y ambiental (bacterias diazótrofas y metanótrofas) así como distintos factores que pueden afectarlas (variedad de arroz utilizada, inundación del cultivo, tipo de suelo y su historia agrícola). Los resultados obtenidos revelaron que en las hojas de las tres variedades de arroz más sembradas en Uruguay la estructura de la comunidad de bacterias endófitases muy similar. Mientras Brevundimonas fue el género dominante en el análisis molecular (clonado del gen 16S rRNA) Pantoea ananatis y Pseudomonas syringae fueron las especies más frecuentemente aisladas en las tres variedades de arroz y en las dos zafras analizadas Se observó que la inundación como práctica habitual de manejo del cultivo de arroz tiene efectos muy importantes sobre la abundancia, diversidad y composición de las comunidades de bacterias diazótrofas endófitas de raíz. La abundancia de genes nifH, codificante para la dinitrogenasa reductasa, y su diversidad aumentan sustancialmente después de la inundación. Mientras que antes de inundar prevaleció la clase Gamaproteobacteria (66% de genoteca del gen nifH, dominando Stenotrophomonas con el 49% de las secuencias de nifH recuperadas), luego de la inundación se observó una gran diversidad de secuencias de genes nifH de bacterias anaerobias estrictas y facultativas (Spirochaetes, Chlorobi, Deltaproteobacteria y diversos Firmicutes). Estos resultados sugieren que los diazótrofos anaerobios podrían tener un rol importante en la fijación de nitrógeno en este ecosistema una vez que el cultivo de arroz es inundado. Asimismo, los diazótrofos endófitos de hoja se encuentran en una proporción casi 100 veces menor que en raíz y suelo de acuerdo a la cuantificación de genes nifH realizada por q-PCR. Las raíces de plantas de arroz sembradas en suelos de alta fertilidad con historia previa de cultivo de arroz presentaron una abundancia significativamente mayor que las raíces de las plantas sembradas en otros suelos, incluso mayor que aquellas sembradas en suelos similares que no habían sido utilizados previamente para el cultivo de arroz. La abundancia de las poblaciones diazótrofas endófitas de hoja parece ser independiente de las propiedades de los suelos en los las plantas se cultivan. Del mismo modo, no se observaron diferencias significativas en el número de copias del gen nifH en los distintos suelos, a pesar de sus características fisicoquímicas e historia de uso del suelo diferentes. 2 Por otra parte, las bacterias metanótrofas, de gran importancia porque contribuyen a mitigar las emisiones de metano producidas en el ecosistema arrocero, fueron detectadas en bajo número tanto en hojas como en raíces de plantas de arroz crecidas en suelo. Para ambos tejidos se obtuvieron cultivos altamente enriquecidos en metanótrofas capaces de crecer en ausencia de nitrógeno agregado indicando que se trata de metanótrofas diazótrofas. Esta propiedad podría ser importante en la interacción de este grupo fisiológico con la planta. La caracterización molecular de estos enriquecimeintos confirmó que Methylosinus trichosporium, una cepa metanótrofa C9 (aislada previamente de rizósfera de arroz en Uruguay) y Methylocystis hirsuta, se encuentran presentes en los tejidos internos de las hojas de arroz. El estudio realizado permitió generar nuevos conocimientos sobre las comunidades de bacterias endófitas nativas de arroz y profundizar en los factores que las afectan aportando información relevantesobre la interacción bacteria- planta. Asimismo, los conocimientos generados podrían ser útiles en la búsqueda de nuevas cepas con potencial biotecnológico y en la racionalización del uso potencial de inoculantes en el cultivo de arroz. DESIGN: A mixed-method approach was used, which included semi-structured interviews with school principals and a survey of the foods and beverages sold and advertised in the schools. SETTING: Primary and secondary schools in Montevideo (the capital city of Uruguay).ParticipantsSchool principals. RESULTS: The great majority of the schools did not comply with the initiative. Exhibition of non-recommended products was the main cause for non-compliance, followed by advertising of non-recommended products through promotional activities of food and beverage companies. Although school principals were aware of the healthy snack initiative and showed a positive attitude towards it, the majority lacked knowledge about its specific content. Factors underlying compliance with the healthy snacking initiative were related to its characteristics, characteristics of the schools, and external factors such as family habits and advertising. CONCLUSIONS: Results showed that the rationale underlying the selling of products at schools favours the availability of ultra-processed products and constitutes the main barrier for the promotion of healthy dietary habits among children and adolescents. Strategies aimed at facilitating the identification of unhealthy foods and beverages and provision of incentives to canteen managers to modify their offer are recommended. The growing evidence on the negative health effects of ultra-processed products indicates the need to develop strategies to discourage their consumption among infants and children. The development of such strategies requires a deeper understanding of the factors underlying children’s consumption of ultra-processed products. In this context, the present work aimed to analyze factors underlying mothers’ decision to offer meals composed of ultra-processed products during early childhood (6 months to 5 years old). An online survey with 888 mothers was conducted. Participants were shown a series of pictures of ultra-processed meals and were asked to indicate whether they would offer them to their children or not. A follow-up open-ended question was used to explore reasons underlying negative responses. A relevant proportion of mothers indicated that they would offer the ultra-processed meals to their children (48.5 % to 64.5 %). On the other hand, motives for not offering ultraprocessed meals to children were mainly related to food processing and nutritional composition. Several variables of the child (age, food fussiness), the mother (education level and socio-economic status), and the household (presence of children older than 5 years) had a significant effect on the likelihood of mothers offering ultra-processed meals and mentioning different reasons for not doing so. Results stress the need to implement strategies to increase perceived risk of ultra-processed products, particularly among parents with low educational level and from low socio-economic status. The aims of the presentworkwere to determine failure criteria based on consumers’ rejection to purchase for shelf life estimation of minimally processed lettuce, and to compare criteria for whole and cut lettuce. A trained sensory panel and a consumer panel evaluated samples of whole and cut lettuce leaves packaged in passive modified atmosphere. In order to determine failure criteria to estimate sensory shelf lives, sensory attribute intensities corresponding to 25% consumers’ rejection to purchase percentage were calculated using logistic regressions. Failure criteria values were lower for cut lettuce than for whole leaves for all the evaluated attributes, suggesting that consumers reacted differently towards whole and cut lettuce leaves, being stricter towards cut lettuce than towards whole lettuce leaves. These results indicate that sensory limits depended on the product considered and therefore a unique criterion should not be used to estimate the shelf life of both cut lettuce and whole lettuce leaves. Twenty-five percent of the consumers would refuse to purchase cut lettuce if the intensity of the evaluated defectswas over 10% of the measuring scale, whereas scores of 25% of the scale were needed to achieve a 25% of consumer rejection in the case of whole leaves. These failure criteria were stricter than those traditionally used for sensory shelf life estimation of minimally processed lettuce, which might assure the products’ quality at the end of its shelf life. La variabilidad interindividual en la respuesta y toxicidad a tratamientos es un problema muy común y serio, que causa preocupación y puede ser responsable de la reducción de la dosis o discontinuación del tratamiento. La Farmacogenética busca predecir dicha variabilidad, mediante el análisis y uso de la información genética individual o poblacional. El metotrexate(MTX), análogo estructural del ácido fólico, ha sido un fármaco altamente efectivo en tratamientos oncológicos y autoinmunes. Sin embargo presenta una gran variabilidad interindividual en su respuesta farmacológica y toxicidad. Una de las causas de dicha variabilidad es la existencia de polimorfismos en los genes que codifican proteínas transportadoras y dianas del MTX. El objetivo general de este trabajo fue contribuir al conocimiento para un diseño de una terapia personalizada con MTX. Como forma de realizar una aproximación a una vía tan compleja, desde el ingreso del MTX hasta su blanco de acción en la célula, nos propusimos evaluar la distribución y analizar la asociación entre la toxicidad y eficacia del tratamiento y los polimorfismos en los genes que codifican para los transportadores: de folato reducido (SLC19A1) G80A y de solutos de aniones orgánicos de la familia 1B1 (SLCO1B1) T521C; así como también en los genes que codifican para las enzimas involucradas en la actividad del MTX: 5,10- metilentetrahidrofolato reductasa (MTHFR) C677T y A1298C, variación en el número de copias de 28 pb en el gen timidilato sintasa (TYMS) y dihidrofolato reductasa (DHFR) C- 1610G/T, C-680A, A-317G y el polimorfismo que consiste en una inserción deleción de 19 pb. El grupo de estudio consistió en 41 pacientes adultos uruguayos con leucemia linfoide aguda (LLA) y linfoma no-Hodgkin (LNH) agresivo tratados con un protocolo de poliquimioterapia que incluyera mediana y/o altas dosis de MTX y en un grupo control de 55 adultos uruguayos voluntarios sanos. De dichos pacientes se relevaron los parámetros clínicos y de laboratorio obtenidos del seguimiento de la terapia. Dendritic cell-specific ICAM-3 grabbing non-integrin (DC-SIGN) expressed on a variety of DCs, is a C-type lectin receptor that recognizes glycans on a diverse range of pathogens, including parasites. The interaction of DC-SIGN with pathogens triggers specific signaling events that modulate DC-maturation and activity and regulate T-cell activation by DCs. In this work we evaluate whether F. hepatica glycans can immune modulate DCs via DC-SIGN. We demonstrate that DC-SIGN interacts with F. hepatica glycoconjugates through mannose and fucose residues. We also show that mannose is present in high-mannose structures, hybrid and trimannosyl N-glycans with terminal GlcNAc. Furthermore, we demonstrate that F. hepatica glycans induce DC-SIGN triggering leading to a strong production of TLR-induced IL-10 and IL-27p28. In addition, parasite glycans induced regulatory DCs via DC-SIGN that decrease allogeneic T cell proliferation, via the induction of anergic/regulatory T cells, highlighting the role of DC-SIGN in the regulation of innate and adaptive immune responses by F. hepatica. Our data confirm the immunomodulatory properties of DC-SIGN triggered by pathogen-derived glycans and contribute to the identification of immunomodulatory glyans of helminths that might eventually be useful for the design of vaccines against fasciolosis. The adult male transmits some of this alkaloid to the female at mating for eventual incorporation into the eggs. We now show by chemical analyses that the female herself is the first beneficiary of the alkaloid she receives from the male. By the end of mating the male's alkaloid is found already to be generally distributed throughout the female's body (including even the wings), while it is still largely absent from the ovaries. Adverse health consequences are associated with consumption of so-called trans fatty acids (FAs) generated during chemical hydrogenation processes. Naturally-occurring trans fats, including trans vaccenic acid (TVA) derived from a conjugated isomer of linoleic acid (18:2 cis-9, trans-11), (CLA) in contrast, are recognized as having potentially beneficial effects on human health. These natural trans FAs are found in fat from dairy products and meat of ruminants, and have distinct effects compared with trans FAs industrially produced. The lipid composition of cheeses depends on the milk used. In this work was determined CLA and TVA content in milk fat of sheep and cow to be 1.4% and 3.2%, respectively, and from goat, 0.7% and 1.8%. Also, differences in CLA and TVA content were found in fat extracted from samples of the same cheese type made from cow’s milk from the same company but with different production dates during a year. The levels of CLA and TVA found in Uruguayan cheeses were generally higher than levels reported in previous studies with comparable cheeses produced in other countries. Milk fat is well-known to confer specific properties to foods. This in turn will affect the rheology and sensory attributes as food. For this reason, it is of interest to determine the triglyceride (TAG) composition in milk-derived foods. Results show that there are characteristic differences in the TAG composition of the different cheeses. We found minimal differences between cheeses from goat and sheep but found distinct characteristics for TAGs from cow milk-derived cheese.
