A summary of the resource.
A diurnal variation study of carbamazepine (CBZ) and its metabolite carbamazepine-10,11-epoxide (EPOX) was carried out on six healthy volunteers, measuring drug levels in plasma and saliva throughout 34 hours. Four hundred milligrams of CBZ were given to subjects at 8:00 am or 8:00 pm, in a two-way crossover design. Drug and metabolite concentration in plasma and saliva stimulated with parafilm (after 24 hours post-dose) were higher during the night. The EPOX/CBZ metabolic ratio did not show any change between dark and light periods. The non-stimulated saliva/plasma concentration ratio (S/P) was higher during the day, but this quotient did not change when stimulated saliva was considered. Diurnal variation in the body blood flow seems to be a reasonable explanation for those differences. Factors such as protein binding or metabolic pathway are discarded as sources of carbamazepine chronopharmacokinetic variations.

The EPR of radicals obtained by electrolytic reduction of some new analogues of the antiprotozoal drug nifurtimox were measured in DMSO and DMF. The electrochemistry of these compounds was studied using cyclic voltammetry AM1 and INDO semiempirical molecular orbital calculations were performed to obtain the optimized geometries and spin distribution, respectively. Density functional theory was used to rationalize the reduction potentials of these compounds.

A summary of the resource.
Este artículo presenta los resultados de un estudio sobre la producción de compuestos conjugados de una beta-galactosidasa neutra (lactasa), mediante su inmovilización en tiosulfinato-agarosa. Se alcanzaron rendimientos de sobre 90% y los productos obtenidos mostraron buena estabilidad frente a agentes sanitizantes. También, estos productos mostraron ser muy efectivos para la conversión de lactosa en soluciones tamponadas, en lactosuer, en permeatos de lactosuero y en leche. Se realizaron cuatro ensayos en un bioreactor de columna que funcionó en forma continua por diecisiete días en cada experiencia. En todos los casos se logró una conversión del orden del 90%. Después de nueve meses de almacenamiento adecuado, los conjugados mantuvieron su actividad enzimática, demostrando su alta estabilidad.

A summary of the resource.
After reversible immobilization of neutral beta-galactosidase from Kluyveromyces lactis on to thiolsulfinate/thiolsulfonate supports, more than 80 % of the activity was retained. Blocking the remaining reactive groups with glutathione increased the thermal stability of the derivatives almost two-fold. These derivatives achieved a high degree of conversion (85-90 %) of lactose (50g / l) in saline solution, whey, whey permeates, and skimmed milk, either batchwise or in packed beds. They remained fully active upon storage for 10 months in activity buffer at 4 deg C, and when treated with sanitizing agents.

EgDf1 is a developmentally regulated protein from the parasite Echinococcus granulosus related to a family of hydrophobic ligand binding proteins. This protein could play a crucial role during the parasite life cycle development since this organism is unable to synthetize most of their own lipids de novo. Furthermore, it has been shown that two related protein from other parasitic platyhelminths (Fh15 from Fasciola hepatica and Sm14 from Schistosoma mansoni) are able to confer protective inmunity against experimental infection in animal models. A three-dimensional structure would help establishing structure/function relationships on a knowledge based manner. 3D structures for EgDf1 protein were modelled by using myelin P2 (mP2) and intestine fatty acid binding protein (I-FABP) as templates. Molecular dynamics techniques were used to validate the models. Template mP2 yielded the best 3D structure for EgDf1. Palmitic and oleic acids were docked inside EgDf1. The present theoretical results suggest definite location in the secondary structure of the epitopic regions, consensus phosphorylation motifs and oleic acid as a good ligand candidate to EgDf1. This protein might well be involved in the process of supplying hydrophobic metabolites for membrane biosynthesis and for signaling pathways.

Abstract
A summary of the resource.
The behaviour of wheat β-amylase from crude extracts in chromatography on agarose gels substituted with different ligand types was investigated. The enzyme displayed high salt promoted adsorption onto thiophilic gels provided with sulfone-thioether and 2-thiopyridine ligands. Quantitative recovery of the enzyme was easily accomplished by elution with buffer in the absence of Na2SO4. The 3-(2-pyridylthio)-2-hydroxypropylagarose (PyS-gel) also allowed elimination of pigments present in the wheat extract. These pigments showed no adsorption onto the gel, thus regeneration is easily achieved, allowing its re-use. The enzyme also displayed strong salt-dependent adsorption onto adsorbents provided with pyridyldisulfide moieties, but in this case enzyme binding was due to its thiol content since elution was achieved mainly through reduction with DTT. When the enzyme was chromatographed on a series of hydrophobic alkyl ligands in the presence of 0.5 M sodium sulphate, it was partially adsorbed on pentylagarose and quantitatively adsorbed on hexyl-agarose, elution being easily performed by sodium sulphate-free buffer. This behaviour was markedly different from that towards phenyl-Sepharose, to which the enzyme was strongly adsorbed and which required much more drastic elution conditions.

Polarized Sensory Positioning (PSP) is a relatively new methodology for sensory characterization, based on the comparison of samples to reference products, usually called poles. The key step in the implementation of this methodology is the selection of the poles. In this context, the aim of the present work was to gain insights on the strategies used by consumers to complete this task and to study the influence of the number and characteristics of the poles on results from PSP. Three consumer studies were carried out, on three product categories: chocolate-flavored milks, vanilla milk desserts and orange-flavored powdered drinks. In each study, three groups of consumers (n = 40) evaluated samples using one of three sets of two or three poles. After the PSP task consumers indicated how they evaluated the degree of difference between the samples and the poles, and listed the sensory characteristics of the poles they took into account for the evaluation. Consumers tended to evaluate the degree of difference between the samples and each of the poles by evaluating the intensity of one or two main sensory characteristics. Sample configurations obtained using two and three poles did not largely differ, especially when the poles clearly represented the sensory characteristics responsible for the main differences among samples. This work demonstrates that it is possible to unfold a multidimensional sensory space with the use of just two well-selected poles, and that the number of poles should be selected considering the main sensory characteristics that discriminate among samples.

Objective: Tostudy the influence of 2 interpretational aids of front-of-package (FOP) nutrition labels (color code and text descriptors) on attentional capture and consumers’ understanding of nutritional information. Design: A full factorial design was used to assess the influence of color code and text descriptors using visual search and eye tracking. Participants: Ten trained assessors participated in the visual search study and 54 consumers completed the eye-tracking study. Main Outcome Measures: In the visual search study, assessors were asked to indicate whether there was a label high in fat within sets of mayonnaise labels with different FOP labels. In the eye-tracking study, assessors answered a set of questions about the nutritional content of labels. Analysis: The researchers used logistic regression to evaluate the influence of interpretational aids of FOP nutrition labels on the percentage of correct answers. Analyses of variance were used to evaluate the influence of the studied variables on attentional measures and participants’ response times. Results: Response times were significantly higher for monochromatic FOP labels compared with colorcoded ones (3,225 vs 964 ms; P < .001), which suggests that color codes increase attentional capture. The highest number and duration of fixations and visits were recorded on labels that did not include color codes or text descriptors (P < .05). The lowest percentage of incorrect answers was observed when the nutrient level was indicated using color code and text descriptors (P < .05). Conclusions and Implications: The combination of color codes and text descriptors seems to be the most effective alternative to increase attentional capture and understanding of nutritional information.

A summary of the resource.
The levels of superoxide dismutase (SOD) were determined in detergent-soluble, somatic and excretion–secretion (E–S) preparations from adult Fasciola hepatica using the xanthine oxidase system and visualized in substrate gels. Compared to detergent-soluble and somatic extracts, E–S products showed the highest SOD activity (88 ·5 U/mg), indicating active release to the medium in which parasites were maintained. SOD specific activity was also detected at high levels in E–S products from 3-week-old and 5-week-old immature migrating flukes (25 and 143 U/mg, respectively). In all preparations except for the somatic extract, the activity was characterized as cyanide-sensitive CuZn SOD. Differences in SOD isoenzyme profiles between the extracts were observed in native polyacrylamide gel electrophoresis: the somatic and detergent-soluble extracts exhibited 1 band of activity while the E–S products from immature and adults flukes contained 2 and 3 migrating bands, respectively. SOD was purified from the detergent-soluble extract and E–S products of adult worms by a combination of ultrafiltration, gel filtration on Sephacryl S-200 HR and ion-exchange chromatography on QAE Sephadex A-50. The SOD from detergent-soluble extract showed, by SDS–PAGE analysis, 1 band of 16 kDa apparent molecular weight. The SOD from E–S products showed 2 bands of 16 and 60 kDa apparent molecular weight. N-terminal sequence analysis of the 16 kDa band from the detergent-soluble preparation showed some similarity with Schistosoma mansoni cytoplasmic SOD. These enzymes may have a potential role in the evasion of the oxidative burst killing mechanism by immune cells.

The kinetic isotope effects, KIEs, for hydrogen abstraction from the isotopologs CH4, 13CH4, CH3D, and CD4 by chlorine atoms have been studied by the dual-level direct dynamics approach with the MORATE computer program. Low-level calculations of the potential energy surface were carried out at the NDDO-SRP level (in particular the AM1-SRP level), using two different sets of specific reaction parameters labeled SRP4 and SRP13. High-level structural and energetic properties of the reactants, saddle point, and products were obtained at the MP2-SAC and MP2 levels using the 6-311G(2d,d,p) basis set and were used to interpolate corrections to the low-level calculations. The dual-level calculations were carried out using the ICL-Eckart improved interpolated corrections algorithm. Tunneling was included by the microcanonical optimized multidimensional tunneling (μOMT) method, and we find that large-curvature tunneling paths usually provide the dominant contribution, with significant participation of excited vibrational states. Both rectilinear and curvilinear coordinates were applied to the unsubstituted reaction. The 12C/13C KIEs calculated at the MP2-SAC///SRP4 level using MP2 frequencies are in very close agreement with the experimental ones, with values of 1.07 and 1.06 at 243 and 297 K, respectively, as compared to experimental values of 1.07 at both temperatures. For the reaction CH3D + Cl, the calculated H/D KIEs are equal to 1.55 and 1.45 at 223 and 296 K, while the measured values are equal to 1.59 and 1.50, respectively. The H/D KIE for the reaction CD4 + Cl is calculated to decrease from 11 to 4.7 as the temperature increases from 300 K to 450 K, whereas the experimental value decreases from 12 to 3.9. We also make comparison with previous results for the unsubstituted reaction CH4 + Cl.

Molecular dynamics simulations of the zinc finger domain of protein Zif268, in a complex with a high affinity DNA sequence, yields a globally stable system with small yet significant readjustments with persistence time of the order of 1.1ns. The results confirm the quality of the standard GROMOS87 force field with a corrected solvent-to-solute interaction that does not affect the water-water SPC interactions nor the intra-molecular cohesive forces. Specificity determinants are discussed. The simulations of DNA alone, with the same force field, showed the important role played by the solvent and the symmetry of the counterion distribution. (Tapia & Velázquez, J. Am. Chem. Soc., 119, 5934, 1997) In the present work, this feature was retained when appropriate. The results for root mean square deviations and temperature B-factors illustrate the reliability of this approach. The structure of DNA is held by its interactions with the zinc finger protein. This behavior is not much affected by the slow whithering away of finger-1 from DNA. The factors contributing to the molecular stability found in GROMOS' potential energy function appear to be sufficient to yield stable fluctuation patterns when surrounding medium effects are properly included.

In this paper, with the help of a nanosecond-long molecular dynamics trajectory, we show that a fully charged protein−DNA system (Zif268-DNA) in a water environment can be simulated with a general force field (GROMOS) if solvation and counterion effects are appropriately represented. A model exhibiting ionic-atmosphere effects on mobile counterions (Tapia, O.; Velázquez, I. J. Am. Chem. Soc. 1997, 119, 5934−5938) was implemented. The root-mean-square deviations (rmsds) with respect to X-ray structure for the full complex, the protein, and the 12-base pair consensus sequence were 2.0, 1.95, and 1.35 Å, respectively, while the counterions displayed an rmsd from the initially equilibrated position of 1.2 Å. The mean-square fluctuation with respect to the average structure correlated with temperature factors for the protein and DNA; the agreement in trend is good. The results show that GROMOS87 force field with an appropriate representation of coion (ion atmosphere) effects on the counterions, and corrections for hydrophobicity default in the water to solute C12(Ow,Ow)1/2 parameter, previously used for proteins (Daura X.; Oliva, B.; Querol, E.; Avilés, F. X.; Tapia, O. Proteins 1996, 25, 89−103), are sufficient to obtain a fairly realistic simulation of this protein−DNA complex.

En este artículo se describe la sínesis de bislunularina, un compuesto fenólico bisbencílico, aislado de briofitas. Las principales transformaciones incolucran la formación de enlaces carbono-carbono aromático mediante la reacción de Ullman y la formación de enlaces carbono-carbono alifático a través de Wittig. Se usó una simple estructura modelo para optimizar las condiciones de ciclación para obtener biflavona. Basado en los resultados, se sintetizó biflavanona, un precursor avanzado para la preparación de algunos flavonoides naturales.

A summary of the resource.
Albendazole is a poorly water soluble drug, with low oral bioavailability, used in pharmacological treatment of a systemic disease as hydatid parasitosis. Lipidic matrices of Gelucires (44/14 and 35/02) were developed. After "in vitro" studies, one formulation was chosen for a single dose study in 8 healthy volunteers, with a cross-over and randomised design, taking a commercially available tablet as reference. Drug absorption was followed by albendazole sulphoxide dosage in urine by high pressure liquid chromatography. Neither albendazole nor albendazole sulphoxide were recovered in urine after tablet administration while 0.18% (+/- 0.06) of dose was recovered after lipidic matrix administration in the first 24 hours. Besides ageing control were performed up to 18 months post-elaboration. Lipidic matrix with Gelucire 44/14 was revealed as a promising attempt for oral pharmaceutical form in albendazole systemic treatment.

En esta tesis se muestran tanto la caracterización in vitro como in vivo de péptidos y fagos expresando péptidos en su cápside contra el receptor ErbB3. La tecnología aquí presentada permite un rápido cribado a partir de grandes bibliotecas de fagos en una metodología clásica y funcional desde su descubrimiento como lo es la técnica de phage display. La importancia de esta diana celular radica en que la expresión de ErbB3 posee un valor pronóstico, dado que altos niveles de este receptor están asociados con una sobrevida más corta en pacientes que sobrexpresan ErbB2. A su vez, ErbB3 juega un rol compensatorio en la señalización asociada a la resistencia al tratamiento en cáncer de mama en tumores que sobrexpresan ErbB2. Por ello es de gran importancia monitorear la expresión de ErbB3 y diagnosticarlo in vivo, por lo que sería deseable el desarrollo de una agente de imagen que lo reconociese. El principal objetivo planteado en este trabajo fue el descubrimiento de un péptido anti-ErbB3 obtenido a partir de fagos seleccionados por phage display para imagenología molecular de tumores mamarios. En esta tesis se seleccionaron y desarrollaron dos fagos (MSPC4 y MSPC3) y dos péptidos anti-ErbB3 respectivamente. Se investigó la marcación y caracterización de los distintos péptidos y fagos, obteniéndose imágenes SPECT e imágenes con sistemas ópticos. Se desarrollaron tres métodos de marcación a través de sistemas bifuncionales como lo son DOTA y HYNIC. Se obtuvo para todos los complejos marcados con 99mTc, 111In y 177Lu, pureza radioquímica adecuada y estabilidad tanto in vitro como in vivo, así se mantuvo la selectividad y afinidad por el receptor a nivel in vitro. Los sistemas desarrollados en base a los fagos MSPC4 y MSPC3, demostraron tener captación selectiva por el modelo tumoral utilizado. Constituyéndose así como potenciales candidatos a radiofármacos para la detección de tumor de mama a través de imagenología molecular óptica o SPECT.

The structures of three furosemide prodrugs, pivaloyloxymethyl 4-chloro-N-furfuryl-5-sulfamoylanthranilate, C18H21ClN2O7S, butyryloxymethyl 4-chloro-N-furfuryl-5-sulfamoylanthranilate, C17H19ClN2O7S, and isobutyryloxymethyl 4-chloro-N-furfuryl-5-sulfamoylanthranilate, C17H19ClN2O7S, have been determined; the crystals have been shown to be isostructural. The crystal structures are described and compared with that of a related prodrug. The dihedral angle between the two planar rings of each prodrug is close to 70°. The space group is P\overline{1} in each case, and the molecules pack as dimers in infinite chains along one of the crystallographic axes.

Abstract
A summary of the resource.
The composition of the essential oil of Uruguayan Citrus clementine Hort., prepared in the laboratory from the fruit of Nules and Comune cultivars, has been studied. The volatile fraction was analysed by HRGC and HRGC–MS (quadrupole); 69 components were identified; the enantiomeric distribution of β-pinene, sabinene, limonene, linalol and α-terpineol was studied by multidimensional HRGC–HRGC. Polymethoxylated flavones present in the non-volatile residue were analysed by normal-phase HPLC. The results were compared with those of Italian clementine oil.

A summary of the resource.
The recognition profile of the tissue cysts antigens by IgG antibodies was studied during acute and chronic human toxoplasmic infection. Thus the IgG response against Toxoplasma gondii was investigated by immunoblotting in two patients accidentally infected with the RH strain as well as in group of naturally infected patients at acute and chronic phase. There was an overall coincidence of molecular mass among antigens of tachyzoites and tissue cysts recognized by these sera, however, they appear not to be the same molecules. The response against tissue cysts starts early during acute infection, and the reactivity of antibodies is strong against a wide range of antigens. Six bands (between 82 and 151 kDa) were exclusively recognized by chronic phase sera but only the 132 kDa band was positive in more than 50% of the sera analysed. A mixture of these antigens could be used to discriminate between the two infection phases. The most important antigens recognized by the acute and the chronic phase sera were 4 clusters in the ranges 20-24 kDa, 34-39 kDa, 58-80 kDa and 105-130 kDa as well as two additional antigens of 18 and 29 kDa. Both accidentally infected patients and some of the naturally infected patients showed a weak specific response against tissue cyst antigens.
