The geometries and binding energies of CO2 adsorbates on Pt(100) and Pt(111) cluster surfaces were calculated by means of an improved version of the extended Hückel molecular orbital method. The polarization of the surface by an applied electric potential and coadsorption of H atoms were included in the model. For simulated applied potentials in the range −1.0 to 1.0 V, CO2 coordination geometries (side-on, formate) involving two adsorbate atoms bonded to the surface are favored, regardless the surface topology and the presence of coadsorbed H atoms. In agreement with experiment, larger binding energies are always calculated for the Pt(100) cluster surface.

A summary of the resource.
The electrochemical adsorption characteristics of 3-buten-2-one and 2-butanone on porous platinum electrodes were investigated by differential electrochemical mass spectrometry (DEMS) in aqueous 0.5 M H2SO4. The study was conducted through the oxidation and reduction of residues formed at different adsorption potentials (Ead). The maximum adsorption for both organic substances was observed at the potential of zero charge of the interface, that is, ca 0.20 V. At this potential an adsorbate species with the C–O bond lying parallel to the surface is proposed. The sole oxidation product for both molecules was carbon dioxide. Anodic current transients were detected for 3-buten-2-one at adsorption potentials higher than 0.20 V. Between 0.40 and 0.60 V, it is possible to admit a 1–3e− loss during adsorption with a progressive fragmentation of the original molecule at increasing Ead. After three cycles in the H-adatom region, about 75% of the 3-buten-2-one adsorbate is desorbed; the main products being butane, 1-butene and propane. The ratio of these products depends on Ead, as is expected for the fragmentation of the molecule upon increasing potentials. On the other hand, 2-butanone was adsorbed in the 0.10–0.60 V potential range without current transients associated with the dissociative reactions of the molecule. The surface coverage diminished drastically at both sides of the maximum adsorption (0.20 V). Butane was the main reduction product of 2-butanone.

A summary of the resource.
An improved extended Hückel molecular orbital study was conducted to predict the adsorbate configuration of ethylene and acetylene on Au(100) and Au(111) single crystal cluster surfaces at different simulated electric potentials. Three adsorbed configurations on both cluster surfaces were found for ethylene and acetylene, i.e. π, di-σ and μ-bridging species, whose relative contributions to the total organic surface coverage depend on the electric potential. Acetylene is chemisorbed on gold in a π-bonded mode when a positively charged surface is simulated, whereas di-σ and μ-bridging state are the most likely adsorption modes at negative electric potentials, the binding energies on Au(100) being larger than those of Au(111) clusters. Ethylene, on the other hand, is physisorbed on gold at equilibrium and positive electric potentials, however the chemisorption of the molecule is achieved when a negatively charged surface is simulated. Adsorption of ethylene at potentials below that of zero charge of the gold solution interface was observed. The anodic stripping of adsorbed ethylene at potentials lower than 0.20 V in 1 M H2SO4 (0.25 V in 1 M HClO4) exhibits two residues, in the double layer and in the gold oxide region. On the other hand, the adsorption of acetylene took place in two potential regions, one coinciding with the ethylene adsorption domain, and the other involving potential values positive to that of zero charge.

BACKGROUND: Allergic contact dermatitis caused by species of Lithraea genus (Anacardiaceae) is frequent in South America. Nevertheless, it has been scarcely reported in the literature, hitherto the responsible allergens have not been studied in some species. OBJECTIVE: The purpose of this study was to identify and characterize the allergenic compounds of Lithraea molleoides and brasiliensis, and to investigate the existence of cross-reactions with Toxicodendron allergens. METHODS: Twenty-seven South American subjects (17 with previous Lithraea dermatitis and 10 controls without any plant dermatitis) and four North American subjects who are highly sensitive to poison oak were tested with both purified Lithraea molleoides and brasiliensis extracts and poison oak urushiol. Lithraea extracts were analyzed by gas liquid chromatography (GLC) and gas chromatography-mass spectrometry (GC-MS). RESULTS: All 17 Lithraea-sensitive subjects reacted to poison oak urushiol and 13 of them also reacted to Lithraea molleoides and/or brasiliensis extracts. All 4 poison oak sensitive subjects reacted to poison oak urushiol and to Lithraea extracts. In both groups of sensitive subjects, the responses to poison oak urushiol were stronger and occurred at lower concentration than those to Lithraea extracts. The allergenic fraction in both Lithraea species consisted of: 3-pentadecylcatechol, 3-pentadecenylcatechol, 3-heptadecenylcatechol and 3-hepta-dec-dienilcatechol. CONCLUSION: We concluded that Lithraea molleoides and brasiliensis allergens are closely related to urushiol, although their eliciting potential seems to be lower in comparison with poison oak urushiol, even for Lithraea-sensitive subjects.

A summary of the resource.
The invasive stages of the parasitic trematode Fasciola hepatica release proteinases into the medium in which they are mantained. In this study, we investigated the interaction of F. hepatica excretory/secretory (E/S) products and 2 cysteine proteinases (CL1 and CL2) purified from these products with extracellular matrix and basement membrane macromolecules. Fasciola hepatica E/S products contained collagenolytic activity on fibrillar types I and III collagen as well as basement membrane type IV collagen. CL1 and CL2 were capable of degrading acid-soluble type III and type IV collagen but not insoluble type I collagen. In contrast, neither the E/S products nor the purified CL1 and CL2 showed elastinolytic activity. Fibronectin and laminin were degraded by E/S products and by CL1 and CL2. Sequence analysis of fibronectin degradation products showed that the fragments obtained corresponded to complete biologically active domains. These results indicate that the cysteine proteinases secreted by F. hepatica may be involved in the process of tissue invasion by the parasite.

A summary of the resource.
The present work describes the preparation and characterization of iscoms from tegumental antigens of Echinococcus granulosus protoscoleces, and their use as immunogens in mice by intranasal and subcutaneous routes. Iscoms given at 10 mg doses evoke a significant antibody response when administered i.n. and a strong and long lasting antibody response by s.c. route. The Ag administered i.n. in a non-adjuvanted form as a booster was not immunogenic. The i.n. route of immunization induced higher IgA serum titre in relation to IgG than the s.c. route and antisera of slightly higher avidity. Also, ten fold lower IgG2a/IgG1 and ten fold higher IgG3/IgG1 ratios were observed for the i.n. protocol compared to the s.c. one. Moreover, the mucosal route of immunization induced more efficiently antibodies of both isotypes directed to carbohydrate epitopes. The differences between immune response generated by i.n. and parenteral immunizations should be taken into consideration, particularly in those cases in which protective antigens are glycosylated.

A summary of the resource.
Although the traditional wine industry of Uruguay is virtually unknown in the international wine market, wine sales are now increasing in some of the most important markets of Europe and North America. Because of strong domestic consumption and a quiescent R&D situation in recent decades, the pioneer vision of high quality that was bom in the 1880s was not realised. The strategy of promoting the typical grape Tannat (Harriague), best known for making high‐quality Madiran reds in southwest France, is starting to result in successful competitive reds which are only found in Uruguay. This paper presents a summary of the history of the Uruguayan wine industry, beginning with the first Tannat vines planted in the 1870s by the Frenchman Harriague, who with a British family developed the most technically advanced winery of those days, including a bacteriological laboratory. With the programme of training for winemakers and the recently started R&D projects in the University of the Republic, it is hoped that the Uruguayan wine industry, based on quality wines at a good price, will continue opening markets and reach its target of 300 000 cases by the year 2000. The country could occupy an interesting position within the quality wines of the Mew World in the near future.

A summary of the resource.
In order to prepare the imine derived from condensation of glycine ethyl ester and 5-nitrothiophene-2-carboxaldehyde, another compound was obtained instead. This product was the enamine, ethyl 2-amino-3-(5-nitro-2-thienyl)-2-propenoate, resulting from the condensation between the methylene of the glycine derivative and the heterocyclic carbonyl group. The reactivity of related amines and heterocyclic aldehydes was studied in order to get more insight about the mechanism of enamine formation. It was found that the nitro-heterocyclic group and the easy of enolization of the aminoacidic ester played an important role.

Fatty acid-binding proteins (FABPs) are candidate molecules for vaccines against several parasitic platyhelminths. A FABP from the cestode Echinococcus granulosus (EgDf1) was expressed in Salmonella vaccine strains as a C-terminal fusion to fragment C of tetanus toxin (TetC) by using expression vector pTECH. The fusion protein was equally expressed in several attenuated vaccine strains derived from bacteria with different genetic backgrounds and different attenuating mutations. Single-dose immunization experiments with the aroA Salmonella typhimurium strain SL3261 carrying the pTECH-EgDf1 construct were conducted with mice, using both the intravenous and the oral routes. Surprisingly, the antibody response to EgDf1 and the antigen-specific cytokine production in spleen cells were stronger in mice immunized orally. Furthermore, immune mouse sera strongly reacted with fixed sections of the worm's larval stage. Analysis of the isotype distribution of the specific anti-EgDf1 antibodies showed similar production of immunoglobulin G1 (IgG1) and IgG2a together with specific IgA antibodies. In addition, stimulation of spleen cells from mice immunized with the different constructs with either Salmonella lysate, TetC, or EgDf1 showed that, together with Th1-related cytokines (gamma interferon and interleukin 2 [IL-2]), significant levels of a Th2 cytokine (IL-5) were produced specifically, indicating a Th2 component to the response to the Salmonella carrier and to the recombinant antigens. Salmonellae expressing the TetC-rEgDfl fusion are currently under evaluation as potential vaccines against E. granulosus.

Les inhibiteurs de la calcineurine (ICN) sont les immunosuppresseurs les plus employés en transplantation d’organe et, malgré leur toxicité et leur efficacité imparfaite, ils resteront les premières options thérapeutiques dans un avenir proche. Leurs effets présentent une large variabilité intra- et inter-individuelle, qui n’est pas expliquée par les différences de doses, de concentrations ou d’aires sous la courbe des concentrations en fonction du temps, ce qui limite les bénéfices du suivi thérapeutique pharmacologique et suggère que d’autres facteurs contribuent à la variabilité de la réponse. Comme les ICN ciblent la fonction des lymphocytes T (LT), la concentration des ICN dans les LT a été étudiée comme marqueur intermédiaire de la fonction immunitaire. Effectivement, les concentrations intra-lymphocytaires de TAC chez des transplantés hépatiques montraient une plus forte association avec le rejet aigu que les concentrations dans le sang total, probablement parce qu’elles intègrent la variabilité inter-individuelle d’entrée du médicament dans les LT. De la même manière, des études antérieures ont montré que les concentrations intra-hépatiques de TAC étaient très bien corrélées avec les scores de rejet de Banff, alors que les concentrations résiduelles dans le sang ne l’étaient pas. Les deux pourraient représenter des biomarqueurs pharmacocinétiques intéressants, quoique difficiles à mesurer. Pour se qualifier comme index d’activité immunosuppressive des ICN, un biomarqueur pharmacodynamique (PD) devrait être directement affecté par l’activité du médicament, c'est-à-dire ne pas être trop éloigné de la cible du médicament et en même temps proche de ses effets cliniques. Comme de nombreuses preuves accréditent le concept que la suppression des LT est le mécanisme clé par lequel les ICN induisent une immunosuppression permettant de prévenir le rejet cellulaire, la fonction et l’activation des LT sont des candidats séduisants pour les stratégies de suivi PD. Toutefois, à ce jour aucun biomarqueur PD ne présente tous les prérequis idéaux, c'est-à-dire est à la fois non-invasif, fiable, sensible, spécifique, reproductible et disponible rapidement. Afin d’identifier des biomarqueurs PD très spécifiques de l’inhibition de la calcineurine et reflétant une part importante de la variabilité inter-individuelle, nos travaux avaient pour objectifs d’explorer la pharmacodynamie des ICN, la force et la variabilité de la Ofelia Noceti Penza | Thèse de doctorat | Université de Limoges | 2015 9 of 298 transmission du signal le long de l’axe calcineurine, ainsi que les étapes où les sources de variabilité PD internes (génétiques) ou externes sont les plus influentes. Dans ce but, nous avons mesuré simultanément la translocation de NFAT1 dans le noyaux des cellules mononuclées du sang périphérique (PBMC) (NFTA1 étant l’isoforme prédominante de NFAT dans les LT quiescents ou activés), l’expression intracellulaire d’IL-2 dans les sous-populations lymphocytaires CD3+, CD4+ et CD8+, et l’expression membranaire de CD25 (IL-2Rα), un marqueur de surface de l’activation des LT. Un essai clinique non-interventionnel a été mis en place chez des volontaires sains (VS), des patients inscrits en liste d’attente de transplantation hépatique (PLA) et des patients transplantés hépatiques (PTH). Une question différente a été étudiée dans chaque groupe : HS et PLA ont été inclus pour étudier la réponse des LT au tacrolimus (TAC) exvivo dans des conditions de stimulation, ainsi que les sources génétiques potentielles de variabilité PD, respectivement dans des conditions physiologiques et pathologiques. Les PTH ont été recrutés pour étudier les relations pharmacodynamiques, pharmacogénétiques et pharmacocinétiques dans des situations d’administration de tacrolimus ou de ciclosporine, c'est-à-dire d’activité PD résiduelle mesurée ex-vivo en conditions de stimulation ou non. Le petit groupe de PTH (n=6) inclus juste avant la transplantation et suivi de façon répétitive pendant la première année post-greffe était prévu pour explorer les relations entre PD des ICN et réponses cliniques. L’étude chez les VS (n=35) : a exploré la PD du TAC au long de la voie calcineurine en exposant des PBMC ex-vivo au médicament ; a modélisé la transmission du signal au long de cette voie ; a examiné la variabilité inter-individuelle des paramètres pharmacodynamiques du TAC ; et a étudié les sources de cette variabilité et leur contribution à chaque étape de la voie calcineurine. De plus, elle nous a permis d’évaluer la variabilité analytique de nos techniques ainsi que la variabilité intra-individuelle des paramètres PD du TAC. Les PLA (n = 19) nous ont permis de confirmer les résultats obtenus chez les VS, ainsi que de tester l’influence potentielle de leur maladie initiale sur la pharmacodynamie ex-vivo du TAC. Les buts de l’étude transversale chez des PTH (n=80) étaient d’explorer la variabilité inter-individuelle de la PD des ICN dans des situations cliniques réalistes et les sources pharmacogénétiques potentielles de cette variabilité. Ofelia Noceti Penza | Thèse de doctorat | Université de Limoges | 2015 10 of 298 Nous avons mesuré par cytométrie en flux la présence de NFTA1 transloqué dans le noyau des PBMC (qui est très proche de l’activité phosphatase de la calcineurine et probablement plus spécifique que la mesure « directe » de son activité, qui nécessite d’inhiber d’autres activités phosphatases), ainsi que la fonction (IL-2) et l’activation (CD25) des LT comme marqueurs du statut de l’immunité cellulaire. Les PBMC ont été séparés par gradient de centrifugation à partir d’échantillons de sang total recueillis avant l’administration du médicament et à jeun. Des aliquots frais de 106 PBMC dans 100 μl de RPMI supplémenté ont été employés pour toutes les analyses. Les PBMC des VS et des PLA ont été exposés ex-vivo pendant 30 min à des concentrations de TAC entre 0 et 50 ng/mL avant chaque mesure. Les mesures de NFAT1 étaient précédées d’une stimulation polyclonale par du phorbol 12-myristate 13-acétate (PMA) à 50 ng/mL et du calcium ionophore (I) à 2.5 μg/ml, à 37°C dans une atmosphère humidifiée avec 5% CO2 pendant 30 min, pour garantir la translocation de NFAT1 vers le noyau. Après deux lavages avec du PBS 1X froid, les cellules étaient incubées dans la glace pendant 30 min avec du tampon de lyse PIPE (pH 7,4 dans le PBS 1X) pour perméabiliser les noyaux cellulaires. Après deux nouveaux lavages avec de la BSA à 1% dans le PBS 1X, 50 μL d’un mélange d’anticorps anti-NFAT1 à 5μg/ml dans le PBS 1X étaient ajoutés au milieu pour 30 min d’incubation dans la glace. Puis des Ig polyclonales à 0,33% marquées au PE (fragment F(ab’)2 de chèvres anti- IgG(H+L) de souris) étaient ajoutées. Après 15 min d’incubation dans la glace à l’abri de la lumière, le signal fluorescent résultant des noyaux de PBMC marqués a été mesuré par cytométrie de flux et l’intensité de fluorescence moyenne de NFTA1 dans les noyaux isolés utilisée pour les analyses statistiques. La mesure d’IL-2 intra-cellulaire a nécessité 5h d’activation polyclonale avec du PMA/I et de la bréfeldine à 1 μg/ml (pour arrêter la sécrétion de protéines) à 37% en atmosphère humidifiée avec 5% de CO2. Les cellules étaient alors : i) marquées avec des marqueurs de surface anti-LT humain (CD3 PerCP-Cy5.5, CD4 FITC, CD8 PE-Cy7); ii) fixées : iii) perméabilisées (IntraPrep) ; iv) marquées avec des anticorps PE de rat anti-IL-2 humains et avec des IgG2a κ PE de rat comme contrôle isotypique. Pour la mesure d’expression de CD25, des aliquots de PBMC ont été incubés pendant 72h à 37°C et 5% de CO2 en présence de 7,5 μg/ml de Concanavaline A dans du RPMI 1640 1X. Ofelia Noceti Penza | Thèse de doctorat | Université de Limoges | 2015 11 of 298 Après deux lavages additionnels dans le PBS 1X, un marquage direct était réalisé avec : CD3 PerCP–Cy5.5, CD4 PE, CD8 PE-Cy7 et CD25 APC. L’analyse par cytométrie de flux était pratiquée après 30 min d’incubation dans le noir et deux lavages avec de la BSA dans du PBS 1X. De plus, pour les PLA et PTH, un aliquot de PBMC a été analysé sans stimulation ex-vivo (NS condition) pour NFAT1, IL-2 et CD25. L’expression d’IL-2 et CD25 a été exprimée en pourcentage de cellules fluorescentes sur le nombre total de cellules CD4+, CD8+ ou CD3+, respectivement. Reinvestigation of the phenolic constituents from a diethyl ether extract of gametophytes of <em>Pellia epiphylla</em> yielded pellepiphyllin, 7-hydroxypellepiphyllin, perrottetin E, perrottetin E-11-methyl ether, 14′-hydroxyperrottetin E, 10′- hydroxyperrottetin E, 10′-hydroxyperrottetin E-11-methyl ether, 10,10′-dihydroxyperrottetin E and 13′, 13‴-bis (10′-hydroxyperrottetin E), The structures of three bisbibenzyls published earlier are revised. The new structures have been elucidated by two-dimensional NMR-techniques and chemical synthesis. In contrast to previous publications, pellepiphyllin has been found as a constituent of <em>P. epiphylla</em>.

Two new chromenol derivatives, compounds (5) and (6) were isolated from the brown alga Desmarestia menziessi and their structures were elucidated by spectroscopic methods. The already described compounds desmarestial (1), sargadiol I (3), plastoquinone (2) and its hydroquinone form, were also isolated.

Abstract
A summary of the resource.
Alkylisocyanide complexes of copper(I) with formula [Cu(CNR)4]BF4 [where R = 2-methoxyisobutyl and 2-(carbomethoxy)-2-methylethyl] were synthesized by direct reaction of cuprous chloride with ligand. The stability of solid complexes and their aqueous solutions were tested by HPLC. Comparative studies were carried out on electronic, IR and 1H NMR spectroscopy. Crystal structures of [Cu(MIBI)4]BF4 and [Cu(CPI)4]BF4 were determined from X-ray data and show a tetrahedral arrangement of ligands around the central copper atom in both compounds.

Abstract
A summary of the resource.
Blepharocalyx tweediei leaf oil, obtained by hydrodistillation, was analyzed by GC and GC/MS. Fifty four components were identified in the oil. The main components were: 1,8-cineole (55.7%), caryophyllene oxide (4.1%) and α-terpineol (3.9%). Eighteen sesquiterpene hydrocarbons were identified in the sesquiterpene fraction of the oil.

Abstract
A summary of the resource.
The analysis of industrial and laboratory-prepared bergamot oils produced in Uruguay during the 1995 production season was carried out by GC and GC/MS. The composition of the Uruguayan bergamot oils were compared with those of Italian bergamot oils. They were found to be very similar; however, they could be differentiated from their psoralen and coumarin contents. The enantiomeric ratio of linalool and linalyl acetate was also studied by GC using a β-cyclodextrin column. The results were similar to that found in Italian oils.

Abstract
A summary of the resource.
The composition of 31 genuine Uruguayan lemon oils obtained by industrial processing (FMC on line) during the 1995 season is reported. The samples were representative of all production areas situated in the North and South of Uruguay. The volatile fraction was analysed by HRGC–FID and GC–MS; the enantiomeric distribution of β-pinene, sabinene, limonene, linalol, terpinen-4-ol and α-terpineol was studied by multidimensional HRGC–HRGC; coumarins and psoralens present in the non-volatile residue were analysed by normal phase HPLC. The results relative to the volatile fraction were compared with those obtained for Uruguayan oils produced in the 1992 and 1993 seasons and for Italian FMC oils. Coumarins and psoralens have been analysed for the first time in Uruguayan oils; their content is compared with that of Italian oils.

Hyptis floribunda leaf oil, obtained by hydrodistillation, was analyzed by GC and GC/MS. Forty-three components were identified in the oil. The main components were germacrene B (40.2%), β-caryophyllene (9.9%), germacrene D (9.8%) and α-copaene (8.0%).

A new, efficient and economic method employing Medium Pressure Liquid Chromatography (MPLC) for the isolation of the two majorSolanum tuberosum L. glycoalkaloids (α-solanine and α-chaconine) is described. Potato peelings are homogenised with 5% acetic acid, the glycoalkaloids purified by filtration through an XAD-2 column and then by precipitation from the aqueous solution. The resulting glycoalkaloid fraction was purified by MPLC using a Silica Gel column and a CHCl3:MeOH:2% NH4OH mixture (70∶30∶5) as mobile phase to yield pure α-chaconine and a-solanine. This methodology can be used to obtain glycoalkaloids for enthomology and toxicological research where large amounts of these compounds are required.

Hydatid disease, which affects humans and various livestock species, is caused by the larval stages of the cestode parasite Echinococcus granulosus. The disease involves the growth of a fluid-filled cyst in the host's internal organs. The hydatid cyst wall is generally thought to preclude the access of host immune cells to the cyst, while allowing passage of macromolecules. Thus, evasion of humoral effector mechanisms, in particular the complement system, may be crucial to parasite survival. Evasion of complement might involve inhibition of the C3b deposition step, which can in principle be achieved through parasite-derived or host-derived and parasite-bound regulatory molecules. In this work, extracts from bovine hydatid cysts were analyzed for the presence of factor H-like function, namely the cofactor activity for the breakdown of amidated bovine C3 by bovine factor I. Both this activity and antigenically detectable bovine factor H were found in most extracts. The cofactor activity was retained by an anti-bovine factor H Ab column; as no factor H-cross-reactive parasite molecules were detected in the active extracts, the activity was indeed due to host factor H. The pattern of extraction of host factor H from the parasite tissue indicated that at least part of it was selectively bound to the cyst wall, and that a strong charge component was involved in the binding. Our results indicate that the selective concentration of host factor H from plasma/interstitial fluid may contribute to complement evasion by E. granulosus.

Four fatty acid binding proteins (FABPs) have been described in 4 parasitic platyhelminths: Schistosoma mansoni, Schistosoma japonicum, Fasciola hepatica and Echinococcus granulosus. FABPs form a multigenic family of cytosolic proteins widely distributed in metazoan tissues, the function of which is still poorly understood. These helminth proteins have recently received attention, since there are reports to indicate that S. mansoni and F. hepatica FABPs may be protective antigens. In addition, these proteins could play a major role in the parasites' life-cycles because platyhelminths are unable to synthesize de novo most of their lipids. We have undertaken phylogenetic and structural analyses of platyhelminth FABPs in an attempt to characterize features of biological relevance. Phylogenetically, these FABPs appear to be more closely related to those of vertebrate heart, mammary gland, muscle, retina, skin, brain and myelin, although no clear functional relationships were established between them. We describe several conserved motifs characteristic of specific groups of FABPs. Hydrophilicity, flexibility and accessibility analyses revealed several major putative epitopes for the E. granulosus FABP, EgDf1, that appear to be centred in loops of the EgDf1 3-dimensional structure modelled by molecular replacement.

Abstract
A summary of the resource.
The Ilex paraguariensis St. Hil. (yerba mate) saponins effectively inhibit the passive diffusion of cholic acid through dialysis membranes. The formation of large macromolecular mixed micelles between cholic acid and yerba mate saponins could explain this effect. The observed inhibition of the passive diffusion of cholic acid used as a model of bile acids, could have an antihypercholesterolaemic effect in vivo due to an increased sterols excretion.
