<p>Antibiotic resistance is a major threat to global public health. β-lactamases, which catalyze breakdown of β-lactam antibiotics, are a principal cause. Metallo β-lactamases (MBLs) represent a particular challenge because they hydrolyze almost all β-lactams and to date no MBL inhibitor has been approved for clinical use. Molecular simulations can aid drug discovery, for example, predicting inhibitor complexes, but empirical<br />molecular mechanics (MM) methods often perform poorly for metalloproteins. Here we present a multiscale approach to model thiol inhibitor binding to IMP-1, a clinically important MBL containing two catalytic zinc ions, and predict the binding<br />mode of a 2-mercaptomethyl thiazolidine (MMTZ) inhibitor. Inhibitors were first docked into the IMP-1 active site, testing different docking programs and scoring functions on multiple crystal structures. Complexes were then subjected to molecular<br />dynamics (MD) simulations and subsequently refined through QM/MM optimization with a density functional theory (DFT) method, B3LYP/6-31G(d), increasing the accuracy of the method with successive steps. This workflow was tested on two IMP-1: MMTZ complexes, for which it reproduced crystallographically observed binding, and applied to predict the binding mode of a third MMTZ inhibitor for which a complex<br />structure was crystallographically intractable. We also tested a 12-6-4 nonbonded interaction model in MD simulations and optimization with a SCC-DFTB QM/MM approach. The results show the limitations of empirical models for treating these systems and indicate the need for higher level calculations, for example, DFT/MM, for reliable structural predictions. This study demonstrates a reliable computational pipeline that can be applied to inhibitor design for MBLs and other zinc-metalloenzyme systems.</p>

A summary of the resource.
Los glicosidos han despertado un gran iDteres debido a su participacion en multiples procesos biologicos. EI uso de glicosidasas constituye tma alternativa atractiva a su compleja sintesis quimica, permitiendo la formaci6n de enlaces glicosidicos en un solo paso de reaccion y con un control completo de la configuracion del centro anomerico. Como la sintesis de giic6sidos frecuentemente requiere el uso de solventes organicos, las glicosidadas a see utilizadas deben ser estables en dichas condiciones. Una de las estrategias mas utilizadas para la estabilizaci6n de enzimas es su inmovilizacion. Esto a su vez facilita la separacion de la enzima del medio de reaccion, propiedad esencial en procesos de sintesis cineticarnente controlados. Se estudiaron dos estrategias de inmovilizacion de f3-galactosidasas. La primera a traves de la union de grupos amino de la enzima con grupos aldehido del soporte, y la segunda mediante la formaci6n de puentes disulfuro entre grupos tiol expuestos de la enzima y estructuras tiol-reactivas del soporte. La inmovilizacion sobre geles glutaraldehido genero derivados enzimciticos con rendimientos de inmovilizaci6n superiores al 60%, buenas actividades especificas y termicarnente mas estables que la enzima soluble. Se logro la estabilizacion de la f3-galactosidasa de K. lactis en presencia de acetona y DMF (18% v/v). La estabilidad del derivado de f3-galactosidasa de A. oryzae frente al efecto de los cosolventes, aunque no logro mejorar la correspondiente estabilidad de la enzima soluble, fue superior a la del derivado de K factis, Esto 10 biro potencialmente aplicable a la sintesis enzimatica en rnedio orgcinico. Las propiedades de los derivados obtenidos por inmovilización de la enzima sobre soportes tiolS\llfinato-agarosa fueeon difeeentes, dependiendo del origen de la enzima y de la modificación quimica realizada previa a la inmovilizacion. Se obtuvieron derivados con buenas actividades especificas y rendimientos de inmovilizacion superiores al 60 %, pero solo se logro una mayor estabilidad en presencia de cosolventes orgcinicos para el caso del derivado de la f3-galactosidasa de K. factio'S obtenido con la enzima reducida. Se estudio la sintesis de distintos galactósidos a partir de 0-nitrofellil-I3-D-galactopiran6sido (ONPG) y lactosa como dadores del grupo galactosilo y distintos aceptores (4-metilumbeliferona, xilosa, etilenglicol). Se obtuvo 4-metilumbiliferil-f3-D-galactopiranosido con un rendimiento muy bajo. Es destacable la sintesis de galactosil-xilosa con un rendirniento de un 16% para la enzirna soluble, el que aurnent6 a un 28% cuando se utilizó la enizrna inmovilizada, obteniendose una rnezcla de regiois6rneros (f3-D-galactopiranosil (1-1) D-xilopiranosa y f3-D-galactopiranosil (1-4) D-xilopiranosa 0 f3galactopiranosil (1-5) D-xilofuranosa). A su vez resulto interesante la produccion de galactosilletilenglicol con rendiIlljentos de hasta un 70% y 100% de transglicosilacion.

Abstract
A summary of the resource.
This study investigated the effect of hunger on consumers’ visual attention during a food choice task, and the role of time orientation (i.e., present and future orientation) in this interplay. A lab-based eye-tracking experiment including 102 participants was conducted, with hunger as the manipulated factor (hungry, satiated). Participants in the satiated condition were served a breakfast buffet before the experimental tasks, whereas participants in the hungry condition were served the buffet after completion of the tasks. Both groups were exposed to a set of planograms depicting supermarket shelves and were asked to choose an option they could consider buying, while their eye movements were recorded. Stimuli included non-food items as well as the key category of interest, Swedish crisp bread. After completion of the eye-tracking recordings, participants indicated their time orientation as well as their height and weight, which were used to calculate body mass index (BMI). The results revealed that hunger (vs. satiation) increased participants’ present orientation and visual attention towards bread, but decreased their future orientation, with participants’ present orientation mediating the effect of hunger on visual attention. Additional exploratory analyses revealed a positive correlation between participants’ BMI and their present orientation. Taken together, the results offer several fruitful avenues for future research, which may be used to promote public health. Moreover, the findings contribute to the literature by documenting that hungry individuals do not necessarily make more rapid decisions in drive-relevant domains; rather, they may actually devote a larger share of their attentional resources in the food domain, given their desire to acquire food

A summary of the resource.
Con el objetivo de desarrollar potenciales radiofarmacos de 99mTc para imagenes de zonas de hipoxia, se han preparado varios complejos conteniendo el farmac6foro paranitroaromatico. Dicho farmac6foro en condiciones de hipoxia es capaz de sufrir una reducci6n irreversible, transformandose en grupos citot6xicos como el aminobencilo y aminobenceno, que quedarian atrapados dentro de la celula. Como sistema de uni6n del farmac6foro al centro metalico se emplearon los complejos mixtos "3+1" de Tc(V) y los complejos tricarbonilicos de Tc(I). Los complejos mixtos "3+1" fueron preparados empleando cantidades equimoleculares de un ligando tridentado aminoditi61ico y un coligando ti61ico monodentado conteniendo el farmacoforo nitroaromatico. La estructura de estos complejos es del tipo TcO-[(S-(CH2Hs)-NR-(C2Hs)-S + S-R]. EI estudio estructural de los complejos mixtos, realizado empleando los complejos analogos de Re estable, verific6 que en todos los casos se formaron las estructuras esperadas. Los estudios de voltamperometria indicaron que la reducci6n del grupo nitro y el core monooxo ocurre a potenciales que se pueden alcanzar en medios biol6gicos. EI ensayo de citotoxicidad revel6 la importancia de la presencia del grupo dialquilamina en la estructura del ligando tridentado para la citotoxicidad del complejo final. Los complejos de 99mTc fueron preparados por sustituci6n empleando el 99mTc-glucoheptonato como precursor, obteniendo en todos los casos una pureza radioquimica superior al 90%, con una estabilidad de por 10 menos 6 hs. Los estudios de biodistribuci6n en ratones CD1 normales, realizados entre 5 min. y 24 horas post-inyecci6n, mostraron el perfil esperado para compuestos lipofilicos, incluyendo eliminaci6n mayorotaria por via hepatobiliar. La potencialidad para imagenes oncol6gicas fue evaluada en ratones portadores de sarcomas inducidos entre 30 min. y 24 horas post-adm. La captaci6n fue moderada (1.2 %/g a los 30 min.), pero la relaci6n tumor/musculo (TIM) alcanz6 valores entre 1.5 y 2 en el periodo estudiado. Imagenes centellograticas muestran similares resultados. Los complejos tricarbonilicos de 99mTc fueron obtenidos mediante sustituci6n a partir del precursor fac-99mTc-[(COh(H20hr, preparado por reducci6n del pertecneciato en atm6sfera de CO. Para esta etapa se emplearon dos ligandos, uno de los cuales actuaria como tridentado a traves de un sistema donor del tipo NNO EL segundo ligando actuaria como bidentado dejando una posici6n de coordinaci6n libre, este ultimo tam bien se va a emplear conjuntamente con un coligando monodentado adecuado como el CN, para formar un complejo mixto "2+1" tricarbonilico. Para el caso del ligando tridentado se obtuvo una especie con una pureza radioquimica superior al 90%, la que result6 estable por 10 menos 6 hs. Los estudios quimicos permitieron corroborar la formaci6n de la estructura propuesta. Los estudios "in vitro" revelaron que el complejo presenta muy baja citotoxicidad. La voltamperometria muestra que la reducci6n del grupo nitro ocurre a potenciales alcanzables en los medios biol6gicos. Los estudios de biodistribuci6n en animales normales dieron los resultados esperados para un complejo de lipofilicidad intermedia, presentado una eliminaci6n por via hepatobiliar y urinaria y una rapida depuraci6n de sangre y tejidos blandos. Los estudios en animales portadores de tumores revelaron una muy baja captaci6n (0,2%/g a los 30 min.), pero una relaci6n TIM favorable debido a la rapida eliminaci6n. EI complejo tricarbonilico con el ligando bidentado di6 lugar a la formaci6n de varias especies, las cuales pudieron ser restringidas mediante el agregado de un coligando monodentado adecuado como el CN. Los estudios estructurales con Re estable no permitieron la elucidaci6n de la estructura formada pero los datos obtenidos indican la ausencia del grupo nitro en los complejos finales. Los complejos obtenidos no presentaron propiedades 6ptimas para ser utilizados como radiofarmacos de diagn6stico de hipoxia, una de las posibles formas de continuaci6n de este trabajo seria la de realizar la uni6n del mismo farmac6foro al metal a traves de otros sistemas de marcaci6n. Pero dado los resultados similares obtenidos para las estrategias empleadas parece mas adecuado intentar un cambio de farmac6foro para aumentar la selectividad.

BACKGROUND: The differential characteristic of sparkling wine is the formation of foam, which is dependent, among other factors, on yeast autolysis, aging and oenological practices. In this study, we analyzed the effects of yeast strain, nutrient supplementation to the base wine and aging process on the sparkling wine composition and its foamability. RESULTS: We determined that the addition of inorganic nitrogen promoted nitrogen liberation to the extracellular medium, while the addition of inactive dry yeast to the base wine caused an increase in the polysaccharide concentration and foaming properties of the sparkling wine. The use of synthetic and natural base wines allowed us to discriminate that the differences in high-molecular-weight polysaccharides and oligosaccharides could be attributed to the yeast cells and that the higher nitrogen content in the natural wine could be due to external proteolysis. CONCLUSION: The practices of nitrogen addition and supplementation of inactive dry yeast could modulate the main characteristics of the sparkling wine and be a critical element for the design of this kind of wine.

A pesar de la indiscutible conveniencia de la utilización del biodiesel como combustible alternativo, la viabilidad de la implementación de este producto depende en gran medida de la posibilidad de elaborarlo a un costo competitivo con el diesel de petróleo. El sebo vacuno es una materia prima de bajo costo que puede permitir la producción de un biodiesel económicamente competitivo con el diesel de petróleo, principal inconveniente cuando el biodiesel se produce con aceites vegetales. Esta ventaja económica del sebo vacuno contrasta con la desventaja inherente a la elevada dureza del material y a los problemas de fluidez que dicho biodiesel presenta a las temperatura de invierno del Uruguay. En el presente trabajo se estudió el contenido de sólidos de los ésteres obtenidos a partir del sebo vacuno y diferentes alcoholes (metanol, etanol, 1-propanol, 2-propanol, 1-butanol y 2-butanol), a distintas temperaturas de interés para el clima uruguayo, los que constituyen un tipo de biodiesel. También se estudiaron mezclas de estos ésteres con ésteres obtenidos a partir de aceite de soja. El menor grado de linealidad de las moléculas de monoalquil ésteres obtenidas a partir de alcoholes de alto peso molecular y de alcoholes secundarios permite preparar un biodiesel a partir de sebo vacuno con características de fluidez similares a las de un biodiesel convencional.

A summary of the resource.
Lead bromide polycrystalline films were grown by the physical vapor deposition method (PVD). Glass 1″x1″ in size, uncoated, and coated with Indium Tin Oxide (ITO), was used as substrate and rear contact. The starting material was evaporated at temperatures from 395°C to 530°C under high vacuum atmosphere (6 x 10-3 Pa) and during 8 days. The substrate temperature was prefixed from 190°C to 220°C. Film thickness yielded values from 40 to 90 μm. Optical microscopy and scanning electron microscopy (SEM) were performed on the films. Grain size resulted to be from 1.0 to 3.5 μm. SEM and X-ray diffraction indicate that films grow with a preferred orientation with the (0 0 l) planes parallel to the substrate. The Texture Coefficient (TC) related to the plane (0 0 6) was 7.3. Resistivity values in the order of 1012 Ωcm were obtained for the oriented samples, but a strong polarization indicates severe charge transport problems in the films. Film properties were correlated with the growth temperature and with previous results for films of other halides.

La variabilidad interindividual en la respuesta y toxicidad a tratamientos es un problema muy común y serio, que causa preocupación y puede ser responsable de la reducción de la dosis o discontinuación del tratamiento. La Farmacogenética busca predecir dicha variabilidad, mediante el análisis y uso de la información genética individual o poblacional. El metotrexate(MTX), análogo estructural del ácido fólico, ha sido un fármaco altamente efectivo en tratamientos oncológicos y autoinmunes. Sin embargo presenta una gran variabilidad interindividual en su respuesta farmacológica y toxicidad. Una de las causas de dicha variabilidad es la existencia de polimorfismos en los genes que codifican proteínas transportadoras y dianas del MTX. El objetivo general de este trabajo fue contribuir al conocimiento para un diseño de una terapia personalizada con MTX. Como forma de realizar una aproximación a una vía tan compleja, desde el ingreso del MTX hasta su blanco de acción en la célula, nos propusimos evaluar la distribución y analizar la asociación entre la toxicidad y eficacia del tratamiento y los polimorfismos en los genes que codifican para los transportadores: de folato reducido (SLC19A1) G80A y de solutos de aniones orgánicos de la familia 1B1 (SLCO1B1) T521C; así como también en los genes que codifican para las enzimas involucradas en la actividad del MTX: 5,10- metilentetrahidrofolato reductasa (MTHFR) C677T y A1298C, variación en el número de copias de 28 pb en el gen timidilato sintasa (TYMS) y dihidrofolato reductasa (DHFR) C- 1610G/T, C-680A, A-317G y el polimorfismo que consiste en una inserción deleción de 19 pb. El grupo de estudio consistió en 41 pacientes adultos uruguayos con leucemia linfoide aguda (LLA) y linfoma no-Hodgkin (LNH) agresivo tratados con un protocolo de poliquimioterapia que incluyera mediana y/o altas dosis de MTX y en un grupo control de 55 adultos uruguayos voluntarios sanos. De dichos pacientes se relevaron los parámetros clínicos y de laboratorio obtenidos del seguimiento de la terapia.

Background Individual variability is among the causes of toxicity and interruption of treatment in acute lymphoblastic leukemia (ALL) and severe non-Hodgkin lymphoma (NHL) patients under protocols including Methotrexate (MTX): 2,4-diamino-N10-methyl propyl-glutamic acid. Methods 41 Uruguayan patients were recruited. Gene polymorphisms involved in MTX pathway were analyzed and their association with treatment toxicities and outcome was evaluated. Results Genotype distribution and allele frequency were determined for SLC19A1 G80A, MTHFR C677T and A1298C, TYMS 28 bp copy number variation, SLCO1B1 T521C, DHFR C− 1610G/T, DHFR C-680A, DHFR A-317G and DHFR 19 bp indel. Multivariate analysis showed that DHFR-1610G/T (OR = 0.107, p = 0.018) and MTHFR677T alleles (OR = 0.12, p = 0.026) had a strong protective effect against hematologic toxicity, while DHFR-1610CC genotype increased this toxicity (OR = 9, p = 0.045). No more associations were found. Conclusions The associations found between gene polymorphisms and toxicities in this small cohort are encouraging for a more extensive research to gain a better dose individualization in adult ALL and NHL patients. Besides, genotype distribution showed to be different from other populations, reinforcing the idea that genotype data from other populations should not be extrapolated to ours.

Methotrexate (MTX), a structural analog of folic acid, is widely employed in the treatment of different cancers and autoimmune diseases. Despite the successful results observed, the main disadvantage lies in interpatient variability in the pharmacokinetic and pharmacodynamic parameters. In particular, adverse events and toxicities induced by MTX are a matter of concern and can be the cause of dose reduction or treatment discontinuation. Among the different approaches to reduce MTX therapeutic limitations, pharmacogenomics contributes by considering the effect of inherited genetic differences on those parameters. This review provides an update on MTX pharmacogenomics. It reports the contribution of main gene polymorphisms involved in the influx, efflux, cellular effect, and elimination on MTX toxicity and efficacy, on all the diseases treated with this drug. From the analysis of the data presented in this review, we concluded that only gene polymorphisms MTHFR rs1801133, SLC19A1 rs1051266, and TYMS rs34743033 could influence clinical decision-making.

<p>Considering the importance of texture in consumers’ acceptance of many dairy products, particularly milk desserts, it is important to rely on methodologies to understand consumers’ texture perception.The aim of the presentwork was to study the relationship between consumers’ texture perception of milk desserts and trained assessors’ data. Six commercial milk desserts were evaluated by a panel of 100 consumers, divided into two groups of 50, which evaluated the texture of the desserts using intensity scales for five texture attributes or a check-all-that-apply question. Concurrently, seven texture attributes of the desserts were evaluated by a trained panel. Results showed that both consumer profiling methodologies were able to identify differences in the texture characteristics of the desserts, providing very similar sensory maps.High correlation between textural attributes evaluated by consumers and trained assessors was found, indicating that consumers were able to evaluate the attributes in a similar way as trained assessors did.</p>

Marketing of unhealthy products has been identified as one of the main characteristics of the food environment that negatively affects children's eating patterns. Restrictions on advertising of unhealthy foods to children have already been imposed in different countries. However, marketing strategies are not limited to broadcast and digital advertising, but also include package design. In this context, the current study aimed to describe the food products targeted at children and sold in supermarkets in Montevideo, Uruguay, in terms of package design and nutrient profile. Two supermarkets in Montevideo were selected for data collection. In each supermarket, all products targeted at children were identified. Products were analyzed in terms of package design and nutritional profile, considering the Pan American Health Organization Nutrient Profile Model. A total of 180 unique products were identified, which included a wide range of product categories. The great majority of the products corresponded to ultra-processed products with excessive amounts of sodium, free sugars, total fat, saturated fat, and/or trans fat, which are not recommended for frequent consumption. Several marketing strategies were identified in the design of packages to attract children's attention and drive their preferences. The most common strategies were the inclusion of cartoon characters, bright colors, childish lettering, and a wide range of claims related to health and nutrition, as well as the products' sensory and hedonic characteristics. The study's findings provide additional evidence on the need to regulate packaging of products targeted at children.

For most food products, shelf life is determined by changes in their sensory characteristics. A predetermined increase or decrease in the intensity of a sensory characteristic has frequently been used to signal that a product has reached the end of its shelf life. Considering all attributes change simultaneously, the concept of multivariate shelf life allows a single measurement of deterioration that takes into account all these sensory changes at a certain storage time. The aim of the present work was to apply survival analysis to estimate failure criteria in multivariate sensory shelf life testing using two case studies, hamburger buns and orange juice, by modelling the relationship between consumers' rejection of the product and the deterioration index estimated using PCA. In both studies, a panel of 13 trained assessors evaluated the samples using descriptive analysis whereas a panel of 100 consumers answered a “yes” or “no” question regarding intention to buy or consume the product. PC1 explained the great majority of the variance, indicating all sensory characteristics evolved similarly with storage time. Thus, PC1 could be regarded as index of sensory deterioration and a single failure criterion could be estimated through survival analysis for 25 and 50% consumers' rejection. The proposed approach based on multivariate shelf life testing may increase the accuracy of shelf life estimations.

Well-being is one of the ultimate goals people pursue in their life and, consequently, it could have the potential to contribute to a better understanding of consumer food choices. Sensory and consumer scientists are expected to play a key role in the development, validation, and use of methodologies to evaluate perception of well-being in a food-related context. However, research on the topic is still incipient. In this chapter, methodological approaches to measuring consumer-perceived well-being in a food-related context are presented and remaining challenges on the topic are discussed.

Sodium reduction strategies that fully or partially compensate for the effect of reducing sodium on the sensoy profile of foods have been proposed as a cost‐effective alternative to decrease dietary sodium intake at the population level. The present work aimed at exploring the effect of partially replacing NaCl with KCl on the sensory characteristics and hedonic perception of toast bread. Equivalent salty concentration of NaCl : KCl blends were estimated relative to different NaCl concentrations, using paired comparisons with a panel of trained assessors. The influence of partially replacing NaCl by KCl on the dynamic sensory profile of toast breads was assessed using the Temporal Check‐all‐that‐apply approach with a trained panel. Finally, a consumer study was carried out to evaluate consumers' sensory and hedonic perception of toast breads produced by partially replacing 30% NaCl with KCl. Results from the present work suggest that KCl can be deemed effective at compensating for saltiness intensity in sodium reduced toast breads. Partial replacement of 30% NaCl with KCl did not significantly affect the sensory characteristics of toast breads. However, NaCl replacement equal or higher than 40% caused an increase in bitterness, off‐flavor, and metallic flavor. Heterogeneity in consumer hedonic reaction toward partial replacement of NaCl with KCl was observed, but mainly associated to differences in texture perception.

Objective: The information displayed on the packages of feeding bottles and teats commercialized in Montevideo (Uruguay) was analyzed using content analysis with the goal of identifying key marketing practices that may discourage breastfeeding.<br />Design: The study was conducted as part of the periodic assessment performed by the Uruguayan government to monitor the marketing of breast-milk substitutes. All the feeding bottles and teats sold in 44 retail outlets selling breast-milk substitutes were purchased. The information available on the packages was analyzed using content analysis and descriptive statistics.<br />Setting: Montevideo, Uruguay.<br />Results: A total of 197 feeding bottles and 71 teats were found. The majority of the packages included information to enable caregivers to adequately use the products, including recommended age, instructions on how to use the products and instructions on the use of hygienic practices. However, the packages frequently included information that implied that bottle-feeding was equivalent to breastfeeding, particularly from a physiological perspective, or that idealized product use. Idealizations included ability to reduce colic, improvements in the feeding experience and improvements in children’s health, wellbeing and development. Statements on the superiority of breastfeeding were infrequent.<br />Conclusions: Results from the present work showed the high prevalence of marketing practices on the packages of feeding bottles and teats that may discourage breastfeeding. Stricter and more detailed regulations seem necessary to enable caregivers to make informed feeding decisions for infants.

Food donation has expanded globally and has become a common form of food assistance in emerging countries. However, research on food donation experiences in these settings is still scarce. Purpose In this context, the objectives of the present work were as follows: (i) to explore the views of Uruguayan stakehold ers on food donation as a strategy to reduce food waste, (ii) to identify barriers and enablers for food donation in Uruguay, and (iii) to explore the organizational characteristics of Uruguayan food banks. Methods A qualitative approach based on two data collection strategies was used: documentary content analysis of ofcial governmental documents and in-depth semi-structured interviews with key stakeholders related to food waste and food donations in Uruguay. Results Food donation emerged as a central strategy for food waste reduction in the view of Uruguayan stakeholders, mainly because it was perceived as an efcient strategy to reduce food insecurity. The complexity of food donation was acknowledged by Uruguayan stakeholders and who identifed several barriers for its implementation related to economic considerations, the characteristics of the donated products, legal and commercial considerations, and organizational characteristics of food banks. Conclusion Although most barriers were aligned with results from previous studies conducted in developed countries, some specifcities to the Uruguayan context were also identifed. Clear and detailed legal frameworks to ensure that the donated products reach the target population along with nutritional requirements for the foods to be donated were deemed necessary.
