Two digestive proteases from Parona signata (Palometa) were isolated from gastric and pyloric caeca tissues and characterized. The stomach enzyme was inhibited by pepstatin and was classified as pepsin. Palometa pepsin purified from a Sephadex G-100 column appeared as two bands on an SDS-PAGE gel and exhibited optimum activity at pH 3.5, and 37C. Palometa pepsin was inhibited by pepstatin to a similar exient as porcine pepsin. An enzyme isolated from the pyloric caeca was shown to be trypsin based on its molecular weight, its ability to hydrolyze the synthetic substrates, N-α-benzoyl-arginine-p-nitroanilide (BAPA), and tosyl-arginine methyl ester (TAME) and inhibition by the known trypsin inhibitors, SBTI, TLCK, PMSF and benzamidine. The purified trypsin from palometa pyloric caeca yielded a single band with a molecular mass of 24 kDa. Optima trypsin activity was obtained at pH 8.5 and the enzyme was stable over a pH range of 3 to 11. Palometa trypsin activity was stable for 30 min at 50C, but lost 50% of its activity after 30 min at 60C. The optimum temperature for activity was 65C. The biochemical properties of Palometa trypsin and pepsin were discussed in relation to the varying environment of Palometa in the Rio de la Plata estuary. A southwest Atlantic croaker protease was purified from the pyloric caeca by ammonium sulfate fractionation, acetone precipitation and affinity chromatography. Croaker trypsin activity was stable between pH 5 and pH 11 for 30 min at 0C, 10C and 25C and its maximal activity against BAPA was at pH 9.5. Thermostability was observed to about 55C for 30 min at pH 7.8 and its temperature optimum was 60C. Substrate turnover number was 850 BAPA units per μmol trypsin, and the Kmwas 0.081 mM at 25C. For the hydrolysis of TAME, Vmaxwas 9273 units per μmol trypsin and the Km was 0.155 mM at 25C. The catalytic efficiency Vmax/Kmwas higher than that of trypsin from fish living in colder waters. Fluorescence lifetime and rotational behaviour of 1,6-diphenyl-1,3-5-hexatriene (DPH) and 1-(4-trimethylammoniumphenyl)-6-phenyl-1,3,5 hexatriene (TMA-DPH) were used to study the influence of several monofunctional pentacyclic triterpenes on the structural and dynamic properties of dipalmitoylphosphatidylcholine (DPPC) bilayers. The plant monoalcohols α-amiryn, lupeol and taraxerol, the semisynthetic (H-18-α)-α-amiryn, and the ketone taraxerone were studied in comparison with cholesterol. Among them, only α-amiryn is incorporated at as high levels as cholesterol into DPPC bilayers. At 50 mol%, it results in the phase transition vanishing. Pentacyclic monoalcohols are more potent than cholesterol to decrease the DPPC gel phase order, but less potent than it to increase the liquid–crystalline state order. They also produce more packing defects than cholesterol in DPPC bilayers. These effects correlate with the absence of planarity in the pentacyclic skeleton, which would obstruct the `all trans' packing of acyl chains in the gel state and Van der Waals interactions in the liquid–crystalline state. Taraxerone has little effect on DPPC bilayers, probably due to its difficulty to form hydrogen bonds. The carabid beetle Galerita lecontei has a pair of abdominal defensive glands that secrete a mixture of formic acid, acetic acid, and lipophilic components (long-chain hydrocarbons and esters). Formic acid, at the concentration of 80%, is the principal constituent. The beetle ejects the secretion as a spray, which it aims accurately toward parts of the body subjected to assault. At full capacity, the glands store 4.5 mg of formic acid (3% of body mass), enough for upward of six ejections. The beetle reloads the glands at a rate of 126 μg of formic acid per day. For the approximately 500 secretory cells of the glands, this means an hourly output of 10 ng of formic acid per cell, or about 5% of cell volume. Replenishing empty glands to their full formic acid load takes the beetle an estimated 37 days. Replenishing the 0.7 mg of formic acid expended in a single discharge takes 5.5 days. Unknown phosphorated compounds, different from organophosphorus pesticides, were detected during GC analyses of citrus oils; the chromatographic profile was similar to a triarylphosphate mixture. Standard mixtures of triarylphosphates were analyzed and mass spectra were obtained; the results showed that the contamination was due to triphenylphosphate and tritolylphosphate isomers essentially. Blood samples and questionnaire background data were collected from 96 children (age 2–14 years) living in urban, suburban, or rural areas with varying traffic intensity and industrial lead pollution in Uruguay. Spot samples of tap water were collected from the homes of 44 children, and samples of top soil were taken from seven areas. Samples of air-borne dust were collected in central and suburban Montevideo. Blood lead concentrations (B-Pb) in children ranged between 47 and 191 (mean 96) μg/L and exceeded in 36% of the children 100 μg/L, the intervention level adopted by the United States Centers for Disease Control. Lead in tap water ranged from 0.2 to 230 (mean 15) μg/L and exceeded in 39% of the samples the maximum level recommended by WHO, 10 μg/L. Lead pipes were used in parts of the water supply systems. Lead in air varied between different locations from 0.15 to 1.7 μg/m3, highest in the very center of Montevideo. The median soil lead ranged from 6 to 2100 μg/g and was highest in industrially polluted areas. At multiple regression analysis, B-Pb was significantly associated only with age (P= 0.032) and traffic intensity at school (P= 0.045). No significant impact on B-Pb of lead in water or soil could be established. The antibody response was followed during 68 weeks in 17 Balb/c mice intraperitoneally (i.p.) infected with Echinococcus granulosus protoscoleces (PSC) and in three mice i.p. immunized with dead PSC. Titres of antibodies recognizing peptidic and glucidic PSC epitopes, as well as their isotypic and avidity profiles were followed by ELISA. In addition, antigen recognition patterns were analysed by immunoblot. The response against carbohydrate epitopes was dominant in infected and immunized mice but stronger in the first group. Infected mice showed similar profiles of specific IgG and IgM with maximum titres from week 38 to 53. Although IgG1 and IgG3 were the predominant antibody subclasses, the ratio of IgG1/IgG3 antibody titres as well as antibody avidity decreased during the experiment, encompassing a decrease in recognition of peptidic epitopes. Immunized mice did not show significant levels of specific IgM and, after week 15, showed IgG titres lower than the infected mice. IgG1 was the predominant IgG subclass during all the experiment with background levels of IgG3. The mean Ab avidity was high and showed no significant changes during immunization. Different patterns of response were thus produced by dead and developing live parasites. Although high avidity IgG1 antibodies were early found in both cases, lower avidity IgG3 antibodies were increasingly produced afterwards only in infected animals. The isotype switch and avidity decrease observed only during infection are consistent with a possible parasitic mechanism to evade host immunity. The effects of destruction of carbohydrate moieties of Echinococcus granulosus antigen by sodium periodate treatment, and/or blocking phosphorylcholine specific antibodies by the addition of free reagent, were investigated to improve sensitivity and specificity of enzyme-linked immunoabsorbent assay (ELISA) for diagnosis of hydatid disease. Sera were collected, in Uruguay and Sweden, from patients with confirmed hydatidosis, non-hydatid tapeworm infections, other disorders, and healthy donors. ELISA performed with sodium periodate or free phosphorylcholine increased sensitivity, but decreased specificity. Western blot analysis showed reduced recognition of the 38 kDa antigen following both treatments, and of the 15.5 kDa antigen after periodate treatment. Addition of free phosphorylcholine enhanced the recognition of two antigens, 26 and 50 kDa, which may represent new cross-reacting epitopes. A new moderately thermophilic proteolytic anaerobe, strain UT, was isolated from mesophilic granular methanogenic sludge. The cells were spore-forming, motile rods that were 0.4 pm wide and 2.4 to 4 pm long and stained gram negative. Electron micrographs of thin sections revealed the presence of an atypical gram-positive cell wall. Optimum growth occurred at 55°C and at pH values between 7.0 and 7.5, with a doubling time of 30 min. The DNA base ratio of guanine plus cytosine was 31 mol%. The bacterium fermented proteins mainly to acetate, hydrogen, formate, and branched-chain fatty acids. Several amino acids, including glutamate, aspartate, arginine, histidine, threonine, methionine, and branched-chain amino acids, were also utilized. Glutamate was degraded to acetate, formate, hydrogen, and alanine. In addition, the strain degraded carbohydrates, including glucose, fructose, mannose, cellobiose, and starch, to acetate, ethanol, formate, lactate, and hydrogen. The results of a 16s rRNA sequence analysis phylogenetically placed strain UT in the low-guanine-plus-cytosine-content subgroup of the gram-positive phylum. We propose to classify the described strain in the genus Caloramator as a new species, Caloramator proteoclasticus. The type strain of C. proteoclas- ticus, strain U, has been deposited in the Deutsche Sammlung von Mikroorganismen as strain DSM 10124. Five glycoalkaloids were identified from the aerial parts of wild Solanum commersonii Dun. ex Poir growing in Uruguay and their structures elucidated mainly by chemical and spectroscopic methods. In addition to the already known commersonine, demissine, tomatine and dehydrocommersonine, a new glycoalkaloid, identified as δ5-demissine, solanid -5-en-3-O- {β-D-Glcp-(1→2)-[β-D-Xylp-(1→3) ]β-D-Glcp-(1→4)-β-D-Galp } was found. Tomatine and dehydrocommersonine have not been reported earlier as components of this plant. The glycoalkaloid composition found in the wild plants studied differ significatively from those described earlier, and have some implications in taxonomy and its use in potato breeding. An analysis of static dipole polarizability of one atomic anion (F - ) and three diatomic molecules (H 2 , N 2 and HF) has been performed at the density functional theory (DFT) level. Dunning correlation-consistent basis sets, ranging from the smallest cc-pVDZ to the aug-cc-pV5Z were employed systematically for all the molecules. Additionally, special types of basis sets, derived specifically for the calculation of polarizabilities in an ab initio environment, were employed in some cases. Calculations of the polarizabilities were performed at the DFF level employing the BLYP, B3LYP and B3PW91 exchange-correlation potentials, and the accuracy of the results was judged against experimental data on the one hand, and the polarizabilities calculated at the Hartree-Fock (HF), MP2, MP4(SDTQ), QCISD and CCSD levels on the other. In all the cases studied, it was observed that the quality of the results followed the order (from worst to best) HF < MP2 < BLYP < B3LYP < B3PW91 MP4 QCISD CCSD, demonstrating that DFT (B3PW91 in particular) is a convergent alternative to highly correlated post-Hartree-Fock methods. High-level ab initio (MP2/6-311++G(2d,2p) geometry, Gaussian-2, MP4(SDTQ) and QCISD(T) binding energies) and density-functional (Becke3LYP/6-311++G(2df,2pd)) calculations have been performed on the charge-transfer complex between water and carbon dioxide. The complex appears to have two equivalent non-planar minima of Cs symmetry. Minima are separated by transition states with C1 symmetry, whereas the totally planar structure with C2v symmetry is a second-order transition state. All the critical points lie at approximately the same energy (less than 0.05 Kj mol−1 difference). Therefore, the experimentally observable structure should be planar. The best equilibrium intermolecular distance for this complex calculated at the MP2/6-311++G(2d,2p) level is 2.800 Å. Our best estimate of the observable intermolecular distance (corrected for anharmonicity) is 2.84 Å, in agreement with the experimentally derived value of 2.836 Å. Our best estimate of the binding energy at the QCISD(T) level, taking into account the variation of the distance owing to anharmonicity and the use of more sophisticated theoretical treatments, is −12.0 ± 0.2 kJ mol−1. Our best estimate of the barrier to internal rotation, also at the MP2/6-311++G(2d,2p) level, is 4.0 kJ mol−1, outside the error limits of the experimental determination (3.64 ± 0.04 kJ mol−1). Density functional theory at the level employed here gives an equilibrium intermolecular distance that is too large (2.857 Å), a binding energy that is too small (8.1 kJ mol−1), attributable neither to geometry nor to the basis set, and also a barrier to internal rotation that is slightly too small (3.39 kJ mol−1). The overall picture is, however, reasonably good. A method termed "PERFEXT", based on sequential perfusion and detergent extraction of lymphoid and non-lymphoid organs, has been developed for the quantitative measurement of cytokines produced at a local level in a given tissue. In vivo treatment of mice with Staphylococcus enterotoxin B (SEB) or lipopolysaccharide (LPS) served as the model systems. Interleukin-2 (IL2) and interferon-gamma (IFN gamma) levels were monitored by ELISA analysis of extracted samples. After local footpad (FP) injection with SEB, spleen and serum IL2 levels peaked at 2-4 h, while IL2 levels peaked at around 4-8 h in both FP and popliteal lymph nodes. SEB injection resulted in increased IFN gamma levels both in the FP and the draining lymph node. The detection of cytokines in the intestine allows for the application of the method at mucosal sites as well, provided enzyme inhibitors are present during the extraction procedure. After FP injection with LPS, IFN gamma production was significantly increased in the draining lymph node and was detectable in the FP, whereas IL2 was undetectable in any organ examined. Uno de los problemas más importantes que afronta la humanidad es la contaminación de los recursos hídricos y la disponibilidad de agua potable para vastos sectores de la población. En muchos casos estos contaminantes tienen efectos nocivos sobre distintas formas de vida, constituyendo un riesgo ambiental y sanitario que se agudiza frente a la creciente demanda de agua de calidad. Por esta razón, el desarrollo de procesos que permitan la eliminación de contaminantes del agua resulta una contribución al desarrollo sustentable. Los métodos convencionales para el tratamiento de aguas resultan ineficientes en el caso de contaminantes recalcitrantes y/o de elevada toxicidad. Para este conjunto de contaminantes se requieren técnicas no convencionales más efectivas. Entre las técnicas no convencionales se encuentran los Procesos Avanzados de Oxidación (PAO), efectivos en la oxidación prácticamente total de un gran número de contaminantes orgánicos resistentes a los tratamientos convencionales. Entre los PAO más interesantes se encuentra el proceso foto-Fenton que consiste en la generación de radicales hidroxilo, de alto poder oxidante, mediante la acción combinada de hierro y peróxido de hidrógeno en una solución irradiada con luz adecuada. Este proceso presenta como desventaja la necesidad de remover el hierro en solución luego del tratamiento, lo que encarece el proceso. Esto puede evitarse utilizando sólidos catalíticamente activos en este tipo de procesos, lo que facilita la separación y recuperación del catalizador y posibilita su reutilización. Es así como en los últimos años se han desarrollado los denominados procesos foto-Fenton heterogéneos. En Uruguay existe un yacimiento de arcillas montmorilloníticas de alta pureza, ubicado en Bañado de Medina, Departamento de Cerro Largo. La presente Tesis está orientada a la obtención de catalizadores activos en procesos tipo foto-Fenton, a partir de esta arcilla montmorillonítica, con miras a valorizar un recurso natural nacional y v favorecer el desarrollo de un proceso que contribuya a resolver problemas de contaminación del agua. Se prepararon catalizadores incorporando hierro como especie activa en la arcilla mediante la técnica de pilareado, obteniendo arcillas pilareadas con hierro (Fe-PILCs). Se recurrió a diferentes técnicas para caracterizar la arcilla y los catalizadores. Se estudió la influencia de diferentes parámetros de preparación en las propiedades de los catalizadores y en su desempeño catalítico en el proceso foto-Fenton, aplicado a la degradación del colorante naranja de metilo como compuesto modelo. Se estudió la influencia del fenómeno de adsorción del compuesto a degradar sobre el catalizador y del pH del medio en la actividad catalítica. Para ello se utilizaron compuestos modelo con diferentes propiedades: azul de metileno, naranja de metilo y fenol. Además se ensayó la aplicación del proceso foto-Fenton heterogéneo empleando una de las Fe-PILCs, a la degradación de atrazina en medio acuoso. Este herbicida es uno de los contaminantes del agua producto de la actividad agropecuaria de nuestro país. La Tesis aborda también el estudio de la eficiencia en el uso de la radiación del proceso foto-Fenton heterogéneo con Fe-PILCs como catalizadores, mediante la determinación de la eficiencia aparente y la eficiencia cuántica del proceso. Para ello se evaluó la actividad catalítica en la degradación del contaminante 2-clorofenol, se cuantificó el flujo de radiación que ingresa al medio de reacción y la velocidad de absorción de fotones en el mismo. Esto requirió la determinación de las propiedades ópticas de las Fe-PILCs en suspensión mediante medidas espectrofotométricas y la resolución de la Ecuación de Transferencia de Radiación utilizando un método numérico. Además de los anticuerpos convencionales, los camélidos poseen anticuerpos carentes de cadena liviana en los que un único dominio forma el sitio de unión al antígeno. Este dominio, denominado Nanobody o VHH, es uno de los fragmentos de reconocimiento de antígeno más pequeños conocidos en la naturaleza (~15 kDa) y se caracterizan por su alta estabilidad, solubilidad y especificidad. Estas características únicas de los VHHs junto a su facilidad de manipulación mediante biología molecular convierten a los VHH en una prometedora alternativa a los anticuerpos convencionales o fragmentos de anticuerpo para numerosas aplicaciones biotecnológicas. Este trabajo de tesis tuvo como horizonte establecer tecnología local para la generación de bibliotecas de nanobodies en fagos filamentosos a partir de linfocitos B de llamas (Lama glama). Asimismo se desarrollaron metodologías que facilitan la selección y caracterización de los nanobodies de interés contra una variedad de antígenos que van desde pequeñas moléculas hasta complejas estructuras celulares. La funcionalidad de los nanobodies seleccionados se demostró en diferentes aplicaciones biotecnológicas. En la primera parte del trabajo se presenta el desarrollo de un vector de alto nivel de expresión y un sistema de biotinilacion in vivo de los nanobodies que facilita el proceso de screening y caracterización paralela de cientos de clones en forma sistematizada y rápida. El sistema tiene gran versatilidad y facilita el estudio de la reactividad de los nanobodies mediante citometría de flujo, ELISA, microscopía, inmunoprecipitación, etc. Como primer aplicación de este sistema se desarrolló un método de ranqueo de afinidad relativa por el antígeno de la población de nanobodies aislados en la etapa de selección inicial (panning) y un sistema que permite la búsqueda de pares de nanobodies para la detección del antígeno en inmunoensayos tipo sándwich, lo que permitió el desarrollo de un ELISA ultrasensible para la cuantificación de la epoxi-hidrolasa humana. 2 En la segunda parte del trabajo se describe la preparación de una biblioteca de nanobodies contra células dendríticas murinas, y la adaptación de la plataforma de descubrimiento de nanobodies para aislar anticuerpos contra receptores celulares nativos utilizando células enteras para las etapas de aislamiento y caracterización. De esta forma se aislaron anticuerpos contra el componente CD11b del receptor Mac-1, la región no polimórfica del MHC-II y contra el receptor CD45, constituyendo un desarrollo de gran relevancia debido a que, a diferencia de los anticuerpos convencionales, los nanobodies pierden drásticamente la reactividad contra su antígeno ante pequeños cambios conformacionales. Debido a eso los nanobodies seleccionados contra receptores recombinantes inmovilizados sobre poliestireno (placa de ELISA) muy raramente reaccionan en forma cruzada con el receptor nativo en las células. A pesar de esto último, y dada la importancia de dirigir la selección utilizando receptores recombinantes, se mostró que la cuidadosa elección de la forma de inmovilización del receptor recombinante, sí permite aislar nanobodies contra receptores nativos. Esto fue demostrado seleccionando nanobodies contra la cadena CD11c del receptor CDR4 que es un marcador de células dendríticas. Por último se realizó una prueba de concepto sobre la utilidad de los nanobodies como dominios de funcionalización, en este caso para conferir funciones efectoras a nanobodies con capacidad de neutralizar la toxina tetánica. Una vez aislados los nanobodies anti-toxina, se prepararon anticuerpos biespecíficos fusionándolos a los nanobodies anti- CD11b, MHCII y CD45, y se estudió su poder de neutralización y biodistribución in vivo, encontrando un dramático incremento del poder neutralizante cuando los anticuerpos neutralizantes se funcionalizaron con el nanobody anti-CD11b (que reacciona con un receptor endocítico), pero no con el anti-MHC-II o anti-CD45. Thiolsulphinate groups have been introduced on to pearl-polymerized agarose, cross-linked dextran, microcrystalline cellulose, polyacrylamide-dextran co-polymers, co-polymerized (ethylene glycol)-methacrylate, acrylic beads and porous glass. The procedure consists of a thiolation and two oxidation reactions. Depending on the conditions and type of solid phase used, degrees of substitution ranging from 60 to 500 μmol of thiolsulphinate groups per g of dried support were obtained. These groups provide the solid phase with the property of reversible covalent binding of thiol compounds via disulphide-bond formation. The experimental conditions for the introduction of thiolsulphinate groups on to agarose have been optimized, as this is one of the most widely used supports in chromatography. With agarose, degrees of thiolsulphinate substitution ranging from 100 to 500 μmol/g of dried derivative can be easily prepared from the same thiol-agarose batch by varying the concentration of the reagent used in the second oxidation step. The thiolsulphinate-agarose had the ability to bind up to 500 μmol of low-Mr thiols/g of dried gel.
