A summary of the resource.
The purpose of the present research was further investigate the reproducibility of check-all-that-apply (CATA) questions for sensory product characterization. Evaluations obtained when such questions are used by consumers are rarely replicated and therefore reproducibility of the data may be at risk. Results from the present work, which included five studies, each with 100–200 consumers across a range of product categories, revealed that sensory product characterizations obtained using CATA questions with consumers are highly reproducible. Hence, the research confirms previous research by Jaeger, Chheang, et al. (2013) and extends it to the use of CATA terms in randomised presentation order as has been recommended to avoid satisficing response behavior. In the future, if CATA studies are conducted without replication and researches seek to examine the reliability of CATA data, the use of a posterior bootstrapping re-sampling approach is suggested.

A summary of the resource.
The glycobiology of the cestodes, a class of parasitic flatworms,<br />is still largely unexplored. An important cestode<br />species is Echinococcus granulosus, the tissue-dwelling<br />larval stage of which causes hydatid disease. The E. granulosus<br />larva is protected from the host by a massive<br />mucin-based extracellular matrix termed laminated layer<br />(LL). We previously reported (Biochemistry 48:11678,<br />2009) the molecular structure of the most abundant LL<br />O-glycans, comprising up to six monosaccharide residues.<br />These are based on Cores 1 and 2, in cases elongated by a<br />chain of Galpβ1-3 residues, which can be capped by<br />Galpα1-4. In addition, the Core 2 GlcNAcp residue can<br />be decorated with the Galpα1-4Galpβ1-4 disaccharide.<br />Larger glycans also detected contained additional HexNAc<br />residues that could not be explained by the structural repertoire<br />described above. In this work, we elucidate, by<br />mass spectrometry (MS) and nuclear magnetic resonance<br />(NMR), six additional glycans from the E. granulosus LL<br />between six and eight residues in size. Their structures are<br />related to those already described but in cases bear GlcNAcpβ1-6 or Galpα1-4Galpβ1-4GlcNAcpβ1-6 as ramifications<br />on the core Galpβ1-3 residue. We also obtained<br />evidence that noncore Galpβ1-3 residues can be similarly<br />ramified. Thus, the new motif together with the previous<br />information may explain all the glycan compositions<br />detected in the LL by MS. In addition, we show that the<br />anti-Echinococcus monoclonal antibody E492 (Parasite<br />Immunol 21:141, 1999) recognizes Galpα1-4Galpβ1-<br />4GlcNAcp (the blood P1-antigen motif). This explains the<br />antibody’s reactivity with a range of Echinococcus tissues,<br />as the P1-motif is also carried on non-LL N-glycans and<br />glycolipids from this genus.<br />

A summary of the resource.
The 11th Pangborn Sensory Science Symposium, which took place in Gothenburg (Sweden), in August 2015 ended with a plenary session where areas of direction for future research in sensory and consumer science were suggested by four speakers. Within their chosen topic the speakers presented three statements which the audience voted on (ranking task) in terms of importance for the future of the field. On the topic of understanding individual variation in sensory perception, the statement endorsed as most important was: “We must engage with other specialists, e.g., geneticists and neuroscientists, to understand the mechanism behind individual variation in perception”. With regard to the role of context and situation in future research and the top ranked statement for this topic was: “We must increase the number of real-life consumer studies for more ecological validity”. With regard to consumers’ decision making processes, the statement endorsed as most important was: “We must develop new methodological approaches to understand the heuristics that influence food choice”. Finally, on the topic of future sensory and consumer research in industry, the top ranked statement was: “We must better capture consumer responses in context (at the moment, in the environment, etc.).” Three broad themes emerged: increasing the ecological validity of sensory and consumer research, doing inter-disciplinary research and accounting for individual differences (perception and decision making). These coincide with the trend from past decades towards a broadening of this science domain.

Abstract

Front-of-package (FOP) nutrition labelling schemes use different framing to highlight nutrient content and promote healthier food choices. Health logos and nutritional warnings can be positioned on opposite sides of a continuous that goes from highlighting healthful products (gain-frame) to highlight unhealthful products (lossframe), respectively. The aim of the present work was to evaluate the impact of health logos and nutritional warnings on consumers’ choice of products with lower sugar content, and the impact of the two FOP nutrition labelling schemes on consumers’ associations with sugar. An online study with 1232 people was carried out. Participants were randomly assigned to three experimental conditions: control (no information), health logos and nutritional warnings. They were presented with a series of three packages of different product categories featuring the corresponding information about sugar content and they had to indicate the product they would buy. After completing the choice tasks, they had to complete a word association task about sugar. Results confirmed that the inclusion of FOP nutrition labelling schemes containing information about sugar content encouraged consumers to select the product with lower sugar content. A larger proportion of participants selected the product with the lowest sugar content when packages featured nutritional warnings compared to when they featured health logos. Results from the word association task showed that nutritional warnings made the negative health effects of sugar more salient on consumers’ mind. These results suggest that FOP nutrition labelling schemes based on a loss-frame to sugar content may be more efficient at encouraging healthier food choices than gain-frame schemes.

Introduction: The identification of chemical compounds that interfere with SARS CoV-2 replication continues to be a priority in several academic and pharmaceutical laboratories. Computational tools and approaches have the power to integrate, process and analyze multiple data in a short time. However, these initiatives may yield unrealistic results if the applied models are not inferred from reliable data and the resulting predictions are not confirmed by experimental evidence. Methods: We undertook a drug discovery campaign against the essential major protease (MPro) from SARS-CoV-2, which relied on an in silico search strategy –performed in a large and diverse chemolibrary– complemented by experimental validation. The computational method comprises a recently reported ligand-based approach developed upon refinement/learning cycles, and structure-based approximations. Search models were applied to both retrospective (in silico) and prospective (experimentally confirmed) screening. Results: The first generation of ligand-based models were fed by data, which to a great extent, had not been published in peer-reviewed articles. The first screening campaign performed with 188 compounds (46 in silico hits and 100 analogues, and 40 unrelated compounds: flavonols and pyrazoles) yielded three hits against MPro (IC50 ≤ 25 μM): two analogues of in silico hits (one glycoside and one benzo-thiazol) and one flavonol. A second generation of ligand-based models was developed based on this negative information and newly published peer-reviewed data for MPro inhibitors. This led to 43 new hit candidates belonging to different chemical families. From 45 compounds (28 in silico hits and 17 related analogues) tested in the second screening campaign, eight inhibited MPro with IC50 = 0.12–20 μM and five of them also impaired the proliferation of SARS-CoV-2 in Vero cells (EC50 7–45 μM). Discussion: Our study provides an example of a virtuous loop between computational and experimental approaches applied to target-focused drug discovery against a major and global pathogen, reaffirming the well-known “garbage in, garbage out” machine learning principle.

Abstract
A summary of the resource.
Gas chromatographic determinations are well established as part of undergraduate laboratory experiments on instrumental analysis (1-5). One objection (5) leveled at some of the suggested experiments is that the determinations do not include the derivatization steps that are almost universal in any "real life" situation |b thus giving the students an erroneous feeling of simplicity. We propose an experiment that does have a derivatization step using readily available reagents; that requires limited manipulative skills |b centring the attention on the method itself; that can be completed within the time constraints of a normal laboratory course; and that

Abstract
A summary of the resource.
Volatile stimuli can be detected by specific protein receptors from olfactory epithelium and trans mitted to the brain where finally are interpreted as the sense of smell. The interpretation of odor response complexity provokes the continuous development of instrumental methods for objectively analyze them. Gas chromatography-olfactometry (GC-O) introduction was a milestone in the odor research of, coupling a resolution technique of volatiles mixtures (gas chromatography) with the selectivity and sensitivity of the human olfactory system. GC-O generates a list of the odor descriptor of each peak in a GC run (“aromagram”), indicating which GC peaks (chromatographic resolved components) have or not given aromas/odors associated. The GC-O technique has been profusely employed for the evaluation of aromas in food matrices, but its application to essential oil study is still scarce for the characterization of specific odor active compounds which contribute to the overall olfactory impression. In this chapter an overview of data able to be obtained applying different GC-O methodologies to essential oils are presented, including the procedures and fundaments involved. The information presented does not intend to be exhaustive, but provides examples available in the literature and references to further reading.

Workflows based on gas and liquid chromatography coupled to mass spectrometry for the identification of toxic pyrrolizidine alkaloids present in plants were developed and applied to Echium plantagineum L., Boraginaceae, extracts. GC–MS based determinations need reduction and derivatization steps prior to MS analysis, which is performed using a Full Scan and Single Ion Monitoring sequence for screening, identification and quantification purposes. The LC–(ESI)–MS/MS determination was performed directly from the extract without derivatization. Acetyl lycopsamine, echimidine, echimidine N-oxide, echiumine, echiumine N-oxide, lycopsamine, lycopsamine N-oxide, 7,9-ditigloylretronecine N-oxide and a not reported PA of m/z 466, were identified and quantified in E. plantagineum extracts, through three operating modes of LC-QTRAP: precursor ion scan, enhanced product ion scan and multiple reaction monitoring. Precursor ion scan detects all the ions that give rise to a daughter ion at m/z 120, the presence of the parent pyrrolizidine alkaloid is confirmed through its MS2 spectrum (enhanced product ion) and quantified by multiple reaction monitoring. These workflows are general approaches to study chemical families using GC/LC-MS. For extracts suspicious of containing pyrrolizidine alkaloids, they are suitable tools for the quality and safety control of food, feed as well as phytotherapeutics.

Plant defensins comprise a large family of antimicrobial peptides (AMP) with a wide range of biological functions. They are cysteine-rich molecules, highly sequence diverse but with a conserved and stable structure. In this work, a defensin gene (EcgDf1) was isolated from Erythrina crista-galli, a legume tree native from South America. The predicted peptide presents eight cysteines, with a γ-core motif GXCX3-9C and six cysteines distributed like the typical defensin αβ motif. The mature EcgDf1 coding sequence was heterologously expressed in Escherichia coli strains and purified by affinity chromatography. Possible dimer and oligomers of EcgDf1 were visible in SDS electrophoresis. Moreover, its 3D structure, determined by homology modeling, docking, and molecular dynamics simulations, was found to be compatible with the formation of homodimers between the β3 and β1-loop-α1, leaving the β2-loop-β3 free to interact with lipid membranes. The purified recombinant peptide inhibited the growth of several critical plant and human pathogens, like the opportunistic fungi Candida albicans and Aspergillus niger and the plant pathogens Clavibacter michiganensis ssp. michiganensis, Penicillium expansum, Botrytis cinerea, and Alternaria alternata. EcgDf1 is a promising candidate for the development of antimicrobial products for use in agriculture and medicine.

An efficient and facile general method for the synthesis of conduritol C analogs, taking advantage of an enantioselective biocatalysis process of monosubstituted benzenes, is described. The absolute stereochemical patterns of the target molecules (−)-conduritol C, (−)-bromo-conduritol C, and (−)-methyl-conduritol C were achieved by means of chemoenzymatic methods. The stereochemistry present at the homochiral cyclohexadiene-cis-1,2-diols derived from the arene biotransformation and the enantioselective ring opening of a non-isolated vinylepoxide derivative permitted the absolute configuration of the carbon bearing the hydroxyl groups at the target molecules to be established. All three conduritols and two intermediates were crystallized, and their structures were confirmed by X-ray diffraction. The three conduritols and intermediates were isostructural. The versatility of our methodology is noteworthy to expand the preparation of conduritol C analogs starting from toluene dioxygenase (TDO) monosubstituted arene substrates.

Bacterial diseases affecting wheat production in Uruguay are an issue of growing concern yet remain largely uninvestigated in the region. Surveys of 61 wheat fields carried out from 2017 to 2019 yielded a regional collection of 63 strains identified by 16S rRNA gene analysis as Xanthomonas spp. A real-time PCR protocol using species-specific primers previously reported allowed the identification of 44 strains as X. translucens, the causal agent of bacterial leaf streak (BLS) in wheat and other cereal crops. Multilocus sequence analysis (MLSA) of four housekeeping genes (dnaK, fyuA, gyrB, and rpoD) revealed that these strains were most closely related to X. translucens pv. undulosa, the pathovar that is most commonly associated with BLS of wheat. Multilocus sequence typing (MLST) was applied to examine the genetic diversity among X. translucens strains. Strains were assigned to four different sequence types, three of which have been previously reported globally. Additionally, 17 Xanthomonas strains not belonging to X. translucens were obtained from diseased wheat leaves. Phylogenetic analysis showed that these strains are closely related to Xanthomonas prunicola, and clustered together with previously uncharacterized Xanthomonas strains isolated from wheat in Minnesota, US. In planta pathogenicity assays carried out on a BLS susceptible wheat cultivar showed that X. translucens pv. undulosa strains caused brown necrosis symptoms typical of BLS, while non-translucens Xanthomonas sp. strains elicited an atypical symptom of dry necrosis. These findings suggest that local wheat fields are affected by X. translucens pv. undulosa, and by a new wheat pathogen within the Xanthomonas genus.
