Two multiple response optimization methodologies were applied and compared to obtain an optimum combination medium formulation for culturing the yeast Rhodotorula graminis: (1) desirability function, and (2) pareto-optimal front by means of genetic algorithms (POF-GA). The concentration of microbial lipids, a raw material for the production of biodiesel, was modelled by the response surface methodology and then maximized through the application of a methodology based in genetic algorithms. The results obtained by applying POF-GA (one of the first times this approach was applied to a microbiological process) were confirmed by using the well-known Derringer's desirability function and by carrying out independent experiments. Then, it was verified that POF-GA is a viable option to be applied in biotechnological processes and that molasses may be employed as a source for culture media employed to produce microbial lipids, representing important economical savings since molasses is a raw material from sugarcane industrialization. Fijiviruses replicate and package their genomes within viroplasms in a pro cess involving RNA-RNA and RNA-protein interactions. Here, we demonstrate that the 24 C-terminal residues (C-arm) of the P9-1 major viroplasm protein of the mal de Río Cuarto virus (MRCV) are required for its multimerization and the formation of viroplasm like structures. Using an integrative structural approach, the C-arm was found to be dis pensable for P9-1 dimer assembly but essential for the formation of pentamers and hexamers of dimers (decamers and dodecamers), which favored RNA binding. Although both P9-1 and P9-1DC-arm catalyzed ATP with similar activities, an RNA-stimulated ATPase activity was only detected in the full-length protein, indicating a C-arm-mediated interaction between the ATP catalytic site and the allosteric RNA binding sites in the (do)decameric assemblies. A stronger preference to bind phosphate moieties in the dec amer was predicted, suggesting that the allosteric modulation of ATPase activity by RNA is favored in this structural conformation. The factors affecting the innate susceptibility to Echinococcus granulosus infections are largely unknown. We assessed the interaction of healthy human neutrophils with protoscoleces (PSC) and antigen B (AgB) of E. granulosus by analysis of CD11b upregulation and H2O2 production by flow cytometry. PSC induced neutrophil activation, but their viability was not affected. In contrast, no effects were observed with AgB in both assays. Neutrophil-enriched fractions were also incubated with PSC or AgB, and interleukin 8 (IL-8) production was measured by ELISA. Significant increment in IL-8 production was detected only in supernatants from neutrophilenriched fractions cultured with PSC. The possible effect of a prior incubation with AgB on the phorbol myristate acetate-induced activation was also evaluated. No changes were observed in CD11b expression, but the H2O2 production was significantly reduced in platelet-activating factor (PAF)-primed neutrophils. The synthesis and spectroscopic characterization (UV–Vis, IR, EPR) of a new copper complex with a betablocker propranolol (1-(isopropylamino)-3-(1-naphthyloxy)-2-propanol) are presented. Besides, the Xray crystal structure of [Cu(propranolol)2]�2H2O is determined, showing two different copper ions, one coordinated through two S propranolol isomers and the other through two R isomers. The effect on the heart contraction force and on the heart rate plus the block of response to adrenaline of the complex and the free ligand, were studied. The effect of [Cu(propranolol)2]�2H2O on contractility was very similar to that of the free propranolol while the reduction on the heart rate is approximately 30% of the reduction obtained with the free ligand. This is an encouraging result since the search of new beta-blocker drugs that have lesser effect on heart rate is one of the important topics in cardiac pharmacology. Niobium-based oxides are being increasingly evaluated as materials for energy storage applications. Additionally, the use of these oxides as cathodes in aqueous electrolytes has shown promise. Based on this, the pseudocapacitive behavior of protonic niobate nanowires in an aqueous acidic electrolyte (1 M H2SO4) was evaluated for the first time. The material was obtained in two simple sequential steps. First, hydrothermal synthesis resulted in sodium niobate; second was ionic exchange (in two concentrations of 2 M and 0.1 M HNO3), where the protonic niobate was obtained. The resulting protonic niobate was characterized by FEG-SEM, the results demonstrated that the morphology of the oxide was concentration-dependent in the ionic exchange step, and EDS analysis was used to validate the procedure. Using DRX, Raman spectroscopy, and FTIR analysis, the transformation of sodium niobate to protonic niobate was evidenced. The electrochemical tests demonstrated that the protonic niobate presented pseudocapacitive behavior when employed as the cathode in 1 M H2SO4, and the ionic exchange in 2 M HNO3 promoted a better specific capacitance, reaching 119.8 mF·cm−2 at a 1 mA·cm−2 current density. Estudios previos mostraron que las características senso-riales, especialmente el sabor, son claves en la aceptación de las mandarinas por los consumidores. Se llevaron a cabo dos estudios durante la estación de cosecha de mandarinas (media y tardía) aplicando la metodología «marque todo lo que corresponda» (CATA) y con la participación de al menos 100 consumidores. La aceptabi-lidad de los cultivares que fueron más aceptados se situó entre 6,5 y 7,7 en una escala hedónica de 9 puntos, evidenciando una reacción hedónica positiva. Los nuevos híbridos presentaron puntajes de aceptabilidad simi-lares o superiores a los cultivares de referencia Tango, Gold Nugget, Murcott y Ortanique. A su vez, los resul-tados mostraron que el color naranja, la forma regular, la piel lisa, el dulzor, la jugosidad, el sabor intenso y el sabor típico a mandarina son características claves que determinan la aceptabilidad de las mandarinas, y que la apariencia externa no es un buen predictor del sabor. Limited shelf life of bakery products, caused by microbial deterioration, is a concern for industries due to economic losses. Fungal spoilage of sponge cakes industrially produced in Montevideo was caused mainly by Penicillium species, in particular by Penicillium crustosum. The combination of different hurdles was studied to inhibit P. crustosum growth in sponge cakes. A full factorial design was performed to study the effect of the concentration of potassium sorbate, pH, packaging atmosphere and storage time. The results showed that packaging atmosphere and storage time were the significant factors in the ranges tested. No growth was detected in cakes stored in modified atmosphere packaging (MAP) (N2:CO2 50:50) at room temperature (25 °C) for 15 days. The effect of MAP on P. crustosum growth in cakes at room temperature was compared with the effect of air-packaging and storage at low temperature (4 °C) for 30 days. P. crustosum growth was not detected in cakes packaged in MAP, whereas it was detected after 20 days in cakes packaged in air and stored at 4 °C. This growth was quantified by a specific real time PCR developed in this work. Specific primers were designed using the sequence of β-tubulin gene of P. crustosum as a target and PCR conditions were adjusted to ensure specificity. PCR efficiency was 107%, with a detection limit of 0.0014 ng of DNA. The qPCR method presented here, resulted specific and sensitive enough to detect the growth of P. crustosum even before biodeterioration signs were visible. Trifluoroacetonitrile (CF3CN) is a decomposition product from the promising dielectric heptafluoro isobutyronitrile (C4F7N) under partial discharge process, so can be used to detect failures in gas-insulated switchgear. In this work, we study through density functional theory (DFT) calculations the adsorption of CF3CN on pristine aluminium nitride nanosheets (AlNNS) and defective Al-vacancy (vAl) and N-vacancy (vN) AlNNS, in order to explore the potential of these nanomaterials as sensors for this gas. Our energetic analysis shows negligible adsorption energy for the pristine surface but strong interactions can be observed between the molecule and the monovacancy-defected surfaces, with adsorption energy values estimated to be about − 2.2 and − 3.1 eV for CF3CN/vN-AlNNS and CF3CN/vAl-AlNNS, respectively, which suggests chemisorption processes. Furthermore, our geometric and bonding studies revealed the formation of new N—Al interactions between gas and monovacancy-defected surfaces, with serious distortions for the vAl-AlNNS nanosheet. The electronic density of states and band structure analysis reveals moderate band gap variations after gas adsorption. In contrast, significant changes in the work function and the magnetic moment of defective AlNNS were computed after gas exposition, for all optimized adsorption configurations.
