Abstract

Benzenoids are compounds associated with floral and fruity flavours in flowers, fruits and leaves and present a role in hormonal signalling in plants. These molecules are produced by the phenyl ammonia lyase pathway. However, some yeasts can also synthesize them from aromatic amino acids using an alternative pathway that remains unknown. Hanseniaspora vineae can produce benzenoids at levels up to two orders of magnitude higher than Saccharomyces species, so it is a model microorganism for studying benzenoid biosynthesis pathways in yeast. According to their genomes, several enzymes have been proposed to be involved in a mandelate pathway similar to that described for some prokaryotic cells. Among them, the ARO10 gene product could present benzoylformate decarboxylase activity. This enzyme catalyses the decarboxylation of benzoylformate into benzaldehyde at the end of the mandelate pathway in benzyl alcohol formation. Two homologous genes of ARO10 were found in the two sequenced H. vineae strains. In this study, nine other H. vineae strains were analysed to detect the presence and per cent homology of ARO10 sequences by PCR using specific primers designed for this species. Also, the copy number of the genes was estimated by quantitative PCR. To verify the relation of ARO10 with the production of benzyl alcohol during fermentation, a deletion mutant in the ARO10 gene of Saccharomyces cerevisiae was used. The two HvARO10 paralogues were analysed and compared with other α‐ketoacid decarboxylases at the sequence and structural level.

Bacterial wilt, caused by Ralstonia solanacearum, is a major disease affecting potato (Solanum tuberosum) production worldwide. Although local reports suggest that the disease is widespread in Uruguay, characterization of prevalent R. solanacearum strains in that country has not been done. In all, 28 strains of R. solanacearum isolated from major potato-growing areas in Uruguay were evaluated, including 26 strains isolated from potato tubers and 2 from soil samples. All strains belonged to phylotype IIB, sequevar 1 (race 3, biovar 2). Genetic diversity of strains was assessed by repetitive-sequence polymerase chain reaction, which showed that the Uruguayan strains constituted a homogeneous group. In contrast, inoculation of the strains on tomato and potato plants showed, for the first time, different levels of aggressiveness among R. solanacearum strains belonging to phylotype IIB, sequevar 1. Aggressiveness assays were also performed on accessions of S. commersonii, a wild species native to Uruguay that is a source of resistance for potato breeding. No significant interactions were found between bacterial strains and potato and S. commersonii genotypes, and differences in aggressiveness among R. solanacearum strains were consistent with previously identified groups based on tomato and potato inoculations. Moreover, variation in responses to R. solanacearum was observed among the S. commersonii accessions tested.

Methotrexate (MTX), a structural analog of folic acid, is widely employed in the treatment of different cancers and autoimmune diseases. Despite the successful results observed, the main disadvantage lies in interpatient variability in the pharmacokinetic and pharmacodynamic parameters. In particular, adverse events and toxicities induced by MTX are a matter of concern and can be the cause of dose reduction or treatment discontinuation. Among the different approaches to reduce MTX therapeutic limitations, pharmacogenomics contributes by considering the effect of inherited genetic differences on those parameters. This review provides an update on MTX pharmacogenomics. It reports the contribution of main gene polymorphisms involved in the influx, efflux, cellular effect, and elimination on MTX toxicity and efficacy, on all the diseases treated with this drug. From the analysis of the data presented in this review, we concluded that only gene polymorphisms MTHFR rs1801133, SLC19A1 rs1051266, and TYMS rs34743033 could influence clinical decision-making.

Efflux pumps can make a significant contribution to the capacity of bacteria to resist the action of antibiotics. Certain efflux pumps also recognize antimicrobial agents that are present in their respective hosts and their ability to export toxic agents could enhance bacterial survival during infection prior to appearance of cellular or humoral host defensive systems. This review is concerned with the principal efflux pumps possessed by two closely related strict human pathogens, Neisseria gonorrhoeae and Neisseria meningitidis. Specific emphasis is placed on the organization of the structural genes encoding the mtr and far efflux pumps, the substrates (often host- derived) recognized by these pumps, and the cis- and trans -acting transcriptional factors that regulate efflux pump gene expression in gonococci and meningococci. The overriding theme of this review is that the efflux pumps possessed by these pathogens likely contribute to their pathogenic mechanisms by providing a means to escape a number of antimicrobial compounds that bathe mucosal surfaces

Genetic fingerprint profiles, the type and content of glycoalkaloids (GAs) and hemagglutination (HAG) activity against red cells were analysed in accessions of Solanum commersonii, collected from different locations in the south of Uruguay. Thirty-nine accessions from 21 geographically distinct areas were studied. Random Amplified Polymorphic DNA (RAPD) analysis revealed a high degree of genetic diversity among the accessions used in the study and effectively discriminated among all of the accessions analysed. There was a very high diversity in the type as well as the concentration of GAs in the samples. Strong HAG activity against rabbit red cells was detected in all the S. commersonii tuber extracts analysed. Such activity was specifically inhibited by N,N#-diacetylchitobiose and N,N#,N$-triacetylchitotriose. Differences in the levels of specific HAG activities were found in the different extracts, which might indicate different levels of the lectin specific for N-acetylglucosamine (Glc-NAc) and its oligomers, in the tubers. Nitric oxide donor tocopherol analogs were found to be incorporated in low-density lipoprotein to release nitric oxide into the hydrophobic core of the lipoprotein, thus inhibiting lipid oxidation processes associated with atheroma plaque formation. Previously, we studied their cytotoxicity against human and murine macrophages as first selection for in vivo studies. Herein, we examined both the in vitro mutagenic and DNA-damage effects of selected compounds to further evaluate drug potential. While the compounds of interest were nongenotoxics in both experimental tests (Ames and alkaline comet), one of the potential blood metabolites exhibited genotoxicity (alkaline comet test), and the furazan derivative was mutagenic (Ames test). Two selected (nitrooxy and furoxan) compounds were studied in long- and short-term in vivo treatment, and in these conditions, animal toxicity was not evidenced, suggesting the possibility of these compounds as potential antiatherogenic drugs.

<p>Methotrexate (MTX), an important chemotherapeutic agent, is often accompanied with mucositis. The occurrence and severity are unpredictable and show large interindividual variability. In this study, we review and meta-analyze previously studied genetic variants in relation to MTX-induced mucositis.<br />We conducted a systematic search in Medline and Embase. We included genetic association studies of MTXinduced<br />mucositis in cancer patients. A meta-analysis was conducted for single nucleotide polymorphisms (SNPs) for which at least two studies found a statistically significant association.<br />A total of 34 SNPs were associated with mucositis in at least one study of the 57 included studies. Two of the seven SNPs included in our meta-analysis were statistically significantly associated with mucositis: MTHFR c.677C > T (recessive, grade ≥3 vs grade 0–2, OR 2.53, 95 %CI [1.48–4.32], False Discovery Rate[FDR]- corrected p-value 0.011) and MTRR c.66A > G (overdominant, grade ≥1 vs grade 0, OR 2.08, 95 %CI<br />[1.16–3.73], FDR-corrected p-value 0.042).</p>

Issatchenkia terricola 0621 is a non-Saccharomyces yeast strain isolated from Tannat grapes from Uruguayan vineyards; it stands out for its ability to produce high levels of β-glucosidase activity, which contributes to the aromatic complexity of wines. To delve into the potential oenological applications of this strain, its high-quality genome was obtained and explored, allowing the main central carbon and nitrogen metabolic pathways to be reconstructed. I. terricola is able to utilise glycerol as the sole carbon source in a way that has not previously been described for yeasts. The genes of the fermentome and those involved in stress resistance during winemaking were also identified, and differences were found when compared to S. cerevisiae, which may explain why I. terricola is unable to complete fermentation. The pathways responsible for natural aroma synthesis were also reconstructed, and the production of aromatic acids, alcohols, esters, acetates and lactones was verified experimentally. Finally, sequences encoding for β-glucosidases, in addition to the previously characterised one, were identified in the genome. The work presented here lays the groundwork for experimental research focused on the dissection of the metabolism of a native non-Saccharomyces strain and its application for oenological and biotechnological purposes.

Hanseniaspora is the main genus of the apiculate yeast group that represents about 70% of the grape-associated microflora. Hanseniaspora vineae is emerging as a promising species for quality wine production compared to other non-Saccharomyces Wines produced by H. vineae with Saccharomyces cerevisiae consistently exhibit more intense fruity flavours and complexity than wines produced by S. cerevisiae alone.In this work, genome sequencing, assembling and phylogenetic analysis of two strains of H. vineae shows that it is a member of the Saccharomyces complex and it diverged before the Whole Genome Duplication (WGD) event from this clade. Specific flavour gene duplications and absences were identified in the H. vineae genome, as compared to 14 fully sequenced industrial S. cerevisiae genomes. The increased formation of 2-phenylethyl acetate and phenylpropanoids such as 2-phenylethyl and benzyl alcohols might be explained due to gene duplications of H. vineae aromatic amino acid aminotransferases (ARO8, ARO9) and phenylpyruvate decarboxylases (ARO10).Transcriptome and aroma profiles under fermentation conditions confirmed these genes were highly expressed at the beginning of stationary phase coupled to the production of their related compounds. The extremely high level of acetate esters produced by H. vineae compared to S. cerevisiae is consistent with the identification of six novel proteins with alcohol acetyltransferase (AATase) domains. The absence of the branched-chain-amino-acid transaminases (BAT2) and acyl-CoA/ethanol O-acyltransferases (EEB1) genes, correlates with H. vineae reduced production of branched-chain higher alcohols, fatty acids and ethyl esters, respectively. Our study provides sustenance to understanding and potentially utilizing genes that determine fermentation aromas.Importance The huge diversity of non-Saccharomyces yeasts in grapes is dominated by the apiculate genus Hanseniaspora Two native strains of H. vineae applied to winemaking due to their high oenological potential in aroma and fermentation performance, were selected to obtain high quality genomes. Here, we present a phylogenetic analysis, and the complete transcriptome and aroma metabolome of H. vineae during three fermentation steps. This species produced significantly richer flavour compound diversity compared to Saccharomyces, such as benzenoids, phenylpropanoids, and acetate derived compounds. The identification of six proteins, different from S. cerevisiae ATF, with diverse acetyl transferase domains in H. vineae offers a relevant source of native genetic variants for this enzymatic activity. The discovery of benzenoid synthesis capacity in H. vineae provides a new eukaryotic model to dilucidate an alternative pathway to that catalysed by plants' phenylalanine lyases.

The identification of six proteins, different from S. cerevisiae ATF, with diverse acetyltransferase domains in H. vineae offers a relevant source of native genetic variants for this enzymatic activity. The discovery of benzenoid synthesis capacity in H. vineae provides a new eukaryotic model to dilucidate an alternative pathway to that catalyzed by plants' phenylalanine lyases.

Isolation and characterization of common scab (CS) pathogen Streptomyces spp. from Uruguayan potato tubers and soil samples were done in response to significant economic losses due to CS on potato in autumn 2010. Seventy of the 331 isolates were classified as pathogenic owing to their ability to induce necrosis on tuber disks and stunting of radish seedling. Streptomyces spp. causing CS on potato in Uruguay were found to represent a range of different species by virtue of their diverse morphological and physiological traits as well as rep-PCR, rpoB phylogenetic analysis, and multi-locus sequences analysis. We identified isolates primarily as Streptomyces scabiei, S. acidiscabies, and S. europaeiscabiei. However, some of the pathogenic isolates still remain to be identified at the species level. This highlights the need for improved methods for discrimination among pathogenic Streptomyces species. The presence of Streptomyces pathogenicity island (PAI) genes was analyzed, including genes encoding for thaxtomin synthetase (txtA, txtB), tomatinase (tomA), and a necrosis protein (nec1). Among the isolates that were pathogenic, 50% contained the four pathogenicity genes, 33% had an atypical composition of PAI marker genes, and 17% did not contain any genes. The absence of the genes reported to be involved in thaxtomin biosynthesis (txtA, txtB) was confirmed by whole-genome sequencing of two representative strains of this group. This finding suggests the participation of other virulence factors in plant pathogenicity.

A summary of the resource.
The chemical composition of Aloysia virgata var. platyphylla oil obtained from aerial parts of different plants populations growing wild in San Lorenzo (Province of Corrientes, Argentina) were compared with the essential oil of the same species from Rio Empedrado. These selected zones represent similar ecological and geographical units of soils. In order to evaluate the constancy of the oil’s composition for this species in the selected region, samples were collected at different seasonal stages. For a more precise identification and description of chemical biodiversity of this medicinal and aromatic plant, the analysis and characterization of enantiomers from sabinene, β-caryophyllene and germacrene D has also been evaluated. Using this approach, it has been possible to establish the relationships between the chemical variability and the determinant environmental factors in order to characterize the species ecologically.

Abstract
A summary of the resource.
Cis-trans isomerization studies were performed on commercial oleic acid and refined sunflower oil. Reactions were carried out at constant temperature, under inert atmosphere (without hydrogen), using an industrial nickelcontaining hydrogenation catalyst. With sunflower oil, hydrogen transfer and polymerization were concurrent with the formation ofrans isomers.

Abstract
A summary of the resource.
The kinetics of the geometrical isomerization of oleic and palmitoleic acids both contained in U.S.P. oleic acid were studied. Sulfur powder was used as a catalyst. The methyl esters of fatty acids were analyzed by GLC with 15 OV-275 columns. The sulfur-catalyzed isomerization at 180 and 225 grC proceeds via two consecutive mechanisms. The position of equilibrium is reached by the second mechanism. For this at any particular initial concentration of sulfur |b the pseudo-first-order rate dependence on substrate for a reversible reaction holds. The full rate has been shown to be proportional to the initial sulfur concentration taken to the 1.2 power. The rate constants at both temperatures and the activation energies were calculated.

Abstract
A summary of the resource.
This study investigated the effect of hunger on consumers’ visual attention during a food choice task, and the role of time orientation (i.e., present and future orientation) in this interplay. A lab-based eye-tracking experiment including 102 participants was conducted, with hunger as the manipulated factor (hungry, satiated). Participants in the satiated condition were served a breakfast buffet before the experimental tasks, whereas participants in the hungry condition were served the buffet after completion of the tasks. Both groups were exposed to a set of planograms depicting supermarket shelves and were asked to choose an option they could consider buying, while their eye movements were recorded. Stimuli included non-food items as well as the key category of interest, Swedish crisp bread. After completion of the eye-tracking recordings, participants indicated their time orientation as well as their height and weight, which were used to calculate body mass index (BMI). The results revealed that hunger (vs. satiation) increased participants’ present orientation and visual attention towards bread, but decreased their future orientation, with participants’ present orientation mediating the effect of hunger on visual attention. Additional exploratory analyses revealed a positive correlation between participants’ BMI and their present orientation. Taken together, the results offer several fruitful avenues for future research, which may be used to promote public health. Moreover, the findings contribute to the literature by documenting that hungry individuals do not necessarily make more rapid decisions in drive-relevant domains; rather, they may actually devote a larger share of their attentional resources in the food domain, given their desire to acquire food
