<p>Detection of sulfuryl fluoride (SO2F2) and thionyl fluoride (SOF2) can be used to find out the degradation of sulfur hexafluoride (SF6), indicating gas insulation switchgear equipment failures. In the present work, we study through first-principle DFT calculations the SOF2 and SO2F2 adsorption on aluminium nitride nanosheets (AlNNS), with the aim of to evaluate the performance of this nanomaterial as gas sensor. First, we performed an energetic, geometric and bonding analysis, obtaining considerable adsorption energy values for SO2F2 and SOF2 (−1.63 and −1.93 eV, respectively), which suggests these gases are chemisorbed on the AlN surface.<br />Furthermore, certain SeF bonds were broken during adsorption, while new AleF and SeN overlaps were developed. Simultaneously, an important weakening was observed for AleN bonds close to the adsorption site. The adsorption selectivity of AlNNS toward SO2F2 and SOF2 was also studied, noting that other molecules normally present in air show a lower reactivity.<br />Band structure and electronic density of states were realized in order to evaluate the sensing capacity of AlNNS surface. The obtained plots show a notorious narrowing of the band gap after gas adsorption, particularly in the case of SOF2, so an important change in the conductivity of AlNNS could be expected during gas exposition. Charge density analysis reveals an important charge transfer from AlNNS to both adsorbed gases, which suggest that this nanomaterial could behave as an n-type semiconductor. These results seem to indicate that<br />AlNNS could act with good sensibility and selectivity for SOF2 and SO2F2 detection, showing high potential as gas sensing material.</p>

Cell death mechanisms in Trypanosoma cruzi have not been disclosed in detail though different conventional techniques have been used in the classification of parasite-cell death type. Nuclear magnetic resonance (NMR) has successfully been used as a tool to evaluate the onset of apoptosis in a number of higher eukaryote-cell models analysing the ratio of CH2/CH3 integration from the visible mobile lipids (VML). Surprisingly, this versatile non-invasive spectroscopy technique has never been employed with this purpose in T. cruzi. In the present study it is shown that under different parasite deathconditions the ratio CH2/CH3 varied drastically. Thus, T. cruzi epimastigotes in apoptotic conditions increase significantly this ratio while in necrotic as well as in autophagic situations the parasites maintain the VML, CH2/CH3 ratio, in normal values. Additionally, other VML markers commonly used in these studies, such as the change in the region of methylcholine moiety, -N+(CH3)3, exhibited different particular patterns according to the type of cell death. Our results suggest that the 1H NMR-VML technique is an adequate tool to discriminate different T. cruzi death pathways.

In this work the characterization of P-29, a novel 29 kDa antigen from Echinococcus granulosus is reported. E. granulosus was identified while looking for parasite antigens distinct from those present in hydatid cyst fluid. A monoclonal antibody (mAb 47H.PS) prepared against protoscolex components revealed that P-29 is localized to the tegument and rostellum of protoscoleces, and to the germinal layer of the cyst, but it is absent in hydatid cyst fluid or adult worm extracts. Several internal fragments of P-29 showed sequence identity to the amino acid sequence encoded by Eg6, a partial gene sequence reported to code for an epitope of antigen 5 (Ag5), one of the major diagnostic antigens of the parasite. We confirmed that Eg6 encodes a sub-fragment of P-29 by mapping the epitope of mAb 47H.PS, and isolating the full length P-29 cDNA. Since Eg6 had been postulated to encode a fragment of Ag5, we specifically studied the relationship of P-29 and Ag5 by: (i) examining the cross-reactivity displayed by different mAbs; (ii) comparison of their peptide finger prints; and (iii) a comparative study of their diagnostic value. Our results prove unequivocally that P-29 and Ag5 are immunologically related, but different proteins, raising several questions on the current knowledge of Ag5

Monoclonal antibodies (MoAbs) specific for unique epitopes of the catalytic domain of cruzipain (Crz) were used to develop a two-site sandwich ELISA specific for native Crz. In addition, the authors developed a sandwich ELISA that allowed the detection of the protease C-terminal domain (CT) using a combination of a MoAb specific for the CT and rabbit anti-Crz IgGs. Both assays were sensitive with detection limits of 2 ng/ml and 0.7 ng/ml, respectively. The assays were assessed for applicability in detection of antigens in serum and urine from experimentally infected BALB/c mice. The antigens were already detectable in serum by the third week after infection, reached their peak by week four, and decreased during the chronic phase of the infection. Throughout the infection the relative amount of CT detected was several-fold higher than that of native Crz, and the data demonstrate that the cT exposes highly immunogenic epitopes that are absent in native Crz. Since these observations have a potential application in diagnosis, the authors analysed the degree of cross-reactivity with antigens from T. rangeliT. bruceiLeishmania mexicana and L. panamensis, and determined that the assays were highly specific. Measurable amounts of the CT were also recorded in urine samples.

In the present study we describe the production and characterization of a panel of monoclonal antibodies (MoAbs) directed against cruzipain (Crz), the major cysteine proteinase from Trypanosoma cruzi. The five MoAbs, BD6, BF2, CG2, CH8, and DC10 were analysed with respect to affinity and specificity. None of the MoAbs cross-reacted with papain, which has regions of high homology with Crz. Treatment of the antigen with periodate did not affect the binding of the MoAbs. suggesting that they bind to the polypeptide moiety of Crz. CH8 recognized a continuous epitope located at the C-terminal extension of the proteinase that appeared to be highly immunogenic. Although the rest of the MoAbs recognized epitopes located in the catalytic domain, the enzymatic activity of Crz was not impaired by the binding of the MoAbs. Characterization of the antibody-binding sites revealed the presence of at least four separate epitopes.

A summary of the resource.
The efficiency of supercritical carbon dioxide for the selective removal of the more important pesticides from commercial lanoline was studied. Lanoline solubility values from 0.03 to 2.50 mg/g CO2 were obtained, corresponding to CO2 densities from 498.4 to 857.3 kg/m3, respectively. The original lanoline showed the following pesticide profile: diazinon 66 ppm, methyl-pirimifos 4.6 ppm and ethion 140 ppm. By an extraction carried out at 25 MPa and 45 °C, using a mass ratio solvent to substrate of 19.5 g CO2/mg lanoline, only 3.4% of lanoline was co-extracted and the pesticide concentrations were reduced to 0.47 ppm for diazinon, less than 20 ppb for methyl-pirimifos and 0.2 ppm for ethion. These results shows that supercritical CO2 is a very convenient solvent for the selective extraction of the main pesticides from lanoline, without the occurrence of substantial lanoline losses during the process.

El derivado nitrado del ácido araquidónico (AA), el ácido nitroaraquidónico (NO2AA) presenta actividades antiinflamatorios celulares durante la activación de los macrófagos. Sin embargo, se sabe poco acerca de la capacidad del NO2AA para regular la producción de especies reactivas de oxígeno (ROS) en condiciones proinflamatorias. Una de las respuestas inmediatas tras la activación de macrófagos implica la producción de radical superóxido (O2•⎯) por la enzima NADPH oxidasa fagocítica (NOX2). Como el AA es capaz de activar a la NOX2, proponemos que NO2AA puede modular la activación de los macrófagos mediante al inhibir la formación de O2•⎯ por NOX2. Cuando los macrófagos se activaron en presencia de NO2AA, se observó una inhibición significativa de la actividad NOX2 evaluadas por la reducción de citocromo c, la luminiscencia de luminol, la fluorescencia del Amplex Red y por citometría de flujo. Este proceso también se produce en el fagosoma, sin afectar la viabilidad celular ni la fagocitosis. El NO2AA disminuyó la producción de O2•⎯ de forma dependiente de la dosis y del tiempo de incubación siendo un efecto reversible. La inhibición observada no fue debida a una disminución de la fosforilación de las subunidades citosólicas (p40phox y p47phox). Sin embargo, se observó una reducción de la migración a la membrana de p47phox lo que implica la prevención del correcto montaje de la enzima activa en la membrana. Los efectos observados in vitro se comprobaron en un modelo inflamatorio agudo in vivo mediado por tioglicolato, donde la inyección subcutánea de NO2AA fue capaz de disminuir la actividad NOX2 en los macrófagos de ratones tratados, confirmando los efectos observados en los estudios celulares. Trabajos recientes indican la participación de la Proteína Disulfuro Isomerasa (PDI) en la activación de NOX2. Creemos que el NO2AA podría afectar directamente a la PDI provocando la inhibición de la actividad NOX2. El NO2AA inhibió las actividades tanto reductasa como chaperona de la PDI de forma dependiente de la dosis y del tiempo de incubación siendo independiente del •NO liberado por el NO2AA. Como los nitroalquenos son potentes electrófilos y la PDI tiene residuos de cisteína críticos para su actividad, podría formarse un aducto covalente reversible entre NO2AA y PDI. Hemos demostrado la modificación covalente reversible de PDI por NO2AA en residuos de cisteínas usando el ensayo de PEG-switch, así como LCMS/ MS, determinando la modificación de residuos de cisteína del sitio activo de 4 PDI. Además, los estudios de microscopía confocal demostraron que el pretratamiento con NO2AA en macrófagos activados con PMA presenta un cambio en la distribución intracelular de PDI así como cambios en la estructura de actina. El inhibidor de PDI Tiomuscimol presentó un efecto similar al nitroalqueno tanto en la distribución de PDI como en la inhibición de NOX2, lo que indica que la inhibición de NOX2 por NO2AA puede ser debido a la inhibición de la PDI. Asimismo, la colocalización de p47phox con PDI se vio disminuida en presencia del NO2AA, pudiendo afectar la función de PDI sobre la actividad NOX2. En resumen, nuestros resultados implican que las modificaciones covalentes por NO2AA en PDI podrían afectar la función moduladora de PDI en la NOX2, evitando su interacción con p47phox y su migración a la membrana, necesarios para la activación de la NOX2.

Our research group has made incursions into the scarcely known coordination chemistry of rhenium(II). The literature shows that Re(II) mononuclear complexes are attractive in molecular magnetism due to high magnetic anisotropy because of a significant spin-orbit coupling, making them a potential source for new molecule-based magnets. In this work, we present the preparation of four novel Re(II) compounds of general formula NBu4 [Re(NO)Br4 (L)] [NBu4 + = tetra-n-butylammonium: L = imidazole (1), pyrazole (2), 1,2,4-triazole (3) and 1H-tetrazole (4)]. The four compounds were fully characterized by single-crystal X-ray diffraction, infrared spectroscopy, and cryomagnetic measurements in the temperature range of 1.8–300 K. Their crystal structures consist of mononuclear [Re(NO)Br4 (L)]− complex anions and NBu4 + cations. Each Re(II) ion is six-coordinate with a linear nitrosyl group and one monodentate nitrogen-donor (L), which are trans-positioned, plus four bromide groups, building a tetragonally distorted octahedral surrounding. The inter-anionic contacts were thoroughly analyzed using Hirshfeld surface analyses (plots over the dnorm, shape index, and 2D fingerprints). Cryomagnetic measurements show that these complexes behave as quasi-magnetically isolated spin doublets with weak antiferromagnetic interactions at low temperatures. The magnetic behavior of Re(II) was modeled by the influence of the ligand field, tetragonal distortion, spin-orbit coupling, and covalence effects. In addition, the antiferromagnetic exchange coupling was correlated to the nature of the intermolecular interactions.

To date, there are a few technologies for the development of noncompetitive immunoassays for small molecules, the most common of which relies on the use of anti-immunocomplex antibodies. This approach is laborious, case specific, and relies upon monoclonal antibody technology for its implementation. We recently demonstrated that, in the case of monoclonal antibody-based immunoassays, short peptide loops isolated from phage display libraries can be used as substitutes of the anti-immunocomplex antibodies for noncompetitive immunodetection of small molecules. The aim of this work was to demonstrate that such phage ligands can be isolated even when the selector antibodies are polyclonal in nature. Using phenoxybenzoic acid (PBA), a major pyrethroid metabolite, as a model system, we isolated the CFNGKDWLYC peptide after panning a cyclic peptide library on the PBA/anti-PBA immunocomplex. The sensitivity of the noncompetitive enzyme-linked immunosorbent assay (ELISA) setup with this peptide was 5-fold (heterologous) or 400-fold (homologous) higher than that of the competitive assay setup with the same antibody. Phage anti-inmunocomplex assay (PHAIA) was also easily adapted into a rapid and highly sensitive dipstick assay. The method not only provides a positive readout but also constitutes a major shortcut in the development of sensitive polyclonal-based assays, avoiding the need of synthesizing heterologous competing haptens.

The impact of the use of herbicides in agriculture can be minimized by compliance with good management practices that reduce the amount used and their release into the environment. Simple tests that provide real time on-site information about these chemicals are a major aid for these programs. In this work, we show that phage anti-immunocomplex assay (PHAIA), a method that uses phage-borne peptides to detect the formation of antibody-analyte immunocomplexes, is an advantageous technology to produce such field tests. A monoclonal antibody to the herbicide clomazone was raised and used in the development of conventional competitive and noncompetitive PHAIA immunoassays. The sensitivity attained with the PHAIA format was over 10 times higher than that of the competitive format. The cross-reactivity of the two methods was also compared using structurally related compounds, and we observed that the two-site binding of PHAIA "double-checks" the recognition of the analyte, thereby increasing the assay specificity. The positive readout of the noncompetitive PHAIA method allowed adaptation of the assay into a rapid and simple format where as little as 0.4 ng/mL clomazone (more than 10-fold lower than the proposed standard) in water samples from a rice field could be easily detected by simple visual inspection.

Analytical pyrolysis (Py–GC/MS) at 500◦C was applied to study wood composition of Eucalyptus species(Eucalyptus grandis, Eucalyptus dunnii and Eucalyptus benthamii) which are relevant for pulping inUruguay. Multivariate data treatments mainly principal component analysis and discriminant analy-sis were used to explore differences between the original wood cultivars. Multivariate analyses withautomatic backwards variable selection indicated that simplified methoxyphenol patterns (up to 10compounds) are sufficient for wood discrimination in terms of species and geographical origin butalso with purposes of forecasting the ease of delignification of the resulting pulps measured as activealkali. No additional chemotaxonomical accuracy was achieved when the data sets were enlarged withcarbohydrate-derived products. On the other side, discriminant or forecasting models (partial leastsquares and multiple linear regression) were much less significant when based on individual diag-nostic compounds, groups of compounds, or the classical syringyl-to-guaiacyl (S/G) ratio. Principalcomponent analysis indicated that the variability in lignin composition due to bioclimatic variations(spatial replications) was more significant than that due to phylogenetic differences (species andcultivars).

Biological control may benefit from the behavioral manipulation of natural enemies using volatile organic compounds (VOCs). Among these, herbivore-induced plant volatiles (HIPVs) provide potential tools for attracting or retaining predators and parasitoids of insect pests. This work aimed to characterize the VOCs emitted by pear plants in response to attack by Cacopsylla bidens (Hemiptera: Psyllidae), a major pest in pear orchards, to compare these with VOCs induced by a leaf chewing insect, Argyrotaenia sphaleropa (Lepidoptera: Tortricidae), and to evaluate the behavioral response of Chrysoperla externa (Neuroptera: Chrysopidae) to HIPVs from pear plants damaged by either herbivore. The results demonstrated that plants damaged by the pear psylla emitted VOC blends with increased amounts of aliphatic aldehydes. Leafroller damage resulted in increased amounts of benzeneacetonitrile, (E)-4,8-dimethylnona-1,3,7-triene, β-ocimene and caryophyllene. In olfactometer bioassays, larvae of C. externa were attracted to herbivore-damaged plants when contrasted with undamaged plants. When plant odors from psylla-damaged were contrasted with those of leafroller-damaged plants, C.externa preferred the former, also showing shorter response lag-times and higher response rates when psylla-damaged plants were present. Our results suggest that pear plants respond to herbivory by modifying their volatile profile, and that psylla-induced volatiles may be used as prey-specific chemical cues by chrysopid larvae. Our study is the first to report HIPVs in pear plants attacked by C. bidens, as well as the attraction of C. externa to psyllid-induced volatiles.

We have previously reported that alfa-dictalediol monoacetate(1)a new diterpene possessing a novel carbon skeletonwas the major metabolite of Dictyota dichotoma(Huds). Lamour collected off GüimarTenerifeCanary Islands. From the remaining metaboliteswe isolated a diterpene triol to which we tentatively assigned the structure 2. Our interest in this compound was rekindled when Ishitsuka et al. described a related diterpene monoacetate 3 from Pachydictyon coriaceum. We have subsequently confirmed the relationship between the two compounds by comparison of spectral data for samples of the triacetate 4.

The iboga alkaloids scaffold shows great potential as a pharmacophore in drug candidates for the treatment of neuropsychiatric disorders. Thus, the study of the reactivity of this type of motif is particularly useful for the generation of new analogs suitable for medicinal chemistry goals. In this article, we analyzed the oxidation pattern of ibogaine and voacangine using dioxygen, peroxo compounds, and iodine as oxidizing agents. Special focus was placed on the study of the regio- and stereochemistry of the oxidation processes according to the oxidative agent and starting material. We found that the C16-carboxymethyl ester present in voacangine stabilizes the whole molecule toward oxidation in comparison to ibogaine, especially in the indole ring, where 7- hydroxy- or 7-peroxy-indolenines can be obtained as oxidation products. Nevertheless, the ester moiety enhances the reactivity of the isoquinuclidinic nitrogen to afford C3-oxidized products through a regioselective iminium formation. This differential reactivity between ibogaine and voacangine was rationalized using computational DFT calculations. In addition, using qualitative and quantitative NMR experiments combined with theoretical calculations, the absolute stereochemistry at C7 in the 7- hydroxyindolenine of voacangine was revised to be S, which corrects previous reports proposing an R configuration.

<p>Hanseniaspora vineae is a non-Saccharomyces yeast that has a powerful impact on the sensory profile of wines. Its effect on the aromatic profile of non-aromatic grape varieties, such as Albillo Mayor (Vitis vinifera, L.), during vinification is a useful biotechnology to improve sensory complexity. Fermentation in steel barrels using Hanseniaspora vineae and sequential inoculation with Saccharomyces cerevisiae have been used to study the formation of terpenes and cell lysis in the production of Albillo white wines. The GC-MS analysis profile shows a significant effect of H. vineae fermentation on the contents of terpenes (3), mainly in linalool (>3), -citronellol (>4), geraniol (>2) and-terpineol (2). The contents of several polyoxygenated terpenes and some volatile phenols with a spicy aroma were increased during fermentation. In summary, Hanseniaspora vineae releases a large number of cell wall polysaccharides during fermentation that affect wine palatability and structure.<br />Hanseniaspora vineae is a powerful bio-tool to enhance the fruitiness, floral notes and freshness in non-aromatic white varieties.</p>

SARS-CoV-2 surveillance of viral populations in wastewater samples is recognized as a useful tool for monitoring epidemic waves and boosting health preparedness. Next genera tion sequencing of viral RNA isolated from wastewater is a convenient and cost-effective strategy to understand the molecular epidemiology of SARS-CoV-2 and provide insights on the population dynamics of viral variants at the community level. However, in low- and mid dle-income countries, isolated groups have performed wastewater monitoring and data has not been extensively shared in the scientific community. Here we report the results of moni toring the co-circulation and abundance of variants of concern (VOCs) of SARS-CoV-2 in Uruguay, a small country in Latin America, between November 2020—July 2021 using wastewater surveillance. RNA isolated from wastewater was characterized by targeted sequencing of the Receptor Binding Domain region within the spike gene. Two computa tional approaches were used to track the viral variants. The results of the wastewater analy sis showed the transition in the overall predominance of viral variants in wastewater from No-VOCs to successive VOCs, in agreement with clinical surveillance from sequencing of nasal swabs. The mutations K417T, E484K and N501Y, that characterize the Gamma VOC, were detected as early as December 2020, several weeks before the first clinical case was reported. Interestingly, a non-synonymous mutation described in the Delta VOC, L452R, was detected at a very low frequency since April 2021 when using a recently described sequence analysis tool (SAM Refiner). Wastewater NGS-based surveillance of SARS-CoV-2 is a reliable and complementary tool for monitoring the introduction and preva lence of VOCs at a community level allowing early public health decisions.This approach allows the tracking of symptomatic and asymptomatic individuals, who are generally under reported in countries with limited clinical testing capacity. Our results suggests that waste water-based epidemiology can contribute to improving public health responses in low- and middle-income countries.

Among small cellular molecules, certain inositol phosphates (InsPs) excel at complexing Fe3+ in a safe manner in vitro. In particular Ins(1,2,3)P3 is probably an important chelator of iron cations not strongly bound to proteins, which avoids iron redox cycling and the associated production of free radicals. In this work we report the first thermodynamic data of the Fe3+/Fe2+ redox potential in the presence of Ins(1,2,3)P3 which demonstrate the capability of this ligand to inhibit iron(III) reduction in the intracellular media. The Fe3+–Ins(1,2,3)P3 speciation was also studied by 31P NMR measurements, focusing on the structure of the complexes. The work is completed by theoretical calculations which lead us to predict the geometries of the species at different pH values and their relative stabilities. The influence of the ring conformation in the Fe3+ complexed forms is also discussed.

Equine chorionic gonadotrophin (eCG) is a hormone of practical value in veterinary medicine and animal production. Here we report a novel preparation procedure based on its direct adsorption onto anionic-exchange resins in a batch-wise mode. The active plasma is previously conditioned to reduce pH and ionic strength to required levels. After the adsorption stage, a 90% recovery of the initial eCG is achieved, with a concentration factor of about 50 and an enrichment factor around 500, with high preservation of biological activity. Further purification is carried out by cation-exchange column chromatography. The recovery for the whole process is higher than 70%, and the final potency of the preparation is close to 4000 IU/mg. The process is well suited for its application to the industrial scale

<p>Baccharis dracunculifolia DC. (Asteraceae) is an aromatic plant species widely distributed across and along South American countries where it grows in different environments, even being considered, in some cases, as a hard to control weed. Its essential oil is highly appreciated in the fragrance/perfumery industry by its woody, foral, and green notes; mainly attributed to (E)-nerolidol and other sesquiterpene alcohols. Recent research has demonstrated a wide range of applications for the essential oil besides its aromatic properties. Although B. dracunculifolia cannot be considered as a medicinal plant for most of the indigenous communities in South America (except for Brazil), modern pharmacological research has demonstrated its high potential for the development of new therapeutic drugs, mainly with antiulcer, anti-infammatory, antiparasitic and anticancer effect. The special literature regarding B. dracunculifolia is profuse and thus, this chapter attempts to summarize the principal knowledge about this species.</p>

Ibogaine is a potent psychedelic alkaloid that has been the focus of intense research because of its intriguing anti-addictive properties. According to anecdotic reports, ibogaine has been originally classified as an oneirogenic psychedelic; i.e., induces a dream-like cognitive activity while awake. However, the effects of ibogaine administration on wakefulness (W) and sleep have not been thoroughly assessed. The main aim of our study was to characterize the acute effects of ibogaine administration on W and sleep. For this purpose, polysomnographic recordings on chronically prepared rats were performed in the light phase during 6 h. Animals were treated with ibogaine (20 and 40 mg/kg) or vehicle, immediately before the beginning of the recordings. Furthermore, in order to evaluate associated motor behaviors during the W period, a different group of animals was tested for 2 h after ibogaine treatment on an open field with video-tracking software. Compared to control, animals treated with ibogaine showed an increase in time spent in W. This effect was accompanied by a decrease in slow wave sleep (SWS) and rapid-eye movements (REM) sleep time. REM sleep latency was significantly increased in animals treated with the higher ibogaine dose. While the effects on W and SWS were observed during the first 2 h of recordings, the decrement in REM sleep time was observed throughout the recording time. Accordingly, ibogaine treatment with the lower dose promoted an increase on locomotion, while tremor and flat body posture were observed only with the higher dose in a time-dependent manner. In contrast, head shake response, a behavior which has been associated in rats with the 5HT2A receptor activation by hallucinogens, was not modified. We conclude that ibogaine promotes a waking state that is accompanied by a robust and long-lasting REM sleep suppression. In addition, it produces a dose-dependent unusual motor profile along with other serotonin-related behaviors. Since ibogaine is metabolized to produce noribogaine, further experiments are needed to elucidate if the metabolite and/or the parent drug produced these effects.
