We present the first worldwide study on the apple (Malus × domestica) fruit microbiome that examines questions regarding the composition and the assembly of microbial communities on and in apple fruit. Results revealed that the composition and structure of the fungal and bacterial communities associated with apple fruit vary and are highly dependent on geographical location. The study also confirmed that the spatial variation in the fungal and bacterial composition of different fruit tissues exists at a global level. Fungal diversity varied significantly in fruit harvested in different geographical locations and suggests a potential link between location and the type and rate of postharvest diseases that develop in each country. The global core microbiome of apple fruit was represented by several beneficial microbial taxa and accounted for a large fraction of the fruit microbial community. The study provides foundational information about the apple fruit microbiome that can be utilized for the development of novel approaches for the management of fruit quality and safety, as well as for reducing losses due to the establishment and proliferation of postharvest pathogens. It also lays the groundwork for studying the complex microbial interactions that occur on apple fruit surfaces. Over the last few years, interest in collecting an increasing variety of information in order to acquire a deeper and more comprehensive understanding of the consumer experience with products has steadily grown. The present study was designed to develop a methodological approach to explore the Global Profile of products within a category. A product-specific questionnaire was developed using one-on-one online interviews conducted with a modified version of the Repertory Grid Method (RGM) combined with semiotic analysis, EmoSemio, extended to cover all the main dimensions of product experience of the category of processed tomato. A 96 item questionnaire - including liking, sensory properties, emotions, emotional and functional conceptualisations and contextual appropriateness measures - was developed and employed in a home use test with 196 consumers who evaluated 9 products (one per day). All the statements were found to discriminate between products with the exception of one. A Multiple Factor Analysis showed that emotions were highly correlated with functional/emotional conceptualisations, while sensory properties were mainly related to emotional conceptualisations (memories) and uses in the recipes. The information provided by the simultaneous collection of these different dimensions allows to go beyond liking and may be used in product development and innovation in order to better understand the consumer experience of a product. Las glutation transferasas (GSTs) son una superfamilia de protefnas multifuncionales que cumplen funciones esenciales en la detoxificacion de compuestos tanto endogenos como exogenos. Las GSTs pueden ser c1asificadas en cuatro familias, siendo las citosolicas las estudiadas en mayor profundidad. En parasites helmintos se identificaron varias GSTs citosolicas con roles clave para su sobrevida en el hospedero. Asimismo, no solo la actividad detoxificante es importante sino que las GSTs, particularmente las de c1ase Sigma, estan implicadas en procesos de inmunomodulacion a traves de su actividad prostaglandina sintasa. En este sentido, es de nuestro interes caracterizar este grupo de enzimas en el parasito Echinococcus granulosus. Particularmente, este trabajo describe la identificaci6n y c1asificacion de • una nueva GST de E. granulosus (EgGST2), 105 avances en la caracterizacion cinetica de EgGSTl tipificada previamente y por ultimo, 105 estudios de expresion de las tres EgGSTs identificadas al momento. A partir del transcriptoma de larva de E. granulosus publicado se obtuvo la secuencia de EgGST2. En la Seccion I se muestra la tipificacion de EgGST2. Los estudios de funcion catalftica ubican a EgGST2 indudablemente como miembro de la c1ase Sigma. Por otro lade, se presenta el trabajo orientado a la produccion de la protefna recombinante enzimaticamente activa y finalmente se muestra que la protefna se expresa en protoescolex de E. granulosus. La enzima EgGSTl fue previa mente estudiada por nuestro grupo de trabajo donde se demostro que pertenece a la c1ase Mu. Por otro lado, se realizaron estudios bioqufmicos de la enzima recombinante, donde se analizo el panel de actividades que presenta asf como sus parametros enzimaticos. En la Seccion 1\ se presenta la profundizacion de la caracterizacion cinetica de EgGST1. En este contexto, se realizaron estudios de velocidades iniciales y de inhibicion por producto, cuyos resultados muestran que EgGSTl sigue un mecanisme secuencial aleatorio concordante con las GSTs de mamffero de c1ase Mu descritas. Por otro lado, se determine la constante de disociacion para cada uno de 105 sustratos por analisis de union en equilibrio. Finalmente, experimentos de pH permitieron acceder al pKa del tiolato del GSH en el complejo binario enzima - GSH, siendo este resultado acorde con 10 reportado para las GST que presentan una Tyr catalftica. En ultimo lugar, en la Seccion III, se exponen 105 estudios de expresion de las tres GST identificadas al momento en E. granulosus. Primero, se presenta el analisis bioinformatico de las regiones 5' flanqueantes de 105 tres genes donde se identificaron elementos relacionados con la regulacion de enzimas detoxificantes. Por otro lado, se observo que las tres isoformas presentan una expresion diferencial a nivel del ARN • mensajero cuando 105 protoescolex son expuestos a condiciones de estres oxidativo 0 xenobioticos in vitro. Finalmente, se estudio la expresion de las EgGSTs en el contexto de la infeccion secunda ria murina temprana. EI conjunto de estes resultados pretende contribuir al entendimiento de la interrelacion hospedero - parasite a partir del entendimiento del rol biologico de este versatil grupo de enzimas detoxificantes. Helminths express various carbohydrate-containing glycoconjugates on their surface, and they release glycan-rich excretion/secretion products that can be very important in their life cycles, infection and pathology. Recent evidence suggests that parasite glycoconjugates could play a role in the evasion of the immune response, leading to a modified Th2-polarized immune response that favors parasite survival in the host. Nevertheless, there is limited information about the nature or function of glycans produced by the trematode Fasciola hepatica, the causative agent of fasciolosis. In this paper, we investigate whether glycosylated molecules from F. hepatica participate in the modulation of host immunity. We also focus on dendritic cells, since they are an important target of immune-modulation by helminths, affecting their activity or function. Our results indicate that glycans from F. hepatica promote the production of IL-4 and IL-10, suppressing IFNγ production. During infection, this parasite is able to induce a semi-mature phenotype of DCs expressing low levels of MHCII and secrete IL-10. Furthermore, we show that parasite glycoconjugates mediate the modulation of LPS-induced maturation of DCs since their oxidation restores the capacity of LPS-treated DCs to secrete high levels of the pro-inflammatory cytokines IL-6 and IL-12/23p40 and low levels of the anti-inflammatory cytokine IL-10. Inhibition assays using carbohydrates suggest that the immune-modulation is mediated, at least in part, by the recognition of a mannose specific-CLR that signals by recruiting the phosphatase Php2. The results presented here contribute to the understanding of the role of parasite glycosylated molecules in the modulation of the host immunity and might be useful in the design of vaccines against fasciolosis. The effect of H2O2-induced oxidative stress on the viability of the yeast antagonist, Cystofilobasidium infirmominiatum, as well as the effect of exogenous glycine betaine (GB) on yeast viability under oxidative stress, was determined. GB treatment improved the tolerance of C. infirmominiatum to oxidative stress. Compared to untreated control yeast cells, GB-treated cells showed less accumulation of reactive oxygen species (ROS) and a lower level of protein oxidation in response to oxidative stress. Additionally, GB-treated yeast exhibited greater biocontrol activity against Penicillium expansum and a faster growth in wounds of apple fruits stored at 25 °C compared to the performance of untreated yeast. The activities of antioxidant enzymes, including catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPX) of C. infirmominiatum were elevated by GB treatment. Plant domestication by selective breeding may reduce plant chemical defense in favor of growth. However, few studies have simultaneously studied the defensive chemistry of cultivated plants and their wild congeners in connection to herbivore susceptibility. We compared the constitutive glycoalkaloids (GAs) of cultivated potato, Solanum tuberosum, and a wild congener, S. commersonii, by liquid chromatography coupled to mass spectrometry. We also determined the major herbivores present on the two species in field plots, and tested their preference for the plants and their isolated GAs in two-choice bioassays. Solanum commersonii had a different GA profile and higher concentrations than S. tuberosum. In the field, S. tuberosum was mostly attacked by the generalist aphids Myzus persicae and Macrosiphum euphorbiae, and by the specialist flea beetle Epitrix argentinensis. In contrast, the most common herbivore on S. commersonii was the specialist sawfly Tequus sp. Defoliation levels were higher on the wild species, probably due to the chewing feeding behavior of Tequus sp. As seen in the field, M. persicae and E. argentinensis preferred leaf disks of the cultivated plant, while Tequus sp. preferred those of the wild one. Congruently, GAs from S. commersonii were avoided by M. persicae and preferred by Tequus sp. The potato aphid performed well on both species and was not deterred by S. commersonii GAs. These observations suggest that different GA profiles explain the feeding preferences of the different herbivores, and that domestication has altered the defensive capacity of S. tuberosum. However, the wild relative is still subject to severe defoliation by a specialist herbivore that may cue on the GAs. Tannins are polyphenolic compounds extensively present in plants and used by food industry as processing aids. Due to the heterogeneity of plant sources, actions involved in food processing and tannin commercial costs can be different. In the last years different approaches aimed at correctly identifying the tannin botanical origin have been developed, in order to satisfy the industry’s request to verify product labels. This work aimed to define the glycosidic simple phenolic profile of a large selection of monovarietal commercial tannins of different origin, using a high-resolution untargeted approach. Using accurate mass, isotopic pattern and MS/MS fragmentation, 167 precursors, 89 as monoglycosylated and 78 as diglycosylated derivatives were tentatively identified in tannins, validating the untargeted approach with 3 custom-synthesized glycosidic precursors. En esta tesis se estudi6 la aplicaci6n de la granulaci6n por fusi6n como alternativa frente a los procesos por via humeda y via seca comunmente utilizados en la industria farmaceutica. Se investigaron excipientes de uso frecuente en formas farmaceuticas s61idas de administraci6n oral y equipos adecuados entre los de uso habitual en la industria farmaceutica local. Se disenaron dos procesos de granulaci6n por fusi6n que se diferencian en la fuerza de corte impartida allecho s6lido a granular durante la etapa de fusi6n: 1. proceso en lecho m6vil, que utiliza la tecnologia mas clasicamente empleada para granulaci6n por via humeda en la industria farmaceutica local: mezcladores de velocidad media 0 fuerza de corte media, en este caso con camisa, para formar masas aglomeradas, seguido de equipos formadores de granulos, como los granuladores oscilantes. Impone movimiento de fuerza de corte media al s6lido. 2. proceso en lecho estatico, que emplea un secador de cabina para la primera operaci6n tecnol6gica, seguido de una regularizaci6n por tamizado manual para terminar la granulaci6n. No imparte movimiento al s6lido. Los procesos fueron aplicados al s6lido inactivo almid6n de maiz, asi como a los principios activos carbamacepina y ketoprofeno. Los granulados fueron caracterizados en relaci6n a sus propiedades fisicomecanicas, incluyendo la aptitud para la compresi6n. Los comprimidos asi obtenidos tambien fueron caracterizados en relaci6n a sus propiedades fisicomecanicas. En el caso de los comprimidos conteniendo principios activos, la caracterizaci6n incluy6 ademas, las propiedades fisicoquimicas, en particular la liberaci6n de los activos. Entre los estudios de compresi6n se evalu6 tambien, la capacidad de estos procesos de granulaci6n por fusi6n para generar granulados inactivos aptos para ser empleados como excipientes de compresi6n directa. Para ello se fabricaron comprimidos de liberaci6n inmediata con la droga modele hidroclorotiazida. Los agentes granulantes seleccionados para lIevar a cabo las granulaciones por fusi6n mediante los procesos disenados comprenden excipientes tanto hidr6filos como hidr6fobos, usados solos 0 en combinaci6n: polietilenglicol 6000, povidona K30, palmitoestearato de glicerilo, acido estearico y monoestearato de glicerilo. A 10 largo del trabajo se emplearon herramientas estadisticas multivariadas, en concreto el analisis de componentes principales (peA) y el analisis de cluster, de forma de evaluar su uso para el estudio de los resultados de las propiedades farmacotecnicas ensayadas. Mediante el empleo de estas herramientas se busc6 contar con tratamientos de datos que facilitaran el analisis del importante numero de variables y la gran cantidad de datos generados normalmente por la cantidad de muestras que se ensayan en la evaluaci6n de granulados y comprimidos. Los resultados del presente trabajo permiten concluir que los dos procesos de granulaci6n por fusi6n (proceso en lecho m6vil y proceso en lecho estatico): J.- son capaces de dar productos de calidad farmacotecnica aceptable para varias materias primas de uso farmaceutico, mejorando el flujo y la compresibilidad de las mismas. J.- son sencillos, emplean equipos simples y con uso bien establecido en la industria farmaceutica, requieren control de pocas variables durante la producci6n y anticipan un escalado y una validaci6n de proceso simples. J.- pudieron utilizarse para producir comprimidos de Iiberaci6n inmediata con carbamacepina y con ketoprofeno, asi como de liberaci6n prolongada con ketoprofeno. J.- son capaces de dar granulados para ser usados como diluyentes de compresi6n directa. Por otra parte, las herramientas multivariadas seleccionadas para la evaluaci6n de los productos obtenidos probaron ser de utilidad para la interpretaci6n de los datos recogidos, facilitando el estudio de las propiedades de los granulados y la selecci6n de procesos de producci6n mas eficientes. Granulomas are responses to persistent nonliving bodies or pathogens, centrally featuring specialized macrophage forms called epithelioid and multinucleated giant cells. The larval stages of the cestode parasites of the Taeniidae family (Taenia, Echinococcus) develop for years in fixed tissue sites in mammals. In consequence, they are targets of granulomatous responses. The information on tissue responses to larval taeniids is fragmented among host and parasite species and scattered over many decades. We attempt to draw an integrated picture of these responses in solid tissues. The intensity of inflammation around live parasites spans a spectrum from minimal to high, parasite vitality correlating with low inflammation. The low end of the inflammatory spectrum features collagen capsules proximal to the parasites and moderate distal infiltration. The middle of the spectrum is dominated by classical granulomatous responses, whereas the high end features massive eosinophil invasions. Across the range of parasite species, much observational evidence suggests that eosinophils are highly effective at killing larval taeniids in solid tissues, before and during chronic granulomatous responses. The evidence available also suggests that these parasites are adapted to inhibit host granulomatous responses, in part through the exacerbation of host regulatory mechanisms including regulatory T cells and TGF-β. The impact of native grape microbiota on wine identity is not completely elucidated. In this work, we explored this issue using microbial communities prepared from V. vinifera (Malbec) and V. labrusca (Isabella) grapes as starters for the fermentation of Malbec grape juice. Analyses of early-stage reconstituted homologous and heterologous fermentations revealed that each Vitis-specific microbial starter influenced the volatile profiles of the resulting fermented grape juice. Changes in the relative abundance of the three main non-Saccharomyces yeast species identified (Hanseniaspora uvarum, Hanseniaspora opuntiae, and Starmerella bacillaris) were observed throughout these fermentations, which could explain the differences in their volatile profiles. Growth parameters determined for yeast isolates from these species, obtained from the Isabella and Malbec microbiotas, showed no growth differences in either Malbec or Isabella grape juices.
