Entire seeds of Chenopodium quinoa Willd are a rich protein source and are also well-known for their high saponin content. Due to their amphiphily quinoa saponins are able to form intricate micellar aggregates in aqueous media. In this paper we study the aggregates formed by self-association of these compounds from two quinoa saponin fractions (FQ70 and FQ90) as well as several distinctive nanostructures obtained after their complexation with different ratios of cholesterol (CHOL) and phosphatidylcholine (PC). The FQ70 and FQ90 fractions were obtained by reversed-phase preparative chromatography. The structural features of their resulting aggregates were determined by Dynamic Light Scattering (DLS) and Transmission Electron Microscopy (TEM). Novel nanosized spherical vesicles formed by self-association with mean diameter about 100-200 nm were observed in FQ70 aqueous solutions whereas worm-like micelles an approximate width of 20 nm were detected in FQ90 aqueous solutions. Under experimental conditions similar to those reported for the preparation of Quillaja saponaria ISCOM matrices, tubular and ring-like micelles arose from FQ70:CHOL:PC and FQ90:CHOL:PC formulations, respectively. However, under these conditions no cage-like ISCOM matrices were observed. The saponin composition of FQ70 and FQ90 seems to determine the nanosized structures viewed by TEM. Phytolaccagenic acid, predominant in FQ70 and FQ90 fractions, is accountable for the formation of the nanosized vesicles and tubular structures observed by TEM in the aqueous solutions of both samples. Conversely, ring-like micelles observed in FQ90:CHOL:PC complexes can be attributed to the presence of less polar saponins present in FQ90, in particular those derived from oleanolic acid.<br /><br />

Geogenic As in groundwater has been recently studied in different aquifers of Uruguay. However, no background epidemiology studies are available regarding environmental population exposure. As a consequence, it was necessary to conduct scientific research in order to study arsenic as an environmental health issue in the country. This research needs have resulted in joint studies of experts from both the geosciences and biosciences. The groups have coordinated studies to assess the risks of exposure to environmental arsenic in Uruguay with common research objectives and, now, this is a matter of common interest through a multidisciplinary approach. The aim of this work is to present a review of these studies conducted in Uruguay.

A saponin fraction extracted from Quillaja brasiliensis leaves (QB-90) and a semi-purified aqueous extract (AE) were evaluated as adjuvants in a bovine viral diarrhea virus (BVDV) vaccine in mice. Animals were immunized on days 0 and 14 with antigen plus either QB-90 or AE or an oil-adjuvanted vaccine. Two-weeks after boosting, antibodies were measured by ELISA; cellular immunity was evaluated by DTH, lymphoproliferation, cytokine release and single cell IFN-γ production. Serum anti-BVDV IgG, IgG1 and IgG2b were significantly increased in QB-90- and AE-adjuvanted vaccines. A robust DTH response, increased splenocyte proliferation, Th1-type cytokines and enhanced production of IFN-γ by CD4(+) and CD8(+) T lymphocytes were detected in mice that received QB-90-adjuvanted vaccine. The AE-adjuvanted preparation stimulated humoral responses but not cellular immune responses. These findings reveal that QB-90 is capable of stimulating both cellular and humoral immune responses when used as adjuvant.

Herein, we investigate the structural, electronic and mechanical properties of zigzag graphene nanoribbons in the presence of stress by applying density functional theory within the GGA-PBE (generalized gradient approximation-Perdew–Burke–Ernzerhof) approximation. The uniaxial stress is applied along the periodic direction, allowing a unitary deformation in the range of ±0.02%. The mechanical properties show a linear response within that range while a nonlinear dependence is found for higher strain. The most relevant results indicate that Young’s modulus is considerable higher than those determined for graphene and carbon nanotubes. The geometrical reconstruction of the C–C bonds at the edges hardens the nanostructure. The features of the electronic structure are not sensitive to strain in this linear elastic regime, suggesting the potential for using carbon nanostructures in nano-electronic devices in the near future.

<p>A bioequivalence study in 16 Caucasian healthy volunteers (eight male, eight female), comparing plasma drug concentrations after a single oral dose of lopinavir and ritonavir (400 and 100 mg, respectively), was carried out following a two-period, two-sequence, two-treatment, randomized crossover design. Formulations were given 15 min after a moderate-fat breakfast in order to diminish both the intrinsic highlyvariable performance and the sex differences observed in bioequivalence trials under fasting conditions. Ninety percent confidence intervals for the Test/Reference (T/R) ratio of geometric means for area under concentration–time curve (AUC) and maximum concentration (CMAX), either for lopinavir or ritonavir, were within the range of 0.80–1.25. Coprandial administration of formulations not only reduced the number of subjects required for bioequivalence assessment, reducing both ethical and economic cost of the trial, but also the sex differences in the T/R ratio of means</p>

A summary of the resource.
This narrative review revises the scientific evidence of recent years on healthy eating in children and adolescents, making sense of promising avenues of action, from a food system perspective. A conceptual framework is provided to better understand how eating habits of children and adolescents are shaped to identify key multisectoral approaches that should be implemented to promote healthier diets. The following influencing factors are discussed: individual factors (physiological and psychological factors, food preferences and food literacy competencies), factors within the personal and socio-cultural food environments, external food environments, and the supply chain. In each section, the main barriers to healthy eating are briefly discussed focussing on how to overcome them. Finally, a discussion with recommendations of actions is provided, anchored in scientific knowledge, and transferable to the general public, industry, and policymakers. We highlight that multidisciplinary approaches are not enough, a systems approach, with a truly holistic view, is needed. Apart from introducing systemic changes, a variety of interventions can be implemented at different levels to foster healthier diets in children through fostering healthier and more sustainable food environments, facilitating pleasurable sensory experiences, increasing their food literacy, and enhancing their agency by empowering them to make better food related decisions. Acknowledging children as unique individuals is required, through interpersonal interactions, as well as their role in their environments. Actions should aim to enable children and adolescents as active participants within sustainable food systems, to support healthier dietary behaviours that can be sustained throughout life, impacting health at a societal level.

Durante este proyecto de tesis se ha desarrollado un novedoso soporte bifuncional para la inmovilizaci6n de proteinas, que permite combinar las ventajas de la uni6n sitio especifica con las de la inmovilizaci6n covalente multipuntual. EI mismo contiene: i) una pequena proporci6n de gropos tiol-reactivos, y ii) un alto numero de gropos ep6xidos. La optimizaci6n de su sintesis se llev6 a cabo utilizando ditiotreitol 0 sulfuro s6dico como agentes tiolantes, y resinas acrilicas (Eupergit C, EP-Sepabeads) como matrices epoxiactivadas. La inmovilizaci6n en este tipo de soportes es un proceso en dos etapas. Primero la proteina es inmovilizada en forma reversible a traves de un intercambio tiol-disulfuro entre los gropos tiol de la enzima y los gropos tiol-reactivos del soporte bajo condiciones suaves de temperatura (24°C) Y pH (7.0). Luego, en una segunda etapa los gropos ep6xido remanentes del soporte son capaces de formar uniones' irreversibles intramoleculares con los gropos nucleofilicos superficiales de la enzima (principalmente residuos de lisina), debido a su proximidad. Este mecanismo en dos etapas fue verificado por medio del estudio del proceso de inmovilizaci6n de proteinas modelo: i) que presentan naturalmente gropos tiol superficiales (~-galactosidasa de Escherichia coli), y ii) cuyos gropos tiol necesitan ser generados por reducci6n de disulfuros nativos (~-galactosidasa de Kluyveromices lactis) 0 par tiolaci6n con SPDP (penicilina G acilasa de E. coli y lipasa de Rhizomucor miehei). A su vez, estudios de inactivaci6n termica demostraron que las uniones adicionales multipuntuales que se establecen durante la segunda etapa de la inmovilizaci6n, son las responsables de un incremento en la estabilidad de las enzimas inmovilizadas con respecto alas enzimas solubles 0 alas inmovilizadas iInicamente a traves de enlaces disulfuros. Dado que la presencia de gropos sulfhidrilo en la superficie de la estructura proteica es una condici6n esencial para el uso de este nuevo soporte mixto, en el desarrollo de esta tesis se estudiaaran en forma complementaria dos estrategias para la generaci6n de gropos tiol: i) por reducci6n de puentes disulfuros, y ii) por mutagenesis dirigida. Es asi que por un lado, se evalu6 el uso de tiopraapil-agarosa, para la obtenci6n de gropos sulfhidrilo por reducci6n de disulfuros proteicos, como una altemativa al uso de reductores solubles. A su vez, con el fin de poder controlar totalmente la orientaci6n proteica sobre los soportes desarrollados, se estudi6 la viabilidad de introducir residuos de cisteina en la PGA de E. coli por medio de mutagenesis dirigida. Si bien los resultados obtenidos son muy preliminares, 10s mismos alientan a la realizaci6n de investigaciones que permitan estudiar el efecto que podria tener en la estabilidad enzimMica, el inmovilizar la PGA por distintas zonas de su superficie en los soportes desarrollados.

Este artículo presenta los resultados de un estudio sobre la producción de compuestos conjugados de una beta-galactosidasa neutra (lactasa), mediante su inmovilización en tiosulfinato-agarosa. Se alcanzaron rendimientos de sobre 90% y los productos obtenidos mostraron buena estabilidad frente a agentes sanitizantes. También, estos productos mostraron ser muy efectivos para la conversión de lactosa en soluciones tamponadas, en lactosuer, en permeatos de lactosuero y en leche. Se realizaron cuatro ensayos en un bioreactor de columna que funcionó en forma continua por diecisiete días en cada experiencia. En todos los casos se logró una conversión del orden del 90%. Después de nueve meses de almacenamiento adecuado, los conjugados mantuvieron su actividad enzimática, demostrando su alta estabilidad.

A summary of the resource.
In this manuscript, we present a new bifunctional support containing epoxide and thiol-reactive groups for its use in protein immobilization. In a first step, the proteins are reversibly immobilized by reaction of its thiol groups with the thiol-reactive groups of the support under mild experimental conditions (pH 7.0, 24 °C). Then, the remaining epoxides of the support can form irreversible bonds with nucleophile surface groups of the already immobilized protein in a rapid way. The partial derivatization of EP-Sepabeads (a commercial matrix containing 120 ímol epoxy groups/g drained support) was optimized, using dithiotreitol (DTT) as thiolating agent. It was possible to achieve a partial thiolation of the support proportional to the concentration of DTT used (3, 8, and 15 ímol SH groups/g wet support). The remaining epoxide content after the thiolation treatment was high (e.g., nearly 70% for the highest thiolation degree). High immobilization yields were obtained for the three model enzymes selected (60% for Penicillin G acylase, 65% and 100% for K. lactis and E. coli â-galactosidase, respectively). In all cases, no significant immobilization onto an unmodified epoxy support was found, thus demonstrating that the first step of attachment takes place through thiol-disulfide exchange reactions. In the case of the bifunctional support, progressive formation of enzymesupport attachments involving the epoxy groups was showed by the irreversible covalent attachment of the proteins on the support. The promotion of this multipoint covalent immobilization required long incubation periods at basic pH values.

The controlled and partial modification of epoxy groups of Eupergit C and EP-Sepabeads with sodium sulfide has permitted the preparation of thiol-epoxy supports. Their use allowed not only the specific immobilization of enzymes through their thiol groups via thiol– disulfide interchange, but also enzyme stabilization via multipoint covalent attachment. Penicillin G acylase (PGA) from Escherichia coli and lipase from Rhizomucor miehei were used as model enzymes. Both enzymes lacked exposed cysteine residues, but were introduced via chemical modification under very mild conditions. In the first moments of the immobilization, a certain percentage of immobilized protein could be released from the support by incubation with DTT; this confirms that the first step was via a thiol–disulfide interchange. Moreover, the promotion of some further epoxy-enzyme bonds was confirmed because no enzyme release was detected after some immobilization time by incubation with DTT. In the case of the heterodimeric PGA, it was possible to demonstrate the formation of at least one epoxy bond per enzyme subunit by analyzing with SDS-PAGE the supernatants obtained after boiling the enzyme derivatives in the presence of mercaptoethanol and SDS. Thermal inactivation studies showed that these multipoint enzyme-support attachments promoted an increase in the stability of the immobilized enzymes. In both cases, the stabilization factor was around 12–15-fold comparing optimal derivatives with their just-thiol immobilized counterparts.

Moreover, the temperature presented a dramatic effect on enzyme immobilization rate and different enzymes become immobilized at very different rates. Finally, it was not possible to detect immobilization of mono aminated compounds at neither alkaline nor neutral pH values. In fact, these supports may be used to purify enzymes due to these great differences in immobilization rate (e.g., glutamate racemase). These special features are not shared with other supports usually utilized for covalent immobilization of enzymes (e.g., glutaraldehyde, cyanogen bromide supports) that are able to immobilize proteins by just one very stable bond and suggesting an unique mechanism for glyoxyl agarose supports. In fact, all these results suggested that each individual amino-glyoxyl bond is very weak. Therefore, the first immobilization of proteins on supports activated with glyoxyl groups, in absence of reducing reagents, needs the simultaneous establishment of, at least, two attachments between the protein and the support. This mechanism promotes that proteins become immobilized by the area/s where the highest lysine residues density is located and explain the high enzyme stabilization usually achieved by using this immobilization technique.

The larval stage of Echinococcus granulosus causes the chronic infection known as cystic echinococcosis, deploying strong inhibitory mechanisms on host immune responses. Using experimental intraperitoneal infection in C57BL/6 mice, we carried out an in-depth analysis of the local changes in macrophage populations associated with chronic infection. In addition, we analyzed T cells and relevant soluble mediators. Infected animals showed an increase in local cell numbers, mostly accounted for by eosinophils, T cells, and macrophages. Within macrophage populations, the largest increases in cell numbers corresponded to resident large peritoneal macrophages (LPM). Monocyte recruitment appeared to be active, as judged by the increased number of monocytes and cells in the process of differentiation towards LPM, including small (SPM) and converting peritoneal macrophages (CPM). In contrast, we found no evidence of macrophage proliferation. Infection induced the expression of M2 markers in SPM, CPM, and LPM. It also enhanced the expression of the co-inhibitor PD-L1 in LPM, SPM, and CPM and induced the co-inhibitor PD-L2 in SPM and CPM. Therefore, local macrophages acquire M2-like phenotypes with probable suppressive capacities. Regarding T cells, infection induced an increase in the percentage of CD4+ cells that are PD-1+, which represent a potential target of suppression by PD-L1+/PD-L2+ macrophages. In possible agreement, CD4+ T cells from infected animals showed blunted proliferative responses to in vitro stimulation with anti-CD3. Further evidence of immune suppression in the parasite vicinity arose from the observation of an expansion in FoxP3+ CD4+ regulatory T cells and increases in the local concentrations of the anti-inflammatory cytokines TGF-β and IL-1Ra.

A summary of the resource.
Cystic echinococcosis is caused by the tissue-dwelling larva (hydatid) of Echinococcus granulosus sensu lato. A salient feature is that this larva is protected by the acellular laminated layer (LL). As the parasite grows, the LL sheds abundant particles that can accumulate in the parasite's vicinity. The potential of LL particles to induce inflammation in vivo has not been specifically analysed. It is not known how each of its two major components, namely highly glycosylated mucins and calcium inositol hexakisphosphate (InsP6) deposits, impacts inflammation induced by the LL as a whole. In this work, we show that LL particles injected intraperitoneally cause infiltration of eosinophils, neutrophils and monocytes/macrophages as well as the disappearance of resident (large peritoneal) macrophages. Strikingly, the absence of calcium InsP6 enhanced the recruitment of all the inflammatory cell types analysed. In contrast, oxidation of the mucin carbohydrates caused decreased recruitment of neutrophils. The carbohydrate-oxidised particles caused cell influx nonetheless, which may be explained by possible receptor-independent effects of LL particles on innate immune cells, as suggested by previous works from our group. In summary, LL particles can induce acute inflammatory cell recruitment partly dependent on its mucin glycans, and this recruitment is attenuated by the calcium InsP6 component.

Oil shales from Mangrullo Formation (Uruguay) was assessed as a potential fuel for energy production. The assessment is based on a comprehensive characterization of the material from a thermal perspective. Co-combustion of this type of fuel with biomass waste can enhance the combustion performance and decrease hazardous gas emissions. The low heating value (3.2 MJ kg−1) of the Uruguayan oil shale indicates that it could be mixed with biomass to obtain better results. Stratified sampling of drill cores was used to obtain a representative sample. The morphological and structural characteristics of the oil shale were studied by X-ray diffraction and scanning electron microscopy. Chemical composition (hydrocarbons and minerals) of oil shales was investigated by X-ray diffraction, X-ray fluorescence, proximate and ultimate analysis and Fourier transform infrared spectroscopy. Rock Eval analysis was performed to measure richness and maturity of the Uruguayan oil shale, the results of the TOC content (8.93%) and the hydrogen index (525 mg HC/ g TOC) indicated that it constitutes an excellent source rock. The kinetic of the combustion and thermal properties of oil shale were studied. The activation energy of the different reaction stages was calculated using the Flynn-Wall-Ozawa model, then we obtained the pre-exponencial factor and the reaction order optimizing the theoretical model using the experimental data. Results indicated that the combustion of the oil shale exhibits multiple reaction stages with activations energies that varies between 152.2 and 316.4 kJ mol−1.

The bronze bug, Thaumastocoris peregrinus Carpintero & Dellapé (Heteroptera: Thaumastocoridae), is an exotic emerging pest in Eucalyptus commercial forests in South America, Africa, and southern Europe. Information on the chemical communication system and reproductive ecology of this insect is scant, and it may be relevant for designing management strategies for eucalypt plantations. Males emit large amounts of 3-methyl-2-butenyl butyrate, which attracts conspecific adult males but not females. To learn more about the biological function of this putative male-produced pheromone, we quantified this compound in volatile emissions collected from males, females, and couples, in three 4-h collecting periods during the morning, afternoon, and night of a single 24-h cycle. Our results showed that virgin males emit 3-methyl-2-butenyl butyrate in a diel time pattern, with an almost sevenfold difference between the afternoon emission peak compared to morning or night hours. In addition, we show that in the presence of females, males emit the compound in the same amounts throughout the photocycle. While a definite function cannot yet be attributed to the emission of 3-methyl-2-butenyl butyrate by T. peregrinus males, our findings point to an intraspecific function, possibly one related to male–male competition.
