También, estos productos mostraron ser muy efectivos para la conversión de lactosa en soluciones tamponadas, en lactosuer, en permeatos de lactosuero y en leche. Se realizaron cuatro ensayos en un bioreactor de columna que funcionó en forma continua por diecisiete días en cada experiencia. En todos los casos se logró una conversión del orden del 90%. Después de nueve meses de almacenamiento adecuado, los conjugados mantuvieron su actividad enzimática, demostrando su alta estabilidad. Selenium is a trace element for most organisms; its deficiency and excess are detrimental. Selenium beneficial effects are mainly due to the role of the 21st genetically encoded amino acid selenocysteine (Sec). Selenium also exerts Sec-independent beneficial effects. Its harmful effects are thought to be mainly due to non-specific incorporation in protein synthesis. Yet the selenium response in animals is poorly understood. In Caenorhabditis elegans, Sec is genetically incorporated into a single selenoprotein. Similar to mammals, a 20-fold excess of the optimal selenium requirement is harmful. Sodium selenite (Na2SeO3) excess causes development retardation, impaired growth, and neurodegeneration of motor neurons. To study the organismal response to selenium we performed a genetic screen for C. elegans mutants that are resistant to selenite. We isolated non-sense and missense egl-9/EGLN mutants that confer robust resistance to selenium. In contrast, hif-1/HIF null mutant was highly sensitive to selenium, establishing a role for this transcription factor in the selenium response. We showed that EGL-9 regulates HIF-1 activity through VHL-1, and identified CYSL-1 as a key sensor that transduces the selenium signal. Finally, we showed that the key enzymes involved in sulfide and sulfite stress (sulfide quinone oxidoreductase and sulfite oxidase) are not required for selenium resistance. In contrast, knockout strains in the persulfide dioxygenase ETHE-1 and the sulfurtransferase MPST-7 affect the organismal response to selenium. In sum, our results identified a transcriptional pathway as well as enzymes possibly involved in the organismal selenium response. A montmorillonite was pillared separately with chromium and aluminum polyoxocations (Cr–C and Al–C respectively) and also with an equimolar mixture of both polioxocations (Al–Cr–C). The resulting solids were characterized by scanning electron microscopy, energy dispersive spectrometry, thermal analysis, Fourier transform infrared spectroscopy, X-ray diffraction and nitrogen adsorption at �196 �C. The inclusion of polycations in the interlamellar space led to modifications in the properties of the clay. An eleven fold increase in specific surface area with respect to the calcined starting clay was found for the Al–C, whereas a twofold increase was observed for the Cr–C; the value for the Al–Cr–C was only slightly higher than that for the latter. Diffractograms of the Cr–C showed the occurrence of a Cr2O3 phase. Catalytic activity in oxidative dehydrogenation of propane was assayed in the temperature range from 350 �C to 450 �C using Ar/C3H8/O2 (80/10/10) as reactant gas mixture. Propylene selectivity was found to be temperature- dependent, showing a maximum in the studied temperature range for the catalyst containing only aluminum, whereas remaining practically constant for the Cr-containing catalysts. Highly promising propylene yields at low temperature were obtained with the catalyst containing Al and Cr (Al–Cr–C): 9.3% at 350 �C and 10.3% at 450 �C. In a previous work we have identified two possible structures for the radical cation obtained by ionization of hydrogen‐bonded formaldehyde–water complexes [Coitiño et al., J. Am. Chem. Soc. 115, 9121 (1993)], a hydrogen‐bonded and an addition‐like complexes. We observed that the results were highly dependent on the method of calculation employed. Inclusion of correlation was crucial for obtaining the correct structures of some of the complexes. In this work we used high‐level ab initio calculations in order to predict the equilibrium structure of these two complexes, the possibility of its existence in gas phase, and the infrared spectrum to be expected in each case. A series of progressively more sophisticated basis sets was used to assess the effect of the quality of the calculations on the expected results. Also, full geometry optimization with each basis set was performed at the second‐order Mo/ller–Plesset level, and correlation energy was calculated at the fourth‐order Mo/ller–Plesset level to assess the contribution of this factor to the global result. Confirming our previous results, we found that correlation affects the hydrogen‐bonded radical‐cation complex more than the addition one, due to the different bonding patterns in each of them. Both complexes are stable—toward decomposition to the fragments or to CO+H+H3O+—by several kcal/mol at all levels of theory. The hydrogen‐bonded complex is more stable than the additional one by a respectable amount (13 kcal/mol at the highest level used here), lending support to our previous analysis of the reactions of the former as the main channels for evolution of the formaldehyde–water radical cation. The H‐bonded complex [H3O+...HCO⋅] presents two characteristics, very intense absorptions which should allow identification of this radical cation if present in the experimental setup. These transitions are also present in the HCO⋅ radical but their intensity is enhanced by an order of magnitude due to the coupling with the proton in H3O+. We conclude that the combination of stability and characteristic infrared transitions should make this radical‐cation complex a relatively easy target for experimental determination. The observed tendency is that the CCSD[T] method overestimates the correlation energy when we compare it against the CCSDT method. The average CCSDT ) CCSD[T] difference is 0.42 kcal/mol with deviations larger than 1 kcal/mol for some molecules, such us SO2, SO3, SCO and CS2. This effect difficults the prediction of enthalpies of formation with chemical accuracy (�1 kcal/mol), since, CCSD[T] in general performs better than CCSDT in the calculation of enthalpies of formation. A lipoprotein fraction from fertile bovine hydatid cyst fluid (FBHCF) was isolated by affinity chromatography on acrylic-heparin. The heparin-binding lipoprotein fraction (HBLF) was proved to be free from host immunoglobulins and albumin. Immunoblotting with confirmed human hydatid sera showed that the major relevant diagnostic bands in FBHCF were also present in HBLF. HBLF or FBHCF were used to sensitize polystyrene latex particles. HBLF-latex showed both higher reactivity with positive hydatid human sera and higher batch-to-batch reproducibility than FBHCF-latex. Titration of sera from 119 surgically confirmed hydatid patients, 48 with other parasitic diseases, 6 with unrelated diseases and 37 healthy subjects was performed using HBLF-latex and conventional ELISA (using FBHCF). Sensitivity and specificity were 83 and 87% for ELISA and 87 and 88% for HBLF-latex. These results indicate that, while being simpler and faster to perform, the HBLF-latex has a similar diagnostic value to that of the conventional ELISA test. Renewable hydrogen produced from biomass as an energy vector appears as one of the most promising alternatives to solve, partially at least, the actual environmental and energetic problems. In this sense, a novel series of Ni catalysts supported on rare-earth zirconates (La and Y) for hydrogen production through bioethanol steam reforming were prepared by sol-gel technique and subsequently calcined at 700 and 950 ◦C. Treatment for Chagas disease, a parasitosis caused by Trypanosoma cruzi, has always been based on two drugs, nifurtimox and benznidazole, despite the toxic side effects described after prolonged prescription. In this work, we study a new prospective antitrypanosomal drug based on vanadium, here named VIVO(5Brsal)(aminophen). We found a good IC50 value, (3.76 ± 0.08) μM, on CL Brener epimastigotes. The analysis of cell death mechanism allowed us to rule out the implication of a mechanism based on early apoptosis or necrosis. Recovery assays revealed a trypanostatic effect, accompanied by cell shape and motility alterations. An uptake mostly associated with the insoluble fraction of the parasites was deduced through vanadium determinations. Concordantly, no drastic changes of the parasite transcriptome were detected after 6 h of treatment. Instead, proteomic analysis uncovered the modulation of proteins involved in different processes such as energy and redox metabolism, transport systems, detoxifying pathways, ribosomal protein synthesis, and proteasome protein degradation. Overall, the results here presented lead us to propose that VIVO(5Brsal)(aminophen) exerts a trypanostatic effect on T. cruzi affecting parasite insoluble proteins. Objective: Systemic inflammatory response syndrome is characterized by increased urinary excretion of selenium and low serum concentration. Repletion by parenteral selenite is the most efficacious form of supplementation. However, the optimum safe dose and mode of administration remain controversial. We aimed to determine pharmacokinetic and pharmacodynamic profiles of selenite and estimate a safe dose to optimize selenium status. Methods: A prospective, randomized, pilot study in 20 patients with systemic inflammatory response syndrome compared a high-dose (HD) group that received a loading dose of selenium as selenite 15.18 mmol over 2 h and thereafter 10.12 mmol/d as a continuous intravenous infusion (CIV) for 10 d with a very–high-dose (VHD) group that received a loading dose of 25.30 mmol over 2 h and thereafter 20.24 mmol as a CIV for 10 d. Clinical outcome was evaluated by length of stay in the intensive care unit, incidence of ventilator-associated pneumonia, and Sequential Organ Failure Assessment score. Results: Patients in group HD (n ¼ 10, age 54 � 23 y) had an Acute Physiology and Chronic Health Evaluation II score of 23 � 5 and a Sequential Organ Function Assessment score of 10 � 2. Those in group VHD (n ¼ 10, age 41 � 19 y) had scores of 21 � 7 and 8 � 3, respectively. Pharmacokinetic concentration/time curves for serum selenium overlapped but were independent of dose, whereas the pharmacodynamics were different, showing maximum glutathione peroxidase activity only with VHD. Glutathione peroxidase decreased after day 7 independently of the selenium dose. Clinical outcomes were similar in both groups. Purpose: To confirm the pharmacodynamics and evaluate the efficacy of high-dose selenium (Se) administered by continuous infusion, following an initial loading bolus of selenite, on clinical outcome in critically ill patients with systemic inflammatory response syndrome (SIRS). Methods: Prospective, placebo-controlled, randomized, single-blinded phase II study in a multidisciplinary university hospital intensive care unit (ICU). Two groups of patients with SIRS, age[18 years, and Acute Physiology and Chronic Health Evaluation (APACHE) II C15 (n = 35) were randomized to receive either placebo or intravenous selenite as a bolus-loading dose of 2,000 lg Se followed by continuous infusion of 1,600 lg Se per day for 10 days. Blood samples were analyzed before randomization (day 0) then at days 3, 7, and 10. Clinical outcome was assessed by Sequential Organ Failure Assessment (SOFA) score. Hospitalacquired pneumonia including ventilator- associated pneumonia (VAP), adverse events, and other safety parameters were monitored as secondary endpoints. Results: SOFA score decreased significantly in the selenite group at day 10 (1.3 ± 1.2 versus 4.6 ± 2.0, p = 0.0001). Early VAP rate was lower in the selenite group (6.7% versus 37.5%, p = 0.04), and hospitalacquired pneumonia was lower after ICU discharge (p = 0.03). Glutathione peroxidase-3 (GPx-3) activity increased in both groups, reaching a maximum at day 7 (0.62 ± 0.24 versus 0.28 ± 0.14 U/mL, p = 0.001) in the selenite group. No adverse events attributable to selenite were observed. Conclusions: Daily infusion of 1,600 lg Se (as selenite), following an initial bolus of 2,000 lg, is novel and without short-term adverse events. High-dose parenteral selenite significantly increases Se status, improves illness severity, and lowers incidence of hospital-acquired pneumonia including early VAP for SIRS patients in ICU. Developing electrode materials with high specific electrical capacitance and low electrical resistance enables an increase of energy accumulated in the supercapacitor. In addition, it is expected that the electrode material presents a simple procedure for preparation with low production cost and be friendly to the environment. In this work, a cheap and simple electroless process was involved to deposit chemical species of iron on a no expensive textile activated carbon fiber felt with interesting properties for application as supercapacitor electrode. The resulting binary composite (Fe@ACF) supercapacitor electrode possesses a high specific capacitance of 237 F g−1 at a current density of 0.2 A g−1 and only 3% of loss of its initial specific capacitance after 3000 charge-discharge cycles. Human milk (HM) constitutes the first immunological barrier and the main source of nutrients and bioactive components for newborns. Immune factors comprise up to 10% of the protein content in HM, where antibodies are the major components (mainly IgA, IgG, and IgM). In addition, antibacterial enzymes such as lysozyme and immunoregulatory factors such as soluble cluster of differentiation 14 (sCD14) and transforming growth factor β2 (TGF-β2) are also present and play important roles in the protection of the infant's health. Donor milk processed in HM banks by Holder pasteurization (HoP; 62.5°C, 30 min) is a safe and valuable resource for preterm newborns that are hospitalized, but is reduced in major immunological components due to thermal inactivation. We hypothesized that high hydrostatic pressure (HHP) and high-pressure homogenization (HPH) are 2 processes that can be used on HM to reduce total bacteria counts while retaining immunological components. We studied the effects of HHP (400, 450, and 500 MPa for 5 min applied at 20°C) and HPH (200, 250, and 300 MPa, milk inlet temperature of 20°C) applied to mature HM, on microbiological and immunological markers (IgA, IgG, IgM, sCD14, and TGF-β2), and compared them with those of traditional HoP in HM samples from healthy donors. The HHP processing between 400 and 500 MPa at 20°C reduced counts of coliform and total aerobic bacteria to undetectable levels (<1.0 log cfu/mL) while achieving approximately 100% of immunological component retention. In particular, comparing median percentages of retention of immunological components for 450 MPa versus HoP, we found 101.5 versus 50.5% for IgA, 89.5 versus 26.0% for IgM, 104.5 versus 75.5% for IgG, 125.0 versus 72.5% for lysozyme, 50.6 versus 0.1% for sCD14, and 88.5 versus 61.1% for TGF-β2, respectively. Regarding HPH processing, at a pressure of 250 MPa and inlet temperature of 20°C, the process showed good potential to reduce coliforms to undetectable levels and total aerobic bacteria to levels slightly above those obtained by HoP. The median percentages of retention of immunological markers for HPH versus HoP were 71.5 versus 52.0%, 71.0 versus 27.0%, 104.0 versus 66.5%, and 30.9 versus 0.2%, for IgA, IgM, IgG, and sCD14, respectively; results did not significantly differ for lysozyme and TGF-β2. This study highlights the development of a high peak capacity, high-throughput (HTP) approach for a target list of 62 organic contaminants in an environmental river water matrix on a HPLC conventional 400 bar system. Key separation metrics were evaluated: (a) peak capacity, (b) total analysis time and (c) mobile phase consumption. An average peak width of 0.10 min with a total analysis time (inclusive of the column wash and re-equilibration) of 10 minutes revealed the increased productivity and performance of the parallel segmented flow (PSF) column technology HPLC-MS strategy, comparable to UHPLC peak widths and analysis times, achieved at a significantly lower backpressure (<400 bar). The operation of the PSF in a narrow-bore scale format (internal diameter of 2.1 mm) resulted in a conservative HPLC scale total mobile phase consumption of 15 mL for the column separation; only 5.5 mL of this volume was exposed to the ion source per injection. Mobile phase consumption was higher compared to that of UHPLC, but on the other hand it achieved higher peak capacity. Three representative compounds (atrazine, diclofenac and fluazuron) with differing retention and ionisation properties were studied in detail in terms of detection sensitivity. The majority of the ion ratios for the standards in the river water matrix were within ±30% of the average ion ratios. The largest ion suppression occurred for atrazine (−25% matrix effect), a pesticide notorious for poor ionization and poor peak shape issues. The lowest response transitions at 1 μg L−1 for the extracted ions in the river water matrix of atrazine, diclofenac and fluazuron had signal to noise ratios ≥3, with the exception of diclofenac where 5 μg L−1 was the lowest calibration level. The peak area's calibration curve slope and standard deviation in the detector's response determination of limit of quantification (LOQ) were between 5.2 and 30.4 μg L−1. The use of phage display peptide libraries allows rapid isolation of peptide ligands for any target selector molecule. However, due to differences in peptide expression and the heterogeneity of the phage preparations, there is no easy way to compare the binding properties of the selected clones, which operates as a major “bottleneck” of the technology. Here, we present the development of a new type of library that allows rapid comparison of the relative affinity of the selected peptides in a high-throughput screening format. As a model system, a phage display peptide library constructed on a phagemid vector that contains the bacterial alkaline phosphatase gene (BAP) was selected with an antiherbicide antibody. Due to the intrinsic switching capacity of the library, the selected peptides were transferred “en masse” from the phage coat protein to BAP. This was coupled to an optimized affinity ELISA where normalized amounts of the peptide−BAP fusion allow direct comparison of the binding properties of hundreds of peptide ligands. The system was validated by plasmon surface resonance experiments using synthetic peptides, showing that the method discriminates among the affinities of the peptides within 3 orders of magnitude.
