Helminth parasite infections are associated with predominant Th2-type cytokine responses, and parasite glycoconjugates have been recognized as partially responsible for such immune bias. It has been proved that Echinococcus granulosus evokes a Th2-type cytokine pattern characterized by a high production of IL-4, IL-5, IL-6 and IL-10, and no or mild IFN-c levels in animal models and in patients with cystic echinococcosis, respectively. Here, we show that E4+ (a glycoconjugateenriched fraction from E. granulosus protoscolex) stimulated the secretion of a high concentration of IL-6, followed by IL-10 and TNF-a by normal peritoneal B cells. We determined that E4+ bound to the surface of peritoneal B cells and induced their activation and, also, triggered the differentiation of peritoneal B cells into IgM-, IgG2b- and IgG3-secreting cells in a T-independent way. Interestingly, the IgM released by E4+-stimulated peritoneal B cells from normal mice recognized protoscolex antigens. Results showed that, after the encounter with antigens from E. granulosus protoscolex, peritoneal B cells are a source of Th2-type cytokines and polyclonal antibodies, some of which recognize parasite antigens, suggesting that peritoneal B cells can condition the outcome of the infection. In anoxic environments, degradation of organic matter (OM) results in strong fractionation of carbon isotopes, with formation of 13C-depleted CH4. Propionate and acetate are important products of OM fermentation. Propionate is further fermented to acetate. Acetate is a direct precursor of CH4, the remainder usually being produced from H2-mediated CO2 reduction. There is a paucity of data for the turnover of acetate and, even more so, propionate. We therefore analyzed the d13C values of organic carbon, propionate, acetate and the methyl (Me) group of acetate (acetate-Me) during the production of CH4 in anoxic incubations of various flooded and non-flooded soils and various lake sediments. Incubation in the presence of CH3F, which inhibits CH4 production from acetate, allowed exclusion of isotope effects during aceticlastic methanogenesis. Despite the variation inherent in the wide diversity of sample type and origin, the data collectively showed that the d13C value of acetate was only marginally different (�2 &plusmn; 5&permil;) from that of OM, while propionate was depleted in 13C relative to total acetate (�6 &plusmn; 5&permil;). Acetate-Me was generally depleted in 13C relative to total acetate (�8 &plusmn; 5&permil;). Thus, isotopic enrichment factors during the degradation of OM to total propionate and acetate were much smaller than those during hydrogenotrophic and aceticlastic methanogenesis or the intramolecular difference in d13C between the carboxyl (CO2H) and Me of acetate, so that the d13C value of OM may be used as a proxy when data for acetate are not available. Nutritional warnings corresponded to separate black octagonal signs containing the word &lsquo;Excess&rsquo; followed by the corresponding nutrient: total fat, saturated fat, sugars and sodium.</p> <p>Participants were asked to purchase a lunch for themselves using a simulated food ordering website.</p> <p><br />Setting: Online study in Uruguay.</p> <p><br />Participants: Convenience sample of 447 Uruguayan participants, recruited using social media.</p> <p>Results: In the control condition, 76 % of the participants selected a dish or a beverage with excessive content of at least one nutrient in the simulated food ordering website. When nutritional warnings were included, this percentage significantly decreased to 62 % (P = 0&middot;002). An in-depth understanding of people&rsquo;s experiences, beliefs and thoughts about cooking and homemade meals is essential for the design of a more effective discourse to encourage cooking from scratch. In this context, the objective of the present work was to explore the social representations of homemade meals and cooking. An online survey was conducted with 974 Uruguayan adults, recruited using an advertisement on Facebook. Participants were asked to write down the first words that came to their mind when thinking of homemade meals and cooking. Responses were coded into categories using inductive coding. The structure of the social representations was explored using the frequency of mention and rank of appearance of the categories. Results showed<br />that the social representations of homemade meals were largely consensual among participants and mainly related to pleasure, health, and wellbeing. However, the social representations of cooking did not have a clear structure, which raised diverse associations related to homemade foods, positive emotions but also to lack of convenience in terms of time and effort. Results suggest public campaigns aimed at encouraging people to cook from scratch should position cooking as enjoyable task and diminish perception of lack of time and effort association. Digital marketing has seen a rapid rise in the last decade as a consequence of the increased popularity of social media. However, few studies so far have analyzed the prevalence and persuasive power of digital marketing of ultra-processed products. The present study aimed at: (i) analyzing the content of Instagram posts of ultra processed products through the lens of the heuristic-systematic model, and (ii) evaluating the influence of the content of the posts on user interactions. A search for Instagram accounts of ultra-processed products was per formed using a master list of products commercialized in the two most popular online supermarkets in Uruguay. For each of the identified Instagram accounts, all the content posted in a 6-month period was recorded (August 15th, 2020 to February 15th, 2021). The posts were analyzed using content analysis based on inductive coding, and gradient boosting models (GBMs) were used to address the second study objective. A total of 2178 Instagram posts promoting specific ultra-processed products or brands were identified. The posts included a diverse set of cues to trigger both systematic and heuristic processing. References to the pleasure derived from product con sumption as well as health-related cues were most prevalent. The GBM showed that references to contests and raffles and invitations to interact encouraged users to engage with the posts through posting comments. In this work, we made a review about the application of Raman spectroscopy for quality control of honey and other food samples. Simultaneously, it was carried out a new research evaluating silver nanoparticles (AgNPs) obtained by green synthesis for detection of the antibiotic Oxytetracycline (OTC), through Surface Enhanced Raman Spectroscopy (SERS). Glucose was used as reducing and capping agent in the formation of AgNPs in alkaline media. The frequencies found, belonging to the antibiotic at 20, 15, 10 and 5 ppb, could be visibly differentiated from those corresponding to honey, being unnecessary to perform any extraction procedure or tedious manipulation of the sample. Theoretical frequencies were obtained by Density Functional Theory calculations and compared with the normal modes from SERS measures, founding an excellent agreement between them. The analysis shows a preference for the adsorption of OTC on silver cluster through oxygen atoms. Opal phytolith, geochemical and sedimentological data from two cores taken in Pe&ntilde;a Lagoon, a small water body on the Atlantic shore of Uruguay, are presented in this paper. The choice of this specific sampling location aimed to obtain high resolution records to infer late Holocene environmental and climatic changes in southeastern Uruguay, in order to improve previous models for early and middle Holocene. The combined record of two cores extends to 2500 14C BP, with a sedimentation rate ranging between 0.6 and 2 mm y&minus;1. A multiproxy approach allowed the identification of changes in dynamics, particularly organic matter and sediment input variability from the watershed. With this information, differences in the amount and extent of environmental signals were evaluated and a comprehensive climate change model was generated, especially for the last 1500 years. Three main climatic events were distinguished. The oldest, which lasted until 700 AD, was characterized by temperate and humid conditions. The second extended until 1200 AD and warmer and wetter conditions were inferred, but also a colder and drier pulse was observed in between. This second event was assigned to the Warm Medieval Period (WMP). During the third event, assigned to the Little Ice Age (LIA), which extended until the present, a high variability in climate was observed, but also three dry/cold phases reaching their maxima by 1300, 1600 and 1900 AD were inferred. Objective: To determine the pharmacokinetic parameters of phenobarbital in infants, using saliva as a biologic fluid, and to correlate the parameters obtained from saliva data with those obtained from plasma data. Design: A prospective, randomized study. Setting: Hospitalized patients at the Medical Center Pereira Rossell, a pediatric hospital in Montevideo, Uruguay. Patients: Sixteen infants with seizure disorders were included in the study. None of them was treated with other medications. Interventions: A direct intravenous loading dose of phenobarbital 10 mg/kg was administered, followed by a maintenance dosage of 5 mg/kg/d (once- or twice-daily dosing) given 12 hours after the loading dose. Main Outcome Measures: Saliva and plasma samples were obtained 6 and 12 hours after the loading dose and 3 days after the initiation of the maintenance dose (trough sample). The samples were analyzed by HPLC and the elimination half-life (t1/2), the volume of distribution (Vd), and the percentage of unbound drug in plasma (UDP) were calculated. Results: The t1/2 obtained from plasma and saliva data was 30 hours; the Vd was 0.73 L/kg from plasma data and 2.4 L/kg from saliva data; and the UDP was 75 percent. Trough concentrations showed no significant difference between treatments. Conclusions: Saliva is a useful biologic fluid to determine phenobarbital pharmacokinetic parameters, mainly in pediatric patients. Moreover, a single daily dose of phenobarbital is sufficient to obtain therapeutic concentrations. Many dairy industries have been using SBR wastewater treatment plants because they allow optimal working condition to be reached. However, to take advantage of SBR capabilities, strong process automation is needed. The aim of this work is to study the factors that influence SBR performance to improve modelling and control. To better understand the whole process we studied the kinetic modelling, the carbon removal mechanism and the relation between reactor performance, aerobic heterotrophic activity and bacterial population dynamics (by terminal restriction fragment length polymorphisms of 16S rDNA, T-RFLP). The heterotrophic activity values presented high variability during some periods; however, this was not reflected on the reactor performance. As sludge health indicator, the average activity in a period was better than individual values. Although all the carbon removal mechanisms are still unclear for this process, they seemed to be influenced by non-respirometric ways (storage, biosorption, accumulation, etc.). The variability of heterotrophic activity could be correlated with the bacterial population diversity over time. Despite the high variability of the activity, a simple kinetic model (pseudo ASM1) based on apparent constant parameters was developed and calibrated. Such modellisation provided a good tool for control purposes. The NS2B-NS3 protease has been identified as an attractive target for drug development against Zika virus (ZIKV) and combined drug repurposing and structure-based virtual screening has improved the development of antiviral drugs. In this study, we performed a structure-based virtual screening of 1861 Food and Administration (FDA) approved drugs available in DrugBank by the selection and docking validation of crystal structure of ZIKV NS2B-NS3 protease (PDB ID 5H4I) using Glide and DOCK 6 software. The antihistaminic chlorcyclizine (Grid score &minus;24.8 kcal/mol) exhibited the most promising interaction with NS2B-NS3 protease in comparison to crystallography ligand (Grid score &minus;15.6 kcal/mol) by interaction to Tyr161 by hydrophobic interactions in the binding site of NS2B-NS3 which is recognized as an important amino acid in substrate molecular recognition. Cytotoxicity and global antiviral activity assay in Vero cells by MTT method showed that chlorcyclizine reduced the ZIKV induced cytopathic effect (EC50 of 69.0 &plusmn; 7.3 &mu;M and SI = 1.9), and explicit molecular dynamics simulations implemented on a NAMD program indicated great stability of chlorcyclizine in protease binding site, suggesting the repurposing of chlorcyclizine as a promising finding in anti-ZIKV drug development. Eukaryotic diphthine synthase, Dph5, is a promiscuous methyltransferase that catalyzes an extraordinary N,O-tetramethylation of 2-(3-carboxy-3-aminopropyl)-l-histidine (ACP) to yield diphthine methyl ester (DTM). These are intermediates in the biosynthesis of the post-translationally modified histidine residue diphthamide (DTA), a unique and essential residue part of the eukaryotic elongation factor 2 (eEF2). Herein, the promiscuity of Saccharomyces cerevisiae Dph5 has been studied with in silico approaches, including homology modeling to provide the structure of Dph5, protein&ndash;protein docking and molecular dynamics to construct the Dph5-eEF2 complex, and quantum mechanics/molecular mechanics (QM/MM) calculations to outline a plausible mechanism. The calculations show that the methylation of ACP follows a typical SN2 mechanism, initiating with a complete methylation (trimethylation) at the N-position, followed by the single O-methylation. For each of the three N-methylation reactions, our calculations support a stepwise mechanism, which first involve proton transfer through a bridging water to a conserved aspartate residue D165, followed by a methyl transfer. Once fully methylated, the trimethyl amino group forms a weak electrostatic interaction with D165, which allows the carboxylate group of diphthine to attain the right orientation for the final methylation step to be accomplished.
