A novel epoxyquinol analog was prepared by molecular simplification of monomeric and dimeric scaffolds. A feasible methodology for the insertion of side chains into cyclohexenone skeleton was developed. Insertion of the hydroxymethyl side chain was achieved through a-sulfonylcarbanion chemistry. The alkenyl chain was inserted through palladium cross-coupling strategy.

A summary of the resource.
The synthesis of 3-hydroxy-2-hexanone and 2,3-hexanediol, two components of the aggregation pheromone of several cerambycid species, is disclosed in here. Starting from 2-hexanone, through an α-hydroxylation using (diacetoxyiodo)benzene, 3-hydroxy-2-hexanone is obtained in good yield. Further reduction of this compound, gives 2,3-hexanediol in excellent yield. A study of the α-hydroxylation reaction of several alkylketones using an hypervalent iodine reagent is also disclosed in here. The synthesis of optically active compounds (R)- and (S)-3-hydroxy-2-hexanone was achieved starting from 2-hexanone with nitrosobenzene and l- and d-proline respectively, in several reaction media.

A summary of the resource.
<p>The growing incidence of non-communicable diseases makes the search for natural sources of bioactive compounds a priority for such disease prevention/control. Achyrocline satureioides<br />(‘marcela’), a plant rich in polyphenols and native to Brazil, Uruguay, Paraguay, and Argentina, could be used for this purpose. Data on its antidiabetic/antiobesity properties and cellular uptake of bioactive compounds are lacking. The potentiality of non-thermal technologies such as highhydrostatic<br />pressure (HP) to enhance polyphenol extraction retains attention. Thus, in the present study aqueous and ethanolic marcela extracts with/without assisted-HP processing were chemically characterized and assessed for their in vitro antioxidant capacity, antidiabetic and antiobesity activities, as well as cellular cytotoxicity and uptake on intestinal cell monolayers (TC7-cells, a clone of Caco-2 cells). Aqueous and ethanolic conventional extracts presented different polyphenolic<br />profiles characterized mainly by phenolic acids or flavonoids, respectively, as stated by reverse phase-high-performance liquid chromatography (RP-HPLC) analyses. In general, ethanolic extracts presented the strongest bioactive properties and HP had none or a negative effect on in vitro bioactivities comparing to conventional extracts. TC7-cell viability and cellular uptake demonstrated in conventional and HP-assisted extracts, highlighted the biological effects of marcela bioactive<br />compounds on TC7-cell monolayers. TC7-cell studies showed no HP-induced cytotoxicity. In sum, marcela extracts have great potential as functional ingredients for the prevention/treatment of chronic diseases such as diabetes.</p>

Toxicology is nowadays an interdisciplinary science, including various areas such as Clinic, Experimental, Forensic, Analytic, Occupational, Environmental and Social Toxicology. Apart from the inter- and multidisciplinary approach, there is now growing conscience on the importance of the transdisciplinary view,which transcends and goes through the various disciplines, collecting their common aspects. The Toxicology and Environmental Hygiene Department of the Faculty of Chemistry, University of the Republic, gives a course called “Analytic Toxicology and Legal Chemistry”, in which the students write and present orally a monography. The themes of the course and their treatment may constitute an exercise of acquisition of the transdisciplinary approach. The aim of the present study is to propose a methodology to evaluate this competence in the students, given the analysis of their performance in previous instances of teaching of this course. Previous result,s through a period of five years, show a satisfactory performance of the students, with average marks higher than 80% of the possible maximum for the written and above 75%. for the oral part. This antecedent points to an incipient acquisition of the transdisciplinary approach, which must be evaluated in future instances of this course, through the requirements for the students monography and a suiting questionnaire.

The perfume industry is increasingly interested in developing perfume classification systems based on their odor character profile to improve consumer shopping experience. However, concerns have been raised regarding the external validity of the classifications performed by experts and trained assessors. In this context, the aim of the present work was to evaluate the influence of training on perfume odor characterization based on a sorting task, by comparing sensory maps of perfumes and descriptions provided by trained assessors and consumers. Fifteen commercially fine female perfumes were evaluated by trained assessors and consumers using a sorting task, followed by a description phase. Trained assessors and consumers provided similar sample configurations. However, there was no perfect agreement in their classifications and some differences in the conclusions regarding similarities and differences between samples were identified. Although the descriptions provided by trained assessors and consumers to characterize the odor profile of the perfumes were different, the most frequently mentioned terms were similar and were used in a similar way

In vitro the yeast exhibited a variety of different antagonistic mechanisms against the phytopathogenic fungus including: volatile organic compounds (VOCs) production, β-1, 3 glucanase and protease activity, inhibition of spore germination, and the competition for saccharose, glucose, fructose and total carbohydrates. Results in situ indicated that disease incidence and lesion diameter of anthracnose on papaya var. Maradol fruit was significantly reduced with a higher concentration of D. hansenii. Our study indicates that D. hansenii is an excellent agent of control to anthracnose disease on papaya var. Maradol fruit.

Investigaci6n y desarrollo de nuevas N-acilhidrazonas bioactivas 11 Resumen El agrupamiento N-acilhidrazona ha sido descrito como un fannacOforo con capacidad de proveer varias actividades farmaco16gicas a traves de su interacci6n con diferentes dianas. A su vez, los heterociclos derivados de furoxano son reconocidos por su actividad bio16gica frente a distintos blancos terapeuticos. En el presente trabajo de tesis doctoral se ha realizado el disefio y sintesis quimica de nuevos compuestos hibridos que incluyen el agrupamiento N-acilhidrazona unido a heterociclos derivados del sistema metilfuroxano, fenilfuroxano y benzofuroxano. Los nuevos compuestos han sido desarrollados empleando procedimientos sinteticos sencillos y eficientes y la caracterizaci6n estructural de los mismos se ha realizado mediante tecnicas espectrosc6picas habituales. Estos compuestos hibridos se han evaluado en diferentes modelos bio16gicos y a traves de ellos ha sido posible identificar nuevos lfderes con elevados indices de selectividad frente a Mycobacterium tuberculosis, Trypanosoma cruzi y Leishmania amazonensis. Los estudios fannaco16gicos han permitido identificar nuevos compuestos con actividades analgesicas y anti-inflamatorias que actuan principalmente a traves de su capacidad de inhibir la via NF-KB y la liberaci6n de Interleuquina-8. Los compuestos que en este trabajo se identificaron con mejor perfil bio16gico, presentan en general, baja citotoxicidad y no son mutagenicos. Esto los ubica como nuevos prototipos para la realizaci6n de futuras optimizaciones moleculares.

We studied in vitro cytokine production by peripheral blood mononuclear cells (PBMC) from patients with primary and recurrent hydatid disease when cells were incubated with mitogen (PHA) and antigen from hydatid cyst fluid (HCFAg); levels of specific IgE, IgG4 and eosinophil counts were also measured in sera. When specifically stimulated, PBMC from patients produced higher levels of IL-2 (P < 0·02), IFN- g (P < 0·0028) and IL-5 (P < 0·01) than those from uninfected donors, whereas IL-10 levels were comparable. Notably, IL-5 was also produced in higher levels (P < 0·01) by PBMC from patients when incubated with PHA. The IL-5:IFN- g ratio was significantly greater (P < 0·02) when measured in response to specific stimulation than it was for PHA-stimulated cultures. These cytokine data suggest a bias towards a Th2-response which is in agreement with the high levels of IgG4 and IgE observed. The polarized response appears to be related to clinical status, as differences between patients with primary infec- tion and those with relapse of disease were demonstrated, with significantly higher levels of IgE (P < 0·003), IgG4 (P < 0·04) titres and eosinophil counts (P < 0·04) in the latter; in addition a tendency to an increased production of IL-5 buy lower IFN- g was also observed in this group. These results merit further study as they are suggestive of a putative role of Th2-like responses in susceptibility to reinfection by E. granulosus

A summary of the resource.
Significant protective immunity against secondary hydatidosis in mice was achieved by immunization with a preparation of surface molecules of E. granulosus protoscoleces with Freund's incomplete adjuvant (PSEx-IFA). Study of PSEx-IFA immunogenicity demonstrates that glucidic epitopes evoke mainly IgM responses while peptidic epitopes evoke mainly IgG responses; however both types of epitopes elicit both types of responses. Analysis of the possible association between susceptibility or resistance to infection and antibody responses after challenge was also performed. Cyst fluid antigen (CFAg) specific antibody titres on month eight after challenge correlated with number and size of cysts. On the other hand no correlation was observed between protection and PSEx specific IgG titres either on the day of challenge or one month later. Nonetheless, immunoblot analysis revealed that some PSEx molecules were recognized on day 30 after challenge only by sera from immunized mice.

Autoimmunity is a feature of celiac disease (CD) with tissue transglutaminase (tTG) as a major autoantigen. A correlation between gynecological-obstetric disorders in CD patients and the presence of circulating antibodies anti-tTG that inhibited tTG activity was reported. Serum anti-tTG antibodies were detected in a non-obese diabetic (NOD) mouse model of type I insulin-dependent diabetes mellitus and Sjögren's syndrome, two comorbid states with CD. Since pregnancy complications have been described in NOD mice, we evaluated the ability of anti-tTG antibodies to affect the functions of tTG relevant to the normal course of an early pregnancy like extracellular matrix assembling and apoptotic cell phagocytosis by macrophages. Circulating IgG antibodies against tTG were detected in NOD mice with titers that decreased at early pregnancy; interestingly, the in vitro transamidating activity of tTG was reduced by NOD serum samples. Particularly, anti-tTG antibody inhibited apoptotic cell phagocytosis by peritoneal macrophages from pregnant NOD mice that express the enzyme on surface. Evidence provided support for a role for anti-tTG antibodies through reduced transamidating activity and reduced apoptotic cell clearance by the macrophages of pregnant NOD mice.

Hybrid vinylthio-, vinylsulfinyl-, vinylsulfonyl- and vinylketo-benzofuroxans developed as anti-trypanosomatid agents, against Trypanosoma cruzi and Leishmania spp., have showed low micromolar IC50 values. The synthetic route to access to these derivatives was an efficient Wittig reaction performed in mild conditions with polymer-supported triphenylphosphine (PS-TPP). Additionally, the benzofurozan analogues, deoxygenated benzofuroxans, were prepared using PS-TPP as reductive reagent in excellent yields. The trypanosomicidal and leishmanocidal activities of the benzofuroxan derivatives were measured and also some aspects of their mechanism of action studied. In this sense, inhibition of mitochondrial dehydrogenases activities, production of intra-parasite free radicals and cruzipain inhibition were studied as biological target for the anti-trypanosomatid identified compounds. The trypanosomicidal activity could be the result of both the parasite-mitochondrion function interference and production of oxidative stress into the parasite

Bladder cancer is a worldwide problem and improved therapies are urgently needed. In the search for newer strong antitumor compounds, herein, we present the study of three nitric oxide-releasing compounds and evaluate them as possible therapies for this malignancy. Bladder cancer cell lines T24 and 253J were used to evaluate the antiproliferative, antimigratory, and genotoxic effects of compounds. Moreover, we determined the NF-κB pathway inhibition, and finally, the survivin downregulation exerted by our molecules. The results revealed that compounds 1 and 3 exerted a high antiproliferative activity against bladder cancer cells through DNA damage and survivin downregulation. In addition, compound 3 reduced bladder cancer cell migration. We found that nitric oxide donors are promising molecules for the development of a new therapeutic targeting the underlying mechanisms of tumorigenesis and progression of bladder cancer

Neglected diseases represent a major health problem. It is estimated that one third of the world population is infected with tuberculosis and additionally Leishmaniosis and Chagas disease affect approximately 30 million people. N-Acylhydrazone moiety is a repeated functional group present in several prototypes and drug candidates for these neglected diseases. On the other hand, furoxan system has been studied as pharmacophore for Leishmaniosis and Chagas diseases. Here we report on the design and preparation of forty hybrid furoxanyl N-acylhydrazones and on their activity on Mycobacterium tuberculosis, H37Rv and MDR strains, Trypanosoma cruzi, and Leishmania amazonensis. Among them, four derivatives displayed excellent to good selectivity indexes against the three different microorganisms. Hybrid compound N0-(4-phenyl-3-furoxanylmethylidene)isoniazide 9 showed the best antibacterial profile with MIC value 4.5 lesser than the value for the reference isoniazid against MDR strain. Furoxanyl N-acylhydrazone (E)-2-methyl-N0-(4-phenyl-3-furoxanylmethylidene)-4H-imidazo[1,2-a] pyridine-3-carbohydrazide 15 was ten-fold more potent against T. cruzi Amastigotes than the standard drug nifurtimox. On the other hand, derivatives (E)-N0-(5-benzofuroxanylmethylidene)benzo[d][1,3] dioxole-5-carbohydrazide 25 and (E)-N0-(4-hydroxy-3-methoxyphenylmethylidene)-3-methylfuroxan-4- carbohydrazide 37 emerged as leads for the development of new leishmanicidal agents. The adequate stability, in simulated biological system and plasma, and the lack of mutagenicity of these derivatives allow us to propose them as candidates for further pre-clinical studies.

En este trabajo de tesis se han explorado dos metodologías en la búsqueda de la obtención de nanopartículas de boratos de zinc. Estas metodologías fueron planteadas para lograr sintetizar fases de boratos de zinc en distintos rangos de temperaturas, lo que brinda grandes posibilidades en el desarrollo de aplicaciones tecnológicas. Empleando la síntesis por combustión de gel asistida se pretendió sintetizar boratos de zinc a temperatura mayores a 250 °C, para los cuales se esperaba obtener nanopartículas que fueran importantes en el desarrollo de nuevos aditivos de materiales que se preparan a altas temperaturas, por ejemplo, plásticos. A partir de las síntesis asistidas por micelas reversas, se pretendió obtener boratos de zinc a baja temperatura, por ejemplo, los aminoboratos de zinc [Zn(NH3)3B4O5(OH)4]·H2O (ZB1) y Zn3(H2B3O7)2·2NH3·4H2O (ZB2), reportados previamente durante mi trabajo de Licenciatura en Química. Mediante la metodología de combustión de gel asistida se logró sintetizar la fase ZnB4O7. Los resultados presentados para estas síntesis son contundentes respecto a los procesos químicos que tienen lugar en el sistema estudiado. El medio reductor ofrecido por la alta concentración de amonio da lugar a la formación de carbono amorfo que permanece como principal impureza en las muestras obtenidas, no siendo posible la obtención de nanopartículas de boratos de zinc. La obtención de ZB2 por métodos de baja temperatura fue posible a partir de un sistema coloidal mantenido por agitación formado por agua, CTAB, hexano y diferentes alcoholes como cosurfactantes. Para estos sistemas se obtuvo la fase cristalina ZB2 con tamaños entre 100 y 1000 nm, reportándose tamaños menores a 150 nm cuando se empleó isopropanol como cosurfactante. También se sintetizó la fase ZB2 en un medio de micelas reversas. Este último es uno de los logros más importantes de este trabajo, ya que se consiguió estabilizar un sistema de micelas reversas y obtener un producto con un tamaño de partícula menor a 150 nm. Este trabajo es un aporte importante en la química básica de estos sistemas, abriendo puertas a futuros estudios de síntesis en sistemas de micelas reversas. La metodología utilizada para determinar las condiciones de formación de micelas reversas es extrapolable a cualquier sistema de obtención de compuestos químicos a partir de una solución acuosa con parámetros de concentración y fuera iónica adecuados. Los estudios microbiológicos realizados sobre la fase ZB2 permitieron confirmar la actividad antifúngica y antibacteriana presentada por este compuesto y su dependencia con el tamaño de partícula

<p>Glutaric acid and adipic acid are dicarboxylic acids (DCAs) that are commonly found in atmospheric aerosols and cloud droplets. Within this study, the temperature- and pH-dependent rate constants of aqueous-phase OH radical reactions<br />of these two DCAs were determined through the competition kinetics method. The following Arrhenius expressions were derived for the temperature dependency of the OH radical reaction with glutaric acid: k(T, H2A) = (3.9 ± 0.1) × 1010 × exp[(−1270 ± 200 K)/T], k(T, HA−) = (2.3 ± 0.1) × 1011 × exp[(−1660 ± 190 K)/T], k(T, A2−) = (1.4 ± 0.1) × 1011 × exp[(−1400 ± 170 K)/T]. Similarly, for adipic acid: k(T, H2A)<br />= (7.5 ± 0.2) × 1010 × exp[(−1210 ± 170 K)/T], k(T, HA−) = (9.5 ± 0.3) × 1010 × exp[(−1200 ± 200 K)/T], k(T, A2−) = (8.7 ± 0.2) × 1010 × exp[(−1100 ± 170 K)/T] (in units of L mol−1 s−1). Density functional theory (DFT) calculations were performed to calculate the energy barrier of the H atom abstraction. The calculation results show that the energy barriers at the Cβ-atoms of the two DCAs are much lower than those at the Cα-atoms, indicating that the H atom abstractions predominantly occur at the Cβ-atoms. The increased ·OH rate constant in the case of the deprotonated form can be explained by the reduction of energy barrier, which is predominately caused by the variation of the inductive effect of the carboxyl group. As an important sink in the atmosphere, the degradation of glutaric acid and adipic acid by ·OH under atmospheric conditions can be accurately described by the Arrhenius expressions obtained.</p>

A summary of the resource.
The effect of H2O2-induced oxidative stress on the viability of the yeast antagonist, Cystofilobasidium infirmominiatum, as well as the effect of exogenous glycine betaine (GB) on yeast viability under oxidative stress, was determined. GB treatment improved the tolerance of C. infirmominiatum to oxidative stress. Compared to untreated control yeast cells, GB-treated cells showed less accumulation of reactive oxygen species (ROS) and a lower level of protein oxidation in response to oxidative stress. Additionally, GB-treated yeast exhibited greater biocontrol activity against Penicillium expansum and a faster growth in wounds of apple fruits stored at 25 °C compared to the performance of untreated yeast. The activities of antioxidant enzymes, including catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPX) of C. infirmominiatum were elevated by GB treatment. Results indicate that the elicitation of antioxidant response by GB may contribute to improvements in oxidative stress tolerance, population growth in apple wounds, and biocontrol activity of C. infirmominiatum.
