Neglected diseases represent a major health problem. It is estimated that one third of the world population is infected with tuberculosis and additionally Leishmaniosis and Chagas disease affect approximately 30 million people. N-Acylhydrazone moiety is a repeated functional group present in several prototypes and drug candidates for these neglected diseases. On the other hand, furoxan system has been studied as pharmacophore for Leishmaniosis and Chagas diseases. Here we report on the design and preparation of forty hybrid furoxanyl N-acylhydrazones and on their activity on Mycobacterium tuberculosis, H37Rv and MDR strains, Trypanosoma cruzi, and Leishmania amazonensis. Among them, four derivatives displayed excellent to good selectivity indexes against the three different microorganisms. Hybrid compound N0-(4-phenyl-3-furoxanylmethylidene)isoniazide 9 showed the best antibacterial profile with MIC value 4.5 lesser than the value for the reference isoniazid against MDR strain. Furoxanyl N-acylhydrazone (E)-2-methyl-N0-(4-phenyl-3-furoxanylmethylidene)-4H-imidazo[1,2-a] pyridine-3-carbohydrazide 15 was ten-fold more potent against T. cruzi Amastigotes than the standard drug nifurtimox. On the other hand, derivatives (E)-N0-(5-benzofuroxanylmethylidene)benzo[d][1,3] dioxole-5-carbohydrazide 25 and (E)-N0-(4-hydroxy-3-methoxyphenylmethylidene)-3-methylfuroxan-4- carbohydrazide 37 emerged as leads for the development of new leishmanicidal agents. The adequate stability, in simulated biological system and plasma, and the lack of mutagenicity of these derivatives allow us to propose them as candidates for further pre-clinical studies. La nanobiotecnología se ha beneficiado recientemente del uso de nanobiocatalizadores inmovilizados aplicados a la biorremediación, bioprocesos sintéticos y biomedicina, entre otros campos. La integración enzimática en materiales híbridos permite capitalizar las ventajas sinérgicas de los diferentes materiales estructurales para mejorar las propiedades del biocatalizador. En esta tesis, hemos estudiado la síntesis de nanopartículas nanohíbridas [silica biomimética + nanopartículas magnéticas (MNPs)] como un vehículo de peroxidasa de rábano picante (HRP) para su uso en la terapia dirigida de profármacos de enzimas (DEPT). DEPT usa enzimas exógenas introducidas artificialmente en el cuerpo con el objetivo de convertir profármacos en su forma activa en el sitio deseado del organismo y, por lo tanto, tiene una ventaja sobre las terapias estándar no específicas por su selectividad potencial. El uso de enzimas enzapsuladas o unidas a nanopartículas (NPs) ha ganado interés en este tipo de terapia ya que permite mejorar la estabilidad y reducir la inmunogenicidad de la enzima terapéutica, a la vez que mejorar la bioacumulación y retención de la enzima en el tejido tumoral. Esto es debido al efecto de retención y permeabilidad aumentada (EPR) por la presencia de una abundante vascularización inmadura en el tumor que hace que de forma pasiva se acumulen mayor cantidad de NPs que en tejidos sanos. Por tanto, las enzimas terapéuticas transportada por NPs tienen una mayor facilidad en acumularse en tejidos tumorales que la misma enzima administrada en forma libre. Sin embargo, las NPs no pueden evitar la distribución no selectiva en el cuerpo, sobre todo en órganos que para filtrar la sangre también usan el efecto EPR (hígado, bazo)., Por lo tanto, el uso exitoso de modalidades de DEPT que utilicen nanomateriales como vehículos de la enzima terapéutica depende de la implementación de estrategias que puedan proporcionar un control espaciotemporal sobre su actividad enzimática. Para esto, en esta Tesis se ha optimizado la co-encapsulación de MNP junto con la enzima terapéutica HRP en nanopartículas de sílica biomimética. Se eligió HRP debido a su capacidad para convertir el ácido indol-3-acético (hormona vegetal) en radicales peroxilados con actividad citotóxica en varias líneas celulares tumorales. Sin embargo, la HRP tiene una temperatura óptima de 45ºC y a la temperatura corporal su actividad biocatalítica no es máxima. Debido a que las MNPs pueden absorber energía en presencia de un campo magnético alterno externo (AMF) y liberarla al medio ambiente en forma de calor, nuestra hipótesis de trabajo se centra en lograr utilizar este calor para 3 alcanzar la temperatura óptima de la HRP de forma localizada (en una escala nanométrica), y así incrementar su actividad enzimática. Tradicionalmente, el desencadenar el aumento macroscópico de la temperatura de tejidos tumorales ha sido la estrategia más difundida para utilizar el gradiente muy alto de temperatura que se genera por calentamiento magnético en pocos nanómetros desde la superficie de la MNPs. Esto ha permitido el desarrollo de una nueva terapia para el tratamiento del cáncer, conocida como hipertermia magnética (MHT), ya que las células tumorales tienen una mayor sensibilidad al calor que las células normales. En esta Tesis sin embargo proponemos aprovechar este gradiente de temperatura localizado alrededor de las MNPs no para desencadenar que se eleve de forma macroscópica la temperatura de un tejido tumoral sino para modular la aactividad de enzimas co-entrapadas con las MNPs en nanohíbridos de sílica biomimética. El uso de silica biomimética en el nanohíbrido se planteó bajo la premisa de utilizar un medio para estabilizar la enzima, que permita su probable inmunoaislación y, como se demostró más tarde, alcanzar a su vez un posible efecto de aislamiento térmico que disminuya la rápida disipación de calor que ocurre a pocos nm de la superficie de las MNPs y así sea posible modular la actividad de enzimas que no se encuentren unidas a la superficie de las MNPs. Es así que efectivamente la estrategia de co-encapsulación facilitó un aumento selectivo en la actividad biocatalítica de HRP al alcanzar localmente su temperatura óptima. Para llegar a esta conclusión, hemos optimizado la síntesis de los nanohíbridos variando diferentes parámetros tales como: i) tamaño del catalizador aminado requerido para la formación de NP de sílica (polietilenimina MW ~ 1300, 2000, 25000, 60000 Da), ii) metodología de inmovilización de la HRP (adsorción / unión covalente en MNP antes de la encapsulación, co-encapsulación con MNP), iii) relación HRP/ MNP. La inmovilización de HRP y los rendimientos de actividad después del atrapamiento son similares con o sin MNP (~ 60 ± 4% y 78 ± 1, respectivamente). Sin embargo, la estabilidad térmica de la enzima (50ºC) aumentó 532 (Factor de estabilización) cuando la HRP se co-encapsula con MNP. El uso de PEI de diferentes tamaños no afectó los rendimientos de la inmovilización de HRP ni el tamaño (diámetro hidrodinámico) de los híbridos. Sin embargo, las MNP parecen actuar como plantillas para la deposición de sílice ya que su co-encapsulación con HRP reduce el tamaño radio de los biohíbridos (de ~ 800-1000 a 500-600 nm). Todos los nanohíbridos obtenidos pudieron oxidar IAA, pero no se observó citotoxicidad en 4 ausencia del profármaco usando diferentes líneas celulares. También hemos estudiado el efecto del tamaño del catalizador aminado requerido para la formación del nanobiocatalizador (polietilenimina MW ~ 1300, 2000, 25000, 60000 Da), en su eficiencia de calentamiento magnético macroscópico (SAR, Specific Absorption Rate o tasa de absorción específica) y nanoscópico (activación enzimática). Los nanohíbridos (500-600 nm) mostraron activación enzimática (250-300%) sin un aumento significativo de la temperatura macroscópica y conservaron su actividad completa después de la aplicación de un ciclo de AMF (829 kHz, 25.2 mT) durante 20 min. Sin embargo, para que los nHs desarrollados puedan tener potencial para ser utilizados en DEPT es clave que presenten buena estabilidad coloidal en medios biológicos complejos. Para ello fue necesario bloquear grupos amino primarios presentes en la superficie de los mismos con glucosa. Este monósocaráido no solo permitió mejorar la estabilidad coloidal de los nHs en medio de cultivo completo, pero también sirve de biomoléculta para vectorización activa de células tumorales. Los nanohíbridos funcionalizados con glucosa lograron diferencias significativas en la reducción de la viabilidad celular de una línea celular de cáncer de colon humano (HCT-116) después de la exposición a un ciclo de AMF (80% versus 30% de reducción en controles sin aplicación de AMF). Estos resultados muestran por primera vez que es posible controlar de forma remota la actividad de HRP co-atrapada con MNP mediante calentamiento magnético para una potencial aplicación en terapia antitumoral DEPT. The main goal of this mini-review is to provide an updated state-of-the-art of the hybrid organic-inorganic materials focusing mainly on interface phenomena involving ionic and electronic transport properties. First, we review the most relevant preparation techniques and the structural features of hybrid organic-inorganic materials prepared by solution-phase reaction of inorganic/organic precursor into organic/inorganic hosts and vapor-phase infiltration of the inorganic precursor into organic hosts and molecular layer deposition of organic precursor onto the inorganic surface. Particular emphasis is given to the advances in joint experimental and theoretical studies discussing diverse types of computational simulations for hybrid-organic materials and interfaces. We make a specific revision on the separately ionic, and electronic transport properties of these hybrid organic inorganic materials focusing mostly on interface phenomena. Finally, we deepen into mixed ionic-electronic transport properties and provide our concluding remarks and give some perspectives about this growing field of research. Salto district, a key horticultural area in Uruguay, has crucial water sources such as the Guarani Aquifer System, the Salto-Arapey Aquifer System (SAAS), and the San Antonio Creek (SAC). Agricultural and livestock activities have increased in recent years, leading to demographic growth and housing expansion. Concerns about water quality arise due to limited studies on environmental exposure to harmful chemicals and microbiological factors from agriculture, inadequate wastewater treatment, and septic tank seepage in rural areas. The present study collected hydrochemical background and new sampling data for hydrogeochemical and water quality evaluation (28 parameters) from SAAS (73 wells) and SAC (6 sites). Results indicated groundwater from SAAS is mainly sodium bicarbonate type, RAS index shows low saline waters and microbiological contamination were detected in half of the sampled wells, As levels (4.0–49.3 μgL−1), total phosphorus (P) (50–178.5 mg/L), and nitrate (NO− 3 ) (7.0–168.5 mg/L). SAC hydrochemistry revealed high levels of P (77.5–163.5 μg/L), which could be one of the factors that promote the appearance of signs of eutrophication towards the mouth of the Uruguay River. Spatial distribution of total arsenic (As) in SAAS and water quality evaluation of SAC assess future human health risks. These results represent a challenge to foster Uruguayan water education and raise public awareness about the importance of using safe water for human consumption and irrigation. In this work, an upflow anaerobic packed bed reactor configuration to produce hydrogen using cheese whey as the substrate was tested. The microbiological composition was linked to the reactor operation.<br /><br />Three different organic loading rates of 22, 33, and 37 g COD/L-d were used at a fixed hydraulic retention time of 24 h. The increase of the organic loading rate from the initial value of 22 g COD/L-d to a value of 37 g COD/L-d and the adjustment of the pH to values greater than 5 had a positive effect on the hydrogen production and values of up to 1 L H2/L-d were achieved. The production of hydrogen was stable under all of the investigated conditions, and the problems frequently reported in this kind of reactors (bed clogging, methanogenesis and solvent production) were not observed during operation.<br /><br />The highest hydrogen yield was 1.1 mol H2/mol lactose far from the maximum theoretical value (8 mol H2/mol lactose), but similar to previous works using cheese whey as substrate. In this sense, hydrogen produced through steam reforming of a biomass-derived biofuel, such as bio-oil, appears as an interesting energetic vector. The aim of this work is the hydrogen production through steam reforming of small organic model compounds present in the aqueous fraction of bio-oil (ethanol, acetic acid, acetone) and, for a better approximation to a real system, emulate such aqueous fraction by a representative mixture of these. Ternary mixed oxides Ni-La-Me catalysts (Me = Ce, Ti, Zr) were prepared, characterized through X-ray diffraction, N2 adsorption–desorption isotherms, thermogravimetric analysis, cyclic voltammetry, and scanning electron microscopy, and tested in the steam reforming reactions. Results indicate that, due to the intrinsic H2O<br />adsorption capacity of ZrO2 as well to a higher number of oxygen vacancies in the catalyst structure, Ni-La-Zr presents interesting properties for its potential applicability as industrial catalyst for hydrogen production through steam reforming. AB2 alloys have been extensively studied as Ni-MH battery anodes. In the hydriding process, hydrogen diffusion inside the crystal lattice is the determining step, so a comparison between gas phase and electrochemical environment was carried out. As concern, we have found structural changes as a consequence of hydriding in terms of segregation of different stable phases and variations in unit cell parameters in each alloy. Maximum hydrogen storage capacity and hydrogen molar volume for studied alloys were evaluated by PCT curves at different temperatures. Resulting values were compared with electrochemical PCT (ECT plots). Results obtained for both methods were similar, with a hydrogen molar volume value close to 30 cm3 mol-1 being higher for AB2M0 in accordance with the reported capacity values. Diffusion time constants were determined from GITT, achieved data were similar to previously reported by EIS and HRD. AB2M1 alloy showed the best kinetic behavior, depicting a time constant value an order of magnitude lower than AB2M0. This fact could be attributed to the diffusion coefficient and/or the hydrogen diffusion length mean path. No significant differences in thermodynamic parameters were observed between both alloys, therefore, the diffusion coefficients should be similar. The existent research literature of Dye Sensitized Solar Cells (DSSC) is mainly focused on the dye but there are very few studies focused in the semiconductor. Here, we present the performance of hydrogen titanate nanotubes (HTNT), showing an enhancement of the solar cell efficiency from 5.95% to 7.63%, respect to anatase as photoelectrode. This enhancement could be explained in terms of open-circuit voltage, directly associated to the value of the HTNT band gap (Eg = 3.3 eV), the relative position of the edge conduction band; and the increase of the short-circuit current, directly associated to the enhancement of the specific area of the photoelectrode. All these characteristics are in very good concordance with our theoretical modelling of the system, based on First Principles calculations. We expect that this system could be further characterized, utilizing different dyes or absorbers, in order to evaluate the potential use of hydrogen titanate nanotubes for solar cell applications. The production of H2-rich gaseous mixtures from steam and oxidative steam reforming of crude glycerol was investigated on catalysts with general formula Ni(12 wt.%)-La2(Ce1-xZrx)2O7 (x = 0, 0.5, 1). The catalysts were prepared by the polymerized complex method based on the Pechini-type reaction route and characterized by a variety of physical and spectroscopic techniques. Samples were calcined in air at 850 °C for 4 h. All catalysts showed a well-defined NiO phase and a mixed oxide with a general formula A2B2O7 (A: La; B: Ce and/or Zr). NiO was completely reduced to metallic Ni on all catalysts under pure H2 at 650 °C. Ni reducibility is favored by the presence of Ce. Redox properties of Ce can also explain a synergetic effect leading to the higher basicity of the catalyst containing Ce and Zr (x = 0.5). Catalytic tests were performed at 650 °C by feeding the fixed-bed tubular reactor with a crude glycerol:water solution containing 30 wt% glycerol. The best results were achieved on the catalyst displaying the highest basicity (Ni-La2(Ce0.5Zr0.5)2O7 both under steam and oxidative reforming conditions. Removal of carbon and alkali metals by simple procedures allows to a significant recovery of the initial catalytic activity without changes in their chemical and phase structure. Herein, we investigate the chemisorption of hydrogen on double wall carbon nanotubes �DWCNT� employing density functional theory and periodic boundary conditions. In agreement with recent investigations based on Lennard-Jones potentials, we found that the �n,m�@�n+9,m� combination is favored for tubes with small diameters. The C–H binding energies determined for the �16,0� single wall carbon nanotubes �SWCNT� are nearly identical to those computed for the �7,0�@�16,0� and �8,0�@�16,0� DWCNTs. For both of the latter we found that interlayer interaction modifies the band structure of the inner tube. In the case of hydrogenated DWCNTs, the electronic structure of the inner tube experiences very small changes at high coverages �50%�. However, at lower hydrogen coverages �3%–25%� changes are observed in the electronic structure of the inner tube. In agreement with recent experimental results we conclude that, for heavily functionalized DWCNTs, the electronic properties of the inner tube remain unchanged. For zigzag SWCNTs, the band gap becomes larger upon increase in hydrogen coverage; at 50% of coverage the hydrogenated �16,0� SWCNT has a band gap of 3.38 eV. First-principles density functional theory calculations are performed to study the stability and electronic properties of hydrogenated and fluorinated two-dimensional sp3 boron phosphide (BP) and boron arsenide (BAs). As expected, the phonon dispersion spectrum and phonon density of states of hydrogenated and fluorinated BX (X = P, As) systems are found to be different, which can be attributed to the different masses of hydrogen and fluorine. Hydrogenated BX systems bear larger and indirect band gaps and are found to be different from fluorinated BX systems. These derivatives can be utilized in hydrogen storage applications and ultrafast electronic devices. Finally, we investigated the stability and electronic properties of stacked bilayers of functionalized BP. Interestingly, we found that these systems display strong interlayer interactions, which impart strong stability. In contrast with the electronic properties determined for the fluorinated/hydrogenated monolayers, we found that the electronic properties of these bilayers can finely be tuned to a narrow gap semiconductor, metallic or nearly semimetallic one by selecting a suitable arrangement of layers. Moreover, the nearly linear dispersion of the conduction band edge and the heavy-, light-hole bands are the interesting characteristics. Furthermore, the exceptional values of effective masses assure the fast electronic transport, making this material very attractive to construct electronic devices. Background: Scintigraphic studies have been performed to assess the release, both in vitro and in vivo, of radiotracers from tablet formulations. Four different tracers with differing physicochemical characteristics have been evaluated to assess their suitability as models for drug delivery. Methods: In-vitro disintegration and dissolution studies have been performed at pH 1, 4 and 7. Invivo studies have been performed by scintigraphic imaging in healthy volunteers. Two hydrophilic tracers, (99mTc-DTPA) and (99mTc-MDP), and two lipophilic tracers, (99mTc-ECD) and (99mTc-MIBI), were used as drug models. Results: Dissolution and disintegration profiles, differed depending on the drug model chosen. In vitro dissolution velocity constants indicated a probable retention of the radiotracer in the formulation. In vivo disintegration velocity constants showed important variability for each radiopharmaceutical. Pearson statistical test showed no correlation between in vitro drug release, and in vivo behaviour, for 99mTc-DTPA, 99mTc-ECD and 99mTc-MIBI. High correlation coefficients were found for 99mTc-MDP not only for in vitro dissolution and disintegration studies but also for in vivo scintigraphic studies. Conclusion: Scintigraphic studies have made a significant contribution to the development of drug delivery systems. It is essential, however, to choose the appropriate radiotracers as models of drug behaviour. This study has demonstrated significant differences in release patterns, depending on the model chosen. It is likely that each formulation would require the development of a specific model, rather than being able to use a generic drug model on the basis of its physicochemical characteristics. The derivatives were characterized by kinetics parameters, co-solvent (ethanol and acetone) and temperature stability. Hydrophilization achieved important co-solvent stabilization in all cases. One of the most remarkable results obtained was a 25-fold increase in the half-life of the A. oryzae Glut90-Pal-Pam derivative in 50% (v/v) acetone. Stabilization achieved in very drastic co-solvent concentrations is directly related to the hydrophilization of the nano-environment. The KM values show that the hydrophilic shell appears to behave as an open structure and may create a “partition effect” that protects the enzymes from denaturation. SnS2 has adequate properties for photovoltaic applications in solar cells. For this purpose, we studied the influence of the capping agents pyridine and aniline in the nanoparticles hydrothermal syntheses. These capping agents are suitable for use in hybrid organic- inorganic solar cells. Different ratios of these agents with respect to the metal precursor were tested. Hexagonal and disc-type nanoparticles were obtained. Lower concentrations of capping agent increased the proportion of hexagons, while diminishing their size. The obtained products are adequate in size, dispersion of size, and stabile in suspension in chloroform to use them in hybrid solar cells. Moreover, the use ofpyridine allowed for more stable suspensions, and therefore better results for this application. We synthesized HgS nanostructures by the hydrothermal method in order to use them as electron acceptors in hybrid organic-inorganic solar cells. We employed different mercury sources (HgO and Hg(CH3COO)2) and polyvinylpyrrolidone (PVP) or hexadecanethiol (HDT) as stabilizing/capping agent for controlling size, crystallinity, morphology and stability of the obtained nanostructures. We also used thiourea as sulfur source, and a temperature of 180 °C during 6 h. Synthesized nanostructures were characterized by powder X-Ray Diffraction, Diffuse Reflectance Infrared Fourier Transform and Transmission Electron Microscopy. When PVP acts as stabilizing agent, the mercury source has influence on the size -but not in morphology- of the beta-HgS obtained nansostructures. HDT has control over nanostructures’ size and depending on the relation Hg:HDT, we obtained a mixture of alpha and beta HgS which can be advantageous in the application in solar cells, due their absorption in different spectral regions. The smallest nanostructures obtained have a mean diameter of 20 nm when using HDT as capping agent. Also, we deposited the aforementioned nanostructures onto flat glass substrates by the spin coating technique as a first approach of an active layer of a solar cell. The depositions were characterized by atomic force microscopy. We obtained smaller particle deposition and higher particle density -but a lower area coverage (5%) – in samples with HDT as capping agent. This work presents promising results on nanostructures for future application on hybrid solar cells. Further efforts will be focused on the deposition of organic-inorganic layers. A rod-shaped and Gram-stain-negative bacterial strain, 1BT, was isolated from an air sample collected at King George Island, maritime Antarctica. Strain 1BT is strictly aerobic, psychrophilic, catalase-positive, oxidase-positive and non-motile. Growth of strain 1BT is observed at 0–20 °C (optimum, 10 °C), pH 6.0–8.0 (optimum, pH 8.0) and in the presence of 0–1.0% NaCl (optimum, 0.5 % NaCl). Phylogenetic analysis based on 16S rRNA gene sequences places strain 1BT within the genus Hymenobacter and shows the highest similarity to Hymenobacter antarcticus VUG-A42aaT (97.5 %). The predominant menaquinone of strain 1BT is MK-7 and the major fatty acids (>10 %) comprise summed feature 3 (C16 : 1  ω7c and/or C16 : 1  ω6c; 32.5 %), iso-C15 : 0 (17.6 %) and anteiso C15 : 0 (12.3 %). The polar lipid profile consists of the major compounds phosphatidylethanolamine, phosphatidylserine, two unidentified aminolipids and one unidentified phospholipid. The DNA G+C content based on the draft genome sequence is 61.2 mol%. Based on the data from the current polyphasic study, 1BT represents a novel species of the genus Hymenobacter , for which the name Hymenobacter artigasi sp. nov. is suggested.
