We report a new procedure for the synthesis of aliphatic and aromatic beta-methoxy enones by a two-carbon homologation of carboxylic acids using previously unknown methoxydiazoethane. These substrates are of general interest as protected beta-keto aldehydes substrates for hetero-Diels-Alder reaction reagents for the synthesis of gamma-pyrones trans trans-dienones |b or potential synthons for the direct preparation of regioselectively substituted ketones from carboxylic acids (by alkylation of the available alfa-position hydrolysis and deformylation)

Abstract
A summary of the resource.
Submicron peanut oil-in-water emulsion stabilised with whey proteins, was processed by ultra-high-pressure homogenisation at 200 MPa (first-stage, 2-pass homogenisation) and an initial emulsion temperature (Tin) of 24 °C. Retinyl acetate (RAC) was selected as a model of a lipophilic biomolecule to be carried by submicron droplets. For comparison, micelles of retinyl acetate (RAC-micelles) were prepared at atmospheric pressure using polysorbate 80 (Tween® 80) as emulsifier. Both types of bio-particles displayed submicron diameters. Their behaviour was investigated on TC7-cell monolayers to characterise their influence on monolayer integrity (TER measurements), cell metabolic activity (MTT-assay) and cell membrane integrity (LDH-leakage). TC7-cell exposure to submicron droplets UHPH-elaborated as shuttles for RAC, and stabilised with proteins, did not impair TC7-cell viability compared with control cells (without deposit) or cells exposed to RAC-micelles. Such results demonstrated UHPH-processing innocuity in terms of Novel Food Regulation and implementation of UHPH as a novel technology. TC7-cell internalisation of whey proteins coating submicron oil droplets was observed using confocal microscopy and fluorescent probes. Cellular uptake of RAC was determined for both systems, after extraction from TC7 cells and HPLC quantitation. Results revealed RAC incorporation into TC7 cells and cellular turn-over of RAC into retinol for both systems without previous in vitro digestion. RAC bioaccessibility appeared yet better in the form of RAC-micelles than after entrapment into oily submicron droplets. Nevertheless, submicron droplets stabilised by proteins displayed a better physical stability than did RAC-micelles, which is the main feature for further developments of carrier systems.

Three novel Re(IV) compounds, the mononuclear complex Bu4N[ReBr5(Hpyzc)] (1) and the heterobimetallic complexes [ReBr5(l-pyzc)M(dmphen)2]·2CH3CN [M = Co (2), Ni (3)] (Hpyzc = 2-pyrazinecarboxylic acid, dmphen = 2,9-dimethyl-1,10-phenanthroline), have been synthesized and their crystal structures determined by single-crystal X-ray diffraction. The structure of 1 consists of [ReBr5(Hpyzc)]− complex anions and tetrabutylammonium cations, Bu4N+. TheRe(IV) is surrounded by five bromide anions and a N-donor Hpyzc monodentate ligand, in a distorted octahedral environment. The structures of 2 and 3 consist of dinuclear units [ReBr5(l-pyzc)M(dmphen)2], with the metal ions linked by a pyzc bridge ligand, being bidentate toward M(II) and monodentate toward Re(IV). The environment of Re(IV) is the same as in 1, whereasM(II) is six-coordinate, being surrounded by four nitrogen atoms of two bidentate dmphen ligands and one oxygen atom and one nitrogen atom of the pyzc anion. The magnetic properties of 1–3 were investigated in the temperature range 2.0–300 K. 1 shows the expected magnetic behavior for a mononuclear Re(IV) complex with a weak intermolecular antiferromagnetic coupling at low temperatures. The bimetallic complexes exhibit an intramolecular ferromagnetic coupling between Re(IV) and the M(II) ion (Co, Ni).

A summary of the resource.
The reaction of CrCl36H2O with the ligand bis(3,5-dimethylpyrazol-1-yl)methane (L) yielded the cationic complex [(Cr(L)(H2O)2Cl2]+, which crystallized as the chloride trihydrate [(Cr(L)(H2O)2Cl2]Cl·3H2O. The chromium complex was characterized by elemental analysis, electrical conductivity, Infrared and Ultraviolet/Visible spectroscopy. The crystal structure determination using single-crystal X-ray diffraction showed a chromium center in a distorted octahedral coordination sphere. In the crystal, the packing was directed by OH⋯(O,Cl) hydrogen bonds and weak CH⋯O interactions to build a monoclinic P21/c supramolecular structure. The complex showed excellent properties as an initiator for the ring opening polymerization of є-caprolactone (CL) under solvent-free conditions. The obtained polymer showed high crystallinity (89.9%) and a decomposition temperature above 475 °C. In addition, the new complex was evaluated against epimastigotes from Trypanosoma cruzi (T. cruzi) strains. The results indicated that this complex has a high activity against this parasite with a minimum inhibitory concentration 50 (MIC50) of 1.08 μg/mL. Interestingly, this compound showed little effect on erythrocytes, indicating that it is not cytotoxic. These results provide interesting contributions to the design of metal complexes by using simple and accessible ligands with activity against T. cruzi and with potential applications in the polymerization of CL.

El presente proyecto tiene como objetivo optimizar las metodologías analíticas para evaluar la presencia de metales y semimetales tóxicos en la matriz cabello a los efectos de ser utilizado como matriz biológica para la determinación de elementos tóxicos, evaluando su desempeño como posibles biomarcadores de exposición en población presuntamente expuesta. Para ello, en primera instancia, se puso a punto una metodología para el monitoreo y tratamiento de tóxicos metálicos y semimetálicos en una matriz compleja como lo es el cabello. Se optimizaron las metodologías para el tratamiento de la muestra y para las determinaciones de los elementos en estudio se utilizó la técnica de espectroscopía de absorción atómica. Se logró desarrollar un protocolo de digestión para determinación de cuatro elementos, plomo, mercurio, arsénico y manganeso, en forma simultánea y su posterior validación. Luego se realizó un estudio en población presuntamente expuesta a los tóxicos estudiados en el asentamiento Aquiles Lanza, con un enfoque basado en los pilares de investigación de la Ecosalud: transdisciplinaridad, participación social, pensamiento sistémico, equidad social y de género, sustentabilidad y acción. En el estudio poblacional, se encontraron niveles de plomo cuantificables, mientras que del resto de los analitos, si bien se detectaron, los niveles estuvieron por debajo de los límites de cuantificación. Del análisis estadístico, surge que podrían existir correlaciones con el sexo de los niños entre otras variables estudiadas. El estudio realizado, ha contemplado una integración global, utilizando los principios de la Ecosalud. Es el primer proyecto en Uruguay que plantea desde un comienzo, la integración de factores ambientales, sociales, toxicológicos y analíticos sentando las bases para futuros proyectos

A summary of the resource.
<span>An analytical method for sample preparation and subsequent determination of Cd, Pb and Se in beef using aerosol dilution by inductively coupled plasma-mass spectrometry was developed. The use of high matrix introduction technology allowed the introduction of a complex sample into the plasma with higher content of total dissolved solids a residual acidity of 4% and better tolerance to matrix effects. Trueness was verified by the analysis of two reference materials. At the 95% significance level, results were statistically equivalent to the certified or informed values. Analytical precision expressed as relative standard deviation was less than 9% in all cases. After validation, a total of 12 beef samples from Uruguay were analyzed. Obtained levels for Cd and Pb were far below the maximum limits established by international regulations in all samples. Also, results showed that beef from this region is a good source of Se in the range 0.120–0.870 mg kg</span><span>−1</span><span> with an interesting contribution to the daily intake of this essential element.</span>

Se determinó el contenido de selenio (Se) antes y después del proceso de cocción en tres cortes de carne vacuna (asado, cuadril y bola de lomo) provenientes de 20 animales. Los distintos cortes fueron sometidos a dos puntos de cocción. Cada corte fue dividido en varias porciones y se realizó la cocción en plancha y horno. El tratamiento de las muestras consistió en una digestión asistida con microondas y posterior determinación de Se mediante espectrometría de emisión atómica con plasma inducido por microondas acoplado a generación de hidruros (HG-MIP OES). El método analítico propuesto fue validado, resultó adecuado para el fin definido y presentó ventajas respecto a los métodos habitualmente utilizados en los laboratorios de análisis. Los puntos de cocción se establecieron evaluando la pérdida de peso (desde 8 hasta 35%). Los resultados indican que la carne vacuna aporta entre 20 y 26% del requerimiento diario de Se de un adulto. Este porcentaje se mantiene sin cambios significativos aun luego de una pérdida de peso de hasta un 35%, por lo que se concluye que la cocción no influye en el aporte de selenio a la dieta.

Ultrasound‐assisted extraction (UAE) methods for food analysis are recognized as being in accordance with the principles of green analytical chemistry (GAC). Reference standard methods generally use microwave‐assisted extraction (MAE) for sample digestion; however, the use of ultrasonication processes has many advantages. Herein, a simple, and economical method for the determination of Cd and Pb in fruits and vegetables for food safety monitoring is reported. This method was optimized using a standard reference material (SRM, spinach leaves) through a multivariate experimental design (optimal conditions: 0.5 g of sample, 15 min of sonication, and 20.00 mL of 2.5 mol L−1 HNO3) and subsequently validated. The limits of detection obtained were one or two orders of magnitude lower than those achieved with MAE, making this a promising technique for contaminants monitoring. The UAE method was then tested on vegetable samples (apples, carrots, tomatoes, and lettuce) obtained from commercial sources in Uruguay and a comparison was made with samples treated by MAE. Both processes yielded comparable results, and the levels of Cd and Pb in all samples were below the maximum allowed concentration (MAC) established by the World Health Organization (WHO). In addition, to provide insight into the mechanism by which the ultrasonic process aids extraction, confocal optical images of the surface of the SRM were obtained. The leaves were examined after treatment at different sonication times (5–20 min). The confocal microscopy images show the effects of the ultrasonic treatment on the plant tissue, including desaturation, erosion, and abrasion, which increased with increasing sonication time. In conclusion, the UAE method meets and overcome the requirements of a standard method for food safety assurance.

Abstract

Motor neuron degeneration and neuroinflammation are the most striking pathological features of amyotrophic lateral sclerosis (ALS). ALS currently has no cure and approved drugs have only amodest clinically therapeutic effect in patients. Drugs targeting different deleterious inflammatory pathways in ALS appear as promising therapeutic alternatives. Here, we have assessed the potential therapeutic effect of an electrophilic nitroalkene benzoic acid derivative, (E)-4-(2-nitrovinyl) benzoic acid (BANA), to slow down paralysis progression when administered after overt disease onset in SOD1G93A rats. BANA exerted a significant inhibition of NF-κB activation in NF-κB reporter transgenicmice and microglial cell cultures. Systemic daily oral administration of BANA to SOD1G93A rats after paralysis onset significantly decreased microgliosis and astrocytosis, and significantly reduced the number of NF-κB-p65-positive microglial nuclei surrounding spinal motor neurons. Numerous microglia bearing nuclear NF-κB-p65 were observed in the surrounding of motor neurons in autopsy spinal cords from ALS patients but not in controls, suggesting ALS-associated microglia could be targeted by BANA. In addition, BANA-treated SOD1G93A rats after paralysis onset showed significantly ameliorated spinal motor neuron pathology as well as conserved neuromuscular junction innervation in the skeletal muscle, as compared to controls. Notably, BANA prolonged post-paralysis survival by ~30%, compared to vehicle-treated littermates. These data provide a rationale to therapeutically slow paralysis progression in ALS using small electrophilic compounds such as BANA, through a mechanism involving microglial NF-κB inhibition.

Al administrar un medicamento a un individuo se busca producir una respuesta clínica específica a través de un efecto farmacológico determinado en gran parte por la respuesta farmacocinética, es decir, por las cantidades de fármaco presentes a lo largo del tiempo en los diferentes tejidos y fluidos del cuerpo humano. Medicamento y organismo conforman un complejo sistema que debe ser correctamente interpretado para cumplir con los objetivos terapéuticos de eficacia y seguridad al diseñar un tratamiento farmacológico. La natural variabilidad fisiológica que presentan los individuos a lo largo de una población exige racionalizar la instauración de los mismos, ya que posee un gran impacto en el destino de un fármaco administrado. El sexo como característica fisiológica divide a la población en dos mitades, su relevancia en el campo de la medicina es altamente notoria, hombre y mujer al interaccionar con un medicamento componen sistemas diferentes. Es de esperar entonces que ciertas características específicas al sexo del paciente puedan condicionar el éxito de un tratamiento farmacológico. Durante esta tesis doctoral se analizó la influencia del sexo en la respuesta farmacocinética de principios activos correspondientes a diversos grupos farmacológicos dividiendo el análisis en dos bloques: su efecto sobre la evaluación de biodisponibilidad de medicamentos y su efecto sobre la disposición de fármacos. La diferente discriminación de marcas comerciales de medicamentos que pueden hacer hombres y mujeres, y sus consecuencias sobre las conclusiones de un ensayo de bioequivalencia promedio no habían sido analizadas hasta el momento en profundidad. El análisis realizado demuestra la importancia de tener en cuenta el sexo en la evaluación de bioequivalencia y de diseñar ensayos clínicos a modo de obtener conclusiones sobre ambos grupos. Los resultados hallados aumentan la evidencia respecto a diferentes respuestas farmacocinéticas en hombres y mujeres. Se incorpora la recirculación de fármacos como proceso de disposición relevante, y se introducen modelos no lineales de efectos mixtos para describir y cuantificar su contribución. Al indicar un medicamento, el sexo del paciente debe ser considerado ya que de acuerdo a las características del mismo podría tener un significativo efecto sobre el resultado clínico del tratamiento.

Although sex-related differences in gastrointestinal physiology have been vastly reported, its impact on drug oral<br />bioavailability and bioequivalence (product discrimination) is often ignored. On this work results from an<br />average bioequivalence study between tablets containing 600 mg of the antiretroviral efavirenz (EFV), carried<br />out with 14 healthy subjects (8 female and 6 men) in a randomized 2-period, 2-treatment crossover design,<br />are analyzed from a sex-based approach. Sequences were balanced within each sex group. Considering all<br />subjects, no differences were observed on EFV absorbed amount, as shown by the estimated 90CI of the AUC96<br />Test/Reference bioequivalence ratio (T/R): 0.950–1.05. However, results were not conclusive due to the 90CI<br />for CMAX T/R was 0.743–1.07. Over this parameter, a significant sex-by-formulation interaction was detected:<br />90CI CMAX T/R was 0.838–1.36 in women and 0.540–0.920 in men; with a 52% relative difference between<br />point estimates. Formulation differences were therefore evidenced only by male subjects. In vitro dissolution<br />and disintegration tests for both products were carried out in two aqueous media: A) SLS 0.25% and B) HCl/KCl<br />pH 1.2. T/R results for dissolution efficiency and tablet disintegration times of formulations in both A and B<br />media were highly correlated with CMAX T/R bioequivalence results observed in women and men respectively,<br />showing that a dissimilar gastrointestinal environment between sexes affected EFV oral absorption. This work<br />shows how sex-by-formulation interaction can affect bioequivalence conclusions. Sex effect on product discrimination<br />should be specially disclosed in bioequivalence studies, mainly for drugs aimed to be given to both sexes.

Background: The goal of this study was to develop an in vivo-in vitro (IVIV) correlation, both in men and women, which allows constructing a model to predict bioequivalence assessments for drugs with narrow absorption windows. Besides, pharmacokinetic and pharmacodynamic equivalences were also investigated. Furosemide was chosen as a prototype. Methods: Twelve healthy Caucasian volunteers (8 women and 4 men) participated in a relative bioavailability study. Two oral formulations [Lasix® (Reference, R) and Furosemide EFA® (Test, T)] were administered under fasting conditions. Urinary excretion of unchanged drug (PK), and of chloride, sodium and potassium (PD) wasmonitored throughout time. PK and PD parameters were calculated from each respective excretion rate versus time curve. In vitro dissolution testing of both formulations was carried out using the USP apparatus 2 and 4 with fixed and variable dissolution media. Results: T and R could be considered bioequivalent since the 90% confidence intervals for the T/R ratio of geometric means for the area under the urinary drug excretion rate versus time curve and for the maximum excretion rate were within the 0.80-1.25 bioequivalence interval. However, T had faster initial absorption and higher levels in women, while R displayed such characteristics in men. Closer IVIV correlations in women were obtained when apparatus 4 with variable biorelevant dissolution media were used [going from fasting state simulated gastric fluid to fasting state simulated intestinal fluid]. Since R had faster disintegration time than T, a shorter stay of R under gastric conditions was required in order to obtain a good IVIV correlation in men. Saluretic effect displayed a typical clockwise hysteresis loop for the PKPD correlation assessed through chloride-versus-furosemide urinary excretion rates. Even though a higher amount of furosemide was excreted with the urine in men, differences in the excretion of electrolytes between sexes were almost negligible. Conclusions: Sex-differences in the gastrointestinal transit of formulations, under fasting conditions, determined the extent and the rate of furosemide absorption. The prolongation of the absorption process by mean of slowing the gastric emptying would make the formulation more effective. The USP-4 apparatus with variable dissolution media was able to discriminate the formulations even between sexes, becoming a promissory in vitro dissolution testing to predict bioequivalence.
