Recombinant single domain antibodies (nanobodies) constitute an attractive alternative for the production of neutralizing therapeutic agents. Their small size warrants rapid bioavailability and fast penetration to sites of toxin uptake, but also rapid renal clearance, which negatively affects their performance. In this work, we present a new strategy to drastically improve the neutralizing potency of single domain antibodies based on their fusion to a second nanobody specific for the complement receptor CD11b/CD18 (Mac-1). These bispecific antibodies retain a small size (~30 kDa), but acquire effector functions that promote the elimination of the toxin-immunocomplexes. The principle was demonstrated in a mouse model of lethal toxicity with tetanus toxin. Three anti-tetanus toxin nanobodies were selected and characterized in terms of overlapping epitopes and inhibition of toxin binding to neuron gangliosides. Bispecific constructs of the most promising monodomain antibodies were built using anti Mac-1, CD45 and MHC II nanobodies. When co-administered with the toxin, all bispecific antibodies showed higher toxin-neutralizing capacity than the monomeric ones, but only their fusion to the anti-endocytic receptor Mac-1 nanobody allowed the mice to survive a 10-fold lethal dose. In a model of delayed neutralization of the toxin, the anti- Mac-1 bispecific antibodies outperformed a sheep anti-toxin polyclonal IgG that had shown similar neutralization potency in the co-administration experiments. This strategy should have widespread application in the development of nanobody-based neutralizing therapeutics, which can be produced economically and more safely than conventional antisera.

Twitter data is emerging as a source of insight regarding food-related consumer behaviour, and as such merit consideration regarding inclusion in the set of methodologies used by sensory and consumer researchers. Contributing to the evaluation of the pros/cons of Twitter as a research tool, we present a case study on the topic: “what people say when they tweet about different eating situations”. Rather than focusing on specific food/beverage products, we consider eating/drinking more widely, and hereby adopt a broader lens through which to evaluate this data gathering platform. Using the search words breakfast, lunch, dinner and snack, a total of 69,961 tweets were retrieved, of which 48,746 corresponded to original tweets and were subject to automated word analysis. However, manual content analysis of a subset of 16,000 tweets randomly selected provided more detailed insight. Mirroring results from past food choice research, information about what was consumed, when, where, with whom, and why was contained within the tweets, hereby confirming the potential of Twitter data as a source of information about food choice decisions. Despite access to free data, labour costs associated with data analysis were high and other limitations were also identified. While the content of tweets was largely free of written emotional expressions, emoticons were used with greater frequency. Some tweets also contained references to the (un)healthiness of foods/beverages being consumed and may be useful for exploring consumer perceptions of health/wellbeing, a topic which, like emotions, is currently attracting considerable attention. Overall, we find that Twitter data merit inclusion in the researcher’s toolbox, but that it is no panacea.

Astringency is one of the most important sensory characteristics of red wine. Although a hierarchically structured vocabulary to describe the mouthfeel sensations of red wine has been proposed, research on consumers' astringency vocabulary is lacking. In this context, the aim of this work was to gain an insight on the vocabulary used by wine consumers to describe the astringency of red wine and to evaluate the influence of wine involvement on consumers' vocabulary. One hundred and twenty-five wine consumers completed and on-line survey with five tasks: an open-ended question about the definition of wine astringency, free listing the sensations perceived when drinking an astringent wine, free listing the words they would use to describe the astringency of a red wine, a CATA question with 44 terms used in the literature to describe astringency, and a wine involvement questionnaire. When thinking about wine astringency consumers freely elicited terms included in the Mouth-feel Wheel, such as dryness and harsh. The majority of the specific sub-qualities of the Mouth-feel Wheel were not included in consumer responses. Also, terms not classified as astringency descriptors were elicited (e.g. acid and bitter). Only 17 out of the 31 terms from the Mouth-feel Wheel were used by more than 10% of participants when answering the CATA question. There were no large differences in the responses of consumer segments with different wine involvement. Results from the present work suggest that most of the terms of the Mouth-feel Wheel might not be adequate to communicate the astringency characteristics of red wine to consumers.

Herein, we employed first-principle calculations to study the structure and reactivity of dual-doped graphene. The new materials were derived from graphene by replacing two carbon atoms with one 2p element (B, N or O), and one 3p element (Al, Si, P or S). A total of 12 dual-doped graphenes were used to perform a comparative study and identify the most promising materials for the development of new catalysts and anchoring nanoparticles. The structural analysis indicated that in all cases, except SiB, the dopants prefer to replace a CC bond. Yet, not always the dopants are bonded, presenting an edge-like bonding. As regards chemical reactivity, in general the introduction of two atoms remarkably increases reactivity as compared to graphene. The most prominent examples are Al–O, S–N, P–O and Si–B(para) codoped graphene which present reactivity higher than perfect and monodoped graphenes. This effect was attributed to the strong charge redistribution induced by the dopants. In most cases the heteroatoms are the most reactive sites, but in others the carbon atoms are more reactive, as in Al–O and Si–B codoped graphene. The combination of 2p and 3p dopants offers the possibility of adjusting over a wide range the reactivity of graphene.<br /><br />

The C60H28 buckycatcher (BC) is an excellent host for fullerenes. This receptor features two corannulene pincers which trap C60/C70 via π stacking interactions. Although, the formation of C60@C60H28 complexes is readily observed, the dimerization of C60H28 is not a competitive process, even at high concentrations. By means of first principle calculations, we have studied the thermodynamics of the polymerization of BCs and the formation of fullerene complexes. The results obtained with the M06-2X, B97-D, B3LYP-D3BJ, PBE-D2, and PBE-D3 functionals indicated that the interaction energy of (C60H28)2 is larger than the one computed for C60@C60H28, by 8–10 kcal/mol. Because of the greater number of atoms, and due to the presence of more hydrogens, the inclusion of free energy corrections lowers the energetic separation between (C60H28)2 and C60@C60H28, even though the dimer maintains its position as being slightly more bound than that of the C60@C60H28. Our calculations indicated that up to the C60H28 trimer could be formed with a free energy change larger than that corresponding to the dimerization and fullerene complexation processes. Finally, we found that the inversion of the corannulene pincers attached to the cyclooctatetraene core is 2–3 kcal/mol lower than that corresponding to free corannulene. We expect that this work can motivate new investigations that may lead to the observation of C60H28 polymers. © 2015 Wiley Periodicals, Inc.

9,10-bis(1,3-dithiol-2-ylidene)-9,10-dihydroanthracene (exTTF) is commonly used as pincer in fullerene receptors. In order to understand how this molecule interacts with itself, and thus gain insight in the overall interaction in fullerene receptors which use exTTF as pincer, we studied the exTTF dimer at the M06-2X and ωB97XD levels of theory. The results indicate that this supramolecular complex displays an interaction energy which is larger than the one determined for corannulene. However, the most important difference between both fullerene pincers is not the interaction energy but the number of conformations available, which are larger for the exTTF-based dimer. In effect, at least four completely different structures of the exTTF dimer present interaction energies larger than that computed for corannulene. For this reason, exTTF is expected to: a) adopt strongly dispersion-bound conformations when it is used as pincer to construct fullerene receptors and b) intricate crystallographic structures when stacked with fullerenes.

Antifeedant and repellent effects of two different extracts from native Solanum species Solanum bistellatum and Solanum sisymbrifolium on Cryptotermes brevis were evaluated. The extracts obtained, particularly the dichloromethane extracts and the enriched fraction of sugar esters from S. sisymbrifolium and S. bistellaltum, showed antifeedant and repellent action against the termite. The antifeedant effect of dichloromethane extract from S. sisymbrifolium at the concentration of 25.00mg/mL reached 100%, while the repellent action of the dichloromethane extracts and that of the enriched fraction of sugar esters at the concentration of 1.00mg/mL was higher than 90%. In case of S. bistellatum, the antifeedant effect of the dichloromethane extract and the enriched fraction of sugar esters at the concentration of 25.00mg/mL was 43.29% and 57.46%, respectively. The repellent action of the dichloromethane extracts and of the enriched fraction of sugar esters at a concentration level of 2.50mg/mL was higher than 92%.

Los helmintos parásitos son un grupo de metazoarios muy diverso y complejo, cuya principal característica en común es su habilidad de generar infecciones crónicas en sus hospederos naturales. Dentro de las helmintiasis, la equinococosis quística (EQ) es una parasitosis causada por el estadío larvario de Echinococcus granulosus, y es considerada una zoonosis de gran relevancia por afectar tanto al ser humano como a una amplia variedad de especies ganaderas productivas. Interesantemente, la infección por E. granulosus es capaz de durar varios años de manera asintomática en sus hospederos intermediarios, por lo que se ha postulado que el parásito tiene la habilidad de modular la respuesta del hospedero en favor de su sobrevida. En este sentido, los fenómenos inmunológicos responsables de la resistencia o susceptibilidad del hospedero a la infección han sido escasamente estudiados. Por lo tanto, el presente trabajo se realizó a efectos de profundizar en el conocimiento de las respuestas inmunes potencialmente protectoras desencadenadas por hospederos intermediarios. Para ello, se realizaron estudios en humanos y modelos murinos focalizados en demostrar que las respuestas inmunes montadas por ciertos hospederos tendrían un papel crucial en limitar el establecimiento y/o desarrollo de la EQ. En primer lugar, se caracterizó el perfil serológico de una cohorte de individuos residentes en una zona endémica para EQ en Uruguay; que si bien fueron serológicamente positivos, no fueron diagnosticados imagenológicamente como pacientes con EQ durante un período de seguimiento de 2 años. En forma paralela, se caracterizaron también sendas cohortes de pacientes con EQ confirmada por cirugía y de donantes sanos. Los resultados mostraron que los individuos seropositivos pero normales por ultrasonido para EQ, presentaron un perfil serológico particularmente interesante, capaz de diferenciarlos de los dos grupos control. Asimismo, las características de dicho perfil sugieren que son capaces de mediar potenciales funciones efectoras antiparasitarias. This paper describes a non-destructive “peak parking” protocol in order to assess the axial heterogeneity of an in situ modified monolithic column for high performance liquid chromatography; a “gradient stationary phase” was designed whereby the ligand density decreases along the length of the rod in the “forward flow” configuration. The results of multi-location peak parking demonstrated a consistent increase in peak variance from the 1 cm position of the column to the 9 cm location. This increase in band broadening supported the theory of a decreasing ligand density along the length of this gradient column. This is consistent with efficiency measurements performed in both the forward and reverse flow directions, with an improved efficiency (15% increase in N m−1) in the reverse direction. These results are consistent with theoretical investigations into stationary phase gradients.

1. The uricosuric drug benzbromarone is extensively metabolized in man and two main metabolites are formed: the previously characterized 1″-hydroxybenzbromarone (metabolite M1) and an arylhydroxybenzbromarone (metabolite M2) of unknown structure. A dimethyl derivative was isolated from urine after methylation and was characterized by gas chromatography-mass spectrometry (g.l.c.-m.s.) and high resolution nuclear magnetic resonance spectroscopy as 4″-O-methyl-6-methoxybenzbromarone; the structure of M2 therefore is 6-hydroxybenzbromarone. 2. A minor metabolite was similarly characterized as 1″-oxobenzbromarone by comparison with authentic synthetic samples and is a product of biodegradation and not an artifact derived from the in vitro oxidation of 1″-hydroxybenzbromarone. Further minor metabolites were detected and were provisionally characterized by g.l.c.-m.s. after derivatization and include: 2″-hydroxybenzbromarone (an isomer of 1″-hydroxybenzbromarone); 1″,6-dihydroxybenzbromarone; dihydroxy-aryl-benzbromarone; and two structure isomers of 6-hydroxybenzbromarone. Debrominated metabolites were not detectable. 3. Benzbromarone is hydroxylated in vivo at the prochiral centre C1″to 1″-hydroxybenzbromarone; analysis of 1″-hydroxybenzbromarone from plasma and urine extracts by h.p.l.c. using a chiral column revealed that two peaks were eluted which showed a mean enantiomeric ratio of 2.1 for plasma and 7.3 for urine; these data demonstrate that the formation and elimination of this metabolite is enantioselective; the absolute configuration of the 1″-chiral centre is presently unknown.

A summary of the resource.
Several oral ivermectin (IVM) formulations for use in sheep are available in the pharmaceutical veterinary market in different countries. All of them are indicated at the same dose rate to treat the gastrointestinal nematodes. However, there is a lack of information on the relative systemic exposure (plasma bioavailability) and clinical efficacy among oral formulations routinely used in sheep. The main goal of the work reported here was to perform a pharmaco-parasitological assessment of three different IVM oral formulations in lambs infected with multiple resistant gastrointestinal nematodes. The comparative drug systemic exposure (IVM plasma concentrations) and nematodicidal efficacies (clinical efficacy) in lambs were determined for a reference (RF) and two different test (T1, T2) IVM oral formulations. One hundred and fifty six (n= 156) healthy Corriedale lambs, naturally infected with multiple resistant gastrointestinal nematodes were allocated into four experimental groups (n=39). Animals in each group received treatment (200 μg/kg) with either the RF, one of the test IVM formulations or were kept as untreated control. Blood samples were collected over 15 days post-treatment (n=8). The IVM plasma concentrations were measured by high performance liquid chromatography with fluorescence detection. The faecal nematode egg count reduction test (FECRT) (n=39) and evaluation of the clinical efficacy were performed at day 14 post-treatment (n=6), where a predominance of IVM highly resistant nematodes was observed. A rapid quantitative TLC method for the determination of glycoalkaloids in potato leaves and tubers is described. The method includes a microscale extraction, a simple clean-up step and quantification by TLC scanning. The method is valuable for the determination of glycoalkaloids when many samples have to be evaluated as in potato breeding programmes.

<p>The rate of the aerobic metabolism of pyruvic acid by bakers' yeast cells is determined mainly by the amount of undissociated acid present. As a consequence, the greatest rate of oxidation was observed at pH 2.8. Oxidation, at a slow rate, started at pH 1.08; at pH 9.4 there was no oxidation at all. The anaerobic metabolism, only a fraction of the aerobic, was observed only in acid solutions. There was none at pH values higher than 3.</p>
<p>Pyruvic acid in the presence of oxygen was oxidized directly to acetic acid; in the absence of oxygen it was metabolized mainly by dismutation to lactic and acetic acids, and CO<sub>2</sub>. Acetic acid formation was demonstrated on oxidation of pyruvic acid at pH 1.91, and on addition of fluoroacetic acid. Succinic acid formation was shown by addition of malonic acid. These metabolic pathways in a cell so rich in carboxylase may be explained by the arrangement of enzymes within the cell, so that carboxylase is at the center, while pyruvic acid oxidase is located at the periphery.</p>
<p>Succinic and citric acids were oxidized only in acid solutions up to pH 4. Malic and α-ketoglutaric acids were not oxidized, undoubtedly because of lack of penetration.</p>

<p>Oxidizing agents of sulfhydryl groups such as iodosobenzoate, alkylating agents such as iodoacetamide, and mercaptide-forming agents such as cadmium chloride, mercuric chloride, <em>p</em>-chloromercuribenzoate, sodium arsenite, and <em>p</em>-carboxyphenylarsine oxide, added in small concentrations to a suspension of sea urchin sperm produced an increase in respiration. When the concentration was increased there was an inhibition. These effects are explained by postulating the presence in the cells of two kinds of sulfhydryl groups: <em>soluble sulfhydryl groups</em>, which regulate cellular respiration, and <em>fixed sulfhydryl groups</em>, present in the protein moiety of enzymes. Small concentrations of sulfhydryl reagents combine only with the first, <em>thus</em> producing an increase in respiration; when the concentration is increased, the fixed sulfhydryl groups are also attacked and inhibition of respiration is the consequence.</p>
Other inhibitors of cell respiration, such as cyanide and urethanes, which do not combine with —SH groups, did not stimulate respiration in small concentration.

A rapid quantitative TLC method for the determination of glycoalkaloids in potato leaves and tubers is described. The method includes a microscale extraction, a simple clean-up step and quantification by TLC scanning. The method is valuable for the determination of glycoalkaloids when many samples have to be evaluated as in potato breeding programmes.

Experiments dealing with protein fractionation are typically included in biochemistry laboratory courses. This knowledge is important for basic and applied research purposes as well as for industrial production. Very often, teachers use artificial mixtures of proteins which differ substantially in some physicochemical properties (eg solubility, isoelectric point, molecular size). However, students prefer to start working from raw materials, since this gives them a feeling for the solving of a real task. Enzymes in cereal seeds have been investigated from a fundamental point of view for their significance in physiology, biochemistry, genetics and technology/ In this way, amylolytic enzymes are of practical importance in industrial process such as brewing, malting, and baking. In the brewing of beer, amylolytic enzymes (mainly a- and 13-amylases) participate at different stages of the process; their presence is involved in the production of fermentable sugars and limit dextrins, a-Amylase (EC 3.2.1.1.) attacks ct-1,4 glycosidic bonds in starch essentially at random and is properly classified as an endoenzyme.

A summary of the resource.
Se determinó la incidencia de la infección toxoplásmica en 449 mujeres de 15 a 40 años en Montevideo con la reacción de Sabin y Feldman. Se calculó que entre 2,5 y 4,2 de cada 1000 fetos se infectarian con T.gondii , sufriendo las consecuencias patológicas conocidas. Se comparan los resultados con los obtenidos en otros países. Se discuten los recursos para la prevención o atenuación del daño fetal por Toxoplasma : la educación sanitaria de la embarazada y la determinación del IgM total en sangre del cordón, seguida de la búsqueda de anticuerpos específicos para el grupo de infecciones prenatales (toxoplasmosis, citomegalovirus, rubéola, herpes, sífilis). Se resalta la importancia de proseguir las investigaciones sobre una vacuna para aplicar a las mujeres seronegativas que deseen quedar embarazadas y de la vacunación de los gatos contra la eliminación de ooquistes ya factible.

A summary of the resource.
Previous studies have demonstrated that oral administration to cats of tissue cysts of the oocyst-negative mutant strain of Toxoplasma gondii , T-263, induces immunity to oocyst shedding following challenge. Experiments were designed to compare the levels of protection induced by T. gondii T-263 when tissue cysts, bradyzoites released from tissue cysts, and tachyzoites were administered to cats. In 1 experiment, groups of cats received 2 oral doses of intact tissue cysts or released bradyzoites of T. gondii T-263 and were challenged 47 days later with the oocyst-producing strain of T gondii T-265. All cats seroconverted following immunization and none of them shed oocysts following challenge. In a second experiment, groups of cats received tachyzoites of T. gondii T-263 as a single oral dose and either 1 or 2 intraduodenal doses; they were challenged 60 days after the last vaccination. All cats seroconverted following immunization. Following challenge, all cats shed oocysts except for 2 of 7 cats that received 2 intraduodenal doses of tachyzoites. Thus, orally administered bradyzoites of T. gondii T-263, either contained in intact tissue cysts or liberated from cysts, induced immunity to oocyst shedding. In contrast, tachyzoites did not completely protect against oocyst shedding, even when delivered directly to the duodenum and despite the development of high antibody titers
