La academia ha reconocido la necesidad de tener en cuenta tanto las métricas ambientales como las económicas a la hora de desarrollar procesos que, en última instancia, encontrarán aplicación en la fabricación de productos químicos. Las aminas quirales son porciones de moléculas que constituyen un grupo de "estructuras preferidas" (“privileged scaffolds”) que son frecuentes en los productos naturales, fármacos, agroquímicos y otros productos químicos, y que se incluyen habitualmente en el diseño de bibliotecas para el descubrimiento de nuevas drogas. También se conoce que el 80 % de los 20 fármacos más recetados en medicinas son aminas quirales, lo que hace que estos compuestos sean valiosos componentes básicos en la industria farmacéutica. Esta tesis tiene como objetivo principal investigar nuevos biocatalizadores en síntesis para la producción de aminas quirales, utilizando tanto enzimas reportadas como nuevas enzimas aún no identificadas. Mediante análisis de motivos aminoacídicos conservados detectados en secuencias disponibles y análisis de motivos de iminoreductasas reportados en paralelo a nuestro trabajo, se realizó una búsqueda de nuevas IRED. Como resultado, se han encontrado varias secuencias que cumplen con las características para ser potenciales enzimas reductoras de iminas. Además, se aplicó un enfoque de “genome mining and motif análisis”, para la selección de potenciales secuencias R-ω-TA. A partir de 423 candidatos, la secuencia R-ω-TA de Capronia semiimmersa fue seleccionada, clonada y expresada en Escherichia coli . La enzima expresada demostró aceptar D-alanina así como también isopropilamina como grupo aminodonor y tener actividad sobre una variedad importante de cetonas. Ademas, ha sido demostrada la aplicación de una cascada quimioenzimática novedosa para la síntesis regio y estereoselectiva de un panel de pirrolidinas 2,5-disustituidas a partir de las correspondientes 1,4-dicetonas. Al mismo tiempo, se logró establecer un sistema de desracemización de aminas en una misma reacción, con cinco biocatalizadores actuando paralelamente de manera simple y práctica. De esta forma se logran obtener las aminas enantioméricamente puras a partir de una mezcla racémica o a partir de la amina con estereoquímica opuesta. Por otra parte, se logró ampliar los sistemas enzimáticos aplicados a la síntesis de aminas quirales, para ello se utilizó un nuevo tipo de transaminasa bifuncional denominada α,ω- DTAs, perteneciente a la metabolización de la putrescina. Este nuevo sistema, requiere solamente 3-5 equivalentes de aminodonor en el medio de reacción en contraste a los 50-500 equivalentes de grupo aminodonor utilizados normalmente.

Chiral amines are essential precursors in the production of biologically active compounds, including several important drugs. Among the biocatalytic strategies that have been developed for their synthesis, the use of ω-transaminases (ω-TA) appears as an attractive alternative allowing the stereoselective amination of prochiral ketones. However, the problems associated with narrow substrate specificity, unfavourable reaction equilibrium and expensive amine donors still hamper its industrial application. The search for novel enzymes from nature can contribute to expand the catalytic repertoire of ω-TA and help to circumvent some of these problems. A genome mining approach, based on the work described by Höhne et al., was applied for selection of potential R-ω-TA. Additional criteria were used to select an enzyme that differs from previously described ones. A candidate R-ω-TA from Capronia semiimmersa was selected, cloned and expressed in Escherichia coli. Interestingly, alignment of this enzyme with previously reported TA sequences revealed the presence of two additional amino acid residues in a loop close to the active site. The impact of this change was analysed with a structural model based on crystallized R-ω-TAs. Analysis of the substrate specificity of R-ω-TA from C. semiimmersa indicates that it accepts a diversity of ketones as substrates yielding the corresponding amine with good yields and excellent enantioselectivity. The expressed enzyme accepts isopropylamine as amine donor what makes it suitable for industrial processes.

Using the flexible ligand oxydiacetic acid (H2oda), ten new heteropolynuclear compounds with general formula [Ln2Zn3(oda)6]·xH2O (Ln = lanthanide ion, oda = oxydiacetate) were synthesized and structurally characterized. They were prepared by the direct reaction of oda in aqueous solution with stoichiometric amounts of Zn and Ln salts, followed by slow organic solvent diffusion. Two different structures were obtained: the larger lanthanides Pr, Nd, Eu and Tb gave compounds belonging to the hexagonal system, space group P6/mcc; whereas, the smaller ones, Ho, Er, Yb and Y formed cubic systems, space group Fd[3 with combining macron]c. In the case of Dy, the two polymorphs were obtained by slight variations in the synthetic conditions. The phase change observed is discussed in comparison with previously reported [Ln2M3(oda)6]·xH2O compounds (M = bivalent metal ion). Luminescence studies were also carried out. Compounds containing Eu and Tb showed the most intense luminescence, also bearing relatively long lifetimes for the excited state deactivation. The luminescence profiles of both Dy systems are independent of the hexagonal or cubic arrangements, due to the similar environment of the Ln having the same point symmetry.

The design of white-light emitting materials for solid-state lighting technology is governed by the combination of independent red, green and blue emitting centers. Many mixed-lanthanide single phase white-light source materials have been produced so far by a careful trial and error combination of lanthanide ions during the synthetic procedure. In this work we propose a simple and efficient method for the direct preparation of the desired material with the optimal combination of metal ions, avoiding the trial and error multiple synthetic approach. In the proposed strategy, the emission profiles of the isostructural complexes of each lanthanide ion for a chosen excitation wavelength are compared relative to their ion density and the necessary final composition of the material is calculated in order to obtain the white-light CIE coordinates. Using this method, we prepared a white-light emitting material which is a metal-organic framework built from oxydiacetate ligand, lanthanide ions (Eu, Tb and Dy emitting red, green and blue respectively) and zinc (a closed shell metal ion that has proven to be ideal for optical applications). The compound [Dy1.60Eu0.38Tb0.02Zn3(oda)6(H2O)6]·12H2O was prepared and fully characterized by single crystal x-ray diffraction, its optical properties were also measured. The proposed rational mixing of lanthanide ions could be useful for the preparation of white-light source materials in analogous systems, provided the single-lanthanide compounds are isostructural along the series and no optical interaction is operative among ions.

Severe infections with potentially fatal outcomes are caused by parasites from the genera Trypanosoma and Leishmania (class Kinetoplastea). The diseases affect people of remote areas in the tropics and subtropics with limited access to adequate health care. Besides insufficient diagnostics, treatment options are limited, with tenuous developments in recent years. Therefore, new antitrypanosomal antiinfectives are required to fight these maladies. In the presented approach, new compounds were developed and tested on the target trypa nothione synthetase (TryS). This enzyme is crucial to the kinetoplastids’ unique trypa nothione-based thiol redox metabolism and thus for pathogen survival. Preceding studies have shown that N5 -substituted paullones display antitrypanosomal activity as well as TryS inhibition. Herein, this compound class was further examined regarding the structure-activity relationships (SAR). Diverse benzazepinone derivatives were designed and tested in cell based assays on bloodstream Trypanosoma brucei brucei (T. b. brucei) and intracellular amastigotes of Leishmania infantum (L. infantum) as well as in enzyme-based assays on L. infantum TryS (LiTryS) and T. b. brucei TryS (TbTryS). While an exchange of just the sub stituent in the 9-position of paullones led to potent inhibitors on LiTryS and T. b. brucei para sites, new compounds lacking the indole moiety showed a total loss of activity in both assays. Conclusively, the indole as part of the paullone structure is pivotal for keeping the TryS inhibitory and antitrypanosomal activity of this substance class.

A summary of the resource.
This paper aim to evaluate the ultrafiltration (UF) process for constituents recovery from whey. Sequences of factorial designs were performed by varying temperature (5 to 40°C) and pressure (1 to 3 bar), to maximize the proteins concentration using membrane of 100kDa in dead end system. Based on the best result new experiments were performed with membrane of 50kDa and 10kDa. With the membrane of 50 the protein retention was about 3 times higher than the membrane of 100kDa. The concentrated obtained by UF membrane of 10kDa, 10°C and 2 bar in laboratory scale showed a mean protein retention of 80 %, greater protein solubility, emulsion stability and the identification of β-lactoglobulins (18.3 kDa) and α-lactalbumin fractions (14.2kDa). Therefore, the use of membrane of 100 and 50kDa are became a industrially recommendable alternatives to concentration of whey proteins, and/or as a previous step to the fractionation of whey constituents using membrane ≤10kDa, aiming at future applications in different areas (food, pharmaceutical, chemical, etc.).

The importance of professionalism in teaching practices for achieving high-quality education has been acknowledged. Becoming a professional teacher is a long-term commitment but it is essential to promote high standards of professionalism from the beginning of teacher candidate formation. In addition, as professionalism in teaching is closely linked to the concept of “teacher as a researcher”, formative research seems to be a potent strategy for engaging future teachers in developing research abilities, essential for understanding and analyzing their educational practices. In this sense, the objective of this work is to explore the influence of a formative research approach on the development of research competencies by applying an action-research approach with a longitudinal cut. Thus, an intervention based on a ubiquitous strategy of formative research was implemented during the final three years of education of a teacher’s training institution in Uruguay. The teacher candidate development of research competencies was assessed along the process. The obtained results demonstrated the effectiveness of implementing formative research to enhance the research skills of teacher candidates. The assessment of overall research competencies exhibited a statistically significant and substantial positive change (Cohen’s d = 0.92). The impact was most pronounced among teacher candidates with the least developed initial research competencies, particularly for those in the early stages of their careers. This suggests that candidates engaged in the formative research strategy throughout their studies might achieve even higher levels of research competencies. Disaggregating the results by gender and for most of the specific subjects showed similar effects.In addition, within the framework of ubiquitous learning, the emphasis on the use of digital tools during the intervention led to an increase in the development of the research skills related to these tools for the studied sample. This is the first example of a quantitative approach that leads to concrete conclusions related to the effect of a research formative strategy for teacher candidates in Uruguay.

Abstract
A summary of the resource.
Zinc borates are very important industrial materials with applications that include fire retardants and preservatives in wood. In this work, we report the preparation of three novel zinc borates: [Zn(NH3)3B4O5(OH)4]·H2O (ZB1), Zn3(H2B3O7)2·2NH3·4H2O (ZB2), and [Zn(NH3)4][B4O5(OH)4]·4H2O (ZB3). The solid phases were characterized by elemental analysis, X-ray diffraction, infrared and Raman spectroscopy, scanning electron microscopy and thermal analysis. The crystal structures of ZB1 (monoclinic, Cc, a = 12.1972(8), b = 7.8314(5), c = 12.1441(8) Å and β = 107.404(5)°) and ZB3 (orthorhombic, Pbca, a = 15.0796(9), b = 11.8853(5) and c = 16.7606(8) Å) were determined. They are novel neutral Zn-polyborate complexes [Zn(NH3)3B4O5(OH)4] hydrate and [Zn(NH3)4][B4O5(OH)4] tetrahydrate salt, respectively. The complete assignment of infrared and Raman spectra was performed theoretically using DFT calculations. For ZB2 (obtained as a polycrystalline phase), powder X-ray diffraction confirmed a single phase and allowed the determination of the unit cell parameters and lattice type (rhombohedral, a = 36.78076 (6), c = 12.20052 (3) Å) with an expected formula Zn3(H2B3O7)2·2NH3·4H2O suggesting that this compound is a complex triborate. Furthermore, a low-cost scalable synthetic procedure for ZB1 starting from zinc oxide and boric acid in ammonia solution with a high yield is also reported.

A summary of the resource.
The local crystal structure of Y3NbO7 was probed using neutron total scattering. Five different structural models were fit to the experimental pair distribution function of this material up to 15 Å. Fits of defect fluorite (Fm3m), pyrochlore (Fd3m), and orthorhombic C weberite models (C2221) were attempted based on prior structural investigations of Y3NbO7 and related fluorite-based materials. Orthorhombic P weberite (P2221) and monoclinic weberite models (P1121) were derived via symmetry reduction of the orthorhombic C weberite structure. All models except monoclinic weberite failed to reproduce the experimental pair distribution function. Misfits were clearly visible for peaks arising from atom pairs within the first coordination shell (defect fluorite and pyrochlore) and beyond (orthorhombic C and P weberite). By contrast, monoclinic weberite (γ ≈ 91.8◦) provided an excellent fit, with no major misfits observed. This unit cell features distorted YO8 dodecahedra, YO7 monocapped octahedra, and NbO6 octahedra as building blocks. Comparison between the topologies of the cationic and anionic substructures in monoclinic weberite and its parent orthorhombic C structure revealed displacements of the metal atoms along the [100] direction and a less compact spatial distribution of oxygen atoms along the a axis. The monoclinic weberite model presented herein may serve as a starting point to revisit structural modeling of fluorite-based materials with challenging local structures

En este trabajo de tesis se han explorado dos metodologías en la búsqueda de la obtención de nanopartículas de boratos de zinc. Estas metodologías fueron planteadas para lograr sintetizar fases de boratos de zinc en distintos rangos de temperaturas, lo que brinda grandes posibilidades en el desarrollo de aplicaciones tecnológicas. Empleando la síntesis por combustión de gel asistida se pretendió sintetizar boratos de zinc a temperatura mayores a 250 °C, para los cuales se esperaba obtener nanopartículas que fueran importantes en el desarrollo de nuevos aditivos de materiales que se preparan a altas temperaturas, por ejemplo, plásticos. A partir de las síntesis asistidas por micelas reversas, se pretendió obtener boratos de zinc a baja temperatura, por ejemplo, los aminoboratos de zinc [Zn(NH3)3B4O5(OH)4]·H2O (ZB1) y Zn3(H2B3O7)2·2NH3·4H2O (ZB2), reportados previamente durante mi trabajo de Licenciatura en Química. Mediante la metodología de combustión de gel asistida se logró sintetizar la fase ZnB4O7. Los resultados presentados para estas síntesis son contundentes respecto a los procesos químicos que tienen lugar en el sistema estudiado. El medio reductor ofrecido por la alta concentración de amonio da lugar a la formación de carbono amorfo que permanece como principal impureza en las muestras obtenidas, no siendo posible la obtención de nanopartículas de boratos de zinc. La obtención de ZB2 por métodos de baja temperatura fue posible a partir de un sistema coloidal mantenido por agitación formado por agua, CTAB, hexano y diferentes alcoholes como cosurfactantes. Para estos sistemas se obtuvo la fase cristalina ZB2 con tamaños entre 100 y 1000 nm, reportándose tamaños menores a 150 nm cuando se empleó isopropanol como cosurfactante. También se sintetizó la fase ZB2 en un medio de micelas reversas. Este último es uno de los logros más importantes de este trabajo, ya que se consiguió estabilizar un sistema de micelas reversas y obtener un producto con un tamaño de partícula menor a 150 nm. Este trabajo es un aporte importante en la química básica de estos sistemas, abriendo puertas a futuros estudios de síntesis en sistemas de micelas reversas. La metodología utilizada para determinar las condiciones de formación de micelas reversas es extrapolable a cualquier sistema de obtención de compuestos químicos a partir de una solución acuosa con parámetros de concentración y fuera iónica adecuados. Los estudios microbiológicos realizados sobre la fase ZB2 permitieron confirmar la actividad antifúngica y antibacteriana presentada por este compuesto y su dependencia con el tamaño de partícula.

Los desafios que envuelve la sintesis de productos naturales son numerosos. No solo implican aumentar el conocimiento acerca de los mismos, sino que ademas, tienen como motivaci6n para el quimico sintetico el descubrir nuevas reacciones asi como tambien mejorar otras ya conocidas. EI mejor conocimiento de los detalles estructurales que gobiernan los procesos biol6gicos ha permitido el diseno y sintesis de analogos que emulan la actividad exhibida por los compuestos naturales, con la posibilidad incluso de aumentar el interes biomedico de los mismos. En general, la busqueda de modelos sinteticos sencillos utilizando metodologias de simplificaci6n molecular, con una actividad biol6gica de~eada, es uno de los grandes desaflos de la Quimica Farmaceutica moderna. Este trabajo de Tesis se enmarca en un proyecto general tendiente a la construcci6n de metabolitos marinos activos, 0 bien, simplificaciones moleculares de los mismos. Esto implica trabajar sobre aspectos sinteticos, estructurales y de actividad biol6gica de los mismos. Los Phorboxazoles son macr61idos de origen marino que presentan una marcada actividad biol6gica. Su esqueleto carbonado sin precedentes ha hecho que muchos grupos de investigaci6n se hayan interesado en los mismos, tanto en 10 que respecta al logro de su sintesis total, asi como tambien, al desarrollo de estudios tendientes a obtener modelos simplificados de los mismos. Se presentan aqui, un conjunto de metodologias sinteticas para la preparaci6n de modelos sencillos, analogos a dominios parciales de Phorboxazoles (Fig. 1), asi como tambien sus correspondientes estudios estructurales y conformacionales. Las estrategias sinteticas estan basad as en a) construcci6n del resto oxaz61ico usando metodologias de ciclodeshidrataci6n-oxidaci6n de ~-hidroxiamidas y b) estudios sinteticos para la construcci6n de 2,4,6 tetrahidropiranos trisustituidos por tres vias: i) expansi6n electrofflica de oxiranos, ii) ciclaci6n tipo Prins y Iii) Hetero Oiels-Alder . Se presentan ademas, resultados de actividad antihelmintica, citotoxicidad y propiedades acomplejantes de algunos compuestos seleccionados.

Con el fin de analizar la presencia de metalo-β-lactamasas en nuestro medio, se incluyeron en este estudio aislamientos de Pseudomonas aeruginosa causantes de infecciones nosocomiales en un centro hospitalario del Uruguay, en el período comprendido entre abril y setiembre de 2008. En un aislamiento se detectó la presencia del gen codificante de la metalo-β-lactamasa VIM-2 asociado a un integrón de clase 1 y del gen codificante de una β-lactamasa de espectro extendido CTX-M-2. Esta es la primera comunicación de la presencia de los genes blaCTX-M-2 y blaVIM-2 en un mismo aislamiento de P. aeruginosa. A pesar de que las carbapenemasas ya han sido ampliamente documentadas en varias partes del mundo, esta es la primera comunicación de una metalo-β-lactamasa adquirida con actividad carbapenemasa en bacterias patógenas encontradas en el Uruguay.

<p>El objetivo principal de la presente tesis fue la generación de nuevas moléculas bioactivas mediante reacciones multicomponente (RMC), reacciones de Passerini y de Ugi, siguiendo principios de la química verde y el estudio preliminar de su actividad anticancerígena.<br />En este siglo, uno de los mayores desafíos de la química orgánica y la química médica, es la obtención de nuevos compuestos orgánicos estructuralmente diversos, de manera rápida y eficiente, para estudiar sistemas biológicos y acelerar el proceso de descubrimiento de fármacos. La química también enfrenta retos como la reducción del impacto ambiental de los residuos generados y el manejo adecuado de recursos ambientales. En este sentido, en el presente trabajo de tesis se estudiaron y optimizaron rutas sintéticas basadas en RMC con alta eficiencia atómica, utilizando condiciones de reacción alternativas amigables con el medio ambiente (empleo de disolventes verdes o reacciones en ausencia de disolvente) y en muchos casos minimizando el consumo energético y el tiempo de reacción mediante el empleo de irradiación de microondas o ultrasonido.<br />En la actualidad una estrategia terapéutica muy prometedora contra el cáncer es el diseño y desarrollo de potenciales fármacos híbridos. Es así que se diseñaron y sintetizaron tres series de compuestos mediante hibridación de tres farmacóforos con potencial actividad antitumoral: grupos liberadores de óxido nítrico (GLNO), funcionalidad nitroalquenilo y análogos de tocoferol. Se sintetizaron 76 compuestos derivados de RMC de Passerini o de Ugi, además de compuestos que se utilizaron como componentes de partida, siempre que fuera posible, en condiciones verdes. Se obtuvieron en general con rendimientos de moderados a buenos y en algunos casos, excelentes.<br />La quimioteca así generada fue sometida a una evaluación biológica primaria in vitro con el objetivo de estudiar su actividad antiproliferativa frente a un panel de 6 líneas celulares tumorales humanas. Como aproximación al estudio del mecanismo de acción de estas familias de compuestos se llevaron a cabo experimentos de: i) apoptosis; ii) análisis del ciclo celular; iii) capacidad de liberar óxido nítrico; iv) efecto sobre la activación del inflamasoma NLRP3. Para ello se seleccionaron algunos de los compuestos más prometedores en cuanto a su relación actividad antiproliferativa /citotoxicidad. De los 76 compuestos sintetizados entre las tres series, 8 resultaron con una actividad biológica relevante. Entre ellos destaca el rol de los furoxanos (GLNO) como farmacóforo antitumoral, en los compuestos 12l y el 13e, con valores de IC50 entre 0.11 a 3.5 μM y 0.021 a 3.0 μM respectivamente en las diferentes líneas celulares tumorales. Los 8 compuestos destacados, presentaron además selectividad hacia células tumorales sobre células normales, demostrando ser prometedores candidatos para el desarrollo de potentes y selectivos agentes terapéuticos contra el cáncer.</p>

En este siglo, los químicos se han encontrado con un nuevo desafío: producir de manera eficiente los materiales necesarios para mantener la calidad de vida de los seres vivos, reduciendo los efectos negativos sobre el medio ambiente y la salud humana. En este sentido, es fundamental el rol del docente en la formación de futuros científicos que puedan generar de manera segura y eficiente productos y procesos no tóxicos, hacia el camino de un futuro sustentable. En este trabajo se describe el uso de una reacción multicomponente, la reacción de Passerini, como un experimento de laboratorio en un curso de grado de Química Orgánica. Es un proceso rápido, eficiente y simple que permite obtener productos complejos a partir de moléculas sencillas y en un paso de reacción. Diferentes parámetros fueron analizados para determinar cuán amigable es esta reacción con el medio ambiente, en las condiciones ensayadas. El estudiante tendrá la posibilidad de discutir sobre química verde, desarrollo de moléculas bioactivas, síntesis orientada a la diversidad, reacciones multicomponente y química en disolución acuosa. A summary of the resource.
A one-pot efficient, practical and eco-friendly synthesis of tocopherol analogues has been developed using water or solvent free conditions via Passerini and Ugi multicomponent reactions. These reactions can be optimized using microwave irradiation or ultrasound as the energy source. Accordingly, a small library of 30 compounds was prepared for biological tests. The evaluation of the antiproliferative activity in the human solid tumor cell lines A549 (lung), HBL-100 (breast), HeLa (cervix), SW1573 (lung), T-47D (breast), and WiDr (colon) provided lead compounds with GI50 values between 1 and 5 μM. A structure–activity relationship is also discussed. One of the studied compounds comes up as a future candidate for the development of potent tocopherol-mimetic therapeutic agents for cancer.

The aim of this work was to assess the influence in the diagnostic value for human hydatid disease of the composition of bovine hydatid cyst fluid (BHCF) obtained from fertile (FC) and non-fertile cysts (NFC). Eight batches from FC and 5 from NFC were prepared and analysed with respect to chemical composition: total protein, host-derived protein, carbohydrate and lipid contents. No differences were observed in the first two parameters but carbohydrate and lipid contents were shown to be higher in batches from FC than in those from NFC. Bands of 38 and 116 kD in SDS-PAGE profiles were observed to be present in BHCF from FC only. Two pools were prepared from BHCF batches obtained from FC (PFC) and NFC (PNFC), respectively. Antigen recognition patterns were analysed by immunoblot. Physicochemical conditions for adsorption of antigens to the polystyrene surface (ELISA plates) were optimized. The diagnostic value of both types of BHCF as well as the diagnostic relevance of oxidation of their carbohydrate moieties with periodate were assessed by ELISA using 42 serum samples from hydatid patients, 41 from patients with other disorders, and 15 from healthy donors. Reactivity of all sera against native antigen were tested with and without free phosphorylcholine. The best diagnostic efficiency was observed using BHCF from periodate-treated PFC using glycine buffer with strong ionic strength to coat ELISA plates.

Carquejyl acetate (CA), the main component of the essential oil from Baccharis trimera was isolated and characterized by using Fourier Transform infrared (FT-IR) and Raman (FT-Raman), Ultraviolet–Visible (UV–Visible), Electronic Circular Dichroism (ECD), Mass, Hydrogen and Carbon Nuclear Magnetic Resonance (1H- and 13C NMR) and 2D 1H-1H gCOSY, 1H-13CgHSQC, 1H-13CgHMBC spectroscopies. Two conformers were found in the potential energy surface (PES) for CA in gas and aqueous solution phases by using the hybrid B3LYP method with the 6-31G* and 6-311++G** basis sets and, only one of them, C2, was the most stable and with higher populations in both phases. Natural bond orbital (NBO) studies evidence the high stability of CA as compared with carquejol while atoms in molecules (AIM) analyses reveals the different characteristics of the H bonds formed showing CA higher topological properties. The gap values suggest that carquejol is most reactive than CA while higher values in the global electrophilicity and nucleophilicity indexes are observed for C2, as compared with carquejol. Very good correlations between the experimental FTIR, FT-Raman, UV–Visible and 1H and 13C NMR spectra with their theoretical counterparts were found. The ECD spectrum predicted for C2 reveals the absolute configuration of CA. The complete assignment of the 84 normal vibration modes of CA is presented.

Muchos de los procesos en los cuales seria conveniente el uso de enzimas requieren condiciones de trabajo que general mente son adversas para la estabilidad de las mismas. Ademas el alto costa y la dificultad para regenerarlas condicionan sus posibilidades de aplicaci6n. Los estudios realizados en esta tesis se orientaron hacia la busqueda y caracterizaci6n de estrategias de estabilizaci6n de enzimas mediante procesos de ingenieria enzimMica en fase s6lida. En particular se estudi6 la influencia de los siguientes factores: i) inmovilizaci6n de la enzima; ii) modificaci6n post¬inmovilizaci6n de la superficie del soporte; iii) recubrimiento de la enzima inmovilizada con polimeros hidrofilicos. Se determinaron los parametros de inmovilizaci6n de todos los derivados obtenidos, y se hicieron estudios de estabilidad termica y estabilidad en presencia de sistemas de co¬solventes organicos, con cada uno de ellos. Como enzimas modelo se utilizaron tres f3-galactosidasas de diferentes origenes (E. coli, K lactis y A. oryzae). Se estudiaron dos quimicas de inmovilizaci6n distintas: en glutaraldehido (Glut)-agarosa, elegida como metodo de referencia por ser convencional y ampliamente estudiado; y en tiolsulfinato (TSI)¬agarosa, que es un metoda relativamente nuevo, desarrollado y estudiado en nuestro laboratorio, que utiliza condiciones de inmovilizaci6n suaves. En general, los derivados obtenidos con la quimica del TSI tuvieron rendimientos de inmovilizaci6n mayores, y sus KM fueron menos afectadas que las de los derivados obtenidos con la quimica del glutaraldehido. Sin embargo, estos ultimos exhibieron mejores propiedades de estabilidad termica y frente a solventes, 10 que los hace mas adecuados para biocataIisis bajo condiciones agresivas. Estos resultados demostraron la relevancia de la quimica de inmovilizaci6n en el disefio del biocatalizador. Para modificar las caracteristicas de la superficie del soporte TSI-agarosa, luego de la inmovilizaci6n de la enzima se hizo reaccionar el exceso de grupos TSI con cuatro tiol¬compuestos. De esta manera se consigui6 construir en el entomo de la enzima, cinco nano¬ambientes con caracteristicas diferentes. El comportamiento diferencial de los derivados obtenidos en los estudios de estabilidad termica y frente a solventes organicos, sugiere que cada nano¬ambiente contribuye de forma distinta a la estabilidad de la enzima, dependiendo de las condiciones de desnaturalizaci6n. Es decir que resulta posible modelar las caracteristicas de la superficie del soporte, con el fin de maximizar la estabilidad de la enzima para usarla en una aplicaci6n en particular. La inmovilizaci6n en Glut-agarosa demostr6 ser efectiva para alcanzar estabilizaci6n termica, aun a altas temperaturas; sin embargo no siempre result6 efectiva frente al aumento de la concentraci6n de los co-solventes. La hidrofilizaci6n de los derivados inmovilizados en Glut-agarosa con polimeros de polialdehido-dextrano y poliamina-dextrano, permiti6 una importante estabilizaci6n frente a concentraciones altas de co-solventes; no obstante, la estabilizaci6n alcanzada frente al aumento de la temperatura fue menor. Uno de los resultados mas destacables fue el aumento de la vida media del derivado hiper-hidrofilizado de la enzima de A. oryzae, en presencia de 50% v/v de acetona, en un factor de 25 respecto al derivado sin modificar. Los resultados obtenidos muestran el potencial de esta estrategia de hidrofilizaci6n como una herramienta uti! para alcanzar estabilizaci6n adicional de las enzimas inmovilizadas, frente a condiciones drasticas de concentraci6n de co¬solventes. Los estudios realizados en esta tesis permiten afirmar que es posible el disefio orientado de biocatalizadores en funci6n de sus aplicaciones, mediante diversos procesos de ingenieria de la enzima en fase solida.

Human milk (HM) constitutes the first immunological barrier and the main source of nutrients and bioactive components for newborns. Immune factors comprise up to 10% of the protein content in HM, where antibodies are the major components (mainly IgA, IgG, and IgM). In addition, antibacterial enzymes such as lysozyme and immunoregulatory factors such as soluble cluster of differentiation 14 (sCD14) and transforming growth factor β2 (TGF-β2) are also present and play important roles in the protection of the infant's health. Donor milk processed in HM banks by Holder pasteurization (HoP; 62.5°C, 30 min) is a safe and valuable resource for preterm newborns that are hospitalized, but is reduced in major immunological components due to thermal inactivation. We hypothesized that high hydrostatic pressure (HHP) and high-pressure homogenization (HPH) are 2 processes that can be used on HM to reduce total bacteria counts while retaining immunological components. We studied the effects of HHP (400, 450, and 500 MPa for 5 min applied at 20°C) and HPH (200, 250, and 300 MPa, milk inlet temperature of 20°C) applied to mature HM, on microbiological and immunological markers (IgA, IgG, IgM, sCD14, and TGF-β2), and compared them with those of traditional HoP in HM samples from healthy donors. The HHP processing between 400 and 500 MPa at 20°C reduced counts of coliform and total aerobic bacteria to undetectable levels (<1.0 log cfu/mL) while achieving approximately 100% of immunological component retention. In particular, comparing median percentages of retention of immunological components for 450 MPa versus HoP, we found 101.5 versus 50.5% for IgA, 89.5 versus 26.0% for IgM, 104.5 versus 75.5% for IgG, 125.0 versus 72.5% for lysozyme, 50.6 versus 0.1% for sCD14, and 88.5 versus 61.1% for TGF-β2, respectively. Regarding HPH processing, at a pressure of 250 MPa and inlet temperature of 20°C, the process showed good potential to reduce coliforms to undetectable levels and total aerobic bacteria to levels slightly above those obtained by HoP. The median percentages of retention of immunological markers for HPH versus HoP were 71.5 versus 52.0%, 71.0 versus 27.0%, 104.0 versus 66.5%, and 30.9 versus 0.2%, for IgA, IgM, IgG, and sCD14, respectively; results did not significantly differ for lysozyme and TGF-β2. The HPH at 300 MPa produced higher inactivation of immunological components, similar to values achieved with HoP.

A summary of the resource.
We studied the comparative antioxidant and anti-glycation activities of Mentha x piperita L., Baccharis trimera Iless. and Peumus boldus Mol, in order to evaluate their potential interest as ingredients in functional foods. The total content of polyphenol compounds was determined by Folin-Ciocalteau assay as their antioxidant and anti-glycation capacities, using ABTS and ORAC for the first one and a model with methylglyoxal and bovine serum albumin for the latter. Then, paraoxonase 1 (PON 1) arylesterase activity was measured as well as apolipoprotein A-1 (ApoA-1) structure by SDS-PAGE, in the presence of an oxidative agent and the herbal extracts. Finally, the same procedure was applied to high density lipoprotein (HDL) particles using a Lipoprint kit. Results show that herbal extracts have a considerable amount of total polyphenols, thus a high antioxidant activity and a considerable anti-glycation activity. Furthermore, these extracts restore PON 1 activity as well as the original configuration of apoA-1 and the distribution of HDL subclasses, favouring anti-atherogenic particles. These herbal extracts are interesting targets to use as ingredients in functional foods.

Las enfermedades crónicas no trasmisibles (ECNT) son enfermedades no infecciosas ni contagiosas. Constituyen la principal causa de muerte tanto en Uruguay como en el mundo. Éstas incluyen: el cáncer, las enfermedades cardiovasculares (ECVs), la diabetes y las enfermedades respiratorias crónicas. Este trabajo se enfocó principalmente al estudio de la aterosclerosis, principal factor de riesgo para el desarrollo de ECVs. La aterosclerosis es una reacción inflamatoria crónica de la pared arterial, en cuyo origen y avance participan desde componentes del sistema inmune, hasta la dieta ingerida por el individuo. Los productos de la glicación avanzada (advanced glycation end products, AGEs), son un grupo de compuestos grande y heterogéneo, que se originan en la reacción de Maillard. Estos compuestos juegan un rol crucial en el desarrollo de ECNT mediante los siguientes mecanismos: a) el entrecruzamiento de proteínas alterando sus funciones y propiedades; b) activación de cascadas de señalización pro-inflamatorias mediante interacción con receptores específicos, o mediante mecanismos no dependientes de receptores; c) generación de compuestos oxidantes y pro-inflamatorios. Los compuestos fitoquímicos son compuestos bioactivos presentes en alimentos de origen vegetal. Entre éstos se encuentran los compuestos fenólicos, los cuales presentan una gran actividad antioxidante y antiglicante inhibiendo la formación de AGEs, y por ende disminuyendo el estrés oxidativo y las respuestas pro-inflamatorias. Incluso, se ha visto que algunos de estos compuestos presentan actividad antiaterómica, manteniendo la actividad antioxidante de partículas de lipoproteína de alta densidad (HDL), al evitar la oxidación de la enzima paraoxonasa 1 (PON-1) que contienen en su superficie. Estos compuestos con propiedades bioactivas, pueden ser utilizados en la formulación de alimentos funcionales, de forma de prevenir el desarrollo de ECNT. Por lo tanto, el principal objetivo de este trabajo fue el de obtener extractos inhibidores de la formación de los mismos a partir de hierbas medicinales de uso frecuente en Uruguay (Mentha x piperita L., Baccharis trimera Iless. y Peumus boldus Mol.), así como evaluar las diferentes vías de formación de compuestos AGEs provocados por la dieta. Conjuntamente, se estudió su comportamiento a lo largo del tracto gastrointestinal, de forma de determinar si la bioactividad se mantiene al alcanzar el intestino. Para efectuar este estudio, se realizaron extractos de las hierbas antes mencionadas utilizando dos solventes: agua destilada a 95°C (3 minutos) y etanol 95% a temperatura ambiente (24 horas). A su vez, se analizó la eficacia de un método de extracción alternativo (ultrasonido) a diferentes tiempos (1, 2 y 3 horas). Los resultados obtenidos demostraron en primer lugar, que la aplicación de ultrasonido no presenta diferencias significativas a la hora de extraer compuestos bioactivos, por lo que se seleccionaron los extractos en agua destilada a 95°C durante 3 minutos y en etanol 95% durante 24 horas. Todos los extractos seleccionados (figura 0), presentaron considerable XII contenido de compuestos fenólicos totales, así como una gran actividad antioxidante (frente a radicales ABTS•+ o peroxilo), antiglicante y anti-inflamatoria, incluso luego de haber sido sometidos al proceso de simulación gastrointestinal. Se sugiere que los extractos evitan la oxidación de la PON-1 en presencia de AAPH y SIN-1, manteniendo así su actividad arilesterasa. Asimismo, parecen afectar la distribución de subclases de partículas de HDL. En conclusión, surge de nuestros resultados que los extractos acuosos y etanólicos de las tres hierbas estudiadas, son muy buenos candidatos como fuente de compuestos fenólicos con capacidades bioactivas muy interesantes, pudiendo ser utilizados como ingredientes en alimentos funcionales.

Glutathione transferase enzymes (GSTs) constitute a major detoxification system in helminth parasites and have been related to the modulation of host immune response mechanisms. At least three different GSTs classes have been described in Platyhelminthes: Mu, Sigma and Omega. Mining the genome of Echinococcus multilocularis and the ESTs databases of Taenia solium and E. granulosus identified two new GSTs from the cestode E. granulosus, named EgGST2 and EgGST3. It also revealed that the Omega class of GSTs was absent from the Taenidae family. EgGST2 and EgGST3 are actively expressed in the parasite. In order to know the origin of these new GSTs, in silico analyses were performed. While EgGST2 is classified as belonging to the Sigma class, the data obtained for EgGST3 allowed a less clear interpretation. The study of the evolutionary relatedness based on the C-terminal domain sequence, gene structure conservation and three-dimensional structure predictions, suggests that EgGST3 is derived from the Platyhelminthes' Sigma-class cluster. Interestingly, the N-terminal domain displays some characteristic Omega-class residues, including a Cys residue that is likely to be involved in the catalytic mechanism. We discuss different evolutionary scenarios that could explain the observed patterns.
