By means of first-principles calculations, we investigate the reduction chemistry of monolayer boron nitride (2D BN) and graphene. The adsorption of alkali atoms dramatically increases the reactivity of 2D BN, Li being the most powerful agent and closely followed by K and Na. Interestingly, the reactivity enhancement in 2D BN is 3.6 times larger in comparison with graphene. On average, Li adsorption enhances the adsorption energies by 66.0 and 18.2 kcal/mol for 2D BN and graphene, respectively. The higher increment in adsorption energies observed for 2D BN can be attributed to the significant difference in alkali charge donation to pristine and functionalized 2D BN. In contrast, for graphene, this difference is much smaller. The increase of reactivity strongly depends on the functional group added as well as on the alkali used. The reduction chemistry facilitates the addition of functional groups to 2D BN which cannot be added without the presence of alkalis. We conclude that reduced 2D BN is more reactive than reduced graphene when alkalis act as reducing agents. Los ácidos grasos trans (AGT) se encuentran en aceites parcialmente hidrogenados. Una alta ingesta de ellos provoca consecuencias adversas sobre la salud, en particular, aumentando el riesgo de enfermedades cardiovasculares. La Organización Panamericana de la Salud (OPS), integrante de la Organización Mundial de la Salud (OMS), recomendó un contenido máximo de 2% de ácidos grasos trans en aceites vegetales y margarinas untables, y un máximo de 5% para las grasas y los aceites que se utilizan como ingredientes en alimentos. Por su parte, el Mercosur estableció como obligatoria la inclusión del contenido total de ácidos grasos trans en el etiquetado nutricional de los alimentos envasados. En consecuencia, resulta imprescindible analizar los alimentos que contienen AGT y sus propiedades térmicas para su posible sustitución. A diferencia de los AGT de origen industrial, se debe valorizar a los ácidos grasos trans "naturales" como el trans-vaccénico (TVA – Trans Vaccenic Acid) y el ácido linoleico conjugado (CLA – Conjugated Linoleic Acid). Ambos son reconocidos por su efecto beneficioso en la salud humana y aparecen en las grasas lácteas y en las carnes de rumiantes que no han sufrido modificación industrial. En el presente trabajo se analizó la composición en ácidos grasos y el comportamiento térmico del material graso extraído de productos de panaderías (panes con grasa y galletas malteadas). El máximo valor de AGT totales encontrado en uno de los materiales grasos para la elaboración de productos de panadería fue 17,3%. Sin embargo, la mayoría de ellos presentó valores cercanos al 7%, y el AGT principal encontrado fue el TVA con un promedio de 4,3%. Extracts from hydatid cyst fluid (HCF-ext), protoscoleces (PSC-ext) and hydatid cyst membrane (HCM-ext) activated human C producing C3 conversion and generating the C5b6 complex and the terminal C complex (TCC). HCM-ext showed much lower C activating activity than PSC-ext and HCF-ext. Moreover, its ability to generate C5b6 and TCC was lower than its ability to convert C3. On the other hand, PSC-ext and HCF-ext proved to be good C activators when their specific C activating activities were compared with that of inulin. However, PSC-ext produced lower levels of TCC than those produced by HCF-ext, in spite of the fact that both produced practically the same levels of C3d and C5b6. These results may be consistent with the existence of several mechanisms of C modulation involved in the defence of the parasite against host C damage. El control de la inflamación por el parasito debe ser un proceso multifactorial, que regule las diferentes vias por las que la inflamación se inicia y mantiene. Entre estas se encuentra el sistema complemento (C), un conjunto de proteinas plasmaticas y de superficie celular, que se activa sobre superficies no propias para desencadenar una serie de efectos biol6gicos, entre ellos, la inducci6n de inflamaci6n. Los antecedentes sobre el C y E. granulosus indican que, durante el desarrollo de la forma infectante a quiste, la superficie expuesta al hospedero se transforma en no activadora del C. La pared del quiste (PQH) constituye la interfase con el hospedero, y su inercia frente al C radica, en gran medida, en las propiedades de su capa mas extema, la capa laminar (CL). Esta es una estructura acelular, rica en glucidos, que se visualiza a nivel ultraestructural como una mall a de fibras sobre las que se disponen granulos de tamaiio definido, densos a los electrones. Los granulos se forman en las celulas de la PQH y son exocitados hacia la CL. La búsqueda de mecanismos de control de la activación del C por la PQH habia conducido a la detecci6n de una actividad que interferia con una de las vias de activación del sistema, la via altema (VA). El objetivo central de este trabajo fue la caracterización de esta actividad, a traves de la identificación del (de los) componente(s) responsable(s) y de la elucidación del mecanismo de inhibici6n. El componente activo se purificó a partir de un extracto de PQH mediante metodos cromatograficos y, utilizando diferentes tecnicas analiticas, se identificó como myo-inositol hexakisfosfato (IP6). Esta molecula, ubicua en los eucariotas y de función desconocida, habia sido descrita como exclusivamente intracelular. Sin embargo, se demostr6 que E. granulosus secreta al medio extracelular grandes cantidades de IP6 insolubilizado con calcio, dando lugar asi al elemento granular de la CL. La acumulaci6n de IP6 calcico ocurre Unicamente en la interfase con el hospedero y es una caracteristica no compartida con otra especie de su genero, E. multilocularis. Los resultados sugieren que el IP6 en la CL no esta relacionado al control de la activación del C por la PQH. Pese a esto, en forma insoluble y en presencia de Ni++, el compuesto bloquea diferentes eventos de la activación de la VA, a traves de la interacción con uno de sus componentes, el factor B. Los resultados del trabajo aportan información valiosa a la biologia, tanto del parasito, como de la propia molécula. No se ha establecido el papel del IP6 en la CL; los experimentos futuros deberan esc1arecer si el IP6 cumple una función meramente estructural, 0 esta involucrado en la relación con el hospedero. The ubiquitous intracellular molecule myo-inositol hexakisphosphate (IP6) is present extracellularly in the hydatid cyst wall (HCW) of the parasitic cestode Echinococcus granulosus. This study shows that extracellular IP6 is present as its solid calcium salt, in the form of deposits that are observed, at the ultrastructural level, as naturally electron dense granules some tens of nanometers in diameter. The presence of a calcium salt of IP6 in these structures was determined by two different electron microscopy techniques: (i) the analysis of the spatial distribution of phosphorus and calcium in the outer, acellular layer of the HCW (the laminated layer, LL) through electron energy loss spectroscopy, and (ii) the observation, by transmission electron microscopy, of HCW that were selectively depleted of IP6 by treatment with EGTA or phytase, an enzyme that catalyses the dephosphorylation of IP6. Diabetes pathogenesis encompasses oxidative stress, inflammation, insulin malfunctioning and partial or total insulin secretion impairment, which leads to a constant hyperglycemia. Polyphenols are known to possess bioactive properties, being Tannat grape skin a natural and sustainable source of these compounds. The present study aimed to find out the bioaccessibility of health-promoting molecules composing a multifunctional extract from Tannat grape skin obtained under hydro-alcoholic-acid conditions. The identification of phenolic compounds in the samples was performed by ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS). Subsequently, the samples were in vitro digested mimicking the human oral gastrointestinal conditions and the bioactivity of the digest (antioxidant, anti-inflammatory and modulation of glucose metabolism) was assessed. E�ect on glucose metabolism was estimated by measuring carbohydrases activity and the functionality of glucose transporters of small intestine cells in presence and absence of the digested extract. Flavonoids, phenolic acids and phenolic alcohols were the major phenol compounds detected in the extract. The bioaccessible compounds protected the intestinal cells and macrophages against the induced formation of reactive oxygen species (ROS) and nitric oxide (NO). In addition, glucose transporters were inhibited by the digested extract. In conclusion, the bioaccessible compounds of the extract, including phenols, modulated key biochemical events involved in the pathogenesis of diabetes such as oxidative stress, inflammation and glucose absorption. Rice is an important source of methane (CH4) and other crops may be sources of nitrous oxide (N2O), both of which are powerful greenhouse gases. In Uruguay, irrigated rice rotates with perennial pastures and allows high productivity and low environmental impact. A long-term experiment with contrasting rice rotation intensification alternatives, including rice–soybean and continuous rice, was recently carried out in an Argialboll located in a temperate region of South America. To know if rotation systems influence soil microbial activity involved in CH4 and N2O emissions, the abundance and potential rate for gas production or consumption of microbial populations were measured during the rice crop season. CH4 was only emitted when rice was flooded and N2O emission was not detected. All rotational soils showed the highest rate for methanogenesis at tillering (30 days after rice emergence), while for methanotrophy, the maximum rate was reached at flowering. The abundance of related genes also followed a seasonal pattern with highest densities of mcrA genes being observed at rice flowering whereas pmoA genes were more abundant in dry soils after rice harvest, regardless of the rotation system. Differences were foundmainly at tillering when soils with two consecutive summers under rice showed higher amounts ofmcrA and pmoA gene copies. The potential denitrification rate was highest at the tillering stage, but the abundance of nirK and nirS genes was highest in winter. Regarding ammoniumoxidation, bacterial amoA abundance was higher in winter while the archaeal amoA gene was similar throughout the year. A strong influence of the rice growth stage was registered for most of the parameters measured in rice paddy soils in this no-till rice intensification experiment. However, differences among rotations begin to be observed mainly at tillering when the abundance of populations of the methane and nitrous oxide cycles seemed to respond to the rice intensification. The performance of a group of density functional methods of progressive complexity for the description of the ClO bond in a series of chlorine oxides was investigated. The simplest ClO radical species as well as the two isomeric structures XClO/ClOX for each X=H, Cl and O were studied using the PW91, TPSS, B3LYP, PBE0, M06, M06-2X, BMK and B2PLYP functionals. Geometry optimizations as well as reaction enthalpies and enthalpies of formation for each species were calculated using Pople basis sets and the (aug)-cc-pVnZ Dunning sets, with n=2-6. For the calculation of enthalpies of formation, atomization as well as isodesmic reactions were employed. Both the precision of the methods with respect to the increase of the basis sets, as well as their accuracy, were gauged by comparing the results with the more accurate CCSD(T) calculations, performed using the same basis sets as for the DFT methods. The results obtained employing composite chemical methods (G4, CBS-QB3 and W1BD) were also used for the comparisons, as well as the experimental results when they are available. The results obtained show that error compensation is the key for successful description of molecular properties (geometries and energies) by carefully selecting method and basis sets. In general, expansion of the one-electron basis set to the limit of completeness does not improve results at the DFT level, but just the opposite. The enthalpies of formation calculated at the CCSD(T)/aug-cc-pV6Z for the species considered are generally in agreement with experimental determinations, and the most accurate derived theoretically up to present. Different sources of error in the calculations are discussed in detail. Logos on environmental sustainability could consist of an effective strategy to provide consumers with accurate, understandable and trustworthy information to encourage them to buy environmentally sustainable wines. However, the large number of different logos indicating environmental sustainability available in the market raises the question ofwhether their associated messages are successfully conveyed to consumers. In this context, the aim of the present exploratory study was to investigate how Burgundy wine consumers perceive a series of logos indicating environmental sustainability in wine production. Fourteen logos available in the French market were selected: three logos being specific to wine and eleven nonspecific. The logos were presented to 127 wine consumers from Dijon area (France), following an incomplete balanced block design. For each logo, participants had to answer the question: “What does a bottle of wine with this logo suggest to you?”. Responses were qualitatively analyzed and grouped into different categories. Chi-square tests and Correspondence analysiswere used to identify the relationship among logos and categories. Results showed large differences in how consumers perceived the logos. Biodyvin, the former European AB and the French AB organic logos were the logos that most successfully conveyed their messages, being strongly associated to organic wine. Most logos did not communicate a message related to environmental sustainability, which reaffirms the need to provide consumers with adequate information on environmental sustainability and to conduct further research on this subject. The aim of this study was to analyze in detail the changes produced by the enzymatic interesterification of BT/RBO blends (beef tallow/rice bran oil) at different proportions, as an alternative for production of trans free fats. It was observed that the increase in the oil content produced a range of the content of saturated fatty acids from 20 to 38% in the blend and a range of polyunsaturated fatty acids from 35 to 18%. In TAG composition, the SUU and SSS type (S: saturated fatty acid; U: unsaturated fatty acid) increased in concentration as a result of interesterification process and this effect was more noticeable when the blend was richer in oil, arriving at 19 and 4% respectively in 50:50 BT/RBO blend. These variations in composition greatly improved the plastic range of BT. The process studied produced new trans free fatty materials with improving suitability as food ingredients. El glicerol es un subproducto inevitable de la producción de biodiésel. Una gran cantidad de este producto es generado durante la transesterificación de grasas y aceites con alcohol. El exceso de oferta y la limitada demanda de este producto afectan directamente los costos de producción del biodiésel. Sin embargo, el glicerol puede ser convertido a productos de más alto valor agregado mediante procesos puramente químicos o mediante biotransformaciones. Los bioprocesos verdes, tales como las reacciones biocatalíticas, son capaces de modificar selectivamente sustratos polifuncionales debido a su alta eficiencia catalítica, selectividad inherente y fácil o nulo procesamiento downstream. Además, las biotransformaciones se realizan bajo condiciones suaves con menores requerimientos energéticos, ofreciendo tecnologías medioambientalmente limpias. Las nuevas tendencias en bioconversiones marcan el acoplamiento de procesos biocatalíticos enzimáticos a procesos fermentativos como una solución de gran potencial en la biosíntesis secuencial de productos biotecnológicos. En esta tesis se propuso una ruta biosintética mixta novedosa para la síntesis de ácido L-láctico a partir de glicerol crudo, subproducto de la industria del biodiésel. Nuestro enfoque incluyó el uso de biocatalizadores inmovilizados microbiológicos y enzimáticos mediante estrategias que aportaron estabilidad y la posibilidad de reutilización en aras de conversiones más verdes y económicas. Las enzimas catalizan un importante número de reacciones, a menudo con regio o estero selectividad muy precisa, hacienda posible una variedad de procesos catalíticos que no serían posibles o son impracticables por síntesis convencional. El uso aplicado de enzimas en biocatálisis se afianza año a año con la aparición de nuevos y mejores biocatalizadores que facilitan la producción de moléculas de interés biotecnológico. Cada vez son más comunes los procesos en los que se combinan varias enzimas actuando secuencialmente. El uso de sistemas enzimáticos acoplados puede mejorar la cinética de las reacciones, disminuir la pérdida de intermediarios o evitar la inactivación o inhibición de una de las enzimas del sistema por eliminación rápida de algún intermediario inactivante. Sin embargo, la naturaleza soluble de las enzimas presenta restricciones específicas para su uso aplicado: pobre estabilidad, dificultad de separación, contaminación de productos y reuso limitado. La inmovilización de enzimas es usada comúnmente para mejorar estas limitaciones ya que muchas veces provee efectos estabilizantes, facilita su separación del medio de reacción y su reuso. En este trabajo de Maestría hemos abordado el estudio de sistemas enzimáticos que acoplados funcionan para producir moléculas de interés industrial. Nuestros estudios incluyeron la inmovilización de todas las enzimas de las vías sintéticas seleccionadas, caracterización y utilización in vitro. Muchos de los biocatalizadores utilizados debieron ser expresados de manera recombinante y purificados parcialmente previamente a su utilización. Las estrategias de inmovilización utilizadas incluyeron interacción covalente y atrapamiento físico. Además de utilizar agarosa como soporte de inmovilización se planteó el uso de un nuevo soporte para la unión de enzimas: las nanopartículas de sílica biomimética. Para mejorar las propiedades de este soporte para la unión de proteínas hemos propuesto una estrategia de síntesis novedosa que incluye moldes proteicos y genera nanopartículas más pequeñas y más homogéneas en tamaño. Para todas las enzimas con las que se trabajó se lograron preparar inmovilizados activos, en algunos casos muy estables, alcanzándose factores de estabilización sin precedentes en la literatura. Nuestros estudios permitieron el acoplamiento de 5 enzimas que en una secuencia no natural funcionaron en la producción enantioselectiva del L-ácido láctico a partir de mezclas racémicas de Alanina. Además, la co-inmovilización en un mismo soporte de dos enzimas que en la naturaleza funcionan secuencialmente en la vía de síntesis del antibiótico butirosina B, permitió la producción del antibiótico in vitro a partir de un Tesis de Maestría en Química Lic. Erienne Jackson sustrato alternativo que debimos sintetizar químicamente. Asimismo, hemos sentado las bases para la inmovilización de uno de los sistemas enzimáticos más complejos de la naturaleza, las poliquétido sintasas demostrando mediante el seguimiento de actividades parciales su estabilización frente al complejo no inmovilizado. En todos estos sistemas hemos demostrado que un diseño adecuado de los procesos de inmovilización facilita el acoplamiento de múltiples enzimas trabajando en tándem para la síntesis in vitro de moléculas de interés biotecnológico. In response to the growing use of consumers for sensory product characterisation, methodological research contributing to development of best practise guidelines is ongoing. We focus here on concurrent elicitation of hedonic and sensory product characterisation by check-all-that-apply (CATA) questions. Jaeger et al. (2013b) reported that CATA questions only caused weak and transient bias of co-elicited hedonic scores. In the current research six studies were conducted, in which more than 700 consumers took part. Five product categories were tested (rice crackers, lite bread, cheese, kiwifruit, black currant drinks) with 4–7 samples per study. In none of these studies was evidence obtained suggesting bias of hedonic scores and it is now possible to conclude with greater certainty that co-elicitation of hedonic scores and product attribute information using CATA questions is unlikely to bias hedonic scores. A second result of the current research was that the use of designs that rotate presentation order of CATA terms was not associated with hedonic bias, and neither was the use of the forced Yes–No CATA question format. In future research, in light of a strong dominance of positive CATA terms used in these studies, we recommend studying more thoroughly the influence of positive/negative/neutral words in CATA lists as a possible source of hedonic bias. An exploratory component to this research suggested that consumers perceive the concurrent elicitation of hedonic and CATA responses as easy, but that too many samples may make the task tedious.
